The sperm heads of frozen/thawed horse spermatozoa were injected into ovulated mouse oocytes where they decondensed and formed paternal pronuclei in which the DNA damage has been evaluated. The motile spermatozoa exhibit none or minor level of DNA damage and the xenogenic zygotes cleave to the two-cell embryonic stage with regular nuclei and no micronuclei (MNs). On the other hand, the immotile spermatozoa exhibit typically high levels of DNA damage and blastomeres in two-cell stage embryos contain multiple MNs. Sperm during the freezing and thawing process is damaged by oxidative stress. Thus, its antioxidant scavenger is essential for sperm survival and death in frozen-thawed semen. We used melatonin and silymarin in experiments. Our study aimed to identify the effect of melatonin and silymarin on the motility and viability of sperm, reactive oxygen species (ROS), and nitric oxide (NO) production in frozen-thawed boar semen. Melatonin and silymarin were treated alone and co-treated in the fresh boar semen. Boar semen was collected using the gloved-hand method from ten crossbred pigs, and the age of the experimental ten male pigs was 28.7 ± 3.2 months. We evaluated sperm viability using SYBR-14 and PI kit, and ROS and NO production were detected by DCF-DA and DAF-2, respectively. The sperm motility was not significantly different between non-treatment and treatment. ROS and NO production in frozen-thawed sperm were decreased by melatonin and silymarin (P < 0.05). Moreover, silymarin significantly reduced NO production more than melatonin (P < 0.05). Melatonin (46.37 ± 1.10) and silymarin (46.13 ± 0.80) enhanced the viability of sperm (P < 0.05). We suggest that melatonin and silymarin are essential antioxidants in semen cryopreservation for protecting sperm damage and maintaining sperm viability. Melatonin and silymarin may be useful antioxidants in freezing boar sperm.

In the present work freezing and melting characteristics of water seeded with chemically functionalized graphene nano-platelets in a vertical cylindrical capsule was experimentally studied. The volume percentage of functionalized graphene nano-platelets was varied from 0.1% to 0.5% with an interval of 0.1%. The stability of the synthesised samples were carried out by zeta potential distribution. The thermal conductivity of the nanocomposite samples were experimentally measured using transient hot wire method. A maximum enhancement of ~24% in the thermal conductivity was observed for the 0.5% volume percentage in the liquid state while a ~53% enhancement in the solid state. Freezing and melting behaviour of water dispersed with graphene nanoplatelets were carried out using a cylindrical stainless steel capsule in a constant temperature bath. The bath temperatures considered for studying freezing characteristics were considered to be −6 °C and −10 °C, while to study the melting characteristics the bath temperature was set as 31 °C and 36 °C. The freezing and melting time decreased for all the test conditions when the volume percentage of GnP increased. The freezing rate was enhanced by ~ 43% and ~32% for the bath temperatures of −6 °C and −10 °C respectively at 0.5 vol % of graphene loading. The melting rate was enhanced by ~42% and ~63% for the bath temperature of 31 °C and 36 °C respectively at 0.5 vol % of graphene loading.

We tested the bioactivity of a dalbavancin + heparin (DH) vs. dalbavancin (D) against biofilms of Staphylococcus aureus ATCC43300 (MRSA) and Staphylococcus epidermidis ATCC35984 (MRSE) comparing DH median value of cfu counts and metabolic activity with that obtained for D before and during storage under freezing up to 6 months. The addition of heparin to a dalbavancin lock solution did not affect its bioactivity against staphylococcal biofilms irrespective of its preservation time under freezing.

The objective of this study was to evaluate the effect of the addition of atomized black Maca in the cryopreservation medium of epididymal spermatozoa of alpaca species. Fifteen pares of testes and epididymis from alpacas of reproductive age were collected from a local (Huancavelica) slaughterhouse and transported in NaCl 0,9% at 10°C to the laboratory. Each sample of epididymal sperm was divided into six groups: 1) Fresh, 2) Yolk medium (YM), 3) YM+10mg/ml Maca, 4) YM+20mg/ml Maca, 5) YM+30mg/ml Maca, and 6) YM+ Resveratrol (antioxidant positive control). Sperm cryopreservation was performed through the slow-freezing method. Markers associated with functionality such as motility, vitality and plasma membrane integrity. Cellular markers associated with oxidative damage, such as DNA fragmentation, total ROS production and lipid peroxidation, were analyzed before and after freezing. The results showed that the supplementation with black Maca at 20mg/ml improved the sperm motility, viability and integrity of the membrane. Similarly, total ROS production decreased with Maca at 10 mg/ml (25 ± 2.23%) and 20mg/ml (20 ± 2.52%) (P<0.05). However, DNA fragmentation and lipid peroxidation were similar in all groups. These results suggest that Maca at 20mg/ml has a protective effect during the freezing/thawing process of epididymal sperm of alpaca species.

Global climate has undergone significant changes in the past 50-60 years. The transition from stable climate conditions to current warming has significantly influenced permafrost landscapes and infrastructure in cold regions. Arctic and Subarctic landscapes had distinct reactions to these changes. The objective of this study is to analyze the regional aspects of climate and permafrost changes in Yakutia's Arctic and subarctic regions from the past 50-60 years. Our analysis focused on climate change patterns during different periods, including cold period, pre-warming, and modern warming. We analyzed data on mean annual air temperature, thawing and freezing indices, and precipitation. During these periods, we determined the reaction's characteristics in various landscape regions. The most significant impact on permafrost began in the mid-2000s, as our observations confirmed. Global warming directly causes permafrost temperature to rise, the active layer to thicken, and cryogenic processes to develop. The conducted research provides the information needed to study the effects of global warming on permafrost and adapt socio-economic conditions in northern Yakutia.

This paper provides an overview of the social reasons that lead women to consider egg cryopreservation, as well as the attitudes and knowledge of women towards this procedure. Methods: For the creation of this article, a literature review was carried out both in the existing medical literature and in search engines. The key points are 1. The main reasons women choose to delay childbearing are the lack of a suitable male partner, education and career advancement, and financial instability. Women feel societal and family pressure to have children, even though they may not feel ready. 2. Women's attitudes towards egg cryopreservation vary. While some are open to the idea, many are opposed to it, especially when it is used for non-medical reasons. There are concerns about the success rates, health risks, and ethical implications of the procedure. 3. The legal and regulatory framework around egg cryopreservation differs across countries. Some countries allow it only for medical reasons, while others have more permissive policies. The issue of who should bear the financial cost of the procedure is also debated. 4. There is a need to better educate women, as well as healthcare providers, about fertility decline and the options available for preserving fertility, including egg cryopreservation.

The problem of intensification of melt crystal growth process has been analyzed using CdTe as an actual material. Numerical simulation of 100 mm diameter CdTe crystal growth by VGF technique has been carried out. The heat-mass transfer was controlled by introducing of low-frequency oscillating baffle into the melt, so called axial vibrational control (AVC) technique. The baffle configuration has been optimized to destroy solid «tails», which were formed near the crucible walls at high cooling rates due to the low thermoconductivity and the corresponding latent heat. Analysis of CdTe homogeneity range showed that during fast crystal cooling Te micro precipitations were formed resulting from decay of oversaturated Cd-riched nonstoichiometric solid solution at Bridgman crystal growth technique. A VGF grown CdTe crystal after full crystallization stays inside the phase field of the high temperature wurtzite polymorph. This makes it possible to go through the polymorph transition without Te micro-precipitating using the advantages of VGF specific feature of very slow cooling.

Paraffin wax stores energy in the form of latent heat at a nearly constant temperature during melting and releases this energy during solidification. This effect is used in industrial energy storage. An unusual change in the shape of a melted droplet of paraffin wax placed on a relatively cold glass plate is studied. As the droplet solidifies, its upper surface becomes nearly flat and a dimple is formed in the center of this surface, making the droplet look like a fruit (pumpkins are more commonly shaped like this, but the authors prefer apples). A series of experiments, as well as physical and numerical modeling of the droplet's thermal state, taking into account the formation of a mushy zone between liquidus and solidus, made it possible to understand the role of gravity and gradual increase in viscosity and density of paraffin wax on changing the droplet shape and, in particular, to clarify the mechanism of formation of the dimple on its upper.

In order to study the performance of a new type of cement-based grouting material under the coupling effect of freezing-thawing cycle and sulfate attack, experiments were designed and carried out. The damage mechanism of the material under the coupling effect of freezing-thawing and Na2SO4 solution was analyzed by measuring the mass change, relative dynamic elastic modulus, compressive strength loss and microstructure of the new grouting material. With increasing freeze-thaw cycles, mass loss and compres-sive strength loss of the sample in 15% Na2SO4 solution gradually increase, and the relative dynamic elastic modulus gradually decreases. During a 30-cycle freeze-thaw cycle, the mass loss rate, compressive strength loss rate, and relative dynamic elastic modulus of the sample are 4.17%, 24.59%, and 84.3%, respectively, showing good corrosion resistance and frost resistance durability. Microscopic analysis showed that SO42- caused the decomposition of C-S-H gel in the sample and the formation of Gypsum, and the widening of crack width aggravated the internal deterioration, indicating that the material deterioration rate increased under the dual-factor coupling effect.

Benign design of alkali-activated slag/FA geopolymer paste has intriguing increasing attention for optimizing its service performance. Therefore, the replacement of fly ash (FA) with 10 wt% silica fume (SF) is investigated by mechanical strength after freeze-thawing cycles and microstructure characterization. The results show that an appropriate dosage (30 wt%) of slag is necessary to attain alkali-activated slag/FA geopolymer paste with excellent mechanical performance, and the SF/slag/FA (SF: slag: FA= 10:30:60, wt%) geopolymer paste exerts the compressive and flexural strength of 95.2 and 3.2 MPa, respectively. Meanwhile, the doped 10 wt% SF facilitates the propagation of (N, C)-A–S–H chains, rather than the formation of C-S-H gels, evidenced by the absent exothermic peak at about 861oC from the differential scanning calorimetry (DSC) curves, leading to the increases in the pores volume with a pore diameter<20 nm and tortuosity by mercury intrusion porosimeter (MIP) results, corresponding to an improved freezing-thawing resistance with the residual compressive strength of 52.8 MPa and the weight loss of 10.5% after 300 freeze-thawing cycles. It explores a cost-effective and benign facile approach to designing heating curing-free alkali-activated slag/FA geopolymer paste with good freezing-thawing resistance.

The climate change is likely to modify the freezing-thawing cycles in soils and surface waters of permafrost-affected and subarctic regions. Yet, the change of solution chemical composition during ice formation and evolution of remaining fluids remain very poorly known. Towards better understanding of dissolved (&lt; 0.45 µm) organic carbon, major and trace elements behavior in permafrost peatland environments, here we performed laboratory freezing of peat leachates, from complete freezing to complete thawing in order to quantify the partitioning of solutes between the aqueous solution and the remaining ice. We hypothesized the existence of two main groups of solutes, behaving conservatively or non-conservatively during freezing depending on their incorporation into the ice or ability to coagulate in the form of insoluble minerals or amorphous materials in the fluid phase. An unexpected result of this work was that, despite sizable degree of element concentration in the remaining fluid and possible coagulation of organic, organo-mineral and inorganic compounds, the freezing and subsequent thawing produced the final concentrations of most solutes which were not drastically different from initial concentrations in starting leachates, prior freezing. This demonstrates high stability of dissolved (&lt; 0.45 µm) OC, Fe, Al and some trace metals to repetitive freezing and thawing of surface waters in permafrost peatlands.

Cuticular waxes are a mixture of hydrophobic very-long-chain fatty acids and their derivatives accumulated in the plant cuticle. Most studies define the role of cuticular wax largely based on reducing non-stomatal water loss. The present study investigated the role of cuticular wax in reducing both low-temperature and dehydration stress in plants using Arabidopsis thaliana mutants and transgenic genotypes altered in the formation of cuticular wax. cer3-6, a known Arabidopsis wax-deficient mutant (with distinct reduction in aldehydes, n-alkanes, secondary n-alcohols, and ketones compared to wild type (WT)), was most sensitive to water loss; while dewax, a known wax overproducer (greater alkanes and ketones compared to WT), was more resistant to dehydration compared to WT. Furthermore, cold-acclimated cer3-6 froze at warmer temperatures, while cold-acclimated dewax displayed freezing exotherms at colder temperatures compared to WT. GC-MS analysis identified a characteristic decrease in the accumulation of certain waxes (e.g. alkanes, alcohols) in Arabidopsis cuticles under cold acclimation, which was additionally reduced in cer3-6. Conversely, the dewax mutant showed a greater ability to accumulate waxes under cold acclimation. FTIR spectroscopy also supported observations in cuticular wax deposition under cold acclimation. Our data indicate cuticular alkane waxes along with alcohols and fatty acids can facilitate avoidance of both ice formation and leaf water loss under dehydration stress, and are promising genetic targets of interest.

The determination of the parameters that characterise the air-void system in hardened concrete elements becomes crucial for structures under freezing and thawing cycles. The ASTM C457 standard describes some procedures to accomplish this task, but they are not easy to apply, require specialised equipment such as a stereoscopic microscope and result in highly tedious tasks to be performed. This paper describes an alternative procedure to the modified point-count method described in the Standard that makes use of macro photography. This alternative procedure is successfully applied to a large set of samples and presents some advantages over the traditional method, since the required equipment is less expensive and provides a more comfortable and less tedious procedure for the operator.

In order to clarify the individual role of freezing and frozen storage on the quality of fish, fillets of large-mouth bass (Micropterus salmoides) were subjected to different freezing rates (freezing with -18 °C (A), -60 °C (B), -60 °C with forced air circulation at 2 m/s (C), respectively) followed by frozen storage at -18 °C for 30 and 90 days. Another 2 groups were frozen with -60 °C, followed by storage at -40 °C (D) and -60 °C (E), respectively. Results showed that water-holding and TVBN were mainly affected by storage time. No significant changes were found in free thiol content among treatments. A greater freezing rate and lower storage temperature generally led to lower TBARS. GC×GC-TOFMS revealed a total of 66 volatile compounds which were related to lipid oxidation. PLS-DA showed that fresh samples were separated from the frozen-thawed ones, and fillets in group D &amp; E were relatively close to fresh fillets in the composition of oxidation-related volatiles. In conclusion, freezing rate and storage temperature had significant impact on lipid oxidation and protein denaturation in the fillets of large-mouth bass, while protein oxidation was more affected by freezing rate.

Freezing of gait (FOG), a debilitating symptom of Parkinson’s disease (PD), can be safely studied using the stepping in place (SIP) task. However, clinical, visual identification of FOG during SIP is subjective and time consuming, and automatic FOG detection during SIP currently requires measuring center of pressure on dual force plates. This study examines whether FOG elicited during SIP in 10 individuals with PD could be reliably detected using kinematic data measured from wearable inertial measurement unit sensors (IMUs). A general, logistic regression model (AUC = 0.81) determined that three gait parameters together were overall the most robust predictors of FOG during SIP: arrhythmicity, swing time coefficient of variation, and swing angular range. Participant-specific models revealed varying sets of gait parameters that best predicted FOG for each participant, highlighting variable FOG behaviors, and demonstrated equal or better performance for 6 out of the 10 participants, suggesting the opportunity for model personalization. The results of this study demonstrated that gait parameters measured from wearable IMUs reliably detected FOG during SIP, and the general and participant-specific gait parameters allude to variable FOG behaviors that could inform more personalized approaches for treatment of FOG and gait impairment in PD.

A temperate deep lake, Lake Kuttara, Hokkaido, Japan (148 m deep at maximum) was completely frozen every winter in the 20th century. However, unfrozen conditions of the lake over winter occurred four times in the 21st century, which is probably due to global warming. In order to understand how thermal regime of the lake responds to climate change, its heat storage change was calculated by estimating heat budget of the lake and monitoring water temperature at the deepest point for September 2012–June 2016. As a result, temporal change of the heat storage from the heat budget was very consistent with that from the direct temperature measurement (determination coefficient R² = 0.827). The 1978–2017 data at a meteorological station near Kuttara indicated that there are significant (less than 5% level) long-term trends for air temperature (0.024 °C/yr) and wind speed (−0.010 m/s/yr). A sensitivity analysis for the heat storage from the heat budget estimate and an estimate of return periods for mean air temperature in mid-winter allow us to conclude that the lake could be unfrozen once per about two year in a decade.

Winter frost injury is a major limiting factor for olive cultivation in temperate regions. The response of olive shoots to freezing stress can be used for selecting resistant genotypes to freezing. The electrolyte leakage (EL) and tetrazolium tests (TZ) are commonly used to evaluate dead tissues in cold stress studies. The temperature-response curve of dead tissues to lethal temperature (LT) is measured with models to calculate LT50 and LT90. In this study, we evaluated the accuracy and efficiency of eighteen non-linear regression models (NLRs) in calculating LT50 and LT90 of freezing stress in different olive cultivars at various stages of dormancy. After evaluating the prediction performance of NLR models, it was found that only eight models were suitable for the purpose of this research out of the 18 models examined. The 2p-logistic and Gompertz models were selected for modeling EL and TZ, respectively. Our research findings indicate that the Roughani, Kawi, and Zard varieties of olive trees exhibit the best performance in cold weather conditions. Our findings provide valuable insights into selecting frost-resistant cultivars and designing effective strategies for cold acclimation in olive cultivation.

Asphaltene-resin-paraffin deposition (ARPD) is a complicated and prevalent issue in oil and gas industry, impacting the efficiency, integrity of petroleum extraction, production, transportation and processing systems. Considering all witnessed ARPD problems in Azerbaijan oil fields, this paper proposed a chemical method and optimized the type and the concentration of chemical inhibitors. And then the effect of selected chemical reagent on inhibiting the ARPD amount and thus en-hancing oil recovery was detected by reservoir simulation during both waterflooding and CO2 flooding production. Three new chemical compounds (namely, Chemical-A, Chemical-B and Chemical-C) were examined in laboratory conditions and their impact on rheological properties of high-paraffin oilfield samples of Azerbaijan (X, Y and Z) were investigated. Experimental results show that Chemical-C with concentration of 600 g/t has the best efficiency for alleviating the problems. After adding Chemical-C to the crude oil, the freezing point of oil is decreased from 12 °C to (-4) °C, ARPD amount is declined from 0.185 to 0.016 g and oil effective viscosity is reduced from 16.2 mPa·s to 3.1 mPa·s. It is determined that for water and CO2 flooding, higher injection pressure resulted in reduced asphaltene precipitation. Adding selected ARPD inhibitor, the oil recovery for waterflooding can increase from 52% to 62%, while it can rise from 55% to 68% for CO2 flooding.

In Poland, the mountainous and hilly regions until recently were territories with unbalanced and poor diet quality, especially in early spring. Wild edible plants were or still are influential in supplementing a human diet. One of the earliest wild edible plants to appear in the growing season, characteristic of a broad swath of Europe from Ukraine to Germany, remains wild (bear) garlic (Allium ursinum L.). This paper assesses the impact of the vegetation site and preservation methods on selected parameters of the chemical composition of bear garlic. In addition, the connection between its genome pattern and chemical composition was assessed. The plants were collected in three locations in the Carpathian foothills, and the reference sample was plants collected in the lowlands. The site from which the raw material was harvested has a statistically significant effect on the chemical composition of the preserved leaves of bear garlic. The profiles of the analysed components differed in the material from a different site. Bear garlic leaves from mountainous areas had more analyzed components than those obtained from the lowland sites. Moreover, the material from the lowlands had a different genetic pattern from the leaves from the mountainous areas..

Freezing/thawing indices are important indicators of dynamics of frozen ground on the Qinghai-Tibet Plateau (QTP), especially for areas with limited observations. Based on the numerical outputs of Community Land Surface Model version 4.5 (CLM4.5) from 1961 to 2010, this study compared the spatial and temporal variations between air freezing/thawing indices (2 m above the ground) and ground surface freezing/thawing indices in permafrost and seasonally frozen ground (SFG) across the QTP, after presenting changes of frozen ground distribution in each decade in the context of warming and wetting. The results indicate that an area of 0.60×106 km2 permafrost of QTP degraded to SFG in 1960s-2000s, and the primary shrinkage period occurred in 2000s. The air freezing index (AFI) and ground freezing index (GFI) decreased dramatically at the rates of 71.00℃·d/decade and 34.33℃·d/decade from 1961 to 2010, respectively. In contrast, the air thawing index (ATI) and ground thawing index (GTI) increased strikingly with the values of 48.13℃·d/decade and 40.37℃·d/decade in the past five decades. Permafrost showed more pronounced changes in freezing/thawing indices since 1990s compared to SFG. The changes of thermal regimes in frozen ground shows closely relations to air warming until the late 1990s, especially in 1998 when the QTP has undergone the most progressively warming. However, a sharp increase in the annual precipitation from 1998 seems to take a stronger control role on thermal degradation in frozen ground than the air warming in 2000s. Meanwhile, the prominent vegetation expansion further promotes thermally unstable of frozen ground in this highly wetting period.

A recently developed thermodynamic theory for the determination of the driving force of crystallization and the crystal–melt surface tension is applied to the ice–water system employing the new Thermodynamic Equation of Seawater TEOS-10. The deviations of approximative formulations of the driving force and the surface tension from the exact reference properties are quantified, showing that the proposed simplifications are applicable for low to moderate undercooling and pressure differences to the respective equilibrium state of water. The TEOS-10 based predictions of the ice crystallization rate revealed pressure-induced deceleration of ice nucleation with an increasing pressure, and acceleration of ice nucleation by pressure decrease. This result is in, at least, qualitative agreement with laboratory experiments and computer simulations. Both the temperature and pressure dependencies of the ice–water surface tension were found to be in line with the le Chatelier–Braun principle, in that the surface tension decreases upon increasing degree of metastability of water (by decreasing temperature and pressure), which favors nucleation to move the system back to a stable state. The reason for this behavior is discussed. Finally, the Kauzmann temperature of the ice–water system was found to amount TK=116K, which is far below the temperature of homogeneous freezing. The Kauzmann pressure was found to amount pK=-212MPa, suggesting favor of homogeneous freezing upon exerting a negative pressure on the liquid. In terms of thermodynamic properties entering the theory, the reason for the negative Kauzmann pressure is the higher mass density of water in comparison to ice at the melting point.

Considering the important demand of fresh water and its scarce availability, water desalination is an interesting technology, producing about 44 Mm³/year worldwide, but, in general the most common desalination techniques are highly energy demanding. Freezing-Melting (F/M) desalination uses just up to 70% less thermal energy, but is the least used process mainly, due to the difficulty of the salt separation. This study proposes a model able to analyses the thermodynamic potential that allows the salt diffusion during the F/M process, using an aqueous solution of sodium chloride. This should allow to obtain a sensitive analysis of the process to promote the separation between the high concentration brine and the ice with liquid separation by physical process. The unidimensional model is based on the evolution of both processes: thermal and mass diffusions, depending on temperature and saline gradients, predicting whether the salt will remain inside the ice or not. Thus, the thermal potential is adjusted to frozen only when the salt has been "pushed" towards the brine. Mostly models have base their results on the assumption of a “certain value of saline concentration of the liquid fraction”, value in which there is great disagreement. In this paper the calculations are based on the concentration in the solid-liquid interface, which has been extensively studied and there is a coincidence in those results, being the main advantage of the proposed model.

Winter rapeseed (Brassica rapa) has good chilling and freezing tolerance. INDUCER OF CBF EXPRESSION1 (ICE) plays a crucial role in cold signaling in plant; however, its role in B. rapa remains unclear. In this study, we identified 41 ICE1 homologous genes from 6 widely cultivated Brassica species. These genes exhibited high conservation, with evolutionary complexity between diploid and allotetraploid species. Cold stress induced ICE1 homolog expression, with differences between strong and weak cold-tolerant varieties. Two novel ICE1 paralogs, BrICE1 and BrICE2 were cloned from B. rapa Longyou 6. Subcellular localization assays showed that they localized to the nucleus, and low temperature did not affect their nuclear localization. Overexpression of BrICE1 and BrICE2 increased cold tolerance in transgenic Arabidopsis, and enhanced reactive oxygen species (ROS) scavenging ability. Furthermore, our data demonstrate that overexpression of BrICE1 and BrICE2 inhibited root growth in Arabidopsis, and low temperatures could induce the degradation of BrICE1 and BrICE2 via the 26S-proteasome pathway. In summary, ICE1 ho-mologous genes exhibit complex evolutionary relationships in Brassica species, and are involved in the CBF pathway, and ROS scavenging mechanism in response to cold stress; these regulating mechanisms might be also responsible for balancing the development and cold defense of B. rapa.

With global warming and accelerated degradation of permafrost, the engineering problems caused by the formation of weak zones between the shallow and permafrost layers of soil–rock mixture (S-RM) slopes in permafrost regions have become increasingly prominent. To explore the influence of rock content on the shear strength of the S-RM freezing–thawing interface, the variation in the shear strength for different rock content is studied herein using direct shear tests. In addition, a 3D laser scanner is used for obtaining the topography of the shear failure surface. Combined with the analysis results of the shear band-particle calculation model, the influence of the rock content on the shear strength of the interface is explored. It was found that the impact threshold of the rock content on the interface strength and failure mode is approximately 30%, when the rock content (R) is &gt; 30% and that the shear strength increases rapidly with increasing rock content. When R ≤ 30%, the actual shear plane is similar to waves; when R &gt; 30%, the shear plane appears as gnawing failure. The shear strength of S-RM freezing–thawing interface mainly comes from the bite force and friction between particles. The main reason for the increase in shear strength with increasing rock content is the increase in bite force between particles, which makes the ratio of bite force to friction force approximately 1:1.

Frost heave, which is the volumetric expansion of frozen soil, has great ecological significance, since it creates water storage spaces in soils at the beginning of the growing season in cold temperate forests. To understand the characteristics of frost heave in seasonally frozen soil and the factors that impact its extent, we investigated the frost heave rates of forest soil from different depths and with different soil moisture contents, using both lab-based simulation and in situ measurement in a broadleaved Korean pine forest in the Changbai Mountains (northeastern China). We found that frost heave was mainly affected by soil moisture content, soil type, and gravitational pressure. Frost heave rate increased linearly with soil moisture content, and for each 100% increase in soil moisture content, the frost heave rate increased by 41.6% (loam, upper layer), 17.2% (albic soil, middle layer), and 4.6% (loess, lower layer). Under the same soil moisture content, the frost heave rate of loam was highest, whereas that of loess was lowest, and the frost heave of the uppermost 15 cm, which is the biologically enriched layer, accounted for ~55% of the frost heave. As a result, we determined the empirical relationship between frost heave and freezing depth, which is important for interpreting the effects of frost heave on increases in the storage space of forest soils and for calculating changes in soil porosity.

Inducer of C-Repeat Binding Factor (CBF) expression (ICE), a well-known MYC-type bHLH transcription factor, plays a crucial role in the ICE-CBF-cold regulated (COR) cold signaling pathway. Brassica rapa (B. rapa), an important oilseed and vegetable crop, exhibits strong chilling and freezing tolerance. However, the exact molecular mechanisms by which ICE1 functions in B. rapa remain unclear. Here, 41 ICE1 homologous genes were identified from six widely cultivated Brassica species using a bioinformatics approach. These genes exhibit high conservation but reveal evolutionary complexity between diploid and allotetraploid species. Low-temperature stress induced ICE1 homolog expression, with patterns differing between strong and weak cold-tolerant varieties. Two novel ICE1 paralogs, BrICE1 and BrICE2, were cloned from Longyou 6. They positively regulate cold tolerance in B. rapa via a CBF-dependent pathway, contributing to reactive oxygen species (ROS) scavenfging and osmotic adjustment. Immunoblot analysis revealed that low temperatures induce BrICE1 and BrICE2 degradation via the 26S-proteasome pathway. In summary, ICE1 exhibits complex evolutionary relationships in Brassica species. BrICE1 and BrICE2 positively regulate cold tolerance via the CBF-dependent pathway and ROS scavenging mechanism and are also responsible for balancing the development and cold defense of B. rapa.

Despite being stored at 113 K and atmospheric pressure, LNG cold potential is not exploited to reduce green ships’ energy needs. An innovative system based on three Organic Ranking Cycles integrated into the regasification equipment is proposed to produce additional power and recover cooling energy from condensers. A first-law analysis identified ethylene and ethane as suitable working fluids for the first and the second ORC making available freshwater and ice. Propane, ammonia and propylene could be arbitrarily employed in the third ORC for air-conditioning. An environmental analysis that combines exergy efficiency, ecological indices and hazard aspects for marine environment and ship’s passengers, indicated propylene as safer and more environmentally friendly. The exergy analysis confirms more than 20% of the LNG potential can be recovered from every cycle to produce a net clean power of 76 kW, whereas 270 kW can be saved by recovering condensers' cooling power to satisfy some ship needs. Assuming the sailing mode, a limitation of 162 kg in LNG consumptions was determined, avoiding the emission of 1584 kg of CO2 per day. Marine thermal pollution is reduced by 3.5 times by recovering the working fluids' condensation heat for the LNG pre-heating.

Ice-binding proteins are crucial for adaptation of various organisms to low temperatures. Some of these, called antifreeze proteins, are usually thought to inhibit growth and/or recrystallization of ice crystals. However, prior to these events, ice must somehow appear in the organism, either coming from outside or forming inside through the nucleation process. Unlike most other works, our paper is focused on ice nucleation rather than the behavior of the already existing ice. The nucleation kinetics is studied both theoretically, with special attention paid to ice nucleation on ice-binding surfaces, and experimentally. For experimental studies, we use the ice-binding protein mIBP83, a previously constructed mutant of a spruce budworm Choristoneura fumiferana antifreeze protein. We show that mIBP83 does not affect the ice nucleation temperature in the buffer in test tubes, but hinders the impact of potent ice nucleators of various chemical natures, namely CuO powder and ice-nucleating bacteria Pseudomonas syringae. Additional experiments on human cells show that mIBP83 is concentrated in some regions, but only in cooled cells. Thus, the antifreeze protein not only binds to ice, but also blocks various sites that act or can act as ice nucleators. Such ice-preventing binding may be the crucial biological task of antifreeze proteins.
