Old growth forests are increasingly rare but important carbon sinks which harbour rich biodiversity. Chronic browsing by the white-tailed deer (Odocoileus virginianus) is a major problem in northern temperate forests where deer numbers have increased in recent decades driven by stricter hunting rules and reduced predation necessitating local monitoring of vegetation responses. The objective of this study was to determine the effects of deer exclusion on tree regeneration dynamics and soil nutrient cycling in an old growth Carolinian forest. This was done using exclusion fencing and tip-up mounds at McMaster Forest Nature Preserve and the Sheelah Dunn Dooley Nature Sanctuary in Hamilton Ontario. Tree regeneration was surveyed from thirty 1m x 1m quadrats within exclusion plots and another thirty quadrats from deer browsed areas adjacent to the exclusion plots. Soil samples were taken from each quadrat to analyze browsing impacts on nitrate, phosphate and soil organic matter. Red oak (Quercus rubra) was planted at the top and base of tip-up mounds of varying heights and widths and monitored for deer access and browsing activity. Results show a significantly higher density of woody plants within exclosures compared to non-exclosures (p=0.0089) and twice more abundance of highly palatable species within the exclosures. However, species richness (p > 0.05) and diversity (p > 0.05) were minimally impacted by deer browsing showing a resilient old growth forest. Soil nitrate was consistently higher in the non-exclosures while phosphate was consistently higher within deer exclosures. Finally, more seedlings survived at the top of mounds than the bases showing the potential of tip-up mounds to be a natural method of deer exclusion and a critical avenue for restoring over-browsed forests.

Two putative boar pheromones (Androstenone and Androstenol) were discovered in endangered mouse deer during captive breeding program. This study further examined the molecular characteristics, pheromone synthesis pathway, and the functional role of these pheromones in reproduction of mouse deer. CYP17A1 and CYB5 genes were cloned and expressed in HEK-293, COS-7 cell lines and gonads of mouse deer to investigate CYP17A1 gene’s andien-β-synthase activity towards synthesis of sex pheromones in mouse deer. An enzyme immunoassay was also developed and standardized to measure the fecal androstenone during reproductive cycles of mouse deer. Results showed that mouse deer’s CYP17A1 gene possesses andien-β-synthase activity and could transform pregnenolone into 5,16-androstadien-3β-ol. The expression of CYP17A1 gene upregulated in the testis and ovary, compared to other tissues in mouse deer. Significantly elevated pheromones and estrogens were recorded prior to delivery and postpartum estrus / mating in mouse deer. Further, there were weak correlations between fecal pheromones and estrogens/ androgens in mouse deer during breeding season. The findings suggest that the boar pheromones might play a direct role in the reproductive activities of mouse deer which might be used for breeding of mouse deer elsewhere.

Chronic wasting disease is a fatal transmissible spongiform encephalopathy (TSE) of cervids caused by a misfolded variant of the normal cellular prion protein, and is closely related to sheep scrapie. Variations in a host’s prion gene, PRNP, and its primary protein structure, dramatically affect susceptibility to specific prion disorders, and breeding for PRNP variants that prevent scrapie infection has led to steep declines in the disease in North American and European sheep. While resistant alleles have been identified in cervids, a PRNP variant that completely prevents CWD has not yet been identified. Thus, control of the disease in farmed herds traditionally relies on quarantine and depopulation. In CWD-endemic areas, depopulation of private herds becomes challenging to justify, leading to opportunities to manage the disease in situ. In the present study, we developed a selective breeding program for farmed white-tailed deer in a CWD-endemic area, focused on reducing frequencies of highly susceptible PRNP variants and introducing animals with less-susceptible variants into historically high prevalence areas. We found that breeding followed predictable Mendelian inheritance, and early data support our project’s utility in reducing CWD prevalence. This project represents a novel approach to CWD management, with future efforts building on these findings.

Reproductive abnormalities have been observed in fallow deer populations in Hungary. We supposed mycotoxin contamination to be one of the possible causes because multi-mycotoxin contamination is known to be dangerous even at low toxin levels, especially for young animals. We investigated the spatial pattern of mycotoxin occurrences and the relation of maternal and foetal mycotoxin levels. 72 fallow deer embryos and their mothers were sampled in seven forested regions in Hungary in the 2020/2021 hunting season. We analyzed Aflatoxin (AF), Zearalenone (ZEA), Fumonizin B1 (FB1), DON, T2-toxin concentrations in maternal and foetal liver by ELISA. AF was present in 70% and 82%, ZEA in 41% and 96%, DON in 90% and 98%, T2 in 96% and 85%, FB1 in 84% and 3% of hind and foetus livers, respectively. All mycotoxins passed into the foetus, but Fumonizin B1 only rarely. The individual variability of mycotoxin levels was extremely high, but the spatial differences were moderate. We could not prove a relation between the maternal and foetal mycotoxin concentrations, but we found accumulation of ZEA and DON in the foetuses. These results reflect the possible threats of mycotoxins to the population dynamics and reproduction of wild fallow deer.

Apex predators were extirpated from the UK hundreds of years ago. Practitioners have recently advocated for reintroductions of predators in the UK given their role as keystone species. For a reintroduction to take place, a survey of public desirability needs to be conducted. We aimed to understand the perspectives of people across the United Kingdom on the reintroduction of Eurasian lynxes, grey wolves, and brown bears. We collected data via questionnaires from 78 key stakeholders, such as farmers, general public, nature conservationists, and those in environmental sectors, with these areas being seen as the most affected by these reintroductions. It was organizations which front these stakeholder groups which mainly distributed the survey, as well as social media groups. We ran a path analysis, which highlighted that there was a negative correlation between respondents viewing the species as a danger towards livestock and humans, and the respondent seeing it being good to have that species in their region. Overall, participants who stated that rewilding their region was a priority also stated that it would be good to have Eurasian lynx in their region. That said, 70% of farmers strongly disagreed with the reintroduction of Eurasian lynx. These findings showed that much of the United Kingdom would welcome the reintroduction of Eurasian lynx, but both farmers and the public would fear the loss of livestock that may come along with these species. Support can be gained through education and knowledge, a necessary step needed especially targeting farmers before any reintroduction plans can be made.

Tick borne encephalitis virus (TBEV) is the causative agent of Tick borne encephalitis in humans, a severe zoonosis occurring in the Paleartic region mainly transmitted through ticks belonging to the genus Ixodes. In Italy, TBEV is restricted to few foci in the north-eastern part of the country. This report describes for the first time a case of clinical TBE in a roe deer, occurred in the Belluno province, Veneto region, an area highly endemic for the presence of the virus. The affected roe deer showed ataxia, staggering movements, muscle tremors and persistent teeth grinding causing hypersalivation. At necropsy, the macroscopic picture was inconclusive. RNA of TBEV was detected by real-time RT-PCR. Phylogenetic analysis revealed a close relationship to TBEV of the European subtype, and 100% similarity with a virus from the bordering Trento Province. The histological examination of the midbrain confirmed the viral etiology and specific immunofluorescence indicated the presence of a Flavivirus infection and characterized the pattern of infection in the neurons. This report underlines for the first time the occurrence of clinical encephalitic manifestations due to TBEV in a roe deer, thussuggesting to include this pathogen in the frame of differential diagnosis in this species.

The Korean water deer (WD), the dominant wildlife species in South Korea, is classified as vul-nerable on the IUCN Red List. Beside their several unique characteristics, they have a lower in-cidence of tick bites compared to other Cervidae species, particularly roe deer (RD). In natural environments, ticks exhibit questing behavior by waiting on grasses to climb onto host's hair coat. They use sensory organs to detect signals such as body temperature and chemicals from the host's skin, then move to preferred feeding location. Therefore, we conducted a comparative analysis of the skin tissue and hair features of both species to compare the factors leading to lower incidence of tick bites in WD. Notably, WD had significantly larger blood vessels, sebaceous glands, and sweat glands which maintain the barrier functions of the skin (p≤0.004), irregular scale surfaces in hair cuticles, and stiff and 2.83-times thicker primary hair diameter (p<0.001). Each parameters made the ticks difficult to move onto the epidermis of WD. To our knowledge, this is the first report which is focused on the comparative analysis for skin and hair feature in WD and RD. We suggest that these differences may attribute to evade tick bites in WD.

Hepatitis E virus (HEV) is a zoonotic foodborne virus with an annual infection prevalence of 20 million human cases, which seriously affects public health and economic development in both developed and developing countries. To better understand the epidemiology of HEV in the Centre of Portugal, a cross-sectional study was conducted from 2016 to 2023 with sera samples from wild ungulates. The seroprevalence and risk factors for HEV seropositivity were evaluated in the present study. Specifically, antibodies against HEV were determined by a commercial enzyme-linked immune-sorbent assay (ELISA). Our results show that in the 650 sera collected from 298 wild red deer and 352 wild boars from Portugal, 9.1% red deer (95% confidence interval [CI]: 6.312.9%) and 1.7% wild boar (95% CI: 0.63.3%) were positive for antibodies to HEV. Regarding age, the seropositivity in juvenile wild ungulates was 1.3% (95% CI: 0.273.72%) and 7.2% in adults (95% CI: 4.910.11%). Logistic regression models investigated risk factors for seropositivity. The odds of being seropositive was 3.6 higher in adults than in juveniles (95% CI: 1.7218.11%) and the risk was 4.2 higher in red deer than in wild boar (95% CI: 1.64–10.69%). Both wild ungulate species were exposed to HEV. The higher seroprevalence in red deer suggests that this species may have a major contribution to the ecology of HEV in the Centre of Portugal. Further research is important to understand how wildlife affects the epidemiology of HEV infection in Portugal.

Animal welfare assessment protocols are important for identifying welfare problems in conservation programs. This study aimed to develop a baseline welfare protocol for the assessment of welfare in huemul (Hippocamelus bisulcus) in conservation centers. This protocol is based on the Welfare Quality® (WQ) framework for cattle, considering the available literature on the behaviours, ecology, conservation plans, captive management of this species, and welfare assessment protocols for other ungulate species. As a result, the protocol was specifically developed for huemuls and included four principles, 12 criteria, and 23 animal- and resource-based indicators. The 12 criteria of the WQ protocol were reduced to nine, and three new criteria were added because they were both feasible and essential for welfare assessment in captive huemuls. This protocol is mainly intended to identify welfare problems in endangered species in the context of conservation centers (reproduction, rescue, rehabilitation, or treatment centers). Thus, aggregation of different measures to obtain a global score has not been proposed. However, a scoring system that assigns a value on a 0-2 scale (0= No welfare concern; 1= welfare concern; 2= urgent welfare concern) is proposed for each category. Although further research is still needed to fully validate the protocol, this is the first development of a completed protocol for huemul welfare assessment.

In the lowland forests of south-western Poland, the effect of the fencing of forest regeneration stages, aged up to 20 years, upon the level of damage inflicted by red deer (Cervus elaphus), fallow deer (Dama dama), and roe deer (Capreolus capreolus) in non-fenced patches of this age class of forest was studied. In the studied forest districts (n=4), there was the highest proportion of the mixed coniferous‑deciduous forest types, where the main forest‑forming tree species were pine (Pinus sylvertris), birch (Betula sp.), oak (Quercus sp.), beech (Fagus sylvatica), and spruce (Picea abies). At the forest district level, the percentage of fenced areas in young forest plantations (1‑10‑year old) ranged, on average, from 9.1-30.9%, and that in tickets (11-20‑year old) ranged from 0.0%-10.2%. The percentage of areas of severely damaged young plantations fluctuated between 5.7 and 14.2%, and that of tickets fluctuated between 0.3-12.9%. For young plantations, a statistically significant correlation (“r” ranged from 0.51-0.75) was obtained between percentage of area in fenced plots and the level of damage caused by deer. For thickets, this correlation was obtained solely in Pieńsk Forest District (r = 0.646). In the studied forest districts, the fencing of young plantations and thickets resulted in the reduction of average potential food base for cervids, from 2.9 to 10.0 %. An attempt was made to interpret the level of deer damage on the basis of an index, taking into account the standing crop of deciduous browse and forbs, and the population density of cervids.

Antler is the fastest growing tissue. Because is based on proto-oncogenes, to avoid the risk of cancer, antlers evolved strong anticancer mechanisms, and thus its extract (DVA) is effective also against the few human tumours studied so far. We assessed whether DVA is a general anticancer compound testing direct effects in cells of different tumours: glioblastoma (GBM), colorectal and breast cancer, and leukaemia. DVA reduced viability of tumour but not healthy cells. Mobility decreased at least at the longest test (72 h). Intraperitoneal/oral 200 mg DVA/kg administration in GBM xenograft mice for 28 d reduced tumour weight 66.3% and 61.4% respectively, and also spleen weight. In addition, tumours treated with DVA showed symptoms of liquefactive necrosis. Serum cytokines showed DVA up-regulated factors related with tumour fighting and down-regulated those related with inducing immune tolerance to the tumour. DVA shows general anticancer effects in the lines tested and, in GBM mice, also strong indirect effects apparently mediated by the immune system.

This study employs Principal Component Analysis to examine cranial measurements obtained from both extant and fossil specimens spanning 38 species and comprising over 300 individuals within the subfamily Cervinae. Our findings elucidate craniometric distinctions among cervids characterized by varying body sizes and certain evolutionary adaptations. While our results generally corroborate previous assertions regarding the limited craniometric variability among plesiometacarpal deer, they also unveil specific cranial adaptations within certain genera and species. The Principal Component Analysis of craniometric data revealed that giant and large-sized deer display significantly broader ecomorphological diversity in cranial shape compared to small-sized deer. Secondly, small-sized deer exhibit greater uniformity in their cranial shape, appearing densely clustered on the factorial map. Thus, we conclude that body size imposes ecological constraints, limiting the available niches due to eco-physiological factors. The study demonstrates that endemic insular deer do not evolve consistent craniometric features attributable to insular isolation, while the cranial proportions of medium-to-small sized deer delineate a ubiquitous morphological archetype characteristic of numerous cervid taxa spanning diverse phylogenetic lineages and sharing comparable body sizes. This group of "Dama-like" deer, characterized by similar body size, metabolic rates, ecological needs, and cranial morphometry, is referred to here as the fundamental eco-physiological type, typical of warm regions within the Palearctic and Oriental biogeographic realms..

In the last decade, an upsurge of human leishmaniasis has been reported in the Emilia-Romagna region, Northeast Italy. Epidemiologic data raised doubts about the role of dogs as the main reservoirs for Leishmania infantum. In the present study, a total of 1,077 wild animals were screened for L. infantum DNA in earlobe and spleen samples from 2019 to 2022. The lymph nodes were tested only in the 23 animals already positive in the earlobe and/or spleen. A total of 71 (6.6%) animals resulted positive in at least one of the sampled tissues, including 3/18 (16.7%) wolves, 6/39 (15.4%) European hares, 38/309 (12.3%) roe deer, 1/11 (9.1%) red deer, 8 (4.9%) wild boars, 13/319 (4.1%) red foxes, 1/54 (1.9%) porcupine, and 1/59 (1.7%) European badger. Most of the infected animals (62/71) tested positive only in the earlobe tissue, just 4 animals (2 roe deer and 2 wild boars) tested positive only in the spleen, and 5 animals (3 roe deer and 2 red foxes) resulted positive for both tissues. L. infantum DNA was detected in the lymph nodes of 6/23 ani-mals. L. infantum detection occurred in all seasons associated with low real-time PCR Ct values. Further research is needed in order to clarify the role of wildlife in the re-emerging focus of leishmaniasis in Northeast Italy.

This article investigates the effectiveness of feature extraction and selection techniques for enhancing the performance of classifiers accuracy in Type II Diabetes Mellitus (DM) detection using microarray gene data. To address the inherent high dimensionality of the data by employing three Feature Extraction (FE) methods are used namely, Short-Time Fourier Transform (STFT), Ridge Regression (RR), and Pearson Correlation Coefficient (PCC). To further refine the data, meta-heuristic algorithms like Bald Eagle Search Optimization (BESO) and Red Deer Optimization (RDO) are utilized for feature selection. The performance of seven classification techniques such as Non-Linear Regression - NLR, Linear Regression - LR, Gaussian Mixture Model - GMM, Expectation Maximization - EM, Logistic Regression - LoR, Softmax Discriminant Classifier - SDC, and Support Vector Machine with Radial Basis Function kernel - SVM-RBF are evaluated with and without feature selection. The analysis reveals that the combination of PCC with SVM-RBF achieved a promising accuracy of 92.85% even without feature selection. Notably, employing BESO with PCC and SVM-RBF maintained this high accuracy. However, the highest overall accuracy of 97.14% was achieved when RDO was used for feature selection alongside PCC and SVM-RBF. These findings highlight the potential of feature extraction and selection techniques, particularly RDO with PCC, in improving the accuracy of DM detection using microarray gene data.

Quaking aspen (Populus tremuloides Michx.) landscapes are valued for their biodiversity, water retention, fire mitigation, aesthetics, and recreation opportunities. Across North America, some aspen populations are experiencing population declines as they face uninhibited ungulate browsing, drought, fire suppression, insects, disease, and inappropriate management. Increased human development and recreational use within aspen landscapes can serve as additive stressors, though there is a dearth of literature examining these elements. At a popular recreational area in Utah, USA, identifying the cause of apparent decline within a larger aspen community is complicated by development upstream and recreation-related activities. We sought to: 1) assess the overall condition of the aspen at the site, 2) understand key variables that influence aspen conditions, and 3) elucidate how aspen fitness varies across the site. We collected data from forty-five plots using established aspen sampling methods, including ungulate presence, tree characteristics, soil chemistry, and environmental descriptors. Results suggest that a combination of higher levels of browsing and elevated soil sodium may be causing premature mortality and limiting aspen recruitment in a portion of the study area. These findings will inform future management at this site, as well as similar recreational forest settings experiencing compound stressors.

The evidential value of an mtDNA match between biological remains and its potential donor is determined by the random match probability of the haplotype. This probability is based on the haplotype’s population frequency estimate. Consequently, implementing a population study representative of the population relevant to the forensic case is vital to correctly evaluating the evidence. The emerging number of poaching cases and the limited availability of such data emphasizes the need for improved fallow deer mtDNA population databank for forensic purposes, including targeting the entire mitochondrial control region. By sequencing the whole mitochondrial control region in 138 animals from five populations in Hungary we found four different haplotypes, including one which has not yet been described. Our results, supplemented with data already available from previous research do not support the possibility of determining the population of origin, although some patterns of geographical separation can be distinguished. Estimates of molecular diversity indicate approximately similarly low mtDNA diversity (Hd=0.565 and π = 0.002), compared with data from other countries. The calculated random match probability of 0.547 shows a high probability of coincidence and, therefore, a limited capacity for exclusion. Consequently, it may be necessary to search for polymorphic sites on other mitochondrial gene sections or to sequence the entire mitogenome of fallow deer.
