The European Fish Index EFI+ is the only fish-based multimetric index for the assessment of the ecological status of running waters validated and thus applicable across the European Union. Metrics of the index rely on several attributes of the species present in the fish assemblage, irrespective of their native/alien status. The abundance of alien fish, together with other anthropogenic impacts, is one of the most important threats to the conservation of native fish and ecosystem health and is also an indicator of degraded stream conditions. Therefore, to improve the performance and utility of the EFI+ in regions with high incidence of alien species, the EFI+ was adapted to include the alien fish pressure by averaging the two original metrics with a new metric that reflects the number of alien species as well as the proportional abundance of alien individuals. The application of the adapted index (A-EFI+) is illustrated with data from several Iberian Mediterranean basins and showed similar or stronger correlations than the original EFI+ with anthropogenic pressure (land-use variables) and with other regional fish indices. EFI+ has been invaluable to intercalibrate fish indices across Europe and A-EFI+ is similar but explicitly includes the alien pressure, thus helping to provide a more comprehensive assessment of ecosystem health and to communicate it to society.

: Nanotechnology has expanded what can be achieved in our approach to cancer treatment. The ability to produce and engineer functional nanoparticle formulations to elicit higher incidences of tumor cell radiolysis has resulted in substantial improvements in cancer cell eradication while also permitting multi-modal biomedical functionalities. These radiosensitive nanomaterials utilize material characteristics, such as radio- blocking/absorbing high-Z atomic number elements, to mediate localized effects from therapeutic irradiation. These materials thereby allow for subsequent scattered or emitted radiation to produce direct (e.g., damage to genetic materials) or indirect (e.g., protein oxidation, reactive oxygen species formation) damage to tumor cells. Using nanomaterials that activate under certain physiologic conditions such as the tumor micro-environment can selectively target tumor cells. These characteristics combined with biological interactions that can target the tumor environment allow for localized, radio-sensitization while mitigating damage to healthy cells. This review explores the various nanomaterial formulations utilized in cancer radiosensitivity research. Emphasis on inorganic nanomaterials showcase the specific material characteristics that enable higher incidences of radiation while ensuring localized cancer targeting based on tumor micro-environment activation. The aim of this review is to guide future research in cancer radiosensitization using nanomaterial formulations and to detail common approaches to radiosensization, as well their relations to commonly implemented radiotherapy techniques.

Grazing exclusion (GE) and rest grazing (RG) are important management to restore grassland ecosystems. In order to evaluate the effects and mechanisms of different grazing management on species diversity, vegetation community indices and soil variables were determined in the plots of Qilian Mountains in Gansu Province. The results showed that diversity effects and their regulating mechanisms had space-scale dependence under different grazing management. Species richness and species diversity indices of RG in grassland were significantly higher than that of GE at the regional scale. Additionally, three grazing management in mountain meadows had only a significant effect on species richness, but different management in mountain meadows and temperate steppes had a significantly different effect on species diversity indices. Meanwhile, soil variables only influenced species diversity at the regional scale. Most of community and soil variables at each scale had positive effects on species diversity, except that the direction of biodiversity effects was negative for species coverage, mean plant height, soil porosity (SK) and bulk density (BD) under two contrasting grazing management. In conclusion, choosing RG at the regional scale and select grazing management according to different grassland types at the local scale to restore degraded grassland vegetation.

The mantis shrimp Rissoides pallidus (Giesbrecht, 1910) is among the rarely reported crustacean species in the Central Mediterranean Sea. In December 2020, during a trawl survey off Mazara del Vallo harbour (Strait of Sicily), two specimens of R. pallidus were captured on coastal detritus and coastal terrigenous mud bottoms at about 132 and 152 m depths. This current note, therefore, updates the existing knowledge of R. pallidus in the Strait of Sicily. Additional information specific to biometrics, bottom types, depth preference, habitats, and geographic distribution of this crusta-cean species are provided.

A major driver of declining biodiversity is landcover change leading to loss of habitat. Many studies have estimated large-scale declines in biodiversity, but loss of biodiversity at a local scale due to the immediate effects of development have been poorly studied. California, in particular, is a biodiversity hotspot and has rapidly developed; thus, it is important to understand the effects of development on wildlife. Here, we conducted reconnaissance surveys -- a type of survey often used by consulting biologists in support of environmental review of proposed projects -- to measure changes in relative abundance and richness of vertebrate species in response to urban development. We completed 2 reconnaissance surveys at each of 52 control sites that remained undeveloped at the times of both surveys, and at each of 26 impact sites that had been developed by the time of the second survey. We completed the surveys as part of a before-after, control-impact (BACI) experimental design. Our main interest was on the interaction effect between the before-after phases and control-impact treatment levels, or on the impact of development. We also tested for effects of survey duration, years intervening the surveys in the before and after phases, project area size, latitude, degree of connectedness to adjacent open space, and whether the site was a redevelopment site, infill or not infill. After development, the average number of vertebrate wildlife species we detected declined 48% within the project area, and 66% within the bounds of the project sites. Further, the average number of vertebrate animals we counted declined 90% within the project area, and 89% within the bounds of the project sites. Development impacts measured by the mean number of species detected per survey were greatest for amphibians (-100%), followed by mammals (-86%), grassland birds (-75%), raptors (-53%), special-status species (-49%), all birds as a group (-48%), non-native birds (-44%), and synanthropic birds (-28%). Our results indicated that urban development substantially reduced vertebrate species richness and numerical abundance, even after richness and abundance had likely already been depleted by the cumulative effects of loss, fragmentation, and degradation of habitat in the urbanizing environment. Cumulative effects monitoring is needed, and so are conservation measures to mitigate the effects of urbanization.

In organisms that use reduced sulfur compounds as alternative or additional electron donors to organic compounds, transcriptional regulation of genes for enzymes involved in sulfur oxidation is needed to adjust metabolic flux to environmental conditions. However, little is known about the sensing and response to inorganic sulfur compounds such as thiosulfate in sulfur-oxidizing bacteria. In the Alphaproteobacterium Hyphomicrobium denitrificans, one strategy is the use of the ArsR-SmtB-type transcriptional regulator SoxR. We show that this homodimeric repressor senses sulfane sulfur and that it is crucial for the expression not only of sox genes encoding the com-ponents of a truncated periplasmic thiosulfate-oxidizing enzyme system but also of several other sets of genes for enzymes of sulfur oxidation. DNA binding and transcriptional regulatory activity of SoxR are controlled by polysulfide-dependent cysteine modification. The repressor uses the formation of a sulfur bridge between two conserved cysteines as a trigger to bind and release DNA and can also form a vicinal disulfide bond to orchestrate a response to oxidizing conditions. The importance of the sulfur bridge forming cysteines was confirmed by site directed mutagen-esis, mass spectrometry and gel shift assays. In vivo, SoxR interacts directly or indirectly with a second closely related repressor, sHdrR.

Changes in epitranscriptome with N6‐methyladenine (m6A) modification could be involved in the development of multiple diseases, which might be a prevalent modification of messenger RNAs (mRNAs) in eukaryotes. The m6A modification might be performed through the action of methyltransferases, demethylases, and methylation-binding proteins. Importantly, the m6A methylation may be associated with various neurological disorders including Alzheimer's disease (AD), Parkinson's disease (PD), depression, aging related diseases, and/or aging itself. In addition, the m6A methylation might functionally regulate the eukaryotic transcriptome by influencing splicing, export, subcellular localization, translation, stability, and decay of mRNAs. Neurodegenerative diseases may possess a wide variety of phenotypes depending on the neurons that degenerate at occasions. Interestingly, increasing number of evidences have indicated that m6A modification could modulate the expression of autophagy-related genes and promote autophagy in neuronal cells. Oxidative stresses such as reactive oxygen species (ROS) could stimulate the m6A RNA methylation, which may be also related to the regulation of autophagy and/or the development of neurodegenerative diseases. Both m6A modification and autophagy could also play critical roles in regulating health condition of neurons. Therefore, a comprehensive understanding of the m6A and autophagy relationship in human diseases may benefit in developing therapeutic strategies in the future. This paper reviews advances in the understanding of the regulatory mechanisms of m6A modification in the occurrence and development of neurodegenerative diseases and/or aging, discussing the possible therapeutic procedures related to mechanisms of m6A RNA methylation and autophagy.

Bacterial phylogenetics has largely been determined via 16S rRNA gene sequencing and phylogenetic tree reconstruction. Observed utility of this approach has driven the popularity of the 16S rRNA gene amplicon metagenomics method for profiling and identifying diverse microbes from specific habitats. This work sought to develop universal primers for amplifying the 16S rRNA gene from a consortium of disparate microbial species. Using multiple sequence alignment of the 16S rRNA gene of a variety of microbes, the resulting highly conserved region of the consensus sequence was used for design of universal polymerase chain reaction (PCR) primers for 16S rRNA gene. Application of the universal primers in simulated PCR reveals poor amplification efficiency where only 12 species out of 31 generated an amplicon. BLAST analysis of the resulting amplicons reveals a classification error of 50%. More significantly, analysis of the amplicon length indicates variable read length ranging from 81 to 122 base pair compared to the predicted read length of 100 base pairs. This suggests that the 16S rRNA gene harbours significant hitherto underappreciated sequence diversity, and may have unknown alternative splicing and recombination mechanisms. Overall, results from this study suggests that primer design for 16S rRNA amplicon metagenomics may be application and habitat specific, where it is difficult to design universal primers for all bacterial species. Conceptually, this meant that there may be sequence co-evolution in 16S rRNA gene for microbial species in the habitat where environmental and nutritional conditions impact on 16S rRNA gene structure and sequence. In essence, 16S rRNA gene may habour epigenetics signals at the gene level.

The environment is seldom optimal for plant growth and changes in abiotic and biotic signals, including temperature, water availability, radiation and pests, induce plant responses to optimise survival. The New Zealand native plant species and close relative to Arabidopsis thaliana, Pachycladon cheesemanii grows under environmental conditions that are unsustainable for many plant species. Here we compare the responses of both plant species to different stressors (low temperature, salt and UV-B radiation) to help understand how P. cheesemanii can grow in such harsh environments. The stress transcriptomes were then determined and comparative transcriptome and network analyses discovered similar and unique responses within species between different stresses, and between the two plant species. A number of widely studied plant stress processes were highly conserved in A. thaliana and P. cheesemanii. However, in response to cold stress, Gene Ontology terms related to glycosinolate metabolism were only enriched in P. cheesemanii. Salt stress was associated with alteration of the cuticle and proline biosynthesis in A. thaliana and P. cheesemanii, respectively. Anthocyanin production may be a strategy to cope with UV-B radiation stress in P. cheesemanii only. These results allowed us to construct broad stress response pathways in A. thaliana and P. cheesemanii and identify possible novel plant strategies that help mitigate environmental stress.

Corals are an important habitat for a diverse fauna which is especially rich in branching species. Each colony harbors several species and dozens of individuals forming an integrated symbiotic community. Factors affecting the diversity and abundance of coral symbionts are poorly understood. The present study tested experimentally the relationship between the population density of coral Pocillopora verrucosa and the diversity and abundance of symbionts in planted coral fragments exposed during three months. Colony size and bleaching were also considered as two additional factors. We demonstrated that even at the early stage of the colonies’ formation, the diversity of obligate symbiontson on these corals was higher than in any other studied local faunas of the World Ocean.Species richness and abundance of symbionts were positively related to colony size and density, while no impact of bleaching was found. Species richness and abundance of obligate symbionts increased along with the host population density, while in facultative symbionts, these indexes decreased. We hypothesized that the negative effect of the host density on species richness and abundance of facultative symbionts is caused by the competitive pressure of obligates, which increases with their abundance. Reasons for the different responses of obligate and facultative symbionts to host density were considered.

Maize (Zea mays) is one of the most important staple food and primary source of livestock feed in the world. As the consumption of maize grown on the selenium-enriched soils of Naore Valley is one of the apparent causes of selenosis in the area, this work collected and analyzed total Se, Se fractions, and Se species distributions in maize plant samples, including grains, leaves, stalks, roots, rhizosphere soils, and the most representative parent rock materials from Naore Valley, Ziyang County, China. The Se distribution in soils markedly correlated with the weathered Se-enriched bedrocks, but most of the Se in the analyzed soils is enclosed as recalcitrant residual Se and organic-sulfide bound Se. In contrast, Se in rocks had a comparatively higher bioavailability and is bounded mainly to organic matter and sulfides minerals, with very few of the Se enclosed in the residual fraction. Maize plants might take a large amount of Se from the organic-sulfide bound Se fraction in the Se-rich soils, the weathered products from bedrocks or plant litters. Total Se concentrations in the collected samples were observed in descending order soil>leaf>root>grain>stalk. The predominant Se species detected in maize plants was SeMet. Se inorganic forms, mainly Se(VI), decreased from root to grain and were possibly assimilated into organic forms. Se (IV) was barely present. The natural increases in Se concentration affected mainly leaf and root dry-weight biomass as they are the organs that coped with the highest Se accumulation. This paper offers an insight into the uptake, accumulation, and distribution of Se forms in natural Se-rich maize crops and an opportunity for shifting Se-rich soils from menaces to valuable resources for growing Se-rich agricultural products.

Several extinctions have already been attributed, at least in part, to global warming, as climate change constitutes a serious threat for species living in isolated ecosystems and thus unable to track habitat changes. However, in all these cases extinction was due to human impacts, often directly but generally also through exotic invasive species. For two arboreal land snails in Indian Ocean islands a link has been proposed with decreasing rainfall. The decline (but probably not extinction) of Pachnodus velutinus, a specialist of moist forests on the summits of northwestern Mahé, was most likely caused instead by invasive plants altering its habitat and alien predators decimating the population in the small remaining moist forests. An alternative explanation assuming genetic swamping through hybridization with a species from lower elevations has no basis, as the presumed hybrid constitutes a distinct species able to survive in the altered, dryer habitat. On Aldabra Atoll, the endemic Rhachistia aldabrae was claimed to have been the first extinction due to climate change, but is still extant. No relationship can be detected between number of sightings and annual rainfall, although a weighted measure that takes into account rainfall in previous years suggests a limited impact of weather. Analysis of the sighting record in various ways yields a probability of survival over time that never dropped below 0.3. The decline was caused instead by intense impacts of exotic invasive species. Alternative shortcuts to evaluate extinction rates among poorly known species are shown to be unreliable. Although no contemporary extinction can still be attributed to climate change, indirect and synergistic impacts on biodiversity are expected, especially through promoting biological invasions.

Sexual vs asexual reproduction—unisexual vs bisexual populations—diploid vs polyploid biotypes—genetic vs environmental sex determination: all these natural phenomena are associated with the genus of teleost fish, Carassius. Two Carassius entities with completely different biological characteristics exist: one globally widespread and invasive Carassius gibelio, and the other C. carassius with decreasing trend of natural occurrence. Comprehensive biological and cytogenetic knowledge of both entities, including the physical interactions between them, can help to balance advantages of highly invasive and disadvantages of threatened species. For example, benefits of a wide-ranged colonization can lead toward extinction of native species or be compensated by parasitic enemies and lead to equilibrium. This review emphasizes the comprehensive biology and cytogenetic knowledge and importance of the Carassius genus as one of the most useful experimental vertebrate models for evolutionary biology and genetics. Secondly, the review points out that effective molecular cytogenetics should be used for identification of various species, ploidy levels, and hybrids. The proposed investigation of these hallmark characteristics in Carassius may be applied in conservation efforts to sustain threatened populations in their native ranges. Also, the review focuses on consequences of co-occurrence of native and non-native species and outlines future perspectives of Carassius research.

Huntington’s disease (HD) is a late-onset; progressive, dominantly inherited neurological disorder marked by an abnormal expansion of polyglutamine (poly Q) repeats in Huntingtin (HTT) protein. The pathological effects of mutant Huntingtin (mHTT) are not restricted to the nervous system but systemic abnormalities including immune dysregulation have been evidenced in clinical and experimental settings of HD. Indeed, mutant huntingtin (mHTT) is ubiquitously expressed and could induce cellular toxicity by directly acting on immune cells. However, it is still unclear if selective expression ofmHTT exon1 in neurons could induce immune responses and hemocyte function. In the present study, we intended to monitor perturbations in the hemocytes population and their physiological functions in Drosophila, caused by pan-neuronal expression of mHTT protein. We found that pan-neuronal expression of mHtt significantly alters crystal cells and plasmatocyte count in larvae and adults with disease progression. Interestingly, plasmatocytes isolated from diseased conditions exhibit a gradual decline in phagocytic activity ex vivo at progressive stages of the disease as compared to age-matched control groups. We also observed an increased production of reactive oxygen species (ROS) in plasmatocytes at advanced stages of the disease. In addition, diseased flies displayed elevated reactive oxygen species (ROS) in circulating plasmatocytes at the larval stage and in sessile plasmatocytes of hematopoietic pockets at of disease. All the parameters were monitored progressively, targeting the circulation at larvae stage and hematopoietic pockets in adults at different disease stages, and many alterations were documented in the early stage itself. These findings strongly implicate that neuronal expression of mHtt alone is sufficient to induce non-cell-autonomous immune dysregulation in vivo. Based on these findings, we propose that further insight into the mechanisms through which neuronal expression of mHtt might be inflicting the innate immune responses would facilitate therapeutic inventions aimed at amelioration of HD pathology and improving the quality of life of the patients.

In this paper, impulsive control on a single-species Markovian jumping ecosystem leads to a stability criterion, and the newly-obtained theorems improve the related existing results. Numerical examples illuminate the effectiveness.

Nowadays, the human population is facing the third and may be the worst pandemic caused by human coronaviruses (CoVs). The virus was first reported in Wuhan, China on 31 December 2019 and spread within short time to almost all countries of the world. Genome analysis of the early virus isolates has revealed high similarity with SARS-CoV and hence the new virus was officially named SARS-CoV-2. Since CoVs have the largest genome among all RNA viruses, they can adapt many point mutation and recombination events; particularly in spike gene, that enable these viruses to rapidly change and evolve in nature. CoVs are known to cross the species boundaries by using different cellular receptors. SARS-CoV-2 is believed to originate in bats and transmitted to human being through an ill-defined intermediate host. In the current review, different aspects of SARS-CoV-2 biology and pathogenicity are discussed including virus genetics and evolution, spike protein and its role in evolution and adaptation to novel hosts, and virus transmission and persistence in nature. In addition, the immune response developed during SARS-CoV-2 infection is demonstrated with special reference to the interplay between immune cells and their role in disease progression. We believe that SARS-CoV-2 outbreak will not be the last and spillover of CoVs from bats will continue. Therefore, establishing intervention approaches to reduce the likelihood of future CoVs spillover from the natural reservoirs is a priority.

This study aimed to obtain and characterize extracted hemp oil enriched in cannabidiol (CBD) by decarboxylation of cannabidiolic acid (CBDA) and to give new insights into its antioxidant and anticancer effects. Optimization of CBDA decarboxylation in hemp oil was performed, and CBD and CBDA contents and purities were determined by “flash chromatography”, 1H- and 13C-NMR. The antioxidant properties of CBD-enriched oil were investigated by Fe2+ chelating activity, Fe3+ reducing antioxidant power assay, O2− scavenging activity, HO− scavenging ability and lipid peroxidation inhibitory assay and its cytotoxicity, apoptosis- and oxidative stress-inducing effects on NHDF, MeWo, HeLa, HepG2 and HOS cells were determined. The CBD concentration in hemp oil was increased by CBDA soft decarboxylation optimized at 90°C, for 1 h and resulted oil was capable of reducing iron, scavenging free radicals, and inhibiting lipid peroxidation in cell-free oxidative conditions. CBD-enriched oil promoted NHDF proliferation at up to 15 µg CBD/mL, while inducing apoptosis and ROS production and modulating antioxidant enzymes’ gene expression in cancer cells, being selective for osteosarcoma cells and induced apoptosis by p53- and ROS-independent mechanisms. CBD-enriched hemp oil demonstrated antioxidant properties in oxidative conditions and promoted normal fibroblasts’ proliferation, while inducing apoptosis and ROS production in cancer cells.

Salvia ceratophylloides (Ard.) is an endemic, rare, threatened plant species recently rediscovered in very few individuals in two different sites of South Italy. The study of within-plant variation more than among-plant one is fundamental to understand the plant adaptation to the local conditions, especially in rare species, and consequently to preserve plant biodiversity. Here, we reported the variation of the morpho-ecophysiological and metabolic traits between the sessile and petiolate leaf of S. ceratophylloides plants in two different sites for understanding the adaptation strategies for surviving in these habitats. The S. ceratophylloides individuals exhibited different net photosynthetic rate, maximum quantum yield, light intensity for the saturation of the photosynthetic machinery, stomatal conductance, transpiration rate, leaf area, fractal dimension and some VOCs between the different leaf types. This within-plant morpho-physiological and metabolic variation was depended on the site. These results provide empirical evidence of sharply within-plant variation of the morpho-physiological traits and VOCs profiles in S. ceratophylloides which could be because of adaptation to the local conditions.

The mangrove plant Acanthus ilicifolius and its relative, A. mollis, have been previously proved to possess diverse pharmacological effects. Therefore, evaluating the differentially expressed proteins of these species under tidal flooding stress is essential to fully exploit and benefit from their medicinal values. The roots of A. ilicifolius and A. mollis were exposed to 6 h of flooding stress per day for 10 days. The dry weight, hydrogen peroxide (H2O2) content, anatomical characteristics, carbon and energy levels, and two-dimensional electrophoresis coupled with MALDI-TOF/TOF MS technology were used to reveal the divergent flooding resistant strategies. A. ilicifolius performed better under tidal flooding stress, which was reflected in the integrity of the morphological structure, more efficient use of carbon and energy, and a higher percentage of up-regulated proteins associated with carbon and energy metabolism. A. mollis could not survive in flooding conditions for a long time, as revealed by incomplete cell structures of the roots, less efficient use of carbon and energy, and a higher percentage of down-regulated proteins associated with carbon and energy metabolism. Energy provision and flux balance played a role in the flooding tolerance of A. ilicifolius and A. mollis.

Illumina and nanopore sequencing technologies are powerful tools that can be used to determine the bacterial composition of complex microbial communities. In this study, we compared nasal microbiota results at genus level using both Illumina and nanopore 16S rRNA gene sequencing. We also monitored the progression of nanopore sequencing in the accurate identification of species, using pure, single species cultures, and evaluated the performance of the nanopore EPI2ME 16S data analysis pipeline. Fifty-nine nasal swabs were sequenced using Illumina MiSeq and Oxford Nanopore 16S rRNA gene sequencing technologies. In addition, five pure cultures of relevant bacterial species were sequenced with the nanopore sequencing technology. The Illumina MiSeq sequence data were processed using bioinformatics modules present in the Mothur software package. Albacore and Guppy base calling, a workflow in nanopore EPI2ME and an in house developed bioinformatics script were used to analyze the nanopore data. At genus level, similar bacterial diversity profiles were found, and five main and established genera were identified by both platforms. However, probably due to mismatching of the nanopore sequence primers, the nanopore sequencing platform identified Corynebacterium in much lower abundance compared to Illumina sequencing. Further, when using default settings in the EPI2ME workflow, almost all sequence reads that seem to belong to the bacterial genus Dolosigranulum and a considerable part to the genus Haemophilus were only identified at family level. Nanopore sequencing of single species cultures demonstrated at least 88% accurate identification of the species at genus and species level for 4/5 strains tested, including improvements in accurate sequence read identification when the basecaller Guppy and Albacore, and when flowcell versions R9.4 and R9.2 were compared.

Oxidative stress is known to be critically implicated in the pathophysiology of several neurological disorders, including Alzheimer’s disease and ischemic stroke. The remarkable neurotrophic activity of Gelidium amansii, has been reported consistently in a series of our previous studies, inspired us to investigate whether this popular agarophyte could protect against hypoxia/reoxygenation (H/R)-induced oxidative injury in hippocampal neurons. The primary culture of hippocampal neurons challenged with H/R suffered from a significant loss of cell survival, accompanied by apoptosis and necrosis, DNA damage, generation of reactive oxygen species (ROS), and dissipation of mitochondrial membrane potential (MMP) which were successfully attenuated when the neuronal cultures were preconditioned with GAE, an optimized ethanolic extract of G. amansii. Moreover, the expression of N-methyl-D-acetate receptor subunit 2B (GluN2B), an extrasynaptic glutamate receptor, was significantly repressed in GAE-treated neurons as compared to those without GAE intervention. Together, this study demonstrates that GAE attenuated H/R-induced oxidative injury in hippocampal neurons through, at least in part, a potential neuroprotective mechanism that involves inhibition of GluN2B-mediated excitotoxicity and suppression of ROS production, and suggest that this edible seaweed could be a potential source of bioactive metabolites having therapeutic significance against oxidative stress-related neurodegeneration, including ischemic stroke and neurodegenerative diseases.

Ancient lakes are among the most interesting models for evolution studies, because their biodiversity is the result of a complex combination of migration and speciation. Here, we investigate the origin of single celled planktonic eukaryotes from the oldest lake in the world – Lake Baikal. By using 18S rDNA metabarcoding we recovered 1,427 Operational Taxonomic Units (OTUs) belonging to protists populating surface waters (1-50 meters) and belonging to pico- and nano-plankton size fractions. The recovered communities resembled other lacustrine freshwater assemblages found elsewhere, especially the unclassified protists. However, our results suggest that a fraction of Baikal protists originated evolutionary recently from marine/brackish ancestors. Moreover, our results suggest that rapid radiation may have occurred among some protist taxa, partially mirroring what was already shown for multicellular organisms in Lake Baikal. We found potential species flocks in Stramenopiles, Alveolata, Opisthokonta, Archaeplastida, Rhizaria and Hacrobia. Putative flocks predominated in Chrysophytes, which are highly diverse in Lake Baikal. Some of the species, especially those from these flocks, may be endemic from Lake Baikal, because their 18S rDNA differed > 10% from known DNA. Overall, our study points to novel diversity of planktonic protists in Lake Baikal, some of which may have emerged in situ after evolutionary diversification.

The study was conducted in north wollo, south wollo and orimiya zone, in Eastern Amhara with an aim to assess the resource potential of Acacia seyal in the selected sites. The data from the selected sites were collected using point center quarter method. A total of 90 quadrates from nine study areas with plot size 50m*50m, (22.5ha) were systematically located along each transect, 100 m apart, and was spatially captured with the aid of GPS. At every sampling point, four quadrants (90 degrees) were created, using the transect line and a line perpendicular to it. Species composition Acacia seyal structure and its regeneration status, at points along transects were taken to analyze diversity and target species structure of the sites. The highest and least density of Acacia seyal ha^-1 were attained by Mehale mecharie (148) and Alene sefer (52). The highest shannon weiner diversity and species richness was observed in Alene sefer(Kemessie). Acacia seyal structure in all study sites showed an inverted J shape except lastie gerdao (Gubalafeto). Therefore, Acacia seyal deserves immediate conservation and appropriate management measures in order to get sustainable product and services from the species. Based on the results, awareness creation on the values and management of Acacia seyal, study on the management options of Acacia seyal for firewood, fuel wood, charcoal production and also investigation of gum production techniques of Acacia seyal for the sustainable use of the resource are recommended.

Water hyacinth (Eichhornia crassipes) is an invasive species that has modified ecosystem functioning in the Sacramento-San Joaquin Delta (Delta), California, USA. Studies in lakes and rivers have shown that water hyacinth alters water quality. In tidal systems, such as the Delta, water moves back and forth through the water hyacinth patch so water quality directly outside the patch in either direction is likely to be impacted. In this study, we asked whether the presence or treatment of water hyacinth with herbicides resulted in changes in water quality in this tidal system. We combined existing datasets that were originally collected for permit compliance and long-term regional monitoring into a dataset that we analyzed with a before-after control-impact (BACI) framework. This approach allowed us to describe effects of presence and treatment of water hyacinth, while accounting for seasonal patterns in water quality. We found that although effects of treatment were not detectable when compared with water immediately upstream, dissolved oxygen and turbidity became more similar to regional water quality averages after treatment. Temperature became less similar to the regional average after treatment, but the magnitude of the change was small. Taken together, these results suggest that tidal hydrology exports the effects of water hyacinth upstream, just as river flow is known to transport the effects downstream, creating a buffer of altered water chemistry around patches. It also suggests that although water hyacinth has an effect on dissolved oxygen and turbidity, these parameters recover to regional averages after treatment.

Lineage diversity can refer to the number of genetic lineages within species or to the number of deeper evolutionary lineages, such as genera or families, within a community. Community lineage diversity (CLD) is of interest to ecologists, evolutionary biologists, biogeographers, and those setting conservation priorities. Despite its relevance, it is not clear how to best quantify CLD. With North American tree communities as an example, we test which taxonomic and phylogenetic metrics best measure CLD. We find that phylogenetic metrics outperform taxonomic metrics. Faith’s phylogenetic diversity performs well, but is skewed towards the number of lineages in recent time. The best metric is newly derived here, and termed time integrated lineage diversity (TILD). Mapping the lineage diversity of tree communities across the contiguous United States, we find a spatial pattern differing from that of species richness in key areas. The Pacific Northwest, Great Lakes Region, state of Maine, and south-eastern piedmont and coastal plain forests all emerge as areas high in lineage diversity, but relatively lower in species richness. We urge the consideration of lineage diversity, as well as species richness, when setting conservation priorities.

There have been a great number of investigations about the influence of weak magnetic fields on biological systems, such as isolated cells and whole organisms. This is also a subject of considerable medical concern since old epidemiologic observations have indicated a possible tumorigenic effect of these fields. Their mechanism of action, however, is not firmly established. A large number of biological effects of electromagnetic fields have been attributed either to the production of reactive oxygen species (ROS) or to the entrance Ca²⁺ in the cell. A new biochemical pathway is proposed that covers these two possibilities: the primary effect of the magnetic field would be by the mechanism of radical pairs resulting in the production of ROS; these could activate the ion channels TRPM2 producing cellular inflow of Ca²⁺, which would induce the calcium dependent effects. Thus, a large number of biological effects observed up to the present could be explained.

As a universal pathogen leading to neonatal defects and transplant failure, Human cytomegalovirus (HCMV) has strict species specificity that the inability to using this virus in animals has hampered its pathogenesis study. However, the mechanism of cross-species barrier remains elusive that no non-human cell model has been established to fill this knowledge gap. We observed that primary dermis fibroblasts (TSDF) isolated from the Chinese tree shrew (Tupaia belangeri chinensis), a small laboratory animal with close affinity to primates, were permissive to HCMV replication. In TSDF infected with GFP-expressing HCMV, the green fluorescence and cytopathic effect were observed and the expression of 3 kinetic genes and replication of viral genome were detected. The cell-free viruses produced in TSDF reached 103 pfu/mL at 96 hpi, which were 10-fold lower than in primary human foreskin fibroblasts. Our results demonstrated that TSDF supported low level of lytic replication of HCMV. The TSDF model provides a useful platform for the mechanism study of species barrier of HCMV.

The scene for regional biogeography and human settlements in Central Amazonia is set by the river network, which presumably consolidated in the Pliocene. However, we present geomorphological and sediment chronological data showing that the river network has been anything but stable. Even during the last 50 kyr, the tributary relationships have repeatedly changed for four major rivers, together corresponding to one third of the discharge of the Amazon. The latest major river capture event converted the Japurá from a tributary of the Rio Negro to a tributary of the Amazon only 1000 years ago. Such broad-scale lability implies that rivers cannot have been as efficient biogeographical dispersal barriers as has generally been assumed, but that their effects on human societies can have been even more profound. Climate change and deforestation scenarios predict increasing water levels during peak floods, which will likely increase the risk of future river avulsions. This may have disastrous consequences for the local human societies, especially in those areas where the current floodplains are at only marginally lower elevations than the nearest water divide. We suggest that the prevailing paradigm of rivers as principal structuring elements of Amazonian biogeography needs to be re-evaluated, and that land use planning and civil risk assessment should take the possibility of river avulsions into account.

Background: The official standard for quality control of honey is currently based on physicochemical properties. However, this method is time-consuming, cost intensive, and do not lead to information on the originality of honey. This study aims to classify raw stingless bee honeys by bee species origins as a potential classifier using NMR-LCMS-based metabolomics approach. Methods: Raw stingless bee honeys were analysed and classified by bee species origins using proton nuclear magnetic resonance (1H NMR) spectroscopy and an ultra-high performance liquid chromatography-quadrupole time of flight mass spectrometry (UHPLC-QTOF MS) in combination with chemometrics tools. Results: The honey samples were able to be classified into three different groups based on the bee species origins of Heterotrigona itama, Geniotrigona thoracica, and Tetrigona apicalis. D-Fructofuranose (H. itama honey), β-D-Glucose, D-Xylose, α-D-Glucose (G. thoracica honey), and L-Lactic acid, Acetic acid, L-Alanine (T. apicalis honey) identified via 1H NMR data and the diagnostic ions of UHPLC-QTOF MS were characterized as the discriminant metabolites or putative chemical markers. Conclusion: It could be suggested that the quality of honey in terms of originality and purity can be rapidly determined using classification technique by bee species origins via 1H NMR- and UHPLC-QTOF MS-based metabolomics approach.

Stingless bee beekeeping provides new opportunities to improve the incomes of many households in Malaysia through the sale of honey and other bee products. While Heterotrigona itama is one of the most commonly cultured species of stingless bees, its behavior is not very well understood. Hence, we conducted this study to investigate the behavior of H. itama in exploiting food sources by ascertaining the nectar sugar concentration preferred by the bee. We also aimed to determine the preferred distance of food source from the bee hive. Our results suggest that H. itama prefers high sugar concentrations of 35% and above, and they would fly up to 7 m from the hive to collect food. We discuss how nectar concentration and food distance influence the number of bees exploiting food sources and the overall foraging pattern of H. itama.

Selenocyanates form an interesting class of organic selenium compounds as they serve as multifunctional agents (being the precursors of seleninic acids and diselenides in synthetic chemistry and as antimicrobial and cytotoxic agent in biology) and, due to their similarity with better known thiocyanates promise high biological activity. Yet whilst selenocyanates are common in synthetic chemistry, they are rarely considered in pharmaceutical design. Arylmethyl selenocyanates (1-13) have been synthesized and an insight into their structural properties using X-ray crystallography has been obtained. The compounds subsequently have been evaluated for their potential antimicrobial, nematicidal and cytotoxic activity. ADMET properties in vitro, using mutagenicity (AMES) and permeability (PAMPA) tests, have been determined. The compounds exhibit pronounced activity against various strains of bacteria (both Gram-positive and Gram-negative) and yeasts. Among them, benzylselenocyanate (1) represents the most active anti-ESKAPE agent, with potent antibacterial activity, especially against multidrug resistant MRSA strains (HEMSA 5). Our results demonstrate that the arylmethyl selenocyantes are not only non-mutagenic but also possess moderate cytotoxic activity against cancer cells.

This work updates the characterization of winter fish communities in salt marsh areas of Guadiana estuary (SE-Portugal) and discusses the potential risks of habitat dominance by a non-indigenous species (NIS). To this effect, six field campaigns were carried out during winter season targeting the collection of fish species. Individuals from seven different families (marine and estuarine) were collected although the community was dominated by two estuarine species – the native Pomatoschistus sp. (goby) and the NIS Fundulus heteroclitus (mummichog). Goby controlled the majority of salt marsh habitats, except one specific, marsh pool, where extreme environmental conditions were registered, namely high temperature and salinity. Such conditions may have boosted the intrusion of mummichog in this area. This species is well adapted to a wide range of abiotic factors enabling them to colonize habitats where no predators inhabit. Impacts of mummichog intrusion in the Guadiana salt marsh area are still unpredictable since this is the first recorded in such high density. Nevertheless, in scenarios of increased anthropogenic pressure and, consequently, habitat degradation, there is a potential risk of mummichog spread to other habitats and therefore compete for space and food resources with native species.

We evaluated the mono- and combination-therapy effects of valproic acid (VPA) and doxorubicin (DOX) in hepatocellular carcinoma (HCC) and identified a specific and efficient, synergistic anti-proliferative effect of the VPA and DOX combination in HCC cells, especially HepG2 cells; this effect was not apparent in MIHA cells, a normal hepatocyte cell line. The calculation of the coefficient of drug interaction confirmed the significant synergistic effect of the combination treatment. Concurrently, the synergistic apoptotic cell death caused by the VPA and DOX combination treatment was confirmed by Hoechst nuclear staining and western blot analysis of caspase-3 and poly (ADP-ribose) polymerase (PARP) activation. Co-treatment with VPA and DOX enhanced reactive oxygen species (ROS) generation and autophagy, which were clearly attenuated by ROS and autophagy inhibitors, respectively. Furthermore, as an indication of the mechanism underlying the synergistic effect, we observed that DOX internalization, which was induced in the VPA and DOX combination-treated group, occurred via by the caveolae-mediated endocytosis pathway. Taken together, our study uncovered the potential effect of the VPA and DOX combination treatment with regard to cell death, including induction of cellular ROS, autophagy, and the caveolae-mediated endocytosis pathway. Therefore, these results present novel implications in drug delivery research for the treatment of HCC.

This study investigates the urban ecology of the Red-Whiskered Bulbul (Pycnonotus jocosus) in New Barrackpore, West Bengal, India. We examined the feeding behavior, breeding success, and behavioral patterns of the species in urban habitats. Our results show that the Red-Whiskered Bulbul is adapting to urban environments, with a moderate breeding success rate (64%) and specific behavioral patterns. The species exhibits a significant preference for urban habitats, particularly mango and bamboo trees, and a significant increase in singing activity during the breeding season. Our findings highlight the importance of urban ecology in understanding the behavior and conservation of this species. We suggest that urban green spaces with native tree species and sustainable urban planning practices can support the conservation of the Red-Whiskered Bulbul and other urban-adapted species. This study contributes to our understanding of urban wildlife ecology and conservation in India and has implications for urban planning and biodiversity conservation globally.

Urbanization degrades natural habitats and creates new urban ecosystems, like domestic gardens. The plant composition of these gardens varies with socio-economic factors and urban planning levels. However, the diversity and impact of introduced species are often poorly assessed, causing potential ecological imbalances, particularly in Lubumbashi (DR Congo). The objective was to analyze the spatial structure, plant diversity, propagation strategies, and ecological functions of domestic gardens. Three distinct neighborhoods were selected: planned, unplanned and residential neighborhood. 20 avenues (with 5 plots per avenue) were chosen to represent the diversity within each neighborhood, and stratified random sampling of plots was conducted to analyze gardening practices. Gardens were classified into types, and their vegetation was evaluated based on species origin and ecological impact. The analysis of domestic gardens in Lubumbashi reveals significant variations across different neighborhood types. Residential neighborhoods exhibit larger average garden sizes (315.1 m²), higher species richness (22 species), and larger plot sizes (1032 m²) compared to unplanned and planned neighborhoods, where garden areas and species richness are notably lower. Rectangular gardens dominate in unplanned areas, while planned neighborhoods feature more intentional landscaping elements, such as flowerbeds and hedges. The use of gardens for food production is prominent in planned areas (40.7%), whereas residential neighborhoods prioritize ornamentation (51.4%). The study identifies 232 species across 68 families, with a predominance of exotic species (80%) in all neighborhoods, particularly in unplanned areas (82.25%). The data reveals that Mangifera indica and Persea americana are abundant in all neighborhoods, illustrating their adaptability to different urban contexts. Herbaceous species are most common, followed by woody plants, with vines being sparse. Species dispersal is primarily driven by human activities (anthropochory), accounting for over 85% in all neighborhoods. These findings highlight the strong human influence on the composition and structure of domestic gardens in Lubumbashi, emphasizing the dominance of exotic species and the importance of anthropogenic factors in shaping urban green spaces. Urban policies should incorporate strategies to minimize the negative impacts of exotic species on native flora.

The backwater lowland habitats of large rivers, like the Mississippi River in North America, present complex often inaccessible environments for traditional capture-based fish biodiversity sampling. Our knowledge of the assemblages of fishes that occupy such habitats are often incomplete, and this can compromise management efforts. We employed environmental DNA (eDNA) metabarcoding methods to sample a Mississippi River bottom wetland system to assess ichthyofaunal diversity, and assemblage structure across habitat types, and we compared our results to capture-based survey records for the same habitats. We collected water samples in spring and fall of 2022 from slough, ditch, shallow lake and bayou habitats that varied in depth, vegetation, seasonal variability, and connectivity to the Mississippi River channel. We detected a diverse array of fish species that included 51 taxa. Nearly all species previously documented in the habitats were detected by eDNA metabarcoding, and we increased the number of documented species by more than a third. Most of the species were ubiquitous across the range of habitats, but there was also substantial assemblage structure, with some species exhibiting clear habitat specificity. Fall sampling was limited to the deeper bayou habitats where seasonal variation between spring and fall was minimal. eDNA metabarcode sampling was demonstrated to be effective at detecting invasive species as well as uncommon species, which included several species of conservation concern.

Soil organic matter (SOM) is essential for nutrient cycling and soil carbon (C) accumu-lation, both of which are heavily influenced by the quality and quantity of plant litter. Since SOM dynamics in relation to plant diversity are poorly understood we investigated the effects of willow variety and mixture, and site on the soil C stocks, SOM chemical composition and ther-mal stability. Using pyrolysis-field ionization mass spectrometry we analysed the top 10 cm of soil from two 7-year-old experimental sites in Germany and Sweden, with monocultures and mixtures of two willow varieties (Salix spp.) belonging to different species. Overall, site had the strongest effect on SOM quality. Results showed significant variability across sites for willow identity and mixture effects on C accumulation and SOM chemistry. In the German site (Ros-tock), yearly soil C accumulation was higher (p < 0.05) for variety ‘Loden’ (1.0 Mg C ha-1 year-1) compared to ‘Tora’ (0.5 Mg C ha-1 year-1), whilst in the Swedish site (Uppsala), both varieties ex-hibited similar soil C accumulation rates of around 0.6 Mg C ha-1 year-1. Willow variety identity significantly affected SOM quality at both sites, while mixing had minor effects. Our findings emphasize the significance of site-specific context and variety or species identity in shaping soil C accumulation in willow plantations.

Studies on animal virome have mainly concentrated on chordates and medically significant invertebrates, often overlooking sylvatic mosquitoes, constituting the major part of mosquito species diversity. Despite their potential role in arbovirus transmission, the viromes of sylvatic mosquitoes remain largely unexplored. These mosquitoes may also harbor insect-specific viruses (ISVs), affecting arboviral transmission dynamics. The Cerrado biome, known for rapid deforestation and its status as a biodiversity hotspot, offers an ideal setting for investigating mosquito viromes due to potential zoonotic spillover risks from land use changes. This study aimed to characterize the viromes of sylvatic mosquitoes collected from various locations within Minas Gerais state, Brazil. Total RNA was extracted from mosquito pools of Psorophora albipes, Sabethes albiprivus, Sa. chloropterus, Psorophora ferox, and Coquillettidia venezuelensis species, followed by high-throughput sequencing (HTS). Bioinformatic analysis included quality control, contig assembly, and viral detection. Sequencing data analysis revealed 11 near-complete viral genomes (new viruses are indicated with asterisks) across seven viral families and one unassigned genus. These included: Xinmoviridae (Ferox mosquito mononega-like virus* and Albipes mosquito Gordis-like virus*), Phasmaviridae (Sabethes albiprivus phasmavirus*), Lispiviridae (Pedras lispivirus isolate MG), Iflaviridae (Sabethes albiprivus iflavivirus*), Virgaviridae (Buriti virga-like virus isolate MG and Sabethes albiprivus virgavirus 1*), Flaviviridae (Psorophora ferox flavivirus*), Mesoniviridae (Alphamesonivirus cavallyense isolate MG), and the genus Negevirus (Biggie virus isolate MG virus and Coquillettidia venezuelensis negevirus*). Phylogenetic analyses revealed evolutionary relationships with varying degrees of similarity to known sequences in distinct host species. In some cases, new viruses and potential new genera were discovered, emphasizing the importance of studying sylvatic mosquito virome to understand viral diversity and evolution in ecologically sensitive regions. Moreover, the presence of ISVs and potential novel arboviruses underscores the need for ongoing surveillance and control strategies to mitigate the risk of emerging infectious diseases.

Dams fragment stream habitat and fishways around dams typically serve few species that are strong swimmers or jumpers. We test a prototype wetted ramp designed to allow upstream passage of small-bodied fishes while blocking upstream movement of invasive sea lamprey in the Laurentian Great Lakes. We tested short, smooth ramps with 5-10 mm water depth in various combinations of ramp angle, water flow and swim channel width with the aim to selectively block adult migrating sea lamprey while passing sub-adult white suckers (Catostomus commersonii) and creek chubs (Semotilus atromaculatus). Sea lamprey easily passed a 0.75 ramp at 5 angle, but very few individuals passed a similar ramp at 10 angle and none passed a longer ramp at the 5 angle. Limiting the amplitude of tailbeats in a narrow channel did not hamper lamprey or the other species. Greater water flow, and thereby greater immersion depth on the ramp fostered passage for all species. Smaller-bodied individual of creek chubs and white suckers performed best on the ramp. We show that wetted ramps could be incorporated into fishways at low-head dams to aid passage of smaller-bodied fishes while also blocking the spawning migration of adult sea lamprey.

The retinal pigment epithelium (RPE), which ensures the normal functioning of the neural retina, is a pigmented single-cell layer that separates the retina from the Bruch's membrane and the choroid. There are three main types of pigment granules in the RPE cells of the human eye: lipofuscin granules (LG) containing the fluorescent “age pigment” lipofuscin, melanoprotein granules (melanosomes, melanolysosomes) containing the screening pigment melanin and complex melanolipofuscin granules (MLG) containing simultaneously both types of pigments - melanin and lipofuscin. This review examines the functional role of pigment granules in the aging process and in the development of oxidative stress and associated pathologies in RPE cells. The focus is on the process of light-induced oxidative degradation of pigment granules caused by reactive oxygen species. The reasons leading to increased oxidative stress in RPE cells as a result of oxidative degradation of pigment granules are considered. A mechanism has been proposed to explain the phenomenon of age-related decline in melanin content in RPE cells. The essence of the mechanism is that when the lipofuscin part of the melanolipofuscin granule is exposed to light, reactive oxygen species are formed, which destroy its melanin part. As more melanolipofuscin granules are formed with age and the development of degenerative diseases, the melanin in pigmented epithelial cells ultimately disappears.

The crayfish plague is a devastating disease of European freshwater crayfish and is caused by the oomycete Aphanomyces astaci (Ap. astaci), which was introduced to Europe around 1860. All European species of freshwater crayfish are susceptible to the disease, including the white-clawed crayfish Austropotamobius pallipes. The aquatic mould is primarily spread by North American crayfish species and can also disperse rapidly through contaminated wet gear moved between water bodies. This spread, coupled with competition from non-indigenous crayfish, has drastically reduced and fragmented native crayfish populations across Europe. Remarkably, the island of Ireland remained free from the crayfish plague pathogen for over 100 years, providing a refuge for A. pallipes. However, this changed in 1987 when a mass mortality event was linked to the pathogen marking the pathogen's introduction to the region. Fortunately, the plague did not establish and was not detected again in Ireland until 2015 when a molecular analysis linked a mass mortality event in the Erne catchment to Ap. astaci. Since then, the pathogen has appeared across the island. Between 2015 and 2023, Ap. astaci was detected in 18 Irish water catchments, revealing multiple genotypes. Intriguingly, the pathogen in Ireland exists without its natural host species. The uneven distribution of various genetic lineages strongly suggests human-mediated transport of zoospores via contaminated water equipment as a primary cause of spread. This review details the timeline of these events, Ap. astaci’s introductions into Ireland, its rapid spread, the genotypes documented, the issue of non-indigenous crayfish species in Ireland and management efforts.

Resistance evolution during exposure to non-lethal levels of antibiotics is influenced by various stress responses of bacteria which are known to affect growth rate. Here, we aim to disentangle how the interplay between resistance development and associated fitness costs is affected by stress responses. We performed de novo resistance evolution of wild-type strains and single-gene knockout strains in stress response pathways using four different antibiotics. Throughout resistance development, the increase in minimum inhibitory concentration (MIC) is accompanied by a gradual decrease in growth rate, most pronounced with amoxicillin or kanamycin. By measuring biomass yield on glucose and whole-genome sequences at intermediate and final timepoints, we identified two patterns of how the stress responses affect the correlation between MIC and growth rate. First, single-gene knockout E. coli strains associated with reactive oxygen species (ROS) acquire resistance faster and mutations related to antibiotic permeability and pumping out occur earlier. This increases the metabolic burden of resistant bacteria. Second, the ΔrelA knockout strain which has reduced (p)ppGpp synthesis, is restricted in its stringent response, leading to diminished growth rates. The ROS-related mutagenesis and the stringent response are vulnerabilities within the fitness of resistant strains that can possibly be targeted to prevent development of resistance.

β3-Adrenoceptors mediate several functions in rodents that could be beneficial for the treatment of obesity and type 2 diabetes. This includes promotion of insulin release from the pancreas, of cellular glucose uptake, of lipolysis, and of thermogenesis in brown adipose tissue. In combination, they lead to a reduction of body weight in several rodent models including ob/ob mice and Zucker diabetic fatty rats. These findings stimulated drug development programs in various pharmaceutical companies, and at least nine β3-adrenoceptor agonists have been tested in clinical trials. However, all of these projects were discontinued due to lack of clinically relevant changes of body weight. Following a concise historical account of discoveries leading to such drug development programs we discuss species differences that explain why β3-adrenoceptors are not a meaningful drug target for the treatment of obesity and type 2 diabetes in humans.

Tuna species are an important resource to the people of the Pacific, particularly small island countries like Tonga, that have few alternative natural resources. These countries are dependent on this fishery for their nutrition, recreation, government revenue, employment, welfare and culture. Threatening these benefits, however, are global warming and climate uncertainties that affect the presence and distribution of tuna in the Pacific islands countries and territories. There-fore, one of the most important development goals for Tonga involves managing its tuna fish-eries in order to optimize these benefits within the context of climatic impacts. This paper has two main goals. Firstly, we discuss Tonga’s commitment to implement the existing tuna man-agement policies. This commitment permeates through a range of activities to monitoring catch regulations and regulate locally based foreign vessels to fish in the Economic Exclusive Zone of Tonga. This reflects the implementation of an information-based management framework namely, the Tonga National Tuna Fishery Management and Development Plan that is revised every five years. Secondly, the paper identifies key scientific research programs, which form the basis to design more informed future policies towards tuna management. These researches include : i) examining of the bathymetric features of the island nation’s fishing ground, ii) model-ling the spatio-temporal distribution of tuna and, iii) characterizing tuna habitats within an oceanic environment using biophysical oceanographic data within Geographic Information Sys-tem, and iv) impacts of climate change on tuna

Alzheimer's disease (AD) is a complex disease involving multiple factors such as amyloid plaque formation, mitochondrial dysfunction, telomere length, and oxidative stress. As the disease began 20 years before the onset of clinical symptoms, treatment of AD after its onset is likely to be too late to have a significant effect. Therefore, prevention from an early stage is important. Twendee X®︎ (TwX, Oxicut®︎) is an antioxidant supplement consisting of eight ingredients. TwX has been proven to prevent dementia in patients with mild cognitive impairment (MCI) in a multicenter, randomized, double-blind, placebo-controlled, prospective intervention trial. To elucidate the mechanisms of TwX on brain and cellular function, TwX was administered in in vitro experiments and in a C57BL/6 old wild-type, and spontaneous hyperglycemia model with accelerated cognitive decline, and its effects on diabetes, brain autophagy, neurogenesis, and telomere length were evaluated in comparison with those of the non-treated groups. TwX protected mitochondria from oxidative stress with higher antioxidant capacity than a single antioxidant. TwX not only lowered blood glucose levels, but also suppressed reduction of brain neurogenesis and loss of autophagy. Telomeres in aged mice were significantly longer than those in non-TwX-treated mice. Development of dementia is based on many factors of brain function. TwX was suggested to prevent dementia not only by its antioxidant effects, but also by its effects on diabetes, brain neurogenesis, telomere deficiency, and energy production.

The Puuc Biocultural State Reserve (PBSR is an unique model for tropical dry forest conservation in Mexico. Preserving forest biodiversity and carbon within the PBSR depends on the mainte-nance of low impact productive activities coordinated by multiple communal and private land-owners. In this study, we used state-of-the-art remote sensing data to investigate past spatial patterns in forest clearing dynamics and its relation to forest carbon density and forest plant species richness and diversity in the context of the forest conservation goals of the PBSR. We used a Landsat-based continuous change detection product for the 2000-2021 period and compared it to carbon density and tree species richness models generated from ALOS-2 PALSAR 2 imagery and national scale forest inventory data. The estimated error-adjusted area of detected annual forest clearings from the year 2000 until the year 2021 was 230,511 ha in total (+19,979 ha). The analysis of annual forest clearing frequency and area suggests that although forest clearing was significantly more intensive outside of the PBSR than within the PBSR during the entire 2000-2021 period, there is no evidence suggesting that the frequency and magnitude of forest clearing has changed over the years after the creation of the PBSR in 2011. An emergent hotspot analysis shows, however, that high spatiotemporal clustering of forest clearing events (hotspots) during the 2012-2021 pe-riod was less common than prior to 2011 and these more recent hotspots have been confined to areas outside the PBSR. After comparing forest clearing events to carbon density and tree species richness models, results show that land owners outside the PBSR often clear forests with lower carbon density and species diversity than land owners inside the PBSR. This suggests that, compared to land owners outside the PBSR, land owners within the PBSR might be practicing longer fallow periods allowing forests to attain higher carbon density and tree species richness and hence better soil nutrient recovery after land abandonment. In conclusion, our results show that the PBSR effectively acted as a stabilizing forest management scheme during the 2012-2021 period minimizing the impact of productive activities by lowering the frequency of forest clearing events and preserving late secondary forests within the PBSR. We recommend the continuation of efforts for providing alternative optimal field data collection strategies and modeling techniques to spatially predict key tropical forest attributes. The combination of these models with contin-uous change detection datasets will allow to reveal underlying ecological processes and generate information better adapted to forest governance scales.

Oxidative stress has been implicated in cellular senescence and aging, as well as in the onset and progression of many diverse genetic and acquired diseases and conditions. The search for biomarkers of oxidative stress has become relevant to many biomedical fields using many different methods and approaches. Fluorescence methodology is advantageous because of its simplicity and high sensitivity. Fluorescent probes are frequently used to investigate the role of reactive oxygen species in oxidative stress in experimental or clinical settings, often involving the use of flow cytometry and related single-cell based technologies. To assess the specific role of reactive oxygen species in oxidative stress studies by cytometric methodologies, it is essential to detect and characterize these species accurately. However, the detection and quantitation of individual intracellular ROS is still a challenge, but different and complementary strategies may focus on other endpoints of oxidative stress. In this review we present and discuss briefly the limitations and perspectives of such approaches. Based on our results, we also provide recommendations for proper design of cytometric studies of oxidative stress in order to prevent or minimize experimental errors..

Drought stress is a major production constraint in crops globally. Crop wild relatives are important source of resistance and tolerance for both biotic and abiotic stresses, respectively. A breeding program was initiated to introgress drought tolerance in sunflower through hybridization between the wild species Helianthus argophyllus and the cultigen pool of H. annuus. Selection was carried out in the F2 to F5 segregating populations for high cuticular waxes, smaller leaf area, single heading and high oil content. The selected F5 breeding lines were compared with non-adapted elite sunflower germplasm in a replicated randomized complete block research project using different water treatments comparing fully irrigated (T0), with 75 (T1), 50 (T2) and 25% (T3) irrigation treatments. The comparison between the two types of germplasm showed that drought tolerant breeding lines had comparatively lesser decreases in leaf area, shoot weight and root shoot ratio 10 under the various drought treatments. Leaf area of the drought selected F5 lines were smaller than that of the elite germplasm under controlled irrigated treatments, but they maintained their leaf area under the drought treatments. Several drought resistant promising lines D-2, D-5, and D-27 were identified and showed high leaf area, great root length and increase root to shoot ratio under the highest stressed treatments (T3). Some of the lines could be directly used for the development of drought tolerant hybrids. Combining ability test indicated D-27 (F7) as good general combiner for seed yield plant−1 and oil contents when mated with male line RSIN.82. These hybrids could help to minimize seed yield losses due to water stress and to achieve profitable cultivation of sunflower in 18 arid regions of Pakistan.

Establishment of mixed-forests has gained increasing attention as a way to optimize forest production, to improve ecological benefits and as a safety net for impacts of future climate uncertainties. However, practical knowledge about which species and what proportion of them should be mixed is still lacking. Thus, this study was conducted with the aim of identify suitable species for mixture with Acacia cincinnata. The mixture tested in the present study was A. cincinnata + Eucalyptus robusta (6:4), A. cincinnata + Acacia mangium (3:1) and monospecifc plantation of A. cincinnata established in 2014. After 7 years of growth, we analyzed the effects of species mixture on growth of tree species, understory vegetation and soil physico-chemical properties as well as bacterial community structure and diversity. The results showed that species mixture had no significant effect on growth characteristics, such as diameter and singletree volume of A. cincinnata. However, mixed-species planting increased the total stocking volume compered to monospecific plantation of A. cincinnata. Furthermore, stand mixture significantly increased species diversity, biomass and nutrient stocks in the understory vegetation. The soil of mixed stand of A. cincinnata and A. mangium had the highest C and N contents, whereas the soil of pure A. cincinnata stand had the highest P content. The diversity of soil bacterial community were the highest in the mixed stand of A. cincinnata and E- robusta, followed by pure A. cincinnata stand and A. cincinnata + A. mangium stand. The relative abundance of Proteobacteria and Actinobacteria was higher in soils of mixed stands. Furthermore, the relative abundance of Firmicutes was high in the soil of A. cincinnata + A. mangium while the relative abundance of Verrucomicrobia was high in A. cincinnata + E. robusta stand. As a whole, the study demonstrated that establishing mixed-species plantation enhance the diversity and composition of understory vegetation, soil physico-chemical and soils bacterial community; thereby increasing biodiversity, nutrient cycling and carbon sequestration in the biomass and soil. From the viewpoints of forest productivity and ecological benefits, it is advisable to establish a mixed plantation of A. cincinnata and A- mangium in southern China. As a whole, our work revealed that the sustainability of mixed-species plantation relies on the interactions between soil attributes, vegetation, and bacterial community.

French Guiana mosquito fauna diversity encompasses 242 species, of which nearly half of them belong to Culex (Diptera: Culicidae) genus. Whereas several species from Culex genus are important vectors of arbovirus, scarce studies focus on Culex genus, notably due to the inaccurate morphological identification of field-caught females gender. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) has been reported as a promising method for the identification of mosquitoes. Culex females from French Guiana field were dissected based on morphological classification. Abdomens were used for molecular identification using COI (cytochrome oxydase 1) gene. Legs and thorax of 169 specimens belonging to 13 Culex species,( i.e. Cx. declarator, Cx. nigripalpus, Cx. quinquefasciatus, Cx. usquatus, Cx. adamesi, Cx. dunni, Cx. eastor, Cx. idottus, Cx. pedroi, Cx. phlogistus, Cx. portesi, Cx. rabanicolus and Cx. spissipes) were then submitted to MALDI-TOF MS analysis. A high intra-species reproducibility and inter-species specificity of MS spectra for each mosquito body part tested were noticed. A corroboration of the specimen identification was revealed between MALDI-TOF MS, morphological and molecular results. MALDI-TOF MS protein profiling proves to be a suitable tool for identification of neotropical Culex species and will permit to enhance knowledge on this highly diverse Culex mosquitoes genus.

Borrelia miyamotoi is an emerging tick-borne pathogen in the Northern hemisphere and is the causative agent of Borrelia miyamotoi disease (BMD). B. miyamotoi is vectored by the same hard-bodied ticks as Lyme disease Borrelia, yet phylogenetically groups with relapsing fever Borrelia, and thus has been uniquely labeled a hard tick-borne relapsing fever Borrelia. Burgeoning research has uncovered new aspects of B. miyamotoi in human patients, nature, and the lab. Of particular interest are novel findings on disease pathology, prevalence, diagnostic methods, ecological maintenance, transmission, and genetic characteristics. Herein we review recent literature on B. miyamotoi, discuss how findings adapt to current Borrelia doctrines, and briefly consider what remains unknown about B. miyamotoi.

Earthworm invasions often reduce biodiversity and affect the ability of ecosystems to perform ecosystem functions. Over the past few decades, European lumbricide species have spread widely in natural habitats in Western Siberia, without completely displacing the native species. The aim of the present experiment is to study the survival, reproductive potential, vertical distribution, abundance, and biomass of Aporrectodea caliginosa and Lumbricus rubellus invasive for the region and the native species Eisenia nordenskioldi and their influence on each other. For this purpose, winter and vegetation experiments were conducted in mesocosms simulating the real situation of invasions in the same communities on the same types of soils with the same amount and composition of litter. The authors found that the native species was significantly inferior to A. caliginosa in reproduction rate, number of offspring, and final biomass, but had an advantage over L. rubellus. The native species was positively influenced by the presence of A. caliginosa. In the experiment, there was a significant rejuvenation of the populations of the studied species, an increase in abundance and relative mass under interaction conditions compared to mono variants. The native species had advantages over the invasive species in terms of winter survival. The natural volume of litter used in the experiment was insufficient for L. rubellus. According to the results of the experiment, it can be assumed that the spread of L. rubellus will continue to be limited to non-freezing biotopes rich in organic matter. A. caliginosa will spread in natural communities and agrocenoses of Western Siberia.

Ethnobotany has been, for too long, a descriptive discipline. However, ethnobotanists are increasingly calling for a paradigm shift towards the formulation of unifying theories and hypothesis-driven research in ethnobotany. Here, we formulated a theory, termed time-since-introduction theory, to explain the integration of alien plants into local pharmacopoeias in their recipient environment. This theory suggests that the factor time is paramount in determining which alien plants are more likely to be included in the medicinal flora of the areas they are introduced in. The theory relies on three hypotheses, the availability and versatility hypotheses alongside the residence time hypothesis newly proposed in the present study. We tested this theory by fitting a structural equation model to ethnobotanical data collected on South Africa’s alien woody flora. Although residence time is a direct predictor of medicinal status of alien plants, it is a better predictor when mediated through plant versatility. These findings are in support of the theory, and we consequently proposed a framework with which to understand different paths linking all three hypotheses. Collectively, our study shows the value of time in the development of ethnobotanical knowledge and fully responds to the pressing call for paradigm shift in ethnobotany.

Predator-prey interactions present heightened opportunities for pathogen spillover, as predators are exposed to novel parasites through consumption of prey harboring potentially infectious agents. Epizootics with high morbidity and mortality have been recorded following prey-to-predator spillover events with significant conservation implications, particularly for sensitive species. However, relatively few virulent infections following prey consumption are reported, given the very large number of exposures that presumably occur. Further, many transmitted agents are infectious but clinically silent and thus go unrecognized. Mechanisms that determine outcome of predator exposure to prey-based pathogens therefore represent an important, understudied component of disease dynamics that should be considered in modeling approaches and empirical research to better understand disease risk and emergence, particularly in vulnerable or threatened species.

Biological invasions represent a threat to ecosystems, through competition and habitat destruction, which may result in significant changes of the invaded community. Asparagopsis armata is a red macroalgae (Rodophyta) globally recognized as an invasive species. It is found from the intertidal to shallow subtidal areas, on rock or epiphytic, forming natural vegetation belts on exposed coasts. This study evaluated the variations on native intertidal seaweed and macroinvertebrate assemblages inhabiting rock pools with and without the presence of the invasive macroalgae A. armata. To achieve this, manipulation experiments on Atlantic (Portugal) rock pools were done. Three rock pools were maintained without A. armata by manual removal of macroalgae, and three others were not experimentally manipulated during the study period and A. armata was freely present. In this study the variations between different rock pools were assessed. Results showed different patterns in the macroalgae composition of assemblages but not for the macrobenthic communities. Ellisolandia elongata was the main algal species affected by the invasion of A. armata. Invaded pools tended to show less species richness, showing a more constant and conservative structure, with lower variation of its taxonomic composition than the pools not containing A. armata, where the variability between samples was always higher. Despite the importance of the achieved results, further data based on observation of long-term series are needed, in order to further understand more severe effects of the invader A. armata on native macroalgal assemblage.

Nonhost disease resistance is the most common type of plant defense mechanism against potential pathogens. In this study, the metabolic enzyme formate dehydrogenase (FDH1) was identified to be involved in nonhost disease resistance in Nicotiana benthamiana and Arabidopsis thaliana. In Arabidopsis, AtFDH1 was highly upregulated in response to both host and nonhost bacterial pathogens. Arabidopsis Atfdh1 mutants were compromised in nonhost resistance, basal resistance, and gene-for-gene resistance. The expression patterns of salicylic acid (SA) and jasmonic acid (JA) marker genes after pathogen infections in Atfdh1 mutant indicated that SA is most likely involved in the FDH1-mediated plant defense response to both host and nonhost bacterial pathogens. Previous studies reported that FDH1 localizes to only mitochondria, or both mitochondria and chloroplasts. Our results showed that the AtFDH1 localized to mitochondria and the amount of FDH1 localized to mitochondria increased upon infection with host or nonhost pathogens. Interestingly, the subcellular localization of FDH1 was observed in both mitochondria and chloroplasts after infection with a nonhost pathogen in Arabidopsis. We speculate that FDH1 plays a role in cellular signaling networks between mitochondria and chloroplasts to produce coordinated defense responses such as SA-induced reactive oxygen species (ROS) generation and hypersensitive response (HR)-induced cell death against nonhost bacterial pathogens.

In the wake of the current SARS-CoV-2 pandemic devastating the world, it is imperative to elucidate the comparative genomics of geographically-diverse strains of this novel coronavirus to gain insights into its microevolution, pathogenesis and control. Here we explore the molecular nature, genome-wide frequency, and gene-wise distribution of mutations in three distinct datasets encompassing 68 SARS-CoV-2 RNA-genomes altogether. While phylogenomic analysis revealed parallelism between the evolutionary paths charted by distinct quasispecies clusters of the virus, occurrence of mutations across genomes was found to be non-random. Whereas deletion mutations are extremely scarce and insertions totally absent, of all the instances of single nucleotide substitution detected, the overwhelming majority were transition mutations with cytidine to uridine being the most prevalent type. Propensity of this transition could be attributed to hydrolytic deamination mediated by ultra-violet irradiation or bisulfite reagent, both of which find wide usage as sterilizer/disinfectant. Transversions, albeit few and predominated by the guanosine to uridine form, were found concentrated in loci encoding the structural proteins of the virus, so might confer versatile tissue-colonization potentials. Mutation frequency of the three distinct genome-sets ranged narrowly between 0.07-1.08 × 10^-4 nucleotides positions mutated per nucleotide aligned. Gene-wise mapping of the global mutations illuminated the highly conserved nature of the genes encoding the non-structural proteins Nsp7, Nsp8 (two essential cofactors of the viral RNA-dependent RNA-polymerase) and Nsp9 (Nsp8-interacting single-strand RNA-binding protein), plus the envelope protein E (involved in SARS-CoV-2 assembly, budding and pathogenesis). These mutation-free genomic loci and/or their protein products could be potent targets for future drug designing/targeting.

The Cape Restionaceae species, an endemic of the Fynbos Biome, is threatened by urbanization, alien plant invasion, agricultural expansion, and groundwater extraction. This is further worsened by the semi-arid conditions and hydrological variability factors, which influences species niche dynamics. Therefore, it is important to assess and monitor the Restionaceae species for preservation of their endemism and richness. This study models the hydrological niche and distribution changes of Restionaceae species at the New Years Peak (NYP) at microclimate level for biodiversity conservation. MaxEnt modelling and GIS analytical approaches were applied at various stages in niche modelling process as follows: (i) microclimatic input raster layers’ generation, (ii) ecological modelling and hydrological niche manipulation, and (iii) spatial distributional change mapping. The hydrological niches of the Restionaceae were effectively examined under the recent climate and compared with RCP2.6 and RCP8.5 future climate scenarios as the microscale environmental inputs. The results showed that most of the studied Restionaceae species positioned themselves along a hydrological gradient. Each species tolerated a range of hydrological conditions, which formed their hydrological niche. Changing climate would cause both positive and negative species range shifts. The study assists in plant species conservation and future climate change impact analysis on endangered plant species.

Candida spp. are the most common cause of fungal infections worldwide. The taxonomy of Candida is controversial and has undergone recent changes due to novel genetically related species. Therefore, some complexes of cryptic species have been proposed. In clinical settings, the correct identification of Candida species is relevant since some species are associated with high resistance to antifungal drugs and increased virulence. This study aimed to identify the species of four Candida complexes (C. albicans, C. glabrata, C. parapsilosis, and C. haemulonii) by molecular methods. This is the first report of six cryptic Candida species in Honduras: C. dubliniensis, C. africana, C. duobushaemulonii, C. orthopsilosis, and C. metapsilosis, and it is also the first report of the allele hwp1-2 of C. albicans sensu stricto. It was not possible to demonstrate the existence of C. auris among the isolates of the C. haemulonii complex. We also propose a simple method based on PCR-RFLP for the discrimination of the multi-resistant pathogen C. auris within the C. haemulonii complex.

The Populus L. genus unites tree species, which are botanically grouped into several sections. The species successfully hybridize both in the same section and between some sections. The poplar hybridization widely occurs in nature and in variety breeding. Therefore, the development of poplar species specific molecular markers is very actual. The effective markers for trees of the Aigeiros Daby section have been recently developed using the polymorphism of the 5S rDNA non-transcribed spacers (NTSs). In this article, the 5S rDNA NTS based markers were designed for several species of the Leuce Daby section. The alb9 marker amplifies one fragment with DNA matrix of P. alba and P. × canescens (natural hybrid P. alba × P. tremula). The alb2 marker works the same way except the case with P. bolleana. In this case, the amplification of three fragments was observed. The tremu1 marker amplification is detected with DNA matrix of P. tremula and P. × canescens. Thus, the developed markers may be applied as useful tool for the P. alba, P. tremula, P. × canescens and P. bolleana identification in such areas of plant science as botany, dendrology, genetics of populations, variety breeding etc.

The research objectives was to determine the plants vegetation profile in forest revegetation and Mount Arjuna springs. The method used was descriptive method. While based on the techniques and tools used to research, the author uses the survey method to obtain facts that occur in the research area, namely in the area around the springs of Arjuna mount, Pasuruan, East Java, Indonesia. Data obtained in the field are then processed and analyzed using GIS, ArcView 3.3 and Google Earth programs. The results of the study explain the profile of plant vegetation, namely plant stratification, bird wealth, taxonomic wealth and plant density have a significant effect on forest revegetation and springs in Arjuna mount. The forest vegetation profile has a positive and significant effect on the preservation of springs. The better the profile of forest vegetation, the preservation of springs will be better, and vice versa.

Data on the seasonally dry tropical forests of Mexico have been examined in the light of statistical mechanics. The results suggest a division into two classes of species. There are drifting populations of a cosmopolitan class capable of existing in most dry forest sites; these have a statistical distribution previously only observed (globally) for populations of alien species. A high proportion of species found only at a single site are endemic and these prefer sites comparatively low in species richness.

Hymenoscyphus albidus is a native fungus in Europe where it behaves as a harmless decomposer of leaves of common ash. Its close relative Hymenoscyphus fraxineus was introduced into Europe from Asia and currently threatens ash (Fraxinus sp.) stands all across the continent causing ash dieback. H. fraxineus isolates from Europe were previously shown to harbor a mycovirus named Hymenoscyphus fraxineus Mitovirus 1 (HfMV1). In the present study, we describe a conspecific mycovirus that we detected in H. albidus. HfMV1 was consistently identified in H. albidus isolates (mean prevalence: 49.3%) which were collected in the sampling areas before the arrival of ash dieback. HfMV1 strains in both fungal hosts contain a single ORF of identical length (717 AA) for which a mean pairwise identity of 94.5% was revealed. The occurrence of a conspecific mitovirus in H. albidus and H. fraxineus is most likely the result of parallel virus evolution in the two fungal hosts. HfMV1 sequences from H. albidus showed a higher nucleotide diversity and a higher number of mutations compared to those from H. fraxineus, probably due to a bottleneck caused by the introduction of H. fraxineus in Europe. Our data also points to multiple interspecific virus transfers from H. albidus to H. fraxineus, which could have contributed to the intraspecific virus diversity found in H. fraxineus.

Asthma is a chronic inflammatory airway disease induced by many environmental factors. The inhalation of allergens and pollutants promote the reactive oxygen species (ROS) production leading to airway inflammation, hyper-responsiveness and remodeling in allergic asthma. The effects of asthma medications on ROS production are unclear. The present study investigated the anti-ROS effects of current asthma medications including inhaled corticosteroid (ICS; budesonide and fluticasone), leukotriene receptor antagonist (LTRA; montelukast), long acting β2 agonists (LABAs; salmeterol and formoterol) and a new extra-LABA (indacaterol). The human monocyte cell line THP-1 cells were pre-treated with different concentrations of the asthma medications at different time-points after hydrogen peroxide (H₂O₂) stimulation. H₂O₂ production was measured with DCFH-DA by flow cytometry. Montelukast, fluticasone and salmeterol suppressed H₂O₂-induced ROS production. Indacaterol enhanced H₂O₂-induced ROS production. Budesonide and formoterol alone had no anti-ROS effects, but the combination of these two drugs significantly suppressed H₂O₂-induced ROS production. Different asthma medications have different anti-ROS effects on monocytes. The combination therapy with LABA and ICS seemed not be the only choice for asthma control. Montelukast may be also a good supplemental treatment for the poorly-controlled asthma because of its powerful anti-ROS effects. Our findings provide a novel therapeutic view in asthma.

One of the earliest hallmarks of plant immune response is production of reactive oxygen spe-cies (ROS) in different subcellular compartments, which regulate plant immunity. A suitable equilibrium, which is crucial to prevent ROS over-accumulation leading to oxidative stress, is maintained by salicylic acid (SA), a chief regulator of ROS. However, ROS are not only acting downstream of SA signaling, but were also proposed to be a central component of a self-amplifying loop that regulates SA signaling as well as the interaction balance between dif-ferent phytohormones. The exact role of this crosstalk, the position where SA interferes with ROS signaling and ROS interferes with SA signaling and the outcome of this regulation depend on the origin of ROS but also on the pathosystem. The precise spatiotemporal regulation of or-ganelle specific ROS and SA levels determine the effectiveness of pathogen arrest and is there-fore crucial for a successful immune response. However, the regulatory interplay behind still remain poorly understood, as up till now, the role of organelle specific ROS and SA in HR-conferred resistance has mostly been studied by altering the level of a single component. In order to address these aspects, a sophisticated combination of research methods for monitoring the spatiotemporal dynamics of key players and transcriptional activity in plants is needed, and will most probably consist of biosensors and precision transcriptomics.

Astragalus plants belong to the legume family and have obvious rhizosphere effect. The northwestern area of Yunnan China is the main distribution region of Astragalus plants with high application value, but low recognition and resource reserves. The aim of this research is to explore the species diversity of cultivable rhizosphere fungi derived from A. acaulis, A. forrestii, and A. ernestii growing in special high-cold environment of northwest Yunnan, and discover anti-infective components from these fungi. A total of 93 fungal strains belonging to 38 species in 18 genera were isolated and identified. Antibacterial and antimalarial screening yielded 10 target strains. Among them, the ethyl acetate crude extract of the fungus Aspergillus calidoustus AA12 from A. acaulis fermentation substance showed a broad antibacterial spectrum and the best antimalarial activity. Further chemical investigation led to the first discovery of seven compounds from the species of A. calidoustus, including a sesterterpine 6-epi-ophiobolin G (1), two sesquiterpenes penicisochroman A (2), pergillin (3) and 7-methyl-2-(1-methylethylethlidene)-furo[3,2-H]isoquinoline-3-one (4), and three polyketides trypacidin (5), 1,2-seco-trypacidin (6) and questin (7). Compound 1 exhibited moderate to strong antibacterial activity against six Gram-positive pathogens with the minimum inhibitory concentration (MIC) ranging from 25 to 6.25 μg/mL, and prominent inhibitory effect on the biofilm of Streptococcus agalactiae at MIC value of 3.125 μg/mL. This compound also displayed potent antimalarial activity against Plasmodium falciparum strains 3D7 and chloroquine-resistant Dd2 at the half maximal inhibitory concentration (IC50) values of 3.319 and 4.340 µmol/L at 72 h, respectively. This study contributed to our understanding of the cultivable rhizosphere fungi Astragalus-derived in special high-cold environment and further increased the library of fungi available for natural anti-infectious products screening.

Mango industry played important roles in the economy of tropical and subtropical regions in China. Embryo abortion, flesh rotten, pericarp stain and so on always result in heavy loss. Transgenic breeding is a potential way for solving these problems. However, dependable protocol for mango transgene has not been reported until now. In this research, we found that after mango embryogenic callus were infected by Agrobacterium, most callus necrotized and died. No seedling was differentiated from the survived callus. Contents of 106 compounds in mango callus infected by Agrobacterium were found significantly different from those in control. The top 7 up-regulated compounds were gallic acid, phosphoenolpyruvic acid, dihydrotricetin, ubiquinone, dihydrokaempferol, D-erythrose-4-phosphate and cyanidin. The expressions of ROS-related genes, gallic acid synthesis genes, and defense related genes in callus infected by Agrobacterium were significantly higher than those in control. The growth of Agrobacterium on culturing plate pipetting gallic acid was inhibited. Mango callus infected by Agrobacterium transformed with type III secretion system gene cluster (T3SS) and effector gene AvrPto isolated from Pseudomonas syringae pv. tomato strain DC3000 brown and necrotize slower than those infected by Agrobacterium transformed with net vector (NV). The contents of ROS in callus in-fected by Agrobacterium (T3SS-AvrPto) were less than those in callus infected by Agrobacterium (NV). The expressions of genes participated in immune responses in mango callus infected by Agrobacterium (T3SS-AvrPto) were also less than those in mango callus infected by Agrobacterium (NV). The transformation rate of mango callus infected with Agrobacterium (T3SS-AvrPto-gfp) was 1.6 %. No callus infected with Agrobacterium (NV-gfp) was transformed successfully. The efficiency of mango transformation can be improved by constructing Agrobacterium strains harboring type III secretion system gene cluster and its effector gene.

Glycogen storage disorders (GSDs) are a group of inherited metabolic disorders characterized by defects in enzymes involved in glycogen metabolism. Deficiencies in enzymes responsible for glycogen breakdown and synthesis can impair mitochondrial function. For instance, in GSD type II (Pompe disease), acid alpha-glucosidase deficiency leads to lysosomal glycogen accumulation, which secondarily impacts mitochondrial function through dysfunctional mitophagy, which disrupts mitochondrial quality control generating oxidative stress. In GSD type III (Cori disease), the lack of the debranching enzyme causes glycogen accumulation and affects mitochondrial dynamics and biogenesis by disrupting the integrity of muscle fibers. Malfunctional glycogen metabolism can disrupt various cascades thus causing mitochondrial and cell metabolic dysfunction through various mechanisms. These dysfunctions include altered mitochondrial morphology, impaired oxidative phosphorylation, increased production of reactive oxygen species (ROS), and defective mitophagy. The oxidative burden typical of GSDs compromises mitochondrial integrity and exacerbates the metabolic derangements observed in GSDs. The intertwining of mitochondrial dysfunction and GSDs underscores the complexity of these disorders and has significant clinical implications. GSD patients often present with multisystem manifestations, including hepatomegaly, hypoglycemia, and muscle weakness, which can be exacerbated by mitochondrial impairment. Moreover, mitochondrial dysfunction may contribute to the progression of GSD-related complications, such as cardiomyopathy and neurocognitive deficits. Targeting mitochondrial dysfunction thus represents a promising therapeutic avenue in GSDs. Potential strategies include antioxidants to mitigate oxidative stress, compounds that enhance mitochondrial biogenesis, and gene therapy to correct the underlying mitochondrial enzyme deficiencies. Mitochondrial dysfunction plays a critical role in the pathophysiology of GSDs. Recognizing and addressing this aspect can lead to more comprehensive and effective treatments, improving the quality of life of GSD patients. This review aims to elaborate on the intricate relationship between mitochondrial dysfunction and various types of GSDs. The review presents challenges and treatment options for several GSD

Temporal changes of population densities and species richness of three main pollinator groups: moths and butterflies (Lepidoptera), bees, wasps and sawflies (Hymenoptera) and hoverflies, horse-flies, tachninids and bee flies (Diptera) were investigated in the Carpathian Basin. Maintaining pollinator divesrity is a crucial factor for preserving our biodiversity and ecosystems, furthermore several pollinator species has strong economic role in maintaning crop and fruit cultures. Our conclusions are based on our three or four decades of faunistic surveys in various regions of the Carpathian Basin. Analysing and comparing our data with the historical data of the last 50 years, we concluded, densities of some pollinators were declined during the past decade and the half (Symphyta, hoverflies), although populations of several species of Mediterranean-origin were grown (Aculeata) and even new species were migrated from the warmer regions. In numerous cases, this decrease was dramatic: more than 90% decrease of certain butterfly species were detected. On the other hand, the composition of pollinator fauna significantly changed due to the disappearance of some montaneous or mesophyle species. The main reason of decrease of pollinator communities is partly the climatic change and partly anthropogenic factors. Our conclusion: in our region, the pollinator crisis is present, but moderate; however, there is clear sign of the gradual transition of our pollinator fauna towards the Mediterranean type.

Reactive oxygen species (ROS) play an important role in biological processes and offer hope for the development of effective treatments for ROS-dependent diseases, including cancer. The interaction of hydrogen peroxide with proteins stimulates the creation of chemical molecules that can weaken or stop the pathological process in cancer. However, reliable drugs have not yet been described. To understand the reason for the failure, we studied the behavior of ROS-containing compounds in cancer cells. The epidermal growth factor receptor (EGFR) is activated by interaction with a ligand protein, as well as by binding the chemical structure of hydrogen peroxide to the catalytic cysteine in the intracellular domain. Both mechanisms trigger signaling pathways in cells. The EGFR can be inactivated by protein tyrosinase phosphatase PTP-1B, which itself is activated by hydrogen peroxide. A simple in-gel fluorescence technique was used to demonstrate the rapid binding of hydrogen peroxide to a large number of proteins, including both activating and inactivating target proteins. Natural defense systems take longer to action and apparently cannot prevent hydrogen peroxide from binding to protect cells from exposure to unwanted proteins. Therefore, superoxide dismutase, which produces hydrogen peroxide, is not a suitable target for disease therapy.

Synanthropic flora (weeds, spontaneously developed plants) are plants that accompany humans, appearing as a result of anthropopressure as well as after its cessation. The potential of synanthropic flora in shaping green areas usually brings many measurable benefits that depend on land management in cities. The study aimed to determine the preferences and attitudes of adults towards synanthropic flora in settlement units. The article presents survey results from 447 city residents. The obtained results were statistically analyzed using the k-means clustering method to identify segments of respondents with similar attitudes and preferences toward synanthropic flora. Mann-Whitney U tests were employed to determine statistical differences in the demographic variables among the analyzed clusters of respondents. Additionally, multiple regression analysis was performed to identify the opinions and attitudes towards synanthropic flora, that may influence their greater acceptance in the vicinity of residential areas. The results obtained in multiple regression allow for the statement that the respondent’s evaluation of the level of support for the introduction of more synanthropic vegetation into urban greenery (Yw) grows with the increase of acceptance of synanthropic plants occurrence in surroundings. Respondents’ opinions on the subject of the presence of synanthropic plants were studied using such variables concerning: synanthropic vegetation found in turfgrasses has a positive impact on the visual attractiveness of a site (X1), synanthropic plant communities present themselves better than others (X2), presence of synanthropic vegetation in an estate improves the quality of life in that location (X3), introduction of synanthropic flora into estate arrangements is a positive (X4), support for the use of synanthropic flora for educational purposes - as elements of playgrounds or in the form of environmental workshops (X5). Residents’ acceptance of synanthropic flora in green areas is at a relatively high level.

DNA barcoding has emerged as a transformative tool in the realm of biodiversity assessment, facilitating rapid and precise species identification. This review article delves into the evolving landscape of DNA barcoding, with a particular focus on the emergence of mini-barcodes and alternative genes as innovative approaches. Traditional DNA barcoding, anchored in the COI gene, has significantly advanced our understanding of species diversity; however, inherent challenges persist, especially when dealing with degraded samples or taxonomic groups where COI falls short. The introduction of mini-barcodes, characterized by their reduced sequence length, has revolutionized DNA barcoding by expanding its applicability. We explore the utility of mini-barcodes in various scenarios, ranging from DNA extraction from ancient or fragmented specimens to in-field identification using portable sequencing technologies. Case studies and practical insights into their effectiveness are presented, emphasizing their role in overcoming traditional limitations. Alternative genes, a complementary facet of DNA barcoding, offer a novel avenue for enhancing taxonomic resolution and accuracy. This review showcases instances where researchers have harnessed alternative genes to overcome COI's constraints, particularly in diverse taxonomic groups, such as fungi, plants, and microorganisms. The selection of these genes is discussed, along with their potential to uncover hidden biodiversity and improve species delimitation. Examining the broader implications of mini-barcodes and alternative genes, we unveil their pivotal roles in advancing conservation efforts, forensic science, and citizen science initiatives. The adaptable nature of these approaches empowers a diverse range of stakeholders, from scientists conducting critical research to concerned citizens contributing to our collective understanding of the natural world. Looking ahead, we envision an exciting future for DNA barcoding, as these innovations continue to evolve. Integration with cutting-edge sequencing technologies, data analysis methods, and the harnessing of big data hold immense potential. Nonetheless, challenges related to standardization, scalability, and data sharing persist.

Chemo-resistance is a substantial challenge in the realm of cancer treatment that requires exploring new therapeutic approaches for effective mitigation. Achieving this goal requires examination of the molecular mechanisms involved in both tumor growth and therapeutic interventions. The potential of NRF2 (Nuclear factor E2-related factor 2) in addressing resistance to chemotherapy across diverse cancer types highlights its value as a promising therapeutic approach based on cancer characteristics. Manipulating the NRF2 signaling pathway has a dual impact, offering promise for both preventing and treating cancer, as well as inhibiting carcinogenesis. The influence of the NRF2/KEAP1 pathway on the progression of tumor formation and resistance to drugs has been well-documented. The interplay between the NRF2 signaling pathway and processes such as endoplasmic reticulum (ER) stress, unfolded protein response (UPR), and autophagy plays a crucial protective role. A deeper understanding of NRF2's role in the modulating these pathways is necessary to develop novel approaches for improving chemotherapeutic efficacy. This article discusses the significance of the NRF2-KEAP1 pathway in preventing/promoting cancer and resistance mechanisms to various chemotherapeutic agents, with a focus on the complementary effects of antioxidants via NRF2-mediated signaling pathways. This study aims to provide a molecular basis for targeting NRF2 via inhibitors/activators as promising therapeutic strategies to overcome chemo-resistance.

: Intracellular tripeptide glutathione is an important agent of cell survival in hypoxia. Glutathione covalently binds to SH-groups of hemoglobin cysteine residues, protecting them from irreversible oxidation and changes its affinity to oxygen. Reduced glutathione (GSH) can also form a non-covalent complex with hemoglobin. Previously, we showed that hemoglobin tetramer has four noncovalent binding sites of glutathione GSH molecules inside, two of which are released during hemoglobin transition to deoxy form. In this study, we characterized the conserved cysteine residues and residues of non-covalent glutathione binding sites in the sequences of a number of hypoxia tolerant and hypoxia-sensitive mammals. The alpha subunit of all species considered was found to have no conserved cysteines, whereas the beta subunit contains Cys94 residue, which is conserved across species and whose glutathionylation changes the affinity of hemoglobin for oxygen 5-6-fold. In addition, the key residues of noncovalent glutathione binding sites in both alpha and beta subunits are absolutely conserved in all species considered, suggesting a common mechanism of hemoglobin redox regulation for both hypoxia-sensitive and hypoxia-tolerant mammals. The solvent accessibility of all HbA and HbB residues in oxy- and deoxy- forms was analyzed. The key residue betaCys94 is solvent accessible only in the deoxy form.

Given the dramatic increase of L. sceleratus population in southeastern Aegean Sea, there is growing interest in assessing the toxicity of this pufferfish and the factors controlling its tetrodotoxin (TTX) content. In the present study, liver, gonads, muscle and skin of L. sceleratus specimens from Rhodes island, Greece were subjected to multi-analyte profiling using liquid chromatography - tandem mass spectrometry (LC-MS/MS) in order to quantitate TTX and evaluate whether this biotoxin interrelates with hormones. TTX and its analogues 4-epiTTX, 11-deoxyTTX, 11-norTTX-6-ol, 4,9-anhydroTTX and5,11-6,11-dideoxyTTX were detected in all tissue types. Liver and gonads were the most toxic tissues, with the highest TTX concentrations being observed in the ovaries of female specimens. Only 22% of the analyzed muscle samples were non-toxic according to the Japanese toxicity threshold (2.2 μg TTX eq g-1), confirming the high poisoning risk from from the inadvertent consumption of this species. Four steroid hormones (i.e., cortisol, testosterone, androstenedione and β-estradiol) and the gonadotropin-releasing hormone (GnRH) were detected in the gonads. Androstenedione dominated in female specimens, while GnRH was more abundant in males. A positive correlation of TTX and its analogues with β-estradiol was observed. Though, a model incorporating sex rather than β-estradiol as independent variable was proved more efficient in predicting TTX concentration, implying that other sex-related characteristics are more important than specific hormone-regulated processes.

Global climate change, dominated by climate warming, is seriously affecting the balance of global ecosystems, but the risk of species extinction is particularly high in low-altitude mountain areas. To clarify the response of the endemic and critically endangered species Ilex nanchuanensis to climate change, this study used the MaxEnt model to simulate and predict the potential habitat of I. nanchuanensis during the Last Interglacial, Last Glacial Maximum, the current period, and two future periods (the 2050s and 2070s). The results showed that the hottest monthly minimum temperature is the most important climatic factor affecting the geographical distribution of I. nanchuanensis. Furthermore, I. nanchuanensis will be at risk of population shrinkage and extinction in the future, with the center of mass moving further northwest as concentrations of greenhouse gases increase, especially in the 2070s, when its geographical distribution shrinks the most under the RCP6 scenario. Global warming is causing habitat degradation for many species and will continue to migrate to higher altitudes and latitudes in the future, seriously affecting the global ecosystem balance. Therefore, to actively respond to the impacts of climate change, protected areas should be established around the geographical distribution centers of species, and core, buffer, and experimental areas should be scientifically and rationally delineated for the conservation and cultivation of germplasm resources.

Vegetation classifications on large geographic scales are necessary to inform conservation decisions and monitor keystone, invasive, and endangered species. These classifications are often effectively achieved by applying models to imaging spectroscopy, a type of remote sensing, data, but such undertakings are often limited in spatial extent. Here we provide accurate, high-resolution spatial data on the keystone species Metrosideros polymorpha, a highly polymorphic tree species distributed across bioclimatic zones and environmental gradients on Hawai'i island, using airborne imaging spectroscopy and LiDAR. We compare two tree species classification techniques, support vector machine (SVM) and spectral mixture analysis (SMA), to assess their ability to map M. polymorpha over 28,000 square kilometers where differences in topography, background vegetation, sun angle relative to the aircraft, and day of data collection, among others, challenge accurate classification. To capture spatial variability in model performance, we applied gaussian process classification (GPC) to estimate the spatial probability density of M. polymorpha occurrence using only training sample locations. We found that, while SVM and SMA models exhibit similar raw score accuracy over the test set (96.0% and 93.4%, respectively), SVM better reproduces the spatial distribution of M. polymorpha than SMA. We developed a final 2 m x 2 m M. polymorpha presence dataset and a 30 m x 30 m M. polymorpha density dataset using SVM classifications that have been made publicly available for use in conservation applications. Accurate, large-scale species classifications are achievable, but metrics for model performance assessments must account for spatial variation of model accuracy.

For the water bodies of the Lena Delta Nature Reserve, 413 taxa were known. In 14 small tundra reservoirs in its vicinity, we identified 385 taxa, which in the compilatory list made up a significant diversity (666 taxa including definitions to the genus level) of diatoms. Thus, the species composition of diatoms in the reserve and adjacent territories was replenished by 278 species. The predominance of species of the genera Pinnularia (57) and Eunotia (51) at the family and generic levels was revealed. Index of intraspecific variability Ssp./Sp. for the lakes of the reserve was 1.11, and for the lakes of the Tiksi region 1.14, which is typical for high-latitude and high-mountain communities. The number of rare or endangered species varied in different lakes within 1-10, totaling 42 species for the entire study area. Bioindication has shown that potentially threatened species prefer moderate temperatures, slightly acidic or neutral environments free from organic pollution. A comparative analysis of the species composition of diatoms in the vicinity of the Lena Delta and other northern water bodies of Yakutia and the Arctic Islands showed that the species composition of each lake in the Arctic is strictly individual. The indicator characteristics show a certain response of the species composition of diatoms to changes in salinity, pH, and organic pollution. A number of regularities in the spatial distribution of diatoms in the study area were found in connection with the physicochemical parameters of their habitat, height above sea level, and relation to a particular catchment basin. Statistical mapping of indicators of community and habitat diversity revealed a strong reaction to point one-time pollution and made it possible to assume the influence of summer northeast winds on the species composition. We suggest that the high diversity inherent in the diatom lakes of the Tiksi coastal zone, which can even be replenished in further studies, can be considered as a property of coastal biota inherent in ecotones. Since it is in the coastal Tiksi region that a surge in the number of species is observed, this region can be considered not only an ecotone, but also a hotspot of diatom diversity. The results of the study are important for developing the basis for monitoring biodiversity under the conditions of anthropogenic and climatic changes in the Arctic.

Conservation of tropical endemic amphibians largely suffers from Wallacean shortfall, a gap to which predictive species distribution models have contributed significantly to bridge by delineating probable distribution and the underlying suitable habitat within their distributional range. However, rarely is a prediction model ground-truthed to evaluate their predictive performance. Here we present a species distribution modeling approach using maximum entropy algorithm corrected for smaller sample size, in guiding explorative surveys aimed at optimizing survey effort and discovering unrecorded populations of Zhangixalus suffry, a rhacophorid tree-frog endemic to the northeastern part of the Indian subcontinent along with the factors limiting their distribution. With only 16 established historical locality data to model for (after spatial thinning to reduce autocorrelation) and a set of environmental predictors (climatic, topographic, and landscape composition), our model prediction enabled the successful discovery of seven new population records from unreported landscapes, extending its southernmost distributional limit over a considerable distance. The final composite distribution model combining all the locality records (n=23) predicted similar core areas of suitable habitat consistent with the known geographic distribution of the species but showed poor representation under existing coverage of Protected Area (PA) network in the Region with only 7% representation of suitable habitat under protection. Habitat suitability of a site was significantly governed by factors related to precipitation (precipitation seasonality and precipitation of the warmest quarter) and topographic factors that can influence it (elevation and aspect). This corroborates with the known ecology of Rhacophorid frogs, especially concerning their seasonal explosive reproductive strategy and foam nest-building behavior.Through this study, we propose explorative surveys guided by species distribution models to expedite unknown population discovery of rare, tropical endemic amphibians and using such taxa as surrogates in identifying conservation priority zones that can be directly applied to reserve design and conservation and management planning.

Genetic, developmental, biochemical, and environmental variables interact intricately to produce sex differences. The significance of sex differences in cancer susceptibility is being clarified by numerous studies. Epidemiological research and cancer registries have revealed over the past few years that there are definite sex variations in cancer incidence, progression, and survival. However, oxidative stress and mitochondrial dysfunction also have a significant impact on the response to treatment of neoplastic diseases. Young women may be more protected from cancer than men because most of the proteins implicated in the regulation of redox state and mitochondrial function are under the control of sexual hormones. In this review, we describe how sexual hormones control the activity of antioxidant enzymes and mitochondria, as well as how they affect several neoplastic diseases. The molecular pathways that underlie the gender-related discrepancies in cancer that have been identified may be better understood, which may lead to more effective precision medicine and vital information on treatment options for both males and females with neoplastic illnesses.

Major disturbances fundamentally alter ecosystems through the transformation of both biotic and abiotic factors. The eruption of Mount St. Helens in 1980 resulted in a cataclysmic restructuring of its surrounding landscapes. The Pumice Plain is one of these landscapes, where tree species such as Sitka willow (Salix sitchensis) and their dependent communities have established along newly-formed streams. Thus, the study of these dependent communities provides a unique opportunity to investigate factors influencing metacommunity assembly during true primary succession. We analyzed the influence of landscape connectivity on metacommunity assembly through a novel application of circuit theory, alongside the effects of other factors such as stream locations, willow leaf chemistry, and leaf area. We found that landscape connectivity structures community composition on willows across the Pumice Plain, while our other factors had varied effects. Most importantly, multiple levels of spatial habitat structure linked via landscape connectivity can predict the presence of organisms lacking high rates of dispersal, such as the invasive stem-boring poplar weevil (Cryptorhynchus lapathi). This is critical for management as we show that the maintenance of a heterogeneous mixture of landscape connectivities and resource locations can facilitate meta-community dynamics to promote ecosystem function and mitigate the influences of invasive species.

B-cell acute lymphoblastic leukemia (B-ALL) is a hematologic disorder characterized by the abnormal proliferation and accumulation of immature B-lymphoblasts arrested at various stages of differentiation. Despite advances in treatment, a significant percentage of pediatric patients with precursor B-ALL still relapse. Therefore, alternative therapies are needed to improve the cure rates for pediatric patients. TPEN (N, N, N’, N’-tetrakis(2-pyridylmethyl)-ethylenediamine).is a pro-oxidant agent capable of selectively inducing apoptosis in leukemia cells. Consequently, it has been suggested that TPEN could be a potential agent for oxidative therapy. However, it is not yet known whether TPEN can selectively destroy leukemia cells in a more disease-like model, for example, the bloodstream and bone marrow (BM), in vitro. This investigation is an extension of a previous study that dealt with the effect of TPEN on ex vivo isolated/purified refractory B-ALL cells. Here, we evaluated the effect of TPEN on whole BM from nonleukemic patients (control) or pediatric patients diagnosed with de novo B-ALL or refractory B-ALL cells by analyzing the hematopoietic cell lineage marker CD34/CD19. Although TPEN was innocuous to nonleukemic BM (n=3), we found that TPEN significantly induced apoptosis in de novo (n = 5) and refractory B-ALL (n = 6) leukemic cell populations. Moreover, TPEN significantly increased the counts of cells positive for the oxidation of the stress sensor protein DJ-1, a sign of the formation of H2O2, and significantly increased the counts of cells positive for the pro-apoptotic proteins TP53, PUMA, and CASPASE-3 (CASP-3), indicative of apoptosis, in B-ALL cells. We demonstrate that TPEN selectively eliminates B-ALL cells independent of age, diagnosis status (de novo or refractory), sex, karyotype, or immunophenotype. Understanding TPEN-induced cell death in leukemia cells provides insight into more effective therapeutic oxidation-inducing anticancer agents.

Glufosinate, a glutamine synthetase (GS) inhibitor, often provides variable weed control depending on environmental conditions such as light, temperature and humidity at the time of application. Midday applications normally provide improved efficacy compared to applications at dawn or dusk. We investigated the physiological, molecular, and biochemical basis for the time-of-day effect on glufosinate efficacy in Amaranthus palmeri. GS1 and GS2 gene expression and protein abundance were assessed in different parts (young leaves, old leaves, and roots) of plants incubated in the dark compared to those in the light. The turnover of GS total activity was also evaluated overtime following glufosinate treatment at midday compared to dusk application. The results suggest that GS in A. palmeri is less expressed and less abundant in the dark compared to in the light. Midday application of glufosinate under intense light conditions in the hours following application provide full control of A. palmeri plants. Consequently, these plants are unable to recover GS activity by de novo protein synthesis. Full activity of GS is required for complete inhibition by the irreversible inhibitor glufosinate. Therefore, glufosinate applications should always be performed in the middle of the day when sunlight is intense, to prevent weed escapes from the herbicide treatment.

The horse chestnut leaf miner Cameraria ohridella (Lepidoptera: Gracillariidae) is an invasive pest of horse chestnut and has spread through Europe since 1985. The horse chestnut leaf blotch Guignardia aesculi (Botryosphaeriales: Botryosphaeriaceae) is a fungal disease that also seriously damages horse chestnut trees in Europe. The interaction between the leaf miner and the fungus has not yet been sufficiently described. Therefore, the aim of the present study was to assess leaf damage inflicted to horse chestnut by both C. ohridella and G. aesculi during the vegetation season and to model their interaction. The damage to leaf area was measured monthly from May to September 2013 in České Budějovice, the Czech Republic. A simple phenomenological model describing the expected dynamics of the two species was developed. The study revealed a significant effect of sampling site and sampling period on the damage caused by both the pest and the fungus. The mathematical model indicates that infestation by C. ohridella is more affected by G. aesculi than vice versa. The possible mechanisms affecting the relationship between G. aesculi and C. ohridella are discussed.

Bacteria currently included in Rhizobium leguminosarum are too diverse to be considered a single species, so we can refer to this as a species complex (the Rlc). We have found 429 publicly available genome sequences that fall within the Rlc and these show that the Rlc is a distinct entity, well separated from other species in the genus. Its sister taxon is R. anhuiense. We constructed a phylogeny based on concatenated sequences of 120 universal (core) genes, and calculated pairwise average nucleotide identity (ANI) between all genomes. From these analyses, we concluded that the Rlc includes 18 distinct genospecies, plus 7 unique strains that are not placed in these genospecies. Each genospecies is separated by a distinct gap in ANI values, usually at around 96% ANI, implying that it is a 'natural' unit. Five of the genospecies include the type strains of named species: R. laguerreae, R. sophorae, R. ruizarguesonis, "R. indicum" and R. leguminosarum itself. The 16S ribosomal RNA sequence is remarkably diverse within the Rlc, but does not distinguish the genospecies. Partial sequences of housekeeping genes, which have frequently been used to characterise isolate collections, can mostly be assigned unambiguously to a genospecies, but alleles within a genospecies do not always form a clade, so single genes are not a reliable guide to the true phylogeny of the strains. We conclude that access to a large number of genome sequences is a powerful tool for characterising the diversity of bacteria, and that taxonomic conclusions should be based on all available genome sequences, not just those of type strains.

In late 2018, an epidemic myxomatosis outbreak emerged on the Iberian Peninsula leading to high mortality in Iberian hare populations. Soon, a recombinant virus (MYXV-Tol or ha-MYXV) was identified, harboring a 2.8 kb insertion containing evolved duplicates of M060L, M061L, M064L, and M065L from MYXV. Since 2017, 1616 rabbits and 82 hares were tested by a qPCR directed to M000.5L/R gene, conserved in MYXV and MYXV-Tol/ ha-MYXV strains. A subset (20%) of the positive samples was tested for the insert with MYXV being detected in rabbits and recombinant MYXV in hares. Recently, two wild rabbits found dead in South Portugal, showing skin oedema and pulmonary lesions tested positive for the 2.8 Kb insert. Sequencing showed 100% similarity with the insert sequences described in Iberian hares from Spain. Viral particles were observed in the lungs of both rabbits by electron microscopy, and isolation in RK13 cells showed virus infectivity. Despite the analysis of recombinant MYXV genomes may predict its ability to infect rabbit, routine analyses showed species segregation for the circulation of MYXV and recombinant MYXV in wild rabbit and in Iberian hares, respectively. This study demonstrates, however, that recombinant MYXV can effectively infect and cause myxomatosis in wild rabbits and domestic rabbits, which raises serious concerns for the future of the Iberian wild leporids and emphasizes the need to continue monitoring MYXV and recombinant MYXV in both species.

Urbanization has impacted biodiversity and ecosystems at a global scale. At the same time, it has been recognized as a driver of the gap between humans and nature. The lack of direct contact with nature can deteriorate several aspects of human wellbeing, and change knowledge and attitudes of people towards the environment. However, this phenomenon is still poorly understood in Megacities outside developed countries. Here, we explore the relationship between ecological knowledge and self-reported wellbeing in an important urban park in Santiago, Chile. We conducted semi-structured surveys to park users to explore their notions, preferences, ecological knowledge of plants and birds and self-reported wellbeing. Citizens associated urban parks mainly with “nature”, and particularly with the presence of trees and plants. Trees were recognized as the most relevant elements of urban parks, in turn, birds were ranked as the less relevant. Regarding ecological knowledge, respondents correctly identified an average of 2.01 plants and 2.44 birds out of a total of 10 for each taxon, and exotic species were more likely to be recognized. Park users also reported high scores for self-reported wellbeing. Interestingly, variance of self-reported wellbeing scores tended to increase at low levels of ecological knowledge of trees, but no significant relationship was detected with knowledge of birds, nor native species. These results suggest that parks can positively contribute to bring people closer to nature. Ecological knowledge was related to self-reported wellbeing. Improving ecological knowledge can be critical to restore the relationship between humans and nature in megacities.

Trees in residential landscapes provide many benefits, but can injure persons and damage property when they fail. In hurricane-prone regions like Florida, USA, the regular occurrence of hurricanes has provided an opportunity to assess factors that influence the likelihood of wind-induced tree failure and develop species failure profiles. We assessed open-grown trees in Naples, Florida, following the passage of Hurricane Irma in September 2017 to determine the effect of relevant factors on the degree of damage sustained by individual trees. Of 4,034 assessed individuals (n = 15 species), 74% sustained no damage, 4% sustained only minor damage (i.e., minimal corrective pruning needed), 6% sustained significant damage (i.e., major corrective pruning needed), and 15% were whole tree failures (i.e., overturned trees or trees requiring removal). The proportion of individuals in each damage category varied among species, stem diameter at 1.4 m above ground, and the presence of utility lines, which was a proxy for maintenance. We compared our results with the findings of seven previous hurricanes in the region to explore species’ resilience in hurricanes.

SARS-CoV-2 causes severe pneumonia epidemics and probably originated in horseshoe bats, but the intermediate host is unknown. The interaction of SARS-CoV-2 spike protein and its acceptor protein ACE2 is an important issue in determining viral host range and cross-species infection, while the binding capacity of Spike protein to ACE2 of different species is unknown. Here, we used the atomic structure model of SARS-CoV-2 and human ACE2 to assess the receptor utilization capacity of ACE2s from different species including cats, chimpanzees, dogs, cattles. Our results show, domestic cats (Felis catusc) and dogs (Canis lupus familiaris) are more susceptible to infection by SARS-CoV-2 and that they can efficiently transmit the virus to previously uninfected animals that are housed with them. Especially, cats could be a choice of animal model for screening antiviral drugs or vaccine candidates against SARS-CoV-2.

Interest in drone solutions in forestry applications is growing. Using drones, datasets can be captured flexibly and at high spatial and temporal resolutions when needed. In forestry applications, fundamental tasks include the detection of individual trees, tree species classification, bio-mass estimation, etc. Deep Neural Networks (DNN) have shown superior results when comparing with conventional machine learning methods such as Multi-Layer Perceptron (MLP) in cases of huge input data. The objective of this research was to investigate 3D convolutional neural networks (3D-CNN) to classify three major tree species in a boreal forest: pine, spruce, and birch. The proposed 3D-CNN models were employed to classify tree species in a test site in Finland. The classifiers were trained with a dataset of 3039 manually labelled trees. Then the accuracies were assessed by employing independent datasets of 803 records. To find the most efficient set of feature combination, we compare the performances of 3D-CNN models trained with hyperspectral (HS) channels, RGB channels, and canopy height model (CHM), separately and combined. It is demonstrated that the proposed 3D-CNN model with RGB and HS layers produces the highest classification accuracy. The producer accuracy of the best 3D-CNN classifier on the test dataset were 99.6%, 94.8%, and 97.4% for pines, spruces, and birches, respectively. The best 3D-CNN classifier produced ~5% better classification accuracy than the MLP with all layers. Our results suggest that the proposed method provides excellent classification results with acceptable performance metrics for HS datasets. Our results show that pine class was detectable in most layers. Spruce was most detectable in RGB data, while birch was most detectable in the HS layers. Furthermore, the RGB datasets provide acceptable results for many low-accuracy applications.

This article argues for five correctives to the current ethical debate about speciesism, and proposes normative, conceptual, methodological and experimental avenues to move this debate forward. Firstly, it clarifies the Principle of Equal Consideration of Interests and points out limitations of its scope. Secondly, it disambiguates between ‘favouritist’ and ‘species-relative’ views about moral treatment. Thirdly, it argues that not all moral intuitions about speciesism should be given equal weight. Fourthly, it emphasizes the importance of empirical research to corroborate statements about ‘folk speciesism’. Fifthly, it disambiguates between the moral significance of species and the moral status of their individual members. For each of these issues, it is shown that they have either been overlooked, or been given inapt treatment, in recent contributions to the debate. Building on the correctives, new directions are proposed for ethical inquiry into the moral relevance of species and species membership.

Freshwater fish represent half of all fish species and are the most threatened vertebrate group. Given their considerable passion and knowledge, aquarium hobbyists can play a vital role in their conservation. CARES is made up of many hobbyist organizations, whose purpose is to encourage aquarium hobbyists to the most endangered or extinct-in-the-wild freshwater fish to help ensure their survival. We found the CARES priority list contains nearly six hundred species from twenty families and two dozen species extinct-in-the-wild. The major families were typically the ones with largest hobbyist affiliations such as killifish, livebearers, and cichlids; which alone were half of CARES species. CARES contained every IUCN threatened species of Pseudomugilidae and Valenciidae, but only one percent of threatened Characidae, Cobitidae, and Gobiidae species. No Loricariidae in CARES were in the IUCN red list as they were not scientifically described. Tanzania and Mexico contained the largest amount of species, with the latter containing the most endemics. A large percent of species were classified differently than the IUCN, including a third of extinct-in-the-wild species classified as least concern by the IUCN. The vast disconnect exemplifies the importance of collaboration and information exchange required between hobbyists, the scientific community, and conservation organizations.

Macrobrachium rosenbergii is a valuable freshwater prawn in Asian aquaculture. In recent years, a new symptom that was generally called as ‘white head’ caused high mortality in M. rosenbergii farms in China. Samples of M. rosenbergii, M. nipponense, Procambarus clarkii, M. superbum, Penaeus vannamei, and Cladocera from a farm suffering from ‘white head’ in Jiangsu Province were collected and analyzed in this study. Pathogen detection showed that all samples were positive for Shrimp hemocyte iridescent virus (SHIV). Histopathological examination revealed dark eosinophilic inclusions and pyknosis in hematopoietic tissue, hepatopancreas and gills of M. rosenbergii and M. nipponense. Blue signals of in situ DIG-labeled LAMP (ISDL) appeared in hematopoietic tissue, hemocytes, hepatopancreatic sinus, and antennal gland. TEM of ultrathin sections showed a large number of SHIV particles with a mean diameter about 157.9 nm. The virogenic stromata and budding virions were observed in hematopoietic cells. Quantitative detection by TaqMan probe based real-time PCR of different tissues in natural infected M. rosenbergii showed that hematopoietic tissue contained the highest SHIV load with a relative abundance of (25.4±16.9)%. Hepatopancreas and muscle contained the lowest SHIV load with a relative abundance at (2.44±1.24)% and (2.44±2.16)%, respectively. Above results verified that SHIV is the pathogen causing ‘white head’ in M. rosenbergii, and M. nipponense and Pr. clarkii are also the susceptible species of SHIV.

It is known that eumelanin (EM) is photoprotective while pheomelanin (PM) is phototoxic. A recent study using a mouse model demonstrated that PM produces reactive oxygen species (ROS) that cause DNA damage and eventually lead to melanomagenesis. A biochemical study showed that PM possesses a pro-oxidant activity. PM consists of benzothiazine (BT) and benzothiazole (BZ) moieties, the former being transformed to the latter by heat or light. In this study, we compared the effects of ultraviolet A (UVA) irradiation using synthetic PMs with different BT to BZ ratios and using various coat color mouse hairs. We found that UVA irradiation of BZ-PM increased glutathione (GSH) consumption and generated more H₂O₂ than UVA irradiation of BT-PM. Non-irradiated controls did not exhibit strong pro-oxidant activities. Upon UVA irradiation, yellow mouse hairs oxidized GSH and produced H₂O₂ faster than black or albino mouse hairs. Next, to examine the mechanism of the pro-oxidant activity of BT-PM and BZ-PM, we examined the pro-oxidant activities of 7-(2-amino-2-carboxyethyl)-dihydro-1,4-benzothiazine-3-carboxylic acid (DHBTCA) and 6-(2-amino-2-carboxyethyl)-4-hydroxybenzothiazole (BZ-AA) as BT and BZ monomers, respectively. Their pro-oxidant activities were similar, but a large difference was seen in the effects of ROS scavengers, which suggests that the redox reactions may proceed via singlet oxygen in BZ-AA and via superoxide anions in DHBTCA. These results show that UVA enhances the pro-oxidant activity of PM, in particular BZ-PM.

Although the signaling function of Na/K ATPase is been studied for decades, the chasm between the pumping function and the signaling function of Na/K – ATPase is still an open issue. This article explores the relationship between ion pumping and signaling with attention to the amplification of oxidants through this signaling function. Starting with some experimental observations published by our laboratories and others, we develop some predictions regarding cellular oxidant stress.

Crotoxin (CTX), the predominant toxin in Crotalus durissus terrificus snake venom (CdtV), has anti-inflammatory and immunomodulatory effects. Despite its inhibitory action on neutrophil migration and phagocytosis, CTX does not directly affect the production of reactive oxygen species (ROS) by the neutrophils. In contrast, it enhances the generation of reactive oxygen and nitrogen intermediates by macrophages. Given the importance of macrophage-neutrophil interactions in innate antimicrobial defense, the aim of this study was to investigate the effect of CTX on neutrophil ROS production and killing activity, either through CTX-treated macrophage co-culture or conditioned medium of CTX-treated macrophages. The results showed an important modulatory action of CTX on the neutrophil function as well as neutrophil-macrophage interactions, as demonstrated by the increased production of hydrogen peroxide, hypochlorous acid, nitric oxide and TNF-α, along with the increased fungicidal activity of neutrophils.

The productivity of forests is often considered to be limited by the availability of phosphorus (P). Knowledge of the role of organic and inorganic P in humid subtropical forest soils is lacking. In this study, we used chemical fractionation and 31P nuclear magnetic resonance (NMR) spectroscopy to characterize the form of P and its distribution in undisturbed perhumid Chamaecyparis forest soils. The toposequence of transects was investigated for the humic layer from summit to footslope and lakeshore. The clay layer combined with a placic-like horizon in the subsoil may affect the distribution of soil P because both total P and organic P (Po) contents in all studied soils decreased with soil depth. In addition, Po content was negatively correlated with soil crystalline Fe oxide content, whereas inorganic P (Pi) content was positively correlated with soil crystalline Fe oxide content and slightly increased with soil depth. Thus, Pi may be mostly adsorbed by soil crystalline Fe oxides in the soils. Among all extractable P fractions, the NaOH-Po fraction appeared to be the major component, followed by NaHCO3-Po; the resin-P and HCl-Pi fractions were lowest. In addition, we found no typical trend for Pi and Po contents in soils with topographical change among the three sites. From the 31P-NMR spectra, the dominant Po form in soils from all study sites was monoesters with similar spectra. The 31P-NMR findings were basically consistent with those from chemical extraction. Soil formation processes may be the critical factor affecting the distribution of soil P. High precipitation and year-round high humidity may be important in the differentiation of the P species in this landscape.

This study retrospectively evaluated the prevalence and drug susceptibility of Candida species in patients diagnosed with oral mucosal candidiasis at the Department of Periodontal and Oral Mucosal Diseases, Silesian Medical University, from 2018 to 2022. A total of 1286 patients were examined (788 females, 498 males) through mycological testing to identify Candida strains and assess their drug resistance to amphotericin B, nystatin, flucytosine, econazole, ketoconazole, miconazole, and fluconazole. Among the 958 patients with positive results, Candida albicans accounted for 66.79% of infections, followed by non-albicans Candida (NAC) strains (33.21%), with rare strains identified in 0.82%. Drug resistance in C. albicans was less than 10% for most antifungals but reached 30% for azoles, while NAC strains exhibited more than 40% resistance, with C. krusei being the most resistant. Key risk factors for candidiasis included removable denture use and uncontrolled diabetes mellitus. The study highlights an increase in NAC infections, multi-strain infections, and decreasing efficacy of antifungal treatments, especially for NAC strains, with more severe cases observed in patients with multiple risk factors.

The fishery biology of the invasive Atlantic blue crab Callinectes sapidus in the Mediterranean Sea outside the eastern sectors of the basin has been only recently investigated. Here we studied the population of C. sapidus in the Lesina Lagoon (Adriatic Sea, SE Italy). In total, 838 crabs were captured monthly between February 2021 and January 2022 using fyke nets. Abundances varied seasonally with catches per unit effort ranging between 0 and 1.76 crabs fyke nets-1 d-1 in winter and summer. Spatial abundances estimated in summer by a Carle-Strub procedure ranged between 0.06 and 0.64 crabs m-2. Sex ratio (♂/♀) was close to 1:1; males prevailed only in August and September; ovigerous females occurred from April to August. Males’ size at morphological maturity was smaller than females (110.6-112.3 mm vs. 122.1-123.1 mm). Seasonal von Bertalanffy growth parameters indicated that, compared with males, females showed a shorter maximum lifespan (5 vs. 8 years), a higher growth coefficient K (0.6 vs. 0.4 y-1) and growth performance index ' (4.6 vs. 4.3), while maximum sizes CW∞ (237.8 vs. 232.6 mm) and seasonality indices C (0.62 vs. 0.57) were similar. Furthermore, females showed higher natural and fishing mortalities and exploitation rate. We discussed the results of the present study in the context of the available literature to provide a valuable basis for the implementation of standardised Mediterranean-scale management plans, matching exploitation of C. sapidus with sustainable conservation of coastal ecosystems.

The injury causes resistance in human gastric cancer (GC). This Adverse Outcome Pathway (AOP) entitled “Increases in cellular reactive oxygen species (ROS) and chronic ROS leading to human treatment-resistant gastric cancer (GC)” consists of molecular initiating event (MIE) as “Increases in cellular ROS” and “Chronic ROS”, followed by series of key events (KEs); “porcupine-induced Wnt secretion and Wnt signaling activation”, “beta-catenin activation”, “epithelial-mesenchymal transition (EMT), and adverse outcome (AO) as “human treatment-resistant GC” in the sequence. ROS has multiple roles in disease such as development and progression of cancer, or apoptotic induction causing anti-tumor effects. In this AOP, we focus on the role of sustained levels of chronic ROS to induce the therapy-resistance in human GC. EMT, which is cellular phenotypic change from epithelial to mesenchymal-like features, demonstrates cancer stem cell (CSC)-like characteristics in human GC. EMT is induced by Wnt/beta-catenin signaling, providing the rationale to have Wnt secretion and beta-catenin activation as KEs in the AOP.
