COVID-19 is one of the deadliest respiratory diseases, and its emergence caught the pharmaceutical industry off guard. While vaccines have been rapidly developed, treatment options for infected people remain scarce, and COVID-19 poses a substantial global threat. This study presents a novel workflow to predict robust druggable targets against emerging RNA viruses using metabolic networks and information of the viral structure and its genome sequence. For this purpose, we implemented pymCADRE and PREDICATE to create tissue-specific metabolic models, construct viral biomass functions and predict host-based antiviral targets from more than one genome. We observed that pymCADRE reduces the computational time of flux variability analysis for internal optimizations. We applied these tools to create a new metabolic network of primary bronchial epithelial cells infected with SARS-CoV-2 and identified enzymatic reactions with inhibitory effects. The most promising reported targets were from the purine metabolism, while targeting the pyrimidine and carbohydrate metabolisms seemed to be promising approaches to enhance viral inhibition. Finally, we computationally tested the robustness of our targets in all known variants of concern, verifying our targets’ inhibitory effects. Since laboratory tests are time-consuming and involve complex readouts to track processes, our workflow focuses on metabolic fluxes within infected cells and is applicable for rapid hypothesis-driven identification of potentially exploitable antivirals concerning various viruses and host cell types.

Sustainable soil resource management requires detailed knowledge of soil pollution sources and their share in total pollution level. Spatial pollution caused by the total cumulative atmospheric deposition remains largely unknown, as the biggest pollutant emissions occurred in XIX/XX centuries. The use of ombrotrophic peatlands that are specific isolated ecosystems fed only through atmospheric deposition may serve as its natural archives. Accumulation of Cd and Pb from atmospheric deposition in undisturbed soil layers in relation to their total deposited cumulative loads recorded in the ombrotrophic peat bog was exemplified in the Izera Mountains. area historically heavily affected with the transboundary long-range transmission of pollutants from Germany, Czech Republic and Poland. Balance of deposited Cd and Pb loads in soil in relation to the total cumulative deposition determined from peat records, showed 30% depletion of Cd load in the soil profile due to washout of mobile phases, while that of Pb practically did not decline. The deposited element accumulation and release/depletion in undisturbed soil profiles can thus be quantified in relation to the total cumulative load of atmospheric deposition. This shows a new prospective application of peat bog records as monitors of total cumulative loads of trace elements supplied to soils from atmospheric deposition.

Measures in the SARS-CoV-2 pandemic were based on rough ideas regarding transmission routes of pathogens. Quantified models of physical transmission routes are mostly lacking, a gap to be filled. Vaccines and medicines, important, are not studied here. We first survey main routes, from primary production in the alveoli and intestines to emissions, environmental routes, to exposure and alveolar infection. Next, specific routes are modelled, mostly at a preliminary state, open to systematic improvement. Starting from a standardized emitter, modelling results show extreme differences in potential exposure, in a range covering up to 4 orders of magnitude. The outcomes are pathogen-specific, already different between SARS-CoV-2 and influenza. Extreme exposures may result in smaller spaces; with lower ventilation rates; with a high density of emitting persons per m3; who stay there for several hours; and visitors staying more than a few minutes. In spaces where a build-up of concentrations is low, exposures are low, lowest in open air situations. A main conclusion for the next pandemic is that a quantified model can give strong guidance on where measures are primarily due. For SARS-CoV-2, ventilation can be improved short-term. Longer-term, effective ventilation rules and adaptation of buildings may reduce high exposures substantially.

The astro-structures with the shape of comet and semi-circle, are found in the electronegative inductively coupled plasmas (ICPs), e.g., Ar/O2 and Ar/SF6. They are formed by means of the self-coagulation theory. This theory is built and based on two-dimensional fluid model simulation dynamic data of the above ICP sources. Concretely, quasi-Helmholtz equation is constructed through the free diffusion transport component and a novel negative chemical source term. The absolute value of recombination rate is higher than the attachment rate and so the net source of simulated anion is negative. This is quite a new chemical phenomenon in the electronegative and gaseous discharging plasmas when compared to the previously reported normal positive chemical source that will generate ambi-polar diffusion potential. In electronegative plasma, the self-coagulation is ambi-polar to sustain the electrical neutrality. Besides, when the anions are accelerated by ambi-polar potential before self-coagulated, the self-coagulation is again advective which leads to the formation of blue sheath. At the ambi-polar self-coagulation of high electronegativity, the electrons dynamics are relatively decoupled from ions and meanwhile they have their own relatively weak, spontaneous, and monopolar self-coagulation in the periphery of astro-structure. At the ambi-polar self-coagulation, both the anion and cation are individually treated as mass point models that carry charge (point charge models, more precisely). When the charge amounts of two individual astr-structures, e.g., semi-circle SF3+ and Ar+, are equal, the coagulated profile of heavy species SF3+ collapses by the expelling effect of Coulomb force that exists between the two point-charges. This is an anti-collective behavior of astro-structure, instead of the collective interaction of plasma, since the collapse of coagulated heavy species creates spatially dispersed charge density that cannot be shielded. The simulation shows that the lighter the species, the easier it self-coagulates, determined by the inertial effect of density quantity. The discharge picture of electronegative plasma is combined by a dynamic continuum transport (e.g., forming the ellipse background) and the mechanical balance contained in the static self-coagulation process (proven by dimensional analysis of continuity equation). The popular astro-structure of electronegative ICP source awaits urgently for the validation of experiment.

This paper proposes a paradigm shift in AI governance by integrating Indigenous knowledge systems to foster inclusivity and cultural sensitivity. Traditional AI governance frameworks are primarily Western-centric, often overlooking the community-oriented values and ethical considerations essential to Indigenous cultures. This study highlights principles such as Kaitiakitanga from Māori culture, which emphasizes guardianship and environmental stewardship, and Hózhó from Navajo philosophy, which stresses harmony and balance. By engaging Indigenous leaders and knowledge holders throughout the AI lifecycle, this approach ensures technology aligns with community values, addressing critical issues like data sovereignty, ethical technology use, and cultural sensitivity. This paper underscores the necessity of relational accountability in AI governance, which includes prioritizing the health of ecosystems alongside community welfare. This model advocates for an AI framework that not only rectifies historical marginalization but also empowers Indigenous communities to shape the technologies affecting their lives. Furthermore, integrating Indigenous perspectives strengthens the ethical foundation of AI systems, advancing sustainability, social justice, and cultural responsiveness in technology applications. The proposed framework illustrates how Indigenous ethical concepts can transform AI governance by aligning technological progress with values of ecological stewardship and community welfare, creating an AI landscape that respects and reflects diverse cultural contexts. Ultimately, this research demonstrates that Indigenous knowledge systems can significantly enhance AI’s effectiveness and equity, paving the way for more sustainable and culturally attuned technological solutions.

We introduce the linear subgroup of volume-preserving diffeomorphism as the underlying symmetry to construct an action within the effective field theory framework and the Schwinger-Keldish formalism. The formulated action is posited to effectively incorporate dissipative effects into the Navier-Stokes equations.
