Uncertainty is the main source of risk of geological hazards in tunnel engineering. Uncertainty information not only affects the accuracy of evaluation results, but also affects the reliability of decision-making schemes. Therefore, it is necessary to evaluate and control the impact of uncertainty on risk. In this study, the problems in existing entropy-hazard model such as inefficient decision-making and failure of decision-making are analysed, and an improved uncertainty evaluation and control process are proposed. Then the tolerance cost, the key factor in the decision-making model, is also discussed. It is considered that the amount of change in risk value (R1) can better reflect the psychological behaviour of decision-makers. Thirdly, common attribute decision models, such as the expected utility-entropy model, are analysed, and then the viewpoint of different types of decision-making issues that require different decision methods is proposed. The well-known Allais paradox is explained by the proposed methods. Finally, the engineering application results show that the uncertainty control idea proposed here is accurate and effective. This research indicates a direction for further research into uncertainty, and risk control, issues affecting underground engineering works.

Technological innovation has led to the development of increasingly efficient and complex industrial plants. To manage this complexity, it is necessary to define an integrated vision of the socio-technological system that includes: technological, human and organizational component. Petrochemicals can be considered one of the most complex socio-technical systems that deserve special attention to high risk management, especially during the emergency conditions. Traditional safety management models only consider static systems, while new resilience engineering models evaluate the performance variability developed between different actions. One of the recent development methods is the Functional Resonance Analysis Method (FRAM) that identifies the pairs between the functions. FRAM unfortunately is a qualitative model, this research integrates this model with the Performance Shaping Factors (PSFs) and with the Bayesian approach to identify the performance variability of the system. The analysis aims to develop a system that improves safety analysis. The proposed model is applied in a case study of an emergency in a petrochemical company.

The paper discusses the mechanism of montmorillonite structure alteration and bentonites properties modification (on the example of samples from clay deposit Taganka, Kazakhstan) due to the thermochemical treatment (treatment with inorganic acid solutions at different temperatures, concentrations and reaction times). With the use of the suit of methods certain processes were distinguished: transformation of montmorillonite structure, which appears in the leaching of interlayer and octahedral cations, protonation of the interlayer and OH groups at octahedral sheets. Changes in the structure of the 2:1 layer of montmorillonite and its interlayer result in significant changes in the properties – reduction of cation exchange capacity and an increase of specific surface area. The results of the work showed that bentonite clays retain a significant portion of its adsorption properties even after the long term and intense thermochemical treatment (6M HNO3, 60°C, 108 hours)

MoS2 has long been considered as a promising catalyst for hydrogen production. At present, there are many strategies to further improve its catalytic performance, such as edge engineering, defect engineering, phase engineering and so on. However, at present, there is still a great deal of controversy about the mechanism of MoS2 catalytic hydrogen production. For example, it is generally believed that the base plane of MoS2 is inert, but it has been reported that the inert base plane can undergo a transient phase transition in the catalytic process to play the catalytic role, which is contrary to the common understanding that the catalytic activity is only at the edge. Therefore, it is necessary to further understand the mechanism of MoS2 catalytic hydrogen production. In this article, we summarized the latest research progress on the catalytic hydrogen production of MoS2, which is of great significance for revisited the mechanism of MoS2 catalytic hydrogen production.

Industry 4.0 is an industrial paradigm that is causing changes in form and substance in factories, companies and businesses around the world, and is impacting work and education in general. In fact, the disruptive technologies that frame the fourth industrial revolution have the potential to improve and optimize manufacturing processes and the entire value chain, which can lead to an exponential evolution in the production and distribution of goods and services. All these changes imply that the fields of engineering knowledge must be oriented towards the concept of Industry 4.0, for example Mechanical Engineering. The development of various physical assets that are used by cyber-physical systems and digital twins is based on Mechanics. However, the specialized literature on Industry 4.0 says little about the importance of Mechanics in the new industrial era and more importance is given to the evolution of Information and Communication Technologies and Artificial Intelligence. This article presents a frame of reference about the importance of Mechanical Engineering in Industry 4.0 and proposes an extension to the concept of Mechanics 4.0, recently defined as the relationship between Mechanics and Artificial Intelligence. For the analysis of Mechanical Engineering in Industry 4.0, the criteria of the four driving forces that defined Mechanics in the third industrial revolution was used. An analysis of Mechanical Engineering Education in Industry 4.0 is presented, and the concept of Mechanical Engineering 4.0 Education is improved. Finally, the importance of making changes in the educational models of engineering education is described.

So, we may need to urge relying on computer simulation results without any experimental validation to accelerate engineering innovations. In addition, using high-fidelity computer simulation for engineering innovation acceleration without validation will be promoted if it is credited as a new classification. This subject and a related justification's classification in engineering have been discussed.

This paper reviews the measurement challenges associated with 3D scanning techniques in civil engineering. It explores the practical aspects of scanning buildings and complex surfaces through various case studies. The paper details the conventional use of Terrestrial Laser Scanning (TLS) for reconstructing the technical documentation of a hall. It then describes an unconventional application of this technique for measuring an ETICS (External Thermal Insulation Composite System) wall, aiming to detect microdeformations caused by environmental factors controlled within a climatic chamber. The measurements of the insulated wall were subsequently repeated using a metrological grade laser scanner. Numerical data was analyzed with inspection engineering methods. This approach yielded qualitative and quantitative results. Although the qualitative results were consistent, the quantitative data revealed some inconsistencies. To address discrepancies in the quantitative data, a comparative analysis was performed, which highlighted critical findings and areas of divergence.

Engineering Education needs to be approached in different, innovative ways to meet the needs of society, learners, and education providers. This ranges from new educational programs and innovative pedagogies in individual classrooms, to online and immersive learning, to micro-credentials and modularity along with how to enable rapid adaptation for reskilling and upskilling, to entirely new pathways of restructuring education. Gamification is the application of game elements and digital game design techniques to non-game problems, such as learning challenges. It is about using game-based mechanics, aesthetics, and game thinking to enhance users’ engagement, motivate action, and support learning. At first view, the concept of gamification does not contain game design elements, but with the addition of additional elements such as the video game component that is applied in different contexts, the users are encouraged to participate in learning activities and complete learning challenges successfully. The first step in the process of designing a gamification activity is to think like a game designer. According to Cook (2013), any process can be gamified when the following requirements are covered: (i) the gamified activity is easy to learn, (ii) user progress can be measured, (iii) continuous feedback is provided to users.

The purpose of this study was to develop e-learning activities that integrated sustainability concepts and practices in process engineering education. Two blended courses were developed with two activities evaluated quantitatively and qualitatively to measure student engagement, quality of responses, and incorporation of sustainability in their arguments. Social network analysis and lexical analysis were used to assess students’ participation in discussions and peer reviews. In the online discussion, 97 comments were made averaging 120 words per comment. The participants averaged 3.88 comments, with the majority of comments exhibiting simple and complex argumentation, a deep reflection, and widespread use of terms associated with sustainability such as recycling, pollution, waste, and environment. Furthermore, evaluation of peer reviews revealed that the participants demonstrated they could identify errors and positives in an argument. Therefore, this study demonstrated that e-learning, particularly peer review and online discussion could help chemistry and engineering students understand sustainability.

In our long-term engineering practice, we have found that it is often not enough to use only engineering knowledge to solve the practical problems in the engineering. Therefore, we believe that in the education of engineering students, we should not only pay attention to the teaching of engineering knowledge, but also pay attention to the application of knowledge of Humanities in the engineering, for the students. Organization resilience indicates the capacity of an organization or system to return to its steady condition or exceed it after going through a disruptive event that disrupts its steady condition. Qatar's oil and gas sector has shown remarkable Resilience during the COVID-19 pandemic due to its preparedness and readiness to deal with such a disruption. As a lesson learned from the recent COVID-19 pandemic, local governmental institutions need to support national preparedness and Resilience to handle emergencies and unpredictable crises by learning from the success model of Qatar's oil and gas sector. This study presents the key Resilience Engineering Indicators (REIs) that make this sector resilient and validates the six Resilience Engineering indexes or dimensions adopted from [1], [2]& [3], which include top management commitment, speaking up culture, learning, awareness, being prepared, and flexibility. The study evaluated the performance of these REIs using a 5-point Likert Scale survey questionnaire based on the relevance to resilience dimensions. The results show 10 REIs contributing to the organization's resilience and the top four most important Resilient Dimensions (RDs). Moreover, this is the first study to evaluate and assess the organizational resilience level in Qatar's oil and gas sector. This study's results can be integrated into different organizations' strategies to improve the efforts to enhance national response to disturbances in governance.

It provides tools for dealing with the complexity and dynamics related to the optimisation of physical, natural, and virtual systems management. This paper presents a review of how multi-agent systems and complex networks theory are brought together to address Systems Engineering and management problems. In aerospace, robotics, network engineering and mechanical engineering, there exist complex and urgent challenges that can be abstracted into mathematical optimization problems. Therefore, exploring solutions to these optimization problems is a crucial task. Metaheuristic algorithms have been applied across various fields.The Snake Optimization Algorithm (SO) is an innovative metaheuristic method recognized for its efficient solving capabilities.However, this algorithm encounters challenges such as diminished search efficiency in the later stages and a propensity to become ensnared in local optima. To mitigate these issues, this paper introduces an enhanced Snake Optimization Algorithm (ISO). ISO integrates the RIME Algorithm(RIME)and introduces two effective enhancement strategies to improve its capability to evade local optima.To assess the effectiveness of ISO,the population distribution state was first measured using three algorithms, including Star Discrepancy, and the exploration and exploitation performance was analyzed using the CEC2017 test functions. Subsequently, utilized 23 benchmark functions for testing.The evaluation outcomes indicate that ISO exhibits outstanding performance in terms of convergence velocity and robustness.Additionally, it was applied to engineering fields including UAV path planning,Robot path planning, Wireless sensor network node deployment and Pressure vessel design.In comparison to the SO algorithm, the ISO algorithm demonstrates superior stability, converges earlier, and improves its ability to solve for optimal .These findings highlight the significant potential of the ISO algorithm in diverse test functions and a wide range of interdisciplinary problems.

Plato defined a quality as “some degree of perfection” [49]. Studies on quality have unveiled the internal and external quality factors [17] that significantly impact product quality [4]. Internal factors are invisible to the end customer and are dependent on overall work organisation by the development team [17]. This work organisation is called “the process” [11],[33]. In this article, we will discuss how to address the drone-dedicated software development process to support system verification and ensure product quality by supporting quality characteristics and therefore increase software development awareness and control. We will discuss the process's influence on overall work activities and relationships between those activities by addressing the software development process utilised in the drone-dedicated GNSS-denied navigation system development project. The result was observable after three weeks of starting the software development. It was focused on better team awareness, faster task completion, lower number of development mistakes, and better and faster change response than we observed in other projects that didn’t follow the predefined process.

(1) Background: Due to the high proportion of disadvantaged students in a rural school in Taiwan and the gap between students’ concepts and practices of environmental protection and sustainable energy, four science and mathematics teachers in this school planned an engineering-centered PjBL of sustainable energy curriculum in a Makers Club to enhance students’ creativity, engineering skills, practices of environmental protection and sustainable energy, and learning attitudes; (2) Methods: This study is four-year action research. Teachers and students initiated the idea from rebuilding an old fan in a classroom; (3) Results: The students in the Makers Club improved their engineering skills and created various green-power generation devices (evolved from ventilation ball generator, hydropower, ocean current power generators to tiny, 3D-printing wind power generators). They turned environmental protection and sustainable energy concepts into actions during practices and won awards from science and engineering fairs every year. This creative and supportive atmosphere spread from the club to the whole school and improved the students’ practices of environmental protection and learning attitudes after long-term implementation; (4) Conclusions: The design principles of the engineering-centered PjBL of sustainable energy curriculum played a critical role and were outlined at the end of the study.

Industrialization, intensive farming, rapid population growth and urbanization are the source of a large number of pollutants entering the environment. The current concentration of xenobiotics released into the environment exceeds its natural ability to decompose them. Enzymatic degradation of pollutants seems to be an environmentally friendly process. Due to the wide spectrum of substrate specificity, from inorganic compounds to high molecular weight organic compounds such as PAH or dyes, as well as favorable biochemical properties, laccase has been used in the biological removal of xenobiotics from the environment. It is important to understand the degradation mechanisms of pollutants and to evaluate the final products in terms of their toxicity. The laccase oxidizes the substrates with the simultaneous reduction of molecular oxygen to water, which is the purest reaction co-substrate. That is why it is called a green biocatalyst. The trend is an increase in the production of enzymes related to the intensive development of industry, bioremediation or synthetic chemistry. This leads to the search for laccases with greater activity and stability under extreme conditions. The potential of laccases to degrade xenobiotics can be promoted by improving enzymatic catalytic characterization using protein engineering and other genetic engineering methods.

Bioprinting is a relatively new yet evolving technique predominantly used in regenerative medicine and tissue engineering. 3D bioprinting techniques combine the advantages of creating Extracellular Matrix (ECM) like environments for cells and computer-aided tailoring of predetermined tissue shapes and structures. The essential application of bioprinting is for the regeneration or restoration of damaged and injured tissues by producing implantable tissues and organs. The capability of bioprinting is yet to be fully scrutinized in sectors like the patient-specific spatial distribution of cells, bio-robotics, etc. In this review, currently developed experimental systems and strategies for the bioprinting of different types of tissues as well as for drug delivery and cancer research are explored for potential applications. This review also digs into the most recent opportunities and future possibilities for the efficient implementation of bioprinting to restructure medical and technological practices.

Disease, trauma, and aging account for a significant number of clinical disorders. Regenerative medicine is emerging as a very promising therapeutic option. The design and development of new cell-customized biomaterials able to mimic ECM functionalities represent one of the major strategy to control the cell fate and stimulate tissue regeneration. Recently, hydrogels have received a considerable interest for their use in the modulation and control of cell fate during regeneration processes. Several synthetic bioresponsive hydrogels are being developed in order to facilitate cell-matrix and cell-cell interactions. In this review new strategies and future perspectives of such synthetic cell microenvironment will be highlighted.

A medium scale household in South Africa consumes about 1000 kWh on average, and ae household spend about 3000 ZAR per month on energy supply using grid electricity. Grid electricity. South Africa’s energy is supplied by a huge governmental organization. Statistical analyses indicate that projected increase of the cost of grid electricity in South Africa is about 30% per annum for the next five years. Our research show that Photo-Voltaic (PV) technology can offer a viable alternative, provided a large initial capital outlay can be made. The technology is user-friendly and easy to operate. It is calculated that the cost of electricity, capitalized over 20 years, considering an 80 % supply of needs and assuming 20 sunny days per month, is about 2.80 ZAR. The study also shows that utilizing a hybrid combination of renewable energies, such as combining PV supply with solar thermal energy absorbers, and converting thermal energy to electricity, as energy supply systems, could reduce the current cost of electricity per household in South Africa to about half of national grid supply prize, R1,50 per kWhr. The study also shows that water provides as a most viable medium to store energy, instead of using expensive battery technology to perform such functions.

Coasts are characterized by high biological and ecological productivities, and are theatre of strategic economic activities, including tourism, fishing and aquaculture. Both anthropic and natural hazards can thus affect coasts, causing their deterioration. The monitoring of coasts in terms of evolution, adaptation, contamination and resilience, is a pivotal point composed of both classic measurement approaches, and innovative approaches as remote sensing and the use of unmanned aerial vehicles. In the last years, climate change is increasing its effects all over the world and coasts are a delicate system suffering these effects particularly. Climate change has important impacts on coasts, with a serious consequence represented by sea level rise. The latter causes coastal erosion, leading to ecological and economic issues and with consequences for human health. The Mediterranean Basin, which has always represented an important crossroads of different civilizations and economic and cultural exchanges, is now threatened by the consequences of climate change. This area is decribed as an example of coastal system of strategic importance and subjected to serious challenges. Engineering interventions are countermeasures to cope with coastal erosion and coastal degradation. These countermeasures are aimed at protecting the coasts and the populations living on the coasts. The types of intervention can be defined of hard engineering, as in the case of fixed structures in concrete, and of soft engineering, with structures of protection builted according to the principles of nature. Engineering procedures provide coastal benefits, although they can cause further coastal damage. It is thus necessary to protect the coasts and the same time actions aimed at mitigating climate change must be implemented, according to the rules of sustainability.

The world of education has changed. And for all the trials and challenges of managing education during a pandemic, we have also learned a few things that can make our teaching and learning experiences even better in the future.The pandemic imposed constant balance exercises in all aspects of our daily life, including education. Suddenly the living room of the house turned into a work space, the kitchen replaced the canteen, the children's office became a student seat. There was a new area of unknown, irregular and unpredictable forced experimentation. Now what is right, beneficial and effective is an unanswerable question in the absence of previous experience, and the only way to answer it is by trial and error.Universities, in the context of "emergency" remote teaching, also had to experiment. Professors and students found themselves inside virtual classrooms, facing digital boards, closed cameras and microphones.How could a professor not be frustrated by low levels of interaction during the lesson, and how could a student understand a problem without disrupting the fragile digital teaching flow? What was the learning outcome after each two-hour e-teaching? Was the learner's attention maintained, the learning objective achieved, knowledge transferred, new skills acquired? The answers had to be given through experimentation, creativity and innovation.To stop digital teaching from being monotonous and boring, we tried to turn it into a game in order to engage students and lead to better learning outcomes. Gamification is the use of game mechanics in non-game situations. It involves the use of video game elements, such as leaderboards, levels and badges in non-game activities aimed at improving the user experience and increasing user engagement. We find examples of gamification in various fields, e.g. in physical exercise and health (Pereira et al., 2014; Johnson et al., 2016), in the promotion of products and services (Huotari, & Hamari, 2012) and, of course, in education (Triantafyllou & Georgiadis, 2022). For example, airlines enable travelers to earn points with each trip that they can later redeem, or sports equipment companies have developed apps on smart phones and watches so that users can celebrate their performance with digital trophies, compete and challenge their running friends.In the learning process, gamification includes direct feedback mechanisms for learners, point systems, ranking tables, prizes, badges, progress bars and avatars among others, with the main objectives of mobilizing, encouraging and guiding the participant to achieve learning progress. Game-based learning makes the most of these mechanisms with an a priori architecture, which clearly defines the goal of the game, its basic functions and scenario, the rules of progression, competition and interaction with the application and teammates (Triantafyllou & Sapounidis, 2023).The reason why gamification is of particular interest is that research shows an increased degree of involvement and interest of participants when an environment or activity incorporates elements of gamification (Triantafyllou & Georgiadis, 2022). Game-based learning therefore activates not only cognitive functions, such as attention or perception, but also feelings of excitement, surprise, joy, sadness or even anger. This experience results in the achievement of meaningful learning objectives, such as deepening knowledge and developing intrapersonal (e.g. project scheduling) and interpersonal skills (e.g. collaboration).

Many requirements engineering tools have been existing for gathering, documenting and tracing requirements that can even be further processed for such purposes as analysis and transformation. In this study, we analysed 56 different requirements engineering tools for a comprehensive set of features that are categorised into multiple viewpoints (i.e., project management, specification, collaboration, customisation, interoperability, methodology, knowledge management). The analysis results led to many interesting findings. Some of them are as follows: (i) the project planning and execution activities are rarely supported, (ii) multi-user access and versioning are highly supported, (iii) the top-popular specification technique is natural languages, while precise specification via modeling languages is rarely supported, (iv) requirements analysis is rarely supported, (v) requirements transformation is considered for generating documents only, (vi) tool customisation via the tool integration and API support is highly popular, while customising the notation set is rarely supported, (vii) exchanging requirements is popular in such standards as ReqIF and Excel/CSV, while no single standard are accepted by all the tools, (viii agile development is very common, while other methodologies (e.g., MDE and SPLE) are rarely supported, and (viii) user-guide, telephone, e-mail, videos are the top methods for sharing knowledge. The analysis results will be useful for different stakeholders including practitioners, tool vendors and researchers.

Cardiovascular diseases are the leading cause of death worldwide. Cardiovascular diseases complication can give rise to myocardial infarction which produces cell death by blockage of blood flow, leading to loss of heart function. Current treatments directed at heart repair have several disadvantages such as the lack of donors for heart transplantation or the use of non-bioactive inert materials for replacement of the damage tissue. New treatment strategies involve stimulation of heart tissue regeneration with the use of bioactive materials like chitosan, in combination with cells and biochemical factors. Chitosan scaffolds have the necessary proprieties of biocompatibility, porosity, and biodegradation, that imitates the heart extracellular matrix. Chitosan scaffolds physical proprieties, such as electrical conductivity and mechanical proprieties, can be improved by different preparation techniques and by the functionalization with other materials.

Cellular response to mechanical stimuli is an integral part of cell homeostasis. The interaction of the extracellular matrix with the mechanical stress plays an important role in cytoskeleton organisation and cell alignment. Insights from the response can be utilised to develop cell culture methods that achieve predefined cell patterns, which are critical for tissue remodelling and cell therapy. We report the working principle, design, simulation and characterisation of a novel electromagnetic cell stretching platform based on the double-sided axial stretching approach. The device is capable of introducing a cyclic and static strain pattern on a cell culture. The platform was tested with fibroblasts. The experimental results are consistent with the previously reported cytoskeleton reorganisation and cell reorientation induced by strain. The orientation of the cells is highly influenced by external mechanical cues. Cells reorganise their cytoskeleton to avoid external strain and to maintain intact extracellular matrix arrangements.

This thesis presents the effects of work done on a software project for generative models and spreadsheets, allowing for a quick creation of the conceptual model of the aircraft. The subject of the work is a response to the current trends and needs prevailing in the field of computer design engineering CAD and aviation. In the initial chapters, theoretical issues related to the work being carried out were introduced and the methodology of creating software for construction and verification of the structure of aircraft along with the needs of interchange between databases of generative models was presented. In the next stages, the concepts and selected solutions for the user interface supporting the knowledge base were presented along with a set of procedures for its operation. Furthermore, the method of database integration with the methods of determining design features for the developed generative models and with the Siemens NX system. Furthermore, problems encountered in software development, as well as solution examples for model application are specified. The results obtained and the models generated on their basis were subjected to a strength analysis using Autodesk Inventor software and analysed in terms of meeting the initial assumptions. In the end, conclusions and observations resulting from the effects of the work presented in the project were formulated.

Methacrylated silk (Sil-MA) is a chemically modified silk fibroin specifically designed to be crosslinkable under UV light. This allows the structuring of this material throught additive manufacturing techniques and then to easily prototype patient specific construct. In this study we used Sil-MA to produce single layer crosslinked structures that can be withdrawal and ejected recovering their shape after rehydration. A complete chemical and physical characterization of the material has been conducted. Additionally, we tested the material biocompatibility according to the International Standard Organization protocols (ISO 10993) ensuring the possibility to use it in future trials. The material was also tested to verify its ability to support the osteogenesis. Two different additive manufacturing techniques have been tested (a Digital Light Processing (DLP) UV projector and a pneumatic extrusion technique) to develop Sil-MA grid. Finally, we provide a proof-of-concept that the printed Sil-MA structures are injectable.

Context: Business Process Automation (BPA) is adopted by organizations to improve efficiency, reduce costs, and increase overall business performance. Traditional Business Process Automation (TBPA) is one of the three approaches employed to develop a BPA. TBPA entails developing BPA in a programming language for integrating the relevant applications in the digital ecosystem to execute a given process. Process variability and practitioner unavailability encumber the requirements specification for TBPA software. Objective: This work proposes a log-based approach for TBPA software to make software more adaptable to process changes and reduce reliance on practitioners, by providing a higher alignment among business process requirements and software architecture, and employing process mining to semi-automatically discover the business process during requirements elicitation. Method: The research conducted a case study in a technology institute to assess the approach and report its results in practice. Results: The results revealed significant improvements in adaptability to business process changes and decreased the time spent with practitioners, and, efficiency in development. The approach also presented limitations, including human intervention to accurately obtain the business process, complexity to trace the process into the architecture, data privacy concerns, and risk of network traffic overload. Conclusion: This research demonstrated the effectiveness of traceability between process requirements and software architecture, as well as the use of logs and process mining. These methods made TBPA software enhanced the software adaptability to changes and minimized the reliance on practitioners during requirements elicitation respectively.

Software Engineering education has grown during the past couple of decades as a specialized branch of computer science education to address employment demands. Software Engineering education is constantly evolving because of its dynamic nature. However, there are still some gaps between the skills being taught in the universities and skills required for employment. In this paper, we present our experience of designing and delivering a new module to enhance skills for employment. We have taken a structured approach to analyze the gap through literature review and our personal experiences of engaging with employers of our students. Based on the analysis, we arrived at a set of gaps between what we teach and what the employers of our students need. We prioritized the gaps and envisaged a new module ‘Contemporary Software Engineering’ to address the gaps and subsequently help our students for an easy transition to job. As part of our experience, we present the background, design and delivery of the module and reflect on our journey of delivering the module for a few years.

Spider silks, renowned for their exceptional mechanical and biocompatible properties, offer significant potential as scaffolds in tissue engineering. This paper explores the feasibility of using spidroins, the primary proteins in spider silk, to develop scaffolds that closely mimic the extracellular matrix (ECM) essential for tissue regeneration. Spidroins (spider silk proteins) are particularly valuable due to their unique strength, elasticity, and biocompatibility, which support cellular growth and differentiation. The paper examines current methods for producing recombinant spider silk, including using unicellular hosts like E. coli and multicellular systems like transgenic plants and silkworms. It also examines current scaffolds that utilize spidroins and their shortcomings, such as immunogenicity and protein production, which must be urgently addressed for more practical and effective widespread biomedical applications. In addition, it stresses the need for further research to solve these challenges and fully realize the potential of spider silk in biomedical applications.

Therapeutic protein engineering has revolutionized medicine by enabling the development of highly specific and potent treatments for a wide range of diseases. This review examines recent advances in computational and experimental approaches for engineering improved protein therapeutics. Key areas of focus include antibody engineering, enzyme replacement therapies, and cytokine-based drugs. Computational methods like structure-based design, machine learning integration, and protein language models have dramatically enhanced our ability to predict protein properties and guide engineering efforts. Experimental techniques such as directed evolution and rational design approaches continue to evolve, with high-throughput methods accelerating the discovery process. Applications of these methods have led to breakthroughs in affinity maturation, bispecific antibodies, enzyme stability enhancement, and the development of conditionally active cytokines. Emerging approaches like intracellular protein delivery, stimulus-responsive proteins, and de novo designed therapeutic proteins offer exciting new possibilities. However, challenges remain in predicting in vivo behavior, scalable manufacturing, immunogenicity mitigation, and targeted delivery. Addressing these challenges will require continued integration of computational and experimental methods, as well as a deeper understanding of protein behavior in complex physiological environments. As the field advances, we can anticipate increasingly sophisticated and effective protein therapeutics for treating human diseases.

Halide perovskite materials gain enormous attention for their semiconducting properties, higher power conversion efficiency and potential applications in a wide range field of studies along with their two key limitations: stability and toxicity. In spite of great progress made on the halide perovskites and many promising research developments, overcoming these limitations is not realized yet. Therefore, coordination engineering of new framework for obtaining alternative new halide perovskite materials and fundamental understanding of the coordination chemistry and electronic interactions forming the structure of these newly engineered halide perovskite materials might be one way in order to overcome the issues related to both stability and toxicity. In this review, the current development of halide perovskite families, both lead halide perovskites and lead free halide perovskites followed by coordination engineering of the new frameworks to engineer new halide perovskite materials will be reviewed comprehensively. Moreover, all the concerns of the fundamental ideas of coordination chemistry and electronic interactions that are keys in forming the halide perovskite structures are discussed in detail and form the main aim of this review. Interestingly, we also discuss recent potential energy applications beyond photovoltaic and this review has completed with an essential and open question: ‘what could happen in the future of halide perovskites?’ in order to excite commercial enterprises and research institutions again as well as to get motivating new predictions on the future continuity of this field.

Regenerative Medicine and Tissue Engineering are interdisciplinary fields that combine cell biology, material science, and bioengineering to develop therapies for tissue repair and regeneration. This field has made significant advances in recent years, particularly in the use of stem cells and biomaterials. Advancements in cell-based therapies have involved the use of cells to repair or replace damaged tissues and organs. Tissue engineering involves using scaffolds and growth factors to promote tissue regeneration. However, a major challenge in this field is the formation of adhesions, which can cause complications, such as intestinal obstruction and chronic pain. Adhesion barriers are used to prevent adhesion of tissues and organs during recovery, thereby reducing the risk of complications. Anti-adhesion materials, such as polysaccharides, proteins, and synthetic polymers, are used to create a physical barrier between tissues and prevent adhesion. These materials can be used alone or in combination, but their effectiveness varies.

Due to its distinctive chemical and biological properties, pectin has recently drawn much attention in biomedical and tissue engineering applications. Polymers like pectin with cell-instructive properties are attractive natural biomaterials for tissue repair and regeneration. Besides, bioactive pectin and pectin-based composites exhibit improved characteristics to deliver active molecules. Pectin and pectin-based composites serve as interactive matrices or scaffolds by stimulating cell adhesion and cell proliferation and enhancing tissue remodeling by forming an extracellular matrix in vivo. Several bioactive properties, such as immunoregulatory, antibacterial, anti-inflammatory, anti-tumor, and antioxidant activities, contribute to the pectin's and pectin-based composite's enhanced applications in tissue engineering and drug delivery systems.This paper reviews the promising characteristics of pectin or pectic polysaccharides and highlights its potential applications in drug delivery and tissue engineering. Tissue engineering scaffolds containing pectin and pectin-based conjugates or composites demonstrate essential features such as non-toxicity, tunable mechanical properties, biodegradability, and suitable surface properties. The design and fabrication of pectic composites are versatile for tissue engineering and drug delivery applications.

β-glucosidases (BGLs) play a crucial role in the degradation of lignocellulosic biomass as well as in industrial applications such as pharmaceuticals, foods, and flavors. However, the application of BGLs has been largely hindered by issues such as low enzyme activity, product inhibition, low stability, etc. Many approaches have been developed to engineer BGLs to improve these enzymatic characteristics to facilitate industrial production. In this article, we review the recent advances in BGL engineering in the field, including the efforts from our laboratory. We summarize and discuss the BGL engineering studies according to the targeted functions as well as the specific strategies used for BGL engineering.

Female gynecological organ dysfunction can cause infertility and psychological distress, decreasing quality of life of affected women. Incidence is constantly increasing due to growing rates of cancer and delaying of childbearing age in the developed world. Current treatments are often unable to restore organ function, and occasionally are the cause for female infertility. Alternative treatment options are currently being developed in order to face the inadequacy of current practices. In this review, pathologies and current treatments of gynecological organs (ovaries, uterus, and vagina) will be described. The state-of-the-art of tissue engineering alternatives to common practices are evaluated with a focus on in vivo models. Tissue engineering is an ever-expanding field, integrating various domains of modern science to create sophisticated tissue substitutes in the hopes of repairing or replacing dysfunctional organs using autologous cells. Application to gynecology has the potential of restoring female fertility and sexual wellbeing.

Tissue engineering, also called “regenerative medicine”, refers to attempt to create functional human tissue from cells in laboratory. This is a field that uses living cells, biocompatible materials, suitable biochemical and physical factors and their combinations, to create tissue-like structures.. To date, no tissue engineered skeletal muscle implants have been developed for clinical use, but it may represent a valid alternative to treat volumetric muscle loss in the near future. Herein, we reviewed the literature and showed different techniques to produce synthetic tissues with the same architectural, structural and functional properties of native tissues.

The digital society is an outcome of the Internet which has nearly made everything connected and accessible no matter where or when. Nevertheless, despite the fact that conventional IP networks are complicated and very hard to manage, they are still widely adopted. The already established policies make the network configuration/reconfiguration a complex process that reacts to errors, load, and modifications. The prevailing networks are vertically integrated which makes things more and more complicated: Data planes and control are strapped together. Software-defined networking is a model that is meant to solve this issue by splitting the vertical integration and detaching the network’s control logic from the implicit routers and switches; this could be achieved by reinforcing centralization of network control and making the network programmable. In this work, we worked to implement MPLS networks with SDN, to enhance the traffic engineering over the network, and to minimize the network delay and latency, with minimum cost using three of the different SDN networks. The experiment results showed the advantage of the proposed approach for reducing the network delay, comparing with previous studies. Where the average of network delay in our approach reaches to 3.01 milliseconds.

In tissue engineering scaffolds take the place of the natural extra cellular matrix (ECM). The natural ECM is the extracellular part of animal tissue that usually provides structural support to the animal cells in addition to performing various other important functions. The design aspect along with the choice of the material for the artificial scaffold is very crucial to cell differentiation, adhesion, proliferation, and the transport of the growth factors or other bio molecular signals. In addition to the material and design of the scaffolds, it is necessary to replicate the normal physiological situation if the scaffold has to function as an implant. The cells have to be located in the porous scaffold to form a three dimensional assembly. The article discusses the important factors to be considered while designing a scaffold for tissue engineering and regenerative medicine.

Since engineering discipline is at the forefront of latest innovations and developments, there is an increasing emphasis on the critical responsibilities of engineers towards sustainability. There are several studies investigating the incorporation of sustainability principles into engineering curriculum but studies analyzing the knowledge, attitude, and behavior of engineering students are lacking. A deeper understanding of the current state of students’ knowledge, attitude, and behavior towards sustainability is crucial to determine the type of pro-sustainability trainings, strategies and pedagogical activities needed. Hence, the current study is aimed at assessing the level of sustainability cognizance of Mechanical Engineering students via an online Sustainability Consciousness Questionnaire. The final sample size was 134 students from a Higher Education Institute in India with 90% male responders and 52% from urban areas. Results indicate that 70% of the respondents have heard of the Sustainable Development Goals, only around 32% of the students demonstrate a positive behavior and attitude towards sustainability whereas 30% of the students do not hold any opinion towards sustainability principles. The current exploratory study contributes to the knowledge base about sustainability awareness and consciousness and advocates for measures to insert its principles in engineering courses.

This study examines the impact of prompt engineering on large language models (LLMs), focusing on a comparison between multitasking and single-task prompts. Specifically, we explore whether a single prompt handling multiple tasks — such as Named Entity Recognition (NER), sentiment analysis, and JSON output formatting — can achieve similar efficiency and accuracy to dedicated single-task prompts. The evaluation uses a combination of performance metrics to provide a comprehensive analysis of output quality. Experiments were conducted using a selection of open-source LLMs, including LLama3.1 8B, Qwen2 7B, Mistral 7B, Phi3 Medium, and Gemma2 9B. Results show that single-task prompts do not consistently outperform multitasking prompts, highlighting the significant influence of the model’s data and architecture on performance.

Genetic engineering has revolutionized our ability to modify microorganisms for various applications in agriculture, medicine, and industry. This review examines recent advances in genetic engineering techniques for bacteria, fungi, and oomycetes, with a focus on CRISPR-Cas systems. In bacteria, CRISPR-Cas9 has enabled precise genome editing, enhancing applications in antibiotic production and metabolic engineering. For fungi, despite challenges associated with their complex cell structures, CRISPR/Cas9 has advanced the production of enzymes and secondary metabolites. In oomycetes, significant plant pathogens, modified Agrobacterium-mediated transformation and CRISPR/Cas12a have contributed to developing disease-resistant crops. The review provides a comparative analysis of genetic engineering efficiencies across these microorganisms and addresses ethical and regulatory considerations. Future research directions include refining genetic tools to improve efficiency and expand applicability in non-model organisms. This comprehensive overview highlights the transformative potential of genetic engineering in microbiology and its implications for addressing global challenges in agriculture, medicine, and biotechnology.

With the widespread adoption of 3D scanners, reverse engineering methods for obtaining CAD drawings from physical objects have become increasingly utilized. When converting point data obtained from a 3D scanner into a mesh structure, a smoothing process is typically applied because the point data often contains a lot of noise. However, this can result in the loss of important high-frequency details, or in cases where significant high-frequency information remains, there are limitations in representing the object with basic geometric shapes. In this paper, a method to address this issue by using environment mapping and supervised learning techniques is proposed. By mapping the object to an environment and then using supervised learning to analyze the data, it can accurately identify the basic geometric shapes. This approach reduces the working time and allows for easier design of complex surfaces that were previously difficult to model.

Amazon Web Services (AWS) instances provide a convenient way to run training on complex ’omics data analysis workflows without requiring participants to install software packages or store large data volumes locally. However, efficiently managing dozens of instances is challenging for training providers. We present a set of Bash scripts that make it quick and easy to manage Linux AWS instances pre-configured with all the software analysis tools and data needed for a course, and accessible using encrypted login keys and optional domain names. Creating over 30 instances takes 10-15 minutes. A comprehensive online tutorial describes how to set up and use an AWS account and the scripts, and how to customise AWS instance templates with other software tools and data. We anticipate that others offering similar training may benefit from using the scripts regardless of the analyses being taught.

Object-Oriented Programming (OOP) has long been a dominant paradigm in software development, but it's not without its challenges. One major issue is the problem of tight coupling between objects, which can hinder flexibility and make it difficult to modify or extend code. Additionally, the complexity of managing inheritance hierarchies can lead to rigid and fragile designs, making it hard to maintain and evolve the software over time. This paper introduces a software development pattern that seeks to offer a renewed approach to writing code in Object-Oriented (OO) environments. Addressing some of the limitations of the traditional approach, the Puzzle Pattern focuses on extreme modularity, favoring writing code exclusively in interfaces. Concrete classes are subsequently assembled through the implementation of those interfaces, reducing coupling and introducing a new level of flexibility and adaptability in software construction. The highlighted pattern offers significant benefits in software development, promoting extreme modularity through interface-based coding, enhancing adaptability via multiple inheritance, and upholding the SOLID principles, though it may pose challenges such as complexity and a learning curve for teams.

The evaluation of nanostructured biomaterials and medicines is associated with 2D cultures that provide insight into the biological mechanisms at the molecular level, while critical aspects of the tumor microenvironment (TME) are provided by the study of animal xenograft models. More realistic models that can histologically reproduce human tumors are provided by tissue engineering methodologies of co-culturing cells of varied phenotypes, to provide 3D tumor spheroids that recapitulate the dynamic TME in 3D matrices. The novel approaches of 3D tumor models are combined with tumor tissue engineering (TTE) scaffolds including hydrogels, bioprinted materials, decellularized tissues, fibrous and nanostructured matrices. This review focuses on the use of nanostructured materials in cancer therapy and regeneration, and the development of realistic models for studying TME molecular and immune characteristics. Tissue regeneration is an important aspect of TTE scaffolds for restoring the normal function of the tissues, while providing cancer treatment. Thus, this article reports recent advancements on the development of 3D TTE models for antitumor drug screening, studying of tumor metastasis, and tissue regeneration. Also, this review identifies the great opportunities of 3D TTE scaffolds in the evaluation of the immunological mechanisms and processes involved in the application of immunotherapies.

One of the main challenges of cultured meat is reducing the costs of production, often described as scaling. The main components of the cultivation process include: cells, media, scaffolds and bioreactors. This work presents an alternative to current stainless steel or glass bioreactors and uses food-grade plant-based scaffolds. Bioreactors must be sterile and leak-free to maintain a proper environment for cell growth. Thermoplastic films are a popular material for constructing these fluidic systems, but traditional methods of welding these film are expensive and do not allow rapid prototyping resulting in high research and development costs for companies. In this study, a laser welding method was developed to join thermoplastic films in a way that prevents contamination and leaks, affording effective macro-fluidics fabrication. This technique was tested using polyethylene (PET) films and a laser cutter operating with settings calibrated for the material to be welded or cut. The laser welding method was found to produce strong, leak-free seals in PET films with minimal heat-induced damage to the films. The ability to design fluidics, chambers and ports of various size. Using this method afforded the incorporation of plant-based scaffolds from food-grade plants (Rice). The laser welding method developed in this study provides a reliable, contamination-free method for the rapid fabrication of fluidic systems for cell cultivation. This technique has the potential to be compatible with a vast range of cell types and scaffolds and to be widely adopted in tissue engineering for regenerative medicine and food applications such as cultured meat.

Reverse engineering is applied to identify optimum polymerization conditions for the synthesis of polymer with pre-defined properties. The proposed approach uses multi-objective optimization (MOO) and provides multiple candidate polymerization procedures to reach the targeted polymer property. The objectives for optimization include maximal similarity of molar mass distributions (MMD) compared to the target MMD, minimal reaction time, and maximal monomer conversion. The method is tested for vinyl acetate radical polymerizations and can be adopted to other monomers. The data for the optimization procedure is generated by an in-house developed kinetic Monte-Carlo (kMC) simulator for a selected recipe search space. The proposed reverse engineering algorithm comprises several steps: kMC simulations for the selected recipe search space to derive initial data, performing MOO for a targeted MMD, and identification of the Pareto optimal space. The last step uses a weighted sum optimization function to calculate the weighted score of each candidate polymerization conditions. To decrease the execution time clustering of the search space based on MMDs is applied. The performance of the proposed approach was tested for various target MMDs. The suggested MOO-based reverse engineering provides multiple recipe candidates depending on competing objectives.

In an increasingly complex world there is a real, urgent need for methodologies to enable engineers to model complex sociotechnical systems, as these now seem to describe the majority of systems in use today. This is of course exacerbated by the increasing involvement and augmentation with “black box” AI contributions. Hollnagel produced a methodology, (FRAM) which did allow the analyst in-sights into these systems’ behaviour, but the model-based system engineering applications demand numbers and a quantitative ap-proach. In the last 10 years, this original approach developed to model systems as sets of interactive, interdependent “functions”, (abstracted from agent or component details), has been further developed to the point where it can take the basic data and structures from the current component focussed system engineering “models”, and can pull it all together into dynamic models, (as opposed to static, fixed System Theoretic Process Accimaps), from which analysts can discern how they really work in practice, and predict the emergent behaviours characteristic of complex systems. This paper describes how the FRAM methodology has now been extended to provide these extra, essential attributes. It also describes its implementation using an open-source software, freely available for use and verification on the GitHub site.

Chondroitin is a glycosaminoglycan that has gained widespread use in nutraceuticals and pharmaceuticals, mainly for treating osteoarthritis. Traditionally, it has been extracted from animal cartilage but recently, biotechnological processes have emerged as a commercial alternative to avoid the risk of viral or prion contamination and offer a vegan-friendly source. Typically, these methods involve producing the chondroitin backbone using pathogenic bacteria and then modifying it enzymatically through the action of sulfotransferases. Despite the challenges of expressing active sulfotransferases in bacteria, the use of eukaryotic microorganisms is still limited to a few works using Pichia pastoris. To create a safer and efficient biotechnological platform, we have constructed a biosynthetic pathway for chondroitin production in S. cerevisiae as a proof-of-concept. Up to 125 mg/L and 200 mg/L of intracellular and extracellular chondroitin were produced, respectively. Furthermore, as genome-scale models are valuable tools for identifying novel targets for metabolic engineering, a stoichiometric model of chondroitin-producing S. cerevisiae has been developed and used in optimization algorithms. Our research has yielded several novel targets, such as uridine diphosphate(UDP)-N-acetylglucosamine pyrophosphorylase (QRI1), glucosamine-6-phosphate acetyltransferase (GNA1) or N-acetylglucosamine-phosphate mutase (PCM1) overexpression that might enhance chondroitin production, which will guide future experimental research to develop more efficient host organisms for the biotechnological production process.

Probiotics, living microorganisms with demonstrated health benefits when administered in sufficient quantities, have a rich history as dietary supplements to benefit human health. Recently, understanding of their mechanisms in the gastrointestinal tract has prompted exploration of probiotics in treating human diseases. However, the effective and precise delivery of probiotics remains a significant challenge in therapeutic applications. Here, we review the mechanisms of action of probiotics in human health and the most advanced strategies for efficient probiotic delivery. We also discuss the potential applications of engineered probiotics in disease treatment. This review contributes insights into the evolving landscape of probiotic research for therapeutic applications.

Proteins in the crowded environment of human cells have often been studied regarding nonspecific interactions, misfolding, and aggregation, which may cause cellular malfunction and disease. Specifically, proteins with high abundance are more susceptible to these issues due to the law of mass action. Therefore, the surfaces of highly abundant cytoplasmic (HAC) proteins directly exposed to the environment can exhibit specific physicochemical, structural, and geometrical characteristics that reduce nonspecific interactions and adapt to the environment. However, the quantitative relationships between the overall surface descriptors still need clarification. Here, we used machine learning to identify HAC proteins using hydrophobicity, charge, roughness, secondary structures, and B-factor from the protein surfaces and quantify the contribution of each descriptor. First, several supervised learning algorithms were compared to solve binary classification problems for the surfaces of HAC and extracellular proteins. Then, logistic regression was adopted for the feature importance analysis of descriptors considering model performance (80.2% accuracy and 87.6% AUC) and interpretability. The HAC proteins showed positive correlations with negatively and positively charged areas but negative correlations with hydrophobicity, B-factor, beta structure proportion, roughness, and disordered regions proportion. Finally, the details of each descriptor could be explained concerning adaptative surface strategies of HAC proteins to regulate nonspecific interactions, protein folding, flexibility, stability, and adsorption. This study presented a novel approach using various surface descriptors to identify HAC proteins and provided quantitative design rules for the surfaces well-suited to human cellular crowded environments.

3D Printing (3DP) technology has revolutionized the field of the use of bioceramics for maxillofacial and periodontal applications, offering unprecedented control over the shape, size, and structure of bioceramic implants. In addition, bioceramics have become attractive materials for these applications due to their biocompatibility, biostability, and favorable mechanical properties. However, despite their advantages, bioceramic implants are still associated with inferior biological performance issues after implantation, such as slow osseointegration, inadequate tissue response, and increased risk of implant failure. To address these challenges, researchers have been developing strategies to improve the biological performance of 3D printed bioceramic implants. The purpose of this review is to provide an overview of 3DP techniques and strategies for bioceramic materials designed for bone regeneration. The review also addresses the use and incorporation of active biomolecules in 3D printed bioceramic constructs to stimulate bone regeneration. By controlling the surface roughness, and chemical composition of the implant, the construct can be tailored to promote osseointegration and reduce the risk of adverse tissue reactions. Additionally, growth factors, such as bone morphogenic proteins (rhBMP-2) and pharmacologic agent (dipyridamole), can be incorporated to promote the growth of new bone tissue. Incorporating porosity into bioceramic constructs can improve bone tissue formation and the overall biological response of the implant. As such, by employing surface modification, combining with other materials, and incorporation of 3DP workflow can lead to better patient healing outcomes.

Cardiovascular diseases continuing to be a major contributor to illness and death on a global scale and the implementation of stents has given rise to a revolutionary transformation in the field of interventional cardiology. The thrombotic and restenosis complications associated with stent implantation pose ongoing challenges. In recent years, bioactive coatings have emerged as a promising strategy to enhance stent hemocompatibility and reduce thrombogenicity. This review article provides an overview of the surface engineering techniques employed to improve the hemocompatibility of stents and reduce thrombus formation. It explores the mechanisms underlying thrombosis and discusses the factors influencing platelet activation and fibrin formation on stent surfaces. Various bioactive coatings, including anticoagulant agents, antiplatelet agents, and surface modifications, are discussed in detail, highlighting their potential in reducing thrombogenicity. Evaluation methods for assessing hemocompatibility and thrombogenicity are also reviewed, ranging from in vitro assays to animal models. Recent advances in the field, such as nanotechnology-based coatings and bioactive coatings with controlled drug release systems, are highlighted. Surface engineering of bioactive coatings holds great promise for enhancing the long-term outcomes of stent implantation by enhancing hemocompatibility and reducing thrombogenicity. Future research directions and potential clinical applications are discussed, underscoring the need for continued advancements in this field.

Lipases have tremendous potential for industrial use, particularly, those mostly active against water-insoluble substrates, such as triglycerides composed of long-chain fatty acids. However, in most cases, mutants often need to be constructed to achieve optimal performance for such substrates. Protein engineering techniques have been reported as strategies for improving lipase characteristics by introducing specific mutations in the cap domain of esterases or in the lid domain of lipases, or through lid domain swapping. Here, we have improved the lipase character of a lipase, retrieved from the Marine Metagenomics MarRef Database and assigned to the Actinoalloteichus genus (WP_075743487.1), through site-directed mutagenesis and by substituting its lid domain (FRGTEITQIKDWLTDA) by the one of Rhizopus delemar (previously Rhizopus oryzae) lipase (FRGTNSFRSAITDIVF). Results demonstrated that the redesigned mutants gain activity against bulkier triglycerides such as glyceryl tridecanoate and tridodecanoate, olive oil, coconut oil, and palm oil. The absence of a residue in the new lid, present in the original lid, appears to be a key aspect to the increase in lipase character, although full activity recovery is only obtained with lid swapping.

Focal cartilage defects are a prevalent knee problem affecting people of all ages. Due to its avascular nature, cartilage has limited self-repair capacity, and osteochondral defects can lead to pain and long-term complications such as osteoarthritis. Autologous chondrocyte implantation (ACI) has been a successful surgical approach for repairing osteochondral defects over the past two decades. However, a major drawback of ACI is the de-differentiation of chondrocytes during their in vitro expansion. In this study, we isolated ovine chondrocytes and cultured them in a two-dimensional environment as for ACI procedures. We hypothesised that the 3D scaffolds would support the cells re-differentiation without the need for growth factors and so we encapsulated them into soft collagen and alginate (col/alg) hydrogels. Chondrocytes embedded into hydrogels were viable and proliferated. After 7 days they acquired a rounded morphology and started to aggregate. Gene expression studies showed that the genes associated with chondrogenesis started to be up regulated as early as day one. At 21 days chondrocytes had extensively colonized the hydrogel, forming large cell clusters and started to deposit collagen II and aggrecan with limited collagen type I deposition. These findings highlight the potential of soft col/alg hydrogels to enhance ACI outcomes by creating a favourable microenvironment for chondrocyte reprogramming and re-differentiation, eliminating the dependency on growth factors.

The role of mesenchymal-to-endothelial transition in the angiogenic response is controversial. Toward this, the present study aimed to determine if fibroblasts contribute to angiogenesis. Endothelial differentiation of fibroblasts was induced by culturing MRC-5 cells (human fetal lung fibroblast cells) on top of Matrigel hydrogel or embedded inside the hydrogel. The formation of ca-pillary-like networks in response to angiogenic signals was observed. The tube formation occurs quickly and can be visualized us-ing a phase-contrast inverted microscope, and/or the cells can be treated with DAPI before the assay and tubes can be visualized through fluorescence or confocal microscopy. Furthermore, fibroblasts cultured in a higher concentration invaded the Matrigel hydrogel and formed stem-cell-like spheroids. These spheroids embedded in matrigel matrices of varying densities sprouted to form 3D connective-tissue networks. Collectively, our results highlight the endothelial differentiation capacity of human lung fibroblasts. The results obtained in this work may have an impact on the search for alternative cell sources for vascular tissue engineering and the overcome of obstacles to vascularization of autologous tissue-engineered constructs and the production of functional grafts for clinical use.

Engineers make things, make things work, and make things work better and easier. This kind of knowledge is crucial for innovation, and much of the explicit knowledge developed by engineers is embodied in scientific publications. In this paper, we analyze the evolution of publications and citations in Engineering in a middle-income country such as Mexico. Using a database of all Mexican publications in Web of Science from 2004 to 2017, we explore the characteristics of publications that tend to have the greatest impact; this is the highest number of citations. Among the variables studied are the type of collaboration (no collaboration, domestic, bilateral, or multilateral), the number of coauthors and countries, the language of the article, controlling for a coauthor from the USA and the affiliation institution of the Mexican author(s). Our results emphasize the overall importance of joint international efforts and suggest that publications with the highest number of citations are those with multinational collaboration (coauthors from three or more countries), written in English, and when one of the coauthors is from the USA.

The main purpose of tissue engineering is to fabricate and exploit engineered constructs suitable for the effective replacement of damaged tissues and organs, and able to perfectly integrate with the host’s organism without eliciting any adverse reaction. Ideally, autologous materials represent the best option, but they are often limited due to the low availability of compatible healthy tissues. So far, one therapeutic approach relies on the exploitation of synthetic materials: they exhibit good features in terms of impermeability, deformability and flexibility, but present chronic risks of infections and inflammations. Alternatively, biological materials, including naturally derived ones and acellular tissue matrices of human or animal origin, can be used to induce cells growth and differentiation, which are needed for tissue regeneration: however, this kind of materials lacks satisfactory mechanical resistance and reproducibility, affecting their clinical application. In order to overcome the above-mentioned limitations, hybrid materials, which can be obtained by coupling synthetic polymers and biological materials, have been investigated with the aim to improve biological compatibility and mechanical features. Currently, the interest in these mate-rials is growing, but the ideal ones have not been found yet. The present review aims at exploring some applications of hybrid materials, with particular mention to urological and cardiovascular fields: in the first case, the efforts to find a construct that can guarantee impermeability, mechanical resistance and patency will be herein illustrated; in the second case, the search for impermeability, hemocompatibility and adequate compliance will be been disclosed.

In order to overcome the disadvantages of existing treatments in heart valve tissue engineering, decellularization studies are carried out. The main purpose of decellularization is to eliminate the immunogenicity of biologically derived grafts and to obtain a scaffold that allows recellularization while preserving the natural tissue architecture. SD and SDS are detergent derivatives frequently used in decellularization studies. The aim of our study is to decellularize the pulmonary heart valves of young Merino sheep by using low-density SDS and SD detergents together, and then to perform their detailed characterization to determine whether they are suitable for clinical studies. Pulmonary heart valves of 4-6 month old sheep were decellularized in detergent solution for 24 hours. The amount of residual DNA was measured to determine the efficiency of decellularization. Then, the effect of decellularization on the ECM by histological staining was examined. In addition, the samples were visualized by SEM to determine the surface morphologies of the scaffolds. Uniaxial tensile test was performed to examine the effect of decellularization on biomechanical properties. The results showed DNA removal of 94% and 98% from the decellularized leaflet and artery portions after decellularization relative to the control group. No cell nuclei were found in histological staining and it was observed that the 3-layer leaflet structure was preserved. As a result of the tensile test, it was determined that there was no statistically significant difference between the control and decellularized groups in the UTS and elasticity modulus, and the biomechanical properties did not change. In conclusion, we suggest that the pulmonary valves of decellularized young Merino sheep can be used as a initial matrix in heart valve tissue engineering studies.

Antimicrobial resistance (AMR) is a major global concern; antibiotics and other regular treatment methods have failed to overcome the increasing number of infectious diseases. Bacteriophages (phages) are viruses that specifically target/ kill bacterial hosts without affecting other human microbiome. Phage therapy provides optimism in the current global healthcare scenario with a long history of its applications in humans that has now reached various clinical trials. Phages in clinical trials have specific requirements of being exclusively lytic, free from toxic genes with an enhanced host range that adds an advantage to this requisite. This review explains in detail the various phage engineering methods and their potential applications in therapy. To make phages more efficient, engineering has been attempted using techniques like conventional homologous recombination, Bacteriophage Recombineering of Electroporated DNA (BRED), clustered regularly interspaced short palindromic repeats (CRISPR)-Cas, CRISPY-BRED/Bacteriophage Recombineering with Infectious Particles (BRIP), chemically accelerated viral evolution (CAVE), and phage genome rebooting. Phages are administered in cocktail form in combination with antibiotics, vaccines, and purified proteins, such as endolysins. Thus, phage therapy is proving to be a better alternative for treating life-threatening infections, with more specificity and fewer detrimental consequences.

One of the most interesting groups of fatty acid derivates is the group of conjugated fatty acids, from which the most researched are conjugated linoleic acid (CLA) and conjugated linolenic acid (CLNA), which are associated with countless health benefits. Sex pheromone mixture of some insect species, including tobacco horn-worm (Manduca sexta), are typical for the production of uncommon C16 long conjugated fatty acids with two and three conjugated double bonds, as opposed to C18 long CLA and CLNA. In this study, M. sexta desaturases MsexD2 and MsexD3 were expressed in multiple strains of Y. lipolytica with different genotypes. Experiments with supplementation of fatty acid methyl esters into the medium resulted in production of novel fatty acids. Using GCxGC-MS 20 new fatty acids with two or three double bonds were identified. Fatty acids with conjugated or isolated double bonds or combination of both were produced in trace amounts. Results of this study prove that Y. lipolytica is capable of synthesizing C16 conjugated fatty acids. Further genetic optimization of the Y. lipolytica genome and optimization of the fermentation process could lead to increased production of novel fatty acid derivatives with biotechnologically interesting properties.

Nano/micron-sized bioactive glass (BG) particles are attractive candidates for both soft and hard tissue engineering. They can chemically bond to the host tissues, enhance new tissue formation, activate cell proliferation, stimulate the genetic expression of proteins, and trigger unique an-ti-bacterial, anti-inflammatory, and anti-cancer functionalities. Recently, composites based on bi-opolymers and BG particles have been developed with various state-of-the-art techniques for tis-sue engineering. Gelatin, a semi-synthetic biopolymer, has attracted the attention of researchers because it is derived from the most abundant protein in the body, viz., collagen. It is a polymer that can be dissolved in water and processed to acquire different configurations, such as hydro-gels, fibers, films, scaffolds, etc. Searching "bioactive glass gelatin" in the tile on Scopus renders 80 highly relevant articles published in the last ~10 years, which signifies the importance of such composites. First, this review addresses the basic concepts of soft and hard tissue engineering, in-cluding the healing mechanisms and limitations ahead. Then, current knowledge on gelatin/BG composites including composition, processing and properties is summarized and discussed both for soft and hard tissue applications. This review explores physical, chemical and mechanical features and ion-release effects of such composites concerning osteogenic and angiogenic respons-es in vivo and in vitro. Additionally, recent developments of BG/gelatin composites using 3D/4D printing for tissue engineering are presented. Finally, the perspectives and current challenges in developing desirable composites for the regeneration of different tissues are outlined.

Laser patterning of implant materials for bone tissue engineering purposes has shown to be a promising technique to control cell properties such as adhesion or differentiation, resulting in an enhanced osteointegration. However, the perspective of patterning the bone tissue side interface to generate microstructure effects has never been investigated. In the present study, three different laser-generated patterns were machined on the bone surface with the aim to identify the best surface morphology compatible with osteogenic-related cells recolonization. The laser patterned bone tissue was characterized by electron scanning microscopy and confocal microscopy in order to obtain a comprehensive picture of the bone surface morphology. Cortical bone patterning impact upon cell compatibility and cytoskeleton rearrangement to the patterned surfaces was performed with Stromal Cells from Apical Papilla (SCAPs). Results indicated that laser machining had no detrimental effect upon consecutively seeded cells metabolism. Orientation assays revealed that surface patterning characterized by larger hatch distances was correlated with a higher cell cytoskeletal conformation to the laser-machined patterns. For the first time, to our knowledge, bone is considered and assessed here as a potentially engineered-improvable biological interface. Further studies shall focus on in vivo implications of this direct patterning.

Genome-scale metabolic models (GEMs) play an important role in the phenotype prediction of microorganisms, and their accuracy can be further improved by integrating other types of biological data such as enzyme concentrations and kinetic coefficients. Enzyme-constrained models (ecModels) have been constructed for several species and were successfully applied to increase the production of commodity chemicals. However, there was still no genome-scale ecModel for the important model organism Bacillus subtilis prior to this study. Here, we integrated enzyme kinetic and proteomic data to construct the first genome-scale ecModel of B. subtilis (ecBSU1) using the ECMpy workflow. We first used ecBSU1 to simulate overflow metabolism and explore the trade-off between biomass yield and enzyme usage efficiency. Then, we simulated the growth rate on eight previously published substrates and found that the simulation results of ecBSU1 were in good agreement with the literature. Finally, we identified target genes that enhance the yield of commodity chemicals using ecBSU1, most of which were consistent with the experimental data, and some of which may be potential novel targets for metabolic engineering. This work demonstrates that the integration of enzymatic constraints is an effective method to improve the performance of GEMs. The ecModel can predict overflow metabolism more precisely and can be used for the identification of target genes to guide the rational design of microbial cell factories.

Decision making as a result of system dynamics analysis requires, in practice, a straightforward and systematic modelling capability as well as a high-level of customisation and flexibility to adapt to situations and environments that may vary very much from each other. While in general terms a completely generic approach could be not as effective as ad-hoc solutions, the proper application of modern technology may facilitate agile strategies as a result of a smart combination of qualitative and quantitative aspects. In order to address such a complexity, we propose a knowledge-based approach that integrates the systematic computation of heterogeneous criteria with open semantics. The holistic understanding of the framework is described by a reference architecture and the proof-of-concept prototype developed can support high-level system analysis, as well as it suitable within a number of applications contexts - i.e. as a research/educational tool, communication framework, gamification and participatory modelling. Additionally, the knowledge-based philosophy, developed upon Semantic Web technology, increases the capability in terms of holistic knowledge building and re-use via interoperability. Last but not least, the framework is designed to constantly evolve in the next future, for instance by incorporating more advanced AI-powered features.

Genome-scale metabolic model (GEM) is a powerful tool for interpreting and predicting cellular phenotypes under various environmental and genetic perturbations. However, GEM only consid-ers stoichiometric constraints, and the simulated growth and product yield values will show a monotonic linear increase with increasing substrate uptake rate, which deviates from the experi-mentally measured values. Recently, the integration of enzymatic constraints into stoichiometry-based GEMs was proven to be effective in making novel discoveries and predicting new engineer-ing targets. Here we present the first genome-scale enzyme-constrained model (eciCW773) for Corynebacterium glutamicum reconstructed by integrating enzyme kinetic data from various sources using ECMpy workflow based on the high-quality GEM of C. glutamicum (obtained by modifying the iCW773 model). The enzyme-constrained model improved the prediction of pheno-types and simulated overflow metabolism, while also recapitulating the trade-off between biomass yield and enzyme usage efficiency. Finally, we used eciCW773 to identify several gene modifica-tion targets for L-lysine production, most of which agree with previously reported genes. This study shows that incorporating enzyme kinetic information into the GEM enhances the cellular phenotypes prediction of C. glutamicum, which can help identify key enzymes and thus provide reliable guidance for metabolic engineering.

Strategic management sets the direction of a company for several years ahead. Managers and business owners who create strategy must anticipate and have the ability to see systemically—the paper deals with creating a logistics strategy for a company operating in the industrial sector. The first section summarises the theoretical background for strategy and logistics and current trends affecting logistics processes. The second chapter analyses the current state of the art in logistics strategy development and summarizes its problem areas. The central part of the paper is the proposal of a methodology for logistics strategy development in the industrial area by a progressive approach. The methodology is divided into 5 phases - preparatory, analytical, formulation, implementation, and evaluation and control phases. The methodology is partially validated. The created variants of the logistics strategy include the introduction of such elements that will lead to the gradual development of the Industry 4.0 trend in the given company. This is mainly the first variant called "automation," which includes the introduction of an automatic tractor for material import, the introduction of an automatic system for transport and storage of work in progress, and the introduction of a new logistics information system that will make greater use of already established barcodes (transparency, data in real-time).

The rapid infiltration of fake news is a flaw to the otherwise valuable internet, a virtually global network that allows for the simultaneous exchange of information. While a common, and normally effective, approach to such classification tasks is designing a deep learning-based model, the subjectivity behind the writing and production of misleading news invalidates this technique. Deep learning models are unexplainable in nature, making the contextualization of results impossible because it lacks explicit features used in traditional machine learning. This paper emphasizes the need for feature engineering to effectively address this problem: containing the spread of fake news at the source, not after it has become globally prevalent. Insights from extracted features were used to manipulate the text, which was then tested on deep learning models. The original unknown yet substantial impact that the original features had on deep learning models was successfully depicted in this study.

Glaucoma is a blinding disease largely caused by increased resistance to drainage of fluid from the eye’s anterior chamber, resulting in elevated intraocular pressure (IOP). A major site of fluid outflow regulation and pathology is the trabecular meshwork (TM) at the entrance of the eye’s drainage system. We aimed to characterize the structural and functional properties of a newly developed tissue-engineered anterior segment eye culture model. We hypothesized that repopulation of a decellularized TM with non-native TM cells could restore aspects of normal TM. The decellularization protocol removed all cells and debris while preserving the ECM. Seeded cells localized to the TM region and progressively infiltrated the meshwork ECM. Cells reached a distribution comparable to control TM after four days of perfusion culture. After a perfusion rate increase challenge, tissue-engineered cultures reestablished normal IOPs (reseeded = 13.7±0.4 mmHg, decellularized = 35.2±2.2 mmHg, p < 0.0001). eGFP expressing CrFK control cells caused a high and unstable IOP (27.0±6.2 mmHg). In conclusion, we describe a readily available, storable, and biocompatible scaffold for anterior segment perfusion culture of non-native cells. Tissue-engineered organs demonstrated similarities to native tissues and may reduce the need for scarce donor globes in outflow research.

Impulsive noise is the main limiting factor for transmission over channels affected by electromagnetic interference. We study the estimation of (correlated) Gaussian signals in an impulsive noise scenarios. In this work, we analyze some of the existing as well as some novel estimation algorithms. Their performance is compared, for the first time, for different channel conditions, including the Markov-Middleton scenario, where the impulsive noise switches between different noise states. Following a modern approach in digital communications, the receiver design is based on a factor graph model and implements a message passing algorithm. The correlation among signal samples as well as among noise states brings about a loopy factor graph, where an iterative message passing scheme should be employed. As it is well known, approximate variational inference techniques are necessary in these cases. We propose and analyze different algorithms and provide a complete performance comparison among them, showing that both Expectation Propagation, Transparent Propagation, and the Parallel Iterative Schedule approaches reach a performance close to the optimal, at different channel conditions.

The nature of fluid mechanics makes experimentation an important part of a course taught in the subject. Presented here is the application of a novel, large scale multidisciplinary model of practical education in a fluids engineering laboratory. Advantages of this approach include efficiencies through economy of scale leading to better pedagogy for students. The scale justifies dedicated academic resources to focus on developing laboratory classes and giving specific attention to designing activities that meet learning outcomes. Four examples of applying this approach to fluids mechanics experiments are discussed, illustrating tactics that have been developed and honed through many repeated instances of delivery. “The measurement lab” uses a flow measurement context to teach identifying and managing general experimental uncertainty. New students, unfamiliar with fluid mechanics are guided through a process to gain understanding that can be applied to all future experimental activities. The “pressure loss in pipes” lab discusses the advantage and process for sharing equipment and teaching resources between multiple cohorts. The provision for students is adapted for context, such as the degree program or year of study. The “weirs big and small” lab provides a methodology to teach the power of dimensional analysis to mechanical engineers using a field of fluid mechanics that is outside their usual theoretical studies. Finally, the “spillway design” lab discusses mechanisms to deliver student independent, open ended experiments at scale, without excessive staff resource requirement.

At the start of 2020 the rapid onset of the coronavirus pandemic forced higher education institutions across the world to pivot from face to face to remote teaching. For teaching methods that involve the transmission and dissemination of verbal/visual information between academic staff and students, video technologies provided immediate methods to respond to the restricted access to campus. Practical activities, that usually involve interaction with equipment, presented a greater challenge to adapt for remote delivery. With restrictions on higher education being partially lifted, many institutions worldwide intend to offer blended learning, prioritizing in-person activities that are troublesome to deliver online, such as practicals. Social distancing measures are reducing capacity and placing increased pressure on space, creating a need to optimise limited time students have in the lab and strategies to determine which activities can best utilize this limited resource. Time is constrained, leaving little opportunity to make radical changes to learning and teaching structures. In this publication, The department of Mulicdipalnary Engineering Education (MEE) at the University of Sheffield, utilise their experiences in practical teaching to provide simple, implementable ideas for blended practicals which maximize students’ learning and experiences within the envelope of available resources.

Multidisciplinary Engineering Education (MEE) at the University of Sheffield is dedicated to delivering, at scale, practical teaching to students in the Faculty of Engineering. The COVID-19 pandemic initiated the sudden suspension of face to face teaching required MEE to translate over 600 in-lab practicals to a remote delivery format. With little opportunity to coordinate, academic staff independently adopted a variety of tactics to ensure practical learning outcomes were maintained. Following the reactive response, a proactive reflection was conducted and six categories of tactics for remote practicals have been established. These categories are Provide digital artefacts; Simulated practicals; Synchronous remote participation; Asynchronous participation by proxy; Perform procedure in alternative environment; Remote staff support. The advantages and drawbacks of each of these categories is discussed and it is suggested which tactics are appropriate for particular learning outcomes or operational and environmental outcomes of equivalent in-lab practicals. Further work to comprehensively align outcomes to tactics is proposed and lasting benefit from the analysis can be realized by adopting a principle of Remote Enhanced Practicals.

Molecular engineering research is the fundamental way and the only way for the development of biomaterials. Based on molecular engineering, the biocompatibility of natural conformation and polymer biomaterials was studied. In this paper, we discuss that natural conformation is the basis of protein biological function, and that the synergistic action of peptide chain and side group is the motive force for protein to construct natural conformation and complete biological function. On the basis of the influence of the adsorption of polymer biomaterials on the natural conformation of proteins, the relationship between biocompatibility of biomaterials and protein conformation is further explained. Studies have shown that bismuth molecular materials can only be applied in the market and have their functionality if they have good biocompatibility. Therefore, the biocompatibility evaluation of new materials has important practical significance.

Organ-on-Chip is a game-changing technology born from the convergence of tissue engineering and microfluidic technology. Organ-on-Chip devices (OoCs) are expected to offer effective solutions to persisting problems in drug development and personalized disease treatments. This opinion paper surveys the current landscape in research, development, application and market opportunities for OoCs to help establishing a global and multi-stakeholder OoC ecosystem. Based on a bibliometric study, a market analysis, expert interviews, and panel discussions held at the ORCHID Vision Workshop (Stuttgart, 23 May 2018), we outline presently unmet needs, key challenges, barriers and perspectives of the field, and finally propose recommendations towards the definition of a comprehensive roadmap that could render OoCs realistic models of human (patho)physiology in the near future.

The challenges to sustainability governance across multiple geographical/cultural contexts lead us to the “piecemeal engineering” idea advocated by the philosopher Karl Popper, which explicitly considers context. We argue for adopting the piecemeal engineering approach, augmented by adaptive policies and modern (online) collaboration platforms to maximize the prospects of sustainable practices worldwide. This recommended course is not intended to be a theory in itself. Rather, it is a well-grounded, practical and practicable stop-gap measure in times when complexity and change outpace theories and strategies. We present a philosophical foundation for this “Augmented Popperian Experimentation.” Focusing on The Water Network (the largest collaborative platform for water researchers and professionals), we show that sustainability-oriented organizations in the water realm and others are inching toward the practice we advocate. We discuss implications.

Good transportation systems are pre-requisites to economic development. Materials used for road construction are traditionally classified based of empirically developed archaic tests, often classifying naturally available materials as unsuitable for use in the load-bearing road pavement layers. Consequently, design standards usually require the use of imported materials at considerable costs, severely restricting road network development under scenarios of limited funding. New technologies and test methods based on sound engineering criteria, incorporating basic material sciences can substantially change this scenario. Nano-silane technologies can be utilised to improve naturally available materials to meet the engineering requirements of all layers in road structures. Material test and design methods have been developed and successfully tested in South Africa to build New-age Modified Emulsion (NME) stabilised layers in roads, meeting all engineering requirements. Accelerated Pavement Tests (APT) done on actual roads, proved the concept. This work is based on a scientific approach and identification of various factors that will impact on the successful application of applicable nanotechnology solutions. This paper aims to identify these fundamental factors that are a pre-requisite for the evaluation of nanotechnology solutions to ensure that new technologies are introduced into pavement engineering designs at a low risk to any implementing.

Requirements elicitation from multiple human sources involves information validation and conflicts resolution to deal with uncertainty inherent in the process. Most requirements analysis methods focus on expressing the requirements and ignore the uncertainty inherent in the process of requirements elicitation. This paper build on the work presented by the author in [1,10], where a method for requirements elicitation from multiple sources was presented. This paper gives an overview of the method and presents detailed case studies. The cases studies were designed to illustrate different aspects of the Source Control Method and to determine its feasibility and its practical utility as a problem investigation and information validation technique.

Coastal protection solutions can be categorized as grey, hybrid, or natural. Grey infrastructure includes artificial structures like dykes. Natural habitats like seagrasses are considered as natural protection infrastructure. Hybrid solutions combine both natural and grey infrastructure. Evidence suggest that grey solutions can negatively impact the environment, while natural habitats prevent flooding without such adverse effects and provide many ecosystem services. New types of protective solutions, called biomimetic solutions, are inspired by natural habitats and reproduce their features using artificial materials. Few studies have been conducted on these new approaches. This study aims to quantify wave dissipation observed in-situ above a biomimetic solution inspired by kelps, known for their wave dampening properties. The solution was deployed in full water column nearby Palavas-les-Flots in southern France. A one-month in-situ experimentation showed that the biomimetic solution dissipates around 10% of total wave energy on average, whatever the meteo-marine conditions. Wave energy dissipation is frequency dependent: short waves are dissipated, while low-frequency energy increased. An anti-dissipative effect occurs for forcing conditions with frequencies close to the eigen mode linked to the biomimetic solution's geometry, suggesting that resonance should be considered in designing future biomimetic protection solutions.

Several degraded ecosystems worldwide need to attain a stable vegetal cover after mandatory active restoration; thus, plants soil, and soil biodiverse microbes can be taken into consideration for a successful mission. Among environmental and biological variables, interactions with soil and its microorganisms are critical for permanent plant establishment. Thus, advances in the use of microbial management such as inoculation of mycorrhizas and bacteria become a modern aspect of active restoration programs. New strengths have increased worldwide since most plant species, including grasses, associate to mycorrhizae, Thus, a high percent of restored areas, which were degraded due to the increasing mining or conventional agriculture, rest to be monitored. This study shows current reports on active restoration of degraded landscapes as well as the methodology required for a successful ecosystem restoration. Active restoration has been recognized as the best method to restore tropical forests; however, the higher cost of active restoration, which requires planting seedlings, direct seeding, root inoculation of microorganisms or mitigation of disturbances, which aid the recovery process despite monitoring. On the other hand, the spontaneous recovery of trees that colonize and establish in abandoned fields or natural disturbances, named natural regeneration, is less targeted and can require more time to achieve results. We conducted a web of science search of recent published articles on restoration and analyzed them. We confirmed the important role of AMF on root colonization and soil AMF spores as drivers of restoration of main ecosystems according to previous results from restored sites in Brazil. In the restored sites AMF (more than 8 species) was generally found, while in preserved sites dominated Gigaspora, Scutellospora and Acaulospora, restored sites presented relevant AMF richness as well as the preserved sites. Root colonization by AMF in experimental mined sites was high. Vesicles were frequent in restored sites.; however, colonization intensity was generally the same despite treatments. In general, in the control treatments, values of root colonization remain high.

Regenerative medicine is an interdisciplinary field that combines stem cell technology and tissue engineering to replace or regenerate human cells, tissues, or organs and restore normal functions. The term 'regenerative medicine' was coined by William Haseltine at a 1999 conference on Lake Como. Since its inception in 1968, the field has offered clinical benefits for the regeneration, repair, and restoration of bones, skin, cartilage, neural tissue, and heart, as well as scaffold fabrication. Inorganic biomaterials play a crucial role in regenerative medicine, often surpassing traditional synthetic materials due to their adjustable intrinsic properties, such as size, topography, charge, and chemical stability. These properties enable a flexible cellular response within intracellular matrix environments and provide structural support for cell adhesion and tissue regeneration. Given the wide gamut of near-future potential applications of inorganic biomaterials, this article gives an overview of their emerging roles in stem cell regenerative research, tissue engineering, artificial skin and cartilage regeneration, neural nerve injuries, 3D bioprinting, and development of new inorganic bio-scaffolds. The review also addresses the challenges related to the clinical application and tissue compatibility of inorganic biomaterials, utilizing current state-of-the-art techniques.

Trenchless technologies are utilized for installing new pipes or repairing and replacing old ones, categorized into construction and rehabilitation/replacement methods. With rapid urbanization, new pipe networks must be established in proportion to population growth. Additionally, due to issues like corrosion and aging, a significant portion of storm sewers, waste sewers, gas, and water pipes need annual replacement or rehabilitation. Traditional open-cut methods are no longer viable due to traffic disruption, safety concerns, environmental impacts, and high costs. Trenchless construction methods address these issues effectively. Construction trenchless methods include horizontal earth boring (no worker entry), pipe jacking, and utility tunneling (worker entry). Horizontal earth boring is further divided into horizontal auger boring, horizontal directional drilling, pipe ramming, microtunneling, pilot tube microtunneling, and compaction methods. The first part of the current study reviews and discusses the design criteria for horizontal auger boring, horizontal directional drilling, pipe jacking, pipe ramming, and microtunneling. Based on these unique project requirements, the optimal trenchless technology can be proposed. In the second part of the research, a hierarchical algorithm is proposed, based on literature concepts, to serve as a decision-making tool for selecting the optimal trenchless construction method. Sometimes, multiple methods may be proposed, with the best one chosen based on equipment availability, skilled teams, budget, and sustainability principles. Although trenchless renewal methods are not the focus of this study, their design criteria are provided in the appendices to offer a comprehensive collection of design requirements for all trenchless technologies.

This study presents an innovative method for producing thermosensitive bio-ink from chitosan hydrogels saturated with carbon dioxide and agarose. It focuses on a detailed characterization of their physicochemical properties and potential applications in biomedicine and tissue engineering. The ORO test approved rapid regeneration of the three-dimensional structure of chitosan-agarose composites in the unidirectional bench press simulation test. The diffusion of dyes through chitosan-agarose hydrogel membranes strongly depended on the share of both polymers in the composite and the molecular weight of the dyes. Glucose, as a nutrient marker, also diffused through all membranes regardless of composition. Biocompatibility assessment using MTT tests on 46BR.1N fibroblasts and HaCaT keratinocytes confirmed the safety of the bioink. The regenerative potential of the bioink was confirmed by efficient cell migration, especially HaCaT. Long-term viability studies showed that chitosan-agarose scaffolds, unlike the agarose ones, support cell proliferation and survival, especially 14 days after bioink extrusion. Experiments in a skin wound model in mice confirmed the biocompatibility of tested dressing and the beneficial action of chitosan on healing. Studies on vessel formation in chicken embryos highlight the potential of the chitosan-agarose composition for enhancing proangiogenic effects. This composition meets all entry criteria and possesses excellent biological properties.

Yeasts are gaining increasing attention for their potential health benefits as probiotics in recent years. Researchers are actively searching for new yeast strains with probiotic properties (i.e, Debaryomyces hansenii; Kluyveromyces marxianus; Yarrowia lipolytica; Pichia hudriavzevii; Torulaspora delbrueckii) from various sources, including traditional fermented foods, the human gut, and the environment. This exploration is expanding the pool of potential probiotic yeasts beyond the well-studied Saccharomyces boulardii. Research suggests that specific yeast strains possess properties that could be beneficial for managing conditions like inflammatory bowel disease, irritable bowel syndrome, skin disorders and allergy. Additionally, probiotic yeasts may compete with pathogenic bacteria for adhesion sites and nutrients, thereby inhibiting their growth and colonization. They might also produce antimicrobial compounds that di-rectly eliminate harmful bacteria. To achieve these goals, the approach that uses probiotics for human health is changing. Next-generation yeast probiotics are emerging as a powerful new approach in the field of live biotherapeutics. By using genetic engineering, scientists are able to give these tools specialized capabilities. However, most research on these probiotic yeasts is still in its early stages, and more clinical trials are needed to confirm their efficacy and safety for various health condi-tions. This review could provide a brief overview of the situation in this field.

Biotic stressors pose serious threats to crop yield, jeopardizing food security and resulting in over US$220 billion per year losses to the agriculture industry. Plants activate innate defense mechanisms upon pathogen perception and invasion. The plant immune response comprises numerous concerted steps, including the recognition of invading pathogens, signal transduction, and activation of defensive pathways. However, pathogens have evolved various structures to evade plant immunity. Given these facts, genetic improvements of plants are required for sustainable disease management to ensure global food security. Advanced genetic technologies have provided new opportunities to revolutionize and boost plant disease resistance against devastating pathogens. Furthermore, targeting susceptibility (S) genes, such as OsERF922 and BnWRKY70, through CRISPR methodologies offers novel frontiers to disrupt the molecular compatibility of pathogens and introduce durable resistance against them in plants. Here, we provide a critical overview of advances in understanding disease resistance mechanisms. The review also critically examines management strategies under challenging environmental conditions and R gene-based plant genome engineering systems intending to enhance plant responses against emerging pathogens. This work underscores the transformative potential of modern genetic engineering practices in revolutionizing plant health and crop disease management while emphasizing the importance of responsible application to ensure sustainable and resilient agricultural systems.

The number of engineers who are transitioning into environmental sustainability careers is growing, though a gap still exists between supply and demand. This presents an opportunity for undergraduate engineering students to fulfill the demand as environmental sustainability professionals. This qualitative exploratory study investigated environmental sustainability learning experiences and future career interests in environmental sustainability. The social cognitive theory (SCCT) was utilized as a theoretical lens, exploring undergraduate students’ environmental sustainability interests, related learning experiences and their interest in pursuing a future career in environmental sustainability. Twenty-five undergraduate engineering students in various engineering disciplines were interviewed for this study. Data were analyzed to (1) identify students’ interest in pursuing a career in environmental sustainability, (2) determine if students’ interests have changed since they began their undergraduate studies, and (3) explore how learning experiences have impacted students’ future career choice. The findings posit that exposure to environmental sustainability learning experiences is impactful and plays an important role, impacting students' interests in pursuing careers in sustainability. Results reveal that elements such as personal beliefs and salary considerations inspire career choices. This research contributes to addressing the demand for additional working professionals who are prepared to tackle environmental sustainability issues, highlighting the role of learning experiences in shaping students' career interests.

Conductive hydrogels combine the properties of both hydrogels and electrical conductivity, making them soft, flexible, and biocompatible. These properties enable them to conform to irregular surfaces, stretch and bend without losing their electrical conductivity, and interface with biological systems. Conductive hydrogels can be utilized as conductive traces, electrodes, or as a matrix for flexible electronics. Exciting applications in sensors, tissue engineering and human-machine interaction have been demonstrated worldwide. This review comprehensively covers the progress in this field, focusing on several main aspects: functional materials, performance improvement strategies, and wearable applications in human-related areas. Furthermore, the major approaches and challenges for improving their mechanical properties, conductivity, and long-term stability are systematically summarized.

In his Cynefin classification of system behaviours, Snowden (2007), posits that his Simple (Clear ) and even Complicated system categories are sufficiently understandable to be amenable to the traditional methods of system analysis and modelling. Here, relationships between entities in the system are clear and unambiguous and they behave predictably. So, we can build mathematical “models” to describe how the system is expected to function in different situations. Leaving aside his fourth category, Chaotic, as too hard for the moment, this paper focusses on the third category, Complex systems. Yaneer Bar-Yam [1998] defined "complex systems" as systems that "have multiple interacting components, whose collective behaviour cannot be simply inferred from the behaviour of components." So, the traditional system modelling approach of decomposition into components and attempting to build up a picture of system performance from individual pieces, alone, is no longer appropriate for this class. The problem is that most of the methods we currently employ, and particularly the methods that attempt to make the case that these systems are safe, (such as Fault trees, FMEA. and Root Cause Analysis), rely on this decomposition into components and attempt to make the parts more reliable individually. Even the more systemic approaches, such as Reason’s (1990) Swiss Cheese Model, and Leveson’s System Theoretic Analysis (STPA) (2004), encourage adding layers of protection, barriers, strengthened control loops and better safety critical systems, in the hope that they are making the systems safer. But without a valid model to test the effectiveness of these add-ons, we cannot be sure that they will not have an opposite effect, of making the systems even more complex, unreliable and unpredictable. For complex systems, the careful gathering and documentation of system components and their relationships is still a necessary first step, but we need an additional capacity to model these complex systems to test out and prove they work safely and successfully in the real world. (not just draw fixed component relationships as system “models” (wiring diagrams)) In the last 10 years, an original approach developed by Hollnagel, (2012), to model systems as sets of interactive, interdependent “functions”, (abstracted from and independent of component details), has now been developed to the point where it can take the basic data and structures from the current component focussed system engineering “models”, such as MBSE( Goldberg (1994)), and can pull it all together in a dynamic system model from which analysts can discern how it really works in practice, and predict the emergent behaviours characteristic of complex systems. The paper describes how this methodology developed and how it is currently used to model real situations, to analyse incidents and anticipate events.

For years, effort has been devoted to establishing an effective bull breeding soundness evaluation procedure; usual research on the subject is based on BBSE (Bull Breeding Soundness Examination) methodologies, which have significant limitations in terms of their evaluation procedure, such as high cost, time-consuming and administratively difficult. This research focuses on the creation of a prediction model to supplement and/or improve the BBSE approach, through the study of two algorithms, Clustering and Artificial Neural Network (ANN), to find the optimum Machine Learning (ML) approach for our application, with an emphasis on data categorization accuracy. The tool is designed to assist veterinary medicine and farmers in identifying key factors and increasing certainty in their decision-making during the selection of bulls for breeding purposes, providing data from a limited number of factors generated from a deep pairing study of bulls. Zebu Bulls, European Bulls, and Crossbred Bulls were the general groupings. The data utilized in the model's creation (N =359) considers five variables that influence improvement decisions. This approach enhances decision-making by 12%. ANN obtained an accuracy of 90%; Precision was 97% for satisfying, 92% for unsatisfying, and 85% for Bad.

The following are three successful case studies that affected humanity during the COVID-19 pandemic. The key to their success was their swift adaptability to the new confinement regulations and the unyielding determination of those carrying out their plans. Various extraordinary and prompt efforts were required to make the essential logistical preparations and practice for distance learning a reality. The research activities were also reorganized so the thesis students might carry out a few simple experiments at home under the strict supervision of their advisers and supervisors. The findings revealed that the students who participated in these projects applied all the abilities they had gained before the epidemic. In addition, they included crucial knowledge about the project, problem-based, and research-based learning. Furthermore, because they were the primary implementers of the trials, they learned by planning and performing experiments. The culmination of these efforts was displayed in three scientific publications that were published in index journals with a high level of impact. This latter factor is essential for the prestige and visibility of the university, in addition to the acquisition of financing and grants for scientific research.

In our discipline, some numbers, facts and methods are established and unquestionable. Still, a credible source is hard to find. Likewise, some state-of-the-art papers do not have the right focus, or they do not point at current research trends and future mission statements. Lastly, reliable resources for claims and statements that are arguable are not always found in the typical journals of our field. The paper at hand solves this issue for our field. Through critical review of a vast amount of journal papers, monographs, handbooks, conference proceedings and theses as well as manuals, textbooks and tutorials, numbers, facts, methods, claims statements, state-of-the-art, research and application trends are reflected and substantiated.

Critical sized bone defects and articular cartilage injuries resulting from trauma, osteonecrosis or age-related degeneration are often nonhealing by the physiological repairing mechanisms, thus representing a clinic issue due to the relevant epidemiological incidence. Current treatment approaches consist of autologous or allogenic grafting, which are associated with painfulness, morbidity, risk of infections and rejection. Novel tissue-engineering approaches, aiming at the reconstruction of damaged tissues, have been proposed as alternative solutions to these conven-tional methods. These approaches are based on the combination of three fundamental compo-nents: autologous or allogenic cells, a scaffold and growth-stimulating signals, which are gener-ally referred to as the tissue engineering triad. Three-dimensional polymer networks are fre-quently used as scaffolds to allow cell proliferation and tissue regeneration. In this scenario, cryogels are giving promising results as cell scaffolds over other polymer networks, thanks to their peculiar properties. In particular, cryogels possess an interconnected porous structure and a typical sponge-like behaviour, which facilitate the cellular infiltration and ingrowth. Their properties can be appropriately adjusted to match the requirements of the specific tissue or or-gan that it is intended to regenerate. In this review it is reported the state of the art on the fabri-cation and employment of cryogels in supporting osteo or chondro-genic differentiation for the re-building of more organized tissues. Moreover, it will highlight current progress and future perspectives in the implementation of this technology in the clinical practice.

This work aimed to synthesize new N-rich triazole compounds. In this article we describe the synthesis of three different 1,2,4-triazole-based systems starting from 1,3-diaminoguanidine hydrochloride monohydrate. The structure of the obtained crystals was established by single-crystal X-Ray diffraction. The high nitrogen content of the synthetized compounds makes them building blocks for the application in the High-Energy-Density material science.

In the last two decades, gelatin-based hydrogels have been widely used as tissue engineering scaffolds due to their excellent biocompatibility, biodegradability, easy processability, transparency, non-toxicity, and reasonable structural similarity to the natural extracellular matrix (ECM). However, intrinsic low mechanical properties of gelatin are not structurally and mechanically suitable to support cell growth and proliferation. That’s why various crosslinking strategies including physical, chemical, enzymatic and combination of them as well as networking patterns including double network, interpenetrating network and nano reinforcing mechanism have been utilized to enhance the structural stability and mechanical integrity of gelatin. In this review, the advances in modulating the mechanical properties of gelatin-based hydrogels for the design and development of structurally stable scaffolds for tissue engineering are discussed. The optimized crosslinking parameters with the adequate mechanical properties of gelatin-based hydrogels are reviewed. Gelatin-based scaffolds for a wide range of tissue engineering applications, such as bone, cartilage, cardiac, skin, and nerve tissue engineering are also outlined. Lastly, current challenges and future perspectives in this research field are presented.

Herein are reported the main pros and cons of using the conversational Artificial Intelligence (AI) called “ChatGPT” as a tool to write essays in the academic and scientific community. Also, selected mathematical and scientific problems were solved to determine the effectiveness of this platform. To this end, a systematic case study was designed and conducted to obtain data generated by the chatbot that was further analyzed. Six different topics were chosen for the essay evaluation, and the introduction section was created using this AI. Meanwhile, six mathematical problems related to integrals, z-transform, Laplace transform, and resolution of ordinary differential equations were solved. Furthermore, a scientific problem associated with the structural properties of material was also solved. After data analysis, we concluded that the conversational platform could quickly create written essays with well-structured words. Nonetheless, in almost all cases, the chatbot only used basic information to complete the introduction, and no critical writing was obtained. For the mathematical case study, we observed that under the current status of the ChatGPT, this platform has no ability or skills to solve complex equations. Thus, significant changes need to be conducted to improve the performance of ChatGPT for mathematical solutions.

Mesenchymal stem cells (MSCs) are an attractive therapeutic tool for tissue engineering and regenerative medicine owing to their regenerative and trophic properties. The best-known and most widely used are bone marrow MSCs which are currently being harvested and developed from a wide range of adult and perinatal tissues. MSCs from different sources are believed to have different secretion potentials and production which may influence their therapeutic effects. To confirm it, we performed a quantitative proteomic analysis based on the TMT technique of MSCs from three different sources: Wharton’s jelly (WJ), dental pulp (DP) and bone marrow (BM). Our analysis focused on MSC biological properties of interest for tissue engineering. We identified a total of 611 differentially expressed human proteins. WJ-MSCs showed the greatest variation compared with the other sources. WJ produced more extracellular matrix (ECM) proteins and ECM-affiliated proteins and appeared more able to modulate the inflammatory and immune response. BM-MSCs displayed enhanced differentiation and paracrine communication capabilities. DP-MSC appeared to promote exosome production. The results obtained confirm the existence of differences between WJ, DP and BM-MSC and the need to select the MSC origin according to the therapeutic objective sought.

This research examines the impact of social equity on energy consumption. We constructed a digital twin for residential energy consumption by enriching the synthetic population with real-world surveys and feeding them with other environmental and appliance data to the energy modeling framework. We analyzed household hourly energy consumption data from Albemarle County and Charlottesville City in Virginia, USA, for the year 2019. We used clustering analysis to identify patterns in social equity and energy consumption. The results demonstrated the impact of different residential attributes on energy poverty. Statistical analyses, including ANOVA and Chi-Squared tests, were conducted to test for significant differences between racial groups in quantitative and categorical variables. The study found that race is significant in determining the location and quality of housing. People of color often live in areas with higher pollution and less access to green spaces. Additionally, income levels and the age of the house are influential factors in determining energy efficiency. Future work should focus on collecting and analyzing data at the country level and using qualitative data collection methods to gain a more comprehensive understanding of social equity issues concerning energy consumption. Overall, this study provides valuable insights into the relationship between different residential attributes and energy consumption, which can inform policy development to promote more equitable and sustainable communities.

In this study, 3D printing of poly-l-lactic acid (PLLA) scaffolds reinforced with graphene oxide (GO) nanoparticles via Digital Light Processing (DLP) was investigated to mimic bone tissue. Stereolithography is one of the most accurate additive manufacturing method, but the dominant available materials used in this method are toxic. In this research, a biocompatible resin (PLLA) was synthetized and functionalized to serve the purpose. Due to the low mechanical properties of the printed product with the neat resin, graphene oxide nanoparticles in three levels (0.5, 1, and 1.5 Wt.%) were added with the aim of enhancing the mechanical properties. At first, the optimum post cure time of the neat resin was investigated. Consequently, all the parts were post-cured for three hours after printing. Due to the temperature-dependent structure of GO, all samples were placed in an oven at 85 ° C for different time periods of 0, 6, 12, and 18 hours to increase mechanical properties. The compression test of heat treated samples reveals that the compressive strength of the printed parts containing 0.5,1, and 1.5 % of GO increased by 151,162 ad 235%, respectively. Scaffolds with the designed pore sizes of 750 microns and a porosity of 40% were printed. Surface hydrophilicity test was performed for all samples showing that the hydrophilicity of the samples increased with increasing GO percentage. The degradation behavior of the samples was evaluated in a PBS environment, and it revealed that by increasing GO, the rate of component degradation increased, but the heat treatment had the opposite effect and decreased the degradation rate. Finally, besides improving biological properties, a significant increase in mechanical properties under compression can introduce the printed scaffolds as a suitable option for bone implants.

It is known that people perceive animacy in objects. However, many studies on animacy and emotional expressions are limited in that the investigated motions were created by experimenters themselves. This makes the objective validity unclear. Moreover, it remains unclear what types of movements can express emotions with animacy due to the limited number of investigations examining both animacy and emotional expressions. Therefore, we investigated the motion elements for both animacy perception and emotional expressions using simple objects that lack features of specific living things, such as eyes, ears, tails, and voices in this study. First, we investigated the motion elements for animacy perception and emotional expressions using a robot simulator that enabled participants to create undulatory motions by tuning parameters for speed, height, and randomness. In total, 64 participants created motions in Normal (neutral), Joy, Sad, Relaxed, and Angry conditions. The results showed that the medians of speed and height in Normal, related only to animacy, were 0.5569[Hz] and 3.050cm at the edges/4.575cm at the center. The differences in Joy were 0.4028[Hz] and 3.348cm/5.022cm, in Sad were −0.1652[Hz] and −0.9982cm/−1.497cm, in Relaxed were −0.1979[Hz] and −0.4902cm/−0.7353cm, and in Angry were 0.5212[Hz] and 4.688cm/7.032cm. Second, we investigated whether the motion elements revealed in the first experiment were sufficient to express emotions with animacy, using a robot simulator that reflected the results of the motion element investigation. In total, 44 online participants observed the simulator. The results showed that participants could understand emotional arousal levels at the same time as animacy, but they did not fully understand emotional valence. Our findings provide design guidelines for robots that exhibit emotional expressions and closely interact with humans.
