Current proposals on the temporal sequence in the processing of emotional visual stimuli are partially incompatible with growing empirical data. In the majority of them, the initial evaluation structures (IES) postulated to be in charge of the earliest detection of emotional stimuli (i.e., salient for the individual), are high order structures (i.e., those receiving visual inputs after several synapses). Thus, their latency of response cannot account for the first visual cortex response to emotional stimuli (peaking 80 ms in humans). Additionally, these proposed structures lack the necessary infrastructure to locally analyze the visual features of the stimulus (shape, color, motion, etc.) that define a stimulus as emotional. In particular, the amygdala is defended as the cornerstone IES also in humans, and cortical areas such as the ventral prefrontal cortex or the insula have been proposed as well to intervene in this initial evaluation process. The present review describes several first-order brain structures (i.e., receiving visual inputs after one synapsis), and second order structures (two synapses) that may complement the former, that accomplish with both prerequisites: presenting response latencies compatible with the observed activity at the visual cortex and possessing the necessary architecture to rudimentarily analyze in situ relevant features of the visual stimulation. The visual thalamus, and particularly the lateral geniculate nucleus (LGN), a first-order thalamic nucleus that actively processes visual information, is a good candidate to be the core IES, with the complementary action of the thalamic reticular nucleus (TRN). This LGN-TRN tandem could be supported, also in an ascending, initial evaluation phase, by the pulvinar, a second order thalamic structure, and first-order extra-thalamic nuclei (superior colliculus and certain nuclei of pretectum and the accessory optic system). In sum, the visual thalamus, scarcely studied in relation to emotional processing, is a serious candidate to be the missing link in early emotional evaluation and, in any case, is worth exploring in future research.

In this study we discuss critical issues in modelling the structure and function of lymph nodes (LNs), with emphasis on how LN physiology is related to its multi-scale structural organization. In addition to macroscopic domains such as B-cell follicles and the T cell zone, there are vascular networks which play a key role in the delivery of information to the inner parts of the LN, i.e., the conduit and blood microvascular networks. We propose object-oriented computational algorithms to model the 3D geometry of the fibroblastic reticular cell (FRC) network and the microvasculature. Assuming that a conduit cylinder is densely packed with collagen fibers, the computational flow study predicted that the diffusion should be a dominating process in mass transport than convective flow. The geometry models are used to analyze the lymph flow properties through the conduit network in unperturbed- and damaged states of the LN. The analysis predicts that elimination of up to 60–90 % of edges is required to stop the lymph flux. This result suggests a high degree of functional robustness of the network.

Abstract For over a century the somatic gene mutation theory of cancer has been a scientific orthodoxy. Recent failures of causal explanations using this theory and lack of significant progress in addressing the cancer problem medically have led to a new competition of ideas about just what cancer is. This essay presents an alternative view of cancer as a developmental process gone wrong. More particularly, cancer is a break down in the autopoietic process of organ maintenance and the multi-cellular coordination of tissues. Breast cancer is viewed through a systems science perspective as an example of the importance of framing one’s theoretical assumptions before making empirical judgments. Finally, new understanding of the histoarchitecture of the interstitium is presented as a first principle of cancer: a process of cells coming from cells, invading the space between cells.

Mutations in KRT1 (keratin 1) or KRT10 (keratin 10) underlie a spectrum of diseases known as keratinopathic ichthyoses. Epidermolytic ichthyosis (EI) is caused by heterozygous missense mutations in the genes KRT1 or KRT10, mutations in the gene KRT2 (keratin 2) lead to superficial epidermolytic ichthyosis, and congenital reticular ichthyosiform erythroderma is caused by frameshift mutations in the genes KRT10 or KRT1, which lead to the phenomenon of revertant mosaicism. Epidermolytic ichthyosis is also present in a mosaic pattern known as epidermolytic (acantholytic) nevus, isolated or diffuse. In the latter case, gonadic involvement is possible, leading to a rare pedigree in which a parent with diffuse epidermolytic nevus (linear EI) gives birth to a child affected by EI. We present here an update on the phenotypic presentations of keratinopathic ichthyoses and their molecular mechanisms.

As our understanding of the thalamocortical system deepens, the questions we face become more complex. Their investigation requires the adoption of novel experimental approaches complemented with increasingly sophisticated computational modeling. In this review, we take stock of current data and knowledge about the circuitry of the somatosensory thalamocortical loop in rodents, discussing common principles across modalities and species whenever appropriate. We review the different levels of organization, including the cells, synapses, neuroanatomy, and network connectivity. We provide a complete overview of this system that should be accessible for newcomers to this field while nevertheless being comprehensive enough to serve as a reference for seasoned neuroscientists and computational modelers studying the thalamocortical system. We further highlight key gaps in data and knowledge that constitute pressing targets for future experimental work. Filling these gaps would provide invaluable information for systematically unveiling how this system supports behavioral and cognitive processes.
