Fungal endophytes have been found in all plants surveyed to date, yet for many fungi the function of endophytism is still unknown. The Foraging Ascomycete Hypothesis (FAH) proposes that saprotrophic fungi utilize an endophytic stage in leaves to modify dispersal. Under this hypothesis, leaves can provide food and water during time of environmental scarcity and they can transport the fungi to other substrates upon dehiscence. If the FAH is accurate, then some endophytes should have the ability to colonize saprobic substrates directly from a leaf-endophyte stage, though this has been little studied. To assess this ability, twelve surface-sterilized leaves of a tropical tree (Nectandra lineatifolia Mez) were placed directly on wood and incubated for six weeks. Fungi from the wood were subsequently cultured and identified by ITS sequences or morphology. 477 fungal isolates comprising 26 OTUs were cultured from the wood, the majority of which belong to saprotrophic genera (70.8% of OTUs, 82.3% of isolates). The mean OTU richness per leaf was 5.67. The term viaphyte (literally, “by way of plant”) is introduced and defined as fungi that colonize living leaves as endophytes and use the leaves to transfer to another substrate, such as wood, when the leaves dehisce. These results strengthen the Foraging Ascomycete Hypothesis and expose the possibility that viaphytism plays a significant role in the dispersal of fungal saprotrophs.

The urinary tract, once considered sterile, is now understood to host a diverse community of microorganisms known as the urinary microbiome. This microbiome, which is made up of bacteria, fungi, and viruses, is important for preserving urological health and has been linked to the pathogenesis of a number of urinary tract disorders. In this review, we offer a consolidated overview of the urinary tract bacterial population, including its composition, diversity, and factors influencing its dynamics. We explore its involvement in urinary tract infections, interstitial cystitis/bladder pain syndrome (IC/BPS), urinary stone formation, and other urological diseases, highlighting the importance of understanding its correlation with disease pathology for the development of therapeutic strategies. Additionally, we explore its role in autoimmune diseases of the urinary tract and the potential of leveraging the urinary microbiome for targeted interventions in the treatment of urinary tract infections. Longitudinal studies are emphasized for their ability to elucidate microbial dynamics, establish causality, and ensure consistency and reproducibility across research endeavors. These efforts underscore the necessity for continued research and multidisciplinary collaborations in this rapidly evolving field to advance our understanding and therapeutic strategies for urinary tract disorders.

The cellular cytoskeleton provides the cell with a mechanical rigidity which allows mechanical interaction between cells and the extracellular environment. The actin structure plays a key role in mechanical events like motility, or establishment of cell polarity. From the earliest stages of development, as represented by ex vivo expansion of naïve embryonic stem cells (ESCs), the critical mechanical role of the actin structure is becoming recognized as a vital cue for correct segregation and lineage control of cells and as a regulatory structure that controls several transcription factors. Naïve ESCs have a characteristic morphology and the ultrastructure that underlies this condition remains to be further investigated. Here, we investigate the 3D actin cytoskeleton of naïve mouse ESCs using super resolution optical reconstruction microscopy (STORM). We investigate the morphological, cytoskeletal and mechanical changes in cells cultured in 2i or Serum/LIF media reflecting a homogenous preimplantation cell state and a state that is closer to embarking on differentiation. STORM imaging showed that the peripheral actin structure undergoes a dramatic change between the two media conditions. We also detected micro-rheological differences in the cell periphery between the cells cultured in these two media correlating well with the observed nano-architecture of the ESCs in the two different culture conditions. These results pave the way for linking physical properties and cytoskeletal architecture to cell morphology during early development.

Tissue engineering has the potential for repairing large bone defects, which impose a heavy financial burden on the public health. However, difficulties with O₂ delivery to the cells residing in the interior of tissue engineering scaffolds make it challenging to grow artificial tissues of clinically-relevant sizes. This study uses image-based simulation in order to provide insight into how to better optimize the scaffold manufacturing parameters, and the culturing conditions, in order to resolve the O₂ bottleneck. To do this, high resolution 3D X-ray images of two common scaffold types (salt leached foam and non-woven fiber mesh) are fed into a Lattice Boltzmann Method fluid dynamics and reactive Lagrangian Scalar Tracking mass transfer solvers. The obtained findings indicate that the scaffolds should have maximal surface area-to-solid volume ratios, for higher chances of the molecular collisions with the cells. Furthermore, the cell culture media should be flown through the scaffold pores as fast as practically possible (without detaching or killing the cells). Finally, we have provided a parametric sweep that maps how the molecular transport within the scaffolds is affected by variations in rates of O₂ consumption by the cells. Ultimately, the results of this study are expected to benefit the computer-assisted design of tissue engineering scaffolds and culturing experiments.
