This study investigates the preparation, characterization, and catalytic activity of a novel palladium-doped copper molybdate polyoxometalate (Pd@CuMoO₄). Using a mere 19 ppm of palladium, Pd@CuMoO4 demonstrated excellent yields in both Suzuki-Miyaura and Sonogashira-Hagiwara coupling reactions of aryl halides with boronic acids and alkynes, respectively. The synergistic effects between the different metals in this catalyst were confirmed to be responsible for its high catalytic performance. Additionally, Pd@CuMoO₄ was successfully recovered and recycled multiple times in the Suzuki-Miyaura reaction with minimal loss of activity. The structural integrity of the reused catalyst was evaluated through various analytical techniques.

Aryl chlorides and bromides in the presence of NHC palladium hydroxo dimers of the type [{Pd(µ-OH)Cl(NHC)}2] (where NHC = IPr, SIPr, IMes, SIMes) undergo an efficient and selective Sonogashira cross-coupling with terminal acetylenes. For the coupling of 4-chlorotoluene with phenylacetylene TON = 560000 was observed. The procedure enables high-throughput and selective synthesis of a broad spectrum of unsymmetrically substituted 1,2-diarylacetylenes. Nearly a quantitative amount of HCl was detected within cross-coupling products. Water plays the role of the base and is regenerated in the process so that it does not have to be used in stoichiometric amounts (relative to the limiting reactant). Generation of the active Pd(0) complex proceeds via ethanol oxidation.

Cysteine plays a crucial role in the development of an efficient copper-catalyst system, where its thiol group serves as a strong anchoring site for metal coordination. By immobilizing copper onto cysteine-modified, polydopamine-coated magnetite nanoparticles, this advanced catalytic platform exhibits exceptional stability and catalytic activity. Chemical modification of the polydopamine (PDA) surface with cysteine enhances copper salt immobilization, leading to the formation of the Fe₃O₄@PDA-Cys@Cu nanostructure. This system was evaluated in palladium-free, copper-catalyzed Sonogashira coupling reactions, effectively catalyzing the coupling of terminal acetylenes with aryl halides. Additionally, the Fe₃O₄@PDA-Cys@Cu platform was employed in click reactions, confirming the enhanced catalytic efficiency due to increased copper content. The reusability of the platform was further investigated, demonstrating improved performance, especially in recyclability tests in click reaction, making it a promising candidate for sustainable heterogeneous catalysis.

In the frame of our diversity-oriented research on multitarget small molecule anticancer agents, utilizing convergent synthetic sequences terminated by Sonogashira coupling reactions, a preliminary selection of representative alkyne-tethered vindoline hybrids was synthesized. The novel hybrids with additional pharmacophoric fragments of well-documented anticancer agents, including FDA-approved tyrosine-kinase inhibitors (imatinib and erlotinib) or ferrocene or chalcone units, were evaluated for their antiproliferative activity on malignant cell lines MDA-MB-231 (triple negative breast cancer), A2780 (ovarian cancer), HeLa (human cervical cancer) and SH-SY5Y (neuroblastoma) as well as human embryonal lung fibroblast MRC-5 served as reference non-malignant cell line for the assessment of the therapeutic window of the tested hybrids. The biological assays identified a trimethoxyphenyl-containing chalcone-vindoline hybrid (36) as a promising lead compound exhibiting submicromolar activity on A2780 cells with a marked therapeutic window. These results suggest that ovarian cancer might be considered as a prioritized target of treatment with 36.

Bimetallic PdCu nanoparticles with different Pd:Cu ratios and morphologies can be synthesized and immobilized on a variety of support materials. Accordingly, PdCu nanoparticles can be efficiently applied as heterogeneous catalysts in a large number of organic transformations including C-C coupling and cross-coupling reactions. As related to their favorable electronic and structural interactions, the catalytic performance of PdCu bimetallic nanoparticles may be superior to the monometallic species. The heterogeneous catalysts can be recovered and reused and the presence of copper tends to reduce the cost of the expensive Pd catalyst, which would be beneficial for industrial applications.

Our ongoing research focuses on developing imipridone derivatives capable of complete and selective eradication of cancer cells after relatively short treatment. We have synthetized systematically designed novel hybrids and evaluated their antiproliferative activity on PANC-1 and Fadu cell lines. We have also conducted preliminary studies on the mechanism including colony formation as well as dose-response tests in HEK293T wild-type (WT) and HEK293T CLPP-/- cells. Following gradual structural fine-tuning based on high throughput screening we identified two imipridone hybrids as the most potent derivatives. Their unique substitution pattern includes N-methylated propargylamine and ferrocenyl/phenyltriazole moieties on the benzyl groups attached to opposite sides of the imipridone core. The compounds with IC50 values similar to those of ONC201 were found capable of complete eradication of cancer cells at ca. 4 M, while ONC201 treatment at even higher concentrations left 30-50% of viable cells behind. Both compounds were found to exert equal activity in WT and CLPP-/- HEK293T cells, indicating a ClpP-independent mechanism. Further development is needed to improve the tumor selectivity of the two potent imipridone derivatives in order to develop novel drug candidates that evade resistance and can be applied in a sufficiently broad therapeutic window.
