Subcortical beetles interact with a wide range of semiochemicals released from different sources, including trees, fungi, and insect pheromones. While the attraction of bark beetles, their insect predators, and competitors to bark beetle pheromones is well-studied, the attraction of these beetle communities to other sources of semiochemicals remains poorly understood. We tested the attraction of bark and wood-boring beetles and their predators to host stress volatiles, fungal volatiles, and a mountain pine beetle lure in the field. Host stress volatiles were derived from lodgepole pine trees stressed by three fungal symbionts of mountain pine beetle and two common phytopathogens. Our results showed that bark beetles, particularly mountain pine beetles, show a preference for a combination of fungal volatiles, particularly 2-methyl-1-butanol and mountain pine beetle lures. Without the addition of lures, 2-methyl-1-butanol was also identified as a key fungal volatile in the attraction of mountain pine beetle competitors from the Cerambycidae and Buprestidae families. Predators in the Elateridae and Staphylinidae families showed attraction to host stress volatiles and the healthy tree volatile profile. These findings suggest that these chemicals warrant further field testing for potential use in monitoring and management of subcortical beetle populations.

Background: The COVID-19 pandemic, caused by the novel coronavirus SARS-CoV-2 has posed unprecedented challenges to healthcare systems worldwide. Here, we have identified proteomic and genetic signatures for improved prognosis which is vital for COVID-19 research. Methods: We investigated the proteomic and genomic profile of COVID-19 positive patients (n=400 for proteomics, n=483 for genomics), focusing on differential regulation between hospitalised and non-hospitalised COVID-19 patients. Signatures had their predictive capabilities tested using independent machine learning models such as Support Vector Machine (SVM), Random Forest (RF) and Logistic Regression (LR). Results: This study has identified 224 differentially expressed proteins in hospitalised COVID-19 patients compared to non-hospitalised COVID-19 patients, involved in various inflammatory and immunological pathways. LGALS9 (p-value < 0.001), LAMP3 (p-value < 0.001), PRSS8 (p-value < 0.001) and AGRN (p-value < 0.001), were identified as the most statistically significant proteins. Several hundred rsIDs were queried across the top 10 significant signatures, identifying three significant SNPs on the FSTL3 gene showing correlation with hospitalisation status. Conclusion: Our study has not only identified key signatures of COVID-19 patients with worsened health but has also demonstrated their predictive capabilities as potential biomarkers, which suggests a staple role in the worsened health effects caused by COVID-19.

Sour waters are one of the main aqueous byproducts generated during petroleum refining and require processing in Sour Water Treatment Units (SWTUs) to remove contaminants such as H₂S and NH₃ in compliance with environmental legislations. Therefore, monitoring the composition of SWTU effluents, including acid gas, ammoniacal gas, and treated water, is essential. This study aims to present an AI (artificial intelligence) hybrid-based methodology to develop soft sensors capable of real-time prediction of H₂S and NH₃ mass fractions in the effluents of SWTUs and validate them using real data from industrial units. Initially, a new database based on the dynamic simulation of a two stripping columns SWTU phenomenological model, developed in Aspen Plus Dynamics® V10, was generated, aiming at non-faulty runs, unlike our previous work [1]. Ensemble methods (Decision Trees), such as Gradient Boosting and Random Forest, and Support Vector Machines were compared for soft sensor creation using these simulated data. The best outcome was the development of six soft sensors based on Random Forest with R² greater than 0.87, MAE less than 0.12, MSE less than 0.17, and RMSE less than 0.41. Variable importance analysis revealed that the temperature of the second stage of Column 1 significantly influences the thermodynamic equilibrium of H₂S and NH₃ separation from sour waters, being critical for five of the six soft sensors. After this initial stage using data from the phenomenological model, data from an industrial-scale SWTU were used to develop real soft sensors. The results proved the effectiveness of the conjugated use of physical model and industrial data approach in the development of soft sensor for two-column SWTUs.

Studies of Phytophthora impacts in forests generally focus on individual species without recogni-tion that Phytophthora occur in multispecies communities. We investigated community structure of Phytophthora species in the rhizosphere of Agathis australis (kauri) in Te Wao Nui o Tiriwa / Waitākere Ranges, New Zealand, in the context of kauri dieback disease expression. Soil sampling and tree health monitoring was conducted on 767 randomly selected mature kauri trees. Phy-tophthora species were detected using both soil baiting and DNA metabarcoding of eDNA. Four species were detected with soil baiting (P. agathidicida, P. cinnamomi, P. multivora, and P. pseudo-cryptogea) and an additional three species with metabarcoding (P. kernoviae, P. cactorum/P. aleatoria and an unknown clade 7 species). Phytophthora cinnamomi was the most abundant species and was distributed throughout the forest. Both P. multivora and P. agathidicida were limited to forest edges, suggesting more recent arrivals. P. agathidicida presence was strongly correlated with declining canopy health, confirming its role as the main river of kauri dieback. The limited distribution of P. agathidicida and infrequent detections (11.0% samples) suggests that that this species is spreading as an introduced invasive pathogen and provide hope that with strategic management uninfected areas of the forest can be protected. The frequent detections of P.cinnamomi and P. multivora, from symptomatic trees in the absence of P. agathidicida suggest more research is needed to understand their roles in kauri forest health.

The Australian tropical montane cloud forest (TMCF) is home to approximately 18 percent of the nation’s total vascular plant species. Over the past century, human activity and industrial development have caused global climate changes, posing a severe and irreversible danger to the entire land-based ecosystem, and WTWHA is no exception. The current average annual temperature of WTWHA in FNQ is 24 °C. However, in the coming years (by 2030), the average annual temperature increase is estimated to be between 0.5 and 1.4 °C compared to the climate observed between 1986 and 2005. Looking further ahead to 2070, the anticipated temperature rise is projected to be between 1.0 and 3.2 °C, with the exact range depending on future emissions. We identified 84 plant species endemic to the WTWHA in FNQ, which are already experiencing climate change threats. Few of these plants are used in herbal medicines. This study comprehensively reviewed metabolomics studies conducted on these 84 plant species until now toward understanding their physiological and metabolomics responses to global climate change. This review also discusses: i) recent developments in plant metabolomics studies that can be applied to study and understand the interactions of wet tropical plants with climatic stress, ii) medicinal plants and isolated phytochemicals with structural diversity, and iii) reported biological activities of crude extracts and isolated compounds.

The importance of the national security of supply has been emphasized in recent years due to the COVID-19 pandemic and changes in operating environments. Thanks to autonomy and resiliency in fuel procurement, organizations with decentralized energy plants may be able to quickly adapt to the changing needs of society. The main aim of this study was to increase the sustainability of a Finnish CHP plant's wood procurement by determining the company's profitable wood procurement region while keeping the forests renewable. The income received from the sale of energy and the costs incurred from the procurement of wood were collected from the ERP system as input data for the developed DSS. The environmental sustainability of wood procurement was assessed by considering the additional costs caused through the EEAP mechanism if the carbon sink of the forest worsened due to wood procurement in the area. The supply chains were compared so that the CHP plant managers could adopt the best energy wood value chains. The results showed that updating the procurement strategy would be necessary. The CHP plant managers should purchase energy wood from a larger procurement area than the current one and focus on roadside purchases of wood. The Techno-Economic Wood Payment Ability Model was a useful part of the DSS to guide wood buyers toward sustainable wood procurement by increasing the share of renewable forests.

The author attempts to reconstruct the climatic vicissitudes of the Mediterranean to explain why only broadly distributed Eurasian forest species have penetrated the Mediterranean peninsulas. He has carried out a bibliographic survey of the status of breeding forest birds in Italy over 15 decades (1872-2022) in order to establish an objective long-term trend (stable, increasing, decreasing, etc.). The number of breeding forest birds in Italy amounts to 61; their distribution, with a few exceptions, indicates that they are widespread in Eurasia, but only a small percentage of Eurasian forest species have colonized Italy and the other Mediterranean peninsulas, namely 49 in Iberian, 61 in Italian and 64 in Balkan peninsulas; a small percentage of them (between 10.9 and 14.3%) belongs to trans-Saharan migrants. The similarity between the forest species on the three peninsulas (Iberian, Italian and Balkan) results between 0.45 and 0.48, indicating a certain differ-ence in the overall fauna on the three territories. Not all species have penetrated southwards into the three peninsulas; for example, some that stopped in the Italian Alps have instead arrived to the forests of Greece, at a latitude corresponding to southern Italy, or species that in Italy stopped in the northern Apennines in the other two peninsulas have instead arrived far south. Iberian peninsula and the island of Corsica hold three endemic species among breeding forest birds, Italian and Balkan peninsulas have not endemic species. Overall, the Mediterranean presently hosts mainly neo-endemic taxa among forest bird species; the only paleo-endemics can be considered the three species of nuthatches living in Corsica, Algeria, and Turkey (other than Caucasus and the islet of Lesvos) and Le Vaillant’s woodpecker in the Maghreb (North Africa). Italian forests cover ca. 40% of land surface and since 1980’ are increasing, but 22% of them do not have a natural origin. However, it is difficult to know the true increase of forests, because some of them are fired every year. The presence of some ecologically demanding forest birds depends on the age of the trees, permanent open spaces and other characteristics at the edge of woodland.

This research aimed to improve the interpretation of electrical resistivity (ER) results in concrete bridge decks by utilizing results from multiple nondestructive evaluations (NDE) techniques in machine learning algorithms. To achieve this, a parametric study was first conducted using numerical simulations to investigate the effect of various parameters on ER measurements, such as the degree of saturation, corrosion length, delamination depth, concrete cover, and the moisture condition of delamination. A data set from this study was used to build a machine-learning algorithm based on the Random Forest methodology. This algorithm was then implemented on data collected from a bridge deck in the BEAST® facility. The presented results demonstrate an improvement in the interpretation of ER measurements using data from other NDE technologies.

The concept of ecosystem services (ESs) provides a valuable approach to communicate societies' dependence on natural ecosystems, assisting in the evaluation of measures to protect vegetation in watersheds that promote water-related ESs. Vegetation cover and land use are important factors related to the capacity of ecosystems to provide soil loss regulation and sediment retention services, which are highly relevant for sediment management in watersheds draining into reservoirs with multiple water uses. This study aims to assess the relationship between potential changes in land use and land cover and the sediment retention service provided by riparian vegetation in a watershed draining into a hydroelectric reservoir in the Brazilian Cerrado. Sediment load was selected as a physical variable that affects different water uses in reservoirs, with these uses being dependent on the quality or quantity of stored water. Scenarios were analyzed to represent landscape reconfigurations from a baseline condition in order to account for the recovery of riparian vegetation by the agricultural sector, as foreseen in the Brazilian Forest Code. To map and quantify variations in annual sediment retention, the InVEST model was used, which allows estimating an integrated response of the drainage basin to large-scale land use changes over time and space. The results show that for the drainage basin of the Corumbá IV Hydroelectric Power Plant (HPP), the recovery of riparian areas occupied by annual crops has the potential to generate greater benefits to water use in the reservoir. In the studied catchment area, 14% of the total vegetation deficit in riparian areas is occupied by crops, while 86% is occupied by pastures. This highlights the buffering role of riparian vegetation as an effective practice for managing sediment flows originating from sheet erosion in agricultural landscapes. The methodology employed in this study allows for the quantification and mapping of the effects of soil and water conservation measures in river basins. This enables the prioritization of sectoral efforts for the restoration of native vegetation, considering the highest returns in benefits perceived by water users affected by sediment input in reservoirs. The study's results reinforce the importance of conserving vegetation in riparian areas and their surroundings for sediment retention, highlighting the role of these areas as assets in providing water-related ecosystem services.

The owners of forests with water protection functions in Bulgaria do not receive compensation for their limited right to use wood and non-wood forest products from the forest areas they own. At the same time, the contribution derived from forests with water protection functions is received by water users and water consumers, whereas the costs of managing the forests are borne by their owners. The problem thus defined is not a forestry one, but an economic one, and the purpose of this paper ensues therefrom, namely to propose and test a methodology for valuating the produc-tion function of forest areas with water protection function in Bulgaria, allowing a fair distribu-tion of income between the forest owner and the user of forest ecosystem services. The methodol-ogy is based on the form of forest management and the analytical expression of the economic rela-tionship between a forest owner and a user of forest ecosystem services, constructed using Schen-rock’s formula. It has been tested with actual data on forest areas with water protection functions falling within the administrative and territorial scope of Velingrad municipality and based on the obtained results proposals have been made for the distribution of the contributions generated by forest ecosystems between the forest owner and the user of the forest ecosystem services.

Cumulative effects of landscape disturbance in forested source water regions can alter the storage of fine sediment and associated phosphorus in riverbeds, shift nutrient dynamics and degrade water quality. Here, we examine longitudinal changes in major element chemistry and particulate phosphorus (PP) fractions of river-bed sediment in an oligotrophic river during environmentally sensitive low flow conditions. Study sites along 50 km of the Crowsnest River were located below tributary inflows from sub-watersheds and represent a gradient of increasing cumulative sedi-ment pressures across a range of land disturbance types (harvesting, wildfire, and municipal wastewater discharges). Major elements (Si2O, Al2O3, Fe2O3, MnO, CaO, MgO, Na2O, K2O, Ti2O, V2O5, P2O5), loss on ignition (LOI), PP fractions (NH4CI-RP, BD-RP, NaOH-RP, HCI-RP and NaOH(85)-RP) and absolute particle size were evaluated for sediments collected in 2016 and 2017. While total PP concentrations were similar across all sites, bioavailable PP fractions (BD-RP, NaOH-RP) increased downstream with increased concentrations of Al2O3 and MnO and levels of landscape disturbance. This study highlights the longitudinal water quality impacts of increasing landscape disturbance on bioavailable PP in fine riverbed sediments and shows how the convergence of climate (wildfire) and anthropogenic (sewage effluent, harvesting, agriculture) drivers can produce legacy effects on nutrients.

Emerald ash borer (EAB), Agrilus planipennis, is an invasive beetle of East Asian origin that in North America and Russia killed millions of ash trees (Fraxinus spp.). In September 2020, EAB was de-tected in Saint Petersburg, becoming resonant event for the metropolitan city. The aim of the present study was to investigate occurrence and ecology of EAB in Saint Petersburg. The presence of two distinct enclave populations of EAB was revealed, each of which has (very likely) been established by separate events of “hitchhiking” transport vehicles. Following the invasion, further spread of EAB in Saint Petersburg was slow and locally restricted, main explanation for which is climatic factor. Due to spread by “hitchhiking”, the possibility of EAB further long-distance ge-ographic spread of EAB in the Baltic Sea region (EU) is high, and not only by ground transport (120–130 km distance from EU borders), but also by ferries transporting cars (traditional means of transportation across the Baltic Sea). In certain cases, development of EAB on F. excelsior was more successful (stem portion colonized, larval densities, number of galleries, exit holes, viable larvae, emerged beetles) than in (adjacent) F. pennsylvanica trees. Observed relatively high EAB-sensitivity of F. excelsior therefore questions the efficacy and benefits of the currently ongoing selection and breeding projects against ash dieback (ADB), caused by Hymenoscyphus fraxineus. Inventory, mapping, and monitoring of surviving F. excelsior trees in areas infested by both ADB and EAB are necessary to acquire genetic resource for work on strategic long-term restoration of F. excelsior, tackling (inevitable) invasion of EAB to the EU.

Machine learning (ML) has a large capacity to learn and analyze a large volume of data. This study aimed to train different algorithms to discriminate between healthy and pathologic corneal images by evaluating digitally processed spectral-domain optical coherence tomography (SD-OCT) corneal images. A set of 22 SD-OCT images belonging to a random set of corneal pathologies was compared to 71 healthy corneas (control group). A binary classification method was applied; three approaches of ML were used. Once all images were analyzed, representative areas from every digital image were also processed and analyzed for a statistical feature comparison between healthy and pathologic corneas. The best performance was obtained from transfer learning - support vector machine (TL-SVM) (AUROC = 0.94, SPE 88%, SEN 100%) and transfer learning – random forest (TL- RF) method (AUROC = 0.92, SPE 84%, SEN 100%), followed by convolutional neural network (CNN) (AUROC = 0.84, SPE 77%, SEN 91%) and random forest (AUROC = 0.77, SPE 60%, SEN 95%). The highest diagnostic accuracy in classifying corneal images was achieved with the TL-SVM and the TL-RF models. In image classification, CNN was a strong predictor. This pilot experimental study developed a systematic mechanized system to discern pathologic from healthy corneas.

Recent advancements in Software Defined Networking (SDN) makes it possible to overcome the management challenges of traditional network by logically centralizing control plane and decoupling it from forwarding plane. Through centralized controllers, SDN can prevent security breach, but it also brings in new threats and vulnerabilities. Central controller can be a single point of failure. Hence, flow-based anomaly detection system in OpenFlow Controller can secure SDN to a great extent. In this paper, we investigated two different approaches of flow-based intrusion detection system in OpenFlow Controller. The first of which is based on machine-learning algorithm where NSL-KDD dataset with feature selection ensures the accuracy of 82% with Random Forest classifier using Gain Ratio feature selection evaluator. In the later phase, the second approach is combined with Gated Recurrent Unit Long Short-Term Memory based intrusion detection model based on Deep Neural Network (DNN) where we applied an appropriate ANOVA F-Test and Recursive Feature Elimination feature selection method to improve the classifier performance and achieved an accuracy of 88%. Substantial experiments with comparative analysis clearly show that, deep learning would be a better choice for intrusion detection in OpenFlow Controller.

Farm activities continued sand winning operations and the allocation of plots of land to prospective developers in Ghana pose a serious threat to the forest covers and lifespan of the Forest and game reserves. With all the positive add ups to the country from forests, Ghana has lost more than 33.7%(equivalent to 2,500,000 hectares) of its forest, since the early 1990s between 2005 and 2010, the rate of deforestation in Ghana was estimated at 2.19% per annum; the sixth highest deforestation rate globally for that period. This shows how important forest monitoring can be to the forestry commission in Ghana. Despite the frameworks which have been developed to help Ghana to protect and restore its forest resources, inadequate monitoring systems remain a barrier to effective implementation. In this study, Google earth engine was used to map and analyze the structural changes of forest cover using JavaScript to query and compute Landsat, MODIS and NOAA AVHRR satellite imageries of the study area (Ghana) with spatial resolutions 30m, 250m and 7km respectively. A supervised classification was performed on three multi-temporal satellite imageries and a total of six major land use and land cover classes were identified and mapped. By using random Forest-classification technique, from 1985 to 2018 recorded by NOAA AVHRR, forest cover has decreased by 66% and 2000 to 2018 recorded by Landsat and MODIS 61% and 47% respectively. A decrease in the forest has been as a result of anthropogenic activities in Ghana. A change detection analysis was performed on these images and it was noted that Ghana is losing forest reserves in every 5years. Overlay of the reserved forest of the 2000 and the classified map of 2018 shows vegetation changed during 2000-2018 remarkably. Therefore, forest-related institutions like the Forestry Commission can employ and use this monitoring system on Google Earth Engine for processing satellite images particularly Landsat, MODIS and NOAA AVHRR for forest cover monitoring and analysis for fast, efficient and reliable results.

The research objectives was to determine the plants vegetation profile in forest revegetation and Mount Arjuna springs. The method used was descriptive method. While based on the techniques and tools used to research, the author uses the survey method to obtain facts that occur in the research area, namely in the area around the springs of Arjuna mount, Pasuruan, East Java, Indonesia. Data obtained in the field are then processed and analyzed using GIS, ArcView 3.3 and Google Earth programs. The results of the study explain the profile of plant vegetation, namely plant stratification, bird wealth, taxonomic wealth and plant density have a significant effect on forest revegetation and springs in Arjuna mount. The forest vegetation profile has a positive and significant effect on the preservation of springs. The better the profile of forest vegetation, the preservation of springs will be better, and vice versa.

Results-Based Funding (RBF) for Reducing Emissions from Deforestation and Forest Degradation (REDD+) has become an important instrument for channeling financial resources to forest conservation activities. At the same time, much literature on conservation funding is ambiguous about the effectiveness of existing RBF schemes. Many effectiveness evaluations follow a simplified version of the principal-agent model, although in practice the relation between aid providers and funding recipients is much more complex. As a consequence, intermediary steps of conservation funding are often not accounted for effectiveness studies. This research paper aims to provide a nuanced understanding of conservation funding by analyzing the allocation of financial resources for one of the largest RBF schemes for REDD+ in the world: the Brazilian Amazon Fund. As part of this analysis, this study has built a dataset of information on Amazon Fund projects at unprecedented detail in order to accurately reconstruct the allocation of financial resources across different stakeholders (i.e. governments, NGOs, research institutions), geographies and activities. The results show that stakeholders seem to hold preferences with respect to the type of activities that they support, thereby suggesting that project owners exert much influence on how deforestation reduction is to be attained. There are evidences that governmental organizations lack financial additionality of their projects, which renders the growing share of funding to this type of stakeholder particularly worrisome. By contrast, the geographical distribution of financial resources seemed to follow a more focused rationale as financial support tends to concentrate in areas where deforestation threats are highest. Overall, the allocation of the financial resources from the Amazon Fund reflects an arbitrary support of different projects that adopt very diverging theories of change that are not primarily concerned with attaining further deforestation reductions. As projects owners exert influence on funding effectiveness to some extent, the Amazon Fund may either seek to regulate the allocation of financial resources more actively or adopt funding effectiveness evaluations that account for this influence more comprehensively.

In recent years, online social networks have received important consideration in spatial modelling fields given the critical information that can be extracted from them for events in real time; one of the most latent issues is that regarding various natural disasters such as earthquakes. Although it is possible to retrieve data from these social networks with embedded geographic information provided by GPS, in many cases this is not possible. An alternative solution is to reconstruct specific locations using probabilistic language models, more specifically those based on Name Entity Recognition (NER), which extracts names from a user’s description about an event occurring in a specific place (e.g., a collapsed building on a specific avenue). In this work, we present a methodology to use twitter as a social sensor system for disasters. The methodology scores NER locations with a kernel density estimation function for different subtopics originating from a natural disaster and that maps them into a geographic space is proposed. The proposed methodology is evaluated with tweets related to the 2017 earthquake in Mexico.

Mining for resources extraction may lead to several geological and associated environmental changes due to ground movements, collision with mining cavities and deformation of aquifers. Geological changes may continue in a reclaimed mine area, and the deformed aquifers may entail a breakdown of substrates and an increase in ground water tables, which may cause surface area inundation. Consequently, a reclaimed mine area may experience surface area collapse, i.e. subsidence, and degradation of vegetation health. Thus, monitoring short-term landscape dynamics in a reclaimed mine area may provide important information on the long-term geological and environmental impacts of mining activities. We studied landscape dynamics in Kirchheller Heide, Germany, which experienced extensive soil movement due to longwall mining without stowing, using Landsat imageries between 2013 and 2016. A Random Forest image classification technique was applied to analyse land-use and land-cover dynamics and the growth of wetland areas was assessed using a Spectral Mixture Analysis (SMA). We also analyzed the changes in vegetation health using a Normalized Difference Vegetation Index (NDVI). We observed a 19.9% growth of wetland area within the four years with 87.2% of growth in the coverage of two major waterbodies in the reclaimed mine area. NDVI values indicate that 66.5% of the vegetation of the study area was degraded due to changes in ground water tables and surface flooding. Our results inform environmental management and mining reclamation authorities about the subsidence spots and priority mitigation areas from land surface and vegetation degradation in Kirchheller Heide.

Understory plants are important components of forest ecosystem productivity and diversity. Compared to biomass models of overstory canopy trees, few are available for understory saplings and shrubs and therefore their roles in estimation of forest carbon pools are often ignored. In this study, we harvested 24 understory species including 4 saplings, 9 tree-like shrubs and 11 typical shrubs in coniferous and broadleaved mixed forest in northeastern China and developed the best fit allometric equations of above- and below-ground and total biomass by species-specific or multispecies using morphological measurements of basal diameter, height and crown area as independent variables. The result showed that single basal diameter, height or crown area had good explanatory power for both species-specific and multispecies (p<0.001). The best-fit models included only basal diameter in sapling and tree-like shrubs, and combinations of crown area, height, and basal diameter in typical shrubs. The logarithmic model was most desired among the 4 model forms of linear, quadratic, multiple linear and logarithmic, for species-specific and multispecies. The models we developed should help the estimation of forest ecosystem carbon stocks, especially for belowground component, and provide tools for quantification of individual species biomass of understory plants.

Scientific knowledge evolves in small steps, with occasional backsteps to correct inaccuracies, all occurring within a competitive environment. This perspective for the first time looks at the history of latency-related RNA (LR-RNA) that was later renamed latency-associated transcript (LAT). At the 1986 International Herpesvirus Workshop (IHW) meeting in Leeds, England, Daniel L Rock and Anthony B Nesburn first reported the discovery of human herpes virus 1 (HSV-1) latency-related (LR)-RNA that is antisense to ICP0. Less than a month after the IHW meeting a paper was submitted to Science magazine and 8 months later appeared in print thanking “D. Rock for suggesting RNA complementary to the ICP0 message may be present in latently infected cells”. This perspective is not a review of LAT literature but intends to clarify the timeline of LAT discovery and subsequent breakthroughs such as reactivation, apoptosis, CD8+ T cell exhaustion, and LAT expression in different cell types detected during latency. While many review articles have been written about LAT since 1987, the most comprehensive and balanced review about LAT was written by Dr. David Bloom’s group (1). In this overview, I will discuss our original collaboration with Dr. Dan Rock and subsequent work that our group performed, which is still ongoing. Finally, I will discuss the controversies associated with LAT from its inception to current times.

Adhesive-invasive E. coli have been suggested to be associated with the development of Crohn's disease (CD). It is assumed that they can provoke the onset of the inflammatory process as a result of the invasion of intestinal epithelial cells, and then, due to survival inside macrophages, stimulate chronic inflammation and the development of granuloma. In previous reports, we have shown that passage of the СD isolate ZvL2 on minimal medium M9 supplemented with sodium propionate (PA) as a carbon source stimulates, and with glucose - inhibits the adherent-invasive properties and the ability to survive in macrophages. This effect was reversible and not observed for the laboratory strain K12 MG1655. We were able to compare the isogenic strain AIEС in two phenotypes - virulent (ZvL2-PA) and non-virulent (ZvL2-GLU). We showed that, unlike ZvL2-GLU, the ZvL2-PA activates the production of ROS and cytokines when interacting with neutrophils. The laboratory strain does not cause a similar effect. Differences in neutrophil NADH-oxidase activation and ζ potential for ZvL2-GLU and ZvL2-PA are associated with changes in the abundance of membrane proteins. Using differential 2D electrophoresis and LC-MS, we identified membrane proteins that may be associated with the activation of the virulence properties of the СD isolate ZvL2.

Various vaccines have been developed in response to the SARS-CoV-2 pandemic, and the safety of vaccines has become an important issue. COVID-19 vaccines-related central nervous system inflammatory demyelinating diseases (CNS IDDs) have been reported recently. We present one case of AstraZeneca vaccine-related myelin oligodendrocyte glycoprotein (MOG) antibody-associated disease and literature review another 78 patients published from January 2020 to October 2022. Patients were divided into three vaccine groups (viral vector, mRNA and inactivated vaccines) for further analyses. Among 79 patients with COVID-19 vaccines-related CNS IDDs, 49 (62%) cases received viral vector vaccines, 20 (25.3%) received mRNA vaccines and 10 (12.7%) received inactivated vaccines. Twenty-seven cases (34.2%) were confirmed with autoantibodies, including 15 patients (19%) with anti-MOG, 11 (13.9%) with anti-aquaporin 4 (AQP4), and one (1.3%) with both antibodies. Significantly, more males developed CNS IDDs post viral vector vaccines compared to mRNA and inactivated vaccines. Patients receiving mRNA vaccines were older than other groups. Furthermore, mRNA and inactivated vaccines correlated more with anti-AQP4 antibodies, while viral vector vaccines showed higher MOG positivity. The research suggests potential associations between COVID-19 vaccines-related CNS IDDs and gender, age, and autoantibodies, contingent on vaccine types. Protein sequence analysis implies similarities between the S protein and AQP4/MOG. Further studies may elucidate the mechanisms of CNS IDDs, aiding vaccine selection for specific groups.

This prospective study provides data on long-term humoral immunogenicity of a heterologous off-label vaccine regimen combining the adenoviral vectored ChAdOx1 nCoV-19 from Astra-Zeneca (ChAd) with the mRNA-1273 vaccine from Moderna (m1273) in comparison to two different homologous mRNA vaccine schedules. Of the 316 COVID-19 naïve adult health care workers (HCW) included to complete a survey on vaccine-associated symptoms (VAS), 197 had received the homologous BNT162b2 mRNA vaccine from Pfizer/BioNTech (BNT/BNT), 76 the homologous m1273/m1273, and 43 the heterologous ChAd/m1273 vaccine regimen. Concentration of antibodies against SARS-CoV-2 spike protein in plasma 5-7 months after the second vaccine dose was higher in the m1273/m1273 and ChAd/m1273 than the BNT/BNT vaccine group. The frequency of systemic VAS after first vaccine dose was 86% after ChAd compared to 35% and 39% after BNT and m1273, respectively (p &lt; 0.0001), and after second vaccine dose highest (89%) in the m1273/m1273 group (p &lt; 0.001). Individuals with systemic VAS achieved higher levels of antibodies irrespective of vaccine regimen. In conclusion, VAS serve as a strong predictor of long-term humoral immune response, and the heterologous ChAd/m1273 vaccine regimen provides an at least equal long-term humoral immune response compared with the standard vaccine regimens used in Denmark.

This study explores the classification of seven registered dry bean species using a dataset of 13,611 grain samples characterized by 16 features, including both dimensional and shape measurements. The primary objective was to develop robust Machine Learning models to accurately identify species while addressing challenges related to class imbalance and feature redundancy. A series of classification algorithms, including Decision Trees (DT), Random Forests (RF), k-Nearest Neighbors (KNN), Support Vector Machines (SVM), and Multi-Layer Perceptrons (MLP), were applied to evaluate their performance under various data conditions. Notably, the MLP model exhibited superior performance due to its capacity to capture complex patterns in high-dimensional data, achieving the highest classification accuracy. Random Forest models showed resilience to class imbalance, though misclassifications between similar species highlighted the need for improved feature selection. The results indicate that effective feature engineering and careful model tuning are crucial for enhancing classification accuracy in agricultural applications. The study concludes that future work should aim to expand the dataset and explore advanced data augmentation techniques to further improve model robustness and applicability in food security contexts. This study utilizes a combination of optical PlanetScope imagery and Synthetic Aperture Radar (SAR) data from Sentinel-1 to investigate land cover changes in Dili, Timor-Leste, following Tropical Cyclone Seroja. The primary objective is to evaluate how integrating these datasets enhances disaster monitoring and response in flood-affected areas. The Random Forest classifier, known for its robustness in handling high-dimensional and noisy data, was applied to classify land cover into six distinct classes: vegetation, water, built-up areas, bare soil, clouds, and shadows. The classification was performed across three phases—pre-disaster, post-disaster, and recovery—using Google Earth Engine (GEE). Binary segmentation using Otsu thresholding was applied to the SAR images to refine the classification by delineating water bodies from non-water areas. The study achieved high classification accuracy, with overall accuracy ranging from 97.2% to 98.7%, and the Kappa index, which measures agreement between classified and reference data, remained consistently strong, indicating reliable model performance. Key results include a significant increase in water bodies and extensive damage to vegetation and built-up areas during the post-disaster phase, followed by partial recovery in the subsequent period. Despite the high accuracy, urban areas posed classification challenges due to misclassification between built-up and bare soil categories. This research offers a methodological framework for integrating optical and SAR data with machine learning for land cover change detection in post-disaster scenarios. The societal benefits of this study include improved disaster preparedness, enhanced recovery planning, and valuable insights into flood impact mitigation in regions like Timor-Leste, where technical and data limitations persist.

The land use land cover (LULC) map is extensively employed for different purposes. The LULC maps provided by the Portuguese National Geographic Information System (SNIG) are not ideal for continuous assessment and regular scrutiny. Machine learning (ML) algorithms applied in remote sensing (RS) data has been proven effective in image classification, object detection, and semantic segmentation. Previous studies have shown that Random Forest (RF) and Support Vector Machine (SVM) consistently achieved high accuracy for land classification. In this study, the application of RF and SVM classifiers, and object-based (OBIA) and pixel-based (PBIA) approaches with Sentinel-2A imagery was evaluated using Google Earth Engine (GEE) platform for land cover classification of a burned area in the Serra da Estrela Natural Park (PNSE), Portugal. This aimed to detect the land cover change and closely observe the burned area and vegetation recovery after the 2022 wildfire. The combination of RF and OBIA achieved the highest accuracy in all evaluation metrics. At the same time, a comparison with the Normalized Difference Vegetation Index (NDVI) map and Conjunctural Land Occupation Map (COSc) of 2023 year indicated that the SVM and PBIA map resembled the maps better.

Abstract: In the Western of Mekong Delta belong to Vietnam a amount of alluvium sediments flows from upstream China to Vietnam, they are going from the river branches to in the Mekong Delta, then get out the Western Sea of Vietnam. This sediments accumulated gradually elevation the new land. The increase elevation of the land creates a fertile coastal plain where mangrove forests with a rich ecosystem of plants and animals. Over time, these forests change, with different plant species succeeding each other. This aims of this study to finding plant species in the forest, classification of forest types based on ecological regions, assessement the biodiversity of tree species, and compare the abundance communities, measuring the growth of the forest in these regions. In 2023, a comprehensive survey was conducted by using a systematic approach. The number of survey plots have done depend on the density of the forest, Base on the width of the forest range, the number of survey plots from 500m to 1,000m set up 1-2 plots, sub region set up from 10 to 15 plots. In total, 68 plots have done established in the erea, the area of plot is 100 square meters (10m x 10m). Using the statistical software in forestry to survey and analysis data. The result provided a complete picture of the tree species composition, distribution, and community structure characteristics in each ecological sub-region. The result of survey showed in the sub-region 1 have seven species as Sonneracia ovate, Rhzophora apiculata, Excoecaria agallochha, Avicennia officinalis, Avicennia alba, Agricerasvcoculatum, Bruguiera gymnorhiza. In the sub region 2 have eleven species as Sonneracia caseolaris, Annona reticulata, Lumnitzera littorea, Lumnizera racemosa, Rhizophora apiculata, Excoecaria agallocha, Avicennia officinalis, Avicennia alba, Hibiscus tillacerus, Bruguiera gymorhiza, Aericeras comiculatum. In the sub region 3 have eight as Sonratia caseolaris, Rhizophora mucronata, Rhizophora apiculata, Excoecaria agallocha, Avicennia marine, Avicennia alba, Bruguiera gymorhiza. In the region 4 have ten species as Sonneracia caseolaris, Sonneracia ovate, Lumnizera racemosa, Rhizophoa apiculata, Excoecaria agallocha, Avicennia marina, Avicennia officinalis, Aviecnnia alba, Bruguiera gymorhiza, Bruguiera cylindrica The total species of the mangrove forest in the Western Mekong Delta have 16 species from 11 plant families have been identified. Among these species have 6 dominant species include Avicennia oficinali),Avicennia alba, Rhizophora apiculata, Excoecaria agallocha, Someratia caseolaris, and Bruguiera yipamoriza. Two species easy grow on the new land are Avicennia officinalis Avicennia alba this findings show they can develope on the original new land for the shore of the Western Mekong Delta. The surveyed results also calculated the average of the height, diameter (D1.3), canopy, health of the nature mangrove tree for each sub region and total region. From these results showed the division of foresty structure, the structure of height, diameter (D1.3), canopy, vitality of the sub region and total region in the Western Mekong Delta.

SQL injection attacks are one of the most common types of attacks on web applications. These attacks exploit vulnerabilities in the application’s database access mechanisms, allowing attackers to execute unauthorized SQL queries. In this study, we propose an architecture for detecting SQL injection attacks using a recurrent neural network (RNN) autoencoder. The proposed architecture was trained on a publicly available dataset of SQL injection attacks. Then compared with several other machine learning models, including ANN, CNN, Decision Tree, Naïve Bayes, SVM, Random Forest, and Logistic Regression. The experimental result showed that the proposed approach achieved an accuracy of 94% and an F1 score of 92%, which demonstrate its effectiveness in detecting QL injection attacks with high accuracy in comparison with other models covered in the study.

Fit-for-purpose land administration (FFPLA) seeks to simplify cadastral mapping via lowering the costs and time associated with conventional surveying methods. The approach can be applied to both initial establishment and on-going maintenance of system. In Ethiopia, cadastral maintenance remains an on-going challenge, especially in rapidly urbanizing peri-urban areas, where farmers' land rights and tenure security are often jeopardized. Automatic Feature Extraction (AFE) is an emerging FFPLA approach, proposed as an alternative for mapping and updating cadastral boundaries. This study explores the role of the AFE approach for updating cadastral boundaries in the vibrant peri-urban areas of Addis Ababa. Open-source software solutions are utilized to assess the (semi-) automatic extraction of cadastral boundaries from orthophotos (segmentation), designation of 'boundary' and 'non-boundary' outlines (classification), and delimitation of cadastral boundaries (interactive delineation). Both qualitative and quantitative assessments of the achieved results (validation) are undertaken. A high-resolution orthophoto of the study area and a reference cadastral boundary shape file are used, respectively, for extracting the parcel boundaries and validating the interactive delineation results. Qualitative (visual) assessment verified the completed extraction of newly constructed cadastral boundaries in the study area, although non-boundary outlines such as footpaths and artefacts are also retrieved. For the buffer overlay analysis, the interactively delineated boundary lines and the reference cadastre were buffered within the spatial accuracy limits for urban and rural cadasters. As a result, the quantitative assessment delivered 52% correctness and 32% completeness for a buffer width of 0.4m and 0.6m, respectively, for the interactively delineated and reference boundaries. The study further demonstrated the potentially significant role AFE could assist in delivering fast, affordable, and reliable cadastral mapping. Further investigation, based on user input and expertise evaluation, could help to improve the approach and apply it to a real-world setting.

Falls are extremely damaging to the elderly. The number of elderly who have experienced falls has increased over the years, several of the elderly stay alone or in in badly maintained elderly homes. This makes a low-cost fall detection system a necessity. There has been huge improvements in terms of IoT systems, ML algorithms. Varied data sets have been collected across the world for fall detection. These data sets have a very little in common among them, in terms of user demographics, sensors used, the ADL and Fall activities Hence in this paper we present a data set that has wide user demographics, we used various sensors – such as accelerometer, gyroscope, magnetometer and hear rate. We used wrist worn sensors to collect data. In this paper we present a detailed analysis of the data set we collected using common ML algorithms such as – Naïve Bayes (NB), K Nearest Neighbor (KNN), Logistic Regression (LR), Random Forest (RF) and Support Vector Machines (SVM). We analyzed the performance of these algorithms for variations in accuracy with respect to age, gender, height, weight and health issues and we have identified outliers by analyzing each incorrect prediction. This paper provides the complete details of the data collection methodology, The methods used for analysis and presents the results of analysis in complete detail.

Remote sensing is one of the modern methods that have significantly developed over the last two decades and nowadays provides a new means for forest monitoring. High spatial and temporal resolutions are demanded for accurate and timely monitoring of forests. In this study multi-spectral Unmanned Aerial Vehicle (UAV) images were used to estimate canopy parameters (definition of crown extent, top and height as well as photosynthetic pigment contents). The UAV images in Green, Red, Red-Edge and NIR bands were acquired by Parrot Sequoia camera over selected sites in two small catchments (Czech Republic) covered dominantly by Norway spruce monocultures. Individual tree extents, together with tree tops and heights, were derived from the Canopy Height Model (CHM). In addition, the following were tested i) to what extent can the linear relationship be established between selected vegetation indexes (NDVI and NDVIred edge) derived for individual trees and the corresponding ground truth (e.g., biochemically assessed needle photosynthetic pigment contents), and ii) whether needle age selection as a ground truth and crown light conditions affect the validity of linear models. The results of the conducted statistical analysis show that the two vegetation indexes (NDVI and NDVIred edge) tested here have a potential to assess photosynthetic pigments in Norway spruce forests at a semi-quantitative level, however the needle-age selection as a ground truth was revealed to be a very important factor. The only usable results were obtained for linear models when using the 2nd year needle pigment contents as a ground truth. On the other hand, the illumination conditions of the crown proved to have very little effect on the model’s validity. No study was found to directly compare these results conducted on coniferous forest stands. This shows that there is a further need for studies dealing with a quantitative estimation of the biochemical variables of nature coniferous forests when employing spectral data acquired by the UAV platform at a very high spatial resolution.

1) Background: Climate change may subject forests to climate conditions to which they are not adapted. Elevated temperatures reduce net photosynthesis by increasing respiration rates and increasingly long droughts dramatically increase morbidity. CO₂ enrichment enhances productivity, but it is not clear to what extent CO₂ enrichment can offset the negative effects of elevated temperatures and longer droughts. 2) Methods: We used a mechanistic landscape model to conduct controlled simulation experiments manipulating CO₂ concentration, temperature, drought length and soil water capacity. 3) Results: We found that elevated CO₂ stimulates productivity such that it dwarfs the negative effect caused by elevated temperature. Energy reserves were not as strongly mitigated by elevated CO₂, and mortality of less competitive cohorts increased. Drought length had a surprisingly small effect on productivity measures, but had a marked negative effect on mortality risk. 4) Conclusions: Elevated CO₂ compensated for the negative effect of longer droughts in terms of productivity measures, but not survival measures.

The aim of this study is to evaluate three different strategies to improve classification accuracy in a highly fragmented semiarid area. i) Using different classification algorithms: Maximum Likelihood, Random Forest, Support Vector Machines and Sequential Maximum a Posteriori, with parameter optimisation in the second and third cases; ii) using different feature sets: spectral features, spectral and textural features, and spectral, textural and terrain features; and iii) using different image-sets: winter, spring, summer, autumn, winter+summer, winter+ spring+summer; and a four seasons combination. A 3-way ANOVA is used to discern which of these approaches and their interactions significantly increases accuracy. Tukey-Kramer contrast using a heteroscedasticity-consistent estimation of the kappa covariances matrix was used to check for significant differences in accuracy. The experiment was carried out with Landsat TM, ETM, and OLI images corresponding to the period 2000-2015. A combination of four images was the best way to improve accuracy. Maximum Likelihood, Random Forest and Support Vector Machines do not significantly increase accuracy when textural information is added, but do so when terrain features are taken into account. On the other hand, Sequential Maximum a Posteriori increases accuracy when textural features are used, but reduces accuracy substantially when terrain features are included. Random Forest using the three feature subsets and Sequential Maximum a Posteriori with spectral and textural features had the largest kappa values, around 0.9.

Our goal was to determine the cellular immune response (CIR) among a random sample of the Borriana COVID-19 cohort (Spain) to identify its associated factors and their relationship with infection, reinfection and sequelae. We conducted a nested case-control study using a randomly selected sample of 225 individuals age 18 and older, including 36 individual naïve to SARS-CoV-2 infection, and 189 infected patients. We employed flow cytometry-based immunoassays for intracellular cytokine staining, utilizing Wuhan and BA.2 antigens and chemoluminescence microparticle immunoassay for detection SARS-CoV-2 antibodies. Logistic regression models were used. A total of 215 (95.6%) participants exhibited T-cell response (TCR) to at least one antigen. Positive responses of CD4+ and CD8+ T cells were 89.8% and 85.3% respectively. No difference in CIR was found for naïve and infected patients. Patients who experienced sequelae exhibited higher CIR than those without. A positive correlation was observed between TCR and Anti-Spike IgG levels. Factors positive associated with TCR included A-blood group, number of SARS-CoV-2 vaccine doses received, and Anti-N IgM; factors inversely related were the time elapsed since the last vaccine dose or infection, and B-blood group. These findings contribute valuable insights into the nuance immune landscape shaped by SARS-CoV-2 infection and vaccination.

Fibroblast activation protein (FAP) is a membrane-tethered serine protease overexpressed in the reactive stromal fibroblasts of &gt; 90% human carcinomas, which makes it a promising target for developing radiopharmaceuticals for imaging and therapy of carcinomas. Here, we synthesized two novel (R)-pyrrolidin-2-yl-boronic acid-based FAP-targeted ligands; SB02055 (DOTA-conjugated (R)-(1-((6-(3-(piperazin-1-yl)propoxy)quinoline-4-carbonyl)glycyl)pyrrolidin-2-yl)boronic acid) and SB04028 (DOTA-conjugated ((R)-1-((6-(3-(piperazin-1-yl)propoxy)quinoline-4-carbonyl)-D-alanyl)pyrrolidin-2-yl)boronic acid). natGa- and 68Ga-complexes of both ligands were evaluated in preclinical studies and compared to previously reported natGa/68Ga-complexed PNT6555. Enzymatic assays showed that FAP binding affinities (IC50) of natGa-SB02055, natGa-SB04028 and natGa-PNT6555 were 0.41±0.06, 13.9±1.29 and 78.1±4.59 nM, respectively. PET imaging and biodistribution studies in HEK293T:hFAP tumor-bearing mice showed that while [68Ga]Ga-SB02055 presented with a nominal tumor uptake (1.08±0.37 %ID/g), [68Ga]Ga-SB04028 demonstrated clear tumor visualization with ~1.5-fold higher tumor uptake (10.1±0.42 %ID/g) compared to [68Ga]Ga-PNT6555 (6.38±0.45 %ID/g). High accumulation in the bladder indicated renal excretion of all three tracers. [68Ga]Ga-SB04028 displayed low background level uptake in most normal organs, and comparable to [68Ga]Ga-PNT6555. However, since its tumor uptake was considerably higher than [68Ga]Ga-PNT6555, the corresponding tumor-to-organ uptake ratios for [68Ga]Ga-SB04028 were also significantly greater than [68Ga]Ga-PNT6555. Our data demonstrate that (R)-(((quinoline-4-carbonyl)-D-alanyl)pyrrolidin-2-yl)boronic acid is a promising pharmacophore for the design of FAP-targeted radiopharmaceuticals for cancer imaging and radioligand therapy.

Access to medicines is one of the essential problems in Public Health of low- and middle-income countries (LMICs). The World Health Organization (WHO) defines access to medicines as the possibility of "having continuously accessible and affordable medicines in public or private health facilities that are within a kilometer of the place of residence." Access to medicines, as defined by the WHO, is not fully guaranteed in many LMICs and even in many regions of high-resource countries. The WHO identifies several factors as determinants of limitations in the access to medicines: rational selection, affordable prices, sustainable financing, and reliable health services. The action on these factors makes it possible to improve universal access to medicines with consequent improvement in Public Health. Adequate access to antibiotics and vaccination will avoid a large part of the deaths caused by infectious diseases in the LMICs. However, the emergence of resistance and the difficulties in vaccination campaigns due to socio-political or cultural problems make it challenging to fight many easily treatable infectious diseases. The use and abuse of antibiotics are inevitably associated with the appearance of resistances that make them ineffective. Thus, whereas limited access to antibiotics raises mortality rates from infectious diseases, generalized open access to them ends up eliminating their clinical value. Moreover, the contraction of research in this field for many years has reduced the success in discovering new drugs. Additionally, local market regulations, inadequate selection, inaccessible prices, especially for those of second and third-generation, inefficient health systems, and difficulties of administration and control of prescription compliance, especially in the case of combined therapies, are additional obstacles to universal access to antibiotics. In order to simultaneously improve access to antibiotics and keep resistances under control, it is necessary to develop training and education activities at different social levels (from patients to various Health Care Providers) to complement the national or supranational strategic plans.

Outlier detection is a critical technique across various domains, including statistics, data science, machine learning, and finance. Outliers, data points that differ significantly from the majority, can indicate errors, anomalies, or even new insights. This article provides an in-depth exploration of the primary techniques used to detect outliers, categorized into statistical methods, machine learning-based approaches, and proximity-based methods. We discuss the advantages, limitations, and specific use cases of each technique, highlighting their applicability to different types of datasets. The goal is to equip practitioners with a better understanding of how to identify and handle outliers effectively in real-world data analysis.

This study harnesses the potential of artificial intelligence (AI) focusing on machine learning (ML) to improve decision-making in advanced engineering education. The onset of the COVID-19 pandemic compelled a swift transformation in higher education methodologies, particularly in the domain of course modality. This study aims to align institutional decisions with student and faculty preferences in the face of rapid changes in instructional approaches, particularly learning styles, theoretical, practical, and theoretical-practical, prompted by the COVID-19 pandemic. To ascertain the preferences of students and instructors regarding class modalities across various courses, we utilized the Cognitive Process-Embedded Systems and E-learning conceptual framework. This framework effectively delineates the task execution process within the scope of technology-enhanced learning environments for both students and instructors. This study was conducted in seven Iranian universities and their STEM departments, examining their preferences for different learning styles. After analyzing the variables by different feature selection methods, we used three ML methods—decision trees (DT), support vector machines (SVM), and random forest (RF)—for comparative analysis. The results demonstrated the high performance of the RF model in predicting curriculum style preferences, making it a powerful decision-making tool in the evolving post-COVID educational landscape. This study not only demonstrates the effectiveness of ML in predicting educational preferences but also contributes to understanding the role of self-regulated learning in educational policy and decision-making in higher education.

This manuscript reviews three case studies from Brazil, Panama, and Portugal that illustrate Nature-based Solutions. The Tijuca Forest in Rio de Janeiro is a remarkable story of centuries of forest management that today is recognized as a UNESCO World Heritage Site. The Panama Canal Watershed produces water for Canal operations, electricity generation, and drinking water for half the country´s population. Traditional windmills and weirs near streams in the Alentejo Region, Portugal, have largely been abandoned due to the damming of the Guadiana River. Yet today, weirs are increasingly recognized for their important contribution to water provisioning in this dry region. All have a primary goal related to water provisioning, yet their ecosystem benefits are multiple. The cases offer important lessons for adaptation to climate change, cultural benefits from traditional human activities, and concerns about social equity.

As digitalization expands across all sectors, the economic toll of software defects on the U.S. economy reaches up to $2.41 trillion annually. High-profile incidents like the Boeing 787-Max 8 crash have shown the devastating potential of these defects, highlighting the critical importance of software testing within quality assurance frameworks. However, due to its complexity and resource intensity, the exhaustive nature of comprehensive testing often surpasses budget constraints. This research utilizes a machine learning (ML) model to enhance software testing decisions by pinpointing areas most susceptible to defects and optimizing scarce resource allocation. Previous studies have shown promising results using cost-sensitive training to refine ML models, improving predictive accuracy by reducing false negatives through addressing class imbalances in defect prediction datasets. This approach facilitates more targeted and effective testing efforts. Nevertheless, the generalizability of these models across different projects (cross-project) and programming languages (cross-language) remained untested. This study validates the model's applicability across diverse development environments by integrating various datasets from distinct projects into a unified, using a more interpretable ML approach. The results demonstrate that ML can support software testing decisions, enabling teams to identify up to seven times more defective modules with the same testing effort as a benchmark.

In recent years, computer-aided analysis techniques have emerged as valuable tools in assisting dermatologists by providing objective and efficient analysis of skin cancer images. This paper utilizes the combination of the Contourlet Transform (CT) and Local Binary Pattern (LBP) techniques for accurately recognizing borders, contrast changes, and shapes of skin cancer images. These results often contain many features, leading to high computational costs and potential over-fitting issues. Hence, we applied Particle Swarm Optimization (PSO) to select the most informative and discriminating features, reducing the dimensionality while retaining important information for accurate classification. After reducing the feature set with PSO, we applied these sets to Machine learning classification algorithms: Support Vector Machine (SVM), Random Forest (RF), and Neural Networks (NN). The results show that SVM has the lowest time complexity of 0.0458 seconds, followed by the Neural Network at 0.08730 seconds, and the Random Forest model has the highest time complexity of 0.1622 seconds. The SVM and Neural Network models are faster to train than the Random Forest model, making them more suitable for real-time or latency-sensitive applications. We also compared our proposed model with the state-of-the-art models and obtained the accuracy of 86.9%, which is the highest among the models.

Flaviviruses are a group of positive sense single-stranded RNA viruses predominantly transmitted by arthropods (mainly mos-quitoes), that cause severe endemic infections and epidemics on a global scale. It represents a major cause of systemic morbidity and death that are expanding worldwide. Among this group, Dengue fever, West Nile virus, Yellow Fever, Japanese Encephalitis, and recently Zika virus have been linked to a spectrum of ocular manifestations. The latter encompass subconjunctival hemor-rhage and conjunctivitis, anterior and posterior uveitis (inclusive of vitritis, chorioretinitis, and retinal vasculitis), maculopathy, retinal hemorrhages, and optic neuritis. Clinical diagnosis of these infectious diseases is primarily based on epidemiological data, history, systemic symptoms and signs, and the pattern of ocular involvement. Diagnosis confirmation relies on laboratory testing, including RT-PCR and serological testing. Ocular involvement typically follows a self-limited course but can result in irreversible visual impairment. Effective treatment for flavivirus infections is currently unavailable. Prevention remains the mainstay for arthropod vector and zoonotic disease control. Effective vaccines are available only for Yellow Fever virus, Dengue virus and Japanese Encephalitis virus. This review comprehensively summarizes the current knowledge regarding the ophthalmic mani-festations of the foremost flavivirus-associated human diseases.

Wearable devices have become ubiquitous, collecting rich temporal data that offers valuable insights into human activities, health monitoring, and behavior analysis. Leveraging this data, researchers have developed innovative approaches to classify and predict time-based patterns and events in human life. Time-based techniques allow the capture of intricate temporal dependencies, which is the nature of the data coming from wearable devices. This paper focuses on predicting well-being factors, such as stress, anxiety, positive and negative affect, on the Tesserae dataset collected from office workers. We examine the performance of different methodologies, including deep learning architectures, LSTM, ensemble techniques, Random Forest (RF) and XGBoost and compare their performances for time-based and non-time-based versions. In time-based versions, we investigate the effect of previous records of well-being factors on the upcoming ones. The overall results show that time-based LSTM performs the best among conventional (non-time-based) RF, XGBoost, and LSTM. The performance even increases when we consider a more extended previous period, in this case, 3 past-days rather than 1 past-day to predict the next day. Furthermore, we explore the corresponding biomarkers for each well-being factor using feature ranking. The obtained rankings are compatible with the psychological literature. In this work, we validated them based on device measurements rather than subjective survey responses.

Forest degradation is known to be widespread in the tropics, but is currently very poorly mapped, in part because there is little quantitative data on which satellite sensor characteristics and analysis methods are best at detecting it. To improve this, we used data from the Tropical Forest Degradation Experiment (FODEX) plots in the southern Peruvian Amazon, where different numbers of trees had been removed from four 1 ha forest plots, carefully inventoried by hand and Terrestrial Laser Scanning before and after the logging to give a range of biomass change (ΔAGB) values. We conducted a comparative study of six multispectral optical satellite sensors (WorldView-3, SkySat, SPOT-7, PlanetScope, Sentinel-2 and Landsat 8) at 0.3 – 30 m spatial resolution, to find the best combination of sensor and remote sensing indicator for change detection. Spectral reflectance, the Normalized Difference Vegetation Index (NDVI) and texture parameters were extracted after radiometric calibration and image preprocessing. The strength of the relationships between the change in these values and field-measured ΔAGB (computed in % ha−1) was analysed. The results demonstrate that: (a) texture measures correlates more with ΔAGB than simple spectral parameters; (b) the strongest correlations are achieved for those sensors with spatial resolutions in the intermediate range (1.5 - 10 m), with finer or coarser resolutions producing worse results, and (c) when texture is computed using a moving square window ranging between 9 - 14 m in length. Maps predicting ΔAGB showed very promising results using a NIR-derived texture parameter for 3 m resolution PlanetScope (R2 = 0.97 and RMSE = 1.80 % ha−1), followed by 1.5 m SPOT-7 (R2 = 0.74 and RMSE = 5.25 % ha−1) and 10 m Sentinel-2 (R2 = 0.71 and RMSE = 5.55 % ha−1). Texture models derived from 0.3 m WorldView-3 improved with increasing window size, with highest R2 of 0.62 and RMSE = 6.35 % ha−1 for a window of 14 m in length. The degradation in our field plots is invisible to the 30 m resolution Landsat data. Our findings imply that, at least for lowland Peru, low-medium intensity disturbance can be detected best in optical wavelengths using a texture measure derived from 3 m PlanetScope data. That such data are being collected daily, and currently released free as monthly mosaics over tropical forests as part of the Norway’s International Climate and Forest Initiative (NICFI), is excellent news for monitoring such degradation.

The aim of this work is to explain the behaviour of the multiresistance percentage of Pseudomona aeruginosa in some countries of Europe through a multivariate statistical analysis and machine learning validation, using data from the European Antimicrobial Resistance Surveillance System, the World Health Organization and the World Bank. First, we will use a descriptive analysis and a principal components analysis. Then, we use a k-means clustering to determine the countries and regions that are most affected by the antibiotic resistance. Second, we expand the database by adding some socioeconomic, governance and antibiotic-consumption variables. We then run a data panel regression analysis to determine some functions that relates the multiresistance percentage with those new variables. Finally, we use machine learning techniques to validate a pooling panel data case, using XGBoost and random forest algorithms. The results of the data panel analysis indicate that the most important variables for the multiresistance percentage are corruption control and the rule of law. Similar results are found with the machine learning validation analysis, where the human development index is an additional important variable for the multiresistance percentage.

The presented study demonstrates the bi-sensor approach suitable for rapid and precise up-to-date mapping of forest canopy gaps for the larger spatial extent. The approach makes use of UAV RGB images on smaller areas for highly precise forest canopy mask creation. Sentinel-2 was used as a scaling platform for transferring information from UAV to a wider spatial extent. The various approaches of the improvement of the predictive performance were examined: (I) the highest R2 of the single satellite index was up to 0.57, (II) the highest R2 using multiple features obtained from the single date, S-2 image was 0.624 and, (III) the highest R2 on the multi-temporal set of S-2 images, was 0.697. Satellite indices such as ARVI, IPVI, NDI45, PSSRa, MCARI, CI, RI, and NDTI were the dominant predictors in most of the ML algorithms. The more complex ML algorithms such as SVM, Random Forest, GBM, XGBoost, and Catboost that provided the best performance on the training set exhibited weaker generalization capabilities. Therefore, a simpler and more robust Elastic Net algorithm was chosen for the final map creation.

Tree-killing bark beetles are the most economically important insects in conifer forests worldwide. Yet despite >200 years of research, the drivers of population eruptions or crashes are still not fully understood, precluding reliable predictions of the effects of global change on beetle population dynamics and impacts on ecosystems and humans. We critically analyze potential biotic and abiotic drivers of population dynamics of the European spruce bark beetle (Ips typographus) and present a novel ecological framework that integrates the multiple drivers governing this bark beetle system. We call for large-scale collaborative research efforts to improve our understanding of the population dynamics of this important pest; an approach that might serve as a blueprint for other eruptive forest insects.

Changes in extreme weather patterns are expected under climate change. In this study, a risk assessment was conducted using 4 building damage history datasets and 33 weather datasets (precipitation, wind speed, snow, and temperature) from 230 regions in South Korea to quantitatively analyze and predict building damage caused by potential future natural disasters. Decision tree analysis was used to evaluate building damage risk in 230 regions. The decision tree model to determine the risk of flood, gale, and typhoon was generated, which excluded gales, with less damage. The weight (variable importance) and limit value (damage limit) of the weather variables ware derived using the decision tree model. Using these two factors, we assessed the building damage risk in 230 regions in South Korea until 2100. The number of regions at risk of flood damage increased by more than 30% in average. Conversely, regions at risk of snowfall damage decreased by more than 90%. The regions at risk of typhoons decreased by 57.5% on average, and the number of regions at high risk of typhoon damage increased by up to 62.5% in RCP 8.5. These results can be used as objective data to minimize future building damage throughout South Korea, representing the first step towards sustainable development in the region with respect to disaster response.

Urban sprawl propelled by rapid population growth leads to the shrinkage of productive agricultural lands and pristine forests in the suburban areas and, in turn, substantially alters ecosystem services. Hence, the quantification of urban sprawl is crucial for effective urban planning, and environmental and ecosystem management. Like many megacities in fast growing developing countries, Chennai, the capital of Tamilnadu and one of the business hubs in India, has experienced extensive urban sprawl triggered by the doubling of total population over the past three decades. We employed the Random Forest (RF) classification on Landsat imageries from 1991, 2003, and 2016, and computed spatial metrics to quantify the extent of urban sprawl within a 10km suburban buffer of Chennai. The rate of urban sprawl was quantified using Renyi’s entropy, and the urban extent was predicted for 2027 using land-use and land-cover change modeling. A 70.35% increase in urban areas was observed for the suburban periphery of Chennai between 1991 and 2016. The Renyi’s entropy value for year 2016 was ≥ 0.9, exhibiting a two-fold rate of urban sprawl. The spatial metrics values indicate that the existing urban areas of Chennai became denser and the suburban agricultural, forests and barren lands were transformed into fragmented urban settlements. The forecasted urban growth for 2027 predicts a conversion of 13670.33ha (16.57 % of the total landscape) of existing forests and agricultural lands into urban areas with an associated increase in the entropy value of 1.7. Our findings are relevant for urban planning and environmental management in Chennai and provide quantitative measures for addressing the social-ecological consequences of urban sprawl and the protection of ecosystem services.

Teclistamab (TEC) is the first B-cell maturation antigen-directed bispecific antibody approved by the European Medicines Agency and Food and Drug Administration for triple-class exposed relapsed/refractory multiple myeloma (RRMM). As TEC is increasingly being used in real-world (RW) settings, this study sought to gather existing RW evidence on effectiveness, safety, healthcare resource utilization, and clinical practices associated with TEC. A systematic literature review was performed to identify RW observational studies of adults with RRMM treated with TEC. Sixty-one records representing 41 unique studies were included; sample sizes ranged from 8 to 572 patients. Where reported, median follow-up ranged from 2.3 to 33.6 months and more than 65% of patients would have been ineligible for the pivotal trial of TEC (MajesTEC-1) in all but one study. Across populations with ≥50 patients, overall response rates ranged from 50 to 66% and cytokine release syndrome (CRS) rates ranged from 18 to 64%. Tocilizumab use for CRS management was reported in 14 studies, with two indicating lower CRS rates (13% and 26%) when used prophylactically. Survival and infection outcomes showed wide variability due to short follow-up in most studies. Overall, early RW effectiveness and safety outcomes of TEC were comparable to findings from MajesTEC-1.

We previously reported nano-pulse treatment (NPT), a pulsed power technology, resulted in 4T1-luc mammary tumor elimination and a strong in situ vaccination, thereby completely protecting tumor-free animals against a second live tumor challenge. The mechanism whereby NPT mounts effective antitumor immune responses in the 4T1 breast cancer predominantly immunosuppressive tumor microenvironment (TME) remains unanswered. In this study, orthotopic 4T1 mouse breast tumors were treated with NPT (100 ns, 50 kV/cm, 1000 pulses, 3 Hz). Blood, spleen, draining lymph nodes and tumors were harvested at 4-hour, 8-hour, 1-, 3-, 7-day. Additional studies were conducted over 3-month posttreatment intervals for the analysis of frequencies, death and functional markers of various immune cells in addition to suppressor function of regulatory T cells (Tregs). NPT was verified to elicit strong in situ vaccination (ISV) against breast cancer and promoted both acute and long-term T cell memory. NPT abolished immunosuppressive dominance systemically and in the TME by substantially reducing Tregs, myeloid-derived suppressor cells (MDSCs), and tumor-associated macrophages (TAMs). NPT induced apoptosis in Tregs and TAMs. It also functionally diminished the Treg suppression capacity, likely explained by the downregulation of activation markers, particularly 4-1BB and TGFβ, and a phenotypic shift from predominantly activated (CD44+CD62L-) to naïve (CD44-CD62L+) Tregs. Importantly, NPT selectively induced apoptosis in activated Tregs and spared effector CD4+ and CD8+ T cells. These changes were followed by a concomitant rise in CD8+CD103+ tissue-resident memory T cells and TAM M1 polarization. These findings indicate that NPT effectively switches the TME and secondary lymphatic systems from an immunosuppressive to an immunostimulatory state allowing cytotoxic T-cell function and immune memory formation to eliminate cancer cells and account for the NPT in situ vaccination.

Finding out how cells with the same genome change fates in differentiation commitment is a challenge of biology. We used MCF-7 breast cancer cells treated with the ErbB2 ligand heregulin (HRG), which induces differentiation, to address if and how the constitutive pericentromere-associated domains (PADs) may be involved in this process. PAD-specific repressive heterochromatin (H3K9me3) and active euchromatin (H3K4me3) marking, centromere (CENPA) labelling, qPCR, acridine-orange-DNA structural test, and microscopic image analysis were applied. We found a two-step DNA unfolding, at 15-20 min and 60 min after HRG treatment, coinciding with bi-phasic activation of the early response genes (c-FOS family) and two steps of critical phase transition which were revealed in transcriptome studies. In control, the distribution of PAD number and size displays a power-law scaling with a boundary at the nucleolus. PADs’ clustering correlates with centromere numbers. 15 min after HRG treatment, the unravelling of PADs occurs, coinciding with the first step of euchromatin unfolding. The second step is associated with transcription of long-non-coding-RNA from satellite III DNA. We hypothesize that splitting of the PAD clusters under the critical size threshold of the silencing domain abrupts position effect variegation. It allows the first genome transcription avalanche to occur, starting differentiation commitment.

Herpesviruses usurp cellular stress responses to avoid immune detection while simultaneously promoting viral replication and spread. The unfolded protein response (UPR) is an evolutionarily conserved stress response that is activated when the protein load in the ER saturates its chaperone folding capacity causing an accrual of misfolded proteins. Through translational and transcriptional reprogramming, the UPR aims to restore protein homeostasis; however, if this fails the cell undergoes apoptosis. It is commonly thought that many enveloped viruses, including herpesviruses, may activate the UPR due to saturation of the ER with nascent glycoproteins and thus these viruses may have evolved mechanisms to evade the potentially negative effects of UPR signaling. Over the past fifteen years there has been considerable effort to provide evidence that different viruses may reprogram the UPR to promote viral replication. Here we provide an overview of the molecular events of UPR activation, signaling and transcriptional outputs, and highlight key findings that demonstrate that the UPR is an important cellular stress response that herpesviruses have hijacked to facilitate persistent infection.

Power consumption prediction is a crucial component in enhancing the efficiency and sustainability of building operations. This study investigates the impact of data collection frequency and model selection on the predictive accuracy of power consumption in two distinct building types: an Academic one with 15-minute interval data and an Industrial one with hourly data. Various machine learning models, including Support Vector Machine (SVM) with Radial and Sigmoid kernels, Random Forest (RF), and Deep Neural Networks (DNN), across different data splits and feature sets. Our analysis reveals that higher data collection frequency generally improves model performance, as indicated by lower RMSE, MAPE, and CV values, alongside higher R² scores. The inclusion of more historical power consumption features has also been found to have a more significant impact on the accuracy of predictions than including climate condition features. Moreover, the SVM-Radial model consistently outperformed others, particularly in capturing complex, non-linear patterns in the data. However, the DNN model, while competent in some metrics, showed elevated MAPE values, suggesting potential overfitting issues. These findings suggest that careful consideration of data frequency, features and model selection is essential for optimizing power prediction, contributing to more efficient power management strategies in building operations.

Machine learning (ML) offers potential opportunities to enhance the learning, teaching and assessments within veterinary medical education including but not limited to assisting with admissions processes as well as student progress evaluations. The purpose of this primer is to assist veterinary educators in appraising and potentially adopting these rapid upcoming advances in data science and technology. In the first section, we introduce ML concepts and highlight similarities/differences between ML and classical statistics. In the second section, we provide a step-by-step worked example using simulated veterinary student data to answer a hypothesis driven question. Python syntax with explanations is provided within the text to create a random forest ML prediction model and within each step, specific considerations such as how to manage incomplete student records are highlighted when applying ML algorithms within the veterinary education field. The results from the simulated data demonstrate how decisions by the veterinary educator during ML model creation may impact the most important features contributing to the model. These results highlight the need for the veterinary educator to be fully transparent during the creation of ML models and future research is needed to establish guidelines for handling data not missing at random in medical education, and preferred methods for model evaluation.

Forests, integral to human civilization, hold immense value and play a vital role in maintaining ecological harmony. Despite India's goal of extensive forest coverage, significant progress is still needed. Uncontrolled forest fires pose a severe threat, particularly in Uttarakhand's Pauri Garhwal district. To address this challenge, a comprehensive study examined surface and subsurface hydrological factors influencing the forest fire occurrences, such as elevation, aspect, slope, vegetation, proximity to human settlements, proximity to waterbodies, Active faults and lineament density. A total of 15 such factors were integrated with advanced techniques of remote sensing and GIS and coupled with historical fire data to create a precise forest fire risk map using the support vector machine algorithm. Forest fire risk map was classified into 5 distinct risk zones, Very High Risk (47.38 Km2), High Risk (275.98 km2, Moderate Risk (985.49 km2), Low Risk (1741.17 km2) and Very Low Risk (2374.11 km2) aiding in proactive fire management. By embracing this innovative tool, decision-makers can protect forests, preserve biodiversity, and ensure a sustainable future for generations to come.

Current satellite remote sensing methods struggle to detect and map forest degradation, a critical issue as it is likely a major and growing source of carbon emissions and biodiveristy loss. TanDEM-X InSAR phase height (hϕ) is a promising variable for measuring forest disturbances, as it is closely related to mean canopy height, and thus should decrease if canopy trees are removed. However, previous research has focused on relatively flat terrain, despite the fact that much of the worlds’ remaining tropical forests are found in hilly areas, and this inevitably introduces artifacts in sideways imaging systems. In this paper, we find a relationship between hϕ and aboveground biomass change in four selectively logged plots in a hilly region of central Gabon. We show that minimising the level of multilooking in the interferometric processing chain strengthens this relationship, and that degradation estimates across steep slopes in the surrounding region are improved by selecting data from the most appropriate pass directions on a pixel-by-pixel basis. This shows that TanDEM-X InSAR can measure the magnitude of degradation, and that topographic effects can be mitigated if data from multiple SAR viewing geometries are available.

In recent years, data providers are generating and streaming a large number of images. More particularly, processing images that contain faces have received great attention due to its numerous applications, such as entertainment and social media apps. The enormous amount of images shared on these applications presents serious challenges and requires massive computing resources to ensure efficient data processing. However, images are subject to a wide range of distortions in real application scenarios during the processing, transmission, sharing, or combination of many factors. So, there is a need to guarantee acceptable delivery content, even though some distorted images do not have access to their original version. In this paper, we present a framework developed to estimate the images' quality while processing a large number of images in real-time. Our quality evaluation is measured using an integration of a deep network with random forests. In addition, a face alignment metric is used to assess the facial features. Experimental results have been conducted on two artificially distorted benchmark datasets, LIVE and TID2013. We show that our proposed approach outperforms the state-of-art methods, having a Pearson Correlation Coefficient (PCC) and Spearman Rank Order Correlation Correlation Coefficient (SROCC) with subjective human scores of almost 0.942 and 0.931 while minimizing the processing time from 4.8ms to 1.8ms.

In this work, a hybrid component Fault Detection and Diagnosis (FDD) approach for industrial sensor systems is established and analyzed, to provide a hybrid schema that combines the advantages and eliminates the drawbacks of both model-based and data-driven methods of diagnosis. Moreover, it shines the light on a new utilization of Random Forest (RF) together with model-based diagnosis, beyond its ordinary data-driven application. RF is trained and hyperparameter tuned using three-fold cross validation over a random grid of parameters using random search, to finally generate diagnostic graphs as the dynamic, data-driven part of this system. This is followed by translating those graphs into model-based rules in the form of if-else statements, SQL queries or semantic queries such as SPARQL, in order to feed the dynamic rules into a structured model essential for further diagnosis. The RF hyperparameters are consistently updated online using the newly generated sensor data to maintain the dynamicity and accuracy of the generated graphs and rules thereafter. The architecture of the proposed method is demonstrated in a comprehensive manner, and the dynamic rules extraction phase is applied using a case study on condition monitoring of a hydraulic test rig using time-series multivariate sensor readings.

Solid tumor cells have an altered metabolism that can protect them from cytotoxic lymphocytes. The antidiabetic drug metformin modifies tumor cell metabolism and several clinical trials are testing its effectiveness for the treatment of solid cancers. The use of metformin in hematologic cancers has received much less attention, although allogeneic cytotoxic lymphocytes are very effective against these tumors. We show here that metformin induces expression of Natural Killer G2-D (NKG2D) ligands (NKG2DL) and intercellular adhesion molecule-1 (ICAM-1), a ligand of the lymphocyte function-associated antigen 1 (LFA-1). This leads to enhance sensitivity to cytotoxic lymphocytes. Overexpression of antiapoptotic Bcl-2 family members decrease both metformin effects. The sensitization to activated cytotoxic lymphocytes is mainly mediated by the increase on ICAM-1 levels, which favors cytotoxic lymphocytes binding to tumor cells. Finally, metformin decreases the growth of human hematological tumor cells in xenograft models, mainly in presence of monoclonal antibodies that recognize tumor antigens. Our results suggest that metformin could improve cytotoxic lymphocyte-mediated therapy.

Extracellular vesicles (EVs), such as exosomes or oncosomes are released with molecules unfavorable for survival from cells. In addition, accumulating evidence has shown that tumor cells often eject anti-cancer drugs such as chemotherapeutics and targeted drugs within EVs, a novel mechanism of drug resistance. The EV-releasing, drug resistance phenotype is often coupled with cellular dedifferentiation and transformation, cells undergoing epithelial-mesenchymal transition (EMT) and taking on a cancer stem cell phenotype. Recent studies have shown that the release of EVs is also involved in immunosuppression. The concept of the resistance-associated secretory phenotype (RASP) is reviewed herein.

Navigating growth cones are exposed to multiple signals simultaneously and have to integrate competing cues into a coherent navigational response. Integration of guidance cues is traditionally thought to occur at the level of cytoskeletal dynamics. Drosophila studies indicate that cells exhibit a low level of continuous caspase protease activation, and that axon guidance cues can activate or suppress caspase activity. We base a model for axon guidance on these observations. By analogy with other systems in which caspase signaling has non-apoptotic functions, we propose that caspase signaling can either reinforce repulsion or negate attraction in response to external guidance cues by cleaving cytoskeletal proteins. Over the course of an entire trajectory, incorrectly navigating axons may pass the threshold for apoptosis and be eliminated, whereas axons making correct decisions will survive. These observations would also explain why neurotrophic factors can act as axon guidance cues and why axon guidance systems such as Slit/Robo signaling may act as tumor suppressors in cancer.

Pediatric Autoimmune Neuropsychiatric Disorders Associated with Streptococcal Infections (PANDAS) is one of the most controversial diseases in pediatric rheumatology. Despite its first description being published more than 25 years ago as the sudden and rapid onset of obsessive-compulsive disorder (OCD) and/or tic disorder symptoms as complications of a Group A beta-hemolytic Streptococcus (GAS) infection, precise epidemiological data are still lacking, and there are no strong recommendations for its treatment. Recent advances in the comprehension of PANDAS pathophysiology are largely attributable to animal model studies and the understanding of the roles of Ca++/calmodulin-dependent protein kinase (CaM kinase) II, disrupted dopamine re-lease in the basal ganglia, and striatal cholinergic interneurons. The diagnosis of PANDAS should be made after an exclusion process and should include prepubescent children with a sudden onset of OCD and/or a tic disorder, with a relapsing/remitting disease course, a clear temporal association between GAS infection and onset or exacerbation of symptoms, and the association with other neurological abnormalities such as motoric hyperactivity and choreiform movements. Antibiotic medications are the primary therapeutic modality. Nonetheless, there is a paucity of randomized studies and validated data, resulting in a scarcity of solid recommendations.

The literature lacks consensus on the minimum microbial density required for diagnosing urinary tract infections (UTIs). This study categorized the microbial densities of urine specimens from symptomatic presumptive UTI patients and correlated them with detected levels of immune response biomarkers NGAL, IL-8, and IL-1β. The objective was to identify changes in immune response at different microbial densities and determine an optimal threshold for diagnosing symptomatic UTIs. Midstream clean-catch urine samples were analyzed for microbial identification and quantification using SUC and M-PCR, while NGAL, IL-8, and IL-1β levels were measured with ELISA. NGAL, IL-8, and IL-1β levels were all significantly higher in symptomatic subjects with microbial densities ≥ 10,000 cells/mL by M-PCR (p < 0.0069) or equivalent CFUs/mL by SUC (p < 0.0104) compared to samples with no detectable microbes. With both PCR and SUC, a consensus of two or more elevated biomarkers correlated well with microbial densities > 10,000 cells/mL or CFU/mL, respectively. The association between ≥ 10,000 cells and CFU per mL with elevated biomarkers in symptomatic patients suggests that this lower threshold may be more suitable for diagnosing UTIs. These findings challenge the widely used threshold of 100,000 CFU per mL, which may overlook many active UTI cases.

Background and Aim. Since virus-related hepatocellular carcinoma (HCC) pathogenesis involves liver inflammation, therefore, post-hepatitis C virus (HCV) infection would be a cause for liver cirrhosis that would progress to HCC. Cytotoxic T cells (Tc) are known to be involved in post-HCV complications and HCC pathogenesis. The inhibitory checkpoint Leukocyte-Associated Immunoglobulin-like Receptor-1 (LAIR-1) is expressed on Tc. Therefore, we aimed to determine whether the Tc expression level of LAIR-1 is associated with HCC progression post-HCV and moreover, to evaluate LAIR-1 expression as a non-invasive biomarker for HCC progression in the context of liver cirrhosis post-HCV genotype 4 (G4) in Egyptian patients’ peripheral venous blood liquid biopsy. We studied LAIR-1 expression on Tc related to the progression of liver cirrhosis in a case-controlled study enrolled 64 patients with post-HCV G4-HCC and 37 patients with post-HCV G4-liver cirrhosis. Methods: LAIR-1 expression was analyzed by flow cytometry. Results: LAIR-1 expression on Tc and the percentage of Tc positive for LAIR-1 (LAIR-1+Tc %) were significantly higher in the post-HCV G4-HCC group compared to the post-HCV G4-liver cirrhosis

The bHLH-PAS proteins are a family of transcription factors regulating expression of a wide range of genes involved in different functions, from differentiation and development control, by oxygen and toxins sensing to circadian clock setting. In addition to the well-preserved DNA-binding bHLH and PAS domains, bHLH-PAS proteins contain long intrinsically disordered C-terminal regions, responsible for their activity regulation. Our aim was to analyse the potential connection between disordered regions of the bHLH-PAS transcription factors with posttranscriptional modifications and liquid-liquid phase separation in the context of the disease-associated missense mutations. Highly flexible disordered regions, enriched in short more ordered motives, are responsible for wide spectrum of interactions with transcriptional co-regulators. Based on our in silico analysis and taking into account fact that transcription factors functions can be modulated by posttranslational modifications and spontaneous phase separation, we assume that the location of missense mutations inducing disease states, is clearly related to sequences directly undergoing these processes or to sequences responsible for their activity regulation.

In this paper, we investigate the proftability of investing in movies for retail investors, focusing on the return on investment (ROI) metric, which is calculated as revenue divided by budget. Our analysis encompasses 5000 movies from the movie database TMDB, examining factors such as production scale, genres, key actors, directors, and more. Additionally, we employ textual analysis techniques like Latent Semantic Analysis (LSA) on movie overviews and tags to incorporate movie themes. We then evaluate various supervised classifcation models including Logistic Regression, Random Forest, and Light Gradient Boosting (LightGBM), comparing their performance. Our fndings highlight the signifcance of production scale, team structure, and movie themes in identifying potential high-return opportunities for investors.

The ever-evolving ways attacker continues to im prove their phishing techniques to bypass existing state-of-the-art phishing detection methods pose a mountain of challenges to researchers in both industry and academia research due to the inability of current approaches to detect complex phishing attack. Thus, current anti-phishing methods remain vulnerable to complex phishing because of the increasingly sophistication tactics adopted by attacker coupled with the rate at which new tactics are being developed to evade detection. In this research, we proposed an adaptable framework that combines Deep learning and Randon Forest to read images, synthesize speech from deep-fake videos, and natural language processing at various predictions layered to significantly increase the performance of machine learning models for phishing attack detection.

Accurate agricultural commodity price models enable efficient allocation of limited natural resources, leading to improved sustainability in agriculture. Because of climate change, price volatility and uncertainty for the sector are expected to increase in the future, increasing the need for improved price modeling. With the emergence of machine learning (ML) algorithms, novel tools are now available to enhance the modeling of agricultural commodity prices. This research uses both univariate and multivariate ML techniques to perform probabilistic price prediction modelling for the Canadian beef industry, taking into account beef production, commodity market, and international trade features to enhance accuracy. We apply multivariate algorithms by using support vector regression (SVR), random forest (RF), and adaboost (AB) and univariate algorithms by using autoregressive integrated moving average (ARIMA), seasonal ARIMA (SARIMA), and the seasonal autoregressive integrated moving average with exogenous factors (SARIMAX). We apply these models to monthly fed steer price data between January 2005 and September 2023 and compare predicted prices with observed prices using several validation metrics. The outcomes indicate that both random forest (RF) and adaboost (AB) in multivariate modeling show superior performance in accurately predicting Alberta fed steer prices in comparison to other algorithms. To better account for the variance of the best model performance, we subsequently adopted a probabilistic approach by considering uncertainty in our best-selected ML model. The beef industry can use these improved price models to minimize resource waste and inefficiency in the sector and improve the long-term sustainability prospects for beef producers in Canada.

The Yellow River Basin (YRB) is a crucial ecological zone and an environmentally vulnerable re-gion in China. Understanding the temporal and spatial trends of terraced-field areas (TRAs) and the factors underlying them in the YRB is essential for improving land use, conserving water re-sources, promoting biodiversity, and preserving cultural heritage. In this study, we employed ma-chine learning on the Google Earth Engine (GEE) platform to obtain spatial distribution images of TRAs from 1990 to 2020 using Landsat 5 (1990－2010) and Landsat 8 (2015－2020) remote sens-ing data. The GeoDa software platform was used for spatial autocorrelation analysis, revealing distinct spatial clustering patterns. Mixed linear and random forest models were constructed to identify the driving force factors behind TRA changes. The research findings reveal that TRAs were primarily concentrated in the upper and middle reaches of the YRB, encompassing provinc-es such as Shaanxi, Shanxi, Qinghai, and Gansu, with areas exceeding 40,000 km2, whereas other provinces had TRAs of less than 30,000 km2 in total. The TRAs exhibited a relatively stable trend, with provinces such as Gansu, Qinghai, and Shaanxi showing an overall upward trajectory. Conversely, Shanxi and Inner Mongolia demonstrated an overall declining trend. When com-pared with other provinces, the variations in TRAs in Ningxia, Shandong, Sichuan, and Henan appeared to be more stable. The linear mixed model (LMM) revealed that farmland, shrubs, and grassland had significant positive effects on the TRA, explaining 41.6% of the variance. The ran-dom forest model also indicated positive effects for these factors, with high R² values of 0.983 and 0.86 for the training and testing sets, respectively, thus outperforming the LMM. The findings of this study can contribute to the restoration of the YRB's ecosystem and support sustainable devel-opment. The insights gained will be valuable for policymaking and decision support in soil and water conservation, agricultural planning, and environmental protection in the region.

In Ultra-wideband (UWB)-based wireless ranging or distance measurement, differentiation between line-of-sight~(LOS), non-line-of-sight~(NLOS), and multi-path (MP) conditions are important for precise indoor localization. This is because the accuracy of the reported measured distance in UWB ranging systems is directly affected by the measurement conditions (LOS, NLOS or MP). However, the major contributions in literature only address the binary classification between LOS and NLOS in UWB ranging systems. The MP condition is usually ignored. In fact, the MP condition also has a significant impact on the ranging errors of the UWB compared to the direct LOS measurement results. Though, the magnitudes of the error contained in MP conditions are generally lower than completely blocked NLOS scenarios. This paper addresses machine learning techniques for identification of the mentioned three classes (LOS, NLOS, and MP) in the UWB indoor localization system using an experimental data-set. The data-set was collected in different conditions at different scenarios in indoor environments. Using the collected real measurement data, we compare three machine learning (ML) classifiers, i.e., support vector machine (SVM), random forest (RF) based on an ensemble learning method, and multilayer perceptron (MLP) based on a deep artificial neural network, in terms of their performance. The results show that applying ML methods in UWB ranging systems are effective in identification of the above-mentioned three classes. In specific, the overall accuracy reaches up to 91.9% in the best-case scenario and 72.9% in the worst-case scenario. Regarding the F1-score, it is 0.92 in the best-case and 0.69 in the worst-case scenario. For reproducible results and further exploration, we (will) provide the publicly accessible experimental research data discussed in this paper at PUB - Publications at Bielefeld University. The evaluations of the three classifiers are conducted using the open-source python machine learning library scikit-learn.

Delineating the cropping area of cocoa agroforests is a major challenge for quantifying the contribution of the land use expansion to tropical deforestation. Discriminating cocoa agroforests from tropical transition forests using multi-spectral optical images is difficult due to a similarity in the spectral characteristics of their canopy; moreover, optical sensors are largely impeded by the frequent cloud cover in the tropics. This study explores multi-season Sentinel-1 C-band SAR image to discriminate cocoa agroforests from transition forests for a heterogeneous landscape in central Cameroon. We use an ensemble classifier, random forest, to average SAR image texture features of GLCM (Grey Level Co-occurrence Matrix) across seasons; next, we compare classification performance with results from RapidEye optical data. Moreover, we assess the performance of GLCM texture feature extraction at four different grey level quantization: 32bits, 8bits, 6bits, and 4bits. The classification overall accuracy (OA) of texture-based maps outperformed that from an optical image; the highest OA of 88.8% was recorded at 6bits grey level. This quantization level, in comparison to the initial 32bits in SAR images, reduced the class prediction error by 2.9%. Although this prediction gain may be large for the landscape area, the resultant thematic map reveals the decrease and fragmentation of forest cover by cocoa agroforests. According to our classification validation, the Shannon entropy (H) or uncertainty provides a reliable validation for class predictions and reveals detail inference for discriminating inherently heterogeneous vegetation categories. The texture-based classification achieved a reliable accuracy considering the heterogeneity of the landscape and vegetation classes.

We aimed to develop a non-linear regression model that could predict the fat fraction of the liver (USFF), similar to magnetic resonance imaging proton density fat fraction (MRI-PDFF), based on quantitative ultrasound (QUS) parameters. We measured and retrospectively collected the ultrasound attenuation coefficient (AC), backscatter-distribution coefficient (BSC-D), and liver stiffness (LS) using shear wave elastography (SWE) in 90 patients with clinically suspected non-alcoholic fatty liver disease (NAFLD) and 51 patients with clinically suspected metabolic-associated fatty liver disease (MAFLD). The MRI-PDFF was also measured in all patients within a month of the ultrasound scan. In the linear regression analysis, only AC and BSC-D showed a significant association with MRI-PDFF. Therefore, we developed prediction models using non-linear least-squares analysis to estimate MRI-PDFF based on the AC and BSC-D parameters. We fitted the models on the NAFLD dataset and evaluated their performance in 3-fold cross-validation repeated five times. We decided to use the model based on both parameters to estimate the ultrasound fat fraction (USFF). The correlation between USFF and MRI-PDFF was strong in NAFLD and very strong in MAFLD. According to a receiver operating characteristics (ROC) analysis, USFF could differentiate between <5% vs. ≥ 5% and < 10% vs. ≥ 10% MRI-PDFF steatosis with excellent, 0.97 and 0.91 area under the curve (AUC) accuracy in the NAFLD and with AUCs of 0.99 and 0.96 in the MAFLD groups. In conclusion, USFF calculated from QUS parameters is an accurate method to quantify liver fat fraction and to diagnose ≥ 5% and ≥ 10% steatosis in both NAFLD and MAFLD. Therefore, USFF can be an ideal non-invasive screening tool for patients with NAFLD and MAFLD risk factors.

The aggregate aftermath of persistent inflammation in patients with inflammatory bowel disease (IBD) places them at increased risk for the advancement to colitis-associated colorectal cancer (CACRC). CACRC is preceded by IBD, the highly heterogenous, pharmacologically incurable, pertinacious, reverting/worsening, and immune-mediated inflammatory pathologies of the colon and rectum. The molecular and immunological basis of CACRC is highly correlated with the length/duration and stringency/severity of colon inflammation predisposed by the exogenous/free hemoglobin alpha chain (HbαC) the byproduct of infiltrating immune cells, extravasated erythrocytes, and macrophage erythrophagocytosis. The exogenous free HbαC prompts oxygen-free radical-arbitrated DNA damage (DNAD) through increased cellular reactive oxygen species (ROS) exacerbated by decreased tissue antioxidant defenses. Mitigation of Fenton reaction via pharmaceutical therapy would attenuate the ROS, promote apoptosis, DNAD repair, and subsequent prevent the incidence of CACRC. Three pharmaceutical options that attenuate hemoglobin toxicity include haptoglobin, deferoxamine, and flavonoids (vitamins C/E). Haptoglobin’s clearance rare from plasma is inversely correlated with its size; the smaller the size, the faster the clearance. Thus, the administration of Hp1-1 may prove to be beneficial. Furthermore, deferoxamine’s hydrophilic structure limits its ability to cross cell membranes. Thus, it may be beneficial if administered intracellularly to avoid the higher plasma concentrations and longer incubation periods associated with extracellular administration. Finally, the effectiveness of flavonoids and natural herb antioxidants is associated with high reactivity of hydroxyl substituents. Multiple analyses are currently underway to assess the clinical context of CACRC and outline the molecular basis of HbαC-induced ROS pathogenesis by exposing colonocytes and/or colonoids to HbαC. These cells are then treated with haptoglobin, deferoxamine (DFO), and flavonoids in order to separate free HbαC and measure their impact on hydroxyl radical formation therapies. The molecular pathogenesis of sporadic colorectal cancer (SCRC) i.e., “inflammation-dysplasia-carcinoma” progression sequence is well described, but the immunopathogenesis of CACRC herein reviewed is broadly still in prodromal stage/phase to be validated and understood. Therefore this timely review outlines the molecular and immunological basis of disease pathogenesis and the pharmaceutical intervention as a protective measure for CACRC.

Severe seasonal droughts driven by global climate change significantly alter the cycling of carbon and nutrients in forest ecosystems through disturbing plant physiology and phenology, soil physicochemical properties, and soil microbial activity. While, the investigation into the impacts of floor mass and plant roots on soil microbial biomass within the context of recurrent seasonal droughts is still rare. To investigate the environmental determinants governing soil microbial biomass with the escalating severity of seasonal droughts, we conducted a study on soil microbial biomass in a comprehensive block experiment, which was manipulated to explore the interplay between floor mass and plant roots contributions, crossed with a factorial irrigation treatment, in a montane subtropical moist evergreen broad–leaved forest of southwestern China. Within control and watered plots, we respectively established four experimental subplots with plant roots and floor mass included (R+F+), plant roots included but floor mass excluded (R+F−), plant roots excluded but floor mass included (R−F+), and plant roots and floor mass excluded (R−F−) from June 2019 to May 2023. The study results revealed that soil microbial biomass as well as soil moisture, floor mass, and plant roots showed an apparent single–hump modal within one year. In the comparative analysis of soil microbial biomass fluctuation amplitudes across control and watered plots, a discernible disparity was observed, indicating that significant differences in microbial biomass dynamics between the respective experimental conditions. The pooled data revealed a statistically significant influence of seasonal drought, floor mass, plant roots, and their reciprocal interactions on the soil microbial biomass, highlighting these factors as pivotal determinants of microbial community dynamics. This study elucidates the interactive regulatory mechanisms by which seasonal drought, floor mass, and plant roots collectively modulate soil microbial biomass within tropical and subtropical forests, offering insights into the complex ecological processes governing microbial community dynamics. This interactive regulation might influence the trajectory of plant species and soil microbial communities, facilitating their adaptive development and evolutionary responses.

This article explores the use of vegetation indices (VIs) in remote sensing for estimating standing wood volume and volume increments in forest ecosystems. The study investigates the correlations between various VIs and wood volume across different forest stand age classes, focusing on the effectiveness of commonly used indices such as Squared Ratio Simple Red Edge (SQSR) and Red-Edge Ratio Vegetation Index (RERVI), as well as other typical indicators including Enhanced Vegetation Index (EVI), Red-Edge EVI (RE-EVI), and Green Normalized Difference Vegetation Index (GNDVI). Additionally, the analysis considers a pair of indicators based on blue and near-infrared channels, resembling the Blue-normalized Difference Vegetation Index (BNDVI). The research reveals that quadratic and power-law models of SQSR and RERVI demonstrate higher correlations (>0.40) with wood volume, especially in younger forest stands (>0.80). Furthermore, the study highlights the potential of NDII and NDWI indices for estimating volume increments in certain classes of forest stands (0.70). However, correlations vary across different forest stand classes, suggesting the need for further investigation into the reliability of VIs for monitoring wood volume and volume increments in forest ecosystems. Overall, this research contributes to the understanding of the applicability of VIs in remote sensing for forest management and ecosystem monitoring.

Cervical Cancer affects a large part of female population that makes the prediction of this disease based on Machine Learning (ML) by outmost importance. ML algorithms can be integrated in complex intelligent agent-based systems that can offer decision support to the resident medical doctors or even to experienced medical doctors. For instance can be mentioned the situation when an experienced medical doctor diagnose a case but he/she needs expertise support that is related to another medical specialty. Data imbalance is frequent in healthcare data and has a negative influencing effect in making predictions using ML algorithms. Cancer data generally and cervical cancer data particularly are frequently imbalanced. Based on this fact the study of data imbalance impact on diverse state-of-the-art ML prediction algorithms is important. This research subject is also motivated by the fact that in many research are presented experimental evaluations of algorithms without characterization of the data on that they have been applied. Such characterizations could give clear indication to other researchers regarding the applicability of the algorithms on their specific data. Specifically, if the data have the respective characteristics than are expectable the same performance evaluation results like those in the reported research. For the study we chosed a messy real-life Cervical Cancer dataset available in a recognized data repository included a large amount of missing and noisy values. To identify the best imbalanced technique for this medical dataset, it is compared the performance of eleven important resampling methods combined with the following state-of-the-art ML models: K-Nearest Neighbours (with k values of 2 and 3), binary Logistic Regression, and Random Forest, that are frequently applied in prediction types of researches in healthcare. The studied resampling methods includes seven undersampling methods namely Condensed Nearest Neighbour, Tomek Links, Edited Nearest Neighbours, Repeated Edited Nearest Neighbours, All K-Nearest Neighbours, NearMiss, Neighbourhood Cleaning Rule, and Instance Hardness Threshold, and four oversampling methods namely Synthetic Minority Oversampling Technique (SMOTE), Adaptive Synthetic Sampling Approach for Imbalanced Learning SMOTE, Support Vector Machine SMOTE and Borderline SMOTE. In the case of the dataset for the confidence interval with 95% confidence level was between 9.23 and 16.22, while the imbalance ratio is 12.73. The obtained results show that resampling methods help the learning models to improve the classification ability of cervical cancer. The applied oversampling techniques generally showed better results than undersampling methods. In the case of Logistic Regression classifier had the highest impact on balanced techniques, while Random Forest had promising performance, even before balancing techniques and KNN2 was generally better than KNN3.

Human-to-human communication via the computer is mainly done using a keyboard or microphone. In the field of Virtual Reality (VR), where the most immersive experience possible is desired, the use of a keyboard contradicts this goal, while the use of a microphone is not always desirable (e.g. silent commands during task force training) or simply not possible (e.g. if the user has a hearing loss). Data gloves help to increase immersion within the VR as they correspond to our natural interaction. At the same time, they offer the possibility to accurately capture hand shapes, such as those used in non-verbal communication (e.g. thumbs up, okay gesture, ...) and in sign language. In this paper, we present a hand shape recognition system using Manus Prime X data gloves, including data acquisition, data preprocessing, and data classification to enable nonverbal communication within VR. We investigate the impact on accuracy and classification time of using an Outlier Detection and a Feature Selection approach in our data preprocessing. To obtain a more generalized approach, we also studied the impact of artificial Data Augmentation, i.e., we create new artificial data from the recorded and filtered data to augment the training dataset. With our approach, 56 different hand shapes could be distinguished with an accuracy of up to 93.28%. With a reduced number of 27 hand shapes, an accuracy of up to 95.55% could be achieved. Voting Meta-Classifier (VL2) has proven to be the most accurate, albeit slowest, classifier. A good alternative is Random Forest (RF), which was even able to achieve better accuracy values in a few cases and was generally somewhat faster. Outlier Detection has proven to be a effective approach, especially in improving classification time. Overall, we have shown that our hand shape recognition system using data gloves is suitable for communication within VR.

As renewable energy generation prediction systems have been introduced into the energy trading market, accurate prediction of solar photovoltaic (PV) energy generation has become a crucial challenge for ensuring stable trade of variable energy. Therefore, it is essential to quantitatively investigate and analyze current prediction technology and develop more advanced prediction systems. In this study, three models for predicting PV energy generation were investigated. Multiple regression, random forest, and gradient boosting machine (GBM) models were con-structed for 15 utility-scale power plants throughout South Korea. Model performance was evaluated in terms of root mean square error (RMSE) and mean absolute error (MAE). The multiple regression-based model had an RMSE of 12.00% and MAE of 9.06%. The random forest model had an RMSE of 11.69% and MAE of 8.61%. The GBM model had an RMSE of 11.34% and MAE of 8.18%, indicating its superior accuracy among the three models in predicting PV energy generation. These findings provide valuable insights that may contribute to the development of advanced PV energy generation models and their application in grid operations.

The identification of informal settlements in urban areas is an important step in developing and implementing pro-poor urban policies. Understanding when, where and who lives inside informal settlements is critical to efforts to improve their resilience. This study aims to analyse the capability of machine-learning (ML) methods to map informal areas in Jeddah, Saudi Arabia, using very-high-resolution (VHR) imagery and terrain data. Fourteen indicators of settlement characteristics were derived and mapped using an object-based ML approach and VHR imagery. These indicators were categorised according to three different spatial levels: environ, settlement and object. The most useful indicators for prediction were found to be density and texture measures, (with random forest (RF) relative importance measures of over 25% and 23% respectively). The success of this approach was evaluated using a small, fully independent validation dataset. Informal areas were mapped with an overall accuracy of 91%. Object-based ML as a hybrid approach performed better (8%) than object-based image analysis alone due to its ability to encompass all available geospatial levels.

Spinocerebellar ataxia type 2 (SCA2) is caused by polyglutamine expansion in Ataxin-2 (ATXN2). This factor binds RNA/proteins to modify metabolism after stress, and to control calcium (Ca2+) homeostasis after stimuli, thus exerting crucial neuroprotection for cerebellar ataxias and corticospinal motor neuron degeneration. Our Atxn2-CAG100-Knock-In mouse faithfully models features observed in patients at pre-onset, early and terminal stages. Here, its cerebellar global RNA profiling revealed downregulation of signaling cascades to precede motor deficits. Validation work at mRNA/protein level defined alterations that were independent of constant physiological ATXN2 functions, but specific for RNA/aggregation toxicity, and progressive across the short lifespan. Earliest changes were detected at 3 months among Ca2+ channels/transporters (Itpr1, Ryr3, Atp2a2, Atp2a3, Trpc3), IP3 metabolism (Plcg1, Inpp5a, Itpka), and Ca2+-Calmodulin dependent kinases (Camk2a, Camk4). CaMKIV–Sam68 control over alternative splicing of Nrxn1, an adhesion component of glutamatergic synapses between granule and Purkinje neurons, was found affected. Systematic screening of pre/post-synapse components, with dendrite morphology assessment, suggested early impairment of CamKIIα abundance together with weakening of parallel fiber connectivity. These data reveal molecular changes due to ATXN2 pathology, impacting communication and excitability of cerebellar neurons. Discovery of such risk versus progression markers improves the assessment of pre-symptomatic treatments in SCA2 and related disorders.

This article compares the predictive capabilities of six models namely the Linear Discriminant Analysis (LDA), Logistic Regression (LR), Support Vector Machine (SVM), XGBoost, Random Forest (RF) and Deep Neural Network (DNN) to predict the default behaviour of credit card holders in Taiwan using data from the UCI machine learning database. Python programming language was used for data analysis. Statistical methods were compared with machine learning algorithms using the confusion matrix measured in metric terms of prediction accuracy, sensitivity, specificity, precision, G-mean, F1 score, ROC and AUC. The dataset contains 30,000 credit card user’s information with 6636 default observations and 23,364 non-default cases. The study results found that modern machine learning methods outperformed traditional statistical methods in terms of predictive performance measured by F1 score, G-mean and AUC. Traditional methods like logistic regression were marginally better than linear discriminant analysis and support vector machines in terms of predictive performance measured by area under the receiver operating characteristic curve. In the modern machine learning methods, deep neural networks were better in most of the predictive performance metrics than XGBoost and Random Forest methods.

Several factors impact the durability of concrete bridge decks, including traffic loads, fatigue, temperature changes, environmental stress, and maintenance activities. Detecting problems such as corrosion, delamination, or concrete degradation early on can lower maintenance costs. Nondestructive evaluation (NDE) techniques can detect these issues at early stages. Each NDE method, meanwhile, has limitations that reduce the accuracy of the assessment. In this study, multiple NDE technologies were combined with machine learning algorithms to improve the interpretation of half-cell potential (HCP) and electrical resistivity (ER) measurements. A parametric study was performed to analyze the influence of five parameters on HCP and ER measurements, such as degree of saturation, corrosion length, delamination depth, concrete cover, and moisture condition of delamination. The results were obtained through finite element simulations and used to build two machine learning algorithms, a classification algorithm and a regression algorithm, based on the Random Forest methodology. The algorithms were tested using data collected from a bridge deck in the BEAST® facility. Both machine learning algorithms were effective in improving the interpretation of ER and HCP measurements using data from multiple NDE technologies.

Satellite-based data classification performance remains a challenge for research community in the field of land use/land cover mapping. Here we investigated supervised per-pixel classifications performance under different scenarios, based on single and seasonal multispectral data combina-tions of different sensors (Landsat-8 OLI and Sentinel-2 MSI). In case of Landsat, seasonal spectral indices (EVI and NDMI) were included. A typical Mediterranean watershed with a complex landscape comprised of various forest and wetland ecosystems, crops, artificial surfaces, and lake water was selected to test our approach. All available geospatial data from national databases (Forest Map, LPIS, Natura2000 habitats, cadastral parcels, etc.) are used as ancillary data for clas-sification training and validation. We examined and compared the performance of ML, RF, KNN and SVM classifiers under different scenarios for land use/land cover mapping, according to Co-pernicus Land Cover nomenclature. In total, eight land use/land cover classes were identified in Landsat-8 OLI and nine in Sentinel-2 MSI for an acceptable overall accuracy over 85%. A com-parison of the overall classification accuracies shows that Sentinel-2 overall accuracy was slightly higher than Landsat-8 (96.68% vs. 93.02%). Respectively, the best-performed algorithm was ML in Sentinel-2 while in Landsat-8 was KNN. However, machine-learning algorithms have similar results regardless the type of sensor. We concluded that best classification performances achieved using seasonal multispectral data. Future research should be oriented towards integrating time-series multispectral data of different sensors and geospatial ancillary data for land use/land cover mapping.

Tree species composition is an important key element for biodiversity and sustainable forest management, and hyperspectral data provide detailed spectral information, which can be used for tree species classification. There are two main challenges for using hyperspectral imagery: a) Hughes phenomena, meaning by increasing the number of bands in hyperspectral imagery, the number of required classification samples would increase exponentially, and b) in a more complex environment, such as riparian mixed forest, focusing on spectral variability per pixel may not be adequate for definability of tree species. Therefore, the focus of this study is to assess spectral-spatial dimensionality reduction of airborne hyperspectral imagery by using minim noise fraction (MNF) transformation, and object-based image analysis (OBIA). An airborne prism experiment (APEX) hyperspectral imagery was used. A study area was a riparian mixed forest located along the Salzach river, and six tree species including Picea abies, Populus (canadensis and balsamifera), Fraxinus excelsior, Alnus incana, and Salix alba were selected. A machine learning algorithm random forest (RF) was used to train and apply a prediction model for classification. Using a spectral dimensionality reduced APEX, a pixel-level classification was also done. According to a confusion matrix, the object-level classification of MNF-derived components achieved the overall accuracy of 85 %, and kappa coefficient of 0.805. The performance of classes according to producer’s accuracy varied between 80% for Fraxinus excelsior, Alnus incana, and Populus canadensis to 90% for Salix alba and Picea abies. Comparison the results to a pixel-level classification, showed a better performance of object-level classification (an overall accuracy of 63% and Kappa coefficient of 0.559 were achieved for pixel-level classification). The performance of classes using pixel-based classification varied 45 % for Alnus incana to 80% for Picea abies. In general, Spectral-spatial complexity reduction using MNF transformation and object-level classification yielded a statistically satisfactory results.

This paper presents a comparative analysis of several decision models for detecting Structured Query Language (SQL) injection attacks, which remain one of the most prevalent and serious security threats to web applications. SQL injection enables attackers to exploit databases, gaining unauthorized access, and manipulating data. Traditional detection methods often struggle due to the constantly evolving nature of these attacks, the increasing complexity of modern web applications, and the lack of transparency in the decision-making processes of machine learning models. To address these challenges, we evaluated the performance of various models, including Decision Tree, Random Forest, XGBoost, AdaBoost, Gradient Boosting Decision Tree (GBDT), and Histogram Gradient Boosting Decision Tree (HGBDT), using a comprehensive SQL injection dataset. The primary motivation behind our approach is to leverage the strengths of ensemble learning and boosting techniques to enhance detection accuracy and robustness against SQL injection attacks. By systematically comparing these models, we aim to identify the most effective algorithms for SQL injection detection systems. Our experiments show that Decision Tree, Random Forest, and AdaBoost achieved the highest performance, with an accuracy of 99.50% and an F1 score of 99.33%. Additionally, we applied SHapley Additive exPlanations (SHAP) and Local Interpretable Model-agnostic Explanations (LIME) for local explainability, illustrating how each model classifies normal and attack cases. This transparency enhances the trustworthiness of our approach in detecting SQL injection attacks. These findings highlight the potential of ensemble methods to provide reliable and efficient solutions for detecting SQL injection attacks, thereby improving the security of web applications.

Xyleborus beetles, a diverse group of ambrosia beetles, present significant challenges to forestry and agriculture due to their damaging burrowing behavior and symbiotic relationships with fungi. This review synthesizes current knowledge on the biology, ecology, and management of Xyleborus. We explore the beetles' life cycle, reproductive strategies, habitat preferences, and feeding habits, emphasizing their ecological and economic impacts. Control and management strategies, including preventive measures, chemical and biological control, and integrated pest management (IPM), are critically evaluated. Recent advances in molecular genetics and behavioral studies offer insights into genetic diversity, population structure, and host selection mechanisms. Despite progress, managing Xyleborus effectively remains challenging. This review identifies future research needs and highlights innovative control methods, such as biopesticides and pheromone-based trapping systems.

Quaking aspen (Populus tremuloides Michx.) is an important deciduous species in western U.S. forests. Existing maps of aspen distribution are based on Landsat imagery and often miss small stands (&lt;0.09 ha or 30 m²), which rapidly regrow when managed or following disturbance. In this study, we present methods for deriving a new regional map of aspen forests using one year of Sentinel-1 (S1) and Sentinel-2 (S2) imagery in Google Earth Engine. We assessed the annual phenology of aspen in the Southern Rockies and leveraged the frequent temporal resolution of S1 and S2 to create ecologically relevant seasonal composites. Additionally, we derived spectral indices and radar textural features targeting the canopy structure, moisture, and chlorophyll content. Using spatial block cross-validation and Random Forests, we assessed the accuracy of different scenarios and selected the best performing set of features for classification. Comparisons were then made with existing landcover products across the study region. The resulting map improves on existing landcover products in both accuracy (average F1-score = 93.1%) and detection of smaller forest patches. These methods enable accurate mapping at spatial and temporal scales relevant to forest management for one of the most widely distributed tree species in North America.

In the dynamic landscape of the telecommunications industry, understanding and mitigating customer churn is paramount for maintaining competitive advantage and sustaining revenue streams. This comprehensive literature review delves into the intricacies of customer churn prediction, a crucial aspect in fortifying the bond between telecom businesses and their customers. The review methodically analyzes 11 articles published between 2000 and March 2024, spotlighting the strategic role of Customer Relationship Management and the prevalence of high churn rates that necessitate a shift from customer acquisition to retention strategies. Furthermore, the review navigates through the challenges inherent in churn prediction, including the complexities of managing large, imbalanced, and noisy datasets and the intricacies involved in model selection and optimization. Despite these challenges, the paper highlights the superior capability of machine learning techniques in handling complex datasets to provide accurate predictions, albeit acknowledging their practical complexities. The main aim of the study is to provide an overview, serving as a knowledge base for researchers interested in machine learning techniques within the realm of telecommunications.

Efforts to enhance quality control (QC) practices in chip seal construction have predominantly relied on single surface frictional metrics such as mean profile depth (MPD) or friction number. These metrics assess chip seal quality by targeting issues such as aggregate loss or excessive bleeding, which may yield low friction numbers or texture depths. However, aggregate loss particularly due to snowplow operations doesn't always result in slippery conditions and may lead to uneven surfaces. The correlation between higher MPD or friction number and superior chip seal quality isn't straightforward. This research introduces an innovative machine learning-based approach to enhance chip seal QC. Using a hybrid DBSCAN-Isolation Forest model, anomaly detection is conducted on a dataset comprising 183,794 20-meter MPD measurements from actual chip seal projects across six districts in Indiana. This results in typical 20 m-segment MPD ranges of [0.9, 1.9], [0.6, 2.1], [0.3, 1.3], [1.0, 1.7], [0.6, 1.9], and [1.0, 2.3] for the respective six districts in Indiana. A two-step QC procedure tailored for chip seal evaluation is proposed. The first step calculates outlier percentages across 1-mile segments, with an established limit of 25% outlier segments per wheel track. The second step assesses unqualified rates across projects, setting a threshold of 50% for 1-mile unqualified wheel track segments. While the results are data-specific, this framework offers pavement construction practitioners a foundational QC standard for chip seal projects.

The import and export of tree seed carries with it risks of inadvertent introduction of pests and pathogens to hitherto unaffected regions. Although trade in seed of specified trees is regulated, phytosanitary requirements for most tree species are minimal, even those related to the most important forest tree species in a given region. A better understanding of the microbiome associated with seed intended for commercial production or ornamental use, and their potential risk with the transport from the source origin of distributors, will help regulatory agencies implement measures to prevent new and emerging risks. In this study we used high throughput sequencing to show that highly diverse fungal communities were associated with seed of 14 different Pinus species obtained from seed banks (seed orchards) and retail sources (online distributors) in North America and Europe. Fungal diversity differed among the 23 seedlots tested. Community composition did not relate to the species of Pinus nor the country of origin. Assigned potential functions based on sequence identity using FUNGuild provided an overall understanding of the likely life strategies associated fungal OTUs. Of those sequences classified to a trophic level, 453 were plant pathogens, with the Dothideomycetes having the highest prevalence. The most common plant pathogens detected included Sydowia polyspora, Lasiodiplodia theobromae, Diplodia intermedia and Diplodia sapinea. The evidence presented here illustrates an urgent need for plant protection authorities, practitioners and the general public to recognize the potential risk of introducing harmful pathogens through innocent transport of seed.

Pine nuts are non-wood forest products (NWFP) with constantly growing market notwithstanding a series of phytosanitary issues and related trade problems. The aim of paper is to review the literature on the relationship between phytosanitary problems and trade development. Production and trade of pine nuts in Mediterranean Europe have been negatively affected by the spreading of Sphaeropsis sapinea (a fungus) associated to an adventive insect Leptoglossus occidentalis (fungal vector), with impacts on forest management activities, production and profitability and thus in value chain organization. Reduced availability of domestic production in markets with growing demand has stimulated the import of pine nuts. China has become a leading exporter of pine nuts, but its export is affected by a symptom associated to the nuts of some pine species: the ‘pine nut syndrome’ (PNS). Most of the studies embraced during the review are associated to PNS occurrence associated to the nuts of Pinus armandii. In the literature review we highlight the need for a comprehensive and interdisciplinary approach to the analysis of the pine nuts value chain organisation, where research on food properties and clinical toxicology be connected to breeding and forest management, forest pathology and entomology and trade development studies.

Bacterial infection of the lower respiratory tract frequently occurs in mechanically ventilated pa-tients and may develop into life-threatening conditions. Yet existing diagnostic methods have moderate sensitivity and specificity, which results in the overuse of broad-spectrum antibiotics administrated prophylactically. This study aims to evaluate the suitability of volatile bacterial me-tabolites for breath-based test diagnosing Ventilation-Associated Pneumonia (VAP). The in vitro experiments with pathogenic microorganisms most prevalent in VAP patients (i.e., Acinetobacter baumannii, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa) were performed to identify bacteria-derived metabolites using specially designed cultivation system enabling headspace sam-pling for GC-MS analysis. Thirty-nine compounds were found to be significantly metabolized by tested species and, therefore, selected for monitoring in the exhaled breath of critically ill, me-chanically ventilated (MV) patients. The emission of volatiles from medical respiratory devices was investigated to estimate the risk of spoiling breath results with exogenous pollutants. Bacterial metabolites were then evaluated to differentiate VAP patients from non-infected MV controls using receiver operating characteristic (ROC) analysis, with AUC, sensitivity, and specificity calculated. Nine bacterial metabolites that passed verification through a non-parametric ANOVA test for significance and LASSO penalization were identified as key discriminators between VAP and non-VAP patients. The diagnostic model achieved an AUC of 0.893, with sensitivity and specificity values of 87% and 82.4%, respectively, being competitive with traditional methods. Further vali-dation could solidify its clinical utility in critical care settings.

To minimise environmental impact, avoid regulatory penalties, and improve competitiveness, energy-intensive manufacturing firms require accurate forecasts of their energy consumption so that precautionary and mitigation measures can be taken. Deep learning is widely touted as a superior analytical technique to traditional artificial neural networks, machine learning, and other classical time series models due to its high dimensionality and problem solving capabilities. Despite this, research on its application in demand-side energy forecasting is limited. We compare two benchmarks (Autoregressive Integrated Moving Average (ARIMA), and an existing manual technique used at the case site) against three deep learning models (simple Recurrent Neural Networks (RNN), Long Short-Term Memory (LSTM), and Gated Recurrent Unit (GRU)) and three machine learning models (Support Vector Regression (SVM), Random Forest, and K-Nearest Neighbors (KNN)) for short term load forecasting (STLF) using data from a Brazilian thermoplastic resin manufacturing plant. We use the grid search method to identify the best configurations for each model, and then use Diebold-Mariano testing to confirm the results. Results suggests that the legacy approach used at the case site is the worst performing, and that the GRU model outperformed all other models tested.

We conducted a retrospective analysis of the immunohistochemical (IHC) staining GRP94, an ER stress protein on large B-cell lymphoma (LBCL) cells, intracellular p53, and 15 factors involved in the metabolism of CHOP regimen :AKR1C3 (HO-metabolism), CYP3A4 (CHOP-metabolism) and HO efflux pumps (MDR1, MRP1). The study subjects were 42 patients with LBCL at our hospital. IHC staining using antibodies against the 17 factors. The odds ratios by logistic regression analysis using a dichotomous variable of CR, non-CR/relapse were statistically significant: for MDR1, MRP1, and AKR1C3. The overall survival (OS) after R-CHOP was compared by the log-rank test. The four groups showed that Very good (5-year OS, 100%) consisted of 4 patients showed negative IHC staining for both GRP94 and CYP3A4. Very poor (1-year OS, 0%), consisted of 3 patients who showed positive results of IHC for both GRP94 and CYP3A4. The remaining 35 patients comprised two subgroups: Good (5-year OS 60-80%.): 15 patients who showed negative staining for both MDR1 and AKR1C3, Poor: 5-year OS, 10-20%) :20 patients who showed positive staining for either MDR, AKR1C3,,MRP1 or p53. The Histological Prognostic Index (HPI) (The four groups: Very poor, Poor, Good, and Very pood) is a breakthrough method for stratifying patients based on factors involved in the development of treatment resistance.

In light of the critical and emerging global challenge posed by the increasing frequency and severity of heatwaves in recent decades, this study demonstrates a practical and robust workflow that bridges the gap in understanding how the effective integration of remote sensing data with ground-based observations can facilitate a comprehensive assessment of spatiotemporal heatwave patterns and trends in the central region of Thailand. This research transcends traditional methods, utilizing air temperature (Tair) and satellite-derived land surface temperature (LST) data from 1981 to 2019. Results exhibit that an analysis of Tair indicates increasing daytime heatwaves in peri-urban areas, with significant trends in heatwave number (HWN), heatwave frequency (HWF), and heatwave duration (HWD), heatwave amplitude (HWA) rises in urban settings while nighttime heatwaves significantly intensify in rural locales. Notably, LST trends reveal varied patterns, with peri-urban areas showing marked daytime increases in heatwave magnitude (HWM), HWA, and HWF. Correlation analysis (p=0.05) highlights strong daytime associations between Tair and LST in rural (HWN, HWF, HWD, r&gt;0.90) and peri-urban (HWM, HWA, r&gt;0.65) regions. Overall, this study advances approaches to the adaptable measurement and spatial-temporal pattern of heatwave-related risk areas, providing insights for decision-makers to develop sustainable practices and strategies for climate change adaptation.

Monitoring crown closure evolution using multi-temporal Light Detection and Ranging (LiDAR) surveys is a method that we expect to be increasingly adopted given the availability of LiDAR sensors and the accumulating survey archives. However, little attention was devoted to comparing crown closure estimates from independent surveys. Although survey parameters cannot be modified after the data collection, we speculate that the error associated to crown closure estimates comparison can be reduced by selecting optimal post-survey parameters. In this study, we compared crown closure estimates of three airborne LiDAR surveys from 2018 (40 pt/m²) used as a reference, and two lower-density surveys from 2016 (4.5 pt/m²) and 2018 (2 pt/m²). We studied the effect of the height threshold used to separate canopy points and the grid resolution, using skewness and variance of lagged difference of crown closure. Crown closure estimates using low height thresholds were more different across surveys, resulting in higher root mean squared error (RMSE), bias and more different variograms. Results show that optimal height threshold was 3 m and grid resolution was 25 m, although there was room for decision (RMSE of 7% and 5%, and bias of 4% and 0% for 2016 and 2018 low-density surveys).

Rotaviruses are highly infectious and typically transmitted by fecal-oral route via in the tropics and leading the cause of diarrheal deaths in children of developing countries, besides causing significant economic impacts like neonatal disease agents of domestic animals. This present report aims to present the clinical and diagnostic findings of two confirmed cases of rotavirus (RV) infection in orphaned Leopardus tigrinus (Schreber, 1775) and Leopardus pardalis (Linnaeus, 1758), the first register of the infection by group A rotavirus in these species. Both felids were rescued in the Pará State Amazon Brazil by the IBAMA (the Brazilian Institute of Environment and Renewable Natural Resources), and treated by veterinarians into intensive care ward in a public Environmental Park of Belém city. After the adaptation period to the quarantine, these animals showed non-specific symptoms of acute fulminant gastroenteritis. Rotavirus group A antigen was identified in blood and faecal samples of L. tigrinus analyzed by immunochromatography (ICG) and immunoassay methods (ELISA) at the Virology Laboratory of the Institute Evandro Chagas. The animals died within few days during the clinical exacerbation unresponsive to current treatment, its necropsies and histopathological analysis were performed in the Laboratory of Veterinary Pathology of the Federal Rural University of Amazonia (UFRA). Despite the compatible pathologic findings of rotavirus infection in both animals, the atypical hemorrhagic character was a curious finding, considering the presumed etiology.
