Tunable hydrogels have gained significant attention in the bioengineering field due to their designer preparation approach. Towards this end, gelatine stands out as a promising candidate owing to the desirable attributes, such as biocompatibility, ability to support cell adhesion and proliferation, biodegradability, along with being cost-effective. This study presents the preparation of a robust gelatine hydrogel employing sacran dialdehyde (SDA) as a natural crosslinker. The resulting SDA-crosslinked gelatine hydrogels (GSDA) display an optimal compressive stress of 0.15 MPa at 50% strain, five times higher as compared to pure gelatine hydrogel. As SDA crosslinking concentration is increased, swelling capacity of GSDA declines. Probing further with FTIR spectroscopy and SEM at the micron-scale unveiled a dual-crosslinking mechanism within the hydrogels. This mechanism encompasses both short- and long-range covalent crosslinking, along with thermo-reversible physical crosslinking, resulting in a significant enhancement of the load-bearing capacity of the fabricated hydrogels.

Antimicrobial resistance (AMR) has become a worldwide health problem. Consequently, there is a global interest in the study of the antimicrobial activity of natural compounds as an alternative to antibiotics. One of the possibilities studied during the last years is the use of bacteriophages and their derived proteins. More specifically, phage lytic proteins or endolysins are specific enzymes that cause cell lysis and can be easily produced and purified after overexpression in bacteria. However, one of the major disadvantages of these proteins is their sensitivity to some environmental conditions that can reduce their efficiency. The use of vesicles for endolysin encapsulation could help overcome this disadvantage, offering an additional protection to the protein and allowing its controlled release and long-term stability.

In recent years, demand for consumption of marine foods, and especially fish, has substantially increased worldwide. The majority of collagen available is sourced from mammalian-derived products. Although fish derived gelatine is a viable alternative to mammalian sourced gelatine, there are some challenges related to the use of fish gelatine including odour, colour, gelling and film forming properties as well as consistency in gelatine amino acid composition. Chemicals used for pre-treatment, as well as extraction conditions such as temperature and time, can influence the length of polypeptide chains that result and the functional properties of the gelatine. Compared to mammalian sources, gelatines derived from fish show notable differences in physical and chemical properties, and great care should be paid to optimization of the production process in order to obtain a product with the best properties for intended applications. The focus of this review is to explore the feasibility of producing gelatine sourced from marine processing by-products using different pre-treatment and extraction strategies with the aim of improving the techno-functional properties of the final product and improving the clean-label status of gelatines. The bioactivities of gelatine hydrolysates are also discussed.

Alginate-gelatin (Alg-Gel) hydrogels have been used experimentally but not clinically associated with mesenchymal stromal / stem cells (MSCs) to guide bone tissue formation. One of the main challenges for its clinical application is optimizing Alg-Gel stiffness to guide osteogenesis. In this study, we investigated how Alg-Gel stiffness could modulate the dental pulp stem cell (DPSCs) attachment, morphology, proliferation, and osteogenic differentiation, identifying the optimal condition to uncouple osteogenesis from the other cell behaviors. An array of Alg-Gel hydrogels was prepared by casting different percentages of Alg and Gel being crosslinked with 2 % CaCl2. We selected two hydrogels, one with 11± 1 kPa called “low” stiffness and one with 55 ± 3 kPa called “high” stiffness. Hydrogel analyses showed that the average swelling rates were 20 ± 3% for low and 35 ± 2% for high hydrogels. The degradation percentage was 47 ± 5% and 18 ± 2% for low and high hydrogels, respectively. Both hydrogel types showed homogeneous surface shape and pro-tein (Alg-Gel) interaction with CaCl2 as assessed by FTIR-ATR and XPS. Cell culture showed good adhesion of the DPSCs to the hydrogels and proliferation. Furthermore, better osteogenic activity was obtained with high-stiffness hydrogels. In summary, this study confirms the possibility of characterizing and optimizing the stiffness of alginate-gelatin gel to guide osteogenesis in vitro without altering other cellular properties of DPSCs.

This study aims to develop low-cost, eco-friendly and circular economy-compliant composite materials by creating three types of magnetorheological suspensions (MRSs) utilizing lard, carbonyl iron (CI) microparticles, and varying quantities of gelatin particles (GP). These MRSs serve as dielectric materials in cylindrical cells used to fabricate electric capacitors. The equivalent electrical capacitance (C) of these capacitors is measured under different magnetic flux densities (B≤160 mT) superimposed on a medium-frequency electric field (f = 1 kHz) over a period of 120 seconds. The results indicate that at high values of B, by increasing the GP content to 20 vol.% decreases the capacitance C up to about one order of magnitude compared to MRS without GP. From the measured data, the average values of capacitance Cm are derived, enabling the calculation of relative dielectric permittivities (ϵr′) and the dynamic viscosities (η) of the MRSs. It is demonstrated that ϵr′ and η can be adjusted by modifying the MRS composition and fine-tuned through the magnetic flux density B. A theoretical model based on the theory of dipolar approximations is used to show that ϵr′, η and the magnetodielectric effect can be coarsely adjusted through the composition of MRSs and finely adjusted through the values B of the magnetic flux density. The ability to fine-tune these properties highlights the versatility of these materials, making them suitable for applications in various industries, including electronics, automotive and aerospace.
