This article focuses on the current state-of-the-art in the area of cellular and molecular biotechnology for over-production of clinically relevant therapeutic growth factors and how the technology can be used for the treatment of osteoarthritis (OA). Transfected and irradiated protein packaging cell lines may be used as “cellular factories” for large-scale production of therapeutic proteins and pro-anabolic growth factors, particularly in the context of cartilage matrix regeneration. We discuss the potential for new innovations in regenerative medicine for degenerative diseases of synovial joints using mammalian protein production platforms, specifically protein packaging cell lines, for over-producing growth factors for cartilage tissue regeneration and give recent examples. Mammalian protein production platforms that incorporate protein packaging cell lines are superior to bacterial expression systems and are likely to have a significant impact on the development of new biological therapies for treating focal cartilage defects and more generally for the treatment of degenerative joint diseases such as OA.

We discuss how the currently available tools and emerging technologies can be used for the regenerative treatment of osteoarthritis (OA). Transfected protein packaging cell lines such as GP-293 cells may be used as “cellular factories” for large-scale production of therapeutic proteins and pro-anabolic growth factors, particularly in the context of cartilage regeneration. However, when irradiated with gamma or x-rays, these cells lose their capacity for replication, which actually makes them safe for use as a live cell component of intra-articular injections. This innovation is already here, in the form of TissueGene-C, a new biological drug which consists of normal allogeneic primary chondrocytes combined with transduced GP2-293 cells that overexpress the growth factor transforming growth factor β1 (TGF-β1). TissueGene-C has revolutionized the concept of cell therapy, allowing drug companies to develop live cells as biological drug delivery systems for direct intra-articular injection of growth factors whose half-lives are in the order of minutes. Therefore, in this paper, we discuss the potential for new innovations in regenerative medicine for degenerative diseases of synovial joints using mammalian protein production platforms, specifically protein packaging cell lines, for over-producing growth factors for cartilage tissue regeneration and give recent examples. Mammalian protein production platforms that incorporate protein packaging eukaryotic cell lines are superior to prokaryotic bacterial expression systems and are likely to have a significant impact on the development of new humanized biological growth factor therapies for treating focal cartilage defects and more generally for the treatment of degenerative joint diseases such as OA, especially when injected directly into the joint.

Background. Escherichia coli commonly causes catheter-related bloodstream infection (C-RBSI) in specific populations. The differential time to positivity (DTTP) technique is the recommended conservative procedure for diagnosing C-RBSIs. Methods. We conducted a retrospective study of episodes in which E. coli was isolated from catheter lumens obtained using the DTTP technique. We analysed microbiological and clinical data based on the DTTP technique as either catheter colonization, C-RBSI, or non-C-RBSI. Results. We included 89 catheter blood cultures classified as follows: catheter colonization, 33.7%; C-RSBI, 9.0%; and non-C-RBSI, 57,3%. Catheter withdrawal was 15.7% without positive catheter-tip cultures. We found no statistically significant differences in catheter type, antibiotic treatment, or clinical outcome among groups, except for the frequency of catheter lock therapy or in the frequency of successful treatment. Mortality was associated with C-RBSI in only 1 patient. Conclusion. E. coli bacteremia diagnosed by the DTTP technique was non-catheter related in most patients. As the majority of the catheters were retained, E. coli bacteremia could not be microbiologically confirmed or ruled out as catheter related by catheter-tip culture. Future studies are needed to assess the profitability of the DTTP technique for diagnosing E. coli C-RBSIs..

Agro-industrial activities generate significant amounts of organic waste and a variety of effluents, thus posing environmental challenges. Viticulture in Argentina, covering 204,847 ha in 2023, faces water scarcity as a limiting factor conditioning its production. This industry produces large volumes of grape marc, sediments, and stalks, which can be valorized into products like alcohol, tartaric acid, and compost. However, these valorization processes generate effluents with high organic load and salinity, further stressing water resources. This study explores the potential of utilizing these effluents to cultivate plant biomass in arid regions (sorghum or perennial pasture), which could serve as bioenergy, animal feed, or composting co-substrates, contributing to circular bioeconomy principles. Sorghum plots showed better establishment and higher dry biomass yield (32.6 Tn/ha) compared to pasture (6.5 Tn/ha). Sorghum demonstrated better tolerance to saline soils and high salinity effluents, aligning with previous studies. Although pasture had lower biomass yield, it was more efficient in nutrient uptake, concentrating more NPK, ash, and soluble salts. Sorghum's higher yield compensates for its lower nutrient concentration. For biomass production, sorghum is preferable, but if nutrient capture from effluents is prioritized, summer polyphytic pastures are more suitable. These results suggest that the final selection between plant biomass alternatives highly depends on whether the goal is biomass generation or nutrient capture.

Glioblastoma multiforme (GBM) is the most frequent and aggressive primary brain tumor. GBM is formed by a very heterogeneous astrocyte population, neurons, neovascularization and infiltrating myeloid cells (microglia and monocyte derived macrophages). The IQGAP1 scaffold protein interacts with components of the cytoskeleton, cell adhesion molecules, and several signaling molecules to regulate cell morphology and motility, cell cycle and other cellular functions. IQGAP1 overexpression and delocalization has been observed in several tumors, suggesting a role for this protein in cell proliferation, transformation and invasion. IQGAP1 has been identified as a marker of amplifying cancer cells in GBMs. To determine the involvement of IQGAP1 in the onco-biology of GBM, we performed immunohistochemical confocal microscopic analysis of the IQGAP1 protein in human GBM tissue samples using cell type-specific markers. IQGAP1 immunostaining and subcellular localization was heterogeneous; the protein was located in the plasma membrane and, at variable levels, in nucleus and/or cytosol. Moreover, IQGAP1 positive staining was found in podosome/invadopodia-like structures. IQGAP1⁺ staining was observed in neurons (Map2⁺ cells), in cancer stem cells (CSC; nestin⁺) and in several macrophages (CD31⁺ or Iba1⁺). Our results indicate that the IQGAP1 protein is involved in normal cell physiology as well as oncologic processes.
