The use of nitrogen (N) fertilizers can have detrimental effects on the environment due to nitrate leaching. This study aims to examine the impact of N fertilizer source and application timing on wheat yield and nitrate leaching in various soil textures. A randomized complete block design (RCBD) was employed to conduct the experiment. The treatments consisted of two N fertilizer sources: ammonium nitrate (AN) and urea (NN), applied at three different times throughout the year, with a control treatment. The three application timings were total application before sowing (T₁), total application in spring (T₂), and half the amount before sowing and the other half in spring (T₃). The experiment was conducted in three different soil textures (sandy loam, silty clay loam, and silty clay), with three replications over three growing seasons: 2018-19 (Y₁), 2019-20 (Y₂), and 2020-21 (Y₃). The results indicated significant effects of different soil textures on grain yield. Specifically, the silty clay loam soil differed significantly from the sandy loam and silty clay soils in terms of grain yield. The interaction effects between the N fertilizer source and application timing were also found to be significant for all soil textures (p<0.01). In sandy loam and silty clay soils, the grain yield increased when using the T₃ application timing compared to the silty clay loam soil. Notably, the silty clay loam soil with the T₂ application timing exhibited the highest grain yield, with a yield of 6863.6 kg ha^-1. Furthermore, the study revealed that N fertilizers and their application timing significantly influenced the protein content in the silty clay and sandy loam soils. The protein percentages were 10.76 and 10.93 for the silty clay and sandy loam soils, respectively, following the T₂ treatment. Additionally, ammonium nitrate (AN) demonstrated a noticeable effect on the percentage of grain protein compared to urea (NN). In conclusion, the application timing of N fertilizers had an impact on grain yield and protein content in different soil textures. Moreover, nitrate concentration increased throughout the experiment, with the highest soil nitrate concentration observed in both fertilizer treatments, particularly in the sandy loam soil.

Compared with maize monocropping systems, maize double-cropping production systems have a much greater nitrogen extraction capacity. This study aims to assess if in these farming systems, animal manures could be applied, using adequate management practices, exceeding the maximum annual amount of livestock manure established in the European Nitrate Directive for vulnerable zones (170 kg N ha-1) without increasing the risk of water nitrate contamination. We compare the risk of nitrate leaching under two fertilisation strategies, one with synthetic fertilisers (M) and the second with maximized application of pig slurry (P). The study was conducted in two contrasting soil types, a deep and heavy-textured soil and a shallow and stony soil. Crop yields and N extractions were not affected by the fertilisation strategies. The nitrate concentrations in the soil below the crop root zone were lower in the deep soil than in the shallow soil, where values exceeded 1000 mg L-1. The soil nitrate concentration was not affected by fertilisation strategies at each site. The results show that pig slurry can be applied above the limit of 170 kg N ha-1, in the conditions of the study up to 360 kg N ha-1, without increasing the risk for nitrate leaching.

The aim of this research involves investigating the elimination of nitrogen ions from groundwater through the application of dynamic permeable reactive barriers (PRB) utilizing A400-nZVI. The aim also implyies determining barrier parameters, as well as assessing the overall retention capacity of nitrogen ions through percolation with a potassium nitrate solution. The research involves obtaining zero valent iron nanoparticles (nZVI), which were synthesized and doped onto an anionic resin support material (A400) through the reduction reaction of ferrous ions with sodium borohydride (NaBH4). This was achieved by preparing a ferrous sulfate solution, contacting it with the ion exchange resin at various solid-liquid mass ratios, and gradually adding sodium borohydride under continuous stirring in an oxygen-free environment to create the A400-nZVI barrier. The outcomes of this study, focusing on the development of permeable reactive barriers composed of nanovalent iron and ion exchangers, demonstrate significant potential in purification processes when appropriately dimensioned. The research specifically evaluated the efficacy of NO3- removal using the A400-nZVI permeable reactive barrier, conducting laboratory tests that simulated a naturally stratified aquifer with high nitrate contamination.

Galdieria maxima is a polyextremophilic alga capable of diverse metabolic processes. Ammonia is widely used in culture media typical of laboratory growth. Recent reports that this species can grow on wastes promote the concept that G. maxima might have biotechnological utility. Accordingly, there is a need to know the range of pH levels that can support G. maxima growth in a given nitrogen source. Here, we examined the combined effect of pH and nitrate/ammonium source on the growth and long-term response of the photochemical process to a pH gradient in different G. maxima strains. All were able to use differing nitrogen sources, despite both the growth rate and photochemical activity were significantly affected by the combination with the pH. All strains acidified the NH4+-medium (pH<3); only G. maxima IPPAS P507 showed reduced capacity in lowering the pH from 6.5. pH was a limiting factor in nitrate uptake at pH≥6.5; noteworthy, at pH 5 on nitrate G. maxima ACUF551 showed a good growth performance, despite the alkalization of the medium.

Nitrate and nitrite are the most extensively used food additives in the food industry for their multifunctional properties, and they have been in the spotlight for decades for its controversial back effects. The conversion of nitrate and nitrite to various nitroso compounds, specifically N-nitrosamine raises the question of the integrity of these additives. Nitrosamine is a well-known carcinogen and mutagen. A large number of studies demonstrate the carcinogenic effects of these salts. Are these findings credible? Nonetheless, the regulatory agencies approve its usage; is it biased? Are we feeding on the carcinogens in processed foods? The current work provides an insight into the basic and toxicological findings of the food additives -nitrate and nitrite.

The results show that seven types of N-nitrosamines: N-nitrosodiethylamine (NDEA), N-Nitrosopiperidine (NPIP), N-nitrosodimethylamine (NDMA), N-nitrosopyrrolidine (NPYR), N-nitrosodipropylamine (NDPA), N-nitrosodibutylamine (NDBA) and N-nitrosodiphenylamine (NDphA) were reduced after heat treatment of pork steaks marinated with L. plantarum MI-LP1, S. carnosus LM-SC1, S. vitulinus LM-SV1. Selected LAB suppressed the formation of putrescine and spermine, lowered acid and peroxide values, which were significantly lower than in the control samples, then P≤0.05 in all cases. The nitrites and nitrates amounts were significantly decreased in pork steaks samples marinated with L. plantarum MI-LP1 and S. carnosus MI-SV1 strains. These results presented that selected lactic acid bacteria could be used for marinating pork steaks to improve quality characteristics and reduce N-nitrosamine formation during grilling.

This brief report presents a comparative analysis to examine differences in the mean dose of nitrate supplementation between studies with positive and negative results on exercise performance. Protocols of at least six days that had positive results used lower nitrate doses (~7.7 mmol), whereas those with negative results used higher doses (~10.2 mmol), showing a statistically significant difference (p < 0.05). These findings suggest long-term supplementation with higher doses (~10.2 mmol) may not improve exercise performance. Further research is essential to understand the underlying mechanisms.

With the rapid development of cities in northwest China, there has been an increasing focus on groundwater pollution in plateau cities, specifically the common occurrence of nitrate pollution. The special climatic, geological, and geomorphological characteristics of plateau and river valley cities contribute to distinct groundwater chemical characteristics. Therefore, the formation and evolution process of groundwater nitrate contamination differs from that of plain cities. To explore these issues, we conducted an analysis of eight major ions in various groups of water samples from rivers, springs, and groundwater in Haidong. By utilizing factor analysis and correlation analysis, we were able to identify the characteristics and formation of groundwater chemistry and nitrate pollution in Haidong. Our findings reveal that the chemical characteristics of groundwater in Haidong are primarily controlled by rock weathering, mineral dissolution, and evaporation, leading to the formation of highly mineralized groundwater. Additionally, the excessive nitrate content in certain areas is a result of domestic sewage discharge and agricultural fertilizer use, exceeding Chinese drinking water health standards. Furthermore, for cities located in valleys, the geological structure significantly impacts the nitrate content of groundwater in different regions. Areas with obstructed groundwater flow tend to have higher nitrate levels, while regions with unobstructed groundwater experience lower nitrate concentrations. Notably, shallow groundxwater is more vulnerable to nitrate pollution compared to deep groundwater. This study holds great significance in understanding the chemical characteristics of groundwater and the formation and evolution of nitrate pollution in highland river valley cities.

The risk of nitrate contamination became a reality for Fairmont in Minnesota, when water rich in NO3-N exceeded the drinking water standard of 10-mg/L. This was unexpected because this city draws its municipal water from a chain of lakes that were fed primarily by shallow groundwater under row-crop land use. Spring soil thaw drives cold water into subsurface pipe where almost no NO3-N reduction occurs. This paper focuses on NO3-N reduction before entering the lakes and no other nitrogen management practices in the watershed. A novel denitrifying bioreactor was constructed behind a sediment forebay which then flowed into a chamber covered by a greenhouse, before entering a woodchip bioreactor. In 2022 and 2023, water depth, dissolved oxygen, and temperature were measured at several locations and continuous NO3-N concentration at the entry and exit of the bioreactor. The results showed better performance at a low water depth with lower dissolved oxygen, and higher water temperature. The greenhouse raised the inlet temperature in 2022 but did not in 2023. The forebay and the greenhouse may have impeded the denitrification process due to the high dissolved oxygen concentrations in the influent and the stratification of dissolved oxygen caused by algae in the bioreactor.

High nitrate and fluoride contamination in groundwater cause a variety of disorders, including methemoglobinemia, teratogenesis, and dental and skeletal fluorosis. The present work assesses the non-carcinogenic health risks posed by nitrate and fluoride in infants, children, and adults using the USEPA method. Groundwater samples were collected from 36 wells and boreholes in three central Saudi Arabia study areas. Nitrate concentrations varied from 0.70 to 47.00 mg/L. None of the 36 studied boreholes had nitrate levels that exceeded WHO guidelines (50.00 mg/L). Fluoride ranged from 0.63 to 2.00 mg/L, and 30.55% of the fluoride samples (11 out of 36) exceeded the WHO recommendations for acceptable drinking water (1.5 mg/L). The average hazard index (HI) values for adults, children, and infants were 0.99, 2.59, and 2.77, respectively. Water samples surpassed the safety level of 1 for adults, children, and infants at 44.44, 97.22, and 100%, respectively. Accordingly, water samples from Jubailah and a few from Wadi Nisah may expose infants, children, and adults to non-cancer health concerns. Infants and children are more vulnerable to non-carcinogenic health risks than adults, possibly due to their lower body weight.

Groundwater contamination by nitrate and organic chemicals (e.g. 1,4-dioxane) is a growing worldwide concern. This work presents a new approach for simultaneously treating nitrate and 1,4-dioxane, which is based on UV sensitization of nitrate and sulfite, and the production of reactive species. Specifically, water contaminated with nitrate and 1,4-dioxane is irradiated by a UV source (< 250 nm) at relatively high doses, to sensitize in-situ nitrate and generate HO•. This leads to the oxidation of 1,4-dioxane (and other organics), and the (undesired) production of nitrite as an intermediate. Subsequently, sulfite is added at an optimized time-point, and its UV sensitization produces hydrated electrons which reacts and reduces nitrite. Our results confirmed the effectivity of the proposed treatment: UV irradiation of nitrate (at > 5 mg N/L) efficiently degraded 1,4-dioxane, while producing nitrite at levels higher than 1 mg N/L (its MCL in drinking water). Adding sulfite to the process after 10 minutes of irradiation reduced the concentration of nitrite, without affecting the degradation rate of 1,4-dioxane. The treated water contained elevated levels of sulfate; albeit at much lower concentration than its MCL. Treating water contaminated with nitrate and organic chemicals (often detected concomitantly) typically requires several (expensive) treatment processes. The proposed approach may present a cost-effective alternative, employing a single system for the treatment of nitrate and organic contaminants

This work aimed to carry out a bioremediation study to evaluate the resistance of Penicillium sp. isolated from kefir grains for the treatment of copper. The fungal culture medium was prepared using a 2% malt-agar medium at pH 7.0 in which Penicillium sp. was inoculated. Penicillium sp. growing in a liquid medium showed a decrease in biomass in presence of Cu(NO3)2 (800 mg.L-1), suggesting that the metal impacts the growth rate of the fungus. Moreover, the combined factors of pH and the presence of the inorganic contaminant impacted the radial growth of the fungus, causing inhibition of 73% at pH 4.0, 75% at pH 7.0, and 77% at pH 9.0 in liquid medium compared to control. However, images obtained with scanning electron microscopy showed the integrity of the fungus cell, even at high doses of copper in the medium. Therefore, it can be concluded that Penicillium sp. isolated from kefir grain can bioremediate the environment and that the harmful effects of heavy metals can be minimized as a result biosorption. Although the growth of Penicillium sp. is inhibited, such retardation requires high doses of copper nitrate, thus ensuring the use of this microorganism for protection against the harmful effects of non-essential copper in the environment.

Biological nitrogen (N) provisioning is a seminal function of the gut microbes in several terrestrial insects, given the unbalanced carbon (C) and N ratios of their diets. Although freshwater insects face comparable dietary N limitations like terrestrial insects, little is known about this function by their gut microbiomes. In this study, we investigated microbial nitrate reduction to ammonium pathways as possible routes of biological N provisioning in two freshwater insects; filter-feeding Hydropsychidae and grazers/collectors Baetidae. After incubation in filtered (microbe-free) artificial stream water (ASW) containing dissolved ¹⁵N-labeled nitrate (treatment) or standard nitrate (control), bulk δ¹⁵N values of treatment samples (Baetidae = 100.62 ± 10.23, mean ± S.E.; Hydropsychidae = 76.82 ± 7.20) were significantly higher than controls (Baetidae = 10.14 ± 0.12 ; Hydropsychidae = 9.03 ± 0.20) in both functional feeding groups (F _{(3, 13)} = 296, P < 0.0001). The treatment δ¹⁵N values are cautiously interpreted as reflecting uptake and incorporation of microbe-derived ¹⁵N-metabolites (¹⁵NH₄ or ¹⁵N-amino acids) into host tissues following nitrate reduction to ammonium pathways in the gut lumen. Microbial nitrate reduction to ammonium activities was assessed via the quantification of dissimilatory (nrfA) and assimilatory (nasA) nitrate reduction to ammonium gene transcripts. There were no significant differences between control and treatment groups within each insect groups. Overall, this study provides a demonstration of the feasibility of applying ¹⁵N-stable isotope analysis for investigating, potential symbiotic functions of freshwater insect gut microbiomes, despite the preliminary nature of the results.

Low efficiency and excessive application of nitrogen (N) fertilizers is a frequent problem in agriculture that impacts the environment. Omeprazole (OMP) has been reported to promote N uptake and assimilation in tomato, basil, and corn. However, information about the effect of omeprazole on N assimilation, recovery and N use efficiency parameters for bean plants is limited. Therefore, the objective of the present study was to determine the effect of foliar application of OMP at 0, 1, 10 and 100 µM on nitrogen assimilation, growth, yield, nitrogen use efficiency parameters and recovery percentage in green bean plants. Green bean plants cv. Strike grown in pots were used. Biomass, yield, nitrate reductase activity, photosynthetic pigments concentration, soluble amino acids and protein concentration, total nitrogen concentration, nitrogen use efficiency parameters and nitrogen recovery were analyzed. The results obtained indicate that the application of OMP at 1 µM increased yield, biomass and promoted N assimilation through higher NR enzyme activity, higher amino acid concentration, higher N use efficiency coefficient and allowed a more efficient nitrogen recovery percentage.

Little is known about the effect of nitrogen nutrition on seedling susceptibility to seed-borne pathogens. We have previously shown that seedlings grown under high nitrate (5 mM) conditions are less susceptible than those grown under low nitrate (0.1 mM) and ammonium (5 mM) in the Arabidopsis-Alternaria brassicicola pathosystem. However, it is not known how seedling metabolism is modulated by nitrogen nutrition, nor what is its response to pathogen infection. Here, we addressed this question using the same pathosystem and nutritive conditions, examining germination kinetics, seedling development, but also shoot ion contents, metabolome and selected gene expression. Nitrogen nutrition clearly altered the seedling metabolome. A similar metabolomic profile was observed in inoculated seedlings grown at high nitrate levels and in not inoculated-seedlings. High nitrate levels also led to specific gene expression patterns (e.g. polyamine metabolism), while other genes responded to inoculation regardless of nitrogen supply conditions. Furthermore, the metabolites best correlated with high disease symptoms were coumarate, tyrosine, hemicellulose sugars and polyamines, and those associated with low symptoms were organic acids (tricarboxylic acid pathway, glycerate, shikimate), sugars derivatives and β-alanine. Overall, our results suggest that the beneficial effect of high nitrate nutrition on seedling susceptibility is likely due to nutritive and signaling mechanisms affecting developmental plant processes detrimental to the pathogen. In particular, it may be due to a constitutively high tryptophan metabolism, as well as down regulation of oxidative stress caused by polyamine catabolism.

: The primary use of the potato is direct consumption. The classification of potatoes into consumption type is made on the basis of consumption evaluation of hydrothermally treated tubers. Nutrient deficiency in the soil, including Mg, contributes to the inhibition of potato growth and development and negatively affects the cooking characteristics of the tubers. Magnesium and biostimulants are responsible for plant nitrogen management. Thus, it is necessary to control the content of harmful nitrates and toxic nitrites in tubers. In potato production, it is also important to maintain appropriate conditions during storage. The purpose of this study was to determine the effect of mineral fertilization with magnesium and a biostimulant preparation during cultivation of edible potato and long-term storage on the traits determining utility-consumption type and the content of harmful nitrogen compounds in tubers. The study was conducted using the early potato variety Satina. In a three-year (2015-2017) three-factor experiment: evaluation date (immediately after harvest and after six months of storage), soil fertilization with mineral magnesium (0, 30, 60, 90 kg MgO ha-1), application of an amino acid biostimulant (0, 1.5, 3.0 l ha-1), five consumption traits of tubers after cooking determining the utility-consumption type of potato were determined. In addition, the consumer safety of the potato was determined based on the nitrate and nitrite content of the tubers. The tendency to overcook tubers significantly depended on all the factors used during potato cultivation. In contrast, the texture and structure of tuber flesh after cooking depended only on mineral fertilization with magnesium. The flesh of the tubers after cooking was most tender (1.0 pt) and firm (1.4 pt) after the application of 60 and 90 kg MgO ha-1, respectively. The moisture level of the flesh after cooking was significantly influenced by the interaction of magnesium fertilization with the biostimulant preparation. The factors of the experiment did not modify the utility-consumption type (B/A), while there was a change in the type in relation to that specified by the grower - general utility (B). The applied cultivation technology and long-term storage did not deteriorate the consumer safety of tubers (&lt;200 mg kg-1) in terms of NO3 (max. 112.1 mg kg-1) and NO2 (max. 1.08 mg kg-1) contents. Organoleptic traits: mealiness, moisture and flesh structure were significantly positively related to nitrate and nitrite content of tubers immediately after harvesting and to nitrite concentration after long-term storage.

Nitrogen fertilization ensures the proper growth of trees. The aim of the study was to evaluate the impact of differentiated nitrogen fertilization on selected parameters. It was assumed that such analysis is an indirect picture of the needs of cherries grown in herbicide fallow. The content of minerals in two layers of the soil, in leaves, and its influence of tree growth, and the content of chlorophyll in leaves were assessed. The experiments were carried out in three different cherry orchards. Three levels of fertilization were applied in each orchard: 0 kg, 60 kg and with 120 kg N ha- 1. As expected the fertilization resulted in an increase in the content of nitrate and ammonium forms of nitrogen in the soil, however, their content was also dependent on precipitation and temperature. Additionally a high nitrogen fertilization increased the content of phosphorus and potassium and decreased the magnesium in the topsoil layer. High nitrogen fertilization caused the decrease of content of phosphorus and potassium in the leaves. The level of calcium and magnesium in leaves increased with fertilization of 60 kg N ha–1 but decreased with the dose to 120 kg N ha–1. The use of nitrogen fertilization increased the vegetative growth of trees measured by leaf area and trunk cross-sectional area. However, the chlorophyll content was not dependent on the amount of nitrogen fertilization.

In order to better understand the biogeochemical cycle of nitrogen in meromictic lakes, which can serve as a model for past aquatic environments, we measured dissolved concentrations of nitrate, nitrite, ammonium and organic nitrogen in deep (39 m maximal depth) subarctic Lake Svetloe (NW Russia). The lake is a rare type of freshwater meromictic water boy with high concentrations of methane, ferrous iron, manganese and low concentrations of sulfates and sulfides in the monimolimnion. In the oligotrophic mixolimnion, the concentration of mineral forms of nitrogen decreased in summer compared to winter, likely due to phytoplankton bloom. The decomposition of the bulk of organic matter occurs under microaerophilic/anaerobic conditions of the chemocline and accompanied by the accumulation of nitrogen in the form of N-NH4 in the monimonimlion. We revealed a strong relationship between methane and nitrogen cycles in the chemocline and monimolimnion horizons. The nitrate concentrations in Lake Svetloe varied in the range from 9 to 13 μM throughout the water column. This fact is rare for meromictic lakes, where nitrate concentrations up to 13 µM are found in the monimolimnion zone down to the bottom layers. We hypothesize, in accord with available data for other stratified lakes, that under conditions of high concentrations of manganese and ammonium at the boundary of redox conditions and below, anaerobic nitrification with the formation of nitrates occurs. Overall, most of organic matter in Lake Svetloe undergoes biodegradation essentially under microaerophilic/anaerobic conditions of the chemocline and the monimolimnion. Consequently, the manifestation of the biogeochemical nitrogen cycle is expressed in these horizons in the most vivid and complex relationship with other cycles of elements.

Copper-based nanoparticles were synthesized using the glycine–nitrate process (GNP) by using copper nitrate trihydrate [Cu(NO₃)₂‧3H₂O] as the main starting material and glycine [C₂H₅NO₂] as the complexing and incendiary agent. The as-prepared powders were characterized through X-ray diffraction (XRD), Brunauer–Emmett–Teller (BET) analysis, X-ray photoelectron spectroscopy, and scanning electron microscopy. Using Cu(NO₃)₂.3H₂O as the oxidizer (N) and glycine as fuel (G), we obtained CuO, mixed-valence copper oxides (CuO + Cu₂O, G/N = 0.3–0.5), and metallic Cu (G/N = 0.7). The XRD and BET results indicated that increasing the glycine concentration (G/N = 0.7) and reducing particle surface area increased the yield of metallic Cu. The effects of varying reaction parameters such as catalyst activity, catalyst dose, and H₂O₂ concentration on nonylphenol-9-polyethoxylate (NP9EO) degradation were assessed. With a copper‐based catalyst in a heterogeneous system, the NP9EO and total organic carbon removal efficiencies were 83.1% and 70.6%, respectively, under optimum operating conditions (pH, 6.0; catalyst dose, 0.3 g/L; H₂O₂ concentration, 0.05 mM). The results suggested that removal efficiency increased with an increase in H₂O₂ concentration but decreased when the H₂O₂ concentration exceeded 0.0.5 mM. Furthermore, the trend of photocatalytic activity was as follows: G/N = 0.5 > G/N = 0.7 > G/N = 0.3. The G/N = 0.5 catalysts showed the highest photocatalytic activity and resulted in 94.6% NP9EO degradation in 600 min.

Nitrate is rich in Mars sediments owing to long-term atmospheric photolysis, oxidation, and deposition coupled with a lack of leaching via rainfall. The Atacama Desert in Chile, which is similarly dry and rich in nitrate deposits, is used as a Mars analog in this study to explore the potential effects of high nitrate levels on microbial growth. Seven study sites sampled across an aridity gradient in the Atacama Desert were categorized into 3 clusters – hyperarid, intermediate, and arid sites, as defined by major elements in the regolith, associated biomass, and precipitation. Intriguingly, the distribution of nitrate concentrations in the shallow subsurface suggests that the buildup of nitrate is not solely controlled by precipitation. Correlations of nitrate with SiO2/Al2O3 and grain sizes suggest that sedimentation rates are also important in controlling nitrate distribution. At arid sites receiving more than 10 mm/yr precipitation, rainfall shows a stronger impact on biomass than nitrate does. However, high nitrate to organic carbon ratios are generally beneficial to N assimilation as evidenced both by soil geochemistry and enriched culturing experiments. This study suggests that even in the absence of precipitation on contemporary Mars, the nitrate levels are sufficiently high to benefit potentially extant Martian microorganisms.

In potato, high levels of nitrogen (N) can lead to excessive vegetative growth at the expense of tuber development, resulting in lower yield and poor quality tubers. We found that Solanum tuberosum CLE4 (StCLE4) is expressed most strongly in the roots on N-rich media and positively regulates root growth of potato in N-deficient conditions. We noted that StCLE4 functions as a negative regulator of normal shoot apex development similar to CLV3 in Arabidopsis. Transcriptomic analysis revealed that overexpression of StCLE4 results in repression of the StIT1 gene, a regulator of potato tuber initiation. Overexpressing StCLE4 stolons were transformed into branches that were similar to milder it1 phenotype. We also found that NIN-like proteins, key regulators of nitrate signaling, bind to the regulatory sequence of StIT1 in yeast one-hybrid assay. Taken together, our findings suggest that StCLE4 regulates shoot, root and stolon growth in potato.

Hypertensive diseases of pregnancy (HDP) represent a global clinical challenge, affecting 5-10% of women and leading to complications for both maternal well-being and fetal development. At the heart of these complications is endothelial dysfunction, with oxidative stress emerging as a pivotal causative factor. The reduction in nitric oxide (NO) bioavailability is a vital indicator of this dysfunction, culminating in blood pressure dysregulation. In the therapeutic context, although antihypertensive medications are commonly used, they come with inherent concerns related to maternal-fetal safety, and a percentage of women do not respond to these therapies. Therefore, alternative strategies that directly address the pathophysiology of HDP are required. This article focuses on the potential of the nitrate-nitrite-NO pathway, abundantly present in dark leafy greens and beetroot, as an alternative approach to treating HDP. The objective of this review is to discuss the prospective antioxidant role of nitrate. We hope our discussion paves the way for using nitrate to improve endothelial dysfunction and control oxidative stress, offering a potential therapy for managing HDP.

Legumes are a diverse and important group of plants that play a vital role in agriculture, food security, and environmental sustainability. Rhizobia are symbiotic bacteria that form nitrogen-fixing nodules on legume roots, providing the plant with a valuable source of nitrogen. Phenolic acids are a group of secondary metabolites produced by plants that have a wide range of biological functions, including defense against pests and diseases, tolerance to abiotic stresses, and nutrient uptake. In the context of climate change and the imperative for sustainable agriculture, this study delves into the dynamic responses of legume species to varying light intensities and their intricate interactions with soil microorganisms. We investigated the impact of light intensity and rhizobial inoculation on the biomass, nitrate reductase, acid phosphatase, and production of phenolic acids in the roots of four legume species, Trifolium repens, Vicia sativa, Ornithopus compressus, and Coronilla juncea. Plants were grown under three light intensity regimes (low, medium, and high) and inoculated with either rhizobia or a non-inoculated control. The results highlights that shaded-light-adapted species, T. repens and V. sativa, increased root exudate production when exposed to high light intensity. This response aligns with their mining strategy, effectively allocating resources to optimize nutrient acquisition under varying conditions. In contrast, species hailing from well- illuminated environments, O. compressus and C. juncea, displayed distinct strategies by significantly increasing biomass under high irradiance, capitalizing on the available light and nutrients. The mining strategy of legumes emerged as a central theme, influencing biomass production, nitrogen dynamics, and enzymatic activities. The strong correlations between biomass and total nitrogen accumulation underscore the role of the mining strategy in efficient nutrient acquisition. Inoculated plants, which rely more on biological nitrogen fixation (BNF), exhibited lower δ15N values, indicative of a successful mining strategy to acquire and utilize atmospheric nitrogen. Enzymatic activities and phenolic acids exhibited significant interspecies variations, reflecting the adaptability of legumes to different light conditions. The findings of this study could be used to develop new strategies for improving legume stress tolerance, nutrient uptake capacity, and rhizosphere health.

Nitrogen (N) strongly affects plant growth and metabolism. For shikonin biosynthesis, although the ammonium toxicity phenomenon has been reported, the effects of nitrogen on the shikonin synthesis remains obscure. In this study, we carried out four different concentrations of NH4+ treatments on Arnebia euchroma hairy roots (AEHR) to clarify the influence of NH4+ on the growth and shikonin accumulation in A. euchroma and the possible mechanism. The results showed that compared with the 0% NH4+ treatment (only nitrate as nitrogen source), 10% NH4+ treatment increased the fresh weight and the dry weight of AEHR, and promoted the synthesis of shikonins; 20% NH4+ treatment started to show the inhibition effects on growth and shikonin accumulation of AEHR and 30% NH4+ treatment exhibited the strongest inhibition effects. With increased percentage of NH4+, AEHRs became shorter and thicker with more branches. To further elucidate the mechanism, we analyzed the time-course of nitrogen assimilation, gene expression level of key enzymes involved in the shikonin biosynthesis pathway, and contents of various endogenous hormones under NH4+ toxicity condition. Results indicated that auxin and cytokinin might regulate the growth and architecture of AEHR under NH4+ toxicity, and jasmonate level was reduced along with the inhibition of shikonin biosynthesis. This first comprehensive investigation into the effects of ammonium/nitrate ratio on shikonin biosynthesis not only provided valuable data for optimizing the in vitro culture and shikonin production in A. euchroma, but also suggested potential fertilizer strategies for its cultivation.

Bare titania and metal promoted TiO2 catalysts were employed in the treatment of nitrates, which are ubiquitous pollutants of wastewater. The results show that the process can be carried out under visible light (from white light LED lamp) and, in the best case, 23.5% conversion of nitrate was obtained over 4 hours with full selectivity towards N2 by employing 0.1 mol% Ag/TiO2 prepared by flame spray pyrolysis. Moreover, the performance was worse when testing the same catalysts with tap water (11.3% conversion), due to the more complex composition of the matrix. At last, it was found that the photoreduction of nitrate can be effectively performed in combina-tion with the photo-oxidation of ammonium without loss in the activity, opening to the possi-bility to treat highly polluted wastewater with a single process. The latter treatment employs the two contaminants simultaneously as electron and holes scavengers, with very good selectiv-ity, in a completely new process that we may call Photo-Selective Catalytic Reduction (Pho-to-SCR).

To investigate this suitability of Black Mustard (Brassica Nigra L.) and Camelina (Camelina Sativa L.) for pulp manufacturing the nitrate-alkaline method was used. The non-wood plants were characterized by chemical analysis, especially lignin, cellulose, ash, extractives and alpha-, beta-, gamma-cellulose. The pulp was cooked in 6% nitric acid and then underwent the extraction by 5% sodium-hydroxide and neutralized by 1% acetic acid. The cooked pulp was characterized by delignification degree – Kappa number. The laboratory sheets were made from this cooked pulp and they were characterized by tensile index, breaking length, smoothness and compared with commonly available papers.

The recently defined and yet rather new topic of Healthy aging is gathering more attention on the global stage. With world’s population getting older - it is rapidly becoming very important to develop and maintain functional abilities in older age and develop mechanisms to protect senior population from chronic diseases. One of the most effective components, as well as, one may call - process involved in, and strongly associated with aging is recently discovered and Nobel prize awarded - nitric oxide as a signaling molecule, which followed by later discoveries showed to have a positive metabolic, immune and anti-inflammatory effects. One of the most debated topics of the last decade in the scientific community is nitrates, one of the pathways involved in nitric oxide production. Thus, the objective of this study was to evaluate effect of different potassium nitrate concentrate supplementation on Drosophila melanogaster longevity. 0,5-3% potassium nitrate medium was analyzed on the life span and motor function in different groups consisting of 100 females fruit flies each. In this assay, female fly species supplemented with potassium nitrate diet showed life span increase by 18.6% and 5.1% with 1% and 2% KNO3- respectively with a positive impact on locomotor function. In conclusion, we found that low concentration of potassium nitrate medium increased lifespan and locomotor function in Drosophila melanogaster.

Genes coding for enzymes of the denitrification pathway appear randomly distributed among isolates of the ancestral genus Thermus, but only in few strains of the species T. thermophilus the pathway has been studied to a certain detail. Here, we review the enzymes involved in this pathway present in T. thermophilus NAR1, a strain extensively employed as a model for nitrate respiration, on the light of its full sequence recently assembled through a combination of PacBio and Illumina technologies in order to counteract the systematic errors introduced by the former technique. The genome of this strain is divided in four replicons, a chromosome of 2,021,843 pb, two megaplasmids of 370,865 and 77,135 bp and a small plasmid of 9,799 pb. Nitrate respiration is encoded in the largest megaplasmid, pTTHNP4, within a region that includes operons for O2 and nitrate sensory systems, a nitrate reductase, nitrate and nitrite transporters and a nitrate specific NADH dehydrogenase, in addition to multiple insertion sequences (IS), suggesting its mobility-prone nature. Despite nitrite is the final product of nitrate respiration in this strain, the megaplasmid encodes two putative nitrite reductases of the cd1 and Cu-containing types, apparently inactivated by IS. No nitric oxide reductase genes have been found within this region, although the NorR sensory gene, needed for its expression, is found near the inactive nitrite respiration system. These data clearly support that partial denitrification in this strain is the consequence of recent deletions and IS insertions in genes involved in nitrite respiration. Based on these data, the capability of this strain to transfer or acquire denitrification clusters by horizontal gene transfer is discussed.

We have studied on the stress measurement by making use of salivary nitrate, which can be a candidate of stress markers, with ion-selective field-effect transistors (ISFETs). ISFETs are suitable for on-site single-drop analysis of salivary nitrate within 10 seconds. However, when ISFETs are used for salivary nitrate, ISFETs have a problem which is called the initial drift. The initial drift makes it difficult for determination of an accurate nitrate monitoring. Thus, the purpose of this study is to suppress an initial drift and to search for new easy polymer to possess more performance of sensor responses than conventional matrix membrane such as PVC. In this research, we investigated ISFETs using specific matrix membrane for example, KP-13, Pellethane^® and P7281-PU. The initial drift was evaluated from the fluctuation of the response values generated by ISFETs which are immersed in saliva or aqueous solution. As a result, P7281-PU showed its suppression effect for the initial drift in the whole saliva and various solutions. Furthermore, the cause of drift may be H⁺ diffusion, and drift suppression effect of P7281-PU may be affected by urethane bond capturing H⁺ in ion-selective membrane. This result suggests a continuous nitrate monitoring and development of wearable sensors.

In this study, it was analyzed that the characteristics of ammonia and their effects on secondary particulate matter (PM) formation by region in Korea during 2020. The NH3 concentration was high in GJ (11.4 ppb) neighboring agricultural area followed by DJ (9.0 ppb) and SE (8.6 ppb) locating in the urban area. On the other hand, BI (2.6 ppb) and JI (4.5 ppb), which are background regions, were lower than other area. Seasonally, ammonia was high in spring and summer, and it generally increased when human activities are active. Therefore, it is believed that ammonia in the atmosphere changes depending on not only local emissions but also temperature-dependent phase distribution characteristics. For SE and GJ, regions with relatively high ammonia concentrations, the effect of ammonia on secondary PM formation was conducted. In both regions, the ammonium-to-sulfate mole ratio tended to increase with an ammonia or PM2.5 concentration increasing. It can be assumed that PM2.5 concentration increases as nitrates are formed under the ammonia sufficient condition. The adjusted gas ratio is generally greater than 4, indicating that there is a lot of free ammonia. Thus, it is estimated that reduction of ammonia would not effective to restrain the nitrate formation.

In addition to absorbing nitrogen from the soil, legumes have the ability to use atmospheric N2 through symbiotic nitrogen fixation. Therefore, legumes have developed mechanisms regulating nodulation in response to the amount of nitrate in the soil; in the presence of high nitrate concentrations, nodulation is inhibited while low nitrate concentrations stimulate nodulation and nitrogen fixation. This allows the legumes to switch from soil nitrogen acquisition to symbiotic nitrogen fixation. Recently, a particular interest has been given to the nitrate transporters, such as NPF or NRT2, having a role in the functioning of nodules. Nitrate transporters of the two model plants, Lotus japonicus and Medicago truncatula, shown to have a positive and/or a negative role in nodule functioning, depending on nitrate concentration, are presented in this review. Also, by exploiting available genomic and transcriptomic data in the literature we have identified the complete PsNPF family in Pisum sativum (69 sequences previously described and 21 new that we have annotated) and putative nitrate transporters candidate for playing a role in nodule functioning in P. sativum.

Indole-3-acetic acid (IAA) is a key vegetative hormone that plays crucial roles in plant growth and development. In this study, the effect of the exogenous application of IAA as a biostimulant in the cultivation of onions (Allium cepa) was investigated. Different IAA concentrations were evaluated to determine their effects on onion growth and chemical parameters. Various physical and chemical parameters such as total fresh weight, plant height, chlorophyll content, nitrates, total phenols, and antioxidant capacity (DPPH) were analyzed. The results revealed that exogenous application of IAA had a significant impact on onion growth and quality. Specifically, it was found that certain concentrations of IAA fostered a greater fresh weight of the bulb, an increase in the levels of phenolic compounds and a greater DPPH. However, the onion response to IAA was dependent on the concentration. In conclusion, the present study provides evidence that the exogenous application of IAA as a biostimulant can improve onion growth and quality. These results are relevant for the development of sustainable agricultural practices and could be oriented towards crop improvement.

In this opinion article, we have analyzed the relevancy of a hypothesis which is based on the idea that in Arabidopsis thaliana jasmonic acid (JA)-mediated defense system against necrotrophic fungi is weakened when NO3- supply is high. Such hypothesis is based on the fact that when NO3- supply is high, it induces an increase in the amount of bioactive ABA which induces the sequestration of the phosphatase ABI2 (PP2C) into the PYR/PYL/RCAR receptor. Consequently, the Ca sensors CBL1/9 - CIPK23 are not dephosphorylated by ABI2, thus remaining able to phosphorylate targets such as AtNPF6.3 and AtKAT1, a NO3- and K+ transporters respectively. Therefore, the impact of phosphorylation on the regulation of these two transporters, could 1) reduce NO3- influx as in its phosphorylated state AtNPF6.3 shifts to low capacity state and 2) increase K+ influx, as in its phosphorylated state KAT1 becomes more active. It is also well known that in the roots K+ loading in the xylem and its transport to the shoot is activated in the presence of NO3-. As such, the enrichment of plant tissues in K+ can impair jasmonic acid (JA) regulatory pathway and the induction of the corresponding biomarkers. The latter are known to be up-regulated under K+ deficiency and inhibited when K+ is resupplied. We therefore suggest that increased K+ uptake and tissue content induced by high NO3- supply modifies JA regulatory pathway, resulting in weakened JA-mediated plant’s defense system against necrotrophic fungi.

Soils are an important COS sink. However they can also act as sources of COS to the atmosphere. Here we demonstrate that variability in the soil COS sink and source strength is strongly linked to available soil inorganic nitrogen (N) content across a diverse range of biomes in Europe. We revealed in controlled laboratory experiments that N fertilisation simultaneously decreases the COS sink strength of soils while increasing the COS production rate. Furthermore, we found strong links between variations in the two gross COS fluxes, microbial biomass and nitrate and ammonium contents, providing new insights into the mechanisms involved. Our findings provide evidence for how soil-atmosphere exchange of COS is likely to vary spatially and temporally, a necessary step for constraining the role of soils and land use in the COS mass budget.

Our study aimed to assess sprinkler irrigation's impact on the activity of selected soil enzymes concerning nitrogen metabolism and oxidation-reduction processes in the soil with different doses of inorganic nitrogen fertilizers. The soil Alfisol was sampled from the experimental field of spring barley in the University Research Center in the central part of Poland in the moderate transitory climate during the growing seasons 2015–2017. The index resistance (RS) was derived to recognise the resistance enzymes of drought. In the maturity phase, nitrate reductase activity was at an 18% higher level in irrigated soils and the activity of other enzymes was higher in no-irrigated treatments by 25% in dehydrogenase, 22% in peroxydase 33% in catalase and 17% in urease case. The development stage of barley influenced nitrate reductase activity. Enzymatic activity has been changing in the examined years of the research, depending on the weather conditions. During the maturity stage, the soil's lower ammonium nitrogen content resulted from higher spring barley uptake due to drought stress. Irrigation probably contributes to increased leaching of nitrate in soil. The highest index of resilience was presented in the soil catalase activity.

Green and sustainable electrocatalytic conversion of nitrogen-containing compounds to ammonia are currently in high demand in order to replace the eco-unfriendly Haber-Bosch process. Model catalysts for the nitrate reduction reaction were obtained by electrodeposition of metal Co, Fe and bimetallic Fe/Co nanoparticles from aqueous solutions onto a graphite substrate. The samples were characterized by the following methods: SEM, XRD, XPS, UV-vis spectroscopy, cyclic (and linear) voltammetry, chronoamperometry and electrochemical impedance spectroscopy. Besides, the determination of electrochemically active surface was also performed for all electrocatalysts. The best electrocatalyst was a sample containing Fe-nanoparticles on the layer of Co-nanoparticles, which showed the Faradaic efficiency 58.2% (E=-0.785 V vs. RHE) at the ammonia yield rate of 14.6 μmol h-1 cm-2. An opinion was expressed on elucidation of the mechanism of coordinated electrocatalytic action of a bimetallic electrocatalyst. This work can serve primarily as a starting point for future investigations on electrocatalytic conversion reactions to ammonia using model catalysts of the proposed type.

Contamination in coastal regions attributed to fluoride and nitrate cannot be disregarded, given the substantial environmental and public health issues they present worldwide. Maintaining water quality is crucial for environmental well-being. This comprehensive study was performed to assess the spatial as well as indexical water quality, identify contamination sources, and evaluate associated health risks pertaining to nitrate and flouride in AlHassa region, KSA. The physiochemical results revealed a pervasive pollution of the overall groundwater. The dominant water type was Na-Cl, indicating saltwater intrusion and reverse ion exchange impact. Spatiotemporal variations in physicochemical properties suggest diverse hydrochemical mechanisms, with geogenic factors primarily influencing groundwater chemistry. The groundwater pollution index varied between 0.8426 and 4.7172, classifying samples as moderately to very highly polluted. Similarly, the synthetic pollution index (in the range of 0.5021-4.0715) revealed that none of the samples had excellent water quality, with various degrees of pollution categories. Nitrate Health Quotient (HQ) values indicated chronic human health risks ranging from low to severe, with newborns being the most vulnerable. Household use of nitrate-rich groundwater for showering and cleaning did not pose significant health risks. Fluoride HQ decreased with age, and children faced the highest risk of fluorosis. The Hazard Index (HI) yielded moderate to high-risk values. Nitrate risks were 1.21 times higher than fluoride risks, as per average HI assessment. All samples fell into the vulnerable category based on the Total Hazard Index (THI), with 88.89% classified as very high risk. This research provides valuable insights into groundwater quality, guiding water authorities, inhabitants, and researchers in identifying safe water sources and vulnerable human populations. The results highlight the need for appropriate treatment techniques and long-term coastal groundwater management plans.

We have performed the first comparative analysis of the potential of two physiologically-diverse model cyanobacteria, Synechococcus PCC 7002 (S.7002) and Synechococcus PCC 7942 (S.7942), for the photosynthetic production of four chemically-different high-value terpenes: two monoterpenes limonene and pinene, and two sesquiterpenes bisabolene and farnesene. We showed, for the first time, that S.7002 and S.7942 can produce farnesene and bisabolene, respectively. Both cyanobac-teria produced farnesene (S.7942 better than S.7002) more efficiently than the other tested terpenes (especially pinene the weakest produced terpene). S.7002 produced limonene better than bisabo-lene, whereas S.7942 produced bisabolene better than limonene. These findings suggest that S.7942 is better suited to produce sesquiterpenes than monoterpenes. Also interestingly, higher levels of terpenes were produced by S.7942 and S.7002 expressing a terpene-synthase gene from both a RSF1010-derived replicating plasmid and a neutral chromosomal site, as compared to ei-ther the plasmid alone or the chromosome alone. These results suggest that in both cyanobacteria the production of terpenes is more limited by the activity of terpene synthases than the abondance of terpene precursors. Finally, higher levels of terpenes were produced by S.7002 growing on urea (a frequent pollutant) as compared to nitrate (or ammonium) the standard nitrogen sources for cyanobacteria (more expensive than urea).

The root-colonizing endophytic fungus Piriformospora indica promotes root and shoot growth of its host plants. We show that growth promotion of Arabidopsis leaves is abolished when the seedlings are grown on media with nitrogen (N) limitation. The fungus neither stimulated the total N content nor did it promote 15NO3- uptake from agar plates to the leaves of the host under N-sufficient or N-limiting conditions. However, when the roots were co-cultivated with 15N-labelled P. indica, more label can be detected in the leaves of N-starved host plants, but not of plants supplied with sufficient N. Amino acid and primary metabolite profiles, as well as expression analyses of N metabolite transporter genes suggest that the fungus alleviates the adaptation of its host to the N limitation condition. P. indica alters the expression of transporter genes which participate in relocation of NO3-, NH4+ and N metabolites from the roots to the leaves under N limitation. We propose that P. indica participates in the plant´s metabolomic adaptation to N limitation by delivering reduced N metabolites to the host, alleviating metabolic N starvation responses, and reprogramming the expression of N-metabolism related genes.

Nitrogen and phosphorous support the ecosystem by supplying nutrients to algae and aquatic plants. Having them in excess results in the eutrophication of waters creating quality problems. In the past, nitrogen has been widely investigated for wells in the context of groundwater flow. However, a national-scale nitrogen assessment in rivers and streams has not received enough attention. In this research, the Wilcoxon rank sum test, as a non-parametric hypothesis testing method, has been applied to nitrogen concentration in the form of nitrate-nitrogen and nitrite-nitrogen in rivers and streams of the Contiguous United States. This approach was particularly selected because of the non-normal and positively skewed nitrogen levels occurring in the surface flow. This method was able to identify the impaired body of waters as well as quantify the confidence, significance, and errors involved. The Northern Appalachians (NAP), Northern Plains (NPL), and Xeric (XER) ecoregions were worsening in the nitrogen-nitrate condition with NAP, and XER needed immediate actions. The nitrite-nitrogen condition did not pose an immediate threat, so mitigation plans should focus more on nitrate-nitrogen remediation. It was shown that the method was superior to the two-sample t-test by yielding lower type II errors.

We report a facile route for the green synthesis of trimethylchitosan nitrate-capped silver nanoparticles (TMCN-AgNPs) with positive surface charge. In this synthesis, silver nitrate, glucose, and trimethyl chitosan nitrate (TMCN) were used as silver precursor, reducing agent, and stabilizer, respectively. The reaction was carried out in a stirred basic aqueous medium at room temperature without the use of energy-consuming or expensive equipment. We investigated the effects of the concentrations of NaOH, glucose, and TMCN on the particle size, zeta potential, and formation yield. The AgNPs were characterized by UV-visible spectroscopy, photon correlation spectroscopy, laser Doppler anemometry, transmission electron microscopy, X-ray diffraction, and X-ray photoelectron spectroscopy. The catalytic activity of the TMCN-AgNPs was studied by the reduction of 4-nitrophenol using NaBH₄ as a reducing agent. We evaluated the antibacterial effects of the TMCN-AgNPs on Acinetobacter baumannii, Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus using the broth microdilution method. The results showed that both gram-positive and gram-negative bacteria were killed by the TMCN-AgNPs at very low concentration (< 6.13 μg/mL). Moreover, the TMCN-AgNPs also showed high antibacterial activity against clinically isolated multidrug-resistant A. baumannii strains, and the minimum inhibitory concentration (MIC) was ≤ 12.25 μg/mL.

In recent years, soil addition with organic amendments, such as biochar and compost, has gained attention as an effective agronomic practice to sustain soil fertility, enhance plant growth and crop yield. Well known are the positive effects of compost on yield of a wide crop varieties, while both positive and negative responses are reported for biochar Therefore, the aim of the study was to verify the effect of biochar mixed with three types of compost on quanti-qualitative response of Swiss chard (Beta vulgaris L. cycla), a leafy green vegetable rich in dietary antioxidants, largely consumed worldwide. A factorial experiment in pots with two factors, including biochar (without biochar and with biochar from vine pruning residues) and compost (without compost, with compost from olive pomace, with vermicompost from cattle manure and with compost from cattle anaerobic digestate), was setup. Two growth cycles were considered and a set of quantitative (height of plants, number, area and fresh weight of leaves) and qualitative parameters (carotenoid, chlorophyll, total N and NO3-content of leaves) were analyzed. Biochar decreased plant growth and NO3- leaf content; on the contrary, it increased total N leaf content, while compost improved all the considered parameters. The interactive effect of biochar and compost was evident only on total N and NO3- leaf content. In our experimental conditions, the compost showed to be the best option to improve Swiss chard growth and increase the phytopigments content, while the biochar-compost mixtures didn’t produce the expected effect.

Nitrates can cause severe ecological imbalances in aquatic ecosystems, with considerable consequences for human health. Therefore, monitoring this inorganic form of nitrogen becomes essential for any water quality management structure. This research aimed at developing a novel Nitrate Portable Measurement System (NPMS) for monitoring nitrate concentrations in water samples. NPMS is a reagent-free ultraviolet system developed using low-cost electronic components. Its operation principle is based on Beer-Lambert law for measuring Nitrate concentrations in water samples through light absorption in the spectral range 295-315 nm. The system is equipped with ready to use ultraviolet sensor, Light Emission Diode (LED), op-amp, microcontroller, liquid crystal display, quartz cuvette, temperature sensor, and battery. All the components are assembled in a 3D-printed enclosure box which allows a very compact self-contained equipment, with high portability, enabling field and near real time measurements. The proposed methodology and the developed instrument were used to analyse multiple nitrate standard solutions. The performance was evaluated in comparison with the Nicolet Evolution 300, a classical UV-Vis spectrophotometer. The results demonstrate a strong correlation between the retrieved measurements by both instruments within the investigated spectral band and for concentrations above 5 mg NO3- /l.

Nitric oxide (NO) is synthesized and is involved in various physiological and developmental process regulation in all kingdoms of life. Fungi have been less explored in terms of endogenous NO biology than mammals, plants, and bacteria. In this review, we summarize and discuss the studies to date on intracellular NO biosynthesis and function in fungi. Two mechanisms for NO biosynthesis, NO synthase (NOS)-mediated arginine oxidation, and nitrate and nitrite reductase-mediated nitrite reduction, are most frequently reported. Furthermore, we summarize the multifaceted functions of NO in fungi, as well as its role as a signaling molecule in fungal growth regulation, development, abiotic stress, virulence regulation, and metabolism. Finally, we present potential directions for future research on fungal NO biology.

Improving the quality of natural water purification is one of the priority areas in the research of scientific communities in the field of ecology. At the same time, the task is to achieve optimal efficiency at a low cost of the technological process. The solution in this case is the use of materials necessary for cleaning, in particular, sorbents from natural raw materials, including agricultural waste. At present, a sufficient number of research results have been published confirming the effectiveness of the sorbent from biochar of various agricultural wastes, as well as from rice straw biochar. This article proposes an innovative method for modifying biochar from rice straw, which allows the use of the material as a sorbent with a disinfecting effect. The method consists in processing biochar in a process activation apparatus (PAP) in a solution of silver nitrate with the release in the form of a carbon nanomaterial with attached metallic silver ions on the surface of biochar particles. Rice straw biochar was mixed in distilled water with silver nitrate AgNO3 and placed in the AAP in a replaceable insert with ferromagnetic particles that have a magnetostrictive effect under the influence of an electromagnetic field. Next, the contact of biochar impregnated with a solution of silver nitrate and ferromagnetic particles was carried out for 2 minutes under electromagnetic influence, after which the treated sample was dried in an oven for 4 hours at t=105ºС, followed by calcination in a muffle furnace at 450ºС for 3 hours. The resulting silver-containing sorbent was subjected to a physicochemical analysis of its properties, photographs of electron microscopy were also obtained, and a bacteriological analysis of the effectiveness of the sorbent on natural water was carried out. According to the results of the analyzes, the silver-containing sorbent is a mesoporous nanomaterial with an average mesopore diameter for desorption of 196 Å, a relative volume of pores with a diameter of up to 900 Å 0.034 cm³/g,, specific surface 7.45 m²/g and a content of metallic silver ions on the surface of the sorbent particles of (Ag) 16.7%, oxygen (O) 54.9%, unexpectedly (С) 13.7%, silicon (Si) 8.1%. Laboratory studies for the bacteriological analysis of natural water consisted in the sorption processing of the selected samples in the carbonization mode, followed by sedimentation after the introduction of the reagent (a mixture of coagulant and flocculant). The analysis was carried out on three indicators - total microbial count (TMC), total coliform bacteria (TCB) and thermotolerant coliform bacteria (TCB). According to the research results, the sorbent showed its disinfecting properties and confirmed its high efficiency (90,48%-100%).

The detection of microplastics (MPs) in drinking water has emerged as a critical environmental concern, with significant implications for public health. However, the variability in identification methods across studies has made it difficult to compare results, underscoring the urgent need for standardized protocols. This study presents a comprehensive meta-analysis and practical ap-proach to standardizing Fourier Transform Infrared Spectroscopy (FTIR)-based methodologies for detecting MPs in drinking water. By systematically reviewing studies from 2019 to 2023, this research identifies the most effective techniques for MP quantification, with an emphasis on the use of 0.45 μm cellulose nitrate filters and Nile red staining for fluorescence detection. The me-ta-analysis demonstrates consistent results across studies, supported by statistical analysis con-firming the reproducibility and reliability of the methodologies employed. Experimental findings highlight that the cellulose nitrate filter offers superior microplastic retention with low back-ground fluorescence, making it ideal for FTIR analysis. Moreover, the Nile red spray method is recommended for its efficiency and uniformity in staining, and random counting proves to be a rapid and reliable quantification technique. This study advocates for the adoption of standard-ized protocols, particularly in filter selection and staining methods, to ensure accurate and com-parable MP detection. The findings contribute valuable insights for optimizing water treatment processes and provide a solid foundation for future research and regulatory frameworks aimed at mitigating microplastic contamination in water resources.

The possibility of deep radiochemical purification of Li2CO3 has been examined in the frame of the purification program of AMoRE collaboration. In this experiment, commercial Li2CO3 was converted into LiNO3. Co-precipitation with inorganic salt-based carriers followed by membrane filtration and sorption using MDM inorganic sorbent methods were tested for the removal of alkaline-earth and transition metals, potassium, magnesium, aluminum, uranium, thorium, and radium. The calcium molybdate-based carrier was the most efficient for removing Th, U, and K. Subsequently, the radium, calcium, and barium contaminations were removed with MDM sorbent. After the impurities’ removal, the final Li2CO3 product was synthesized with NH4HCO3 sludge. The separation factors were derived by means of ICP-MS and HPGe analyses of the initial material, intermediate and final products. The study showed the optimum conditions of co-precipitation and sorption to reach a high yield and radiopurity of lithium carbonate used for low radioactive background experiments.

In Nitrate Vulnerable Zones (NVZ) site-specific techniques are needed to match N availability with durum wheat (Triticum turgidum subsp. durum Desf.) requirements. Enhanced-efficiency fertilizers (EEF) can improve efficient N supply and reduce leaching, thus contributing to sustainable agriculture. To study the effects of rates, sources and timings of nitrogen application, two-year field experiments were carried out at two Mediterranean NVZs of Central Italy (Pisa and Arezzo). The trial compared: i) two N rates: one based on the crop N requirements (NO), the other on the Action Programmes’ prescriptions of the two NVZ (NAP); ii) three N sources (urea, methylene urea (MU), and nitrification inhibitor (NI) 3,4-Dimethylpyrazole phosphate (DMPP); and two top-dressing timings (1st tiller visible and 1st node detectable). Grain yield and yield components were determined, together with N uptake. Results showed that: i) grain and biomass production were reduced with NAP at both locations; ii) urea performed better than slow-release fertilizers; iii) the best application time varied depending on N source and location: at Pisa enhanced-efficiency fertilizers achieved higher yields when applied earliest, while for urea the contrary was true; at Arezzo different N fertilizers showed similar performances between the two application timings. Different behaviors of top-dressing fertilizers at the two localities could be related to the diverse patterns of temperatures and rainfall. Therefore, optimal fertilization strategies vary according to environmental conditions.

The work presents the results of studying dilute aqueous solutions of commercial Ln(NO3)3 · xH2O salts with Ln=Ce-Lu using X-ray diffraction (XRD), IR spectroscopy, X-ray absorption spectroscopy (XAS: EXAFS/XANES) and pH measurements. As a reference point, XRD and XAS measurements for characterized Ln(NO3)3 · xH2O microcrystalline powder samples were performed. The local structure of Ln-nitrate complexes in 20 mM Ln(NO3)3 · xH2O aqueous solution was studied under total external reflection conditions and EXAFS geometry was applied to obtain high-quality EXAFS data for solutions with low concentrations of Ln3+ ions. Results obtained by EXAFS spectroscopy showed significant contraction of the first coordination sphere during the dissolution process for metal ions located in the middle of the lanthanide series. It was established that in Ln(NO3)3 · xH2O solutions with Ln=Ce, Sm, Gd, Yb (c=134, 100, 50 and 20 mM) there are coordinated and, to a greater extent, non-coordinated nitrate groups with bidentate and predominantly monodentate bonds with Ln ions, the number of which increases upon transition from cerium to ytterbium. Cross-relationships between the concentration of solutions, pH value and antimicrobial activity with the type of Ln = Ce, Sm, Gd, Yb were established, as well as the absence of biocidal properties of solutions with a concentration of 20 mM, except for Ln = Yb. The important role of experimental conditions in obtaining and interpreting the results was noted.

Plant N concentration (PNC) has been commonly used to guide farmers in assessing maize (Zea mays L.) N status and making in-season N fertilization decisions. However, PNC varies based on the development stage and therefore a relationship between biomass and N concentration is needed (i.e., critical N dilution curves; CNDC) to better understand when plants are N deficient. A few CNDC have been developed and used for plant N status diagnoses, but have not been tested in the US Midwest. The objective of this study was to evaluate under highly diverse soil and weather conditions in the US Midwest the performance of CNDCs developed in France and China for assessing maize N status. Maize N rate response trials were conducted across eight US Midwest states over three years. This analysis utilized plant and soil measurements at V9 and VT development stages and final grain yield. Results showed that the French CNDC (y=34.0x-0.37, where y is critical PNC, x is aboveground biomass) was better with a 91% N status classification accuracy compared to only 62% with the Chinese CNDC (y=36.5x-0.48). The N nutrition index (NNI), which is the quotient of the measured PNC and the calculated critical N concentration (Nc), based on the French CNDC was significantly related to soil nitrate-N content (R2=0.38-0.56). Relative grain yield on average reached a plateau at NNI values of 1.36 at V9 and 1.21 at VT, but for individual sites ranged from 0.80 to 1.41 at V9 and from 0.62 to 1.75 at VT. The NNI threshold values or ranges optimal for crop biomass production may not be optimal for grain yield production. It is concluded that the CNDC developed in France is suitable as a general diagnostic tool for assessing maize N status in US Midwest. However, the threshold values of NNI for diagnosing maize N status and guiding N applications vary significantly across the region, making it challenging for guiding specific on-farm N management. More studies are needed to determine how to effectively use CNDC to make in-season N recommendations in the US Midwest.

Abstract: Background: COVID-19 is associated with various changes in immunity, including signaling through the pattern-recognition receptors (PRR). However, the role of these processes is poorly understood. The present study aimed to evaluate the relationships of the serum levels of the cyclic dinucleotide 2’3’-cyclic GMP-AMP (cGAMP) marker of PRR activation with immunoglobulin G antibodies against severe acute respiratory syndrome-linked coronavirus (IgG-SARS)-positive status and endothelial dysfunction in apparently healthy subjects and in patients with coronary heart disease (CHD). Methods: The present study investigated selected groups from two cohorts, including cohort 1 of 307 healthy volunteers and cohort 2 of 218 CHD patients. COVID-19 infection was confirmed by the detection of IgG-SARS against SARS-CoV-2 S1 protein receptor-binding domain. Cohort 1 (24–69 years of age; 44.8 ± 8.6 years; 80.4% men) was examined for systematic coronary risk evaluation by European Society of Cardiology (SCORE) starting from 2019 before the onset of the COVID-19 pandemic. Cohort 2 (63±10.9 years of age, 54% men) was processed starting from 2017 (three years prior to the COVID-19 pandemic) in a hospital setting to undergo coronary angiography to assess coronary lesions as Gensini score. The levels of cGAMP and endothelial markers (nitrate and nitrite combined as NOx and endothelin-1) were assessed in the serum to evaluate the associations with IgG-SARS status, SCORE, and extent of coronary lesions by correlation and receiver operating characteristic (ROC) analyses. Results: Serum cGAMP did not discriminate between SARS-positive and SARS-negative healthy subject of cohort 1. Moreover, the level of cGAMP was not associated with endothelial biomarkers in healthy subjects. However, serum cGAMP was associated with atherosclerosis in cohort 2 in patients with CHD (area under the curve (AUC) 0.69; 95% CI 0.587-0.806; P=0.001), indicating that serum cGAMP was lower in patients with coronary stenosis compared to that in patients without coronary lesions. Conclusions: Low levels of serum cGAMP were associated with atherosclerosis and markers of endothelial dysfunction in patients with CHD. However, cGAMP was not associated with endothelial biomarkers or IgG-SASR immune status in healthy subjects, suggesting that cGAMP may be a new biomarker of coronary lesions in the context of sterile inflammation.
