Mancozeb is extensively used fungicide to prevent citrus melanose in most Asian countries, especially in China. So far, however, there have been no reports of thet effect of Mancozeb on the citrus rhizosphere bacterial community. Therefore, this comparative experiment defined the genomic and functional related to community and soil health of 2-years old Citrus unshiu Marc. rhizosphere through amplicon sequencing and chemical analysis. This study evaluated the effect of mancozeb on the chemical properties of citrus-cultivated soil and the richness and diversity of rhizosphere bacterial community. We also investigated the abundance response of rhizosphere bacterial groups to 0, 2, 4, 6 and 8 times application of 2 g mancozeb (active ingredient content, ai.) 600 times diluted with water. Our data revealed that the abundance of rhizosphere-associated bacterial species increased significantly after planting citrus. The relative abundance of Candidatus, Saccharibacteria, Parcubacteria, and Proteobacteria increased with the increase in mancozeb watering times. Meanwhile, the abundance of Nitrospirae decreased with the increase in mancozeb application times. The findings indicated that the chemical properties of the soil and the richness and diversity of rhizosphere bacterial community did not significantly differ across the mancozeb gradients in soil.

Melanophryniscus admirabilis (admirable red-belly toad) is a microendemic and critically endangered species found exclusively along 700 meters of the Forqueta River, in a fragment of Atlantic Forest in southern Brazil. One of the greatest concerns regarding the conservation of this species is the extensive use of pesticides in areas near their natural habitat. In recent years, the adaptation and persistence of animal species in impacted environments have been associated with microbiota. Therefore, the current study aimed to characterize the oral bacterial community of wild M. admirabilis and to address the question of how this community might contribute to toad’s adaptation in the anthropogenic environment and its general metabolic capabilities. In the present study, 11 oral samples collected from wild M. admirabilis were characterized and analyzed via high-throughput sequencing. A total of 181,350 sequences were obtained, resulting in 16 phyla, 34 classes, 39 orders, and 77 families. Proteobacteria dominated (53%) the oral microbiota of toads followed by Firmicutes (18%), Bacteroidetes (17%), and Actinobacteria (5%). No significant differences in microbial community profile from among the samples were reported, suggesting that the dietary restriction may directly influence the bacterial composition. Functional inference of microbiome was performed using PICRUSt2 and important pathways, such as xenobiotic degradation pathways to pesticides and aromatic phenolic compounds were detected, suggesting that the bacterial communities may have important roles in M. admirabilis health and survival in the anthropogenic environment. Overall, our results have important implications for the conservation and management of this microendemic and critically endangered species.

Pito is a traditionally brewed alcoholic beverage in some African countries. It is gaining much prominence and the patronage among the youth. Therefore, samples of the drink were collected every week for six weeks from three different popular brewing sites at Lower Prampram in the Ningo-Prampram District of Accra, Ghana. The samples were processed and examined for bacteria and fungi using the Standard Plate Count (SPC) technique. A total of six different bacteria and a fungus were isolated. The bacteria were Escherichia coli, Klesiella pneumoniae, Shigella spp, Enterobacter aerogenes, Staphylococcus aureus and Pseudomonas aeroginosa, whiles the fungus was Saccharomyces cerevisiae. Total viable counts as well as individual isolates counts in all the pito samples were found to be less than 104 cfu/ml. It is noteworthy that, Saccharomyces cerevisiae, the only fungus isolated is known to be associated with fermentation and the microbes isolated from the pito samples were found to be within the permissible limits. However, these potentially pathogenic microbes, if found in unacceptable limits, from the fermenting samples could merit public health attention. Therefore, periodic screening of pito and their brewers, coupled with education on the maintenance of recommended guidelines concerning food and drink production is encouraged.

Biofertilizer, an environment-friendly and renewable plant nutrient source, has been widely applied and studied to reduce dependency on chemical fertilizers. However, most studies focus on the effects of biofertilizer on the bacterial and fungal communities, and we still lack an understanding of biofertilizer on the protistan community. Here, the effects of biofertilizer application on the composition and interaction of the protistan community in the wheat rhizosphere were investigated based on a 4-year field experiment. Biofertilizer application altered soil physicochemical properties and the protistan community composition (ANOSIM, p < 0.001), and significantly induced an alpha diversity decline. Random forecast and redundancy analysis demonstrated that nitrogenase activity and available phosphorus were the main drivers. Trichomonas classified to the phylum Metamonada was enriched by biofertilizer, and was significantly positive connections with soil nitrogenase activity and some function genes involved in nitrogen-fixation and nitrogen-dissimilation. Biofertilization loosely connected biotic interactions, while did not affect the stability of the protistan community. Besides, biofertilizer promoted the connections of protists with fungi, bacteria, and archaea. Combined with the conjunct biotic network (protist, fungi, bacteria, and archaea) and interactions between protists and soil physicochemical properties/function genes, protists may act as keystone taxa potentially driving soil microbiome composition and function.

To investigate the composition of caproic acid-producing bacterial communities in sub-high-temperature Daqu, we analyzed four different Daqu samples from Sichuan. After enriching the samples with a specialized medium, we performed high-throughput sequencing of 16S rRNA gene amplicons on the metagenomic DNA from both the Daqu samples and the enrichment cultures. Our analysis focused on identifying and characterizing the caproic acid-producing bacterial communities present in Daqu. The results revealed that enrichment increased both the diversity and abundance of potential caproic acid-producing bacteria in Daqu. The predominant caproic acid-producing bacteria, with a relative abundance greater than 1%, included Clostridium sensu stricto 18 (2.19% to 69.00%), Clostridium sensu stricto 15 (1.76% to 7.65%), as well as smaller amounts of Clostridium sensu stricto 12, Clostridium sensu stricto 1, Clostridium sensu stricto 10, Clostridium sensu stricto 7, and Caproiciproducens. Specifically, Q1 Daqu was characterized by the presence of Clostridium sensu stricto 18 and Clostridium sensu stricto 15. Q2 Daqu featured Clostridium sensu stricto 1, Caproiciproducens, and Clostridium sensu stricto 18. Q3 Daqu included Clostridium sensu stricto 18 and Caproiciproducens. Q4 Daqu contained Clostridium sensu stricto 18 and Clostridium sensu stricto 1. These findings indicate that there are variations in the composition of caproic acid-producing bacterial communities among different sub-high-temperature Daqu samples from Sichuan. This preliminary analysis provides a theoretical basis for further studies on caproic acid bacteria in Daqu and offers insights into the development of high-quality Daqu, which may enhance the quality of Nongxiangxing liquor.

The impact of the cell community on its individual components in the prokaryotic realm is often overlooked. However, in the case of intracellular pathogens, where each infected cell can be considered as a single community, understanding how a population adapts to its environment to evolve and successfully propagate is key. Chlamydia infections are characterised by a silent propagation of the bacteria within individual hosts and the wider population. Chlamydia are strict intracellular pathogens residing within a specialised membrane-bound compartment called the inclusion. The life cycle of Chlamydia involves altering between the invasive elementary bodies (EBs) and replicative reticulate bodies (RBs). We have used cryo-soft X-ray tomography to observe individual inclusions, combining excellent resolution (40 nm) and large volume imaging (up to 16 µm). Combined with a semi-automated segmentation pipeline, we were able to consider each inclusion as an individual bacterial niche. Within the inclusion, we identified and classified different forms of the bacteria and confirmed the recent finding that RBs have a variety of volumes (small, large and abnormal). Moreover, we demonstrate that the proportions of these different RB forms depend on the bacterial concentration in the cell demonstrating the impact of the group on its individual component. We conclude that each inclusion operates as an autonomous community which regulates the characteristics of individual bacteria within the inclusion

Bacterial communities in rhizosphere and root nodules have significant contributions to the growth and productivity of the soybean (Glycine max L.). In this report, we analyzed the physiological properties and dynamics of bacterial community structure in rhizosphere and root nodules at different growth stages using BioLog EcoPlate and high-throughput sequencing technology, respectively. The BioLog assay found that the metabolic capability of rhizosphere is in increasing trend in the growth of soybeans as compared to the bulk soil. As a result of the Illumina sequencing analysis, the microbial community structure of rhizosphere and root nodules was found to be influenced by the variety and growth stage of the soybean. At the phylum level, Actinobacteria were the most abundant in rhizosphere at all growth stages, followed by Alphaproteobacteria and Acidobacteria and the phylum Bacteroidetes showed the greatest change. But, in the root nodules Alphaproteobacteria were dominant. The results of the OTU analysis exhibited the dominance of Bradyrhizobium during the entire stage of growth, but the ratio of non-rhizobial bacteria showed an increasing trend as the soybean growth progressed. These findings revealed that bacterial community in the rhizosphere and root nodules changed according to both the variety and growth stages of soybean in the field.

Soil bacteria drive key ecosystem functions, including nutrient mobilization, soil aggregation and crop bioprotection against pathogens. Bacterial diversity is thus considered a key component of soil health. Conventional agriculture reduces bacterial diversity in many ways. Compost tea has been suggested as a bioinoculant that may restore bacterial community diversity and promote crop performance under conventional agriculture. Here, we conducted a field experiment to test this hypothesis in a soybean-maize rotation. Compost tea application had no influence on bacterial diversity or community structure. Plant growth and yield were also unresponsive to compost tea application. Combined, our results suggest that our compost tea bacteria did not thrive in the soil, and that the positive impacts of compost tea applications reported elsewhere may be caused by different microbial groups (e.g., fungi, protists, nematodes) or by abiotic effects on soil (e.g., contribution of nutrients and dissolved organic matter). Further investigations are needed to elucidate the mechanisms through which compost tea influences crop performance.

Phytoremediation, a method of phytomanagement using the plant holobiont to clean up polluted soils, is particularly effective for degrading organic pollutants, such as alkanes and polycyclic aromatic hydrocarbons (PAHS). However, the respective contributions of host plants and their associated microbiota within the holobiont to the efficiency of phytoremediation is poorly understood. Identification of plant-associated bacteria capable of efficiently utilizing these compounds as carbon source while stimulating plant-growth, is a keystone for phytomanagement engineering in order to improve the efficiency of pollutant removal. In this study, we sampled the rhizosphere and the surrounding bulk soil of Salix purpurea and Eleocharis obusta from the site of a former petrochemical plant in Varennes, QC, Canada. Our objectives were to: (i) isolate and identify indigenous bacteria inhabiting these biotopes; (ii) assess the ability of isolated bacteria to utilize alkanes (dodecane and hexadecane) and PAHs (naphthalene, phenanthrene and pyrene) as the sole carbon source, and (iii) determine the plant growth-promoting (PGP) potential of the isolates using five key traits. A total of 438 morphologically different bacterial isolates were obtained, purified, preserved and identified through PCR and 16S rRNA gene sequencing. Identified isolates represent 62 genera, including taxa such as Acinetobacter, Arthrobacter, Bacillus, Enterobacter, Nocardia, Pseudomonas, Rhodococcus, Streptomyces and Variovorax. Approximately, 32% of bacterial isolates, including Arthrobacter, Pseudomonas, Streptomyces, Enterobacter, Nocardia, Acinetobacter and Microbacterium, were able to utilize all five different hydrocarbons compounds. Additionally, 5% of tested isolates belonging to genera Pseudomonas, Acinetobacter, Serratia, Klebsiella, Microbacterium, Bacillus and Stenotrophomonas possessed all five of the tested PGP functional traits. This culture collection of diverse, petroleum-hydrocarbon degrading bacteria, with multiple PGP traits, represents a valuable resource for future use in environmental bio- and phyto-technology applications, including phytoremediation of petroleum hydrocarbons contaminated soils and phytomanagement of anthropized areas.

Russia is one of the largest cereal grain exporters in the world, churning out 34,3 million tons of export grain (wheat, barley and oats) in 2021. Plant infectious pathogens continue to be among the main factors in yield loss in the field and are a danger to the grain exporting industry's ability to expand internationally. This is primarily due to phytosanitary restrictions imposed by nations that monitor the presence and absence of certain phytopathogens in imported goods. Phytosanitary measures prevent the spread of plant pathogens, thus cutting the cost of dealing with them, once the pathogens invade new agricultural regions. This paper is devoted to the detection and identification of bacteria in samples of grain crops of three regions in The Republic of Crimea. The objects of the study were bacterial isolates from plant samples particularly wheat, oats, barley and triticale. The study was conducted in 2021. The identification of the isolates was carried out by sequencing a section 16–23S of the rRNA amplified by PCR with 8UA/519B, 27f/907r and PSf/PSr primers. Nucleotide sequences were deciphered using the Bio Edit program and compared with sequences placed in GenBank (https://blast.ncbi.nlm.nih. gov). The result of identification was considered an organism with maximum similarity. As a result, 38 samples of grain crops were collected, 95 bacterial colonies were isolated, of which 68 were identified to genus level and 22 were identified to species level. Some of the phytopathogens identified include: Agrococcus jenensis, Pseudomonas sp. and Curtobacterium sp. Some of the bacteria identified are beneficial like Ochrobactrum sp. Erwinia sp. and Pantoea sp. had a frequency of 28.95%, with Pantoea agglomerans having a frequency of 18.42%. Ochrobactrum sp. had a frequency of 10.53%. Enterococcus mundtii an frequency of 5.26%. Information about the species composition of bacteria on grain crops can be used to determine the spread of bacteria and their diagnosis and for bioinformatic analysis of genomes in search of species-specific genetic markers.

Biotechnology continues to drive innovation in the production of pharmaceuticals, biofuels, and other valuable compounds, leveraging the power of microbial systems for enhanced yield and sustainability. Genome-scale metabolic (GSM) modeling has become an essential approach in this field that enables a guide for target genetic modifications and optimization of metabolic pathways for various industrial applications. While single-species GSM models have traditionally been employed to optimize strains like Escherichia coli and Lactococcus lactis, the integration of these models into community-based approaches is gaining momentum. Constraint-based metabolic community (CBMc) models, which simulate the interactions between multiple microbial species, offer a more comprehensive understanding of metabolic networks and their potential for improvement of biotechnological processes on pilot- and industrial scales. However, there are few platforms that effectively support both single-species GSM and CBMc models, limiting the accessibility and application of these advanced modeling techniques. In this study, we demonstrate the use of the BioUML platform, one of the few tools that provides simultaneous investigation of both single-species and community models. We harnessed the platform to reconstruct and analyze microbial interactions within a synthetic consortium of widely used biotechnology microorganisms: methanotroph, Methylococcus capsulatus Bath and engineered Escherichia coli strains under oxygen- and nitrogen- limited conditions. Our research highlights the potential of E. coli to reduce the accumulation of inhibitory by-products like acetate, thereby enhancing the metabolic efficiency and growth of M. capsulatus. The study underscores the utility of CBMc models in optimization of microbial consortia for biotechnological applications, offering new strategies for the sustainable production of biofuels, single-cell proteins, and other high-value compounds.

Microbial populations involved in forming the distinctive precipitates of the S, Fe, Mn, and Ca cycles in the San Diego River watershed reflect an interplay between mineralogy of the rocks in the watershed, sparse rainfall, ground- and surface-water anoxia, and runoff of high sulfate, treated imported water. In the less developed headwaters, Temescal Creek tributary emerges from pyrite-bearing metamorphic rocks, and thus exhibits both an oxidized Fe and reduced S cycle. In the middle reaches, the river moves through developed land where treated, imported high sulfate Colorado River water enters from urban runoff. Mast Park surrounded by caliche-bearing sedimentary rocks is a site where marl is precipitating. Cobbles in riffles in the river are coated black with Mn oxide. When the river encounters deep-seated volcanic bedrock, it wells up to precipitate both Fe and Mn oxides at Old Mission Dam. Then, directly flowing through caliche-laced sedimentary rocks, Birchcreek tributary precipitates tufa. Further downstream, at a site that periodically receives full sunlight, a sulfuretum sets up during the summer when the river is deoxygenated. Such a rich geochemistry results in activity of iron and manganese oxidizing bacteria, sulfur oxidizers and reducers, and cyanobacteria precipitating calcareous marl and tufa.

Bacterial biofilms have become a significant and growing threat to human life, nature, and environmental health. The aim of this study is to isolate amylase-producing bacteria from the terrestrial environment (soil) for investigating their general inhibition of some pathogenic human bacterial biofilm. A total of 75 amylase producing isolates were obtained by serial dilution and streaking method. Amylotic activity of these isolates was screened by a starch agar plate method. Isolates were characterized by morphological and biochemical methods. The isolated Bacillus species were B. megaterium (26.7%), B. subtilis (16%), B. cereus (13.3%), B. thuringiesis (10.7%), B. lentus (10.7%), B. mycoides (5.3%), B. alvei (5.3%), B. polymyxa (4%), B. circulans (4%) and Micrococcus roseus (4%). The optimum conditions for amylase production were monitored. Antagonistic activity of these isolates and purified amylase against multidrug-resistant pathogenic human bacteria by agar disk diffusion method. The sensitivity level of some standard antibiotics served as control. Interestingly, it was found that all isolates exhibited great antagonistic activity against the target pathogens. The greatest activity recoded by B. alevi (48 mm) against Staphylococcus aureus (MRSA) and the lowest activity recorded by B. polymyxa (12 mm) against E. coli while low inhibition recorded for amylase. Antibiofilm efficacy of isolates supernatant and purified enzyme also monitored by spectrophotometric methods. The results revealed highly significant inhibition with percentages of 93.6 and 78.8% respectively. So, they represent a good tool for biofilm control in clinical and environmental applications.

Soil improvement methods can result in changes in the microbial community in blueberry soil. However, there have been few reports on the impacts of different soil improvement methods on the microbial function, particularly on endophytic microbe. In this study, we analyzed the response of microbial community composition, microbial function and nitrogen (N) cycle to different improvement methods using high throughput sequencing. We aimed to investigate the best soil improvement method from a microbial perspective. The results showed that the highest microbial diversity was observed in the T4 treatment (peat combined with mushroom bran), followed by the T2 treatment (peat combined with acidified rice husk) both in the rhizosphere and roots. The dominant phyla were Proteobacteria and Actinobacteria, which were most abundance in the T4 and T1 (peat combined with sulfur) treatments in the rhizosphere soil, respectively, but showed the opposite trend in the root endophytic bacterial community. Interestingly, Acidobacterium and Paludibaculum, belonging to the Acidobacteria phylum, were found to have the highest influence according to the correlation network analysis. And these bacteria were most abundant in the T2 treatment in the rhizosphere soil. The rhizosphere soil microbial communities were clustered into two categories: one for T1 and T2 treatments, and another for T3 (mushroom bran) and T4 treatments. Compared to the other treatments, the T1 treatment had the most significant impact on microbial functional pathways in the blueberry roots. T2 treatment promoted the growth of N fixation functional bacteria both in the rhizosphere soil and roots. At the module level, the T2 treatment increased the relative abundance of N fixation and decreased the relative abundance of assimilating nitrate reduction reaction (ANRA), dissimilating nitrate reduction reaction (DNRA), denitrification and completed nitrification in the blueberry rhizosphere soil. Additionally, the T2 treatment increased the abundance of root endophytic microbes involved in N fixation. Overall, our findings suggest that the addition of peat combined with acidified rice husk is the optimal soil improvement method for blueberry cultivation.

Diseases caused by contaminated water consumption and poor hygiene are among the leading cause of death in children, the elderly and people with compromised immune system. The present study aims to assess the drinking water quality of some selected drinking water sources in the chosen study area using water quality index (WQI). Samples of drinking waters were collected from four different sources—river, sachet (packaged), borehole and well in Jabi, Abuja, Nigeria for physicochemical and bacteriological analyses using standard methods. With the exception of pH, river water had the highest content of all the physicochemical parameters examined. Some of the physicochemical parameters such as temperature and pH determined in this study were mostly within the World Health Organization (WHO) standard. The bacteriological analyses revealed that the highest total viable counts of 1.54 (0.12) × 10² (cfu/mL) were obtained in the river sample while the least values of 0.03 (0.00) × 10² (cfu/mL) were obtained in the sachet water. Three bacterial isolates of Escherichia coli, Pseudomonas spp and Aeromonas spp were isolated from well water while Pseudomonas spp and Proteus spp were isolated from borehole water. All other bacteria were isolated from the river. The study demonstrates that the safest drinking water source in Idu district, Jabi, Abuja is the sachet/packaged water while the least safe is the river water due to the presence of significant opportunistic pathogens. The results of this study are beneficial for water quality management and could be used for low-cost effective water quality assessment in Jabi.

Endophytic bacteria, especially those participate in nitrogen fixation, play critical roles in supplying essential nutrients for legume plant growth. Compared to the numerous bacterial microbiome reports targeting legume roots and nodules, little is known about embryonic bacteria that facilitate plant nutrient utilization after seed germination. Here, we collected and investigated endophytic bacterial microbiomes in edible pea (Pisum sativum) embryos using five representative cultivars and a pea sprout control. Twenty-six nitrogen-fixing bacteria (NFBs) were isolated from pea embryos, with three strains found in fresh pea (FGP) and snow pea (SP) exhibiting the strongest nitrogenase activity. Some NFB isolates are also potassium-solubilizing bacteria (KSBs) or phosphorus-solubilizing bacteria (PSBs) utilizing inorganic and/or organic phosphorus. All 26 NFBs showed variable levels of indole-3-acetic acid (IAA) secretion. The nutrient-solubilizing NFBs identified in our research are valuable resources for biofertilizer development. At the microbiome level, high-throughput 16S ribosomal RNA (rRNA) sequencing of 40 bacterial collections from pea embryos revealed similar community structures and species abundances and diversities in most samples. Our embryo-derived endophytic bacterial pool provides a microbiome platform for seed dormancy and germination research of edible peas.

A sustainable solution for crack maintenance in geopolymers is necessary if they are to be the future of modern green construction. This study thus aimed to develop self-healing biogeopolymers that could potentially rival bioconcrete. First, a suitable healing agent was selected from Bacillus subtilis, B. sphaericus, and B. megaterium by directly adding their spores in the geopolymers and subsequently exposing them to a large amount of nutrients for 14 days. SEM-EDX analysis revealed the formation of biominerals for B. subtilis and B. sphaericus. Next, the effect of biochar-immobilization and co-culturing (B. sphaericus and B. thuringiensis) on the healing efficiencies of the geopolymers were tested and optimized by measuring their ultrasonic pulse velocities weekly over a 28-day healing period. The results show that using co-cultured bacteria significantly improved the observed efficiencies, while biochar-immobilization had a weak effect but yielded an optimum response between 0.3-0.4 g/mL. The maximum crack width sealed was 0.65 mm. Through SEM-EDX and FTIR analyses, the biominerals precipitated in the cracks were identified to be mainly CaCO3. Furthermore, image analysis of the XCT scans of some of the healed geopolymers confirmed that their pulse velocities were indeed improving due to the filling of their internal spaces with biominerals. With that, there is potential in developing self-healing biogeopolymers using biochar-immobilized spores of bacterial cultures.

Inflammation which has important functions in human defense systems and in maintaining the dynamic homeostasis of the body has become a major risk factor for the progression of many chronic diseases. Although the applied medical products alleviate the general status, they still exert adverse effects in the long term. For this reason, the solution should be sought in more harmless and affordable agents. Microorganisms offer a wide range of active substances with anti-inflammatory properties. They propose important advantages such as renewable and inexhaustible nature. This review aims at the most recent updates on the microorganisms of different type and genera being carriers of anti-inflammatory activity.

Present worldwide difficulties in healthcare and the environment have motivated the investigation and research of novel materials in an effort to find novel techniques to address the current chal-lenges and requirements. In particular, the use of nanomaterials has demonstrated a significant promise in the fight against bacterial infections and the problem of antibiotic resistance. Metal na-noparticles and carbon-based nanomaterials in particular have been highlighted for their excep-tional abilities to inhibit many types of bacteria and pathogens. In order for these materials to be as effective as possible, synthetic techniques are crucial. Therefore, in this review article, we high-light some recent developments in the design and synthesis of various nanomaterials, including metal nanoparticles (e.g., Ag, Zn, or Cu), metal hybrid nanomaterials, and the synthesis of mul-ti-metallic hybrid nanostructured materials. Following that, examples of these materials’ applica-tions in antimicrobial performance targeted at eradicating multi-drug resistant bacteria, material protection such as microbiologically influenced corrosion (MIC), or additives in construction ma-terials have been described.

Background: Bacterial conjunctivitis is one of the most commonly encountered eye complications in medicine, particularly in developing nations. In Hadhramout Governorate-Yemen, there is no data about bacterial conjunctivitis among patients. Aim: The present study aimed to determine the prevalence rate of bacterial conjunctivitis among patients seeking hospitals in Hadhramout Governorate, Yemen. Methods: A total of 1722 eye swabs were collected from patients attending different Hadhramout hospitals over a period of April to December 2018. The isolation and identification of bacteria are performed using standard bacteriological procedures. In addition, structured questionnaires were used to gather data from patients. Results: Out of 1722 eye swabs, 198 (11.49%) were positive for bacterial conjunctivitis, while 1524 (88.5%) were negative. The high prevalence rate of infection was among female participants and the age group between 31 and 40 years, at 15.25% and 29.29%, respectively. In addition, the prevalence of gram-positive bacteria was significantly greater among study patients than gram-negative bacteria (58.59% vs. 41.41%). The most predominant Gram-positive bacteria were Staphylococcus aureus (26.26%), followed by Streptococci pneumoniae (17.18%), Alpha-hemolytic Streptococci (8.08%), and Staphylococcus epidermidis (7.07%). Whereas the gram-negative bacteria were Pseudomonas aeruginosa (12.1%), followed by Moraxella catarrhalis (10.1%), Haemophilus influenza (7.07%), Klebsiella sp. (6.06%), Enterobacter agglomerans (4.04%), and Neisseria gonorrhoeae (2.02%). Additionally, the majority of study patients suffered from red eyes (100%), tearing eyes (93.93%), itching (90.9%), painful eyes (87.87%), purulent eyes (56.56%), and eye discharge (38.38%). Conclusion: The prevalence rate of bacterial conjunctivitis among patients in Hadhramout, Yemen, is relatively low compared with international investigations. Further investigation is required to assess and identify the antibiotic resistance to these agents that cause conjunctivitis.

Optical immunosensors are one of the most popular category of immunosensors with applications in many fields including diagnostics, environmental and food analysis. The latter field is of particular interest not only for the scientists but also for the regulatory authorities and the public since food is essential for life but can be also the source of many health problems. In this context, the current review aims to provide an overview of the different types of optical immunosensors focusing onto their application for the determination of pathogenic bacteria in food samples. In particular, after the description of main optical transduction techniques, their implementation for the immunochemical determination of bacteria will be discussed. Finally, a short commentary about the future trends in optical immunosensors for food safety applications will be provided.

Orchids are often a mystery because of their close and complex relationships with various microorganisms in the natural environment. Orchids rely on microorganisms to obtain nutrients, affecting their seed germination, protocorm, and adult plant growth. Currently, the majority of relevant research is concentrated on isolating and identifying environmental microorganisms that support orchid development and growth. With the development of metagenomic technology, our understanding of orchid mycorrhizal fungi (OMF) and root-associated bacteria (RAB) has been expanded. New research results and discoveries have emerged, which require a comprehensive assessment to provide a reference for studying microorganisms related to orchids. Therefore, we present a comprehensive summary, identifying significant inadequacies of present methodologies while providing ideas for further research.

Abstract Bacteria and fungi are both used in biological seed treatments. While all have potential uses, some organisms are more widely and successfully used than others. Shelf life is an important consideration. For this reason, organisms that lack cell walls are more difficult to use than ones with long-lasting spores. Bacillus and Trichoderma are both widely effective, have good shelf life, and are frequently used. However, Rhizobiacae lack cell walls, which is a limitation; they are widely used because their symbiosis with legumes facilitates nitrogen fixation which is an important factor that provides economic, agricultural and environmental sustainability. For all organisms, proper formulation is critical for success; this is especially true for Rhizobiacae and other gram-negative bacteria. There are several specialized processes to deliver microbial agents or to enhance their biological activity, such as solid matrix priming and hydroseeding. Biorational chemicals derived from microorganisms are also frequently used. Both living organisms and biorationals provide benefits to plant agriculture. They can control diseases and increase resistance to abiotic stresses such as drought, temperature, salt, and flooding. They also can enhance mineral nutrition and photosynthesis. For these applications, the most effective ones colonize roots internally and provide season-long benefits. These endophytes induce systemic changes in plants’ gene expression and encoding of proteins.

Ticks are key vectors of some important diseases of humans and animals. Although they are carriers of disease agents, the viability and development of ticks are not harmed by the infectious agents due to their innate immunity. Antimicrobial peptides directly protect hosts against pathogenic agents such as viruses, bacteria, and parasites. Among the identified and characterized antimicrobial peptides, defensins have been considerably well studied. Defensins, which contain intramolecular disulfide bridges between cysteine residues, are commonly found among fungi, plants, invertebrates, and vertebrates. The sequence of the tick hemolymph defensin (HEdefensin) gene from the hard tick Haemaphysalis longicornis was analyzed after identification and cloning from a cDNA library. HEdefensin has a predicted molecular mass of 8.15 kDa and a theoretical isoelectric point of 9.48. Six cysteine residues were also identified in the amino acids. The synthetic HEdefensin peptide only showed antibacterial activity against Gram-positive bacteria such as Micrococcus luteus. A fluorescence propidium iodide exclusion assay also showed that HEdefensin increased the membrane permeability of M. luteus. Additionally, an indirect fluorescent antibody test showed that HEdefensin binds to M. luteus. These results suggested that HEdefensin strongly affects the innate immunity of ticks against Gram-positive bacteria.

Salinity is one of the major factors responsible for the deterioration of soils, making them unsuitable for agriculture. The application of halotolerant and halophilic plant growth promoting bacteria (PGPR: Plant Growth Promoting Rhizobacteria) with biological control activities as an inoculant of cultivated plants offers a biological alternative to the use of agrochemicals, particularly when subjected to salt stress. In this perspective, 70 bacterial strains were isolated from saline soils (sebkha) in arid and semi-arid areas of eastern Algeria. Three isolates were selected based on their ability to produce bioactive molecules allowing them to promote plant growth, such as hydrolytic enzymes, indole acetic acid, HCN, NH3, etc. Two of these isolates belong to the genus Serratia and the third is a halophilic Halomonas bacteria. These bacteria were identified based on the 16S rDNA sequence. Antagonism tests against phytopathogenic fungi were carried out. The identification of the antifungal molecules produced by these bacteria was done by high performance liquid chromatography. These bacteria are capable of inhibiting mycelial development against phytopathogenic fungi with rates reaching 80.67% against Botrytis cinerea, 76.22% against Aspergillus niger and 66.67% against Fusarium culmorum for Serratia sp. The strain Halomonas sp. inhibited mycelial growth by production of volatile substances of Aspergillus niger at 71.29%, Aspergillus flavus at 75.49% and Penicillium glabrum at a rate of 72.22%. The identification by HPLC of the antifungal molecules produced by these three bacteria reveals that they are polyphenols, which makes these strains the first rhizobacteria capable of producing phenolic compounds. Finally, pot tests to determine the effectiveness of these strains in promoting wheat growth under salinity stress (125 mM, 150 mM and 200 mM) were carried out. The results revealed that a consortium of two isolates (Serratia sp. and Halomonas sp.) performed best at 125 mM. But, at higher concentrations it was the halophilic bacteria Halomonas sp which gave the best result. In all cases, there was a significant improvement in the growth of wheat seedlings inoculated with the bacteria, compared to non-inoculated controls.

Background: The incidence of metabolic dysfunction-associated steatohepatitis (MASH)-related hepatocellular carcinoma (HCC) is increasing annually with a rise in the incidence of obesity and metabolic syndrome. Based on preliminary reports regarding the potential association of HCC and periodontitis, this study aimed to analyze the involvement of periodontal bacteria as well as the oral and intestinal bacterial flora in MASH-related HCC (MASH-HCC). Methods: Forty-one patients with MASH and 19 with MASH-HCC who completed survey questionnaires, underwent periodontal examinations, and subjected to sample collection (saliva, mouth-rinsed water, feces, and peripheral blood) participated in the study. Bayesian network analysis was used to analyze the causation between various factors, including MASH-HCC, examinations, and bacteria. Results: The occupancy rate of the genus Fusobacterium in the intestinal bacterial flora was significantly higher in in the MASH-HCC group than in the MASH group (p = 0.002), whereas that of Butyricicoccus (p = 0.022) and Roseburia (p < 0.05) was significantly lower. Bayesian network analysis revealed the absence of periodontal pathogenic bacteria and enteric bacteria affecting HCC. However, HCC directly affected the periodontal bacterial species Porphyromonas gingivalis, Tannerella forsythia, Fusobacterium nucleatum, and Prevotella intermedia in the saliva, as well as the genera Lactobacillus, Roseburia, Fusobacterium, Prevotella, Clostridium, Ruminococcus, Trabulsiella, and SMB53 in the intestine. Furthermore, P. gingivalis in the oral cavity directly affected the genera Lactobacillus and Streptococcus in the intestine. Conclusion: MASH-HCC directly affects periodontal pathogenic and intestinal bacteria, and P. gingivalis may affect the intestinal bacteria associated with gastrointestinal cancer.

Fine aroma cocoa (Theobroma cacao) is one of Ecuador's most iconic export products, representing 63% of world production; however, few advances have been made in fermentation processes that greatly influence the development of chocolate´s organoleptic characteristics. The study of starter cultures has been investigated in other countries, which seek to improve organoleptic properties or decrease fermentation time. The aim of this study was to analyze the effect of a native microbial consortium based on two yeasts (Torulaspora delbrueckii and Hanseniaspora uvarum), a lactic acid bacterium (LAB) (Lactobacillus plantarum) and an acetic acid bacterium (AAB) (Acetobacter ghanensis) inoculated at the beginning of the fermentative process, on physical and biochemical variables, microbial population dynamics, and percentage of almond fermentation. The started culture caused changes in sugar and acid content, which in turn generated temperature and pH changes in the dough, resulting in the dynamics of yeast, AAB, and mesophilic microorganisms remaining higher than those of the control throughout the fermentation process; a decrease of filamentous fungi that affect the flavor and quality of beans due to the production of acetic acid or secondary metabolites from yeasts and LAB; and optimized fermentation percentage by 24% higher than spontaneous fermentation in only 96 hours.

The development of a novel and simple inhibition biosensor array for detection of water pollutants based on immobilized bacteria is the main goal of this work. A series of electrochemical measurements (i.e. cyclic voltammograms) were carried out on screen-printed gold electrodes with three types of bacteria, namely Escherichia coli, Shewanella oneidensis, and Methylococcus capsulatus, immobilized via poly L-lysine. For comparison purposes, similar measurements were carried out on bacteria samples in solutions,; also optical measurements (fluorescence microscopy, optical density, and flow cytometry) were performed on the same bacteria in both liquid and immobilized forms. The study of the effect of heavy metal ions (lead), pesticides (atrazine) and petrochemicals (hexane) on DC electrochemical characteristics of immobilized bacteria revealed a possibility of pattern recognition of the above inhibition agents in aquatic environment.

The use of biological methods to control plant disease is an option to reduce the pesticide residues in an environment. The objective of this research is to develop the bio-products for the elimination and/or replacement of pesticides. All the 182 strains of beneficial bacteria were screened from 20 samples of chili. The samples were divided into 8 groups. The strain SP-TU6, SP-TU7, and SP-TU3 were high efficacy inhibited mycelium growth of Colletotrichum capsici P-TU008 with 98.93, 94.62, and 95.69%, respectively as chemical control (carbendazim). The development of 3 bio-products that can promote the growth of economic crops are including Bioproduct 1 = talcum: CaCO3: CMC (9.75:1.5:1), Bioproduct 2 = corn flour: talcum: CaCO3: CMC (10:87.5:1.5:1), and Bioproduct 3 = corn flour: talcum: CaCO3: CMC (20:87.5:1.5:1). All bioproducts showed the best level of solubility in water, distribution, and sedimentation. Found that, the bioproduct 1 was suitable for survival of beneficial bacteria SP-TU3 and SP-TU6 with a population of 4.5x1012 and 3.2x1013 cfu/g respectively, and bioproduct 2 was suitable for strain SP-TU7 (2.4x1012 cfu/g) after storage for 3 months. The strain SP-TU3 in bioproduct 1 can induce seed germination and growth parameter of chili (Capsicum spp.) were 95% and 1.86, respectively, and SP-TU7 in in bioproducts 2 had the highest efficiency to induce seed germination and growth parameter of ruzi grass (Brachiaria ruziziensis) with 62% and 6.51 respectively, and strain SP-TU6 was the most effective germination and growth parameter of B. chinesis and B. oleracea with 98%, 88%, and 3.62, 88.00, respectively.

Due to bacterial variety, complicated relationships, and research technique constraints, investigating the microbiota is difficult. AI and machine learning have expanded microbiome research. Artificial intelligence (AI) systems can efficiently scan large microbiome datasets to help researchers understand microbial populations and their functions. AI-based predictive algorithms can analyze food impacts on microbial communities and forecast illness risks based on gut microbiota composition. AI helps find microbial biomarkers linked to specific health disorders, enabling early illness identification and targeted therapy. AI-driven medication development platforms also modulate microbiome to treat microbiota-related diseases. Understanding microbial populations' involvement in health and illness requires understanding complicated microbial interactions within the microbiome and between it and the host. AI systems improve human health by comprehending these complex relationships. AI in microbiota study must overcome various obstacles. Data quality and AI model interpretability are essential for accurate findings. Diverse and representative datasets avoid biases and strengthen AI-driven conclusions. AI in microbiota research requires data protection, informed permission, and appropriate use of sensitive data. AI is improving our understanding of the human microbiome and its health effects, changing microbiota research. Overcoming difficulties and adhering to ethical norms will enable the proper use of AI-driven findings, leading to microbiome insights and precision medical breakthroughs.

The trillions of bacteria that live in and on our bodies make up the human microbiota, which is a very important part of keeping us healthy and protecting us from pathogenic infections. Dysbiosis is when there are changes in the composition and variety of the microbiota. This can upset the delicate balance between the host and its microbes, making the host more vulnerable to pathogens. Understanding the complicated link between microbiota and pathogens is important for coming up with effective ways to change microbiota and boost the body's defenses against infections. This study looks at how microbiota modulation affects the spread of pathogens and infections. It talks about how the makeup of microbiota affects how susceptible you are to pathogens. It shows how a healthy microbiota can protect you by making it hard for pathogens to grow and take over. The review also goes into detail about how the microbiota protects against pathogens, such as through competitive exclusion, the production of antimicrobial compounds, and the regulation of immune reactions. The study also looks at the idea of dysbiosis and how it is linked to pathogenic shifts. Disruptions in the microbiota can make it easier for pathogens to grow and make it harder for the host to fight off infections. It focuses on the need to recover microbial balance to improve the host's defenses and lower risks caused by pathogens. There is also talk about the use of probiotics, prebiotics, and fecal microbiota transplantation (FMT) as ways to change the microbiome. In conclusion, knowing how the microbiota and pathogens work together in a complicated way can help a lot when it comes to making specific interventions for microbiota modulation. Strategies that restore the balance of microbes and boost the host's defenses could stop pathogens from taking over and making people sick. More study needs to be done in this area to improve therapeutic approaches and use the microbiota to its fullest extent to fight infectious diseases.

Eelgrass meadows have attracted a lot of attention not only for their ability to maintain marine ecosystems as feeding grounds for marine organisms, but also for their potential to store atmos-pheric and dissolved CO2 as blue carbon. This study comprehensively evaluated the bacterial and chemical composition data obtained from eelgrass sediments of different scales along the coast of Japan to investigate the sediment effect on the acclimatization of eelgrass. Regardless eelgrass habitat, approximately 1% Anaerolineales, Babeliales, Cytophagales, and Phycisphaerales were present in the bottom sediment. Sulfate-reducing bacteria (SRB) were present at 3.69 % in eelgrass sediment compared to 1.70 % in bare sediment. Sulfur-oxidizing bacteria (SOB) were present at 2.81% and 1.10% in the eelgrass and bare sediment, respectively. Bacterial composition analysis and linear discriminant analysis revealed that sulfur-oxidizing bacteria detoxified H2S in the eelgrass meadows and that the larger-scale eelgrass meadows had a higher diversity of sul-fur-oxidizing bacteria. Our result indicated that one of the following systems detoxifies the H2S in eelgrass meadows microbial oxidation by sulfur-oxidizing bacteria, oxidation by O2 perme-ated by the physical diffusion of benthos. This study indicated the issues of maintaining sustain-able conservation of the eelgrass bed.

Methicillin-resistant Staphylococcus aureus (MRSA) infections, in particular biofilm-organized bacteria, remain a clinical challenge and a serious health problem. Rifabutin (RFB), an antibiotic of the rifamycins class, has shown in previous work excellent anti-staphylococcal activity. Here, we proposed to load RFB in liposomes aiming to promote the accumulation of RFB at infected sites and consequently enhancing the therapeutic potency. Two clinical isolates of MRSA, MRSA-C1 and MRSA-C2, were used to test the developed formulations, as well as the positive control, vancomycin (VCM). RFB in free and liposomal forms displayed high antibacterial activity, with minimal inhibitory concentrations (MIC) ranging from 0.009-0.013 μg/mL and MBIC50 values ranging from 0.008-0.012 μg/mL, in planktonic and biofilm bacteria, respectively. Confocal microscopy studies demonstrated the rapid internalization of unloaded and RFB-loaded liposomes in the bacterial biofilm matrix. In murine models of systemic MRSA-C1 infection, Balb/c mice were treated with RFB formulations and VCM at 20 and 40 mg/kg of body weight, respectively. Obtained data proved the superior therapeutic effectiveness of RFB formulations compared to Control and VCM groups even using a two-fold lower therapeutic dose as demonstrated by significantly reduced bacterial burden and growth index values in major organs. In addition, RFB formulations were the only formulations associated with 100% survival. Globally, this study emphasizes the potential of RFB nanoformulations as an effective and safe approach against MRSA infections.

Salinity is a major abiotic stress affecting agricultural productivity. This research investigated the impact of mycorrhizal fungi (AMF), bacteria (PGPR), and their combination on pepper plants under salt stress (200 mM NaCl). Conducted in a controlled climate chamber, the study assessed plant height, stem diameter, leaf number, leaf area, leaf weight, shoot fresh and dry weights, SPAD values, leaf relative water content (RWC), leaf water potential, and mineral concentrations (Na, K, Mg, Ca, P, Cu, Mn, Fe, Zn). AMF application significantly increased plant height (39.50 cm) compared to PGPR (37.75 cm) and the control (37.25 cm). While salt stress reduced plant height by 34.90%, AMF and PGPR mitigated these effects, with AMF improving height by 21.65% under saline conditions. AMF and PGPR positively influenced plant diameter, leaf number, and fresh weight. Physiological measurements, including SPAD values, RWC, and osmotic potential, showed that AMF+PGPR applications were most effective in increasing RWC. Nutrient analysis revealed reduced calcium and magnesium levels due to salt stress. In conclusion, AMF and PGPR enhanced pepper plants' growth and physiological responses under salt stress, indicating their potential to improve agricultural practices in saline environments.

Biogas is mainly produced by anaerobic digestion (AD) using organic matter as a substrate. In Mediterranean countries, a promising substrate is lignocellulose biomass of perennial grass Miscanthus x giganteus, due to its potentially high biogas yields, which could be comparable to maize silage. During AD, bacteria convert biomass into more minor compounds, which are further converted to methane by methanogenic archaea. The selection of appropriate microbes for the degradation of the substrate is crucial, and the enhancement of this step lies in the immobilization of microbes on biocarriers. Here, a microbial consortium, de-novo isolated and conditioned to degrade the Mischantus biomass, was immobilized onto several natural biocarriers: natural zeolitized tuff (NZ), ZeoSand®, perlite, and corncob. There was no statistically signifi-cant difference in the numbers of immobilized bacteria between the materials. Therefore, all proved to be suitable for the immobilization of the consortium. In the consortium, five bacterial species with different shares in the consortium were identified: Enterobacter cloacae, Klebsiella pneumoniae, Enterobacter asburiae, Leclercia adecarboxylata, and Exiguobacterium indicum. After im-mobilization on each carrier, the share of each species changed when compared to starting conditions, and the most dominant species was E. cloacae (71-90%), while the share for other species ranged from 2-23%. The share of E. indicum was around 14 % at the start. However, it diminished to less than 1% because it was overgrown during the competition with other bacterial species, not due to an inability to immobilize. For future research, single species could be separately immobilized onto carriers, depending on their degradation capacity, thus creating the targeted and optimized biocarriers.

In marine ecosystems communication among microorganisms is crucial since the distance is significant if considered on a microbial scale. One of the ways used by microorganism cells to reduce this gap is the production of extracellular vesicles. Marine bacteria release extracellular vesicles (EVs), small membrane-bound structures of 50 to 250 nm diameter, into their surrounding environment. The vesicles contain various cellular compounds, including lipids, proteins, nucleic acids, and glycans. EVs may mediate the communication between microorganisms in aquatic environments thus influencing ecosystem function and determining the structure and composition of microbial populations. This review will focus on marine bacterial EVs analysing their structure, composition, functions, and applications.

Medicinal activities carried out within hospitals lead to a high consumption of antibiotics and subsequently result in a high excretion rate of antibiotic residues. These antibiotics consumed in human medicine are not fully absorbed by the body and are often discharged into the environment as biological waste from infected human patients. The significant water consumption in hospitals and the pharmaceutical impact on wastewater from healthcare facilities facilitate the emergence and spread of antibiotic-resistant bacteria and antibiotic-resistant genes in the environment. Hospital effluents could be doubly involved in the dissemination of antibiotic molecules and multidrug-resistant bacteria in various ecosystems. The objective of this study is to characterize hospital effluents by assessing antibiotic concentrations and the diversity of antibiotic-resistant bacteria in these hospital wastewaters and within the environment (aquatic environments; rivers), as well as to inventory the bacteria present and those carrying antibiotic resistance in both hospital wastewater and the environment.

Abstract: Background: The birthrate of Black preterm (BPT) infant is 65% higher than White preterm (WPT) infants with BPT mortality 2.3 times higher. The incidence of culture positive sepsis is as high as 41% in preterms. This study aimed to examine infections and mortality in BPTs and WPTs. Methods: We enrolled 362 preterms born < 32 weeks gestational age (GA) between 2019-2023 in 5 neonatal intensive care unit (NICU)s to study onset of infections, and dispositions; RedCap data were analyzed for descriptive statistics. Results: 226 BPTs (63.5%) and 107 WPTs (30.8%) enrolled, with 29 other than White or Black, 50.5% female, mean GA of 27.67 weeks and 986.74 grams birthweight. Of 426 suspected infections evaluated in enrolled infants, there were 7 infants with early onset sepsis (EOS) and 39 infants with late onset sepsis (LOS). The incidence of EOS was 1.9% overall, 2.2% in BPTs vs 0.9% in WPTs. LOS incidence was 10.8% overall, 11.9% in BPTs and 9.3% in WPTs. Both BPT and WPT infants had ~ 8% gram-positive LOS events and 3% gram-negative LOS events; however, 82% of all gram-negative LOS episodes occurred in BPTs. There were 27 urinary tract infections and 78% were in BPTs. Two cerebral spinal fluid cultures were positive in BPTs. Necrotizing enterocolitis was diagnosed in 5.8% of the sample, with an incidence of 6.1% in BPTs versus 4.7% in WPTs. Death occurred in 6.0% of the enrolled infants with the rate of death being slightly higher in BPTs (7.0%) versus WPTs (5.6%). Conclusions: BPTs had a higher rate of positive cultures, double the gram-negative infections, a much higher rate of urinary tract infec-tions and a higher rate of mortality than their WPT counterparts. This study emphasizes higher risk of morbidity and mortality for BPTs.

Periodic assessment of bacterial contamination is necessary as it allows proper guidance in cases of eye infections through the use of appropriate antibiotics. Due to the extensive use of antibiotic treatment, most of the contamination flora is resistant to the usual ophthalmic antibiotics. The present study shows an updated picture of the susceptibility of Gram-positive and Gram-negative bacteria found on the ocular surface to the most commonly used antibiotic agents in patients undergoing cataract surgery. A total of 993 patients were included in the study with ages between 44 and 98 years old. Conjunctival culture was collected 7 days before cataract surgery. The response of Gram-positive and Gram-negative bacteria to various antibiotic classes like glycopeptides, cephalosporins, carbapenems, fluoroquinolones, aminoglycosides, phenicols, tetracyclines, rifamycins, macrolides and penicillins was assessed. From the tested antibiotics, vancomycin had 97.8% efficacy on Gram-positive bacteria. In the cephalosporin category, we observed an increased resistance of the cefuroxime for both Gram-positive and negative bacteria. Antibiotics that have more than 90% efficacy on Gram-positive bacteria are: meropenem, imipenem, netilmicin, amikacin and rifampicin. On Gram-negative bacteria, we found 100% efficacy of all tested fluoroquinolones, aminoglycosides (except for tobramycin), doxycycline, azithromycin, clarithromycin and chloramphenicol. The current study illustrates patterns of increased resistance of certain bacteria present on ocular surface to some of the commonly used antibiotics in ophthalmological clinical practice. One such revealing example is cefuroxime, which has been highly used as an intracameral antibiotic for the prevention of bacterial endophthalmitis after cataract surgery.

Introduction: Silver nanoparticles have been extensively investigated in dental and orthopedic materials. However, the impregnation of bone graft with silver nanoparticles has been poorly investigated. Objectives: The aim of this study was to evaluate the in vitro antibiofilm activity of a bovine bone impregnated with silver nanoparticles. Methods: Bone scaffolds from cancellous bovine femur were used for the tests and impregnated with silver nanoparticles (50nm) by physical adsorption. Silver nitrate minimal inhibitory and bactericidal concentration were performed Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans, Enterococcus faecalis, Acinetobacter baumannii, and Escherichia coli. Disc diffusion tests for silver nanoparticles susceptibility and quantification of biofilm production on plate and bone with sessile cell count were also performed. Results: All pathogens were susceptible to silver with low minimal inhibitory concentration (0.25 – 4 mg/L). The scaffold impregnated with silver nanoparticles presented a significant reduction in the biofilm cells for all microorganisms with a reduction of more than 3 logs in colony forming units count. Conclusion: Bone scaffolds impregnated with silver nanoparticles can significantly reduce biofilm, and it can be a strategical material to be used as an implant for different approaches.

Fungal infections pose a significant threat to human health, particularly in immunocompromised individuals. Due to several factors, such as the inappropriate use of antimicrobial drugs, the emergence of resistance in microorganisms, and increasing demand for less cytotoxic antifungal agents for immunocompromised patients, antifungal research has gained significant attention in recent years. Antifungal peptides, and among them, cyclic peptides, are a class of antifungal agents that have been under development since the 1960s. Using cyclic peptides as a promising approach for combating antifungal resistance in pathogenic fungi has gained significant attention in recent years. The identification of antifungal cyclic peptides from various sources has been possible due to the widespread interest in peptide research in recent decades. It is increasingly important to evaluate the narrow to broad-spectrum antifungal activity and the mode of action of synthetic and natural cyclic peptides for both synthesized and extracted peptides. This short review aims to highlight some of the antifungal cyclic peptides isolated from bacteria, fungi, and plants, taking into consideration the progress made in peptide research. Additionally, it provides a summary of synthetic cyclic peptides with antifungal activity and discusses the potential future of utilizing combinations of antifungal peptides from different sources. The review underscores the need for further exploration of the novel therapeutic applications of these abundant and diverse cyclic peptides.

Dissolved organic matter (DOM) in the ocean represents about 662 billion tonnes of C, 200 times more than the living biomass. It is produced mainly by microbial primary production. The largest fraction of this DOM is old (>weeks to months) and both chemically and biologically recalcitrant. The remainder is young (seconds to weeks), more labile and surface active. It also changes the rheological properties in the bulk phase of the water and at interfaces including the sea surface microlayer (SML). In order of abundance, this DOM consists of sugars, amino acids, fatty acids and nucleic acids, often incorporated into complex polymers. The DOM molecules are produced by microbial genes, and are further modified by enzymes themselves produced by genes. The properties of ocean water and its interfaces as well as biogeochemical fluxes may thus be modified by ocean microbial genes. These fluxes influence ocean and atmospheric climate, which in return acts on the biota. Therefore the ocean microbial genomes and the fluxes and climates they influence may be subject to Darwinian-type selection. Research programmes need to integrate ocean ecology, rheology, biogeochemistry and genomics, to find the associations among them. Discovery of commercial bioactive molecules may be a bonus.

Antibiotic resistance is a major public health issue that requires a multifaceted approach. One potential source of antibiotic-resistant bacteria is wastewater in developing countries, which often lacks proper treatment infrastructure and can release high levels of antibiotics and antibiotic-resistant bacteria into the environment. This review article summarizes current knowledge on strategies to combat antibiotic resistance in wastewater in developing countries. Our review highlights the importance of improving wastewater treatment infrastructure to effectively remove antibiotics and antibiotic-resistant bacteria, implementing measures to reduce the release of antibiotics into the environment, and monitoring and surveillance to track the presence and spread of antibiotic-resistant bacteria in wastewater. We also discuss the potential challenges and barriers to implementing these strategies and the need for further research to determine their effectiveness in real-world settings. Overall, this review highlights the need for a comprehensive approach to address antibiotic resistance in wastewater in developing countries and underscores the importance of addressing this issue to protect public health.

Polyurethane (PU) is a polymer widely used by humans whose recycling is highly complex due his chemical structure, being limited to incineration or accumulation in landfills. Biodegradation by enzymes and microorganisms has been studied for decades as an effective method of biological decomposition. In this study, Tenebrio molitor larvae (T. molitor) were fed with polyurethane foams, which gut enzymes and microorganisms were capable of degrading the polymer by 35% in 17 days of treatment, producing a weight loss of 14% in the mealworm. Changes in T. molitor gut bacterial community and diversity were observed, which may be due to colonization of species associated with PU degradation. Physical and structural biodegradation in PU by T. molitor compared to virgin PU, was demonstrated by Fourier Transform InfraRed spectroscopy (FTIR), Thermal Gravimetric Analysis (TGA) and Scanning Electron Microphotography (SEM).

Composting is one of the most economical and environmentally safe methods of recycling organic waste. Soil microorganisms play a significant role in decomposition and the availability of plant nutrients. This study was designed to prepare a suitable microbial inoculum and its evaluation on composting heap to decrease the time of waste degradation. The bacteria were isolated and molecular characterized from the soil near composting area by using 16S ribotyping technique. The identified strains of bacillus cereus used as an inoculum gives better results to expedite the degradation of organic waste. The prepared bacterial inoculum with molasses was also compared with commercial inoculum by optimizing physical and chemical parameters (temperature, oxygen, C: N, pH, and moisture content) of composting heap. Monthly reading of these parameters was taken from experimental and control treatments. The highest decomposition rate of organic waste was recorded in treatment A (experimental heap) where molasses and bacterial inoculum were added and less decomposition was observed in treatment D (experimental heap) where no inoculum and molasses were added. It was concluded from the studies that the prepared bacterial inoculum with two strains of Bacillus was effective and prepared mature compost in 2.5 months by increasing decomposition efficiency of organic waste. Furthermore, the prepared compost was also sustainable in its physical and chemical characteristic.

The relevance of bacteria to subjective experiences or qualia is under-appreciated. Here, I make four proposals. Firstly, living systems traverse sequences of active states that determine their behaviour; these states result from competitive coherence, which depends on a connectivity-based competition between a Next process and a Now process whereby elements in the active state at time n+1 are chosen between the elements in the active state at time n and those elements in the developing n+1 state. Secondly, bacteria should help us link the mental to the physical world given that bacteria were here first, are highly complex, influence animal behaviour and dominate the Earth. Thirdly, the operation of competitive coherence to generate active states in bacteria, brains and other living systems is inseparable from qualia. Fourthly, these qualia become particularly important to the generation of active states in the highest levels of living systems, namely, the ecosystem and planetary levels.

Bacterial endophytes interact closely with plant tissues and constitute an essential part of the plant microbiome. These interactions can promote plant growth and elicit specific defense responses against abiotic stresses and pathogen attacks. In this paper, we review the role of endophytic bacteria in modulating defenses of the host rendering the entire plant more resistant to pathogens and pests. The endophyte-induced resistance will probably introduce a new factor when consid-ering plant-pathogen interactions. The impact of the bacterial endosymbionts on the host leading to the priming state is also discussed since it confers a specific adaptation of the plant to the biotic threat.

Here, we report a simple and effective method for separation of gram-negative bacteria using aptamer-modified microbeads and acoustophoresis. As acoustophoresis allows for simultaneous washing and size-dependent separation in continuous flow mode, we efficiently obtained gram-negative bacteria that showed high affinity without any additional washing steps. The proposed device has a simple and efficient channel design, utilizing a long, square-shaped microchannel that shows excellent separation performance in terms of the purity, recovery, and concentration factor. Microbeads (10 µm) coated with the GN6 aptamer can specifically bind gram-negative bacteria. Using acoustophoresis, gram-negative bacteria-bound microbeads and other unbound/contaminants can be separated by size with high purity and recovery. The device demonstrated excellent separation performance, with high recovery (up to 98%), high purity (up to 99%), and a high volume rate (500 µL/min), and a concentration factor of up to 20×. The acoustophoresis microfluidic device also showed binding affinity to multiple strains of gram-negative bacteria, but not to gram-positive bacteria. This study presents a new paradigm for early diagnosis of bacterial infectious diseases. In addition to detecting living bacteria or bacteria-derived biomarkers, this protocol can be extended to monitoring the contamination of water resources, and may aid quick responses to bioterrorism and pathogenic bacterial infections.

Plant growth promoting rhizobacteria (PGPR) are capable to reduce the use of chemical fertilizers input cost of farmer. Keeping in view the study was designed to investigate and evaluate inoculation effect of indigenous rhizospheric bacteria on growth and yield of wheat (Triticum aestivum L.) under in vitro and in vivo conditions using different treatments. Ten potential strains were selected on the basis of their ACC deaminase activity, siderophore production, P-solubilization and production of indole acetic acid (IAA). Further these strains were tested in three different experiments (growth chamber, pot and field). We found significant increase in crop growth response to the inoculants in comparison with un-inoculated control. In pot and field trial we tested PGPR with recommended dose of inorganic fertilizers. The results of present study revealed that inoculation of bacterial strains with wheat seeds significantly increased plant growth and improved crop yield. Results of present study reveal that these strains could be employed in different combinations and can get higher yield in case of half recommended doses of inorganic fertilizers along with consortium of strains in comparison with sole application of recommended dose of fertilizer and with consortium of strains. These strains were further identified by 16Sr RNA gene sequencing, fatty acid profile and biolog. It can be concluded that inoculated bacteria have more potential and contributes in good crop quality, increased yield when they are applied in combination, thus have potential to minimize use of chemical fertilizers.

Extremophilic bacteria are important groups of extremophilic organisms that have been studied during the last years. They are considered as a source of enzymes due to great diversity and can survive under extreme conditions. Many enzymes produced by these microorganisms are of great importance and have found applications in several industries. Due to their activity and stability under extreme conditions, these enzymes offer new alternatives for current biotechnological and industrial applications. They have a wide range of potential uses and have been a nuclear subject of many different investigations. To date, some of the enzymes produced by extremophilic bacteria are currently being assessed thier industrials applications. Despite, benefits that present these enzymes, their potentials remain largely unexplored. These enzymes pose new opportunities for new line of research, and biotechnological applications. This review provides a summary on diversity and biotechnological and industrial applications of some enzymes produced by extremophilic bacteria.

The main purpose of bovine colostrum, being the milk secreted by a cow after giving birth, is to transfer passive immunity to the calf. The calves have an immature immune system as they lack immunoglobulins (Igs). Subsequently, the supply of good quality bovine colostrum is required. The quality of colostrum is classified by low bacterial counts and adequate Ig concentrations. Bacterial contamination can contain a variety of human pathogens or high counts of spoilage bacteria, which becomes more challenging with emerging use of bovine colostrum as food and food supplements. There is also a growing risk for the spread of zoonotic diseases originating from bovines. For this reason, processing based on heat treatment or other feasible techniques are required. This review provides an overview of literature on the microbial quality of bovine colostrum and processing methods to improve its microbial quality and keep its nutritional values as food. The highlights of this review are: high quality colostrum is a valuable raw material in food products and supplements, the microbial safety of bovine colostrum is increased using appropriate processing-suitable effective heat treatment, which does not destroy the high nutrition value of colostrum, the heat treatment processes are cost-effective compared to other methods, and heat treatment can be performed in both small- and large-scale production.

To evaluate the behaviour of the relevant microbial populations during stretched cheese production, the quantitative microbiological analysis was performed during the critical steps of the preparation. The obtained data distributions proved statistically significant increases in all indicators, on average by 4.55 ± 0.64 log CFU/g of presumptive lactococci counts, 4.06 ±0.61 of lactobacilli, 1.53 ± 0.57 log CFU/g of coliforms, 2.42 ± 0.67 log CFU/g of Escherichia coli, 1.53 ± 0.75 log CFU/g of yeasts and moulds, and 0.99 ± 0.27 log CFU/g of presumptive Staphylococcus aureus, from the early stage of milk coagulation until curd ripening (0–24 h). The following steaming/stretching process caused reductions in viable counts with the most significant inactivation effect on coliform bacteria, including E. coli (-4.0 ± 1.0 log CFU/g). Total viable counts and yeasts and moulds showed 2 and almost 3 log reduction (-2.2 ± 1.1 log CFU/g and -2.6 ± 0.9 log CFU/g), respectively. The lowest decreases in presumptive S. aureus counts were estimated at the level of -1.50 ± 0.64 log CFU/g. The counts of yeasts and moulds showed the best indicatory function during the entire storage period of vacuum-packaged cheeses at 6 °C.

Introduction. Bacterial resistance to antibiotics is estimated to cause the major number of deaths by 2050 if we do not find strategies to slow down the rise of drug resistance [1]. Reviews on Mediterranean Wild Edible Plants (MWEPs) with antimicrobial properties are scarce in the mean databases (Pubmed, Scopus and WoS). Hence, we proceeded to a new review of the studies on MWEPs. Methods. We used Wild Edible Plant and Antimicrobial as keywords. We only included the Mediterranean plants, and studies in non-Mediterranean countries, but for plants growing in Mediterranean basin. Exclusion criteria were the document type, studies not concerning plant, plants not edible, not antimicrobial properties, or totally out of topic. Results. Finally, the number of studies reviewed, starting from one hundred and ninety-two, was thirty-eight (19,8%), concerning the antimicrobial properties of seventy-four MWEPs species, be-longing to twenty-five Families. Fifty-seven (77%) out of seventy-four species, proved to be antimicrobial with a stringent threshold selection. Conclusions. The studies are still very heterogeneous. We still know too little about MWEPs properties, but what we already know seriously recommends continuing.

In the last decades, intensive crop management has involved excessive use of pesticides or fertilizers, compromising environmental integrity and public health. Accordingly, there has been worldwide pressure to find an eco-friendly and safe strategy to ensure agricultural productivity. Recently, Plant Growth-Promoting (PGP) rhizobacteria are receiving increasing attention as suitable biocontrol agents against agricultural pests. In the present study, 22 spore-forming bacteria were selected among a salt-pan rhizobacteria collection for their PGP traits and their antagonistic activity against the plant pathogen fungus Macrophomina phaseolina. Based on the higher antifungal activity, strain RHFS10, identified as Bacillus vallismortis, was furtherly examined and cell-free supernatants assays, column purification, and tandem mass spectrometry employed to purify and preliminarily identify the antifungal metabolites. Interestingly, the minimum inhibitory concentration assessed for the fractions active against M. phaseolina, resulted 10 times lower and more stable than the one estimated for the commercial fungicide pentachloronitrobenzene. These results suggest the use of B. vallismortis strain RHFS10 as a potential Plant Growth Promoting Rhizobacteria to efficiently control phytopathogenic fungus M. phaseolina, in alternative to chemical pesticides.

We wanted to investigate the current knowledge on the impact of diet on anti-TNF response in inflammatory bowel diseases (IBD), to identify dietary factors that warrant further investigations in relation to anti-TNF treatment response, and, finally, to discuss potential strategies for such investigations. PubMed was searched using specified search terms. One small prospective study on diet and anti-TNF treatment in 56 patients with CD found similar remission rates after 56 weeks among 32 patients with good compliance that received concomitant enteral nutrition and 24 with poor compliance that had no dietary restrictions (78% versus 67%, p = 0.51). A meta-analysis of 295 patients found higher odds of achieving clinical remission and remaining in clinical remission among patients on combination therapy with specialised enteral nutrition and Infliximab (IFX) compared with IFX monotherapy (OR 2.73; 95% CI: 1.73–4.31, p < 0.01, OR 2.93; 95% CI: 1.66–5.17, p < 0.01, respectively). In conclusion, evidence-based knowledge on impact of diet on anti-TNF treatment response for clinical use is scarce. Here we propose a mechanism by which Western style diet high in meat and low in fibre may promote colonic inflammation and potentially impact treatment response to anti-TNF drugs. Further studies using hypothesis-driven and data-driven strategies in observational, animal and interventional studies are warranted.

Genetic engineering has revolutionized our ability to modify microorganisms for various applications in agriculture, medicine, and industry. This review examines recent advances in genetic engineering techniques for bacteria, fungi, and oomycetes, with a focus on CRISPR-Cas systems. In bacteria, CRISPR-Cas9 has enabled precise genome editing, enhancing applications in antibiotic production and metabolic engineering. For fungi, despite challenges associated with their complex cell structures, CRISPR/Cas9 has advanced the production of enzymes and secondary metabolites. In oomycetes, significant plant pathogens, modified Agrobacterium-mediated transformation and CRISPR/Cas12a have contributed to developing disease-resistant crops. The review provides a comparative analysis of genetic engineering efficiencies across these microorganisms and addresses ethical and regulatory considerations. Future research directions include refining genetic tools to improve efficiency and expand applicability in non-model organisms. This comprehensive overview highlights the transformative potential of genetic engineering in microbiology and its implications for addressing global challenges in agriculture, medicine, and biotechnology.

Background: Bacterial resistance to several antibiotic classes is seen as a grave threat worldwide. Bacterial resistance to antibiotics was mostly caused by the abuse and overuse of antibiotics, particularly penicillins and first-generation cephalosporins, in Egypt's primary healthcare facilities. Aim of the study: To investigate the main resistant bacteria to a few chosen penicillins and first-generation cephalosporins, as well as the characterization of these antibiotics' resistant genes and their detrimental effects in Egypt's main medical facilities. Methodology: Two hundred respiratory and urinary tract infection (RTI and UTI) specimens were gathered from Egyptian primary medical center patients. The turbidimetric assay at 630 nm wavelength, the broth dilution technique, and the nanopore sequencing homology approach were used to characterize the major resistant pathogenic bacteria and their resistant genes to cefadroxil (first generation cephalosporins) and nafcillin (antipseudomonal penicillin). Moreover, adverse outcomes resulting from antibiotic resistance were noted. Results: Out of 200 specimens with RTI and UTI, 186 samples showed the proliferation of resistant bacteria isolates. Enterococcus faecium strain NFC and Staphylococcus aureus strain CDX were among them. Nanopore sequencing confirmed that the antibiotic-resistant gene in Enterococcus faecium was the NFC gene, whereas the antibiotic-resistant gene in Staphylococcus aureus was the CDX gene. Penicillin-binding proteins with modest affinity were encoded by both genes. These bacterial resistance patterns were linked to a low death rate of 5% and a high morbidity rate of 47%. Conclusion: By expressing low-affinity penicillin-binding proteins through resistant NFC and CDX genes, Enterococcus faecium strain NFC and Staphylococcus aureus strain CDX were found to be the predominant bacterial isolates that cause antibiotic resistance to first-generation cephalosporins and penicillins.

Probiotics are becoming more and more popular in the growing of fin and shellfish because these species are consumed worldwide and because different techniques are employed to maximize the productivity and efficiency of fin and shellfish. Probiotic Bacillus is one of the many types of probiotics that are used, but it stands out due to its superior qualities. For instance, because of its ability to sporulate, it can survive in harsh environments, produce antimicrobial substances, and is safe to feed to fish. The different species of Bacillus used in the farming of fin and shell fish were discussed in this review, along with the benefits of using Bacillus spp. as a good substitute in sustainable aquaculture to improve feed utilization, immune system response, stress adaptation, resistance to infectious diseases, tissue preservation integrity, and water quality. Additionally, we discussed the possible risks to safety that Bacillus poses when raising these significant species. For the purpose of future research and development regarding Bacillus application in aquaculture, a summary of the findings of recent studies about the advantages of applying Bacillus to enhance fin and shellfish aquatic animal culture has been provided.

Extensive green roofs provide for many ecosystem services in urban environments. The efficacy of these services is influenced by the substrate characteristics and vegetational composition. Despite their key role in plant performance and productivity, or their contribution to nitrogen fixation or carbon sequestration, green roof microbial communities have received little attention so far. Here, we investigated whether microbial communities in extensive green roof substrates contain a core community, as well as whether they are affected by green roof plant diversity. Eleven green roofs, planted either with a mixture of grasses, wildflowers and succulents (Sedum spp.), or solely species of Sedum, were seasonally sampled to investigate prokaryotic and fungal communities via metabarcoding. Although Sedum-herbs-grasses roofs contained more plant species, we observed no discriminant microbial communities between both roof types, likely due to sharing the same substrate textures and the vegetational layers that became more similar throughout time. Examining key microbial taxa revealed that most taxa are also dominant phylotypes in soils across different biomes. Many bacterial core taxa are capable of nitrogen fixation and most fungal key taxa are stress-tolerant saprotrophs, endophytes, or both. Microbial community composition further revealed high similarities with aerial samples taken in urban environments. Considering the extreme edaphic conditions that periodically arise in green roof substrates, further investigation is warranted to determine the extent to which the dynamic and intriguing microbial communities are influenced by the aeromicrobiome.

A Infectious diseases account for about 3 million deaths per year. The advent of molecular techniques has led to an enormous improvement in their diagnosis, both in terms of sensitivity and specificity and in terms of the speed with which a clinically useful result can be obtained. Digital PCR, or 3rd generation PCR, is based on a series of technical modifications that result in more sensitive techniques, more resistant to the action of inhibitors and capable of direct quantification, without the need for standard curves.This review presents the main applications that have been developed for the diagnosis of viral, bacterial and parasitic infections, and the potential prospects for the clinical use of this technology.

Haemophilus influenzae is one of causative agent of invasive bacterial pathogen that affects both children and adults. Haemophilus influenzae is a pleomorphic gram-negative coccobacillus and it is a common commensal of the upper respiratory tract. It is a human-only pathogen that can cause severe invasive diseases. These bacterial infections can range from mild, such as ear infections, to severe, such as bloodstream infections. The infections typically affect children younger than 5 years old and old age person older than 65 years. They also affect people who are immunocompromised, such as those with certain medical conditions. The highest incidence rates of invasive Haemophilus influenzae disease have recently been discovered in various nations, including North America, Canada, and parts of Europe. In order to monitor the evolving nature of invasive Haemophilus influenzae disease critically reviewed data is required to capture the true status of invasiveness of the Haemophilus influenza diseases. Developing new vaccines against Haemilus influenzae is a potential solution to protect some vulnerable populations against the invasive disease due to this bacterial species. This review article thoroughly investigate recent and up-to-date biomedical science perspective development, innovation, findings, publications and current areas of scientific interest and gap including pathogenicity, diagnosis, multidrug-resistance, Molecular characterization and genetic evolution, epidemiology and immunological characteristics of Haemophilus influenzae, including specific current issues that are affecting the research and development of vaccines to treat Haemophilus influenzae non serotype b diseases and providing insight into how these problems may be overcome.

An event of increasing interest during host-pathogen interactions is the differentiation of patrolling monocytes (MOs) and the polarization of naive MOs into macrophage subsets (MФs). Therapeutic strategies aimed at modulating this event are under investigation. This review focuses on the mechanisms of induction, development, and profile of MФs polarized toward classically proinflammatory (M1) or alternatively anti-inflammatory (M2) phenotypes in response to bac-teria, fungi, parasites, and viruses. It highlights nuclear (PPARγ), cytoplasmic, and cell surface receptors (pattern recognition receptors/PPRs), microenvironmental mediators, and immune signaling. MФs polarize into phenotypes: M1 MФs, activated by IFN-γ, pathogen-associated mo-lecular patterns (PAMPs, e.g. LPS)/DAMPs and membrane-bound PPRs ligands (TLRs/CLRs lig-ands); or M2 MФs (a, b, c and d), induced by IL-4, -10 and -13, antigen-antibody complexes, and helminth PAMPs. The kinetics of M1 and M2 polarization evolve in a pathogen-specific manner, with or without canonicity, and can vary widely. Ultimately, this can result in varying degrees of host protection or more severe disease outcome. While the host is driving an effective MФs po-larization, pathogens are attempting to shift it to increase pathogenicity. The latter results from a bias toward an ineffective M1 or M2 response due to microbial virulence factors, culminating in pathogen evasion of host defenses. Plasticity of M1/M2 phenotypes is also ensured by cellu-lar/genomic reprogramming. Because modulation of this event can occur at multiple points, providing a therapeutic/vaccine design target to boost microbicidal (M1) or tissue repair (M2) response during the inflammation-to-resolution transition; new insights and emerging perspec-tives may have clinical implications.

Water microbial contamination is a serious issue that poses a risk to both animal and human health. 120 water samples collected from main water source and drinkers from a poultry farm. Different bacterial pathogens were isolated from water sources. Escherichia (E.) coli, Pseudomonas (P.) aureoginosa, (Salmonella) S. Typhimurim, Aeromonas (A.) hydrophila at different percentages. Variable degree of bacterial resistance to some commercial disinfectants commonly used to disinfect water system (iodine, terminator and H2O2). Nanoparticles were used to control bacteria in water. About the safety investigation for the prepared nanomaterials, the work results demonstrated that zinc oxide (ZnO) nanoparticles (NPs) exhibit the highest safety profile among the manufactured materials. The median fatal dose (LD50) for ZnO NPs was determined to be 3709 mg/kg body weight. In comparison, the LD50 values for zeolites and nanocomposites were 3251 mg/kg and 2658 mg/kg, respectively. Therapeutic dosages were estimated based on the LD50. Zeolite NPs, ZnO NPs and ZnO/zeolite NPs showing promising results in control of those bacteria. It was concluded that the escalating resistance of bacteria to disinfectants have led to a need to find alternative such as nanoparticles that proved promising results in control of pathogens, particularly it showed a safe effect on laboratory animals.

This study aimed to isolate, identify enteric bacterial species from the migratory seagulls migrated to Libyan cost and to assess the antibiotic resistance of these bacteria. A total of 50 fecal samples were collected from slender-billed gulls in January, 2023 at Farwa Island near the city of Zwara. Bacteria were isolated by conventional culturing method, identified by using the Enterosystem 18R, and antibiotic susceptibility testing was conducted on the isolated bacteria. Only 32 of 46 bacteria were identified using biochemical tests. These identified bacteria belong to six species of Enterobacteriacae namely Citrobacter freundii, Pantoea agglomerans, E. coli, Enterobacter cloacae, Serratia liquifaciens, and Proteus mirabilis, with percentages of (17) 53.125%, (10) 31.25%, (2) 6.25%, (1) 3.125%, (1) 3.125%, and (1) 3.125%, respectively. All isolated bacterial species in this study were 100% sensitive to gentamicin and ciprofloxacin. The highest resistance rate was observed against the antibiotic cefoxitin. The results indicated that C. freundii was the most antibiotic-resistant bacterial species isolated in this study. In conclusion, slender-billed gulls carry multi-drug resistant bacteria. The study recommends implementation of a national program to survey antibiotic-resistant bacteria and expanding scientific research on migratory birds.

Antibiotic resistance is now a first-order health problem, which makes the development of new families of antimicrobials an imperative. These compounds should ideally be inexpensive, readily available, highly active and non-toxic. Here we present the results on the antimicrobial activity of a series of natural and synthetic polyamines with different architectures (linear, tripodal and macrocyclic) and their derivatives containing the oxygen containing aromatic functional groups 1,3-benzodioxol, ortho/para phenol or 2,3-dihydrobenzofuran. The new compounds were prepared through a non-expensive process and their activity was tested against selected strains of yeast, gram-positive and gram-negative bacteria. In all cases, the conjugated derivatives showed antimicrobial activity higher than the unsubstituted polyamines. Several factors, such as the overall charge at physiological pH, the lipophilicity and the topology of the polyamine scaffold were relevant to their activity. The nature of the lipophilic moiety was also determinant on human cell toxicity. The lead compounds showed to be bactericidal and fungistatic, and they were synergic with the commercial antifungals fluconazole, cycloheximide and amphotericin B against the yeast strains tested.

Glucose is an important carbon sources for microbial growth, and its content in infertile soils is essential for the growth of the bacterium. Since the mechanism of oligotrophic bacterium adaptation in barren soils is unclear, the research employed RNA Seq technology to examine the impact of glucose concentration on the low nutrient bacteria BS-G1 in soil affected by desertification. RNA-Seq revealed that differentially expressed genes（DEG）histidine metabolism, glutamate synthesis, HIF-1 signalling pathway, sporulation, and TCA cycle pathway of BS-G1 were significantly enriched in 0.015 g/L glucose concentration, compared with 10 g/L glucose concentration. DEG amino acids system, two-component system, Metal Ion Transport, and nitrogen metabolism system of B. subtilis BS-G1 were significantly enriched in 5 g/L glucose concentration, compared with 10 g/L glucose concentration. In addition, The present study identified its regulation pattern and key genes under the low glucose environment (7 mRNAs, 16 sRNAs). This study primarily investigates the variances in the regulatory pathways of oligotrophic bacteria BS-G1, which holds substantial importance in comprehending the mechanism underlying the limited sugar tolerance of oligotrophic bacteria.

Fermented rice is known as a healthy food due to the presence of lactic acid bacteria as fermentative microorganisms. The study was carried out to discover and characterize the lactic acid bacteria (LAB) from white and red fermented rice (BG varieties). Fermentation was carried out naturally by soaking red, white, raw and cooked rice in water using an earthen pot. Species were confirmed based on 16S r DNA gene sequencing and studied for phenotypic characteristics including morphological, physiological (growth temperature, salt tolerance, milk coagulation) and biochemical characteristics (carbohydrate fermentation pattern) using API 50CH kits. Distinct clusters like cocci (48), diplo cocci (30) and rods (30) shaped bacteria were observed from fermented rice. Five species of lactic acid bacteria including Latilactobacillus curvatus GR Lb1,2 10 and 11 (predominant Bacillus species), Latilactobacillus graminis Lb8, Limosilactobacillus fermentum GRLb17, Weissella confuse GRLb4 and Pediococcus pentosaceus GRLc1 were identified. The base pair length of amplified DNA for the isolates was 1500 Bp. The beneficial physiological and biochemical properties of isolated Lactobacillus species from fermented rice revealed their potential applications in the food industry.

Mosquitoes transmit pathogens that cause human diseases such as malaria, Dengue fever, Chikungunya, yellow fever, Zika fever, and filariasis. Chemical pesticides are effective instruments for reducing disease transmission and managing mosquito populations. However, these chemicals have drawbacks such as high production costs and negative effects on the environment and non-target organisms. Furthermore, as mosquitoes develop resistance to chemical insecticides, they become less effective. As a result, researchers are investigating novel insecticides. Advances in microbial biotechnology have resulted in a wide range of value-added products, including biopesticides for agricultural and public health purposes. In this review, we highlight the mainly untapped biotechnological potential of microorganisms for vector mosquito control and disease transmission reduction. Also, we underline the importance of more basic research and improved translational research methodologies, encouraging efforts to bridge the gap between academic research on biopesticides and public health interventions.

There is significant lack of information on the presence of plant pathogens in aquifer, increasingly used for irrigation during last decades and considered mostly free of them. In this work, we report the presence of bacteria from the Pseudomonas syringae complex (referred as PsyC) including phytopathogenic bacteria, in groundwater of Avignon, France. Their concentration was variable and inversely correlated with water electrical conductivity. Their mean abundance were hundred times lower than in the river Durance, connected with the aquifer but surprisingly, their genetic structure were more homogeneous. Moreover, most strains (97 %) from groundwater were tested as potentially pathogenic on plants, when in the river they were only 71 %. Determinants of this low diversity and high aggressiveness remain to be identified. We conclude that aquifers must be considered potential plant pathogenic reservoirs even if more surveys are needed to understand the real impact on crops during irrigation. These results could be included in prediction models and new approaches to disease forecasting and surveillance and could lead to adaptation of agricultural practices.

Abstract: The overuse of antibiotic prophylaxis during the COVID-19 pandemic would have led to the devel-opment of antibiotic-resistant bacterial strains, thereby increasing the epidemiological burden of antimicrobi-al resistance. The aim of this study was to determine the prevalence and antibiotic resistance pattern of bacte-ria isolated in 02 referral health facilities in Yaoundé before and during the COVID-19 pandemic era. We conducted a retrospective study over a period of 03 years (from 1 January 2019 to 31 December 2021) in the bacteriology units of the Central and General Hospitals of Yaoundé, Cameroon. The review of the services' registers was done to record bacterial isolates (Streptococcus, Staphylococcus, Neisseria meningitidis and Entero-bacteriaceae) as well as their susceptibility to selected, specific and highly potent antibiotics: Cefixime, Azythromycin and Erythromycin. The relationship between each group of resistant bacteria and the antibiotic of interest was determined by simple linear regression; the comparison of the prevalences of before (2019) and during 2 consecutive years of the COVID-19 pandemic onset (2020 and 2021) was done by the Chi2 test of in-dependence. In all, 426 bacterial strains were included. It appeared that the highest number of bacteria iso-lates and lowest rate of bacterial resistance were recorded during the pre-COVID 2019 period in 2019 (160 isolates vs. 58.8% resistance rate). Conversely, lower bacteria stains but greater resistance burden were rec-orded during the pandemic era (2020 and 2021) with the lowest bacteria amount and peak of bacteria re-sistance registered in 2020, the year of COVID-19 onset (120 isolates vs. 70% resistance in 2020 and 146 iso-lates vs.58.9% resistance in 2021). In contrast to almost all others groups of bacteria where the resistance burden was quite constant over years, the Enterobacteriaceae exhibited greater resistance rate during the pandemic period [60%(48/80) in 2019 to 86.9%(60/69) in 2020 and 64.5%(61/95) in 2021)]. Concerning antibiotics, unlike Erythromycin, Azythromycin related resitance increased during the pandemic period and the resistance to Cefixim tends to decrease the year of the pandemic onset (2020) and re-increase one year therafter. A significant association was found between resistant Enterobacteriaceae strains and Cefixime (R= 0.7; P-value= 0.0001) and also, between resistant Staphylococcus strains and Erythromycin (R= 0.8; P-value= 0.0001). It could be that the more frequent use of these antibiotics has increased resistance rate especially in Enterobacteriaceae. Anticromicrobial resistance should be closely monitor during and after COVID pandemic era.

Cyanobacteria constitute pioneer colonizer of specific environments whom settlement in new biotopes precedes the establishment of composite microbial consortia. Some heterotrophic bacteria constitute cyanobacterial partners that are considered as their cyanosphere, being potentially involved in mutualistic relationships through exchange and recycling of key nutrients, and sharing of common goods. Several non-axenic cyanobacterial strains have been recently isolated along with their associated cyanosphere from the thermal mud of Balaruc-les-Bains (France) and the biofilms of the retention basin where they develop. The community structure and relationships among members of the isolated cyanobacterial strains were characterized using a metagenomic approach combined with taxonomic and microscopic description of the microbial consortia. Results provide insights into the potential role and metabolic capabilities of microorganisms of thermal mud-associated cyanobacterial biofilms. Thus, the physical proximity, host-specificity and complimentary functions advocate for their complementarity between cyanobacteria and their associated microbiota. Besides these findings, our results also highlight the great influence of the reference protein database chosen when performing functional annotation of the metagenomes from organisms of the cyanosphere and the difficulty of selecting one unique database that appropriately cover both autotroph and heterotroph metabolic specificities.

Pathogenic Neisseria gonorrhoeae causes the sexually-transmitted infection gonorrhea. N. gonorrhoeae has evolved high levels of antimicrobial resistance (AR) leading to therapeutic failures even in dual-therapy treatment with azithromycin and ceftriaxone. AR mechanisms can be acquired by genetic transfer from closely related species, such as naturally-competent commensal Neisseria species. At present, little is known about the antimicrobial resistance profiles of commensal Neisseria. Here, we characterized the phenotypic resistance profile of four commensal Neisseria species (N. lactamica, N. cinerea, N. mucosa, and N. elongata) against 10 commonly used antibiotics, and compared their profiles to 4 N. gonorrhoeae strains, using disk diffusion and minimal inhibitory concentration assays. Overall, we observed that 3 of the 4 commensals were more resistant to several antibiotics than pathogenic N. gonorrhoeae strains. Next, we compared the penicillin-binding-protein 2 (PBP2) sequences between commensal and N. gonorrhoeae strains. We found mutations in PBP2 known to confer resistance in N. gonorrhoeae also present in commensal Neisseria sequences. Our results suggest that commensal Neisseria have unexplored antibiotic resistance gene pools that may be exchanged with pathogenic N. gonorrhoeae, possibly impairing drug development and clinical treatment.

Multidrug resistant (MDR) bacteria have become a severe problem for public health. Developing new antibiotics for MDR bacteria is difficult, from inception to the clinically approved stage. Here, we have used a new approach; we have modified the antibiotic, ciprofloxacin (CFX), with triphenylphosphonium (TPP, PPh3) moiety via ester- (CFX-ester-PPh3) and amide-coupling (CFX-ester-PPh3), to target bacterial membranes. In this study, we have evaluated the antibacterial activities of CFX and its derivatives against 16 species of bacteria, including MDR bacteria, using minimum inhibitory concentration (MIC) assay, morphological monitoring, and expression of resistance-related genes. TPP-conjugated CFX, CFX-ester-PPh3 and CFX-amide-PPh3 showed significantly improved antibacterial activity against Gram-positive bacteria, Staphylococcus aureus, including MDR S. aureus (MRSA) strains. The MRSA ST5 5016 strain showed high antibacterial activity, with an MIC values of 11.12 µg/mL for CFX-ester-PPh3 and 2.78 µg/mL for CFX-amide-PPh3. The CFX derivatives inhibited biofilm formation in MRSA by more than 74.9% of CFX-amide-PPh3. In the sub-MIC, CFX derivates induced significant morphological changes in MRSA, including irregular deformation and membrane disruption, accompanied by a decrease in the level of resistance-related gene expression. With these promising results, this method is very likely to combat MDR bacteria, through a simple TPP moiety modification of known antibiotics, which can be readily prepared at clinical sites.

Ruminal microorganisms play a pivotal role in cattle nutrition. The discovery of the main microbes responsible for enhancing the gain of weight in beef cattle might be used in therapeutic approaches to increase animal performance and cause less environmental damages. Here, we examined differences in bacterial and fungal composition of rumen samples of Braford heifers raised in a natural grassland from Pampa Biome in Brazil. We aimed to detect microbial patterns in the rumen that could be correlated with the gain of weight. 16S and ITS1 genes were amplified from ruminal samples and sequenced to identify the closest microbial relatives within the microbial communities. A predictive model based on microbes responsible for the gain of weight was build and further tested using the entire dataset. The model detected a set of microorganisms associated with animals in the high gain of weight group, including the bacterial taxa RFN20, Prevotella, Anaeroplasma and RF16 and the fungal taxa Aureobasidium, Cryptococcus, Sarocladium, Pleosporales and Tremellales. Most of these organisms have been correlated to the production of substances that improve the ruminal digestion process. These findings provide new insights about cattle nutrition and suggest the use of these microbes to improve beef cattle breeding.

Polydiacetylene (PDA) inserted in films or in vesicles have received increasing attention due to PDA property to undergo a blue-to-red colorimetric transition along with a change from non-fluorescent to fluorescent upon application of various stimuli. In this review paper, the principle of the detection of various microorganisms (bacteria: directly detected or detected through the emitted toxins or through their DNA, and viruses) and of antibacterial and antiviral peptides based on these responsive PDA vesicles are detailed. The obtained analytical performances, when vesicles are in suspension or immobilized, are given and compared to those of the responsive vesicles mainly based on the vesicle encapsulation method. Many future challenges are then discussed.

One consequence of the global technological advancement in conventional metallurgy is the fast depletion rate of valuable minerals, which are also becoming increasingly difficult to find in pure and economically viable forms. This has spurred more interest in technologies that investigate ability of different microorganisms to mobilize valuable metals from their ores via diverse metabolic processes. This study was carried out therefore to isolate, identify and characterize iron solubilizing bacteria from Iron stones of Agbaja iron ore mining site of Kogi State, Nigeria. Crushed samples in the range of 0.25µm and 0.75µm particle sizes were cultured in a modified 9k media to facilitate bacterial growth and pure cultures were then isolated and sub-cultured for further bioleaching studies. Morphological and biochemical analysis suggests that some of the bacteria identified are members of Acidithiobacillus spp, Pseudomonas spp, and Leptospirillum spp. Studies conducted on pure cultures and mixed consortium of the identified organisms shows that a mixture of the three organisms leached iron ore to about 96.16%. Also results of growth pattern due to bacteria countafter 24-72hours of incubation ranged between 0.1×10³ cfu/ml and 12.3 ×10³ cfu/ml for Acidithiobacillus spp. The need to explore the molecular characteristics of these organisms with a view to generating more information on the quality/quantity of their DNA for future cloning activities was also investigated in this work. DNA was extracted using zymo fungal/bacterial extraction mini prep kit ^TM (cat #6001) and subsequently subjected to 1% agarose gel electrophoresis. Visible bands were obtained with Alpha Innotech Gel Documentation Machine. DNA amplification was carried out using a pettier based thermo cycler PCR machine and electrophoresed on 1.5% agarose gel. Results of the PCR shows a visible band corresponding to 1.5kbp using this primer 27F (5¹-GAGTTTGATCCTGGCTCAG-3¹) and 1492R (5^1-GGTTACCTTGTTACGACT-3¹). DNA purity check shows two of the bacteria possess very good qualities for sequencing for further molecular analysis.

The burden of bacterial bloodstream infections (BSIs) is rapidly increasing in Africa including Rwanda. This is a retrospective study investigates the diversity, distribution, and antimicrobial susceptibility profiles of BSIs bacteria in three tertiary referral hospitals in Rwanda between 2020 and 2022. A total of 1,532 blood culture tests were performed for visiting patients. Overall, the proportions of Gram-negative and Gram-positive bacteria were 48.2% and 51.8, respectively. Staphylococcus aureus was the predominant species accounting for 25% of all Gram-positive BSIs species and Klebsiella species representing 41% of all Gram-negative BSIs species. Antimicrobial susceptibility testing revealed that Amikacin exhibited the highest activity against Enterobacter spp., Serratia spp. and Escherichia coli in &gt;92% of cases and Klebsiella spp. in 75.7%. Meropenem and Imipenem were highly efficacious to Salmonella spp. (100% susceptibility), Enterobacter spp. (96.2% and 91.7%, respectively), and Escherichia coli (94.7% and 95.5%, respectively). The susceptibility of Enterococcus spp., S. aureus, and Streptococcus spp. to Vancomycin was 100%, 99.5% and 97.1%, respectively. Klebsiella spp. was highly sensitive to Colistin (98.7%), Polymyxin B (85.6%), Imipenem (84.9%), and Meropenem (78.5%). We recommend strengthening the implementation of integrated transdisciplinary and multisectoral One Health including AMR-stewardship for the surveillance, prevention, and control of AMR in Rwanda.

Background: Bacteria, among the earliest life forms on Earth, are integral to both human health and environmental ecosystems. They contribute to essential processes such as digestion, nutrient absorption, and immune defense, and play key roles in the biogeochemical cycles that sustain life. Objective: This paper explores the indispensable role of bacteria in human life and environmental health, emphasizing their contributions to digestion, nutrient absorption, immune protection, and ecosystem functioning. It also discusses advances in understanding the human microbiome and their implications for health and therapeutic interventions. Methods: The study reviews existing literature on the human microbiome, probiotics, and the role of bacteria in environmental nutrient cycling. It analyses experimental evidence and case studies, particularly focusing on the impact of fecal microbiota transplantation (FMT) as a therapeutic tool. Metagenomic analysis techniques are examined to highlight the diversity and functionality of gut bacteria. Results: Bacteria are found to be essential for breaking down food, providing vital nutrients, and protecting against pathogens. They play a critical role in maintaining a healthy immune system and are involved in the biogeochemical cycles necessary for ecosystem health. Metagenomic analysis reveals significant variations in bacterial populations associated with health and disease, emphasizing the importance of a balanced microbiota. FMT shows promise as an effective treatment for various diseases, including gastrointestinal, metabolic, and neurological disorders. Conclusions: Bacteria are crucial for human health and environmental stability. Maintaining a balanced microbiota is vital for overall well-being, and interventions such as probiotics and FMT can offer significant health benefits. Future research should focus on further understanding the complex interactions between bacteria and their hosts to develop targeted therapies for various health conditions.

The antimicrobial quantitative structure–activity relationship of plant flavonoids against Gram-positive bacteria was established in our previous works, and the cell membrane was confirmed as a major site of action. To investigate whether plant flavonoids have similar antibacterial effects and mechanisms against both Gram-negative and Gram-positive bacteria, here, the minimum inhibitory concentrations (MICs) of 37 plant flavonoids against Escherichia coli were determined using the microdilution broth method, and then the correlation between their lipophilic parameter ACD/LogP or LogD_7.40 value and their MIC was analyzed. Simultaneously, the correlation between the ACD/LogP or LogD_7.40 value and the MIC of 46 plant flavonoids reported in literature against E. coli was also analyzed. Both sets of results showed that there is a significant correlation between the LogP value and the MIC of plant flavonoids against Gram-negative bacteria. However, it is difficult to effectively predict the MIC of plant flavonoids against Gram-negative bacteria from their lipophilic parameters. By comparing two regression curves derived from plant flavonoids against Gram-negative and Gram-positive bacteria, it was further discovered that the antibacterial activities of most plant flavonoids against Gram-negative bacteria are stronger than those against Gram-positive bacteria when their LogP values are less than about 3.0, but the opposite is true when their LogP values are more than about 3.6. Moreover, this comparison also suggests that unlike mainly acting on the cell membrane of Gram-positive bacteria, plant flavonoids have multiple mechanisms against Gram-negative species, while the cell membrane is also an important action site among them. Combined with the correlation analyses between the enzyme inhibitory activity and the LogP value of the reported flavonoids, it was further suggested that DNA gyrase is another important target of plant flavonoids against Gram-negative bacteria.

Zingiber cassumunar Roxb. is a powerful medicinal plant that has been used as traditional medicine to cure respiratory problems, pain, and inflammation in China, Indonesia, Thailand and other Asian countries by using the crude extracts. The objective of this research is to identify phytochemical composition of Z. cassumunar Roxb. and to analyze antibacterial activity of crude extract, purified compounds, and their microencapsulated products of Rhizome Z. cassumunar Roxb. Identification of phytochemical composition in crude extract of rhizome Z. cassumunar Roxb. was achieved by chromatography-mass spectrophotometer. The major phytochemical composition in crude extract of Z. cassumunar Roxb. is essential oils, including terpinen4-ol (37.7%), β-pinene (20.8%), and (E)-1-(3,4-dimethoxyphenyl)but-1-ene (13.3%). Crude extract of Z. cassumunar Roxb. was purified with silica gel flash column chromatography, resulting two purified compounds. The antibacterial activity of crude extract, purified compounds, and their microencapsulated products of Rhizome Z. cassumunar Roxb. were evaluated against agricultural and foodborne pathogens by using disc agar diffusion and broth microdilution techniques. All of the samples studied (crude extracts, purified compounds, and microencapsulated of Z. cassumunar Roxb.) were effective against all the bacteria. Based on the results of the disc-diffusion assay suggested that amongst the samples studied, purified compounds (compound 1 and 2) and microencapsulated purified compounds (compound 1 and 2) exhibited more effective against all the bacteria compared to the crude extracts. Antibacterial activity of the rhizome of Z. cassumunar Roxb. was contributed mainly by the essential oils components as the active compounds. Gram-negative bacteria (X. oryzae, X. translucens, Pseudomonas spp, E. coli, and S. typhimurium) appeared to the most resistant to the crude extracts, purified compounds, and microencapsulated of Z. cassumunar Roxb. compared to the gram-positive bacteria (S. aureus, B. cereus, and L. monocytogenes). Microencapsulated of the tested samples (crude extract, purified compound 1, and purified compound 2) of the rhizome Z. cassumunar Roxb. exhibited high antibacterial activity with no significantly different with the tested samples without microencapsulation. These results suggest potential antibacterial properties of Z. cassumunar Roxb., which useful for agricultural plant health, food preservation, natural therapies, and pharmaceuticals.

Plastics are very versatile materials that have contributed to the development of society since the 19th century, however, their mismanagement has led to an accumulation of waste in almost every ecosystem affecting the fauna of the planet. In this way recently, studies have shown that some insects might be able to adapt, consuming a wide range of hydrocarbon base polymers. In this work it was studied the adaptive capacity of Zophobas morio larvae when feeding on different synthetic polymers derived from petroleum. Four 30-day larval feeding treatments were carried out with synthetic polymers as expanded polystyrene (PS), low density polyethylene (PBD), polyisoprene (PI) and butyl rubber (BT), in addition a positive control of organic diet was included. Intestinal bacteria were isolated from the treatments and identified by Sanger sequencing. To analyze the chemical composition and physical form of the frass produced, fourier transform infrared spectroscopy (FITR) and images of the feces surface were taken with scanning electron microscopy (SEM) respectively. Zophobas morio larvae were able to consume 54% of PS in 30 days, equivalent to 3.2 mg/d/larva. Nine bacterial strains associated with the decomposition of synthetic polymers were identified in the intestine of the larvae. As for the physicochemical analysis of the feces, FTIR spectra showed the scission of bands corresponding to functional groups of the synthetic polymers in the comparison of the plastic diet treatments versus the feces of antibiotic-treated and plastic-fed larvae, while the comparison of spectra of the plastic and control treatments also identified differences in the absorption peaks. SEM imaging demonstrated that superworm feces differed in dependence on the substrate consumed. The findings demonstrate that Zophobas morio larvae possess a gut biological complex that allows them to feed and survive by consuming various petroleum-derived polymers.

The growing global threat of multidrug-resistant (MDR) bacteria has been exacerbated by inadequate antibiotic consumption and the difficulty in developing new therapeutic strategies. Selective Digestive Decontamination (SDD) is presented as an encouraging measure to prevent the appearance and colonization of MDR bacteria. We compared the pattern of antibiotic consumption in the Intensive Care Unit (ICU) of our hospital before and after the implementation of the SDD protocol. This quasi-experimental study was conducted in an ICU from June-2021 to June-2023. The incidence of nosocomial infections, colonizations, and the number of isolated MDR bacteria, as well as the defined-daily-dose (DDD), were monitored. A total of 3266 patients were included, with 1532 in the pre-intervention group and 1734 in the post-intervention group. The median age was 62 years, 66% were male, and the average APACHE II and SOFA scores were 15 and 6.5, respectively. There was a significant reduction in overall antibiotic consumption in the post-SDD group (p= 0.028), highlighting a decrease in Carbapenemase-Producing Enterobacteriaceae (CPE) (p= 0.0099). There was also a reduction in colonization incidence density and nosocomial infections. Implementing the SDD protocol significantly reduced antibiotic consumption in the ICU, suggesting important implications for infection control and antimicrobial resistance management.

Shrimp is the most important aquaculture industry. Therefore, we determined the effect of nitrifying-probiotic bacteria on water quality, growth, survival, immune response and intestinal microbiota of Litopenaeus vannamei cultured without water exchange. In vitro, only B. licheniformis used TAN, nitrites and nitrates since nitrogen bubbles were produced. TAN decreased significantly in the treatments with bacilli and LAB, but no differences were observed in nitrites. Nitrates were significantly higher in the treatments with bacteria. The final weight and survival of shrimp in the bacterial treatments increased significantly and superoxide anion increased significantly only in LAB treatment. The activity of phenoloxidase decreased significantly in the treatment with LAB and with the mixture of bacilli and LAB. Shrimp treated with bacilli in the water showed lower species richness. The gut bacterial community after treatments was significantly different from that of the control. Linoleic acid metabolism was positively correlated with final weight and superoxide anion, whereas quorum sensing was correlated with survival. Thus, bacilli and LAB in the water of hyperintensive culture systems act as nitrifers, modulate the intestinal microbiota and immune response, and improve the growth and survival of shrimp. This is the first report on Pediococcus pentosaceus and Leuconostoc mesenteroides as nitrifying bacteria.

Recent studies have suggested significant antimicrobial properties of some metallic nanoparticles during the height of antibiotic resistance. The silver nanoparticle (AgNPs) is one of the most prominent candidates due to its noble nature and better tolerance profile in humans. The synthesis of AgNPs was carried out using silver nitrate using five different chemical reduction methods and comparing their activity with silver nitrate, also known to possess antimicrobial properties. AgNPs have been synthesized with varying combination of reducing agents, and subsequently characterized by UV-visible spectroscopy, thermogravimetric analysis (TGA), scanning electron microscopy (SEM), and dynamic light scattering (DLS). Chemical reduction with both trisodium citrate and sodium borohydride provided better particle size and controlled uniformity, whereas hydrogen peroxide was used as a stabilizer to increase the stability of AgNPs. The minimum bactericidal concentration (MBC) for all AgNP formulations was determined against gram-negative E. coli, and AgNPs synthesized with only trisodium citrate were found to be the most effective. The synthesized AgNPs were tested against the resistant strains of E. coli, Salmonella, Klebsiella, Bacillus, and Staphylococcus by the well diffusion method to evaluate and compare their inhibition zones and AgNPs synthesized with polyethylene glycol and polyvinyl pyrrolidine were found to be the most efficient.

Transfer RNA (tRNA) modifications are essential for the temperature adaptation of thermophilic and psychrophilic organisms, as they control the rigidity and flexibility of transcripts. To further understand how specific tRNA modifications are adjusted to maintain functionality in response to temperature fluctuations, we investigated whether tRNA modifications represent an adaptation of bacteria to different growth temperatures (minimal, optimal, and maximal), focusing on closely related psychrophilic (P. halocryophilus and E. sibiricum), mesophilic (B. subtilis), and thermophilic (G. stearothermophilus) Bacillales. Utilizing an RNA sequencing approach combined with chemical pre-treatment of tRNA samples, we systematically profiled dihydrouridine (D), 4-thiouridine (s4U), 7-methyl-guanosine (m7G), and pseudouridine (Ψ) modifications at single-nucleotide resolution. Despite their close relation, each bacterium exhibited a unique tRNA modification profile. Our findings revealed increased tRNA modifications in the thermophilic bacteria at higher temperatures, particularly showing elevated levels of s4U8 and Ψ55 modifications compared to non-thermophilic bacteria, indicating a temperature-dependent regulation that may contribute to thermotolerance. Furthermore, we observed higher levels of D modifications in psychrophilic and mesophilic bacteria indicating an adaptive strategy for cold environments by enhancing local flexibility in tRNAs. Our method demonstrated high effectiveness in identifying tRNA modifications compared to an established tool, highlighting its potential for precise tRNA profiling studies.

Endophytes are ubiquitous organisms that reside within plant organs and contribute to the growth and well-being of plants. Nevertheless, there is limited knowledge regarding the plant growth-promoting endophytes found in medicinal plants. Therefore, the aims of the present work were to isolate some endophytic bacteria associated with roots, stems, leaves and inflorescences of Aerva javanica, collected from the Shada Alasfal Mountain at Albaha region, Saudi Arabia. After that, they were screened for some of their direct and indirect plant growth-promoting mechanisms. Finally, based upon their internal transcribed spacer 16S rRNA gene sequencing, the isolates were identified. The results showed that the selected endophytes differentially produced IAA and ammonia, and solubilized phosphate as direct mechanisms. In addition, they had exhibited variable capacities to produce some lytic enzymes and HCN beside their antifungal activities. Furthermore, they showed abilities to tolerate some stress conditions. The bacterial endophytes were identified by molecular methods and belonged to the genera of Micrococcus, Enterobacter, Pseudomonas, Delftia and Bacillus. The findings of this study indicated that the bacterial endophytes isolated from Aerva javanica are possessing crucial functions to improve plant growth and could be utilized as biofertilizers or bioagents to establish a sustainable crop production system.

This article explores the transformative potential of fermentation in elevating the quality of plant-based matrices to match the desirable attributes of traditional dairy and meat products. As the demand for sustainable products without animal welfare issues increases, fermentation emerges as a key process to enhance the organoleptic properties and nutritional content of plant-based analogs. This study explores the effect of fermentation when applied to legume matrices, focusing on the resulting texture, flavor, and nutritional value. A selection of Bacillus subtilis, lactic acid bacteria (LAB) strains, and combinations thereof, showed potential for improving the aforementioned organoleptic and nutritional characteristics of fermented plant bases. In four different legume-derived matrices, fermentation improved texture, degraded undesirable plant carbohydrates, and removed off-flavor compounds, while producing desirable dairy-associated compounds. The degradation of the undesirable beany off-flavor-causing compound hexanal appears to be a universal phenomenon, as every tested strain as well as their combinations exhibited the capability to decrease the hexanal content albeit with varying efficiency. Some LAB strains were found to be capable of producing carotenoids and might hence have the potential for tailoring fermented plant-based matrices for specific applications, such as yellow cheese or red meat analogs.

Acetic acid bacteria (AAB) are major contributors to the production of fermented vinegar, offering various cultural, culinary, and health benefits. Although the residual unpasteurized AAB after vinegar production are not pathogens, these are necessary the safety evaluations including antibiotic resistance for the use as starters. In this research, we investigated the antibiotics resistance profiles of 26 AAB strains, including various species of Komagataeibacter and Acetobacter, against ten different antibiotics using the E-test method. All strains exhibited resistance to aztreonam and clindamycin. Komagataeibacter species demonstrated a 50% resistance rate to ciprofloxacin, analogous to Acetobacter species, but showed twice the resistance rates to chloramphenicol and erythromycin. Genomic analysis of K. saccharivorans CV1 identified intrinsic resistance mechanisms, such as multidrug efflux pumps, thereby enhancing our understanding of antibiotic resistance in acetic acid-producing bacteria. These findings enhance understanding of antibiotic resistance in AAB for food safety and new antimicrobial strategies, suggesting the need for standardized testing methods and molecular genetic study.

Solar pasteurization, as a means of treating milk in remote rural areas without electrical power, is based on using solar energy to thermally inactivate pathogenic microorganisms at temperatures below the boiling point to maintain or improve milk quality. A solar milk pasteurizer of flat‐plate water‐heating glass acting as the solar collector, connected to a stainless-steel cylindrical milk vat, was constructed and evaluated in Navrongo, in the tropical savanna zone of Ghana, West Africa. The novel approach taken in this project is to integrate the solar collection and heat exchanger into a single unit using materials and fabrication techniques readily available in the developing world. The vat comprises a 1.5 mm thick stainless steel cylindrical tank, a 2.2 cm wide hot water jacket, and an outer layer of 5.0 mm thick aluminium foil insulation. Hot water produced by the collector was used for pasteurizing milk. The optimum quantity of milk that this device could pasteurize under the study conditions was 8 L. The device could pasteurize raw milk to a maximum temperature of 74 °C at 14:00 h GMT. The ambient temperature during the pasteurization ranged from 30 °C to 43 °C. The microbial analysis before and after the pasteurization was done, and the result shows that the total bacteria count before pasteurization was 6.6x10⁶ log CFU/mL per unit volume, and the total bacteria count after pasteurization was 1.0x10² log CFU/mL. By acceptable standards, the total bacteria count after pasteurization is within a safe range for consumption, and coliform counts were negative. The solar milk pasteurization system is cost-effective. Hence, it is appropriate for milk producers in arid pastoral areas without electricity to adopt the solar pasteurizer as an alternative to firewood as a fuel source for milk pasteurization. This approach can potentially create a cottage industry that may also play a crucial role as a water treatment technology that will improve the health of rural populations.

The traditional practice of harvesting and processing raw date palm sap is not only culturally significant but also an essential nutritional source in South Asia. Yet, the potential for bacterial or viral contamination from animals and environmental sources during its collection remains a real and insufficiently studied risk. Implementing improved food safety measures and collection techniques could mitigate the risk of these infections. Additionally, the adoption of advanced food analytical methods has the potential to identify pathogens and uncover the natural bacterial diversity of these products. The advancement of NGS technologies, particularly nanopore sequencing, offers rapid and highly mobile capabilities. In this study, we employed nanopore sequencing for bacterial metabarcoding of a set of raw date palm sap samples collected without protective coverage against animals in Bangladesh in 2021. We identified several bacterial species with importance in the natural fermentation of the product and demonstrated the feasibility of this NGS method in the surveillance of raw palm sap products. We revealed two fermentation directions dominated by either Leuconostoc species or Lactococcus species in these products at the first 6 hours from harvest along with opportunistic human pathogens in the background, represented with low abundance. Plant pathogens, bacteria with the potential for opportunistic human infections and the sequences of the Exiguobacterium genus were also described and their potential role is discussed.

Polyphenols and fermentable fibers have shown favorable effects on gut microbiota composition and metabolic function. However, few studies have investigated whether combining multiple fermentable fibers or polyphenols may have additive beneficial effects on gut microbial states. Here, an in vitro fer-mentation model, seeded with human stool combined from 30 healthy volunteers, was supplemented with blends of polyphenols (PP), dietary fibers (FB) or their combination (PPFB) to determine influence on gut bacteria growth dynamics and select metabolite changes. PP and FB blends independently lead to significant increases in the absolute abundance of select beneficial taxa, namely Ruminococcus bromii, Bifidobacterium spp., Lactobacillus spp. and Dorea spp. Total short chain fatty acid concentrations, relative to non-supplemented control (F), increased significantly with PPFB and FB supplementation, but not PP. Indole and ammonia concentrations decreased with FB and PPFB supplementation, but not PP alone while increased antioxidant capacity was only evident with both PP and PPFB. These findings demonstrated that, while the independent blends displayed selective positive impacts on gut states, combination of both blends provided an additive effect. The work outlines the potential of mixed substrate blends to elicit a broader positive influence on gut microbial composition and function to build resiliency toward dysbiosis.

Nutrition is one of the most influential environmental factor in both taxonomical shifts in gut microbiota as well as development of Type 2 Diabetes Mellitus (T2DM). Emerging evidence has shown that the effects of nutrition on both these parameters is not mutually exclusive and that changes in gut microbiota and related metabolites such as short chain fatty acids (SCFA) and branched chain amino acids (BCAA) may influence systemic inflammation and signaling pathways that contribute to pathophysiological processes associated with T2DM. With this background, our review highlights the effects of macronutrients, carbohydrates, proteins and lipids, as well as micronutrients, vitamins, and minerals, on T2DM, specifically through their alterations in gut microbiota and the metabolites they produce. Additionally, we describe the influences of common food groups, which incorporate varying combinations of these macronutrients and micronutrients, on both microbiota and metabolic parameters in the context of diabetes mellitus. Overall, nutrition is one of the first line modifiable therapies in the management of T2DM and better understanding of the mechanisms by which gut microbiota influence its pathophysiology provides opportunities for optimizing dietary interventions.

Introduction Infections caused by multidrug-resistant bacteria (MDR) occur more frequently after pancreaticoduodenectomy (PD) compared to other abdominal surgeries, and infective complications are a leading cause of postoperative morbidity following PD. Pre-operative biliary stent (PBS) placement often leads to biliary contamination, which in turn plays a significant role in postoperative infections. The aim of this study is to evaluate the impact of MDR contamination on short-term postoperative outcomes in patients undergoing PD and to examine the relationship between MDR bacteria, PBS, and bile contamination. Methods This is a retrospective study based on a prospectively maintained database comprising 825 consecutive patients who underwent pancreaticoduodenectomy (PD) between January 2010 and December 2020 at our center. All procedures were conducted with a standardized resection by a dedicated group of surgeons in a specialized pancreatic surgery unit, and the patients were managed according to the same perioperative enhanced recovery protocol. Results MDR bacteria were present in 17.5% of bile cultures, exclusively within the stented group. In our multivariate analysis, the development of a major postoperative complication (MPC) was correlated with the presence of MDR bacteria in the bile (OR 1.66, 95% CI: 1.1-2.52; p = 0.02). MDR bacteria were detected early in the surgical drainage in 144 out of 825 patients (12.1%), with 72.2% having a previous biliary stent placement and 27.8% without stents (p < 0.001). In our multivariate analysis, the development of an MPC was associated with the presence of MDR bacteria in the drainage (OR=1.81, 95% CI: 1.21-2.73, p= 0.0042). Conclusions We demonstrated that MDR contamination worsens the short-term outcomes of patients undergoing PD. Specifically, when MDR bacteria are present in both the bile and drainage, there is a statistically significant increase in the incidence of major postoperative complications (MPC). Our data suggest that the majority of MDR surgical site infections stem from biliary contamination resulting from the placement of a pre-operative biliary stent (PBS).

DNA extraction for downstream molecular diagnostic applications can be an expensive, time-consuming process. We devised a method to quickly extract genomic DNA from environmental samples based on sodium hydroxide lysis of cells with or without capture by magnetic beads, for subsequent PCR or quantitative PCR. The final DNA extraction method using NaOH is extremely low-cost and can be completed in 10 minutes at room temperature. NaOH extraction was effective for Gram-negative and Gram-positive bacteria in samples from air, soil, sewage, food, laboratory surfaces, and chicken cloacal swabs. The NaOH extraction method was comparable to commercial kits for extraction of DNA from pig fecal samples for 16S amplicon sequencing analyses. We demonstrate that an impinger and portable pump can efficiently capture bacteria in poultry facilities for rapid DNA extraction for quantification of total bacteria and for detection of specific species using qPCR. The air sampling and NaOH extraction procedures are well-suited for routine, high throughput screening, and for metagenomic analyses for specific pathogens, even in resource-limited situations.

Many bacteria have the ability to survive in challenging environments; however, they cannot grow on standard culture media, a phenomenon known as the Viable but Non-Culturable (VBNC) state. Bacteria commonly go into the VBNC state under nutrient-poor environments or under stressful conditions. This review explores the concept of the VBNC state, providing insights into the beneficial bacteria known to employ this strategy. The investigation covers different chemical and physical factors that can induce the latency state, cell features, and gene expression observed in cells in the VBNC state. The revision also covered the significance and applications of beneficial bacteria, methods for evaluating bacterial viability, the ability of bacteria to persist in environments associated with higher organisms, and the factors that facilitate the return to the culturable state. Knowledge about beneficial bacteria capable of entering the VBNC state remains limited, however, beneficial bacteria in this state could face adverse environmental conditions, and return to culturable state when conditions become suitable and continue to exert their beneficial effects. Likewise, this unique feature positions them as potential candidates for healthcare applications, such as the use of probiotic bacteria to enhance human health, applications in industrial microbiology for the production of prebiotics, functional foods, and in the beer and wine industry. Moreover, their use in formulations to increase crop yield and for bacterial bioremediation offers an alternative pathway to harness their beneficial attributes.

Aerococcus urinae is an aerobic Gram-positive coccus that grows as tiny alpha-hemolytic colonies. Actinotignum schaalii is a slow-growing facultative anaerobic Gram-positive rod. These bacteria are part of the urogenital microbiota of healthy patients, but can also be involved in urinary tract infections (UTIs), particularly in elderly men and young children [1–3]. Because A. urinae and A. schaalii are fastidious and are difficult to identify with phenotypic methods, they are underestimated causes of UTIs. Their growth is slow and requires a blood-enriched medium and an incubation time of 48 hours under anaerobic or 5% CO2 atmosphere. Furthermore, accurate identification is only possible using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) or molecular-based methods. In rare cases, these bacteria can be responsible for invasive infections. We describe here the second case of bacteremic UTI caused by both A. schaalii and A. urinae in an 89-year-old woman who presented dyspnea and asymptomatic bacteriuria. This challenging clinical and microbiological diagnosis was made by our laboratory by Gram staining urine with a leucocyte count &gt;50/μL and/or a bacterial count &gt;14/μL, urinary culture on blood agar plate, and direct bacterial identification on positive blood culture bottles (BCBs) by MALDI-TOF MS. After 10 days of antimicrobial treatment consisting of 2g amoxicillin PO t.i.d the patient was discharged with complete clinical and biological recovery. A. schaalii and A. urinae are probably still underestimated causes of UTIs. Microbiologists should assess the presence of these two bacteria in urine using the appropriate culture and identification methods where direct examination is positive for small Gram-positive cocci or rods, and a urinary dipstick is negative for nitrites and associated with leukocyturia.

The continued exploration of the diversity of lactic acid bacteria in little-studied ecological niches represents a fundamental activity to understand the diffusion and biotechnological significance of this heterogeneous class of prokaryotes. In this study, Lactiplantibacillus plantarum (Lpb. plantarum) strains have been isolated from Tunisian vegetable sources, including fermented olive, fermented pepper and from dead locust intestines, subsequently evaluated for their antimicrobial activity against foodborne pathogenic bacteria, including Escherichia coli O157:H7 CECT 4267 and Listeria monocytogenes CECT 4031, as well as against some fungi, including Penicillium expansum, Aspergilus niger and Botrytis cinerea. In addition, their resistance to the oro-gastro-intestinal transit, aggregation capabilities, biofilm production capacity, adhesion to human enterocyte-like cells and cytotoxicity to colorectal adenocarcinoma cell line were determined. Further, the adhesion to tomatoes and the biocontrol potential on this model food matrix were analyzed. It has been found that all the strains were able to inhibit the indicator growth, mostly through organic acids production. Besides, these strains showed promising probiotic traits, including in vitro tolerance to oro-gastrointestinal conditions, adhesion to abiotic surface and to Caco-2 cells. Moreover, all tested Lpb. plantarum strains were able to adhere to tomatoes with similar rate (4.0-6.0 LogCFU/g tomato). The co-culture of LAB strains with pathogens on tomatoes showed that Lpb. plantarum could be a good candidate to control pathogen growth. Nonetheless, further studies are needed to guarantee their use as probiotic strains for biocontrol on food matrices.

Postbiotics are biologically active probiotic microorganisms that have produced non-viable bacterial products in the host. Eating a diet high in probiotic and prebiotic foods may help to ensure that the gut has an appropriate supply of these essential nutrients because postbiotics are produced when probiotics feed on prebiotics. Cell wall components and metabolites produced by living bacteria that have positive effects on the host are referred to as postbiotics. Postbiotics are functional bioactive chemicals that are produced in a matrix during anaerobic fermentation of organic nutrients like prebiotics. Postbiotics are low molecular weight soluble chemicals that are either produced after microbial cell lysis or secreted by live microflora as byproducts of this metabolic process. In general, the producer strains from bacterial and fungal species such as Lactobacillus, Bifidobacterium, Streptococcus, Akkermansia muciniphila, Saccharomyces boulardii, Eubacterium hallii, Faecalibacterium, etc. When handling and selling food items in an industrial setting, postbiotics can be used as functional components in foods, which has a number of advantages, including the potential to be added to some foods that are known to be harmful to probiotic survival. Postbiotic supplements have grown in popularity among customers because of their numerous health advantages, particularly food, cosmetic, and healthcare industries. Classification of postbiotics depends on various factors including type of microorganism, structural composition, and physiological functions. This review presents a succinct introduction, history, and concept of postbiotics, the salient features of postbiotics, classification, production, purification, characterization, biological functions, and applications of postbiotics in the food industry.

Background: Antibiofilm activity of silver nanoparticles has been extensively investigated in common bacteria. Metallo-β-lactamase producing Gram-negative bacteria are hard-to-treat microorganisms with few therapeutic options, and silver nanoparticles were not evaluated on the biofilm of these bacteria. Objectives: The aim of this study was to evaluate the antibiofilm activity of a bone scaffold impregnated with silver nanoparticles in NDM-producing Gram-negative bacilli. Methods: Bone scaffolds from bovine femur were used for the tests and impregnated with silver nanoparticles (50 nm) by physical adsorption. Silver nitrate minimal inhibitory and bactericidal concentrations (MIC and MBC) were performed on NDM-producing Escherichia coli, Klebsiella pneumoniae, and Pseudomonas aeruginosa. Disc diffusion tests for silver nanoparticles susceptibility and quantification of biofilm production on plate and bone with sessile cell count were performed. Results: The MIC results demonstrated that silver nitrate had an antimicrobial effect on all microorganisms, inactivating the growth of isolates from a concentration of 8 µg/mL. MBC results showed that E. coli 16.211 was the only isolate to present MIC different from MBC, with a value of 16 µg/mL. Conclusion: Bone scaffolds impregnated with silver nanoparticles can significantly reduce biofilm, and it can be a strategic material to be used as an implant for different approaches.

(1) Background: Respiratory infections are a major public health problem worldwide, with potentially serious consequences. Indeed, these infections remain one of the main causes of morbidity and mortality in children under 5 years old in developing countries. Etiological information on respiratory infections is crucial for prevention and case management strategies. This systematic review aims to describe the etiology of respiratory infections reported in studies carried out in sub-Saharan African countries; (2) Methods: Using PubMed, HINARI and Google scholar search engines, a systematic search was carried out to identify published articles on the etiology of viral and/or bacterial respiratory infections in sub-Saharan Africa in patients of all ages. We have only considered data from sub-Saharan Africa. Papers published from 2010 to 2021, in English or French have been included in this review; (3) Results: After reviewing 115 articles reporting studies carried out in the African continent, only 32 articles were selected of which, studies were conducted in 15 sub-Saharan African countries, including 6/32 (18.75%) in Cameroon. Twenty (62.5%) were cross-sectional studies, and twenty-four (75%) were hospital-based investigations. In these studies, RT-PCR and culture methods were respectively used for viruses and bacteria investigations. Respiratory syncytial virus was the most frequently identified, with prevalence ranging from 0.6% to 59%, followed by rhinovirus (9.3% -73%), influenza virus (flu) A/B (0.9%-69.1%), and human adenovirus (0.9% - 30.8%). Streptococcus pneumoniae (14.2% - 96%), followed by Haemophilus influenzae type b (2.5% - 54%), and Klebsiella pneumoniae (1.4% - 49.9%) were the most frequently detected bacteria; (4) Conclusions: This review has reported that many pathogens, mainly viruses, are associated with acute respiratory infections in sub-Saharan Africa in both children and adults. Unfortunately, the limited geographical distribution of data across sub-Saharan Africa does not allow most of countries to develop an effective strategy for the prevention and treatment of respiratory infections.
