When a star is born, a protoplanetary disk made of gas and dust surrounds the star. The 1 disk can show gaps opened by different astrophysical mechanisms. The gap has a wall emitting 2 radiation which contributes to the spectral energy distribution (SED) of the whole system (star, disk 3 and planet) in the IR band. As these new-born stars are far away from us, its difficult to know 4 whether the gap is opened by a forming planet. I have developed RHADaMAnTe, a computational 5 astro code based on the geometry of the wall gap coming from hydrodynamical 3D simulations of 6 protoplanetary disks. With this code it is possible to make models of disks to estimate synthetic SEDs 7 of the wall gap and prove whether the gap was opened by a forming planet. I have implemented this 8 code to the stellar system LkCa 15. I found that a planet of 10 Jupiter masses is capable of opening a 9 gap with a curved wall with height of 12.9AU. However, the synthetic SED does not fit to Spitzer IRS 10 SED (ξ² ∼ 4.5) from 5µm to 35µm. This implies that there is an optically thick region inside the gap.

To enhance the peak-shaving capability of the boiler, a mathematical model of hydrodynamic and wall temperature characteristics for the water-cooled wall of a 1000 MW boiler was established. The attributes pertaining to the mass flow distribution, the temperature of the effluent working fluid, pressure drop, and the thermal distribution across the wall surfaces at 30% of the boiler's maximum continuous rating (BMCR) were meticulously computed and subjected to a thorough analytical review. The findings suggest that, under the operation at 30% BMCR load, there is a substantial equilibrium in the flow distribution across the quartet of walls that constitute the water-cooled wall assembly. The maximum mass flow rate deviations in the helical and vertical sections are 1.95% and 3.47%, respectively, showing small flow deviations and reasonable distribution. The temperature deviation in the helical section is 0.3 °C, reflecting the characteristic low thermal deviation in helical tubes. While the temperature deviation at the outlet of the vertical section is higher, it remains within safe limits. The pressure loss across the water-cooled wall system amounts to 0.4 MPa. The peak wall temperature reaches 337.5°C, remaining within the material's permissible safety limits. In-depth performance analysis ensures the safe operating conditions of the water-cooled wall during low load conditions.

Background: Functional restoration of abdominal wall defects represent one of the fundamental challenges of reconstructive surgery. Synthetic grafts or crosslinked animal-derived biological grafts are characterized by significant adverse reactions, which are mostly observed after their implantation. The aim of this study was to evaluate the efficacy of the decellularization protocol to produce a completely acellular full-thickness abdominal wall scaffold. Methods: Full-thickness abdominal wall samples were harvested from Wistar rats and submitted to a 3 cylce decellularization process. Histological, biochemical and DNA quantification analyses were applied to evaluate the effect of the decellularization protocol. Mechanical testing and immunogenicity assessment were also performed. Results: Histological, biochemical and DNA analysis results showed the efficient decellularization abdominal wall samples after the 3rd cycle. Decellularized abdominal wall scaffolds were characterized by good biochemical and mechanical properties Conclusion: The data presented herein confirm the effective production of rat-derived full-thickness abdominal wall scaffold. Expanding this approach will allow the exploitation of the capacity of the proposed decellularization protocol in producing acellular abdominal wall scaffolds from larger animal models or human cadaveric donors. In this way, the utility of biological scaffolds with preserved in vivo remodelling properties may be one step closer to its application in clinical studies.

Lizards in Bulgaria hibernate in winter. Yet, individuals of many species were found active in this period. Cases of winter activity of Podarcis muralis and P. erhardii in the country have been published, but this behavior has not been studied in detail. The purpose of this research was to observe and analyze the winter activity of these two species in the city of Blagoevgrad. The observations took place at two sites in days with high solar radiation in December, January, and February 2015–2017. One of the sites was a typical urban territory, inhabited only by P. muralis. The other site was a suburban area, inhabited by P. erhardii. The number of the active individuals of the Common Wall Lizard recorded was about 8.5 times higher than that of the Erhard's Wall Lizard. The lowest air temperature at which P. muralis was observed was -4°C and for P. erhardii it was +0.5°C. Individuals of both species were registered only at positive substrate temperatures. Representatives of all age groups of P. muralis were recorded often with dorsoventrally flattened bodies while basking. On the other hand, few juveniles of P. erhardii were found active and they flattened their bodies very rarely.

Six Half-ring with varying linewidth from 120nm to 370nm were connected in series on five corners. The magnetization reversal processes were investigated by the measurement of anisotropic magnetoresistances (AMR). The number of switching jumps in the AMR loops, from zero to five, varied with the longitudinal applied field. These discrete jumps result from domain wall nucleating and depinning on the corners. The larger external field applied the fewer number of jumps in the MR curve. This reproducible and particular-respondence of domain wall device in pattern of half-ring wire might be the new promising magnetoelectronic devices.

Steam boiler has a significant role in the industrial sector. The failure in boiler tubes significantly reduces the availability of the plant. Furthermore, deposits on the inner tube wall contribute to such failure by changing the thermal resistance of the wall, which causes a significant increase in the tube wall's temperature and, consequently, lower allowable stress. The presented thermal-fluid model is developed for the water tube. The model is implemented using ANSYS FLUENT to estimate the wall maximum water tube wall temperature considering the deposits layer and then determine the minimum thickness of the tube wall via ASME standards to assure safe operation. The model is verified via a mesh independence test. The minimum tube thickness is recommended and justified given the thermo-mechanical performance of the water tubes at several operating loads of 50, 75, and 100 %. These loads are implemented by different convective heat transfer coefficients of flue gases under different deposit thicknesses, thermal conductivity, and flue gas temperature. The present work contributes to the literature by recommending the minimum thickness of the water wall tubes and highlighting the thermo-mechanical effect of the deposits on water tube resilience under different operating conditions.

C. albicans Dfg5 and Dcw1 are homologous cell wall alpha-1,6-mannosidases with important functions. Our past studies have shown that Dfg5 and Dcw1 function in cell wall biogenesis via cross-linking of glycoproteins into the cell wall, thus playing a key role in cell wall integrity. Additionally, Dfg5 and Dcw1 are required for hyphal morphogenesis. However, the exact functions of Dfg5 and Dcw1 in cell wall integrity, hyphal morphogenesis, and pathogenesis were not known. In this study, we determined the relation of Dfg5 and Dcw1 with Hog1 MAPK, that plays a key role in cell wall integrity via regulation of chitin synthesis in C. albicans. Additionally, we also determined the effect of dfg5 and dcw1 mutations on gene expression of transcriptional regulators of hyphal morphogenesis. Furthermore, we determined the effects of dfg5 and dcw1 mutations on pathogenesis in a mouse model of oral candidiasis. Our results demonstrate that dfg5 and dcw1 mutations, as well as hog1 knock-out mutation result in dysregulation of chitin synthesis resulting in a cell separation defect. The heterozygous and conditional mutations of dfg5 and dcw1 result in decreased levels of Cst20, a positive regulator of hyphal morphogenesis. However, mutations of dfg5 and dcw1 mutations resulted in increased levels of all the five negative regulators of hyphal morphogenesis – Tup1, Nrg1, Mig1, Rbf1 and Rfg1. However, Tup1 levels were significantly higher than other negative regulators indicating that Dfg5 and Dcw1 function in hyphal morphogenesis by repression of Tup1. Finally, dfg5 and dcw1 mutations affected the ability of C. albicans to cause oral candidiasis in mice. Thus, cell wall glycosidases, Dfg5 and Dcw1, are required for virulence and pathogenesis and represent novel drug targets.

Watermelon fruits of different ploidy exhibit significant variations in texture. In order to investigate the primary factors influencing texture differences, this study was conducted to investigate the differences in fruit texture development using homozygous autogamous diploid (2X)lines and their autotriploid (3X) and autotetraploid (4X) lines 'Yixuan'. The research encompassed fruit development, flesh texture profile analysis (TPA), flesh cell wall polysaccharide content, and flesh cell microstructure analysis, specifically in 2X, 3X, and 4X watermelon fruits.The results showed that as watermelon fruits matured, several characteristics exhibited an ascending trend, including fruit weight, TSS (total soluble solids) content, rind hardness, flesh cell size, and cell wall polysaccharide contents such as crude fiber, cellulose, hemicellulose, lignin, and protopectin. Conversely, the flesh texture parameters and soluble pectin content of the flesh cell wall gradually declined. Notably, significant differences in fruit flesh texture and flesh cell structure were observed among fruits of different ploidy at the 32d. Specifically, 2X fruits displayed considerably lower rind hardness, reduced flesh texture (Hardness, Fracturability, Chewiness, Gumminess), and cell density compared to 3X and 4X fruits. Additionally, 2X fruits exhibited larger cell sizes than 3X and 4X fruits. However, no significant differences were observed in the content of flesh cell wall polysaccharide contents across various ploidy levels.These findings suggested that the variation in texture among watermelon fruits of different ploidy can be attributed to the size and arrangement of flesh cells. Importantly, this research provided a foundation for further exploration of the intrinsic regulatory factors and molecular mechanisms contributing to texture variation in polyploid watermelon fruits.

Rammed earth in a built environment has vapor diffusion characteristics and humidity control ability each of which are respectively attributed to the porous structures and the hydrophilic properties. Indeed, these structures and properties, which allows easily absorbing water parti-cles, hence, affecting the durability of a rammed earth wall. This paper presents the water repel-lency method for rammed earth walls which synthesizes siloxane copolymers containing fluo-rine. The water-repellent properties are investigated by measuring the contact angle, water ab-sorption rate, and compressive strength after spray coating with the synthesized siloxane co-polymers on the surface of the rammed earth specimens under study. The water contact angle of the specimen coated with a siloxane copolymer containing 10 mol.% of a silane monomer with a fluorine group is about 140o. The water absorption of the specimen obtained after immersing in water for 24 hours is low at about 3.5 wt.%. In addition, the compressive strength is remained more than 80% of the corresponding strength of the specimen which is not immersed into water. It is confirmed that the use of a 10% by volume of the siloxane copolymer containing the fluo-rine group may assure the water-repellent performance and the economic competitiveness.

Background: Abdominal wall hernias are a common condition that is seen in daily general surgery patients. However, still more researches are needed to be conducted regarding hernias. The aim of this study is to see the prevalence of hernias among patients admitted to the Department of Surgery of a tertiary care center. Methods: A descriptive cross-sectional study was performed among patients who admitted to the department of surgery in a tertiary care center between 1 August 2023 and 20 September 2023. All patients data were extracted to an excel sheet. Results: We had 760 patients who visited in this period. Only 272 patients (27.9 %) were diagnosed with hernias. We found 52.6 % of patients were male and 47.4 % of patients were female. The age between 46- 60 years old was the most to have hernias (97 patients). Inguinal hernias were the common among other types. Diabetes mellitus, hypertension, COPD, and hydrocele were found in 10.7 % of all patients. Conclusion: The prevalence of hernias in this study was nearly to other studies that conducted in the same conditions. Early diagnosis and patient’s education are the corner stone to prevent morbidities and mortalities.

Mycobacterium tuberculosis is the agent of tuberculosis, one of the most important infectious diseases in the world. This microorganism stands out from other bacteria, not only for its extremely high infection capacity, but also for its cellular characteristics that include an extremely resistant and hydrophobic cellular parade the passage of antibiotics. An incredible ability to adapt to adverse conditions inside the host as well as its vast arsenal of virulence factors that allow its survival within the inhospitable environment within the macrophage can be highlighted. This review aims to discuss several aspects of MTB microbiology, genetics, and physiology. We will address in this review details of the metabolism of MTB that allows it to replicate in the active phase and remain viable during latency, as well as the characteristics of its cell wall that contribute to the blockade of the immune response and its resistance to antibiotics.

Since Darwin’s “Power of movement in plants” the precise mechanism of oscillatory plant growth remains elusive. Hence the search continues for the hypothetical growth oscillator that regulates a huge range of growth phenomena ranging from circumnutation to pollen tube tip growth and stomatal movements. Oscillators are essentially simple devices with few components. A universal growth oscillator with only four major components became apparent recently with the discovery of a missing component, notably arabinogalactan glycoproteins (AGPs) that store dynamic Ca2+ at the cell surface. Demonstrably, auxin-activated proton pumps, AGPs, Ca2+ channels and auxin efflux “PIN” proteins, embedded in the plasma membrane, combine to generate cytosolic Ca2+ oscillations that ultimately regulate oscillatory growth: Hechtian adhesion of the plasma membrane to the cell wall and auxin-activated proton pumps trigger the release of dynamic Ca2+ stored in periplasmic AGP monolayers. These four major components represent a molecular PINball machine a strong visual metaphor that also recognizes auxin efflux “PIN” proteins as an essential component. Proton “pinballs” dissociate Ca2+ ions bound by paired glucuronic acid residues of AGP glycomodules, hence reassesses the role of proton pumps. It shifts the prevalent paradigm away from the recalcitrant “acid growth” theory that proposes direct action on cell wall properties, with an alternative explanation that connects proton pumps to Ca2+ signaling with dynamic Ca2+ storage by AGPs, auxin transport by auxin-efflux PIN proteins and Ca2+ channels. The extensive Ca2+ signalling literature of plants ignores arabinogalactan proteins (AGPs). Such scepticism leads us to reconsider the validity of the universal growth oscillator proposed here with some exceptions that involve marine plants and perhaps the most complex stress-test, stomatal regulation.

A fundamental question in biology is how cell shapes are genetically encoded and enzymatically generated. Prevalent shapes among walled bacteria include spheres and rods. These shapes are chiefly determined by the peptidoglycan (PG) cell wall. Bacterial division results in two daughter cells, whose shapes are predetermined by the mother. This makes it difficult to explore the origin of cell shapes in healthy bacteria. In this review, we argue that the Gram-negative bacterium Myxococcus xanthus is an ideal model for understanding PG assembly and bacterial morphogenesis because it forms rods and spheres at different life stages. Rod-shaped vegetative cells of M. xanthus can thoroughly degrade their PG and form spherical spores. As these spores germinate, cells rebuild their PG and reestablish rod shape without preexisting templates. Such a unique sphere-to-rod transition provides a rare opportunity to visualize de novo PG assembly and rod-like morphogenesis in a well-established model organism.

Worldwide, hernia repair represents one of the most frequent surgical procedures encompassing a global market valued at several billion dollars. This type of surgery usually requires the implantation of a mesh that needs the appropriate chemical, physical and biological properties for the type of repair. This review thus presents a description of the types of hernias, current hernia repair methods, and the state of the art of prosthetic meshes for hernia repair providing the most important meshes used in clinical practice by surgeons working in this area classified according to their biological or chemical nature, morphology and whether bioabsorbable or not. We emphasise the importance of surgical site infection in herniatology, how to deal with this microbial problem, and we go further into the future research lines on the production of advanced antimicrobial meshes to improve hernia repair and prevent microbial infections, including multidrug-resistant strains. A great deal of progress has been made in this biomedical field in the last decade. However, we are still far from an ideal antimicrobial mesh that can also provide excellent integration to the abdominal wall, mechanical performance, low visceral adhesion and minimal inflammatory or foreign body reactions, among many other problems.

Transient conjugate heat transfer measurements under varying temperature and velocity inlet boundary conditions at incompressible flow conditions were performed for flat plate and ribbed channel geometries. Therefrom local adiabatic wall temperatures and heat transfer coefficients are determined. Those data are analyzed using typical heat transfer correlations, e.g. Nu=CRe^mPr^n determining the local distribution of C and m. It is shown that they are closely linked. A relationship lnC=A−mB is observed with A and B as modeling parameters. They could be related to parameters in log law or power law representations for turbulent boundary layer flows. The parameter m is shown to have a close link to local pressure gradients and therewith near wall streamlines as well as friction factor distributions. A normalization of the C parameter allows to derive a Reynolds analogy factor and therefrom local wall shear stresses.

The envelope (façade) of a building is the first barrier between the exterior and the interior of the building and withstands the highest variation of temperature and solar radiation. Trombe walls are used to take advantage of that and contribute to the heating of interior air, helping the heating system. In this study, a new Trombe wall design is presented with the aim to contribute significantly to the ventilation of residential buildings. For this an exterior wall equipped with a proposed Trombe system was studied in a numerical simulation analysis. The proposed systems consist of two parts, the air cavities on the exterior of the wall, covered with silica glass, and another layer used for heat transfer, on the interior, comprised of a MPCLBs wall of 115mm. Two cities in Romania were considered to represent the coldest and hottest climate in Romania. Brașov representing the cold climate and Constanța representing the hot climate. In the investigation, both the presence and absence of solar radiation were taken into account. In total 4 cases were established. For all four cases monthly research was made using monthly means temperature, direct and diffuse solar radiation. The results are promising and illustrate that the system works the best during the transitional seasons of spring and autumn. The lower the outdoor temperature the higher the temperature can be increased. Overall, solar radiation accounted for an average of 2 °C increase. The temperature increase varied between 3.4 °C to 15.99 °C for Brașov and between 6.42 °C to 12.07 °C in Constanța.

The construction industry generates significant CO2 emissions, and rebar is a major contributor to this environmental impact. Extensive research has been conducted to address this issue. Recent advancements have introduced algorithms to reduce rebar waste and consumption, demonstrating the feasibility of achieving near-zero rebar cutting waste (N0RCW) by considering special-length rebars. However, conventional lap splices, the most common rebar joint method, consistently consume excessive rebar quantities despite extending beyond their mandated zones. Conversely, couplers eliminate rebar lengths required for lapping splices, reducing the usage of rebar. Applying special lengths and couplers in heavily loaded structures like diaphragm walls can significantly reduce rebar usage and cutting waste, consequently reducing CO2 emissions and associated costs. This study aims to optimize rebar consumption and sustainability in diaphragm wall structures by integrating mechanical couplers with a special-length rebar approach. A case study confirmed a substantial reduction in purchased rebar usage (17.95% and 5.38%) and carbon emissions (15.24% and 2.25%) compared to the original design and previous study, respectively. Broadly implementing the proposed method across various buildings and infrastructure projects could further multiply these benefits, fostering the achievement of SDGs adopted by the United Nations for sustainable construction.

A 17th century wall painting representing a Virgin between two Saints in a noble Italian renaissance palace, Palazzo Gallo in Bagnaia (Viterbo, Italy), was restored in 2021 in the context of a wider restoration campaign interesting the main room of the palace built by cardinal Sansoni Riario. Diagnostic analyses done with traditional characterization techniques (optical microscopy on micro-stratigraphic sections, X-ray fluorescence spectroscopy and Fourier transform infrared spectroscopy) provided the identification of both original painting and restoration materials, while imaging investigations as ultraviolet fluorescence photography, false color images and multispectral mapping provided by hypercolorimetric multispectral imaging (HMI) technique enabled the evaluation of the state of conservation, locating restoration interventions and supporting the monitoring of the cleaning procedure. An altered protective Paraloid-based coating dating from early 2000s had to be removed due to the unpleasant glossy finishing given to the painted surface, making the scene barely readable. To pursue a restoration protocol based on environmental sustainability and green chemistry, enzyme-based gels marketed by Nasier-Brenta© and CTS© companies were tested in different protocols for the cleaning of the mash covering the painting. Although some interesting results were observed, the enzymatic cleaning had a scarce effectiveness with timing beyond a reasonable interval. Traditional chemical solvents as Dowanol PM (methoxy-propanol) and benzyl alcohol were necessary to complete the cleaning of the painting surface.

The A. japonicus industry has expanded significantly, but no research has focused on how to de-termine the age of A. japonicus during farming. Correctly estimating the age of A. japonicus can provide a decision-making basis for the breeding process, and data for the protection of A. japonicus aquatic germplasm resources. DNA methylation levels in the body wall of Apostichopus japonicus at 4 months, 1 year, 2 years, and 3 years old were determined by MethylRAD-Seq, and differentially methylated genes related to age were screened. The results of the study found that 441 and 966 differentially methylated genes were detected at CCGG and CCWGG sites, respectively. As-partate aminotransferase, succinate semialdehyde dehydrogenase, isocitrate dehydrogenase, the histone H2AX, heat shock protein Hsp90, aminopeptidase N, cell division cycle CDC6, Ras GTPase activating protein (RasGAP), slit guidance ligand slit 1, integrin linked kinase ILK, mechanistic target of rapamycin kinase Mtor, protein kinase A Pka, and autophagy-related 3 atg3 these genes may play key roles in the growth and aging process of A. japonicus. This study provided data for identifying the age of A. japonicus.

Influenced by geological, climatic, and natural weathering factors, the Great Wall heritage is sus-ceptible to deformations, posing significant challenges to the comprehensive preservation of the Great Wall sites. To investigate techniques for monitoring deformations in extensive linear cultural heritage sites, such as the Great Wall, this study utilizes the SBAS-InSAR technique for deformation monitoring and analysis. A dataset comprising 161 Sentinel-1A images spanning from March 2017 to January 2022 was chosen for SBAS-InSAR processing, yielding a deformation velocity field. To verify result credibility, a typical mountainous segment spanning approximately 896.53 km within the Great Wall landscape corridor underwent analysis. The findings suggest that around 75.8% of the landscape corridor along the Shanxi section of the Ming Great Wall maintain relative stability, displaying deformation rates varying from -10 to 10 mm/year. The remaining 24.2% of the land-scape corridor experiences notable deformations, including a maximum subsidence rate of 33.1 mm/year and a maximum subsidence of 148.6 mm. This study illustrates the potential utility of the SBAS-InSAR technique for monitoring and evaluating surface deformations in extensive linear cultural heritage sites.

The aim of this study is to test the effectiveness and safety of AE-AE surgery combining otoendoscopy and surgical microscopy in the treatment of acquired pars flaccida cholesteatoma in stages Ib and II of the Japan Otological Society classification occupying the tympanic cavity and attic. A historical cohort study on 65 patients. Of the total, 44 patients were treated by AE-AE surgery using an otoendoscope and 21 with canal wall-up tympanomastoidectomy (CWUT). Patients in whom the AE-AE technique was performed had a lower recurrence rate (9%) compared to those treated with CWUT (38%); p=0.013. In addition, the median time to recurrence was lower in the AE-AE group (4 years [P25-75= 1.25-2-75]) than in CWUT group (2 years [P25-75= 3.25-4.75]); p=0.048. Thresholds were higher in the CWUT group compared to the AE-AE group in pre-surgery (53±16 vs 44±15; p=0.039) and post-surgery (52±18 vs 42±16 dB dB; p=0.042), but not in pre-post-surgery comparisons neither in the AE-AE technique (p= 0.89) nor in the CWUT technique (p= 0.96). We concluded that AE with otoendoscopic support is an effective and safe technique for acquired cholesteatomas occupying box and attic in stages IB and II.

The structure of turbulent flow over non-flat surfaces is a topic of major interest in practical applications in both engineering and geophysical settings. A lot of work has been done in the fully rough regime at high Reynolds numbers where the effect on the outer layer turbulence structure and the resulting friction drag is well documented. It turns out that surface topology plays a significant role on the flow drag especially in the transitional roughness regime and therefore, hard to characterize. Survey of literature shows that roughness function depends on the interaction of roughness height, flow Reynolds number and topology shape. In addition, if the surface topology contains large enough scales then it can impact the outer layer dynamics and in turn modulate the total frictional force. Therefore, it is important to understand the mechanisms underlying drag increase from systematically varied surface undulations in order to better interpret quantifications based on mean statistics such as roughness function. In this study, we explore the mechanisms that modulate the turbulence structure over a two-dimensional (2D) sinusoidal wavy surface with a fixed amplitude, but varying slope. To accomplish this, we model the turbulent flow between two infinitely wide 2D wavy plates at a friction Reynolds number, $Re_{\tau}=180$. We pursue two different but related flavors of analysis. The first one adopts a roughness characterization flavor of such wavy surfaces. The second one focuses on understanding the non-equilibrium near surface turbulence structure and their impact on roughness characterization. Analysis of the different statistical quantifications show strong dependence on wave slope for the roughness function indicating drag increase due to enhanced turbulent stresses resulting from increased production of vertical velocity variance from the surface undulations.

This study was conducted to improve the white index (WI) by preparing thermally expandable microspheres (TEMs) for wallpaper. The thermal properties, foam expansion ratio and WI were studied depending on the particle size of colloidal silica in the preparation of TEMs. As a result, the TEMs with small particles of colloidal silica showed the best results for whiteness and yellowing. Additionally, TGA results indicated that it was highly possible that colloidal silica with small particle sizes was physically or chemically attached to the surface of the TEMs that led to an improvement in whiteness at high temperatures.

Photodynamic inactivation (PDI) combines the nontoxic photosensitizer (PS) and visible light to generate reactive oxygen species (ROS), which can cause oxidative damages in (on) microbial organisms. Previously, we have shown that chitosan can augment the bactericidal efficacy mediated by PDI against bacteria and Candida. In this study, we showed that the antimicrobial action of chitosan to augment PDI relates to the enlargement of cell surface destruction. The microbial cell surfaces exhibit severe irregular shapes after PDI in the presence of chitosan. Furthermore, increase in the concentrations or incubation time of chitosan significantly reduce the amounts of photosensitizers TBO required, indicating that chitosan could be a synergistic agent with PDI against human pathogens. A prolonged lag phase was found in PDI surviving microbial cells, in which chitosan can act to synergistically eradicate the cells. Once the impaired cells rebuild their cellular functions from PDI-induced damage, the increased cytotoxic effect of chitosan disappeared. Together, our results suggest that chitosan with an augmented bactericidal activity after PDI is to inhibit the rehabilitation of PDI surviving cells, leading to microbial death.

The ability to detect the presence as well as classify the activities of individuals behind visually obscuring structures is of significant benefit to police, security and emergency services in many situations. This paper presents the analysis from a series of experimental results generated using a through-the-wall (TTW) Frequency Modulated Continuous Wave (FMCW) C-Band radar system named Soprano. The objective of this analysis was to classify whether an individual was carrying an item in both hands or not using micro-Doppler information from a FMCW sensor. The radar was deployed at a standoff distance, of approximately 0.5 m, outside a residential building and used to detect multiple people walking within a room. Through the application of digital filtering, it was shown that significant suppression of the primary wall reflection is possible, significantly enhancing the target signal to clutter ratio. Singular Value Decomposition (SVD) signal processing techniques were then applied to the micro-Doppler signatures from different individuals. Features from the SVD information have been used to classify whether the person was carrying an item or walking free handed. Excellent performance of the classifier was achieved in this challenging scenario with accuracies up to 94%, suggesting that future through wall radar sensors may have the ability to reliably recognize many different types of activities in TTW scenarios using these techniques.

The accompanying mechanical behavior during the flow of ore-rock dispersion is an important factor leading to the instability and failure of the orepass in the ore storage section. It is of great significance to accurately understand the instability characteristics of the well wall in the ore section, during the ore drawing process by understanding the flow characteristics and internal mechanical transfer mechanism of ore-rock dispersion. Using the discrete element method to analyze the flow characteristics, contact compactness, stress distribution characteristics and contact force probability distribution of the ore-rock dispersion, it realizes the quantitative characterization of the damage degree of ore-rock flow, and reveals the damage mechanism of mine storage wall. The results show that: (1) The friction between ore-rock particles, between ore-rock and well wall is an important reason for the change of the macroscopic flow form of ore-rock from “one” to “V” type. (2) The compactness and stress concentration of ores are increasing with the increase of storage depth, which is shown that with the increase of storage depth, the greater the probability of well wall instability. (3) The probability distribution of contact force strength between particles decreases exponentially in the whole process of ore drawing, in which the strong chain plays a major role in the stability of the dispersion system. (4) The number of overpressure and the overpressure coefficient can be used to quantitatively characterize the wall damage degree under the action of ore-rock flow. The dynamic side pressure decreases periodically in the form of exponential fluctuations, and the dynamic load formed by the ore-rock flow mainly played a role in the lower area of the ore storage section.

Global demand for blueberries has surged in recent years, primarily due to their health benefits. However, blueberries postharvest losses, particularly firmness loss and decay, remain significant challenges. This study applied high concentrations of CO2 (15% and 20%) for 3 days at 1 °C to highbush (Duke) and rabbiteye (Ochlockonee) blueberries to assess their effects on maintaining quality, especially firmness, during low-temperature storage. Various quality parameters were evaluated, including titratable acidity, pH, total soluble solids, weight loss, and decay. The impact of gaseous treatments on firmness was analysed through mechanical parameters and the expression of genes related to cell wall integrity (XTH23, PL8, PG, PM3, EXP4, and VcGH5). The effectiveness of the treatments varied between species. High CO2 levels successfully reduced decay in both cultivars, but only the highbush variety (Duke) showed improvements in firmness. In Duke, CO2 treatments influenced the expression of genes associated to cell wall integrity, such as XTH23, PL8, and GH5. In contrast, the roles of PG and PME in firmness maintenance were minimal, showing no significant differences between treatments. While CO2 did not enhance firmness in Ochlockonee, it effectively reduced weight loss, highlighting the need for tailored postharvest strategies for different blueberry cultivars.

The complex structure of the plant cell wall makes it difficult to use the biomass produced by biosynthesis. For this reason, the search for new strains of microorganisms capable of efficiently degrading fiber is a topic of interest. For these reasons, the present study aimed to evaluate both the microbiological and enzymatic characteristics of the fungus Curvularia kusanoi L7strain. For this, its growth in different culture media was evaluated. Wheat straw mineralization was evaluated by gas chromatography assisted by infrared spectroscopy. The production of endo- and exoglucanase, laccase and peroxidase enzymes in submerged solid fermentation of wheat and sugarcane bagasse were characterized. The strain efficiently mineralized raw wheat straw, showing a significant decrease in signals associated with cellulose, hemicellulose and lignin in the infrared spectra. High enzyme productions were achieved in submerged solid fermentation of both substrates, highlighting the high production of endoglucanases in sugarcane bagasse (2.87 IU/mL) and laccases in wheat (1.64 IU/mL). It is concluded that C. kusanoi L7 is an ascomycete with a versatile enzyme production that allows it to exhaustively degrade complex fibers such as raw wheat straw and sugar cane bagasse, making it a microorganism with great potential in the bioconversion of plant biomass.

Lignin is nature's second most abundant vascular plant biopolymer, playing significant roles in mechanical support, water transport, and stress responses. This study identified 90 lignin biosynthesis genes in the rice based on phylogeny and motif constitution, and they belong to PAL, C4H, 4CL, HCT, C3H, CCoAOMT, CCR, F5H, COMT, and CAD families. Duplication events contributed largely to the expansion of these gene families, such as PAL, CCoAOMT, CCR, and CAD families mainly attributed to tandem and segmental duplication, respectively. Microarray data of 33 tissue samples covering the entire life cycle of rice suggested, fairly high PAL, HCT, C3H, CCoAOMT, CCR, COMT, and CAD gene expression, and rather variable C4H, 4CL, and F5H expression. Some members of lignin-related genes (OsCCRL11, OsHCT1/2/5, OsCCoAOMT1/3/5, OsCOMT, OsC3H, OsCAD2, and OsPAL1/6) were expressed in all tissues examined. The expression patterns of lignin-related genes can be divided into two major groups with eight subgroups, each showing a distinct co-expression in tissues representing typically primary and secondary cell wall constitutions. Some lignin-related genes were strongly co-expressed in tissues typical of secondary cell walls. The combined HPLC analysis showed increased lignin monomers (H, G, and S) in five genotypes from young to old growth stages. Based on 90 genes' microarray data, 27 genes were selected for qRT-PCR gene expression analysis. Four genes (OsPAL9, OsCAD8C, OsCCR8, and OsCOMTL4) were significantly negatively correlated with lignin monomers. Furthermore, eleven genes were co-expressed in certain genotypes during secondary growth stages. Among them, six genes (OsC3H, OsCAD2, OsCCR2, OsCOMT, OsPAL2, and OsPAL8) were overlapped with microarray gene expressions, highlighting their importance in lignin biosynthesis.

Selenium nanoparticles (SeNPs) can mitigate cadmium (Cd2+) toxicity in crop cultivation and aquaculture. This study investigates the impact of SeNPs on Cd-induced toxicity in Chlamydomonas reinhardtii, a model organism, aiming to mitigate Cd damage in crops. SeNPs were synthesized using sodium carboxymethyl cellulose (CMC) as a dispersant and vitamin C to reduce H2SeO3. Characterization by transmission electron microscopy (TEM) and nanoparticle granulator revealed uniform, stable spherical SeNPs. Cd2+ and SeNPs of varying concentrations were introduced into culture media of wild type (CC125mt+) and cell wall defect type (CC400mt+) Chlamydomonas reinhardtii. Growth curves, photosynthetic pigment content, enzyme activities (T-SOD and CAT), MDA content, and ROS levels were assessed. Fluorescence induction kinetics, QA oxidation kinetics, and Cd2+ content in cells were measured to evaluate SeNPs' effect on Cd toxicity. Results showed SeNPs exacerbated Cd2+ toxicity in CC125mt+ by inhibiting growth and increasing catalase activity, indicating enhanced oxidative damage. SeNPs intensified PSII damage by Cd2+ and increased intracellular Cd2+ content, possibly exacerbating toxicity. SeNPs had no effect on Cd2+ toxicity in CC400mt+. Overall, SeNPs exacerbated Cd2+ toxicity in wild type CC125mt+ but not in cell wall defective CC400mt+. These findings caution against indiscriminate use of nano-selenium to mitigate Cd damage, stressing the importance of careful target selection, dosage, and range considerations.

(1) Background: This paper illustrates an innovative methodological approach chosen to study and map the colors of the Medieval wall painting of Santa Maria Antiqua in the Roman forum, one of the pilot sites of the EHEM project (Enhancement of Heritage Experiences: The Middle Ages. Digital Layered Models of Architecture and Mural Paintings over Time); (2) Methods: We chose to use two methods for gathering information about colors and mapping, specifically colorimetry by spot measurements and hypercolorimetric multispectral imaging (HMI) to map those same colors sampled through colorimetry; (3) Results: Chromatic data of all colors in the wall paintings were obtained in the CIELAB color space. In those same points, chromatic similarity maps were performed by the innovative HMI system, a multispectral imaging technique able to obtain also color data information by means of the advanced calibration software named SpectraPick®: this allowed to have a complete knowledge of the color characteristics and distribution; (4) Conclusion: The color measurements and mapping was an important result to improve the reading of Medieval wall paintings, fragmentary and stratigraphically complex, giving a new light in the knowledge of the colors and allowing to better understand the original appearance of the iconographic patterns, reconstructed with colors as faithfully as possible similar to the originals.

Tissue engineered heart valves can grow, repair, and remodel after implantation, presenting a more favorable long-term solution compared to mechanical and porcine valves. Achieving functional engineered valve tissue requires the maturation of human cells seeded onto valve scaffolds under favorable growth conditions in bioreactors. The mechanical stress and strain on developing valve tissue caused by different pressure and flow conditions in bioreactors are currently unknown. The aim of this study is to quantify the wall shear stress (WSS) magnitude in heart valve prostheses under different valve geometries and bioreactor flow rates. To achieve this, the study used fluid-structure interaction simulations to obtain the valve's opening geometries during the systolic phase. These geometries were then used in computational fluid dynamics simulations with refined near-wall mesh elements and ranges of prescribed inlet flow rates. The data obtained included histograms and regression curves that characterized the distribution, peak, and median WSS for various flow rates and opening configurations. The study also found that the upper region of the valve near the commissures experienced higher WSS magnitudes than the rest of the valve.

The centrifugal force field in the hydrocyclone was affected by the concave-wall curvature radius R0, the mechanism of droplet deformation was closely related to the mass transfer efficiency. Numerical simulation and experimental data were collected to reveal the deformation characteristics and mechanism of single droplet crossing concave-wall jet. Normalized interfacial energy γ and stretching performance were provided to investigate the droplet deformation process. The results showed that the droplet was stretched along the streamwise and shrank along the spanwise in the concave-wall jet. The droplet interfacial energy and deformation were the largest when the droplet crossed the jet boundary at t = 0.20 s. Maximum γ value increased with the increase of R0 by 57.3% to 71.4%, and the distance between droplet and concave-wall increased with R0. Q-criterion was exported to show the vortex strength increasing as the decrease of R0 in the jet boundary. The pressure distribution inside the droplet showed that the pressure decreased as R0 increased, while the pressure difference increased along the streamwise and wall-normal direction. The study suggested that the droplet breakup was more difficult for the smaller R0, which was beneficial for liquid-liquid heterogeneous separation.

Candida tropicalis, an opportunistic pathogen, ranks among the primary culprits of invasive candidiasis, a condition notorious for its resistance to conventional antifungal drugs. The urgency to combat these drug-resistant infections has spurred the quest for novel therapeutic compounds, with a particular focus on those of natural origin. In this study, we set out to evaluate the impact of Isoespintanol (ISO), a monoterpene derived from Oxandra xylopioides, on the transcriptome of C. tropicalis. Leveraging transcriptomics, our research aimed to unravel the intricate transcriptional changes induced by ISO within this pathogen. Our differential gene expression analysis unveiled 186 differentially expressed genes (DEGs) in response to ISO, with a striking 85% of these genes experiencing upregulation. These findings shed light on the multifaceted nature of ISO's influence on C. tropicalis, spanning a spectrum of physiological, structural, and metabolic adaptations. The upregulated DEGs predominantly pertained to crucial processes, including ergosterol biosynthesis, protein folding, response to DNA damage, cell wall integrity, mitochondrial activity modulation, and cellular responses to organic compounds. Simultaneously, 27 genes were observed to be repressed, affecting functions such as cytoplasmic translation, DNA damage checkpoints, membrane proteins, as well as metabolic pathways like trans-methylation, trans-sulfuration, and trans-propylamine. These results underscore the complexity of ISO's antifungal mechanism, suggesting that it targets multiple vital pathways within C. tropicalis. Such complexity potentially reduces the likelihood of the pathogen developing rapid resistance to ISO, making it an attractive candidate for further exploration as a therapeutic agent. In conclusion, our study provides a comprehensive overview of the transcriptional responses of C. tropicalis to ISO exposure. The identified molecular targets and pathways offer promising avenues for future research and the development of innovative antifungal therapies to combat infections caused by this pathogenic yeast.

It is generally accepted that the human abdominal wall is comprised of skin, subcutaneous tissues, muscles and their aponeuroses, and the parietal peritoneum. Understanding these layers and their mechanical properties provides valuable information to those designing procedural skills trainers, supporting surgical procedures (hernia repair), and engineering-based work (in silico simulation). However, there is little literature available on the mechanical properties of the abdominal wall in layers or as a composite. This work characterizes the tensile properties of the human abdominal wall by layer and as a partial composite. Tissues were collected from fresh-never-frozen and fresh-frozen cadavers and tested in uniaxial tension at a rate of 5 mm/min until failure. Stress-strain curves were created for each sample, and the values for elastic moduli, ultimate tensile strength, and strain at failure were obtained. The experimental outcomes from this study demonstrated variation of tensile properties within and between tissues. The data also suggest that the tensile properties of composite abdominal walls are not additive. Ultimately, this body of work contributes to a deeper comprehension of these mechanical properties and will serve to enhance patient care, refine surgical interventions, and assist with more sophisticated engineering solutions.

In recent years, with the increase in maritime trade, the necessity of increasing the capacities of the ports has emerged. However, while it is planned to increase the capacities of the ports, it is important that the port continues to operate at the same time. In this respect, the old port structures should not be damaged during the capacity increase. In this study, the strengthening of a port in Guinea is discussed as a case study. In the study, the existing quay wall was evaluated, and geotechnical and structural alternatives of the new structure to be built for capacity increase were evaluated. A combined system was designed as a pile foundation and a reinforced foundation with plastic piles so as not to damage the existing quay wall. The pile capacities obtained as a result of the analyses were verified by loading tests.

The present study provides new insights into the growth of brown algal cell wall by showing that cell wall polysaccharide plays an important role in the process of growth consider- ing the physicochemical characteristic of young and old Cladosiphon okamuranus. To determine its structural variation in detail, the cell wall was sequentially fractionated into five fractions: hot water (HW), ammonium oxalate, hemicellulose-I (HC–I), HC-II, and cellulose, and analyzed physicochemically. Results showed that almost 80% of the total recovery cell wall from both young and old thalli was HW and HC-I contained mainly fucoidan composed of Fuc, GlcA, and sulfate in molar ratios of 1.0: 0.3: 0.6~0.7 and 1.0: 0.3: 0.2~0.3, respectively. Fucoidan in HW was a highly sulfated matrix polysaccharide abundance in young thalli, while fucoidan in HC-I was rich in old thalli and functions as hemicellulose in land plants, crosslink with cellulose and strengthen the cell wall. We found that HW and HC-I were particularly involved in the growth and strength of old thalli appeared to be due to the deposition of HC-I and reducing of water content during the growth process.

The most common method for establishing bioequivalence (BE) is to demonstrate similarity of concentration-time profiles in the systemic circulation, as a surrogate to the site of action. However, the similarity, or otherwise, of profiles from two different formulation in the gastrointestinal tract (GIT) may not be associated with same conclusions based on systemic circulation. Here, we have explored the concordance of BE conclusions (or otherwise) for a set of hypothetical formulations based on drug concentration-profiles in various segments of gut vs those in systemic circulation using virtual trials powered by physiologically-based (PBPK) models. Moreover, by changing physiological and biological attributes of the GIT, the impact of Crohn’s Disease on the BE conclusions was explored. The virtual BE (VBE) was applied to an Entocort EC, a pH/time-dependent release formulation of budesonide, as model drug that is targeted at ileum and colon. Substantial “discordance” between the local and systemic outcomes of VBE was observed. Upper GIT segments were much more sensitive to formulation changes than systemic circulation, where the latter led to false conclusions for BE. The ileum and colon showed a lower frequency of discordance with systemic-derived VBE. Nonetheless, a higher f2 value than the default similarity standard (50) was required to achieve local BE. In the case of Crohn’s Disease, a product-specific f2 value might be needed for drugs such as Entocort EC to ensure local BE, knowing the insensitivity of systemic data to show lack of BE. The results, being limited to the single model drug product, Entocort EC, may not provide a a general and conclusive picture. However, for the first time, it demonstrated the potential discordances between the local gut vs systemic BE.

Common bean has received attention as a model plant for legume studies, but little information is available about the morphology of its pods and the relation of this morphology to loss of seed dispersal and/or the pod string, which are key agronomic traits of legume domestication. Dehiscence is related to pod morphology and the anatomy of pod tissues because of the weakening of the dorsal and ventral dehiscence zones and the tensions of the pod walls. These tensions are produced by the differential mechanical properties of lignified and non-lignified tissues and changes in turgor associated with fruit maturation. In this research, we histologically studied the dehiscence zone of the ventral and dorsal sutures of the pod in two contrasting genotypes for dehiscence and string, by comparing different histochemical methods with autofluorescence. We found that the secondary cell wall modifications of the ventral suture of the pod were clearly different between the dehiscence-susceptible and stringy PHA1037 and the dehiscence-resistant and stringless PHA0595 genotypes. The susceptible genotype had cells of bundle caps arranged in a more easily breakable bowtie knot shape. The resistant genotype had a larger vascular bundle area and fibre cap cells (FCCs), and due to their thickness, the external valve margin cells were significantly stronger than those from PHA1037. Our findings suggest that the FCC area, and the cell arrangement in the bundle cap, might be a partial structure involved in the pod dehiscence of common bean. The autofluorescence pattern at the ventral suture allowed us to quickly identify the dehiscent phenotype and gain a better understanding of cell wall tissue modifications that took place along the bean's evolution, which had an impact on crop improvement. We report a simple autofluorescence protocol to reliably identify secondary cell wall organization and its relationship to dehiscence and string in common bean.

This paper presents a low-cost C-band frequency-modulated continuous wave (FMCW) radar system for use in indoor through-wall metal detection. Indoor remote-sensing applications, such as through-wall detection and positioning, are essential for the comprehensive realization of the internet of things or super-connected societies. The proposed system comprises a two-stage radio-frequency power amplifier, a voltage-controlled oscillator, circuits for frequency modulation and system synchronization, a mixer, a 3-dB power divider, a low-noise amplifier, and two cylindrical horn antennas (Tx/Rx antennas). The antenna yields gain values in the 6.8 ~ 7.8 range when operating in the 5.83 ~ 5.94 GHz frequency band. The backscattered Tx signal is sampled at 4.5 kHz using the Arduino UNO analog-to-digital converter. Thereafter, the sampled signal is transferred to the MATLAB platform and analyzed using a customized FMCW radar algorithm. The proposed system is built using commercial off-the-shelf components, and it can detect targets within a 56.3 m radius in indoor environments. In this study, the system could successfully detected targets through a 4-cm-thick ply board with a measurement accuracy of less than 10 cm.

The study utilizes the energy-flux-vector method to analyze the heat transfer characteristics of natural convection in a wavy-wall porous square cavity with a partially-heated bottom surface. The effects of the modified Darcy number and modified Rayleigh number on the energy-flux-vector distribution and mean Nusselt number are examined. The results show that when a low modified Darcy number with any value of modified Rayleigh number is given, the recirculation regions are not formed in the energy-flux-vector distribution within the porous cavity. Therefore, a low mean Nusselt number is obtained. The recirculation regions do still not form and thus the mean Nusselt number has a low value when a low modified Darcy number with a high modified Rayleigh number is given. However, when the values of the modified Darcy number and modified Rayleigh number are high, the energy flux vectors generate recirculation regions and thus a high mean Nusselt number is obtained.

A numerical study of the flow over a NACA aerofoil is presented in this paper. The numerical simulations are achieved with the computer code CFX and the computational domain is created by the computer tool ANSYS ICEM CFD. The CFX code is based on the finite volume method to solve the equations of mass, momentum and energy. The purpose of this paper is to determine the pressure distribution, flow patterns and the forces acting on the airfoil. Effects of the attack angle and Reynolds number on the velocity and pressure distribution, on the lift and drag coefficients are also explored.

The study examines the behavior of the Mechanically Stabilized Earth (MSE) wall using available reinforcement materials in Danang, Vietnam. The MSE was reinforced by the self-fabricated galvanized steel grids using CB300V steel with 3 cm ribs. The backfill soil is a sandy clay soil from the local area with a low cohesion. A full-scale model with full instrumentation was installed to investigate the distribution of tensile forces along the reinforcement layers. The highest load that caused the wall to collapse due to internal instability (reinforcement rupture) was 302 kN/m2, which is 15 times the design load of 20 kN/m2. The failure surface within the reinforced soil had a parabolic sliding shape which is similar to the theoretical studies. At the failure load level, the maximum lateral displacement at the top of the wall facing was small (3.9 mm) which is significantly lower than the allowable displacements for the retaining wall. Furthermore, a numerical model using FLAC software was applied to simulate the performance of the MSE wall. The modeling results are in good agreement with the physical model. Thus, the self-fabricated galvanized steel grids could be confidentially used in combination with the local backfill soil for the MSE walls.

This paper studies the dynamic response of corner supported modular steel buildings with a core wall system, under progressive collapse scenarios, associated with corner module removals. Since using secondary systems such as concrete core in mid- to high-rise buildings is currently una-voidable, understanding their impact on load transfer between modules during collapse scenarios becomes essential. The designated buildings with four-, eight- and twelve-story were modelled using the macro-model based finite element method in Abaqus. Also, three different locations are considered for the concrete shear core within the building plan, leading to nine various case scenarios. Each vertical and horizontal inter-module connection was modelled by one axial and two shear springs with predefined nonlinear force-displacement behaviour. The local and global buckling, which plays an essential role in the building's stability, was considered to obtain accurate results. Finally, parametric studies on the building response were carried out, including the intra-module connection rigidity and inter-module connection stiffness. The results demonstrated that the core wall could maintain the robustness of modular steel building through two mechanisms dependent on its location within the plan. Also, preventing plastic hinges from forming in beams could be introduced as an anti-collapse mechanism in the corner module removal scenarios

Soil cadmium (Cd) contamination has become a serious problem in China. In this study, the goals of the field experiments were to understand how the application of nitrogen fertilizer at the full heading stage and filling stage reduced the uptake of Cd in rice by affecting the distribution of Cd in iron plaques on the root surfaces and subcellular in the root and flag leaf. The hydroponic culture experiments aimed to explore the effect of interaction or deficiency of nitrogen and cadmium on cadmium accumulation in rice at the late growth stage. The results showed that under the condition of having enough nitrogen supply during the early growth stage, applying nitrogen fertilizer during the full heading stage and filling stage resulted in a significant increase in the concentration of Fe and Cd on the root plaques at the milk stage and mature stage. Additionally, it increased the concentration and proportion of Cd in the soluble fraction of subcellular in the flag leaves at the milk stage. On the other hand, when there was a deficiency of nitrogen, the concentration of Fe in the root plaques increased significantly, while the concentration of Cd in the root plaques decreased significantly. Moreover, the proportion of Cd in the flag leaf cell walls increased significantly. Regardless of whether there was sufficient or deficient nitrogen supply during the early growth stage, applying nitrogen at the full heading stage reduced the Cd concentration in brown rice by 35.11% and 57.25%, respectively. Under hydroponic culture conditions with both Cd exposure and later-stage nitrogen supply significantly increased the Cd concentration in brown rice. However, in the absence of Cd, nitrogen supply significantly reduced the Cd concentration in brown rice. The accumulation of Cd in brown rice was significantly correlated with the accumulation of Fe and Mn at the late growth stage. Our findings indicate that when there is sufficient nitrogen nutrition during the early growth stage of rice, topdressing nitrogen fertilizer at the full heading stage combined with lime with mineral element fertilizers such as Fe and Mn is an effective approach to reduce the presence of cadmium in rice grains.

Amidst the strong demand for wood-based products, and the clamor of environmental consciousness, more functional and green solutions arise to meet both of these goals. Pursuant to this, the study looked into the potential of a bio-waste taken from Cocos nucifera tree known as phellem—the tree’s outermost skin made of dead tissues, to be a main component of a new construction material. This study builds on existing research showing phellem is impermeable, buoyant, elastic, has a slow burn rate, and repels water during precipitation, which supports the hypothesis that phellem may be a potential substitute material for dry wall boards. To test this, three (3) formulations were developed as experimental setups to test the physico-mechanical properties of coconut palm phellem (COPAP): Formulation A (40% COPAP and 60% cement); Formulation B ( 25% COPAP and 75% cement); and lastly, Formulation C that was (10% COPAP and 90% cement). The study tested (1) which among these formulations exhibited the best physico-mechanical properties, then it (2) compared the formulations with common dry wall products available in the market, and subsequently, (3) identified the possible architectural applications of COPAP board as a new building material. Of the three setups, Formulation C exhibited the best physico-mechanical properties in terms of compressive and flexural strength, fire resistivity, and water absorption. It fared better than the particle board in terms of compressive strength, better than wafer and particle board in fire resistivity, performed best in the water absorption test, and was almost equivalent in flexural strength to fiber cement board. With these findings, the study concluded that COPAP board can be used for projects that entail a high number of interior partitions, such as offices, BPO firms and commercial stalls in shopping malls, etc. The findings were conclusive only for interior applications, as the results clearly suggest that the COPAP boards are not load-bearing. Future investigations may explore and assess other properties of COPAP boards like thermal and sound insulation, resistance to rot and other properties that may prove its competitive advantage as a construction material.

The buildup of plaque in the arteries characterizes atherosclerosis, which causes the walls of the arteries to thicken, the lumen to narrow, and the wall to thin in certain areas. These changes can lead to alterations in blood flow, potentially resulting in aneurysms and heart attacks if left untreated. This paper presents a phenomenological model to explain the mechanics of plaque rupture in stenosed bifurcated elastic arteries. The model considers the interaction between the plaque and artery wall, blood flow, mechanical properties of the artery wall and plaque, and hemodynamic forces in the system. Using the Navier-Stokes equations to describe blood flow and elastic properties of artery walls, our study shows that blood flow can become turbulent, leading to backflow, vortices, and possible stagnation. Certain regions can become highly vulnerable and result in elevated heat transfer between blood and arterial walls, which can lead to the rupture of the plaque cap. The study focuses on blood flow features such as velocity profiles and wall displacement on fluid-structure interaction, which are consistent with the literature. Finally, we calculate the wall shear stress (WSS) for minimum and maximum times while considering elastic walls. Our findings may provide valuable insights into the mechanisms of plaque rupture and inform the development of improved diagnostic and therapeutic approaches.

Advanced manufacturing techniques, aimed at implants with high dependability, flexibility, and low manufacturing costs, are crucial in meeting the growing demand for high-quality products like biomedical implants. Incremental sheet forming is a promising flexible manufacturing approach for rapidly prototyping sheet metal components using low-cost tools. Titanium and its alloys are used to shape most biomedical implants because of its superior mechanical qualities, biocompatibility, low weight, and great structural strength. The poor formability of titanium sheets at room temperature, however, limits their widespread use. The goal of this research is to show that gradual sheet formation of a titanium biomedical implant is possible. The possibility of creative and cost-effective concepts for the manufacturing of such complicated shapes with significant wall angles is explored in this study. A numerical simulation based on finite element modeling as well as a design process tailored to metal forming is used to complete the development. The mean of uniaxial tensile tests with a constant strain rate was used to study the flow behavior of the studied material. To forecast the crack, the obtained flow behavior was modeled using the behavior model and failure model.

With the increased use of simulation based training using animal models for the education of surgical and anaesthetic techniques, an increased understanding of the anatomy of such models and how they compare to humans is required. The transversus abdominis plane block is a regional anaesthetic technique that requires an understanding of the abdominal wall anatomy along with proficient ultrasound use. The current review aims to compare the anatomy of the abdominal wall across species, particularly focussing on the pertinent differences within the class of mammals, and secondarily, it aims to address the implications of these differences for simulation based training of the transversus abdominis plane block. To achieve this, the PubMed, Web of Science and Google Scholar databases were searched for relevant literature. The mammalian abdominal wall differs in its musculature, vasculature or innervation from that of amphibians, birds or reptiles, however, among species of mammals, the structure of the abdominal wall follows a similar framework. Particular differences among mammals include the additional muscular layer of the panniculus carnosus found in most mammals other than humans, the variable arterial origins and dominant vascular supply of the abdominal wall and the number of thoracolumbar nerves innervating the abdominal wall. When using animal models for simulation based training, the pig is recommended for the transversus abdominis plane block given its closely homologous abdominal wall structure, availability and larger comparative size.

Joints between walls are very important for structural analysis of each masonry building at the global and local level. This issue was often neglected in case of traditional joints and relatively squat walls. Nowadays the issue of wall joints is becoming particularly important due to the continuous strive for simplifying structures, introducing new technologies and materials. Eurocode 6 and other standards (USA, Canadian, Chinese, and Japanese) recommend inspecting joints between walls, but no detail procedures have been specified. This paper presents our own tests on joints between walls made of autoclaved aerated concrete (AAC) masonry units. Tests included reference models composed of two wall panels joined perpendicularly with a masonry bond (6 models), traditional steel and modified connectors (12 models). A shape and size of test models and structure of a test stand were determined on the basis of the analysis of the current knowledge, pilot studies and numerical analyses of FEM. The analysis referred to the morphology and failure mechanism of models. Load-displacement relationships for different types of joints were compared and obtained results were referred to results for reference models. A mechanism of cracking and failure was found to vary, and clear differences in behaviour and load capacity of each type of joints were observed. Individual working phases of joints were determined and defined, and the empirical approach was suggested to determine forces and displacement of wall joints.

In most studies reported in literature, a flat, constant coefficient is used to convert the amount of optical power required at the input of an optical link into the effective power consumption. This constant coefficient approach is very simple to apply but not accurate, since laser efficiencies are dependent on the optical output power in addition to the operating temperature. In this paper the relationship between laser wall plug efficiency and optical power output is investigated and the effect of this variance on optical links and on-chip laser source electric power consumption is presented

Morphogenesis remains a riddle, wrapped in a mystery, inside an enigma. It remains a formidable problem viewed from many different perspectives of morphology, genetics, and computational modelling. We propose a biochemical reductionist approach that shows how both internal and external physical forces contribute to plant morphogenesis via mechanical stress-strain transduction from the primary cell wall tethered to the plasma membrane by a specific arabinogalactan protein (AGP). The resulting stress vector with direction defined by Hechtian adhesion sites, has a magnitude of a few picoNewtons amplified by a hypothetical Hechtian growth oscillator. This paradigm shift involves stress activated plasma membrane Ca2+channels and auxin-activated H+-ATPase. The proton pump dissociates periplasmic AGP-glycomodules that bind Ca2+. Thus, as the immediate source of cytosolic Ca2+ an AGP-Ca2+ capacitor directs vectorial exocytosis of cell wall precursors and auxin efflux (PIN) proteins. In toto these components comprise the Hechtian Oscillator and also the gravisensor. Thus interdependent auxin and Ca2+ morphogen gradients account for the predominance of AGPs. The size and location of a cell surface AGP-Ca2+ capacitor is essential to differentiation and explains AGP correlation with all stages of morphogenetic patterning from embryogenesis to root and shoot. Finally, evolutionary origins of the Hechtian Oscillator in the unicellular Chlorophycean algae reflect the ubiquitous role of chemiosmotic proton pumps that preceded DNA at the dawn of life.

Multiple engineering projects have confirmed that hydraulic machinery operating in sediment-laden rivers suffers from sediment abrasion. Guide vanes are among the most severely worn flow-passing components and have long been a key research focus in hydraulic machinery. This study conducted wear tests using asymmetric incoming flow with a 10° impact angle and an NACA0012 aluminum alloy airfoil as the research object. The development of wear was analyzed, revealing that it primarily occurs at the airfoil’s leading edge and progresses through three stages: initiation, development, and stabilization. The Finnie model was used to analyze the initial wear mechanism, with calculated results closely matching the latest wear outcomes. In the initial stage, wear rate density was influenced by particle impact velocity, angle, volume fraction, and y-direction shear stress. A low-velocity zone near the impact point, combined with rebounding particles causing secondary impacts, increases particle volume fraction and wear rate density. These secondary impacts are the primary causes of wear on both the upstream and downstream surfaces. Furthermore, flow separation downstream of the leading edge makes this region highly susceptible to wear. This study provides valuable insights for addressing wear in hydraulic machinery for practical engineering applications.

Bicarbonate and CO2 are essential substrates for carboxylation reactions in bacterial central metabolism. In Staphylococcus aureus, the bicarbonate transporter, MpsABC (membrane potential-generating system) is the only carbon concentrating system. A mpsABC deletion mutant can hardly grow in ambient air. In this study, we investigated the changes that occur in S. aureus when it suffers from CO2/bicarbonate deficiency. Electron microscopy revealed that mpsABC has a twofold thicker cell wall thickness compared to the parent strain. The mutant was also substantially inert to cell lysis induced by lysostaphin and the non-ionic surfactant Triton X-100. Mass spectrometry analysis of muropeptides revealed the incorporation of alanine into the pentaglycine interpeptide bridge, which explains the mutant’s lysostaphin resistance. Flow cytometry analysis of wall teichoic acid (WTA) glycosylation patterns revealed a significantly lower -glycosylated and higher ß-glycosylated WTA, explaining the mutant’s increased resistance towards Triton X-100. Comparative transcriptome analysis showed altered gene expression profiles. Autolysin-encoding genes such as sceD, a lytic transglycosylase encoding gene, were upregulated, while genes related to cell wall-anchored proteins, secreted proteins, transporters, and toxins were downregulated. Overall, we demonstrate that bicarbonate deficiency is a stress response that causes changes in cell wall composition and global gene expression resulting in increased resilience to cell wall lytic enzymes and detergents.

Oxidative stress constitutes a main molecular mechanism underlying cardiovascular diseases (CVDs). This pathological mechanism can be triggered by NADPH oxidases (NOXs), which produce reactive oxygen species (ROS). In fact, the different NOXs have been associated with myocardial infarction, atherothrombosis, and stroke. More specifically, we will focus on the implications of NOXs in atherothrombotic stroke. Each NOX member participates in a different way in the several stages of this disease: endothelial dysfunction, immune cell infiltration, foam cell genesis, vascular smooth muscle cells (VSMC) proliferation, and atherosclerotic plaque formation. Additionally, some NOXs are involved in plaque instability, thrombosis, ischemic stroke, and ischemia-reperfusion injury (IRI). Interestingly, the effects of NOXs in this pathology depend on the specific homologue, the cell type in which they are activated, and the stage of the disease. In this review we summarize the most up-to-date information about the implications of vascular NOXs in each of these processes. Finally, we highlight some limitations and future perspectives of the study of NOXs in CVDs.

The seed habit is the most complex and successful method of sexual reproduction in vascular plants. It represents a remarkable moment in the evolution of plants that afterwards spread on land. In particular, seed size had a pivotal role as for evolutionary success and agronomic trait, especially in the field of crop domestication. Given that crop seeds constitute one of the primary products for consumption, it follows that seed size represents a fundamental determinant of crop yield. This adaptative feature is strictly controlled by genetic traits from both maternal and zygotic tissues, although seed development and growth are also affected by environmental cues. Despite being a highly exploited topic for both basic and applied research, there are still many issues to be elucidated as for developmental biology as well as for agronomic science. This review addresses a number of open questions related to cues that influence seed growth and size, and how developmental processes are influenced by them. Besides, new insights on the genetic-molecular control of this adaptive trait are presented.

The earth pressure acting on soilbag reinforced retaining structures subjected to surcharge loads under non-limited states, is crucial for designing these structures. In this study, the earth pressures are investigated through model tests of the soilbag reinforced retaining wall. Findings from these tests reveal a non-linear distribution of lateral earth pressure on the wall when subjected to surcharge loads in non-limited states, with an observed escalation in pressure corresponding to increased surcharge loads. Compression tests on soilbags were conducted to analyze their lateral stress-strain behavior. Insights from these tests facilitated the development of a predictive method for estimating lateral pressure on soilbag reinforced retaining walls under similar conditions. This method, rooted in the principle of force equilibrium in differential elements, has its accuracy and reliability substantiated by the alignment of its predictions with the model test outcomes.

Aluminum (Al) toxicity is the most common factor limiting the growth of alfalfa in acidic soil conditions. Melatonin (MT), a significant pleiotropic molecule present in both plants and animals, has shown promise in mitigating Al toxicity in various plant species. This study aims to elucidate the underlying mechanism by which melatonin alleviates Al toxicity in alfalfa through a combined physiological and transcriptomic analysis. The results reveal that 5 μM MT significantly alleviates Al-induced growth inhibition and preserves the morphology of root tip cells. Moreover, MT application reduces Al accumulation in alfalfa roots and shoots by 28.6% and 27.6%, respectively. Additionally, MT plays a crucial role in scavenging Al-induced excess H2O2 by enhancing the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), consequently reducing malondialdehyde (MDA) levels. The RNA-seq results reveal that MT application significantly upregulates the expression of xyloglucan endotransglucosylase/hydrolase (XTH) and carbon metabolism-related genes, including those involved in the glycolysis process, as well as sucrose and starch metabolism, suggesting that MT application may mitigate Al toxicity by facilitating the binding of Al to the cell walls, thereby reducing intracellular Al accumulation, and by improving respiration and the content of sucrose and trehalose. Taken together, our study demonstrates that MT alleviates Al toxicity in alfalfa by restoring redox homeostasis, enhancing Al binding to the cell wall, and modulating carbon metabolism. This research advances our understanding of the mechanisms underlying MT's effectiveness in mitigating Al toxicity, offering potential strategies for reducing Al accumulation in alfalfa.

There is a substantial lack of data relating to the hygrothermal traits of existing buildings and building materials. Moisture analysis in building envelopes is 1D (one-dimensional), and 2D and 3D effects and feedback can be neglected. Many European standards treat a moisture percentage in building envelopes as surface and interstitial moisture condensation, such as BS EN ISO 10211, BS EN ISO 13788, BS EN ISO 15148, and BS EN 15026. In this manuscript, there was a tendency to capture a relative quantity of superficial moisture, by metering spots on the inner and outer surface of wall lining systems with a moisture meter. The metering specimens were the buildings with different envelopes: a wooden hut-house, a masonry building and a building partially sheeted in stone. The referent heights on wall linings, for taking the moisture data were specified as 50 cm for a wooden and masonry building and 80 cm for a stone-sheeted building, from the ground. The obtained numeric results were palpable with no regard to the current state of the weather outside. The listed conclusions highlight the significance of a well-tailored building envelope, and its dependence on weather, junctions, gaskets, appropriate materials, etc.

Heart attacks and strokes are one of the leading causes of death in the world today, and heart attacks caused by clogged arteries that carry blood to the heart muscle are a significant part of these strokes. These are caused by the accumulation of fat particles in the walls of the arteries and the reduction of blood flow through it over a long process. The process of fat penetration in the underlying layers of the Artery wall has been the focus of many researchers, and various researches and Simulations have been done on it, in each of them, the effect of specific parameters has been considered. In the present study, the effect of blood flow rate on the flow pattern in a bifurcate artery with two ducts has been investigated using FLUENT software with Computation fluid dynamic Method. The effect of the angle between the two ducts of the Artery on the flow pattern has been investigated.

The hygrothermal transfer is very important for the design of a building envelope for thermal comfort and economic and energy analysis of the building envelope. The applications of various materials in building envelope have been studied extensively. The study presents several models for the hygrothermal transfer for various building walls. Several energy and mass conservation equations with different boundary conditions and input considerations were presented in this paper for concrete, bricks and wooden walls. The effect of hysteresis was ignored in developing most model equations, while few considered flow pattern of fluid through the wall surfaces. Due to the flexibility of Luikov models, it formed the basis for modelling the coupled heat and mass transfer for porous material independent of hygroscopic nature with different boundary conditions defined according to the geometry and orientations. The influence of type of wall, orientation, thickness, the density of the material and climatic variations on the temperature and moisture evolutions within the building materials was more pronounced. Literature, presenting imaging models using imagery software like COMSOL multi-physics, CFD etc. were scarce considering that microscopic imagery is now deployed to measure the heat and moisture evolution in materials. Future models should include shrinkage or expansion influence on the fibrous material like wood due to their behaviour under environmental condition.

In this paper, we present and analyze a compact inner-wall grating slot microring resonator (IG-SMRR) with the footprint of less than 13 μm × 13 μm on the SOI platform for label-free sensing, which comprises a slot microring resonator (SMRR) and inner-wall grating (IG). Its detection range is significantly enhanced without the limitation of the free spectral region (FSR) owing to the combination of SMRR and IG. Structural parameters of IG and SMRR are investigated and optimized for favorable transmission properties. The simulation results shows that the IG-SMRR has an ultra-large quasi-FSR of 84.6 nm, and the concentration sensitivities of sodium chloride solutions and D-glucose solutions are up to 960.61 pm/% and 933.06 pm/%, respectively. The investigation on the combination of SMRR and IG is a valuable exploration of label-free sensing application for ultra-large detection range and ultra-high sensitivity in future.

Filamentous fungi such as Aspergillus spp. are opportunistic pathogens, which cause highly invasive infections, especially in immunocompromised individuals. Control of such fungal pathogens is increasingly problematic due to the small number of effective drugs available for treatment. Moreover, the increased incidence of fungal resistance to antifungal agents makes this problem a global human health issue. The cell wall integrity system of fungi is the target of antimycotic drugs echinocandins, such as caspofungin (CAS). However, echinocandins cannot completely inhibit the growth of filamentous fungal pathogens, which results in survival/escape of fungi during treatment. Chemosensitization was developed as an alternative intervention strategy, where co-application of CAS with the intervention catalyst octyl gallate (OG; chemosensitizer) greatly enhanced CAS efficacy, thus achieved ≥ 99.9% elimination of filamentous fungi in vitro. Based on hypersensitive responses of Aspergillus antioxidant mutants to OG, it is hypothesized that, besides destabilizing cell wall integrity, the redox-active characteristic of OG may further debilitate fungal antioxidant system.

Cotton fiber is the leading natural textile material, and fiber elongation plays an essential role in the formation of cotton yield and quality. Although a wealth of components in the molecular network controlling cotton fiber elongation have been reported, a lot of players still need to be functionally dissected to understand the regulatory mechanism of fiber elongation comprehensively. In the present study, an R2R3-MYB transcription factor gene, GhMYB201, was characterized and functionally verified via CRISPR/Cas9 mediated gene editing. GhMYB201 was homologous to Arabidopsis AtMYB60, and both coding genes (GhMYB201At and Dt) were preferentially expressed in elongating cotton fibers. Knocking-out of GhMYB201 significantly reduced the rate and duration of fiber elongation, resulting in shorter and coarser mature fibers. It was found that GhMYB201 could bind and activate the transcription of cell wall loosening genes (GhRDLs), and also β-ketoacyl-CoA synthase (GhKCSs) genes to enhance very-long-chain fatty acid (VLCFA) levels in elongating fibers. Taken together, our data demonstrated that the transcription factor GhMYB201s play essential roles in promoting fiber elongation via activating genes related to cell wall loosening and VLCFA biosynthesis.

This paper investigates the influence of microorganisms and clay minerals on the deterioration process of ancient Egyptian murals in the Al-Qurna region, situated on the west bank of Thebes (Luxor), Egypt. To achieve this objective, an extensive examination of building materials (including bedrock, building stone, mortars, and plasters) from various tombs within the study area was conducted. The investigation employed a multidisciplinary approach, combining micro-biological analysis with advanced techniques such as X-ray diffraction (XRD), X-ray fluorescence (XRF), scanning electron microscopy with energy dispersive spectroscopy (SEM-EDS) as well as petrographic examination. These methods were employed to identify the composition of the materials and to assess the masonry structure’s response to deformation caused by endogenous factors, particularly clay minerals. Additionally, mycological analyses were performed using a swabbing cotton sterile technique, revealing the presence of several species, including Ascosphera apis, Aspergillus tamarii, Aspergillus ochraceus, Doratomyces sp, and Eurotium repens. Notably, swelling clays—such as smectite, illite, and chlorite—along with mixed layers of illite-smectite and illite-vermiculite were detected in the studied samples. The presence of such deleterious clay minerals and fungi imperils wall painting preservation, causing discoloration, surface alteration, physical decay, and structural instability. Informed conservation strategies must address these threats for sustained integrity.

When the epipelic algae group growing on the concrete inclined side wall, which is typical of urban rivers and open diversion channels, enters the water flow, it will seriously affect the func-tion of the rivers or the channels. It is urgent to predict the transport capacity of epipelic algae group with water flow. However, the current research on the transport of pollutants is mostly based on mathematical models, and the study of the transport of flake epipelic algae group is extremely rare. A generalized hydrodynamic experimental model of a flume with sidewall epi-pelic algae group was established. Through a series of hydraulic experiments, the physical movement form and transport capacity of the suspended epipelic algae group on the side wall of the channel were studied qualitatively and quantitatively by taking the thickness, sheet size and flow rate of the epipelic algae group as the influencing factors. The key influencing factors of the suspension of epipelic algae are clarified, so as to quantitatively analyze the critical conditions of its suspension. Through flow scheduling, the probability of epipelic algae gathering some-where is increased. It provides a theoretical basis for realizing the long-distance migration of epipelic algae and optimizing the mathematical model.

We presented unusual case of double left ventricular false aneurysm complicating late presentation AMI successfully treated with the combination of urgent surgical drainage of tamponade, staged PCI of non-culprit lesion, preventive implantation of ICD-DR, guideline directed heart failure medical therapy and delayed but effective reconstructive operation.

Aluminum (Al) and manganese (Mn) toxicity are the top two constraints to crop production in acid soil. Crops have evolved common and specific mechanisms to tolerate the two stresses. In the present study, the responses (toxicity and tolerance) of wheat near-isogenic lines (ET8 and ES8) and their parents (Carazinho and Egret) to Al and Mn were compared by determining physiolog-ical parameters and transcriptome profiling of roots. The results showed that: (1) Carazinho and ET8 exhibited dual tolerance to Al and Mn as compared with Egret and ES8, indicated by higher relative root elongation and SPAD; (2) After entering into roots, Al was mainly distributed in the root and fixed in the cell wall, while Mn was mainly distributed in the cell sap and then trans-ported to leaves. Both Al and Mn stresses decreased the contents of Ca, Mg, and Zn; Mn stress al-so inhibited the accumulation of Fe, while Al showed an opposite effect; (3) Transcriptomic analy-sis identified 5581 differentially expressed genes (DEGs) under Al stress and 4165 DEGs under Mn stress. Among these, 2774 DEGs were regulated by both Al and Mn stresses, while 2280 and 1957 DEGs were exclusively regulated by Al and Mn stress, respectively. GO and KEGG analyses indi-cated that cell wall metabolism responds exclusively to Al, while nicotianamine synthesis exclu-sively responds to Mn. Pathways such as signaling, phenylpropanoid metabolism, and metal ion transport showed commonality and specificity to Al and Mn. TFs, such as MYB, WRKY, and AP2 families, were also regulated by Al and Mn, and WGCNA identified PODP7, VATB2, and ABCC3 as the hub genes for Al tolerance and NAS for Mn tolerance. The identified genes and pathways can be used as targets for pyramiding genes and breeding multi-tolerant varieties.

Haemodynamic simulations are increasingly used to study vascular diseases like Intracranial Aneurysms (IA) and to further develope treatment options. However, due to limited data, some aspects must rely on heuristics, especially at the simulation’s distal ends. In literature, Murray’s Law is often used to model the outflow split based on vessel cross-section area, but this poses challenges for the communicating arteries in the Circle of Willis (CoW). In this study, we contribute by assessing the impact of Murray’s Law in patient-specific geometries featuring IA at the posterior communication. We simulate different domain extensions, representing common modelling choices. We establish Full CoW simulations as a baseline to evaluate the effect of these modelling assumptions on haemodynamic indicators, focusing on IA growth and rupture-related factors like Wall Shear Stress (WSS) and Oscillatory Shear Index (OSI). Our findings reveal qualitative alterations in haemodynamics when not modeling posterior communication. Comparisons between computing the anterior circulation and computing the whole Circle of Willis reveal quantitative changes in WSS may reach up to 80%, highlighting the significance of modelling choices in assessing IA risks and treatment strategies.

Abdominal entry is the critical initial step of laparoscopic surgery as it is associated with unexpected complications. Acceptable working space is a prerequisite for safe minimal access surgery. Direct trocar insertion (DTI) technique has been often reported to be time-consuming and capable of causing injuries. We aim to facilitate conventional DTI employing an easy-to-use self-designed abdominal wall-lifting device, OneShot-M. We tested its safety in a porcine model. 18 pigs were randomized into three groups (n=6): open laparoscopy (Group A), DTI (Group B), DTI using OneShot-M (Group C), suitable for a 5-mm trocar. Outcomes were access time, entry-related minor and major complications. Median access time was shorter in C (43 seconds; range: 35-49 seconds), followed by B (82.5; 72-90) and A (190.5; 182-240). Statistically significant differences in procedural time between techniques were detected (p-value = 0.0005). Pairwise comparisons exhibited a statistically significant difference between A and C in terms of access time. Only one difficulty in entry was seen in C; pigs experienced several occurrences in A and B. This pilot study suggests that OneShot-M appears to be a technically feasible, fast and safe method for pneumoperitoneum induction in a porcine model, as it provides sufficient exposure of the operative field without access-related complications. Further clinical studies are needed.

The construction of large-scale water diversion projects alleviated effectively the current situa-tion of uneven distribution of water resources in China. However, due to the siltation of very fine sediment and organic matter on the side wall of the open channel, and the slow velocity of the side wall flow field, it is easy to produce epipelic algae, which affects water quality. For the prototype observation cannot be used to predict the series of flow in real time, and the mathe-matical model calculation is affected by parameter limitations, these two methods often cannot truly reflect the hydrodynamic characteristics of open channel with epipelic algae. Therefore, by referring to the design parameters of the water diversion project channel, this paper takes the epipelic algae growing on the side wall of the open channel as the research object, and uses the scale of 1:30 to carry out the generalized flume experiment. Through the analysis of the physical characteristics of the prototype sample, and the simulation of the cohesive force between the oblique side wall and the epipelic algae, the multi-group and multi-series hydrodynamic tests are carried out. The flow velocity distribution law and the development and change of the tur-bulent vortex on the side wall are analyzed. It has important scientific guiding significance and practical value for water quantity regulation water regulation and water quality safety protec-tion of long-distance projects.

Some biologically active compounds isolated from sea cucumbers stimulate the body’s immune response through activating immune cells. Immune function is closed related to the integrity intestinal barrier and balanced gut microbiota. However, it is unknown whether daily administration of holothurian wall hydrolysates (HWH) ameliorated intestinal dysbiosis and barrier injury induced by immunodeficiency. This study aimed to investigate the immunomodulatory effect and the underlying mechanism of HWH in cyclophosphamide (CTX)-induced immunocompromised mice. BALB/c mice received CTX (80 mg/kg, intraperitoneally) once a day for 3 days to induce immunodeficiency, and then oral administration of HWH (80 or 240 mg/kg), levamisole hydrochloride (LH, 40 mg/kg, positive control) respectively once a day for 7 days. We utilized 16S rRNA sequencing for microbial composition alterations, histopathological analysis for splenic and colonic morphology, Western blotting for expressions of tight junction proteins (TJs), and quantitative real-time (qRT-PCR) for measurements of pro-inflammatory cytokines. HWH attenuated the immune organ damage induced by CTX, increased the secretions of interleukin (IL)-6, IL-1β and tumor necrosis factor (TNF)-α, and promoted the recovery of goblet cells and the productions of TJs (claudin-1, occludin, and ZO-1) in the colon of the immunocompromised mice. Moreover, HWH promoted the growth of beneficial microorganisms such as Lactobacillus, Lachnospiraceae, Christensenellaceae, and Bifidobacterium, while suppressed the populations of Ruminococcus, Staphylococcus, and Streptococcus. These results demonstrate that HWH elicits intestinal mucosal immunity, repairs the damage to intestinal mucosal integrity, and normalizes the imbalanced intestinal microbial profiles in immunocompromised mice. It may be helpful to identify the biological activities of HWH, and supports the potential as new prebiotics, immunomodulatory agents, and medical additive for intestinal repair.

On the basis of the force and safety factor analysis of traditional retaining wall, a new analytical method of force and displacement of slope is suggested, the numerical theoretical solution of the stress distribution of the sliding body can be obtained under the conditions, which the stresses distribution are satisfied with the differential equilibrium equations, the boundary conditions, the compatibility equation and the macroscopic equilibrium equations. The interface stresses between the sliding body and the retaining wall are continuous, the theoretical solution of retaining wall stress distributions can be gotten, while the stress differential equilibrium equation, the compatibility equation, macroscopic force and moment balance equations are satisfied. The strain and displacement solutions can be obtained by using Duncan Chang and Hooke constitutive equations for the slope and the retaining wall respectively. The transfer station of landfill in the Guandukou Town of Badong County is taken as an example, the results of the sliding body and retaining wall analysis show: The stress and strain solutions of a slope and a retaining wall can be obtained by the proposed method, the forces between the retaining wall and the sliding body include shear stress and normal stress, and the stress distributions change with the deformation of the sliding body, the shape and material of retaining wall can be optimized according to the calculation results，it is feasible for the proposed analysis method of slope with retaining wall design to be run many years.

To live in the information society means to be surrounded by billions of electronic devices full of sensors that constantly acquire data. This enormous amount of data must be processed and classified. A solution commonly adopted is to send these data to server farms to be remotely elaborated. The drawback is a huge battery drain due to high amount of information that must be exchanged. To compensate this problem data must be processed locally, near the sensor itself. But this solution requires huge computational capabilities. While microprocessors, even mobile ones, nowadays have enough computational power, their performance are severely limited by the Memory Wall problem. Memories are too slow, so microprocessors cannot fetch enough data from them, greatly limiting their performance. A solution is the Processing-In-Memory (PIM) approach. New memories are designed that are able to elaborate data inside them eliminating the Memory Wall problem. In this work we present an example of such system, using as a case of study the Bitmap Indexing algorithm. Such algorithm is used to classify data coming from many sources in parallel. We propose an hardware accelerator designed around the Processing-In-Memory approach, that is capable of implementing this algorithm and that can also be reconfigured to do other tasks or to work as standard memory. The architecture has been synthesized using CMOS technology. The results that we have obtained highlights that, not only it is possible to process and classify huge amount of data locally, but also that it is possible to obtain this result with a very low power consumption.

Inertial focusing conditions of fluorescent polystyrene spherical particles are studied at the pointwise level along their pathlines. This is accomplished by an algorithm that calculates a de-gree of spreading function of the particles' trajectories taking streaklines images as raw data. Different confinement ratios of the particles and flow rates are studied and the results are pre-sented in state diagrams showing the focusing degree of the particles in terms of their position within a curve of an asymmetric serpentine and the applied flow rate. In addition, together with numerical simulation results, we present empirical evidence that the preferred trajectories of inertially focused spheres are contained within Dean vortices' centerlines. We speculate about the existence of a new force, never postulated before, to explain this fact.

Damage to peripheral arteries by the atherosclerotic process accounts for about 3% of the world's population, which puts this problem in the category of major social problems. Modified lipoproteins and cholesterol crystals are known to accumulate in the arterial intima and cause the formation of proatherogenic macrophages, foam cells, and inflammation. Defective efferocytosis of apoptotic foam cells leads to the formation of a necrotic core, a sign of failure to resolve inflammation. Resolution of inflammation is mediated by specialized pro-resorption lipid mediators, proteins and signaling gases. Improving the balance between pro-inflammatory and anti-inflammatory factors contributes to the reverse development of local atherogenesis. Active direct intervention in the reconstruction of the adventitial layer of the arterial wall using cholesterol-affinity polysaccharide hydrogels containing a cocktail of growth factors creates conditions for the formation of additional extracellular matrix and reversal of cholesterol mass from the subintimal zone. Creating concentration and electrostatic gradients in the adventitia zone using a hydrogel may be one of the effective ways to degrade early soft atherogenic plaques and locally restore the damaged structure of the vessel wall. Growing scientific interest in the previously insufficiently studied adventitia indicates its important role in angionesis and atherogenesis.

In order to study the way how to improve the spatial action of precast monolithic composite floor slabs, and would replace the cast-in-place surface layer for reduce the weight of structure, using pseudo-dynamic tests on 1/4 scale models of two-span and three-story frame structure. The lateral load tests compared the stresses and displacements with cast-in-place floor frame-shear wall structure (SJ1) and precast monolithic floor frame-shear wall structure with X horizontal braces at the bottom of the floor (SJ2). The results show the X horizontal braces can improve the spatial action. Structural integrity (SJ2) as well as the effective transmission of the horizontal force can be ensured by additional X bracing at the bottom of the rigidity of the floor without cast-in-place concrete topping. The results show that X horizontal braces more effectively transfer horizontal, which provides a beneficial reference for similar research.

Data obtained by Direct Numerical Simulations (DNS) of pressure-driven turbulent channel flow are studied in the range 180 ≤ Reτ≤ 10,000. Reynolds number effects on the Mean Velocity Profile (MVP) and second order statistics are analyzed with a view of finding logarithmic behavior in the overlap region or even further from the wall, well in the boundary layer’s outer region. Values of Kármán constant for the MVPs and the Townsend-Perry constants for the streamwise and spanwise fluctuation variances are determined for the Reynolds numbers considered. A data-driven model of the MVP, proposed and validated for zero pressure gradient flow over a flat plate, is employed for pressure-driven channel flow by appropriately adjusting Coles’ strength of the wake function parameter, Π. There is excellent agreement between the analytic model predictions of MVP and the DNS-computed MVP as well as of the Reynolds shear stress profile. The skin friction coefficient Cf is calculated analytically. The agreement between the analytical model predictions and the DNS-based computed discrete values of Cf is excellent.

We present the full case pictorial assay including preoperative imaging as well as the surgical correction of the uterine anomaly and associated complication of a very rare case of occult unicornuate right uterus with rudimentary non-communicating functioning left horn and its natural evolution following an initially incomplete surgical treatment.

Post-operative pain is the unpleasant sensory and emotional experience after surgery, its origin being both the inflammatory reaction induced by the surgical trauma on the abdominal wall, and the splanchnic pain, induced by the activation of nociceptors of the viscera, which are highly sensi-tive to distension, ischemia and inflammation. Nowadays, it is well recognized that there is a close relationship between the gut microbiome and pain perception, and that microbiome is highly af-fected by both anaesthesia and surgical manipulation. Thus, efforts to restore the disturbed micro-biome by supplementation with beneficial bacteria, namely probiotics, seems to be effective. In this article the knowledge gained mainly from experimental research on this topic is analyzed, the con-cluding message being that each probiotic strain works in its own way towards pain relief.

The ground surface deformation induced by shield tunnels passing through enclosure structure of existing tunnels is a particular underground construction scenario, which is encountered in Wuhan metro line 12 engineering cases in China. The classic ground deformation theory is difficult to accurately predict this ground deformation. This paper develops a semi-analytical method to predict ground heave considering space effect in this engineering condition. Based on improved ground deformation theory, a novel deformation prediction method of ground and enclosure structure is derived combined with Kirchhoff plate theory. Comparing with field deformation measurements, the maximum difference between measured and calculated deformation is 14.6%, which demonstrating that the proposed method can be used to predict the ground heave induced by shield tunnels passing through the enclosure structure of existing tunnels. The parameters of underground diaphragm wall used in Wuhan metro line 12 are further studied in detail. The results show that the ground heaves have positive correlation with embedded ratio of diaphragm wall, but negative correlation with its elastic modulus and thickness. But the thickness and embedded ratio has a limited effect on ground heaves. This study provides a technical reference for optimization setting of enclosure structure in protecting existing building.

Background: The role of NFATC genes expression in BAV progression is not fully understood. Aim of study is to determine the significance of NFATC1 and NFATC4 gene expression for myocardial changes in children with BAV. Methods: In 47 children with BAV the standard Doppler echocar-digraphic characteristics were detected and the expression of the NFATC1 and NFATC4 genes was studied. Results: PWTd, AoPPG, in BAV patients were significantly higher compared to healthy controls (PWTd median(min-max) 9 (7-10) mm vs 7 (6-8) mm), AoPPG median (min-max) 7,79 (2,98-15,09) mm Hg vs 2,94 (2,42-3,72) mm Hg. Expression of NFATC1 gene in BAV children was significantly higher compared to NFATC4 (NFATC1 median (min-max) 70,88 (8,79-106,51) e.u. vs 7,72 (1,74-22,67) e.u., respectively p<0,05). Significant correlation of NFATC1 expression with Ao found (R=+0,53, p<0,05). In BAV patients with PWTd > 8 mm and Ao> 21 mm the NFATC1 expression was significantly higher compare to those with PWTd< =8 mm and Ao<=21 mm NFATC1 median (min-max) 45,49 (5,01-101,52) e.u. vs 15,53 (2,36-44,40) e.u., p<0.05 and 81.11 (20.27-101.10) e.u. mm vs 12.16 (2,40-45.49) e.u., p<0.05, respectively) Conclusion: in children with BAV the high expression of the NFATC1 calcineurin signalling pathway gene is associated with elevated PWTd and Ao.

Production of biofuel from lignocellulosic biomass is relatively low due to the limited knowledge about natural cell wall loosening and cellulolytic processes in plants. Industrial separation of cellulose fiber mass from lignin, its saccharification and alcoholic fermentation is still cost-ineffective and environmental unfriendly. Assuming that the green transformation is inevitable and that the new sources of raw materials for biofuels are needed, we decided to study cell death - a natural process occurring in plants in a context of reducing the recalcitrance of lignocellulose for production of second generation bioethanol. “members of the enzyme families responsible for lysigenous aerenchyma formation were identified during the root hypoxia stress in Arabidopsis thaliana cell death mutants. The cell death regulatory genes, LESION SIMULATING DISEASE 1 (LSD1), PHYTOALEXIN DEFICIENT 4 (PAD4) and ENHANCED DISEASE SUSCEPTIBILITY 1 (EDS1) conditionally regulate the cell wall when suppressed in transgenic aspen. During four years of growth in the field the following effects were observed: lignin content was reduced, the cellulose fiber polymerization degree increased and the growth itself was unaffected. The wood of transgenic trees was more efficient as a substrate for saccharification, alcoholic fermentation and bioethanol production. The presented results may trigger the development of novel biotechnologies in the biofuel industry.

The blackening of cut carrots decreases their shelf life and causes severe economic losses but the molecular and metabolic mechanisms that underpin this phenomenon remain poorly characterized. Studies were therefore undertaken to determine the molecular and metabolic causes of the blackening. The susceptibility of blackening was dependent on the period of time that the crop was stored underground prior to harvest. The structure of the cell walls in the black regions was substantially changed compared to the orange regions. The black regions of carrot batons had decreased immunodetection of xyloglucan, HG-pectin, RG-I pectin, galactan and arabinan but had higher levels of lignin and phenolic compounds compared to the orange regions. Transcript profiling analysis revealed that phytohormone signalling processes were activated in the black regions. Transcripts associated with auxin signalling and ethylene-responsive transcription factors were increased in the black regions. In contrast, the levels of transcripts encoding proteins associated with secondary metabolism were decreased in the black regions. These findings implicate ethylene and auxin-related processes in the control of the primary to secondary metabolism shift that results in lignification and cell wall disruption that underpin the blackening process.

To investigate the impact of a magnetic field on plaque development in a stenotic bifurcated artery, a finite element method is utilized. The blood flow is modelled as a stable, incompressible, Newtonian, biomagnetic, and laminar fluid. Furthermore, the arterial wall is assumed to be linear elastic. The Arbitrary Lagrangian Eulerian (ALE) method is employed to describe the hemodynamic flow in a bifurcated artery under the influence of an asymmetric magnetic field, taking into account two-way fluid-structure interaction coupling. A stable $P_2P_1$ finite element pair discretizes a nonlinear system of partial differential equations that requires a solution. The Newton-Raphson method is utilized to find a solution to the resulting nonlinear algebraic equation system. Numerical modelling is used to simulate the presence of magnetic fields, and the resulting displacement, velocity magnitude, pressure, and wall shear stresses are shown for a range of Reynolds numbers ($Re = 500$, $1000$, $1500$, and $2000$). The results of the numerical analysis demonstrate that the presence of a magnetic field has a significant effect not only on the magnitude of displacement but also on the velocity of the flow. The application of a magnetic field reduces flow separation, extends the recirculation area near the stenosis, and increases wall shear stress.

Influence of Joule heating on magnetic properties, giant magnetoimpedance (GMI) effect and domain wall (DW) dynamics of Fe75B9Si12C4 glass-coated microwires was studied. A remarkable increase in GMI ratio is observed in Joule heated samples. The hysteresis loops of Joule heated samples maintain a rectangular shape, while a slight decrease in coercivity after Joule heating is observed. On the other hand, a modification of MOKE hysteresis loops is observed upon Joule heating. Additionally, DW dynamics improvement after Joule heating is observed. The observed GMI ratio improvement along with the change in MOKE loops and DW dynamics improvement has been discussed considering magnetic anisotropy induced by Oersted magnetic field in the surface layer during Joule heating and the internal stress relaxation. A remarkable GMI ratio im-provement observed in Fe-rich Joule-heated microwires with a rectangular hysteresis loop and fast DW propagation, together with the fact that Fe is a more common and less expensive metal than Co, makes them suitable for use in magnetic sensors.

The building sector is the largest consumer of energy and there are still major scientific challenges in this field. The façade, being the interface between the exterior and interior space, plays a key role in the energy efficiency of a building. In this context, this paper focuses on a ventilated bioclimatic wall for NZEB zero energy buildings. The objective of this study is to investigate an experimental set-up based on a Hot Box allowing characterizing the thermal performances of the ventilated wall. A specific ventilated prototype and an original thermal metrology has been developed. This paper presents the ventilated prototype, the experimental set-up and experimental results on the thermal performances of the ventilated wall. The influence of the air space thickness and the air flow rate on the thermal performances of the ventilated wall is studied.

Blueberry (Vaccinium spp.) has been recognized worldwide as a valuable source of health-promoting compounds, becoming a crop with some of the fastest rising consumer demand trends. Fruit firmness is a key target for blueberry breeding as it directly affects fruit quality, consumer preference, transportability, shelf life, and the ability of cultivars to be machine harvested. Fruit softening naturally occurs during berry development, maturation, and postharvest ripening. However, some genotypes are better at retaining firmness than others, and some are crispy, which is a putatively extra-firmness phenotype that provides a distinct eating experience. In this review, we summarized important studies addressing the firmness trait in blueberry, focusing on physiological and molecular changes affecting this trait at the onset of ripening and also the genetic basis of firmness variation across individuals. New insights into these topics were also achieved by using previously available data and historical records from the blueberry breeding program at the University of Florida. The complex quantitative nature of firmness in an autopolyploid species such as blueberry imposes additional challenges for the implementation of molecular techniques in breeding. However, we highlighted some recent genomics-based studies and the potential of a QTL mapping analysis and genome editing protocols such as CRISPR to further assist and accelerate the breeding process for this important trait.

This study investigates the impact of vertical greenery systems (VGS) applied to heavyweight walls on indoor thermal conditions in a building module situated in the Mediterranean climate of Catania, Italy. Using dynamic simulations in TRNSYS, the research compares the thermal behavior of walls constructed from hollow clay blocks (Poroton) and lava stone blocks with a lightweight wall setup already in place at the University of Catania. The primary focus is on evaluating the VGS’s effect on reducing peak inner surface temperatures and moderating heat flux fluctuations entering the building. The findings indicate that adding a vertical greenery layer to heavyweight walls can decrease the peak inner surface temperature by up to 1.0°C compared to the same wall without vegetation. However, the greenery’s impact on mitigating heat flux fluctuations is less pronounced than in the case of the lightweight wall. This research underscores the potential of green facades in enhancing indoor thermal comfort in regions with climates similar to the Mediterranean, providing valuable insights for sustainable building design and urban planning.

Mycobacterium avium ssp paratuberculosis (MAP) is the causative agent of Johne’s disease (JD) in animals and a suspected cause of Crohn’s disease (CD) in humans. Crohn’s Disease is a progressive, detrimental granulomatous enteritis of which common therapies are targeted to suppressing certain components of the immune system. The efficacy of these treatments can be variable and temporary with no clear process of selection for the individual patient. However, diagnosis of MAP has been elusive in patients with Crohn’s Disease. In animals it can be detected by several different techniques, including bacterial cell culture, polymerase chain reaction (PCR), and immunological targets. To move to an understanding of the possible pathogenicity of MAP and the sup-posed cause of CD in humans, it is necessary to demonstrate the presence of cell wall deficient mycobacterial (CWDM) forms of MAP in blood and tissue cultures from patients with CD. From this, more tailored and accurate therapies can be implemented and be developed to eradicate this infectious organism from humans. Current anti-mycobacterial antibiotics have limited efficacy as MAP is subtly different from tuberculosis. Newer methods, such as vaccination and gut microbiome remediation will be essential as part of combination therapies.

1). BACKGROUND: Cell wall deficient mycobacteria (CWDM) have seldom been isolated in bacterial culture from human subjects. particularly in clinical disease, such as Crohn’s disease. Techniques have focused on the use of polymerase chain reaction (PCR) for the detection of CWDM in association with inflammatory bowel disease. (IBD). This paper is a report of the development of methods to demonstrate the presence of CWDM using the Ziehl-Neelsen (ZN) stain. This is in addition to the detection of the growth of CWDM in fluid media. 2). METHODS: The use of the ZN stain is suitable for the demonstration of CWDM in clinical samples and cultures, when the staining methods that were common prior to the 1970’s, are adopted. Specific culture media are developed and tested in parallel. Alterations in the concentration of basic fuchsin, and changes in the decolourising solutions are necessary. Decolourising solutions such as 20% concentrated sulphuric acid in water are preferable for detection of CWDM in cultures. 3). RESULTS: CWDM can be readily demonstrated using the simple methods outlined in this report. The ability to visualise the presence of CWDM opens the door to the development of specific fluid media for the isolation of CWDM in culture. 4). CONCLUSION: The ZN stain methods optimised for the demonstration of Mycobacterium tuberculosis complex (MTBC) are not suitable for research into infectious causes of inflammatory bowel disease (IBD), as well as for the detection of CWDM in latent tuberculosis. Revisiting the techniques for the detection of CWDM may also refocus attention on inflammatory diseases considered to be idiopathic, but having hallmarks of an infectious process.

This research elucidates the dynamic expression of expansin genes during the wheat grain (Triticum aestivum L.) development process using comprehensive meta-analysis and experimental validation. We leveraged RNA-seq data from multiple public databases, applying stringent criteria for selection, and identified 60,852 differentially expressed genes across developmental stages. From this pool, 28,558 DEGs were found to exhibit significant temporal regulation in at least two different datasets, and were enriched for processes integral to grain development such as carbohydrate metabolism and cell wall organization. Notably, 30% of the 241 known expansin genes showed differential expression during grain growth. Hierarchical clustering and expression level analysis revealed temporal regulation and distinct contributions of expansin subfamilies during the early stages of grain development. Further analysis using co-expression networks underscored the significance of expansin genes, revealing their substantial co-expression with genes involved in cell wall modification. Finally, qPCR validation and grain morphological analysis under field conditions indicated a significant negative correlation between expression of select expansin genes and grain size and weight. This study illuminates the potential role of expansin genes in wheat grain development and provides new avenues for targeted genetic improvements in wheat.

This study discusses the experimental assessment of the in-plane mechanical behavior of a multi-leaf stone masonry wall built from cut stone and reinforced with metal connectors (cramps and dowels). The wall, inspired by conventional multi-leaf stone walls, is meant for use in modern stone masonry buildings. The wall is constructed from two parallel load-bearing walls with a cavity between them, which aims to conceal the installation and insulation needed in modern buildings. The load-bearing walls are connected with cramps and dowels at certain intervals so that the wall works as a single section against horizontal and vertical loads. To characterize the in-plane behavior of the proposed wall, compressive, triplet, and diagonal compression tests were conducted to investigate the compressive strength, shear strength, modulus of elasticity, stiffness, ductility, and energy absorption of the wall. Compared with dry and mortar joint walls, the use of dowels increased the initial shear capacity of the wall by 11 and 19 times, respectively. The application of cramps without curving channels inside the individual stone elements decreased the compressive strength by 18%. The energy absorption of the designed walls with metal connectors was substantially increased to that of the specimens representing conventional stone walls. The results obtained show the applicability of the wall due to its higher shear strength and minimal drop in compressive strength, which is within acceptable limits.

Cytokinesis is accomplished in higher plants by the phragmoplast, creating and conducting the cell plate, to separate daughter nuclei by a new cell wall. The microtubule-severing enzyme p60-katanin plays an important role in the centrifugal expansion and timely disappearance of phragmoplast microtubules. Consequently, aberrant structure and delayed expansion rate of the phragmoplast occur in p60-katanin mutants. Here, the consequences of p60-katanin malfunction in cell plate/daughter wall formation were investigated by transmission electron microscopy (TEM), while deviations in the chemical composition of cell plate/new cell wall were identified by immunolabeling and confocal microscopy, in root cells of the fra2 Arabidopsis thaliana mutant. It was found that, apart from defective phragmoplast microtubule organization, cell plates/new cell walls appeared also faulty in structure, being unevenly thick and perforated by large gaps. In addition, demethylesterified homogalacturonans were prematurely present in fra2 cell plates, while callose content was significantly lower than in the wild-type. Furthermore, KNOLLE syntaxin disappeared from newly formed cell walls in fra2 earlier than in the wild-type. Taken together, these observations indicate that delayed cytokinesis, due to faulty phragmoplast organization and expansion, results in a loss of synchronization between cell plate growth and its chemical maturation.

Tissue clearing methods combined with confocal microscopy have been widely used for studying developmental biology. In plants, ClearSee is a reliable clearing method that is applicable to a wide range of tissues and is suitable for gene expression analysis using fluorescent reporters, but its application to the Arabidopsis thaliana embryo, a model system to study morphogenesis and pattern formation, has not been described in the original literature. Here we describe a ClearSee-based clearing protocol, which is suitable for obtaining 3D images of Arabidopsis thaliana embryos. The method consists of embryo dissection, fixation, washing, clearing, and cell wall staining, and enables high quality 3D imaging of embryo morphology and expression of a fluorescent reporter with the cellular resolution.

Turgor pressure – the intracellular hydrostatic pressure exerted by the cell protoplast on the cell wall – is of critical physiological importance for all organisms that have the cell wall. However, in mycology there is increasing evidence that turgor pressure is not the only and not always the main factor influencing, for example, the tension of the plasma membrane, the size and shape of non-apical fungal cells, and is not even always the determining factor of apical growth. This paper characterizes various aspects of the curtain model, previously proposed to describe the regulation of plasma membrane tension in the hyphae of basidiomycetes. The current understanding of the four main components of the curtain model are outlined: the driving actin cytoskeleton, the elastic cell wall, tight adhesion of the plasma membrane to the cell wall and macroinvaginations of the plasma membrane. All four elements, summed in the single curtain model, complement or replace the physiological functions of turgor, and allow imagining how a non-apical fungal cell maintains its integrity and physiological functionality under changing osmotic and other conditions. Further experimental confirmation of the curtain model is not only of fundamental importance for mycology, but also opens up new prospects in the development of drugs that control the growth and pathogenicity of filamentous fungi.

Expansins are cell wall (CW) proteins that mediate the CW loosening and regulate salt tolerance in a positive or negative way. The transcription of Populus trichocarpa expansin A6 (PtEXPA6) was downregulated upon prolonged duration of salt stress (48 h) after a transient increase induced by NaCl (100 mM). The role of PtEXPA6 in salt tolerance was determined by transferring the PtEXPA6 gene into the hybrid species Populus alba × P. tremula var. glandulosa clone 84K (84K) and Populus tremula × P. alba INRA '717-1B4' (717-1B4). The PtEXPA6-transgenic poplars of 84K and 717-1B4 showed a greater reduction in photosynthesis, stem height and diameter growth under salt stress and a reduced capacity for ROS scavenging by antioxidant enzymes, which is associated with excessive Na+ accumulation in roots and shoots. NMT flux recordings showed that the radial translocation of Na+ salt into the root xylem and real-time Na+ translocation from roots to leaves were greater in PtEXPA6-transgenic poplars than in wild-type poplars. Analysis of comparative contractility and comparative extensibility of intact root tips indicated that PtEXPA6 increased the CW loosening in the transgenic poplars of 84K and 717-1B4. Noteworthy, the PtEXPA6-promoted CW loosening was shown to facilitate radial and longitudinal transport of Na+ in transgenic poplars. We conclude that overexpression of PtEXPA6 leads to CW loosening that facilitates radial translocation of Na+ into the root xylem and subsequent Na+ translocation from roots to leaves, resulting in excessive Na+ accumulation and consequently reduced salt tolerance in transgenic poplars.

(1) Background If placenta percreta (PP) is suspected, one should take into account ultrasound and MRI data, which allow one to assess not only the depth of invasion of the placenta into the myometrium, but also the degree of involvement of the blood vessels of the uteroplacental region and the pelvis, the formation of adhesions and collaterals between organs as well; (2) Methods: The patient with suspected PP was examined by ultrasound and Magnetic Resonance Imaging (MRI) examination. Histological study was performed on paraffin sections (hematoxylin &amp; eosin, Mallory staining); cytotrophoblast was detected with antibodies to cytokeratin-8; (3) Results MRI and ultrasound examinations showed placental invasion extending beyond the serous lining of the uterus involving the posterior wall of the bladder. Elective abdominal delivery was performed at 37 weeks. Surgical excision of the uterine wall with invaded villi and involved fibrous bladder wall were performed. Histological study revealed invasive cytotrophoblast cells in the uterine wall located up to adventitia of the uterine subserosal vessels only. Thinning of the uterine wall up to the serous membrane was detected; (4) Conclusions: Placental villi and trophoblast extended beyond the uterus is not histology equivalent to placenta percreta. To diagnose PP instrumental visualization methods should be used.

As the control brain of the petrochemical plant, blast resistant performance requirements of the petrochemical control room should be guaranteed when the vapor cloud explosion occurs in the petrochemical production process. 3D-Kagome truss core sandwich structure is a kind of blast resistant material with high energy absorption. Considering the influence factors of the radius of the truss core rod and thickness of the upper and lower panels, in this paper, blast resistant performance of a real steel petrochemical control room with 3D-Kagome truss core sandwich wall is analyzed. With the optimization goal of plastic deformation energy and panel displacement, the optimal wall thickness and radius of the truss core rode are obtained. The optimized blast resistant wall is assembled, and the dynamic response of the steel petrochemical control room with the 3D-Kagome truss core sandwich blast resistant wall is analyzed. The simulation results show that the truss core layer does not dissipate the blast energy effectively when the radius ratio of the truss core rod is larger than 2.7% of the total thickness of the wall; with the increase of the thickness of the upper and lower panels, the proportion of plastic deformation energy of the truss core layer increases gradually and then tends to be a stable proportion value, that is about 90%; When the thickness ratio of the panel is 6.7% of the total one of the wall and the radius ratio of the truss core rod is 2.7% of the total thickness of the wall, the optimal blast resistant 3D-Kagome sandwich wall is determined.
