Direct electron transfer (DET)-type bioelectrocatalysis, which couples electrode reactions and catalytic functions of redox enzymes without any redox mediator, is one of the most intriguing subjects studied over the past decades in the field of bioelectrochemistry. In order to realize the DET-type bioelectrocatalysis and to improve the performance, nanostructures of the electrode surface have to be carefully tuned for each enzyme. In addition, enzymes can also be tuned by protein engineering approach for the DET-type reaction. This review summarizes the resent progresses in this field of the research, while taking into consideration of the importance of nanostructure of electrodes as well as redox enzymes. Described are basic concepts and theoretical aspects of DET-type bioelectrocatalysis, significance of nanostructures as scaffolds for DET-type reactions, protein engineering approached for DET-type reactions, and concepts and facts of bidirectional DET-type reactions, from a cross-disciplinary viewpoint.

(1) Background: Transcription factors (TFs) are main regulators of eukaryotic gene expression. The cooperative binding of at least two TFs to genomic DNA is the widespread mechanism of transcription regulation. Massive analysis of co-occurrence of overrepresented pairs of motifs (composite elements, CEs) for different target TFs studied in ChIP-seq experiments can clarify the mechanisms of TF cooperation. (2) Methods: We focused on homotypic CEs representing the same motif models and considered only CEs with spacers. We improved the capability of the Motifs Co-Occurrence Tool (MCOT) to predict asymmetric homotypic CEs in which one of par-ticipating motifs has higher conservation than the other. We categorized the target TFs from the data of M. musculus ChIP-seq and A. thaliana ChIP-seq/DAP-seq according to the structure of DNA-binding domains (DBDs) into classes. (3) Results: For all TF classes the predominance of asymmetric CEs over both symmetric and those with no enrichment in both directions was re-vealed. Target TFs from classes Basic helix-loop-helix factors (bHLH) and Basic leucine zipper factors (bZIP) showed the highest fractions of datasets with asymmetric CEs. We showed that pioneer TFs, despite their DBD types, have a higher significance of asymmetry within homotyp-ic CEs compared to other TFs. (4) Conclusion: Asymmetry within homotypic CEs is a promising new feature decrypting the mechanisms of gene transcription regulation.

Abstract: A recent approach to radiotherapy for prostate cancer is the administration of high doses of radiation to the prostate while minimizing the risk of side effects. Thus, image-guided radiotherapy utilizes advanced imaging techniques and is a feasible strategy for increasing the radiation dose. New radioactive particles are another approach to achieving high doses and safe procedures. Prostate brachytherapy is currently considered as a combination therapy. Spacers are useful to protect adjacent organs, specifically the rectum, from excessive radiation exposure.

This paper presents an approach to the Kirchhoff-Love plate theory (KLPT) using the finite difference method (FDM). The KLPT covers the case of small deflections, and shear deformations are not considered. The FDM is an approximate method for solving problems described with differential equations. The FDM does not involve solving differential equations; equations are formulated with values at selected points of the structure. Generally in the case of KLPT, the finite difference approximations are derived based on the fourth-order polynomial hypothesis (FOPH) and second-order polynomial hypothesis (SOPH) for the deflection surface. The FOPH is made for the fourth and third derivative of the deflection surface while the SOPH is made for its second and first derivative; this leads to a 13-point stencil for the governing equation. In addition, the boundary conditions and not the governing equations are applied at the plate edges. In this paper, the FOPH was made for all of the derivatives of the deflection surface; this led to a 25-point stencil for the governing equation. Furthermore, additional nodes were introduced at plate edges and at positions of discontinuity (continuous supports/hinges, incorporated beams, stiffeners, brutal change of stiffness, etc.), the number of additional nodes corresponding to the number of boundary conditions at the node of interest. The introduction of additional nodes allowed us to apply the governing equations at the plate edges and to satisfy the boundary and continuity conditions. First-order analysis, second-order analysis, buckling analysis, and vibration analysis of plates were conducted with this model. Moreover, plates of varying thickness and plates with stiffeners were analyzed. Finally, a direct time integration method (DTIM) was presented. The FDM-based DTIM enabled the analysis of forced vibration of structures, with damping taken into account. In first-order, second-order, buckling, and vibration analyses of rectangular plates, the results obtained in this paper were in good agreement with those of well-established methods, and the accuracy was increased through a grid refinement.

Apraxia of speech is a persistent speech motor disorder that affects speech intelligibility. Studies on speech motor disorders with transcranial Direct Current Stimulation (tDCS) have been mostly directed to post-stroke aphasia. Only a few tDCS studies have focused on apraxia of speech or childhood apraxia of speech (CAS), and no study has investigated individuals with CAS and people with trisomy 21 (T21, Down Syndrome). This study examined the effects of tDCS combined with a motor learning task in developmental apraxia of speech co-existing with T21 (ReBEC RBR-5435x9). The accuracy of speech sound production of nonsense words (NSWs) during Rapid Syllable Transition Training (ReST) under 10 sessions of anodal tDCS (1.5 mA, 25 cm) over the Broca’s area with cathode over the contralateral region was compared to 10 sessions of sham-tDCS and 4 control sessions in a 20-year-old male individual with T21 presenting moderate-severe childhood apraxia of speech (CAS). The accuracy for NSWs production progressively improved (gain 40%) under tDCS only (sham-tDCS and control sessions showed <20% gain). A decrease in speech severity from moderate-severe to mild-moderate indicated transfer effects in speech production. The speech accuracy under tDCS was correlated with Wernicke’s area activation (P3 current source density), which in turn, was correlated with the activation of the left supramarginal gyrus and the Sylvian Parietal Temporal Junction. Repetitive bihemispheric tDCS paired with ReST may have facilitated the speech sound acquisition in a young adult with T21 and CAS, possibly by recruiting brain regions required for the phonological working memory.

Building a new power plant to address the growing demand for power due to population concentration in the metropolitan area is one of the world's major concerns. However, since a large power plant can not be located around the city due to burden of economic cost, building power plant outside metropolitan and cities is necessary. Therefore, new power generation facilities are promoting policies to provide distributed generator(DG) with a small capacity relatively near the metropolitan. When the DG (photovoltaic, wind farm, etc.) is connected with the grid using medium voltage direct current (MVDC) system, voltage sourced converter(VSC) should supply reactive power to the grid, because of fault ride through(FRT) operation in grid fault. If the voltage drop is severe, the converter should be disconnected from the grid immediately without supplying the reactive power, resulting in a considerable economic loss. In general, superconducting fault current limiter(SFCL) is introduced as a measure to enhance FRT capability. In this paper, we use trigger type SFCL which protects superconducting element and reduces low voltage. On the other hand, the active power unbalance of the DC-link and the DC voltage rise due to the reactive power supply of the grid-side converter. The rise of the DC voltage causes the P (active power), Q (reactive power) control of the converter to deviate, causing malfunction and damage of the DC equipment. Therefore, the rise of the DC voltage must be prevented. In this paper, we consider the suppression the DC voltage rising caused by the FRT operation through the active power tracking control (APTC).

This review focuses on the evidence for neurotherapeutics for Attention Deficit Hyperactivity Disorder (ADHD). EEG-Neurofeedback has been tested for about 45 years with latest meta-analyses of randomised controlled trials (RCT) showing small/medium effects compared to non-active controls only. Three small studies piloted neurofeedback of frontal activations in ADHD using functional magnetic resonance imaging or near-infrared spectroscopy, finding no superior effects over control conditions. Brain stimulation has been applied to ADHD using mostly repetitive transcranial magnetic and direct current stimulation (rTMS/tDCS). rTMS has shown mostly negative findings on improving cognition or symptoms. Meta-analyses of tDCS studies targeting mostly dorsolateral prefrontal cortex show small effects on cognitive improvements with only two out of three studies showing clinical improvements. Trigeminal nerve stimulation has shown to improve ADHD symptoms with medium effect in one RCT. Modern neurotherapeutics are attractive due to their relative safety and potential neuroplastic effects. However, they need to be thoroughly tested for clinical and cognitive efficacy across settings and beyond core symptoms and for their potential for individualised treatment.

Despite great technical capabilities, the theory of non-contact temperature measurement is usually not fully applicable to the use of measuring instruments in practice. While black body calibrations are in practice well established and easy to accomplish, this calibration protocol is never fully applicable to measurements of real objects under real conditions. Currently, the best approximation to real world radiation thermometry is grey body radiation thermometry, which is supported by most measuring instruments to date. Nevertheless, the metrological requirements necessitate traceability, therefore real body radiation thermometry is required for temperature measurements of real bodies. This article documents the current state of temperature calculation algorithms for radiation-thermometers and the creation of a traceable model for radiation thermometry of real bodies that uses a digital twin model of the system of measurement to compensate for the loss of data, caused by spectral integration, which occurs when thermal radiation is absorbed on the active surface of the sensor. To solve this problem, a hybrid model with variable scalar inputs is proposed, in which the scalar input parameters are calculated to correspond to the system of measurement. The method for calculating the effective parameters is proposed and verified with the theoretical model of non-contact thermometry. The sum of effective instrumental parameters is presented for different temperatures to show that the rule of radiation thermometry of grey bodies, according to which the sum of instrumental emissivity and instrumental reflectivity equals to 1, does not hold in radiation thermometry for real bodies. Using the derived models of radiation thermometry, the uncertainty of radiation thermometry due to the uncertainty of spectral emissivity is analysed by simulated measurements along the temperature ranges of various radiation thermometers, enabling traceable non-contact temperature measurements with known measurement uncertainty under any known conditions.

The aim of this paper is to present a comprehensive overview of the main aims and scopes in screening of botanicals, a task which thin-layer chromatography (TLC) is on an everyday basis confronted with and engaged in. Stunning omnipresence of this modest analytical technique (both in its standard format (TLC) and the high-performance one (HPTLC), either hyphenated or not) for many analysts might at a first glance appear chaotic and random, with an auxiliary rather than leading role in research, and not capable of issuing meaningful final statements. Based on these reflections, our purpose is not to present a general review paper on TLC in screening of botanicals, but a blueprint rather (illustrated with a selection of practical examples), which highlights a sovereign and important role of TLC in accomplishing the following analytical tasks: (i) solving puzzles related to chemotaxonomy of plants, (ii) screening a wide spectrum of biological properties of plants, (iii) providing quality control of herbal medicines and alimentary and cosmetic products of biological origin, and (iv) tracing psychoactive plants under forensic surveillance.

The formation and propagation of cracks occurs through irreversible dislocation movements at notches, material defects and grain boundaries. Since this process is partly thermally controlled, the resistance to dislocation movements at low temperatures increases. This slows both fatigue initiation and fatigue crack propagation. From recent experimental data, it can be seen that fatigue crack growth is accelerated below the fatigue transition temperature (FTT) that correlates with the ductile-brittle transition temperature (DBTT) found by well-known fracture mechanics tests, i.e., Charpy impact, fracture toughness, and CTOD. Hence, this study investigates the relation between FTT and DBTT in S500 high-strength steel base material and welded joints at low temperatures using fatigue crack growth, fracture toughness tests as well as scanning electron microscopy. From the tests, an almost constant decrease in fatigue crack propagation rate is determined with decreasing test temperature even below the DBTT.

Cooperating TFs can be revealed through the analysis of co-occurrence of their motifs. Methods: We applied Motifs Co-Occurrence Tool (MCOT) that predicted pairs of spaced or overlapped motifs (composite elements, CEs) for a single ChIP-seq dataset. We improved MCOT capability for prediction of asymmetric CEs with one of participating motifs possessing higher conservation than another does. Results: Analysis of 119 ChIP-seq datasets for 45 human TFs revealed that almost for all families of TFs the co-occurrence with an overlap between motifs of target TFs and more conserved partner motifs was significantly higher than that for less conserved partner motifs. The asymmetry toward partner TFs was the most clear for partner motifs of TFs from ETS family. Conclusion: Co-occurrence with an overlap of less conserved motif of a target TF and more conserved motifs of partner TFs explained a substantial portion of ChIP-seq data lacking conserved motifs of target TFs. Among other TF families, conservative motifs of TFs from ETS family were the most prone to mediate interaction of target TFs with its weak motifs in ChIP-seq.

A retrospective cohort study was performed to determine the likelihood of malignant stroke after mechanical thrombectomy via ASPECTS-PASS score(A-P score), a novel scoring system using ASPECT score and number of arterial-passes. This prediction score is designed to help clinicians take early measures. Malignant stroke is defined as a large MCA infarct with mass effect that develops during the first 5 days following presentation and is associated with approximately 80% mortality. Previous studies have shown an association of low ASPECTS and high number of passes with poor outcome.We performed a retrospective cohort analysis on 78 patients with anterior circulation stroke who underwent mechanical thrombectomy at our hospital. We analyzed the likelihood of developing malignant stroke post thrombectomy in the first 5 days following successful recanalization (TICI ≽ 2b). Malignant stroke is defined as severe symptoms secondary to edema as evidenced by midline shift, cistern engorgement, and hemorrhagic conversion with edema. Patients with ASPECTS of 0-5 were excluded. We used two variables to predict the outcome: ASPECT score and number-of-passes. We used the equation ASPECT score minus number-of-passes (A-P) to arrive at a single number to predict the risk of malignant stroke. A-P scoring scale ranged from 0-9. Our study showed a statistically significant inverse relationship of A-P score and risk of malignant stroke. Approximately 90% of patients with A-P score < 5 developed malignant stroke in our study. A-P score can help predict malignant stroke post thrombectomy even after successful recanalization. Thus, A-P score of ≤ 5 suggests very high risk. (A-P) Score can assist clinicians to take early measures, and thereby prevent worse outcomes.
