Targeted therapy indications for actionable variants in non-small cell lung cancer (NSCLC) have primarily been studied in Caucasian populations, with limited data on Latin American patients. This study utilized a 52-gene next-generation sequencing (NGS) panel to analyze 1,560 tumor biopsies from NSCLC patients in Chile, Brazil, and Peru. RNA sequencing reads, and DNA coverage were correlated to improve the detection of the actionable MET exon 14 skipping variant (METex14). The pathogenicity of MET variants of uncertain significance (VUS) was assessed using bioinformatic methods based on their predicted driver potential. The effects of the predicted driver VUS T992I and H1094Y on MET signaling activation, proliferation, and migration were evaluated in HEK293T, BEAS-2B, and H1993 cell lines. Subsequently, MET inhibitors were tested in 2D and 3D cell cultures, and drug affinity was determined using 3D structure simulations. The prevalence of MET variants in the South American cohort was 8%, and RNA-based diagnosis detected 27% more cases of METex14 than DNA-based methods. Notably, 20% of METex14 cases with RNA reads below the detection threshold were confirmed by DNA analysis. The novel actionable T992I and H1094Y variants induced proliferation and migration through c-Met/Akt signaling. Both variants showed sensitivity to crizotinib and savolitinib, but the H1094Y variant exhibited reduced sensitivity to capmatinib. These findings highlight the importance of RNA-based METex14 diagnosis and reveal the drug sensitivity profiles of novel actionable MET variants from an understudied patient population.

Background: Disruption of alternative splicing (AS) is frequently observed in cancer and it might represent an important signature for tumor progression and therapy. Exon skipping (ES) represents one of the most frequent AS events and in non-small cell lung cancer (NSCLC) MET exon 14 skipping was shown to be targetable. Methods: We constructed a neural network (NN) specifically designed to detect MET exon 14 skipping events using RNAseq data. Furthermore, for discovery purpose we also developed a sparsely connected autoencoder to identify uncharacterized MET isoforms. Results: The NN had 100% Met exon 14 skipping detection rate, when tested on a manually curated set of 690 TCGA bronchus and lung samples. When globally applied to 2605 TCGA samples, we observed that the majority of false positives was characterized by a blurry coverage of exon 14, but interesting they share a common coverage peak in the second intron and we speculate that this event could be the transcription signature of a LINE1-MET fusion. Conclusions: Taken together our results indicate that neural networks can be an effective tool to provide a quick classification of pathological transcription events and sparsely connected autoencoders could represent the basis for the development of an effective discovery tool.

The limits for recording weak signals for electrochemical seismic sensors are determined by their self-noise. At the same time, the noise nature is complex and not fully understood. In this work, for the first time, in addition to the frequency dependences of the noise amplitude, the difference in the phases of the noise currents of the cathodes of an electrochemical sensor has been studied. It has been established that at low frequencies the cathode currents change in an-tiphase. As the frequency increases, the phase difference becomes random. At the same time, the frequency at which the phase difference changes depend on the interelectrode distance; the clos-er the electrodes are located to each other, the greater it is. Physically, this effect is explained by the transition from noise caused by the integral flow of the working fluid to noise associated with vortex flows, the size of which depends on frequency. Thus, a new way of studying noise makes it possible to identify its physical nature and develop new methods of reduction.

The MET proto-oncogene encodes a pivotal tyrosine kinase receptor, binding the hepatocyte growth factor (HGF, also known as scatter factor, SF) and governing essential biological processes such as organogenesis, tissue repair, and angiogenesis. The pleiotropic physiological functions of MET explain its diverse role in cancer progression in a broad range of tumors; genetic/epigenetic alterations of MET drive tumor cell dissemination, metastasis, and acquired resistance to conventional and targeted therapies. Therefore, targeting MET emerged as a promising strategy, and many efforts were devoted to identifying the optimal way of hampering MET signaling. Despite encouraging results, however, the complexity of MET's functions in oncogenesis yields intriguing observations, fostering a humbler stance on our comprehension. This review explores recent discoveries concerning MET alterations in cancer, elucidating their biological repercussions, discussing therapeutic avenues, and outlining future directions. By contextualizing the research question and articulating the study's purpose, this work navigates MET biology's intricacies in cancer, offering a comprehensive perspective.

Nanoscale structuring/printing is of interest for range of applications in 3D subtractive and additive manufacturing (3D^+/-). Basic principles of light field enhancement and control at the nanoscale are overviewed in this section/chapter for bulk, surface, and localised plasmons (1D, 2D, and 3D localisation, respectively). All these plasmons are resonant phenomena which have common Lorentzian spectral lineshape which relates refractive and absorption properties as well as defining the phase of transmitted and scattered light. Localisation of light at the nanoscale creates the possibility of modification with matching resolution. Harnessing this light enhancement can be demonstrated as a "nano-pen" for direct write nanolithography.

Urban public space comfort is vital for enhancing the quality of life and perceptions of livability, especially as climate change increasingly impacts outdoor thermal environments. This study investigates the use of ENVI-met, a 3D microclimate simulation tool, to assess thermal comfort concerning demographic factors like age and gender. While ENVI-met is often used to analyze urban microclimates, there is limited research on its application for simulating heat emission variations across different demographic groups. The findings highlight significant disparities in thermal stress vulnerability: elderly individuals are at higher risk due to elevated maximum PET values, while children experience broader ranges of thermal stress and higher baseline values. Despite cooling effects in the evening, children continue to face greater stress. The study emphasizes the need for tailored management strategies, as morning PET values are consistent but afternoon data reveals children’s ongoing vulnerability. Overall, the research calls for further investigation to enhance urban planning, thermal comfort strategies, and public health interventions amid rising temperatures. It advocates for expanding ENVI-met's capabilities to include diverse demographic factors, promoting a more comprehensive approach to urban resilience and well-being.

CD8+ cytotoxic T lymphocytes (CTLs) play a crucial role in anti-tumor immunity. In a previous study, we identified a subset of murine effector CTLs expressing the hepatocyte growth factor (HGF) receptor c-Met (c-Met+ CTLs) that are endowed with enhanced cytolytic capacity. HGF directly inhibited the cytolytic function of c-Met+ CTLs, both in 2D in vitro assays and in vivo, leading to reduced T cell responses against metastatic melanoma. To further investigate the role of c-Met+ CTLs in a three-dimensional (3D) setting, we studied their function within B16 melanoma spheroids and examined the impact of cell-cell contact on the modulation of inhibitory checkpoint molecules expression such as KLRG1, PD-1 and CTLA-4. Additionally, we evaluated the cytolytic capacity of human CTLs clones expressing c-Met (c-Met+) and compared it to c-Met− CTLs clones. Our results indicated that, similar to their murine counterparts, c-Met+ human CTLs clones exhibited increased cytolytic activity compared to c-Met− CTLs clones, and this enhanced function was negatively regulated by the presence of HGF. Taken together, our findings highlight the potential of targeting the HGF/c-Met pathway to modulate CTL-mediated anti-tumor immunity. This research holds promises for developing strategies to enhance the effectiveness of CTL-based immunotherapies against cancer.

Savolitinib is a highly selective small molecule inhibitor of the mesenchymal epithelial transition factor (MET) tyrosine kinase, primarily developed for the treatment of non-small cell lung cancer (NSCLC) with MET mutations. It is also being investigated as a treatment for breast, head and neck, colorectal, gastric, pancreatic, and other gastrointestinal cancers. In both preclinical and clinical studies, it has demonstrated efficacy in lung, kidney, and stomach cancers. Savolitinib is an oral anti-cancer medication taken as a 600 mg dose once daily. It can be used as a monotherapy in patients with non-small cell lung cancer with MET mutations, and in combination with epidermal growth factor receptor (EGFR) inhibitors for patients who have developed resistance to them. Furthermore, savolitinib has shown positive results in gastric cancer treatment, particularly in combination with docetaxel. As a result, this review aims to validate its efficacy in NSCLC and suggests its potential application in other gastrointestinal cancers, such as pancreatic cancer, based on related research in gastric and renal cancer.

This study assesses Human Thermal Comfort in two selected areas: a Green Infrastructure (GI) area represented by a garden and a high-rise building area, in the Central Business District (CBD) of Melbourne, Australia. Three-dimensional microclimatic modelling software, ENVI-met version 4 was used to simulate the microclimate. The indices of Predicted Mean Vote (PMV), Physiological Equivalent Temperature (PET) and Universal Temperature Climate Index (UTCI) were used to quantify the level of thermal comfort in the research areas. The simulation results showed that at midday, the difference in temperature between the garden area and the high-rise building area was approximately 1°C. Increasing temperatures at midday led to a change in the level of thermal comfort for both the areas, even though it was not significant. In general, the thermal perception in the GI area was slightly ‘cooler’ than in the high-rise building area. The results of the study indicated the important role of GI in improving the thermal comfort in urban areas.

(1) Background: P-cadherin (CDH3) is a transmembrane protein that plays a crucial role in maintaining the structural integrity of epithelial tissue and homeostasis. Its role in carcinogenesis remains a subject of debate, as its behavior can vary depending on the molecular context and the specific tumor cell model under study. In this study, we explored the role of P-cadherin in head and neck squamous cell carcinoma (HNSCC) and the mechanism underlying its function. (2) Methods: We analyzed P-cadherin expression in HNSCC patients using The Cancer Genome Atlas (TCGA) database. For in vitro functional analysis, we conducted proliferation, migration, invasion, and western blot assays after either suppressing or overexpressing P-cadherin. For in vivo functional analysis, we utilized mouse xenograft models. (3) Results: P-cadherin was significantly overex-pressed in tumor samples compared to normal samples in the TCGA-HNSCC cohort. P-cadherin knockdown resulted in decreased proliferation, migration, and invasion compared to control cells, while P-cadherin overexpression increased cell proliferation and migration in HNSCC cells. We discovered that c-Met functioned as an upstream regulator of P-cadherin. Surprisingly, we found that P-cadherin knockdown increased the phosphorylation of c-Met and STAT3. Combining P-cadherin siRNA with the c-Met inhibitor SU11274 resulted in a more effective reduction in HNSCC cell growth, both in vitro and in vivo, compared to either treatment alone. (4) Conclusions: Our study uncovered a previously unknown aspect of P-cadherin-mediated c-Met regulation. The enhanced activation of c-Met/STAT3 following P-cadherin inhibition could be responsible for the survival of resistant tumor cells. Therefore, dual inhibition of P-cadherin and c-Met may be a potentially effective approach for treating HNSCC.

The heavy metal lead (Pb) can irreversibly damage the human nervous system. To help understand Pb-induced damage, we applied a genetically encoded Förster resonance energy transfer (FRET)-based Pb biosensor Met-lead 1.44 M1 to two living systems to monitor the concentration of Pb: induced pluripotent stem cell (iPSC)-derived cardiomyocytes as a semi-tissue platform, and Drosophila melanogaster fruit flies as an in vivo animal model. Different FRET imaging modalities were used to obtain FRET signals, which represented the presence of Pb in the tested samples in different spatial dimensions. Using iPSC-derived cardiomyocytes, the relationship between beating activity (20–24 beats per minute, bpm) determined from the fluctuation of fluorescent signals and the concentrations of Pb represented by the FRET emission ratio values of Met-lead 1.44 M1 was revealed from simultaneous measurements. Pb (50 μM) affected the beating activity of cardiomyocytes, whereas two drugs that stop the entry of Pb differentially affected this beating activity: verapamil (2 μM) did not reverse the cessation of beating, whereas 2-APB (50 μM) partially restored this activity (16 bpm). The results clearly demonstrate a potential of this biosensor system as an anti-Pb drug screening application. In the Drosophila model, Pb was detected within the adult brain or larval central nervous system (Cha-gal4>UAS-Met-lead 1.44 M1) using fast epifluorescence and high-resolution two-photon 3D FRET ratio image systems. The tissue-specific expression of Pb biosensors provides an excellent opportunity to explore the possible Pb-specific populations within living organisms. We believe that this integrated Pb biosensor system can be applied to the prevention of Pb poisoning and advanced research on Pb neurotoxicology.

Research on urban heat mitigation has been growing in recent years with many of the studies focusing on green infrastructure (GI) as a strategy to mitigate the adverse effects of Urban Heat Island (UHI). This paper aims at presenting a review of the range of findings from GI research for urban heat mitigation through a review of scientific articles published during the years 2009-2019. This research includes a review of the different types of GI and its contribution for urban heat mitigation and human thermal comfort. In addition to analyzing different mitigation strategies, numerical simulation tools that are commonly used are also reviewed. It is seen that ENVI-met is one of the modelling tools that is considered as a reliable tool to simulate different mitigation strategies and hence has been widely used in the recent past. Considering its popularity in urban microclimate studies, this article also provides a review of ENVI-met simulation results that were reported in the reviewed papers. It was observed that the majority of the research was conducted on a limited spatial scale and focused on temperature and human thermal comfort.

Background: Capmatinib and savolitinib, selective MET inhibitors, are widely used to treat various MET-positive cancers. In this study, we aimed to determine the effects of these inhibitors on MET-amplified gastric cancer (GC) cells. Methods: After screening 37 GC cell lines, the following cell lines were found to be MET-positive with copy number variation > 10: SNU-620, ESO51, MKN-45, SNU-5, and OE33 cell lines. Nest, we assessed the cytotoxic response of these cell lines to capmatinib or savolitinib alone using cell counting kit-8 and clonogenic cell survival assays. Western blotting was performed to assess the effects of capmatinib and savolitinib on the MET signaling pathway. Xenograft studies were performed to evaluate the in vivo therapeutic efficacy of savolitinib in MKN-45 cells. Results: Savolitinib and capmatinib exerted anti-proliferative effects on MET-amplified GC cell lines in a dose-dependent manner. Savolitinib inhibited the phosphorylation of MET and downstream signaling pathways, such as the protein kinase B (AKT) and extracellular signal-regulated kinase (ERK) pathways, in MET-amplified GC cells. Additionally, savolitinib significantly decreased the number of colonies formed on the soft agar and exerted dose-dependent anti-tumor effects in a MKN-45 GC cell xenograft model. Furthermore, combination of trastuzumab and capmatinib exhibited enhanced inhibition of AKT and ERK activation in human epidermal growth factor receptor-2 (HER2)- and MET-positive OE-33 cells. Conclusions: Therefore, targeting MET with savolitinib and capmatinib efficiently suppressed the growth of MET-amplified GC cells. Moreover, these MET inhibitors exerted synergistic effects with trastuzumab on HER2- and MET-amplified GC cells.

The diversification of life-histories is mediated by cooperation, innovations of biological information, modularity, and heterochrony in developmental processes. These processes are defined, contextualized, and exemplified, studying the evolution of coloniality (i.e. life-history involving modularization of the multicellular individual) in the family of benthic tunicates Styelidae. This study proposes that in these colonial tunicates there is an inter-generational division of labor, where one generation is feeding, a second is developing by morphogenetic processes, and a third is aging by programmed cell death and phagocytosis. The communication system developed in these colonies is mediated, by changes in proportion, location, and gene expression of specialized blood cells. Colonial life-history in animals is related to the reduction of individual size; development of extra-corporeal tissues to interconnect zooids; the inter-generational division of labor; and the reduction of zooid’s individuality. Processes analogous with the widely accepted major evolutionary transitions (METs), suggesting that coloniality could be studied as a MET. The understanding of colonial life-histories could provide information about key mechanisms for life diversification.

Urban air pollution is a significant threat to both the environment and public health. Primarily originating from traffic emissions, particulate matter is identified as the most prevalent and toxic pollutant in cities. In recent years, street vegetation has emerged as a nature-based solution to improve urban air quality. However, to achieve this goal, proper placement of vegetation is crucial. This study critically investigates how different street tree placement strategies affect the local air quality, based on a model calibrated using existing air quality measurements from one of the most polluted streets in Glasgow. We then test the local air quality effects of tree orientation, spacing, shape, porosity, size, species, and wind directions. The findings suggest that trees effectively enhance air quality, particularly when implemented on the leeward side of the street. To achieve optimal effectiveness, the spacing of trees require careful alignment with the aspect ratio, as haphazard planting could lead to unfavorable results. The insights on improving air quality in Glasgow's street canyons, complements the objectives of the ‘Glasgow Avenues Programme’ and can be included in its Low Emission Zones to co-benefit the environment and the wellbeing of the local community.

The Urban Heat Island (UHI) is one of the more serious consequences of urbanization resulting in impacts on thermal comfort levels, heat stress, and even mortality. For Municipal Beirut, implementation of “cool” surface materials and green spaces have been recommended to counterbalance the UHI. This paper builds on previous findings on the topic of non-constructible parcels within the district of Bachoura in Municipal Beirut and examines the possibility of implementing “cool” surface or paving materials and urban vegetation which can improve thermal conditions especially during the summer period and with the viewto project the positive findings of this case study to the entire Municipal Beirut area. A numerical analysis using ENVI-met 4.0 investigates the thermal performance of these non-constructibles further to implementation of high reflective surfaces and urban vegetation within a broad neighborhood scale in Bachoura. Results show reductions in ambient temperatures up to 1K on a summer day.. Within the framework of an integrated approach to planning, this form of urban acupuncture aims for substantial UHI reduction. Energy performance of buildings further to implementation of these mitigation measures is also recommended for future studies and to validate the findings in this paper.

In this work, we have proposed two new methods for the synthesis of [TcO2L4]+ (where L = im-idazole (Im), methylimidazole (MeIm)) complexes using thiourea (Tu) and Sn(II) as reducing agents. The main and by-products of the reactions were determined, and possible reaction mechanisms were proposed. We have shown that the reduction of Tc(VII) with thiourea is ac-companied by the formation of the Tc(III) intermediate and further oxidation to Tc(V). The reac-tion conditions changing can lead to the formation of Tc(VII) and Tc(IV) salts. Seven new crystal structures are described in this work: Tc(V) complexes, salts with Tc(VII) and Tc(IV) anions. For halide salts of Tu the cell parameters were determined. In all obtained compounds, except for [TcO2(MeIm)4]TcO4, there are π-stacking interactions between aromatic rings. An increase in the anion size lead to weaken intermolecular interactions. Halogen bonds and anion-π interactions have also been found in hexahalide-containing compounds. The Hirshfeld surface analysis showed that the main contribution to the crystal packing is made by van der Waals interactions of the H···H type (42.5–55.1%), H···C/C ···H (17.7–21.3%) and hydrogen bonds, which contribute 15.7–25.3% in total.

Most methods for measuring environmental lead (Pb) content are time consuming, expensive, hazardous, and restricted to specific analytical systems. To provide a facile, safe tool to detect Pb, we created pMet-lead, a portable fluorescence resonance energy transfer (FRET)-based Pb biosensor. pMet-lead comprises a 3D-printed frame housing a 405-nm laser diode — an excitation source for fluorescence emission images (YFP and CFP) — accompanied by optical filters, a customized sample holder with a Met-lead 1.44 M1 (the most recent version)-embedded biochip, and an optical lens aligned for smartphone compatibility. Measuring the emission ratios (Y/C) of the FRET component enables Pb detection with a dynamic range of nearly 2 (1.96), pMet-lead/Pb dissociation constant (K_d) 45.62 nM, and limit of detection 40 nM (0.832 μg/dL, 8.32 ppb). To mitigate earlier problems with lack of selectivity for Pb vs. zinc, we preincubated samples with tricine, a low-affinity zinc chelator. We validated pMet-lead measurements of characterized laboratory samples and unknown samples from six regions in Taiwan by inductively coupled plasma mass spectrometry (ICP-MS). Notably, two unknowns had Y/C ratios significantly higher than that of the control (3.48 ± 0.08 and 3.74 ± 0.12 vs. 2.79 ± 0.02), along with Pb concentrations (10.6 ppb and 15.24 ppb) above the WHO-permitted level of 10 ppb in tap water, while the rest four unknowns showing no detectable Pb upon ICP-MS. These results demonstrate that pMet-lead provides a rapid, sensitive means for on-site Pb detection in water from the environment and in living/drinking supply systems to prevent potential Pb poisoning.

Extracellular electron transfer (EET) is key to the success of microbial fuel cells (MFCs). Clostridium sp. often occurs in MFC anode communities, but its ability to perform EET remains controversial. We have employed Clostridium pasteurianum DSM 525 as biocatalyst in a glycerol-fed MFC, designated MFC_DSM. We have also followed the EET of this biocatalyst in the presence of a mediator, namely soluble neutral red (NR), soluble methyl viologen (MV), neutral red film (FNR), or methyl viologen film (FMV). MFC_DSM provided power and current densities (j) of 0.39 mW.cm^-2 and 2.47 mA.cm^-2, respectively, which evidenced that the biocatalyst performs direct electron transfer (DET). Introducing 150.0 µM NR or MV into the MFC_DSM improved the current density by 7.0- and 3.7-fold (17.05 and 8.45 mA.cm^-2), respectively. After 20 cyclic voltammetry (CV) cycles, the presence of FNR in the MFC_DSM anodic chamber provided almost twofold higher current density (30.76 µA.cm^-2) compared the presence of NR in the MFC_DSM. Introducing MV or FMV into the MFC_DSM anodic chamber gave practically the same current density after 10 CV cycles. The MFC_DSM anodic electrode might interact with FMV weakly than with FNR, so FNR is more promising to enhance C. pasteurianum DSM 525 EET within MFC_DSM.

As urban areas expand and populations grow, environmental challenges such as the urban heat island effect, air pollution, and light pollution intensify. The urban heat island effect exacerbates extreme heat conditions, leading to prolonged periods of unhealthy and hazardous environments for both humans and other species. This study investigates the thermal environment associated with the urban heat island effect, focusing on the role of trees in mitigating this issue. Using ENVI-met simulations, the study examines various factors influencing tree cooling effectiveness, including seasonal variations, building shading, transpiration rates, tree placement, and spacing. A new tree-planting strategy is developed based on these findings, aiming to enhance thermal comfort. The study compares the thermal environment of sidewalks under the new tree-planting scheme with the existing arrangement across different months. Results indicate that the new scheme reduces UTCI temperatures by 2.2°C on the hottest day, 0.97°C on the coldest day, and 1.52°C annually in the study area of Los Angeles, demonstrating that the cooling benefits of trees in hot weather outweigh the potential drawbacks in cold weather, highlighting its potential to mitigate the urban heat island effect.

Oil spills may have devastating effects on marine ecosystems, public health, the economy, and coastal communities. Therefore, scientific literature contains various up-to-date, advanced oil spill predictive models, capable to simulate the trajectory and evolution of an oil slick generated by the accidental release from ships, hydrocarbons production, or other activities. To predict in near real time oil spill transport and fate with increased reliability these models are usually coupled operationally to synoptic meteorological, hydrodynamic, and wave models. The present study reviews the available different met-ocean forcings that have been used in oil spill modeling, simulating hypothetical or real oil spill scenarios, worldwide. Nine state-of-the-art oil spill models are critically examined in terms of the met-ocean data used as forcing inputs in the simulation of twenty-four case studies. Results illustrate that most oil spill models are coupled to different resolution, forecasting meteorological and hydrodynamic models, posing, however, limited consideration in the forecasted wave field (expressed as the significant wave height, the wave period and the Stokes drift) that may affect oil transport, especially at the coastal areas. Moreover, the majority of oil spill models lacks any linkage to the background biogeochemical conditions, hence, limited consideration is given in processes like oil biodegradation, photo-oxidation and sedimentation. Future advancements in oil spill modeling should be directed towards the full operational coupling with high-resolution atmospheric, hydrodynamic, wave, and biogeochemical models, improving our understanding in the relative impact of each physical and oil weathering process.

Background/Aim: To evaluate the association between baseline [18F]FDG-PET/CT tumor burden parameters and disease progression rate after first-line target therapy or immunotherapy in advanced melanoma patients. Materials and Methods: 44 melanoma patients who underwent [18F]FDG-PET/CT before first-line target therapy (28/50) or immunotherapy (16/50) were retrospectively analyzed. Whole-body and per-district metabolic tumor volume (MTV) and total lesion glycolysis (TLG) were calculated. Therapy response was assessed according to RECIST 1.1 on CT scan at 3 (early) and 12 (late) months. PET parameters were compared with Mann-Whitney test. Optimal cut-offs for predicting progression were defined using the ROC curve. PFS and OS were studied using Kaplan-Meier analysis. Results: Median(IQR) MTVwb and TLGwb were 13.1 mL and 72.4 respectively. Non-responders patients were 38/44, 26/28 and 12/16 at early evaluation, and in 33/44, 21/28 and 12/16 at late evaluation in the whole-cohort, target and immunotherapy subgroup respectively. At late evaluation, MTVbone and TLGbone were higher in non-responders compared to responder patients (all p<0.037) in the whole-cohort and target subgroup and also MTVwb and TLGwb (all p<0.022) in target subgroup. No significant differences were found for immunotherapy subgroup. No metabolic parameters were able to predict PFS. Controversy, MTVlfn, TLGlfn, MTVsoft+lfn, TLG-soft+lfn, MTVwb and TLGwb were significantly associated (all p<0.05) with OS in both the whole-cohort and target therapy subgroup. Conclusion: Higher values of whole-body and bone metabolic parameters were correlated with poorer outcome, while higher values of whole-body, lymph node and soft tissue metabolic parameters were correlated with OS.

This paper investigates urban microclimate in relation to the typical major streets and mobility layout in Turin, i.e. boulevards with lateral minor roads, called “controviali”, an almost unique urban mobility layout. The aim is to examine the possibility of intervention in relation to the type and scope of mobility for microclimate mitigation and climate adaptation purposes. To this, this paper evaluates to which extent urban morphology and mobility features contribute to determining microclimatic conditions, to finally identify site-specific prioritised strategies to cope with its mitigation. This research is structured in two phases. The first phase is dedicated to urban-scale analysis of the streets with “controviali”, resulting in the creation of a database containing their properties and typology, to finally derive six clusters of boulevards. The second phase involves the selection of one representative case study per cluster and simulation of their microclimate conditions, performed by the software ENVI-met. The results are discussed in relation to the parameters proven in the prior literature review to have a major impact on the microclimate. The assessment of the results indicated which elements affect the climatic conditions the most, allowing for the proposal of different prioritised mitigation strategies in the conclusion.

We have previously developed several successful decellularization strategies yielding porcine cardiac extracellular matrices (pcECMs), which exhibit tissue-specific bioactivity and bioinductive capacity when cultured with various pluri- and multipotent stem cells. Here, we studied the tissue-specific effects of the pcECM on seeded human mesenchymal stem cells (hMSCs) phenotype using reverse transcribed quantitative polymerase chain reaction (RT-qPCR) arrays for cardio-vascular related genes. We further corroborated interesting findings at the protein level (flow cytometry and immunological stains) as well as bioinformatically using several mRNA sequencing and protein databases of normal and pathologic adult tissue expression, as well as during human embryonic organogenesis. We discovered that upon seeding of human mesenchymal stem cells (hMSCs) on the pcECM they displayed partial MET toward endothelial phenotypes (CD31+) and morphologies, which were preceded by an early spike (~day 3 onward after seeding) in HAND2 expression at both the mRNA and protein levels compared to plate controls. CRISPR-Cas9 knockout (KO) of HAND2 and its associated antisense long non-coding RNA (HAND2-AS1) regulatory region resulted in proliferation arrest, hypertrophy, and senescent-like morphology. Bioinformatic analyses revealed that HAND2 and HAND2-AS1 are highly correlated in expression, are expressed in many different tissue types albeit at distinct yet tightly regulated expression levels. Deviation (down or up regulation) from these basal tissue expression levels are associated with a long list of pathologies. We thus suggest that HAND2 expression levels may finetune cell plasticity possibly affecting senescence and mesenchymal-to-epithelial transition states, through yet unknown mechanisms. Targeting this pathway may represent a promising new therapeutic approach for a wide range of diseases, including cancer, degenerative disorders, and aging. Nevertheless, further investigations are required to better understand the molecular players involved, potential inducers and inhibitors of this pathway, and eventually potential therapeutic applications.

Epithelial-mesenchymal transition (EMT) is a reversible cellular program that transiently places epithelial (E) cells into pseudo-mesenchymal (M) cell states. The malignant progression and resistance of many types of carcinomas depends on EMT activation, partial EMT and hybrid E/M status in neoplastic cells. EMT is activated by tumor microenvironmental TGFβ signal and EMT-inducing transcription factors, such as ZEB1/2 in tumor cells. However, reverse EMT factors are less studied. We demonstrate that transcription factor SCAND1 can revert mesenchymal and hybrid E/M phenotype of cancer cells to a more epithelial, less invasive status and inhibit their proliferation and migration. SCAND1 is a SCAN domain-containing protein and hetero-oligomerizes with SCAN-zinc finger transcription factors, such as MZF1, for accessing DNA and transcriptional co-repression of target genes. We found that SCAND1-MZF1 co-expression and interaction correlated with maintaining epithelial features, whereas the simultaneous loss of SCAND1 and MZF1 correlated with mesenchymal features of tumor cells. Overexpression of SCAND1 over endogenous MZF1 in DU-145 prostate cancer cells reverted their hybrid E/M status into cobblestone morphology with increased epithelial adhesion by E-cadherin and β-catenin relocation. Consistently, co-expression analysis in TCGA PanCancer Atlas revealed that both SCAND1 and MZF1 co-express and are negatively correlated with EMT driver genes, including CTNNB1, ZEB1, ZEB2 and TGFBR, in prostate tumor specimens. In addition, SCAND1 overexpression suppressed tumor cell proliferation by reducing the MAP3K-MEK-ERK signaling pathway. Of note, SCAND1-overexpressing DU-145 cells migrated slower than control cells with decreased lymph node metastasis of prostate cancer in a mouse tumor xenograft model. Kaplan-Meyer analysis showed high expression of MZF1 and SCAND1 to correlate with better prognoses in pancreatic cancer and head and neck cancers, although with poorer prognosis in kidney cancer. Overall, these data suggest that the combination of SCAND1-MZF1 complexes may revert the EMT mechanism in cancer to establish an epithelial phenotype. These effects seem to include co-repression of EMT-driver genes and suppression of tumor cell proliferation via inhibition of the MAP3K-MEK-ERK signaling pathway.

The epithelial-mesenchymal (E/M) hybrid state has emerged as an important mediator of elements of cancer progression, facilitated by epithelial mesenchymal plasticity (EMP). We review here evidence for the presence, prognostic significance, and therapeutic potential of the E/M hybrid state in carcinoma. We further assess modelling predictions and validation studies to demonstrate stabilised E/M hybrid states along the spectrum of EMP, as well as computational approaches for characterising and quantifying EMP phenotypes, with particular attention to the emerging realm of single-cell approaches through RNA sequencing and protein-based techniques.

The Epithelial-Mesenchymal-Transition (EMT) phenotype, identified as a significant clinical indicator in cancer, manifests as a biological process where cells transition from epithelial to mesenchymal characteristics. Physiologically, EMT plays a crucial role in tissue remodeling, promoting healing, repair, and responding to various tissue damages. This study investigates the impact of BNE-RRC on oral cancer cells (KB) and reveals its significant effects on cancer cell growth, migration, invasion, and EMT. BNE-RRC induces the epithelial-like morphology in KB cells, effectively reversing EMT to Mesenchymal-Epithelial Transition (MET). Extraordinarily, sustained culture of cancer cells with BNE-RRC for 14 days maintains an epithelial status even after treatment withdrawal, suggesting BNE-RRC as a potential therapeutic agent for cancer. These findings highligh the promise of BNE-RRC as a comprehensive therapeutic agent in cancer treatment by inhibiting cancer cell growth, migration, and invasion, while also orchestrating a reversal of the EMT process. The study proposes that BNE-RRC could be effective agent for cancer treatment.

The emergence of acquired resistance to EGFR-TKIs is almost inevitable even after a remarkable clinical response. Secondary mutations such as T790M and C797S are responsible for the resistance to 1st/2nd generation (1/2G) TKIs and 3G TKIs, respectively. To overcome both the T790M and C797S mutations, novel 4G EGFR-TKIs are now under early clinical development. In this study, we evaluated the efficacy of a 4G EGFR-TKI in the treatment of lung cancer with EGFR mutation as well as explored resistance mechanisms to a 4G TKI. First, we compared the efficacies of seven TKIs including a 4G TKI, BI4020, against Ba/F3 cell models that simulate resistant tumors after frontline osimertinib treatment failure because of a secondary mutation. We also established acquired re-sistant cells to BI4020 by chronic drug exposure. Ba/F3 cells with an osimertinib-resistant secondary mutation were refractory to all 3G TKIs tested (alflutinib, lazertinib, rezivertinib, almonertinib, and befotertinib). BI4020 inhibited the growth of C797S-positive cells; however it was not effective against L718Q-positive cells. Erlotinib was active against all Ba/F3 cells tested. In the analysis of resistance mechanisms of BI4020 resistant (BIR) cells, none harbored secondary EGFR mutations. HCC827BIR cells had MET gene amplification and were sensitive to a combination of capmatinib (MET-TKI) and BI4020. HCC4006BIR and H1975BIR cells exhibited epithelial to mesenchymal transition. This study suggests that erlotinib may be more suitable than 4G TKIs to overcome sec-ondary mutations after front-line osimertinib. We found that off-target mechanisms that cause re-sistance to earlier generation TKIs will also cause resistance to 4G TKIs.
