This study combines two methodologies—key indicators from the Sustainable Livelihoods Framework (SLF) and the Sustainability Assessment of Food and Agriculture Systems (SAFA) approach—to evaluate sustainability among 330 producers from three associations working within the recently recognized Amazonian Chakra system, designated as a Globally Important Agricultural Heritage System (GIAHS). The SAFA framework's four dimensions—Good Governance, Environmental Integrity, Economic Resilience, and Social Well-being—were used to assess sustainability, while the SLF considered key capitals such as financial, natural, and human capitals as critical for understanding livelihood outcomes. The discriminant analysis revealed significant differences across the associations, with Kallari and Wiñak performing better in financial management, environmental practices, and governance compared to Tsatsayaku, which exhibited weaker outcomes in these domains. Key indicators such as chakra income, price determination, and civic responsibility were the most effective in distinguishing the groups. The income analysis further highlights that Tsatsayaku reported the highest total income but relies heavily on non-Chakra sources, while Kallari and Wiñak showed stronger reliance on Chakra income, which aligns with their more sustainable agricultural practices. Per-capita income was higher for Tsatsayaku, but extreme poverty levels remained similar across all associations, underscoring the need for targeted financial interventions. Policy recommendations include promoting access to financial resources and strengthening participatory governance models to enhance sustainability outcomes across associations. For future research, combining the SAFA and SLF provides a comprehensive and robust methodology for assessing sustainability in agricultural systems, enabling a deeper understanding of the interactions between social, economic, and environmental factors in diverse contexts.

Prostate cancer, according to the World Health Organization (WHO), ranks as the fourth most prevalent type in the population, even though it is highly frequent when detected early and promptly treated, resulting in a low mortality rate. However, in some cases, despite adherence to proper clinical protocols, there is a progression to castration-resistant prostate cancer (CRPC). In these cases, as outlined in clinical guidelines such as those developed by the National Comprehensive Cancer Network (NCCN), one approach involves the utilization of the pharmaceutical agent abiraterone to implement androgen deprivation therapy (ADT), aimed at suppressing the production of hormones directly implicated in disease progression. Another noteworthy aspect is the limited literature on clinical protocols and their efficacy concerning the Amazonian indigenous population, which often exhibits unique genomic profiles, with descriptions of variants not yet documented in the literature. Building upon these considerations, this study aims to survey the variants identified in genes associated with the abiraterone signaling pathway, their frequencies in the Amazonian indigenous population, and the correlation of these frequencies with those observed in other global populations. The objective is to contribute to the refinement of clinical practices in the treatment of prostate cancer within this population.

In this work, nine fruits cultivated in the northern Amazon were studied: abiu, acerola, araçá, bacupari, biribá, caçarí, fruta-do-conde, graviola and taperebá, with the objective of carrying out a bromatological and nutritional study of the pulps of fruits studied. Of all of them, are the pulps of graviola (76.83 ± 0.02 kcal 100 g^-1) bacupari (76.83 ± 0.02 kcal 100 g^-1) and fruta-do-conde (46.66 ± 0.02 kcal 100 g^-1). Among the macronutrients, the high concentration of potassium stands out, especially in the graviola (541.16 ± 0.24 mg 100 g^-1) and the biribá (468.21 ± 0.13 mg 100 g^-1). Among the micronutrients, iron concentrations are representative for araçá pulp (3.04 ± 0.02 mg 100g-1), abiu is rich in zinc (3.71 ± 0.02 mg 100 g^-1) and manganese (6.61 ± 0.11 mg 100 g^-1). The presence of cobalt at the level of traces in some of the pulps studied stands out. The Pearson correlation coefficient was evaluated, as well as the statistical treatment by multivariate analysis PCA to establish the correlation between the variables studied.

Ucuùba fat is fat obtained from a plant found in South America, mainly in Amazonian Brazil. Due to its biocompatibility and bioactivity, the Ucuùba fat was used for production of ketoconazole-loaded nanostructured lipid carriers (NLC) in view of an application for the treatment of onychomycosis and other persistent fungal infections. The development and optimization of the Ucuùba fat based NLC were performed using a Box-Behnken design of experiment. The independent variables were surfactant concentration (% w/v), liquid lipids concentration (% w/v), solid lipids concentration (% w/v), while the outputs of interest were particle size, polydispersity index (PDI) and drug encapsulation efficiency (EE). The Ucuùba fat based NLC were produced and the process optimized determining a predictive mathematical model. Applying the model, two formulations with the pre-required particle size, i.e., 30 and 85 nm, were produced for further evaluation. The optimized formulations were characterized and showed a particle size in agreement to the predicted value, i.e. 33.6 nm and 74.6 nm, respectively. The optimized formulations were also characterized using multiple techniques in order to investigate the solid state of drug and excipients (DSC and XRD), particle morphology (TEM) and interactions between the formulation components (FTIR). Furthermore, particle size and surface charge of the formulations was studied during a one-month stability study and did not evidence any significative modification during storage.

The leaves and stems of Bauhinia guianensis Aubl. are used in traditional Amazonian phytotherapy for the treatment of pain and inflammation. This study investigates the anti-inflammatory and antinociceptive effects of hydroethanolic extracts from B. guianensis Aubl. leaves and stems (HELBg and HESBg, respectively) in in vivo models of inflammation and hyperalgesia. Danio rerio experimental animals were submitted to the acute inflammation test, induced by intraperitoneal (ip.) administration of carrageenan 20 μg / animal (abdominal edema), the groups were previously treated orally with saline solution 2 μl / animal (SS), Dimethyl sulfoxide 2 μl / animal (DMSO), Indomethacin 10 mg/kg, HELBg 100 mg/kg and HESBg 100 mg/kg, n = 12 per experimental group, to evaluate inhibition of edema and alterations histopathology of the liver, intestine and kidney of these animals. The antinociceptive effect was observed from the body curvature index and the behavioral responses of Danio rerio, after an experimental protocol for the induction of hyperalgesia, by ip. administration of 10 μl / animal of 2.5% acetic acid, the animals were previously orally treated with saline solution 2 μl/animal (SS), dimethyl sulfoxide 2 μl/animal (DMSO), morphine 2.5 mg/kg, HELBg 100 mg/kg and HESBg 100 mg/kg, n = 5 per experimental group. In carrageenan-induced edema, the group treated with HESBg inhibited edema formation over the 3 hours of the experiment. Maximum edema was inhibited by 54% (p < 0.05) when compared to the control group. Both HELBg and HESBg prevented body curvature index changes (t (df = 3 8) =, 6.96 and t (df = 3, 8) = 6.61, respectively, both p < 0.0001). In the behavioral parameters sensitive to antinociceptive pharmacological modulation, due to the abdominal constriction induced by acetic acid, the administration of HELBg and HESBg resulted in an improvement in swimming activity, by increasing the distance covered (F (df = 3, 16) = 6.50 and F (df = 3, 16)  = 7.72, respectively, both p < 0.0001), decrease in freezing time (F (df = 3, 16)  = 2.04 and F (df = 3, 16)  = 1.28, respectively, both p < 0.0059), increase in the number of ascents to the upper area of the tank (F (df = 3, 16)  = 33.02 and F (df = 3, 16)  = 35.62, respectively, both p < 0.0009 ) and decreased time spent in that area (F (df = 3, 16)  = 101.19 and F (df = 3, 16)  = 103.59, respectively, both p < 0.0038). It is reasonable to suppose that both extracts modulated the variations induced by carrageenan and acetic acid through the inhibition of prostaglandin biosynthesis, decreasing the release of inflammatory mediators and the sensitization of peripheral nociceptors and, consequently, the perception of pain. These results suggest that HELBg and HESBg have anti-inflammatory and antinociceptive activities, probably of peripheral origin and linked to the inhibition of prostaglandin biosynthesis.

The Amazonian shrub Myrciaria dubia (Kunth) McVaugh, known as "camu-camu", produces vitamin C-rich fruits of growing commercial interest. However, sustainable utilization requires assessment and protection of the genetic diversity of the available germplasm. We hypothesized that the ex situ M. dubia germplasm bank assembled from eight river basins across the Peruvian Amazon would harbor substantial genetic diversity and have a genetic population structure. This study aimed to (1) develop new polymorphic microsatellite markers, (2) characterize genetic diversity and validate the hypothesis in the ex situ germplasm, and (3) construct a core subset representing maximum allelic variability. Sixteen polymorphic microsatellite loci were developed using an enrichment approach. The evaluation of 336 genotypes from 43 accessions originating from eight river basins of the germplasm bank corroborated this hypothesis, revealing high gene diversity, with observed heterozygosity ranging from 0.468 to 0.644 and expected heterozygosity from 0.684 to 0.817 at the river basin level. Analysis of molecular variance showed a higher genetic variation within accessions and river basins, at 73% and 86%, respectively, than among accessions and river basins, at 27% and 14 %, respectively. Bayesian clustering detected the presence of ten genetic clusters, with several degrees of admixture among river basins. The Putumayo River basin showed a clear genetic homogeneity. A core collection of 84 genotypes was constructed, thus covering 86.7% of the global allelic diversity. These results have important implications for M. dubia conservation strategies and breeding programs, in demonstrating a need for genetic connectivity between populations but preserving unique genetic resources in isolated basins. These results validate the expected levels of diversity and population subdivision in a crop and stress the need to secure genetically diverse germplasms, underscoring the importance of thorough genetic characterization for ex situ germplasm management.

The Amazon region, with its vast biodiversity and rich natural resources, presents a unique opportunity for the development of sustainable polymeric composites reinforced with residues from both the timber and agro-extractive industries. This article explores the potential of these Amazonian residues, such as sawdust, wood shavings, and agro-industrial by-products like açaí seeds and Brazil nut shells, to enhance the mechanical, thermal, and environmental properties of polymer composites. By integrating these natural materials into polymer matrices, significant improvements in composite performance have been achieved, including increased tensile strength, thermal stability, and biodegradability. The study also highlights the environmental and economic benefits of utilizing these residues, promoting waste reduction and supporting a circular economy in the region. Through case studies and detailed analyses, the article demonstrates the feasibility and advantages of incorporating Amazonian residues into composites for a wide range of applications, from construction materials to consumer goods. This approach not only adds value to the by-products of the Amazonian industries but also contributes to the global effort towards sustainable material development.

The epidemic transmission of Zika virus (ZIKV) in Brazil has been identified as a cause of microcephaly and other neurological malformations in babies of ZIKV-infected women. This study provides a descriptive analysis, since the onset of symptoms to the delivery, of a cohort who were registered as having ZIKV infection in pregnancy, from November 2015 to December 2016. Suspected cases were registered at a referral center for infectious and tropical diseases in Manaus, in the Brazilian Amazonian region. A total of 834 women with suspected ZIKV in pregnancy were included, of whom 91.4% had confirmed pregnancy. Reverse-transcriptase polymerase chain reaction (RT-PCR) confirmed ZIKV infection in 42.2% of the cohort. In 35.2% of the cohort, ZIKV was the sole infection identified. Severe adverse pregnancy outcomes (abortion, stillbirth, or microcephaly) were observed in both RT-PCR ZIKV-positive (4.96%) and ZIKV-negative (2.15%) cases. Women with suspected ZIKV infection were much more likely to have adverse pregnancy outcomes if they were symptomatic during the first trimester of pregnancy (odds ratio 10.5; 95% confidence interval 4.0–27.0; p<0.001). Among pregnant women with suspected ZIKV infection, the occurrence of symptoms in the first trimester is associated with an especially high risk of severe adverse pregnancy outcomes.
