Grape production worldwide is increasingly threatened by grapevine trunk diseases (GTDs). No grapevine cultivar is known to be entirely resistant to GTDs, but susceptibility varies greatly. To quantify these differences, four Hungarian grape germplasm collections containing 305 different cultivars were surveyed to determine the ratios of GTDs based on symptom expression and mortality rate. The cultivars of monophyletic Vitis vinifera L. origin were amongst the most sensitive ones, and their sensitivity was significantly (p<0.01) higher than that of the interspecific (hybrid) cultivars assessed, which are defined by the presence of Vitis species other than V. vinifera (e.g. V. labrusca L., V. rupestris Scheele, V. amurensis Rupr.) in their pedigree. We conclude that ancestral diversity of the grape confers higher degrees of resilience against GTDs.

The wild grapevine, Vitis vinifera subsp. sylvestris grows naturally throughout the northern hemisphere, including the Mediterranean region. Wild grapevines were also observed sporadically across the southern Levant and have been considered a non-native feral plant. Nevertheless, no formal characterization was conducted for wild grapevines in this region; thus, its taxonomical assignment remains elusive. Previously we have shown that the wild grapevine populations growing in northern Israel are genetically separated from the feral domesticated forms. The aim of this work was to comprehensively describe the morphological, anatomical, and ecological traits of wild grapevines naturally thriving in two distinct habitats in Israel. The dioicous nature of the wild grapevine, as well as their flower and pollen morphology, their characteristic sylvestris fruit and seed morphology, in addition to the occurrence of natural germination of seeds in close vicinity to the mother plant, all lead to our conclusion that these plants belong to Vitis vinifera subsp. Sylvestris, and should be included in the Flora Palaestina. These findings, combined with the recently-published genetic evidence for these populations significantly advance our understanding of the species' ecology and persistence under climate change.

The American leafhopper Erasmoneura vulnerata, detected in Europe in early 2000s, has recently become a pest in North-Italian vineyards. Issues were recorded in organic and conventional vine-yards despite the application of insecticides devoted to the control of other pests. Erasmoneura vulnerata completes three generations, and the second generation is frequently associated to large populations. The selection of appropriate active ingredients and the timing of their application is crucial for effective pest control. Field trials were carried out in North-eastern Italy, using a randomized design, to evaluate the impact of insecticides applied against other grapevine leafhoppers on E. vulnerata populations. The beginning of the second generation was selected as best timing for insecticide application. For a number of natural products, two applications were planned. Among the selected insecticides, the most effective were acetamiprid, flupyradifurone and lamb-da-cyhalothrin. Regarding natural products, the most effective was kaolin that could represent an alternative to pyrethrins in organic vineyards. The identification of pest threshold levels and the evaluation of side effects of the most effective insecticides on key natural enemies occurring in vineyards are required

The rest-breaking agent, hydrogen cyanamide (HC), can substitute insufficient chill unit accumulation in Vitis vinifera and induce uniform bud-break; however, due to its toxicity it is being banned. In South Africa, red seedless grapes, including V. vinifera Crimson Seedless (CS), are the largest table grape export group; therefore, replacing HC in V. vinifera CS is crucial. This study aimed to confirm the molecular triggers induced by HC and assess the bud-break-enhancing abilities of commercial plant biostimulants. Forced bud-break assay experiments using V. vinifera CS single-node cuttings and a small-scale field trial were performed. Results demonstrated that increased chill unit accumulation (CUA), reduced HC efficacy. Bud-break started between 10 and 20 days after treatment, irrespective of final CUA. The small-scale field trial found that HC3 % resulted in a slightly delayed bud-break rate, whilst biostimulants were similar to the negative control. The treatment of dormant grapevine compound buds with nitric oxide (NO), hydrogen peroxide (H2O2), and hypoxia trigger dormancy-release to a certain extent, supporting the molecular models proposed for HC action. NO, H2O2, and hypoxia, in combination with PBs, may potentially replace HC; however, this needs to be confirmed in future experiments.

Due to climate change, the sugar content of grapes in Hungary has increased so the high alcohol content alone can make wines disharmonious. In most vintages, this phenomenon is only a problem for early-ripening varieties. In order to prevent and treat this difficulty, we have been carrying out experiments in grape canopy management for four years with the aim of delaying ripening and thus reducing the sugar content of the grapes. The experiments were set up on an early (Pinot noir) and a late (Welshriesling) variety; two treatments (leaf re-moval - LR and short topping - ST) were applied and compared to untreated control in the years 2019-2022. Our results showed that must sugar yield was significantly reduced in all four years and for both of cultivars, while the other measured parameters (yield, acidity, pH, Botrytis infection) were not or lightly affected.

The objective of this study was to evaluate the evolution of the Syrah red and sparkling Moscatel wines stored for 12 months in green, amber and clear bottles. The phenolic compounds profile and antioxidant activity in vitro were determined. Commercial wines were bottled in an automatic filling machine, stored and analyzed every three months for one year. Several phenolic compound families were quantified through reversed-phase high performance liquid chromatography (RP-HPLC) coupled to diode-array detection (DAD) and fluorescence detection (FD). The different bottle colors studied did not influence the evolution of the sparkling Moscatel and Syrah red wines, since the main variations obtained were related to storage time. The main changes were observed in the Syrah wine, where storage time was associated with an increase in hue (h*), decrease in catechins, increase in procyanidins and, most notably, a decrease in the anthocyanin malvidin 3-glucoside. In general, the wines showed good stability in relation to the antioxidant activity in vitro.

Climate change related increases in frequency and intensity of heatwaves affect viticulture, leading to losses in yield and grape quality. We assessed whether canopy architecture manipulation mitigates the effects of summer stress in a Mediterranean vineyard. Muscat of Alexandria grapevines were monitored during 2019-2020. Two canopy shoot positioning treatments were applied: Vertical shoot positioning (VSP); and Modulated shoot positioning (MSP), the west side upper foliage released to promote partial shoot leaning, shading the clusters. Clusters were sampled at pea size (PS), veraison (VER) and full maturation (FM). Measurements included rachis anatomy and hydraulic conductance, and aquaporins (AQP) and stress-related genes expression in cluster tissues. Results show significant seasonal and interannual differences in Kh and vascular anatomy. At VER, Krachis , Krachis+pedicels, and xylem diameter decreased but were unaffected by treatments. The Phloem/Xylem ratio was either increased (2019) or reduced (2020) in MSP, compared to VSP. Most AQP were down-regulated at FM in pedicels, and up-regulated at VER in pulp. A potential maturation shift in MSP was observed, confirmed by the up-regulation of several stress-related genes in all tissues. The study pinpoints the role of canopy architecture in berry water relations and stress response during ripening.

'Cv 'Doña María' table grape is a high-quality table grape variety included in the Protected Designation of Origin (PDO) of the European Union "The bagged grape of the Vinalopó." The PDO stipulates that grape clusters must be protected with paper bags from inclement weather and insects, which helps enhance the final grape quality. However, 'Doña María' is a variety prone to high shattering in the late stages of ripening on the vine and during postharvest. Inorganic calcium treatments are one of the most commonly used tools to reduce this disorder, but the translocation of this mineral from veraison onward has been questioned. In this study, five applications were performed, from veraison to harvest, using sorbitol-chelated calcium (0.7 % + 1.0 %), Ca(NO3)2 (Ca) at 0.7% and sorbitol at 1%. It was observed that bagged grapes (not wetted with the solutions) only increased the concentration of total and bound calcium when treated with Sorbitol-Ca. This resulted in reduced berry drop during cultivation and postharvest and improved fruit firmness. Additionally, it reduced grape metabolism indicators such as respiration rate, weight loss, malic acid degradation, prevented abscisic acid (ABA) and malondialdehyde (MDA) accumulation, and favored the accumulation of secondary metabolites such as total polyphenols, increased antioxidant activity, and sugar content. The application of sorbitol-chelated calcium is an effective and safe tool that enhances fruit quality and prevents losses due to shattering during postharvest.

Hydroalcoholic extracts from Malbec and Torrontés wine pomace (Vitis vinifera L.) originating from the high-altitude vineyards of Argentina's Calchaquí Valleys were characterized. Total phenolics, hydroxycinnamic acids, orthodiphenols, anthocyanins, non-flavonoid phenolics, total flavonoids, flavones/flavonols, flavanones/dihydroflavonols, and tannins were quanti-fied through spectrophotometric methods, with Malbec extract exhibiting higher concentra-tions in most of phytochemical groups when compared to Torrontés. HPLC-DAD identified more than 30 phenolic compounds in both extracts. Malbec displayed superior antiradical ac-tivity (ABTS cation, nitric oxide, and superoxide anion radicals), reduction power (iron, copper and phosphomolybdenum), hypochlorite scavenging, and iron chelating ability, compara-tively to Torrontés. The cytotoxicity assessments revealed that Torrontés affected the viability of HT29-MTX and Caco-2 colon cancer cells by 70% and 50%, respectively, at the highest tested concentration (1 mg/mL). At the same time, both extracts did not demonstrate acute toxicity in Artemia salina or in red blood cell assays at 500 µg/mL. Both extracts inhibited the lipoxygen-ase enzyme, with Malbec also reducing the tyrosinase activity and neither inhibited the xan-thine oxidase. The substantial phenolic content and diverse biological activities in Calchaquí Valleys' pomaces underline their potentialities to be valorized for pharmaceutical, cosmetic, and food industries.

Susceptibility to economically-important diseases of grapes is critical to the evaluation of germplasm recommended for commercial production and for development of sustainable production systems. In 2018-2019, nine cold-hardy grape cultivars including ‘Brianna’, ‘Crimson Pearl’, ‘Itasca’, ‘Louise Swenson’, ‘Marechal Foch’, ‘Marquette’ ‘Petite Pearl’, ‘St. Pepin’, and ‘Verona’ were evaluated on non-treated vines for susceptibility to downy mildew, powdery mildew, black rot, anthracnose, Phomopsis leaf spot and fruit rot, and Botrytis bunch rot. No cultivars were consistently disease-free, and all exhibited some degree of black rot and powdery mildew infection. Relative susceptibility to disease was not consistent across both years, but ‘Brianna’ had greater incidence of black rot and ‘Louise Swenson’ showed lower incidence of powdery mildew in both years. The relatively new cultivars ‘Crimson Pearl’ and ‘Verona’ exhibited comparatively moderate disease susceptibility overall. Growers typically manage diseases with fungicides on commercial farms, so cultivar susceptibility is just one component of a sustainable pest management and production system.

A high performance Driver State Monitoring (DSM) application for the detection of driver drowsiness is presented in this paper. It relies on the usage of an Ensemble of Regression Trees (ERTs) machine learning method that aligns 68 facial landmarks. Special focus is given on the acceleration of the frame processing using reconfigurable hardware. Reducing the frame processing latency saves time that can be used to apply frame-to-frame facial shape coherency rules. False face detection and false shape estimations can be ignored for higher robustness and accuracy in the operation of the DSM application without reducing the frame processing rate that can reach 65 frames per second. The sensitivity and precision in yawning recognition can reach 93% and 97%, respectively. The implementation of the employed DSM algorithm in reconfigurable hardware is challenging since the kernel arguments require large data transfers and the degree of data reuse in the computational kernel is low. Due to this, unconventional hardware acceleration techniques have been employed that can also be useful for the acceleration of several other applications.

Grapevine is one of the most relevant crops in the world being used for economically important products such as wine. Yet, relevant grapevine cultivars are heavily affected by diseases such as the downy mildew disease caused by Plasmopara viticola. Improvements on grapevine resistance are made mainly by breeding techniques where resistance traits are introgressed into cultivars with desired grape characteristics. However, there is still a lack of knowledge on how resistant or tolerant cultivars tackle the P. viticola pathogen. In this study, using a shotgun proteomics LC-MS/MS approach, we unravel the protein modulation of a highly tolerant grapevine cultivar, V. vinifera ‘Regent’, in the first hours post inoculation (hpi) with P. viticola. At 6 hpi, proteins related to defence and to response to stimuli are negatively modulated while at 12 hpi there is an accumulation of proteins belonging to both categories. The co-occurrence of effector-triggered susceptibility (ETS) and effector-triggered immunity (ETI) is detected at both time-points, showing that these defence processes present high plasticity. The results obtained in this study unravel the tolerant grapevine defence strategy towards P. viticola and may provide valuable insights on resistance associated candidates and mechanisms, which may play an important role in the definition of new strategies on breeding approaches.

This study investigates the impact of water stress on grapevines, specifically examining the role of rootstocks and aquaporins. Two experiments on potted plants were conducted in central Chile during the summer, under conditions of high water demand, involving various rootstock genotypes and combinations of Cabernet Sauvignon (CS) grafted onto rootstocks. Significant differences were observed among plants in terms of stem water potential, stomatal conductance, and growth rate. Notably, the CS/CS combination consistently displayed the slowest growth rate, regardless of the irrigation treatment. The study also analyzed the expression levels of plasma membrane intrinsic proteins (PIPs) and tonoplast intrinsic proteins (TIPs) aquaporins in the leaves of grafted plants. Specifically, VvPIP2;2 aquaporins showed reduced expression after 14 days without irrigation, whereas VvTIP1;1 and VvTIP2;1 expression levels correlated positively with gs responses in grafted plants, suggesting their role in modulating water content in leaves under water stress. TIP aquaporins likely play a significant role in the differential responses of CS plants towards nearly isohydric or anisohydric behaviour. The CS/CS combination exhibited near isohydric behavior, correlating with lower TIP aquaporin expression, while the combination of CS onto 1103P and 101-14 showed higher expression, indicating anisohydric behavior. The findings suggest that grafted plants are more resilient to water stress, supporting the idea that rootstocks can mitigate the effects of water stress on the scion.

The Douro Demarcated Region (DDR) has peculiar edaphoclimatic characteristics that provide a suitable terroir for premium wine production. As climate change effects continue to emerge, ensuring productivity and quality becomes increasingly important for viticulturists, as those directly determine their profits. Cultural approaches, such as the use of biostimulants, are actively being developed to mitigate abiotic stress. The main objective of this work was to assess the effect of foliar sprays of a seaweed (Ascophyllum nodosum) based extract (ANE) and glycine betaine (GB) on grape berry quality, bioactive compounds, and antioxidant activity. A trial was installed in a commercial vineyard (cv. Touriga Franca) in the Douro Superior (Upper Douro) sub-region of the Douro Demarcated Region. In 2020, a total of three foliar sprayings were performed during the growing season, namely at pea-size, bunch closure, and veraison. There was a positive effect of both biostimulants (ANE and GB) on the physiological and biochemical performance of cv. Touriga Franca exposed to summer stress. In general, the GB 0.2% spraying was the most promising treatment for this grape cultivar, as it increased berry quality, the concentration of bioactive compounds (total phenolics, flavonoids and ortho-diphenols), and the antioxidant activities. These results revealed the efficacy of biostimulants sprayings as sustainable viticultural practice, improving berry quality under summer stress conditions.

Variability in traits forming the Leaf Economics Spectrum (LES) among and within crop species play a key role in governing agroecosystem processes. However, studies evaluating the extent, causes, and consequences of within-species variation in LES traits for some of the world’s most common crops remain limited. We quantified variation in nine leaf traits measured across 90 vines of five wine grape (Vitis vinifera) varieties at two ontogenetic stages. Grape traits covary along an intraspecific LES, in patterns similar to those documented in wild plants. Across varieties, high rates of photosynthesis (A), and leaf nitrogen (N) concentrations, are coupled with low leaf mass per area (LMA), while the opposite suite of traits defines the “resource conserving end” of this intraspecific LES in grape. Variety identity predicted of leaf physiological (A) and morphological traits (i.e., leaf area and leaf mass), while leaf chemical traits and LMA were best explained by ontogenetic stage. All varieties expressed greater resource conserving trait syndromes (i.e., higher LMA, lower N, lower Amass) later in the growing season. Traits related to leaf hydraulics, including instantaneous water-use efficiency (WUE), were unrelated to LES and other resource capture traits, and were better explained by spatial location. Our results highlight the relative contributions of genetic vs. phenotypic factors in structuring this variation and point to a key role of domestication in governing trait relationships in the world’s crops.

Modification of canopy architecture is fundamental to productivity in grapevine. This research focused on evaluating the impact of canopy openness on photosynthetically active radiation, photosynthetic activity, and the physicochemical properties of fruit in Cabernet Sauvignon grapevines. A completely randomised design was used to compare open and closed canopies, with ten replicates per treatment (20 trees in total), during the vegetative growth period and after harvest. Key measurements included photosynthetically active radiation (PAR), daily light inte-gral (DLI), photosynthetic rate (PR), stomatal conductance (SC), intercellular CO2 concentration (IC), leaf area (LA), transpiration, ambient CO2 concentration, and temperature. Additionally, we assessed fruit quality parameters such as total soluble solids (TSS), glucose-fructose, total titratable acidity (TTA), pH, TSS/TTA, and total phenols (TP). significantly (p≤0.05) increased PAR, DLI, PR, IC, and LA, while after harvest, only PR and IC showed variation. Closed canopies increased CO2/H2O by 62.5%, while H2O/CO2 and temperature were higher in open canopies. Canopy openness increased contacts, gaps and visible sky and reduced leaf area index. Open canopy fruits showed higher TSS, glucose-fructose, pH, TSS/TTA and TP contents. Canopy openness is essential for improving light interception, photosynthetic efficiency and fruit quality in Cabernet Sauvi-gnon grapevine cultivated in northern Mexico.

Environmental degradation leads to a totally unsustainable food system. In addition to this is-sue, the consumption of foods that improve people's health and well-being is recommend-ed. These guidelines create new challenges and the demand for alternatives to meet these needs in the short term. One of the alternatives is undoubtedly the use of by-products of winemaking, namely in the form of grape pomace flour (GPF), which is obtained in a totally sustainable way, requires no further water consumption or agricultural land, and is an ingredient that boasts all the characteristics that meet the circular economy and has colossal potential in promoting nutrition and healthy eating. To verify the benefits of using the Touriga Nacional and Arinto (Vitis vinifera L.) flour varieties, analytical determinations were made to identify and quantify different components, such as nitrogen and extractable phosphorus, ash, moisture, proteins, fats, dietary fibers, total sugars, and phenolic content. GPF could become a valuable ingredient due to its nutritional quality and high content in various polyphenols namely flavonoids and pyranoanthocyanins.

Quebec is the third-largest wine grape producing province in Canada, and the industry is constantly expanding. Traditionally, 90% of the grapevine cultivars grown in Quebec were rustic or semi-rustic and largely dominated by winter hardy interspecific hybrid Vitis sp. cultivars. Over the years, the winter protection techniques adopted by growers and climate changes have offered an opportunity to establish V. vinifera L. cultivars (e.g. Pinot Noir). We characterized the virome of leafroll-infected interspecific hybrid cultivar and compared it to the virome of V. vinifera cultivar to support and facilitate the transition of the industry. A dsRNA sequencing method was used to sequence symptomatic and asymptomatic grapevine leaves of different cultivars. The results suggested a complex virome in terms of composition, abundance, richness, and phylogenetic diversity. Three viruses, grapevine Rupestris stem pitting-associated virus, grapevine leafroll-associated virus (GLR) 3 and 2 and hop stunt viroid (HSVd) largely dominated the virome. However, their presence and abundance varied among grapevine cultivars. The symptomless grapevine cultivar Vidal was frequently infected by multiple virus and viroid species and different strains of the same virus, including GLR virus 3 and 2. Our data shows that viruses and viroids associated with the highest number of grapevines expressing symptoms included HSVd, GLR3 and GLR2, in gradient order. However, co-occurrence analysis revealed that the presence of GLR species was randomly associated with the development of virus-like symptoms. These findings and their implication for grapevine leafroll disease management were discussed.

Plant-based extracts have been recently employed as sustainable tools to improve biotic and abiotic stress tolerance and increase grape quality. However, knowledge about the effect of these extracts on secondary metabolism compounds, that are fundamental for grape and wine quality, is still scarce. In this study, a trial was installed in an experimental vineyard with the variety Touriga Franca located at University of Trás-os-Montes e Alto Douro, Baixo Corgo sub-region of the Douro Demarcated Region, in two growing seasons: 2019 and 2020. Foliar sprayings with the natural products, nettle extract (NE) and Japanese knotweed extract (JKE), were tested to evaluate their effect on downy mildew incidence and severity, on grapevines bioactive compounds contents and antioxidant activity, at veraison and harvest. NE and JKE have stimulated an improvement in the concentration of total carotenoids in leaves and the concentration of total phenolics in berries along with an improved concentration of total phenolics and flavonoids in leaves and berries. Thus, it was verified that these extracts enhance grapevines bioactive compounds and antioxidant capacity and, consequently, the physiological performance of the plant and the quality of the berries. Considering the challenges posed by climate change in the Mediterranean basin viticulture sector, the use of plant extracts might represent a sustainable tool to mitigate the increasing severity of drought, often associated to heatwaves and high irradiation. Moreover, downy mildew incidence results suggest that an integrated disease management approach combining conventional control with alternative fungicides can be used in the future for a more sustainable viticulture.

Soil water balance (SWB) in woody crops is sometimes difficult to estimate with one-dimensional models because these crops do not completely cover the soil and usually have a deep root system, particularly when cropped under rainfed conditions in a Mediterranean climate. In this study, the actual crop evapotranspiration (ETc act) is estimated with the soil water balance model SIMDualKc which uses the dual-Kc approach (relating the fraction of soil cover with the crop coefficients) to improve the estimation of the water requirements of a rainfed vineyard, using data from a deep soil profile. The actual basal crop coefficient (Kcb act) obtained using the SIMDualKc model was compared with the Kcb act estimated using the A&P approach, which is a simplified approach based on measurements of the fraction of ground cover and crop height. Spectral vegetation indices derived from Landsat-5 satellite data were used to determine the fraction of ground cover (fc VI) and thus, the density coefficient (Kd). The SIMDualKc model was calibrated using soil water content (SWC) measurements down to a depth of 1.85 meters, which significantly improved the conditions for using a SWB estimation model. The test of the model was performed using a different SWC dataset. A good agreement between simulated and field measured SWC was observed for both data sets along the crop season, with RMSE<12.0 mm, NRMSE <13%. The calibrated Kcb values were 0.15, 0.60 and 0.52 for the initial, mid-season, and end season, respectively. The ratio between actual (ETc act) and crop evapotranspiration (ETc) was quite low between veraison and maturity (mid-season), corresponding to 36%, indicating that the rainfall was not sufficient to satisfy the vineyards water requirements. Spectral vegetation indices used to compute fc VI were unable to fully track the plants’ conditions when water stress. However, ingestion of data from remote sensing showed promising results that could be used to support decisions making in irrigation scheduling. Further studies on the use of the A&P approach using remote sensing data are required.

The evaluation of suitability for bioclimatic viticulture indices (BVIs) zones, as with any other crop, requires a well-founded knowledge of the spatial variability in climate data, which is also used to assess different grapevine cultivars and to delimit appropriate regions for vineyard production under the current climate and projected climate change. BVIs, on the other hand, are typically calculated using a small number of locations linked by climatic station sites or applied using a coarsely-resolution distributed dataset. Furthermore, often the methods applied to precisely delineate symmetrical regions are not appropriate to generate accurate maps. To provide an analysis using three temperature-based indices and the hydrothermal coefficient (HTC), quantifying their spatial variation, and representing the spatial patterns of each BVI throughout the Jabal Al Arab viticulture—one of the most important Syrian viticulture regions in the Eastern Mediterranean—daily temperature data from 15 meteorological stations and 57 rain gauges, considering the 1984–2014 period, and downscaled future scenarios (RCPs based on CanESM2), considering the 2016–2100 period, were utilized. The statistical method of each BVI was analyzed, and, later, they were mapped by using GIS and a hybrid interpolation (regression-kriging) approach. The regression fitting method revealed that the sum accumulation of heat and the hydro-thermal index during the growing season were highly related to elevation and distance to the seacoast variables; however, the viticulture zones varied spatially depending on which index was used and under suggested future scenarios compared to the current climate. The spatial distribution of climate in the Jabal Al Arab regions exhibits significant variability. The findings suggest that climate change projections indicate a prevalence of warmer conditions in the future. Under the RCP scenarios, the territory can be categorized into up to three bioclimatic classes for both the Heat Index (HI) and the Winter Index-Growing Degree Days (WI-GDD), in contrast to the current climate which has six classes. These results provide valuable insights into the suitability of viticulture within each climatic region and facilitate the identification of homogeneous zones. The utilization of consistent bioclimatic indices and an advanced hybrid interpolation method has enabled the delineation and comparison of bioclimatic variables of Jabal Al Arab with other regions worldwide. Such comparisons should be considered when selecting grapevine varieties and assessing the potential for grape production.

The Folla Redonda grape is indigenous to the region of Rias Bajas in Galicia, Spain. This grape is distinguished by its characteristic acid flavor and pulp of remarkable coloration. Despite the wine obtained from this grape is highly valued by consumers of this region for its surprising acidic notes and thickness, it is important to note that it is not a variety of Vitis vinifera and its production is not legally regulated. In the present study, an evaluation of the physicochemical properties of the Folla Redonda grape (pH, °Brix, °Bé, density, acidity, sugar content, etc.) as well as its content in bioactive compounds (total polyphenols and anthocyanins) was carried out. Additionally, the antioxidant potential of the extracts obtained from this grape was demonstrated. This study proved the interesting organoleptic characteristics of the grape, along with a high content of compounds of interest and an important antioxidant activity comparable to that of Vitis vinifera grapes. Therefore, the grape's natural consumption and its application in different industries such as nutraceutical or pharmacological would result in an important revaluation of the grape and significant health benefits. In case that the production of wine from this variety is legalized, consumers will not only be able to enjoy a wine with a characteristic dense reddish color and important acid tones, but also with interesting concentrations of bioactive compounds.

In vineyards, Botryosphaeria dieback (BD) occurs in young and old plants. In the field, the prevalence and severity of the disease increase proportionally with the age of the vineyards. Among the pathogens that cause (BD), Diplodia seriata is the most prevalent species in Chile and other countries with a Mediterranean climate. To date, no information is available on the sus-ceptibility of adult wood to infection by this pathogen since most of the pathogenicity tests have been carried out on 1- or 2-year-old shoots or detached canes. Therefore, a pathogenicity test was carried out on plants under field conditions, with inoculations in plants, on one-year-old shoots, and 2- and 10-year-old wood in grapevine cv. Cabernet Sauvignon. A pathogenicity test was carried out with two isolates of D. seriata selected in an assay in detached canes that were 2-year-old. The results on plants showed that D. seriata was significantly more aggressive on 10-year-old than on one- or two-year-old tissue. These results were compared with the lesions ob-tained from two-year-old canes. The results of this work are consistent with the damage ob-served in the field, highlight the importance of identifying each pathogenic species that causes (BD) and contribute to the knowledge of the epidemiology of this disease in Mediterranean cli-mates.

A three-year survey was conducted to estimate the incidence of grapevine trunk diseases (GTDs) in Greece and identify fungi associated with the disease complex. In total, 310 vineyards in different geographical regions in northern, central, and southern Greece were surveyed, and 533 fungal strains were isolated from diseased vines. Morphological, physiological and molecular (5.8S rRNA gene-ITS sequencing) analyses revealed that isolates belonged to 35 distinct fungal genera, including well-known (e.g. Botryosphaeria sp, Diaporthe spp., Eutypa sp., Diplodia sp., Fomitiporia sp., Phaeoacremonium spp., Phaeomoniella sp.) and lesser-known (e.g. Neosetophoma sp., Seimatosporium sp., Didymosphaeria sp., Kalmusia sp.) grapevine wood inhabitants. GTDs-inducing population structure differed significantly among the discrete geographical zones. Phaeomoniella chlamydospora (26.62%, n=70), Diaporthe spp. (18.25%, n=48) and F. mediterranea (10.27%, n=27) were the most prevalent in Heraklion, whereas D. seriata, Alternaria spp., P. chlamydospora and Fusarium spp. were predominant in Nemea (central Greece). In Amyntaio and Kavala (northern Greece), D. seriata was the most frequently isolated species (>50% frequency). Multi-genes (rDNA-ITS, LSU, tef1-α, tub2, act) sequencing of selected isolates, followed by pathogenicity tests revealed that Neosetophoma italica, Seimatosporium vitis, Didymosphaeria variabile and Kalmusia variispora caused wood infection with the former being the most virulent. To the best of our knowledge, this is the first report of N. italica associated with GTDs worldwide. This is also the first record of K. variispora, S. vitis and D. variabile associated with wood infection of grapevine in Greece. The potential associations of disease indices with vine age, cultivar, GTD-associated population structure and the prevailed meteorological conditions in different viticultural zones in Greece are presented and discussed.

Biofilm formation and lipopolysaccharide (LPS) are implicated in the pathogenesis of gastrointestinal (GI) diseases caused by Gram-negative such as Escherichia coli and Salmonella enterica serovar Typhimurium. Grape seeds, wine industry by-products, have antioxidant, antimicrobial, and anti-inflammatory activities. In the present study, the protective effect of procyanidin-rich grape seed extract (prGSE), from unfermented pomace of Vitis vinifera cv Bellone, on bacterial LPS-induced oxidative stress and epithelial barrier integrity damages has been studied in a model of Caco-2 cells. The in vitro activity of prGSE against formation of biofilm of S. Typhimurium and E. coli, has been investigated. In vivo, prGSE activity using infected Galleria mellonella larvae has been evaluated. The results showed that the prGSE, if administered with LPS, can significantly reduce the LPS-induced permeability alteration. Moreover, the ability of the extract to prevent Reactive Oxygen Species (ROS) production induced by LPS treatment of Caco-2 cells has been demonstrated. prGSE inhibited the biofilm formation of E. coli and S. Typhimurium. In in vivo activity, increased survival of infected G. mellonella larvae after treatment with prGSE has been demonstrated. In conclusion, grape seed extracts could be used in the reduction of GI damage caused by bacterial endotoxin and biofilm of Gram-negative bacteria.

FLH 13-11 is an F1 interspecific hybrid muscadine grape genotype that was developed to produce new anthocyanins for pigment color stability. This hybrid resulted from a cross between ‘Marsh’ (Vitis munsoniana) and ‘Magoon’ (V. rotundifolia). This report characterizes the anthocyanins produced in fully ripe berries, and reveals a significant difference in total anthocyanin contents from two continuous cropping seasons. High-performance liquid chromatography with a diode array detector (HPLC-DAD) and HPLC-quadrupole time-of-flight tandem mass spectrometry (HPLC-qTOF-MS/MS) were used to profile anthocyanins in berries. The resulting data showed that fourteen anthocyanins were detected, six from 2011 and nine from 2012, with only one produced in both seasons. However, the anthocyanidin profiles of berries were the same. Five anthocyanins were annotated as diglucosides of anthocyanidins based on MS/MS features, including delphinidin 3,5-diglucoside produced in both seasons, cyanidin 3,5-diglucoside mainly formed in 2011, petunidin 3,5-diglucoside, malvidin 3,5-diglucoside, and peonidin 3,5-glucoside only detected in 2012. Also, three anthocyanidin-diglucoside like anthocyanins and three monoglucosides including peonidin 3- glucoside, delphinidin 3- glucoside like, and pelargonidin 3- glucoside like anthocyanins, were detected in 2011 and 2012, respectively. These results indicate that FLH 13-11 can produce both anthocyanidin-diglucosides and -monoglucosides, and their biosynthesis is closely dependent on cropping years.
