Gender and biological sex have distinct impacts on the pathogenesis of type 2 diabetes (T2D). Estrogen deficiency is known to predispose female mice to T2D. In our previous study, we found that high-fat, high-sucrose diet (HFHSD) induces T2D in male mice through the miR-10b-5p/KLF11/KIT pathway, but not in females, highlighting hormonal disparities in T2D susceptibility. However, the underlying molecular mechanisms of this hormonal protection in females remain elusive. To address this knowledge gap, we utilized ovariectomized, estrogen-deficient female mice, fed them a HFHSD to induce T2D, and investigated the molecular mechanisms pathway in estrogen-deficient diabetic female mice, cell lines, and patients with T2D in female. Initially, female mice fed a HFHSD exhibited delayed onset of T2D, but ovariectomy-induced estrogen deficiency promptly precipitated T2D without delay. Intriguingly, insulin (INS) was upregulated, while insulin receptor (INSR) and protein kinase B (AKT) were downregulated in these estrogen-deficient diabetic female mice, indicating insulin-resistant T2D. These dysregulations of INS, INSR, and AKT were mediated by a miR-10a/b-5p-NCOR2 axis. Treatment of miR-10a/b-5p effectively alleviated hyperglycemia in estrogen-deficient T2D female mice, while β-estradiol temporarily reduced hyperglycemia. Consistent with the murine findings, plasma samples from female T2D patients exhibited significant reductions in miR-10a/b-5p, estrogen, and INSR, but increased insulin levels. Our findings suggest that estrogen protect against insulin-resistant T2D in females through miR-10a/b-5p/NCOR2 pathway, indicating the potential therapeutic benefits of miR-10a/b-5p restoration in female T2D management.

Chronic kidney disease (CKD) is a growing public health issue with significant morbidity and mortality rates. Traditional diagnostics, with about 70% accuracy, often delay CKD detection, highlighting the need for more efficient methods. Recent studies indicate a potential link between CKD and auditory health, yet this relationship remains underexplored due to methodological challenges and the complexity of establishing causality. Our research utilizes data spanning from 2000 to 2020 from the National Health and Nutrition Examination Survey (NHANES), covering 12,392 participants, including 2,060 diagnosed with CKD. Through meticulous analysis employing logistic regression and Mendelian randomization, we have unearthed novel insights into the bi-directional associations between hearing impairment and CKD. Furthermore, we developed and validated a machine learning model that surpasses traditional diagnostic approaches in terms of accuracy and predictive power. These findings highlight the innovative integration of auditory examinations with demographic data to enhance CKD detection. Our approach demonstrates the potential of machine learning in transforming diagnostic methodologies, thus offering a significant advancement in the field of nephrology and public health.

We investigated the sleep-wake rhythm of nursery school children with the aim of supporting their health and balanced mental/physical development. We analyzed 4881 children from infant to 6 years of age, using 2-week sleep tables recorded by their guardians. The table contains sleep onset times, wake times, nighttime/daytime sleep duration, and the differences in these between weekdays and weekends. The total sleep decrement of children with age is due to a decrease in daytime sleep only, and nighttime sleep duration remains almost unchanged at about 10 hours, and is therefore called the nighttime basic sleep duration (NBSD). While sleep onset time stabilizes at around 9:30 pm by the age of 2, wake-up times to be before 7 am, resulted in weekdays sleep insufficiency. This lack of sleep is compensated by long naps on weekdays and by catching up sleep on weekends morning, which may be the background to future social-jet-lag. Guardians are encouraged to know their children's exact NBSD and set an ap-propriate bedtime to maintain it on weekdays, which prevent sleep dept and form a daily rhythm of waking up at the same time both on weekdays/weekends. These conditions are believed to maintain balance in mental/physical development and school-social-adaptation.

This study aimed to evaluate the influence of olive cake on the diet of autochthonous Bísaro pigs in processed products. To this purpose, loins and “cachaços” were used and subjected to a manufacturing flowchart to obtain a dry-cured product. Both products were subjected to the same formulation, ingredients, and curing steps. Regarding the physicochemical composition, there were significant differences between the two products for the parameters of aw (p<0.001), moisture (p<0.001), total fat (p<0.001), protein (p<0.001), and haem pigments (p<0.001). As for the diet, it only significantly influences the NaCl content (p<0.05). Neither the product nor the diet had any influence (p>0.05) on the fraction of saturated fatty acid (SFA), monounsaturated fatty acid (MUFA), and polyunsaturated fatty acids (PUFA). However, there is a significant difference when it comes to omega 3 (p<0.05). These fatty acids increased with the addition of olive cake, and the diet with olive cake centrifuged by 25% had the highest value for both products. The increase in this omega 3 resulted in a decrease in the PUFA n-6/n-3 ratio (p<0.01) in the diets with olive cake, compared to the control diets.

The optimal timing for actively discontinuing immune checkpoint inhibitor therapy in long-term responders with recurrent/metastatic head and neck squamous cell carcinoma (R/M HNSCC) remains unresolved. We conducted a retrospective study of 246 patients with R/M HNSCC treated with nivolumab to determine the optimal timing to actively discontinue nivolumab therapy. We examined the point at which progression-free survival (PFS) plateaued in all cases. We compared the prognosis of 19 (7.7%) ongoing cases and 227 (92.3%) discontinued cases and analyzed treatment duration and treatment-free interval (TFI). The 6-year overall survival was 11.8% (median, 12.1), and the 6-year PFS was 15.3% (median, 3.0). The PFS curve remained stable for 3 years. The median duration of nivolumab treatment was 2.9 months (range, 0.03–81.9): Ongoing group, 41.8 (5.6–81.9); Decision group, 36.8 (4.0–70.1); Toxicity group, 30.6 (2.8–64.8); and progressive disease group, 2.0 (0.03–42.9). TFI in the Decision group was 15.1 months (0.6–61.6) and 30.6 months (2.8–64.8) in the Toxicity group. Limited number of long-term responders in R/M HNSCC cases treated with nivolumab exists. Given that PFS plateaued at 3 years, we propose that the optimal duration of treatment for this subset of patients is 3 years.

The growing requirements regarding the safety of using polymers and their composites are related to the emergence of more effective, sustainable, and hazardous-limited fire retardants (FR). Significant amounts of FR are usually required to effectively affect the polymer burning behavior, while the knowledge of their recycling potential is still insufficient. At the same time, concerns are related not only to the reduced effectiveness of flame retardancy but, above all, to the potential deterioration of mechanical properties caused by the degradation of temperature-affected additives under processing conditions. This study describes the impact of four-time reprocessing of bio-based polyamide 11 (PA11) modified with an intumescent flame retardant (IFR) system composed of ammonium polyphosphate (APP), melamine cyanurate (MC) and pentaerythritol (PER) and its composites containing additionally short basalt fibers (BF). Composites manufactured by twin-screw extrusion were subjected to four reprocessing cycles by injection molding, comprehensively analyzing structural, mechanical, and fire behavior changes in each cycle. The obtained results confirmed the safety of using the proposed flame-retardant system for polyamide, processed under the recommended process parameters, without the risk of significant changes in the structure despite the partial increase in flammability caused mainly by the degradation of the polymer.

Bioactive peptides are sequences of amino acids, cut from the parent protein with en-zymes or high pressure processing that can impart a tertiary health benefit to the con-sumer that goes beyond basic human nutrition. This study explores mesopelagic fish in-cluding Maurolicus muelleri and Benthosema glaciale as sources of peptides with an-ti-inflammatory, heart health beneficial, antioxidant and other bioactivities. Bioactive peptides were generated post-catch using different enzymatic hydrolysis processes ap-plied to mesopelagic species caught from experimental cruises targeting these fish. Maurolicus muelleri and Benthosema glaciale were the dominant species processed. Bi-oactive hydrolysates included a Maurolicus muelleri Alcalase hydrolysate and a Maurolicus muelleri autolysis. Hundreds of peptide sequences were identified using mass spectrome-try from active hydrolysates. In silico analysis was used to rank identified peptide se-quences in terms of overall bioactivity using Peptide Ranker, anti-inflammatory activity using PrepAIP, ability to impart Umami flavours with Umami-MRNN and dipeptidyl peptidase IV inhibitory potential using AntiDMPpred . Thirty peptides were selected fol-lowing in silico analysis. Several of the selected peptides had sequence homology with peptides derived from other fish proteins including Atlantic salmon (peptide QCPLHR-PWAL) and thin-lipped mullet (peptide FDAFLPM). Seven peptides predicted to have an-ti-inflammatory, anti-type 2 diabetes or Umami potential using in silico strategies were chemically synthesized and their anti-inflammatory activities confirmed using in vitro bioassays with COX-1 and COX-2 enzymes. The peptide QCPLHRPWAL inhibited COX-1 and COX-2 by 82.90% (+/-0.54) and 53.84% respectively and had a selectivity index greater than 10. This peptide warrant further research as a novel anti-inflammatory/pain relief peptide. Other bioactivities associated with DPP-IV inhibition and Umami flavours de-velopment are discussed. Identified peptides offer potential for use in functional foods or topical agents to prevent pain and inflammation associated with diseases like psoriasis, arthritis and inflammatory bowel disease, however further research in vivo is required.

Primary open-angle glaucoma (POAG) is a progressive optic neuropathy with a complex, multifactorial aetiology. Raised intraocular pressure (IOP) is the most important clinically modifiable risk factor for POAG. All current pharmacological agents target aqueous hu-mour dynamics to lower IOP. Newer therapeutic agents are required as some patients with POAG show a limited therapeutic response or develop ocular and systemic side ef-fects to topical medication. Elevated IOP in POAG results from cellular and molecular changes in the trabecular meshwork driven by increased levels of transforming growth factor β (TGFβ) in the anterior segment of the eye. Understanding how TGF affects both the structural and functional changes in the outflow pathway and IOP is required to de-velop new glaucoma therapies that target the molecular pathology in the trabecular meshwork. In this study we evaluated the effects of TGF-1 and -2 treatment on miRNA expression in cultured human primary trabecular meshwork cells. Our findings are pre-sented in terms of specific miRNAs (miRNA-centric), but given miRNAs work in net-works to control cellular pathways and processes, a pathway-centric view of miRNA ac-tion is also reported. Evaluating TGF-responsive miRNA expression in trabecular meshwork cells will further our understanding of the important pathways and changes involved in the pathogenesis of glaucoma and could lead to the development of miRNAs as new therapeutic modalities in glaucoma.

Visceral leishmaniasis (VL) results from infection by the protozoa Leishmania infantum and L. donovani. HIV-coinfected patients may have a relapsing course of VL, even after secondary prophylaxis with amphotericin B. This study investigated whether host factors would explain the relapses. First, susceptibility to amphotericin B of L. infantum isolates was evaluated in vitro. Then, clinical data and the lipid profile of patients with relapsing and non-relapsing VL were assessed. Susceptibility to amphotericin B was similar between the isolates. CD4+ lymphocytes were reduced in both patients in the first episode and with relapsing VL. Still, the strongest blood cell associated with relapses was low total lymphocyte counts. Total plasma cholesterol, high-density lipoprotein, low-density lipoprotein, and, uniquely, triglycerides of the six individuals in the first episode and 23 with relapsing VL were lower in relapsing patients than those in the first episode. Deceased patients had extremely low, low-density lipoprotein. After CD4+ decrease, lymphocyte CD8+ reduction seem to be the final stage of the immunological failure. The lower lipid concentrations appear to be secondary to the depletion of fat stores by inflammation-induced cachexia provoked by the co-occurrence of both diseases, which can finally lead to death.

Pressure Ulcers (PU) are a debilitating and often painful condition. They are localized lesions on the skin and/or underlying tissues and are common in the elderly, immobile people, diabetics and vascular disease or malnutrition, as well as in those requiring intensive or palliative care. Prevention and treatment of PU involve strategies to optimize hydration, circulation and nutrition. Nutrition plays a key role in pressure ulcer care because wounds require macronutrients and micronutrients to heal. Reports relating to the effectiveness of "Complementary Enzyme Therapy" also in the vulnological field led us to this study, the aim of which was to test the activity of a biodynamic nutritional supplement (Citozym®), characterized by a marked antioxidant activity, administered topically and/or systemically, in patients suffering from PU. The treatment showed a marked reduction in bedsores and in many cases their complete healing. Furthermore, it was possible to observe a lower incidence of side effects compared to conventional therapies. The results obtained, confirmed by various tests, and recognized by the scientific community, allow us to conclude that treatment with Citozym® could represent a new and effective strategy for the treatment of PU.

The Ionian Sea is a crucial intersection for various water masses in the Mediterranean. Its hydrography and dynamics play a significant role in the seawater budgets and biogeochemistry of the neighboring sub-basins. Multiple theories have been formulated to gain a better understanding of the Ionian dynamics. These theories primarily attribute the variability of the near-surface Ionian circulation to internal processes. Here, we utilize horizontal currents and temperature-salinity profiles from the Copernicus reanalysis to examine the contribution of baroclinic modes to the variability of the basin horizontal circulation. Our findings demonstrate that, although the basin vertical structure is characterized by three layers, the primary patterns of the Ionian circulation can be attributed to the first baroclinic mode. This mode, along with the barotropic mode, accounts for over 85% of the overall variability in the Ionian circulation, suggesting that only one of the three interfaces separating the different water masses in the basin is dynamically active. We estimate the depth of this interface to be about 490 m. Additionally, our analysis shows that more than 90% of the kinetic energy over the water column is localized above this interface, indicating that the deep layer of the Ionian is dynamically nearly inert.

Aggressive mature T-cell lymphoma (TCL) is a disease that carries a poor prognosis. The mechanism, at the functional protein level, of the resistance to therapy of TCL has not yet been well clarified. To comprehensively clarify the mechanism, we first reviewed the literature, as mentioned in the Introduction section, on 22 types of representative functional proteins reported as being potentially associated with treatment resistance, mainly in hematologic tumors. Next, we retrospectively analyzed the tumor cell expressions of these 22 functional proteins in 16 randomly selected patients with TCL who had received CHOP (cyclophosphamide, hydroxyl doxorubicin, oncovin, and prednisolone) therapy as first-line treatment. As a result, we propose a new three-group prognostic classification of TCL (the TCL Urayasu Classification) based on the mechanism of resistance to treatment of these tumors. The study subjects included 7 patients (44%) with peripheral T-cell lymphoma, not otherwise specified (PTCL-NOS), 6 patients (38%) with angioimmunoblastic T-cell lymphoma (AITL), and 3 patients (19%) with anaplastic large T-cell lymphoma (ALCL). Immunohistochemistry was performed in paraffin-embedded tumor tissue sections prepared from these patients as antigens using antibodies against the 22 previously reported treatment-resistance-related proteins to determine the tumor cell expression statuses of these proteins. Log-rank test was performed to compare the overall survival (OS) after CHOP therapy of the patients showing positive and negative immunostaining for these proteins, as well as among patients grouped based on other factors. Overall, the median OS of the TCL patients was about 33.5 months. When compared by the disease type, the prognosis was found to be worse in the order of ALCL, AITL, and PTCL. A multivariate analysis showed that expression of glucose-regulated protein 94 (GRP94), a protein that serves as a pro-survival component under endoplasmic reticulum (ER) stress in the tumor microenvironment, was significantly associated with shortened survival. No significant difference was observed for the conventional poor prognostic factors: high-risk International Prognostic Index (IPI) and high-risk Prognostic Index for peripheral T-cell lymphoma, unspecified (PTCL-U) (PIT). However, when GRP94 was combined with six other factors, listed below, as survival-shortening factors, significant differences were observed. The 6 factors were 1) programmed cell death-ligand 1 (PD-L1) and 2) programmed cell death 1 (PD-1), which are involved in immune surveillance; 3) aldo-keto reductase family 1 member C3 (AKR1C3), which is an enzyme that inactivates HO (hydroxyl doxorubicin [H] and oncovin [O]) of CHOP; 4) P53, a tumor suppressor; 5) Glucose-regulated protein 78 (GRP78), an ER stress protein; 6) thymidine phosphorylase (TP), which is an enzyme involved in thymidine catabolism. Based on the combination of GRP94 and the six other factors expressed in the tumors, we propose a new prognostic classification system for TCL (TCL Urayasu classification). Group 1 (group with a relatively good prognosis): GRP94-negative (n = 6; median OS, 88 months; p < 0.01); Group 2 (group with a poor prognosis): GRP94-positive, plus expression of 2 of the 6 aforementioned factors (n = 5; median OS, 25 months; p > 0.05); and Group 3 (group with a very poor prognosis): GRP94-positive, plus expression of at least 3 of the 6 aforementioned factors (n = 5; median OS, 10 months; p < 0.01). Thus, the TCL Urayasu prognostic classification is a simple, useful, and innovative classification that also explains the mechanism of resistance to treatment for each functional protein. Accumulation of more cases and further analyses are expected in the future to enable clinical application, such as use of a stratified treatment approach and treatment using inhibitors.

Pharmacists must acquire the necessary knowledge to provide better drug therapy to patients. In other words, educational programs need to facilitate behavioral changes among pharmacists. Therefore, this study aimed to evaluate how the behavior of pharmacists changed after attending a palliative care educational program. We conducted a web-based questionnaire survey examining the behavior of pharmacists regarding palliative care before participating in the program, two months after participating in the program, and eight months after participating in the program, to determine the behavior and changes over time. For all questions, scores were higher at two and eight months after attending the program than before attending the program (P &lt;0.05). In addition, no significant difference was observed between two and eight months after attending the program for all questions (P =0.504-1.000). The knowledge gained from the educational program was used to repeatedly intervene with patients with cancer, to address the various symptoms they experienced and maintain their behavior. The proven effectiveness of this program serves as a stepping stone for a nationwide rollout across Japan’s 47 prefectures.

Coronavirus disease-19 (COVID-19) has disproportionately affected certain demographics in England, exacerbating existing health disparities. Effective therapeutics are a critical line of defence against COVID-19, particularly for patients at elevated risk for severe disease. Surveillance systems were established to monitor usage of COVID-19 therapeutics in hospital and community settings and inform stewardship. Three antiviral therapies: nirmatrelvir plus ritonavir (Paxlovid®), remdesivir (Veklury®), and molnupiravir (Lagevrio®); and two neutralising monoclonal antibody therapies (nMAbs): sotrovimab (Xevudy®) and casirivimab with imdevimab (Ronapreve®); were in use in England between July 2020 to April 2023. This paper aims to illuminate trends in the utilisation of COVID-19 therapeutics treatment in both hospital and community settings, stratified by the Index of Multiple Deprivation (IMD) in England. Chapter 3 of the English Surveillance Programme for Antimicrobial Utilisation and Resistance (ESPAUR) report 2022 to 2023 also discusses the epidemiological surveillance of these five directly-acting antiviral COVID-19 therapeutics use in England between 2022 to 2023.

Oral bone defects occur as a result of trauma, cancer, infections, periodontal diseases, and caries. Autogenic and allogenic grafts are the gold standard used to treat and regenerate damaged or defective bone segments. However, these materials do not possess the antimicrobial properties necessary to inhibit the invasion of the numerous deleterious pathogens present in the oral microbiota. In the present study poly(ε-caprolactone) (PCL), nano-hydroxyapatite (nHAp), and a commercial extract of Humulus lupulus L. (hops) were electrospun into polymeric matrices to assess their po-tential for drug delivery and bone regeneration. The fabricated matrices were analyzed by scanning electron microscopy (SEM), tensile analysis, thermogravimetric analysis (TGA), FT-IR assay, and in vitro hydrolytic degradation. The antimi-crobial properties were evaluated against the oral pathogens Streptococcus mutans, Porphyromonas gingivalis, and Aggregatibacter actinomycetemcomitans. The cytocompatibility was proved by the MTT assay. SEM analysis proved the nanostructured matrices have suitable fiber size. The present research provides new information about the interaction of natural compounds with ceramic and polymeric biomaterials. The hop extract and other natural or synthetic medicinal agents can be effectively loaded into PCL fibers and has the potential to be used in biomedical applications.

Climate change, growing world population and increasing food demand require higher yields and adaptability of crops, including spring bread wheat. Modern plant breeding based on achievements of genetics has a significant impact on plant resistance to diseases, climate variability and other factors. Genome wide association studies (GWAS) is one of genetics tools for mining of genome loci associated with economically important traits. In the current study, we applied GWAS to reveal QTL for spike productivity and grain quality using a panel of spring varieties of Triticum aestivum L. bred in different decades of the last century in Russia and Germany. The study has enabled to find new genetic markers associated with grain yield and quality. Studying genetic basis of grain yield and quality by comparison of wheat varieties from different breeding periods, including landraces preserved as the source of valuable genetic diversity for breeding and genetic studies, makes it possible not only to understand selection processes in the past, but also to develop strategies for future breeding work.

Infections with multi-drug-resistant (MDR) pathogens in food-animals threaten public health and food security. However, the epidemiology and factors associated with MDR Escherichia coli on Ugandan farms are not well known. This study investigated the prevalence, resistance patterns and factors associated with MDR E. coli carriage on chicken farms. A total of 158 E. coli strains were isolated from 158 chicken farms. The disc diffusion method for susceptibility testing was performed. Clinical breakpoints were interpreted according to Clinical and Laboratory Standards Institute guidelines. MDR was defined as resistance to three or more classes of antimicrobials. MDR E. coli prevalence on chicken farms was 62.7% (95% CI: 55.0 – 70.3). High resistance was observed against ampicillin 79.8% (95% CI: 72.7–85.4), tetracycline 72.8% (95% CI: 65.2–79.2), cotrimoxazole 55.7% (95% CI: 47.8–63.3), and ciprofloxacin 38% (95% CI: 30.7–45.9). Farmers who were males (APR = 0.72, CI: 0.55, 0.93), and attained at least secondary education (APR=0.64, CI: 0.46, 0.88) and administration of recommended antibiotic doses (APR = 0.76, CI: 0.59–0.96) posed a lower risk of MDR E. coli carriage. Farms without footbaths posed a heighten risk (APR=1.48, CI: 1.16–1.88). MDR E. coli were highly prevalent on chicken farms. This study underscores the need for antimicrobial stewardship and improved infection prevention strategies on chicken farms.

Fragile X syndrome (FXS) is the most frequent monogenic form of autism spectrum disorder (ASD). Autistic FXS is caused by loss of the fmr1 gene product, the fragile X mental retardation protein (FMRP), triggering physiological and behavioral abnormalities. It is correlated with clock components for behavioral circadian rhythm. Mutation of this gene causes the disturbances in sleep patterns and circadian behavior commonly observed in patients with autistic FXS, accompanied by frequent dysregulation of melatonin synthesis and melatonin-dependent signaling. These changes impair vigilance, learning and memory, and are also linked to autistic behavior including the abnormal anxiety response. However, although several possible causes, symptoms, and clinical features of ASD have been investigated, the correlation between an altered circadian rhythm and autistic FXS has not been extensively studied. Recent works have highlighted the impact of melatonin on the nervous, immune, and metabolic systems. Even though utilization of melatonin for sleep disorder in ASD has been considered in clinical research, further studies should be aimed at its neuroprotective role in ASD during developmental period. In this review, we focus on the regulatory circuits involved in melatonin dysregulation and circadian system disruption in those with autistic FXS. Additionally, we discuss the neuroprotective effect of melatonin intervention. This may improve neuroplasticity and physical capability. We also review the underlying molecular mechanisms, and suggest that melatonin may be a useful novel treatment for autistic FXS, countering the adverse effects of circadian variation.

Laser-plasma interactions of ultra-short laser pulses with solid targets can generate highly intense pulses of protons and ions with energies of several MeV. The spectra of these particles are spread over a wide range following a Maxwellian distribution. We report on the design and testing of a magnetic chicane for the selection of protons within a limited energy window. It consists in two successive, anti-parallel dipole fields generated by cost-effective, permanent C-magnets with customized configuration and longitudinal positions. The chicane has been implemented into the target vessel of a petawatt laser facility under constraints on the direction of the incoming laser beam and guidance of the outgoing particles through a vacuum port. The separation of protons and carbon ions within distinct energy intervals has been demonstrated and compared to a ray tracing code.

Cardiomyopathies (CMPs) are a group of myocardial disorders that are characterized by structural and functional abnormalities of the heart muscle. These abnormalities occur in the absence of coronary artery disease (CAD), hypertension, valvular disease, and congenital heart disease. CMPs are an increasingly important topic in the field of cardiovascular diseases due to the complexity of their diagnosis and management. In 2023, the ESC guidelines on cardiomyopathies were first published, marking significant progress in the field. The growth of techniques such as cardiac magnetic resonance imaging (CMR) and genetics has been fueled by the development of multimodal imaging approaches. For the diagnosis of CMPs, a multimodal imaging approach, including CMR, is recommended. CMR has become the standard for non-invasive analysis of cardiac morphology and myocardial function. This document provides an overview of the role of CMR in CMPs, with a focus on tissue mapping. CMR enables the characterization of myocardial tissues and the assessment of cardiac functions. CMR sequences and techniques, such as late gadolinium enhancement (LGE) and parametric mapping, provide detailed information on tissue composition, fibrosis, edema, and myocardial perfusion. These techniques offer valuable insights for early diagnosis, prognostic evaluation, and therapeutic guidance of CMPs. The use of quantitative CMR markers enables personalized treatment plans, improving overall patient outcomes. This review aims to serve as a guide for the use of these new tools in clinical practice.

Lamin A, B and C, the nuclear intermediate-filament proteins, play a role in epigenetic regulation. While Lamin B is expressed in all nucleated cells studied, Lamin A/C are transcribed in most somatic cell types except mature B lymphocytes. Since Epstein-Barr virus (EBV), a human gammaherpesvirus, is associated with tumorigenic processes and is known to alter the epigenotype of its host cells, we studied the expression of the LMNA gene and its epigenetic marks in EBV-carrying human lymphoid cell lines. We observed a high lamin A/C mRNA and protein expression in EBV-immortalized lymphoblastoid cell lines (LCLs) and in group III Burkitt lymphoma (BL) lines where hypomethylated first exons were observed with activating histone marks. In most cell lines with low promoter activity a highly methylated first exon could be detected. Our data showed that methylation of the first exon of LMNA was associated with the downregulation of LMNA expression whereas euchromatic histone marks were enriched at active LMNA promoters in EBV-immortalized LCLs. These data suggest a role for viral latency products to activate LMNAp in EBV-infected latency type III B cells in vitro. Expression of lamin A/C may contribute to the establishment of activated B cell phenotype that needs further explorations.

The polymerization mechanism of para-methoxystyrene catalyzed by cationic α-diimine palladium complexes with various ancillary ligands was rigorously examined using density functional theory. In the classical methyl-based α-diimine palladium complex [{(2,6-iPr2C6H3)-N=C(Me)-C(Me)=N-2,6-iPr2C6H3)}PdMe]+ (A+), the 2,1-insertion of para-methoxystyrene is favored over the 1,2-insertion, both thermodynamically and kinetically, during the chain initiation step. The resulting thermodynamically favored η3-π-benzyl intermediates face a substantial energy barrier, yielding only trace amounts of polymer, as experimentally verified. In contrast, the dibenzobarrelene-based α-diimine palladium complex [{(2,6-iPr2C6H3)-N=C(R)-C(R)=N-2,6-iPr2C6H3)}PdMe]+ (R = dibenzobarrelene, B+) shows similar energy barriers for both 2,1- and 1,2-insertions. Continuous 2,1/2,1 or 2,1/1,2 insertions are impeded by excessive energy barriers. However, theoretical calculations reveal that the 1,2-insertion product can seamlessly transition into the chain propagation stage, producing a polymer with high 1,2-regioselectivity. The observed activity of complexes A+ or B+ towards para-methoxystyrene polymerization stems from the energy barrier differences between the 1,2- and 2,1-insertions, influenced by the steric hindrance from the ancillary ligands. Further investigation into the effects of steric hindrance on the chain initiation stage involved computational modeling of analogous complexes with increased steric bulk. These studies established a direct correlation between the energy barrier difference ∆∆G (1,2−2,1) and the van der Waals volume of the ancillary ligand. Larger van der Waals volumes correspond to reduced energy barrier differences, thus enhancing the regioselectivity for para-methoxystyrene polymerization. Moreover, the experimental inertness of complex B+ towards styrene polymerization is attributed to the formation of stable kinetic and thermodynamic 2,1-insertion intermediates, which obstruct further styrene monomer insertion due to an extremely high reactive energy barrier. These findings contribute to a deeper understanding of the mechanistic aspects and offer insights for designing new transition metal catalysts for the polymerization of para-alkoxystyrenes.

Background: Breastfeeding self-efficacy is a critical factor influencing the initiation and continuation of breastfeeding among mothers. Low self-efficacy affects mothers’ ability to sustain breastfeeding. Several studies have investigated the factors influencing breastfeeding self-efficacy, but none have interconnected maternal, infant, and social support factors. Aims: This study aimed to measure the correlation between maternal and infant factors, social support, and breastfeeding self-efficacy. Methods: A cross-sectional approach was employed in Jember, Indonesia, among 200 breastfeeding mothers. The study population consisted of mothers 17-42 years old who were breastfeeding and who met the following inclusion criteria: mothers who have babies aged 1-6 months, term pregnancy history, wanted pregnancy, and baby weight born ≥ 2500 grams. Maternal and infant factors and social support were determined through interviews and questionnaires. Breastfeeding self-efficacy was examined using The Breastfeeding Self-efficacy Scale Short. The multiple logistic analysis was used for multivariate analysis. Results: Of a total of 200 breastfeeding mothers, 63% had low breastfeeding self-efficacy, and 59.5% of mothers did not exclusively breastfeed. The multiple logistic regression indicated that 74.2% of the factors included in the model influenced breastfeeding self-efficacy. Positive correlations were observed between breastfeeding self-efficacy and feeding type (OR=6.603), breastfeeding frequency (OR=6.220), breastfeeding intention (OR=4.993), and family type (OR=4.001). Conclusion: feeding type, breastfeeding frequency, and family support were the important factors that support breastfeeding self-efficacy.

This paper presents the experimental characterization and numerical optimization of the parametric design of inductors used for the induction heating process of metallic samples. Here, the variation of the geometric parameters of the inductor (the distance between the coil and the material, as well as the number of turns and separation between them), was analyzed. In addition, changes in the power input values of the heating system variables were also investigated; and finally, the heating time of the materials was taken into consideration. For the multi-objective numerical optimization process, a full factorial 2K experimental design was devised. Afterward, from preliminary experiments, a response surface was performed, from which the optimal candidates for the heating process were obtained. In the experiments, a methodology was designed for the acquisition and processing of heating data on ASTM E8 tensile test specimens. The main objective of this paper is to determine both the geometric parameters of the coil as well as the power input values of the circuit, which allow for generating uniform heating in the specimens. The objectives of the optimization process are to maximize the heating temperature minimizing the consumption of electrical current in the induction circuit. The data generated in the heating of the specimens were acquired using a Fluke® TI10 thermographic camera, while a BK Precision® regulated power supply was used for the induction circuit. The parameters obtained in the optimization process for the optimal candidates were corroborated through experimental tests. The paper shows the initial (non-optimal) and final (optimal) results obtained from the experimental tests. The heating station created with the optimal coil will be used for mechanical characterization tests on metallic materials at temperatures above room temperature.

This study explores the effectiveness of functional near-infrared spectroscopy (fNIRS) as an implicit measurement tool for evaluating sweetness intensity and affective value. Thirty-two participants tasted sucrose solutions at concentrations of 0.15M, 0.3M, and 0.6M, while both their neural responses were recorded with a 24-channel fNIRS system and their self-reported assessments of sweetness intensity and affective value were collected. The neural fNIRS data were converted into oxygenated hemoglobin (HbO) and deoxygenated hemoglobin (HbR) concentrations using the modified Beer-Lambert Law, and analyzed through univariate activation analysis and multivariable decoding analysis to identify neural activation patterns associated with sweetness perception. The results showed significant activation in the dorsolateral prefrontal cortex (dlPFC) and orbitofrontal cortex (OFC) in response to varying levels of sweetness intensity and affective value, with channels 8, 10, 12, 13, 14, 15, and 17 consistently activated across all sucrose concentrations. As sweetness concentration increased from 0.15M to 0.6M, the number of significantly activated channels rose from seven to eleven, indicating stronger and more widespread neural responses corresponding to higher sweetness intensity. The multivariable decoding analysis further demonstrated the capability of fNIRS in accurately distinguishing positive affective responses, with up to 72.1% accuracy. The moderate positive correlation between explicit self-reports and implicit fNIRS data regarding sweetness intensity further supports the validity of fNIRS as a reliable tool for assessing taste perception. This study highlights the potential of fNIRS in sensory neuroscience, demonstrating its effectiveness in capturing the neural mechanisms underlying sweet taste perception.

Islatravir (ISL) is the first-in-class nucleoside reverse transcriptase translocation inhibitor (NRTtI) with novel modes of action. Data on ISL resistance is currently limited, particularly to HIV-1 non-B subtypes. This study aimed to assess prevalent nucleos(t)ide reverse transcriptase inhibitor (NRTI) resistance mutations in HIV-1 subtype C for phenotypic resistance to ISL. Prevalent single and combinations of NRTI resistance mutations were selected from a routine HIV-1 genotypic drug resistance testing database and introduced into HIV-1 subtype C-like pseudoviruses, which were then tested for ISL susceptibility. Single NRTI resistance mutations were susceptible or showed only a low level of resistance to ISL. This included thymidine analogue mutations (TAMs, i.e., M41L, D67N, K70R, T215FY, K219EQ) and non-TAMs (i.e., A62V, K65R, K70ET, L74IV, A114S, Y115F, M184V). Combinations of M184V with one or more additional NRTI resistance mutations generally displayed reduced ISL susceptibilities. This was more prominent for combinations that included M184V+TAMs, and particularly M184V+TAM-2 mutations. Combinations that included M184V+K65R did not impact significantly on ISL susceptibility. Our study suggests that ISL would be effective in treating people living with HIV (PLWH) failing tenofovir disoproxil fumarate (TDF)/lamivudine (3TC) or TDF/emtricitabine (FTC)-containing regimens but would be less effective in PLH failing zidovudine (AZT) with 3TC or FTC-containing regimens.

This study presents the innovative design and fabrication of a 3D-printed prosthesis arm aimed at enhancing automated hand rehabilitation for targeting hemiplegic patients who cannot open and close their hands. By leveraging surface electromyography (EMG) signals, the prosthesis arm translates muscle contractions in the forearm into precise hand movements, facilitating more effective rehabilitation. A comprehensive analysis of existing prosthesis designs guided the creation of a new, improved prototype. Using a concept scoring evaluation matrix, the best concept design was selected, designed, and fabricated through additive manufacturing. The finished prosthesis improves grip stability by imitating natural finger movements with flexible materials and a tendon system made of elastic ropes and fishing lines. Experimental comparisons between the designed prosthesis arm and a human hand showed significant similarities in flexion angles, though further refinement is needed for measurement accuracy and tactile feedback. The result presents the designed prosthesis arm validity and can be used in automated control rehabilitation in the future. This research also highlights the potential of 3D printing and EMG technology in creating customizable, efficient prosthesis arms for automated rehabilitation, suggesting pathways for future improvements and broader applications.

Multi-agent pathfinding has been extensively studied in the robotics and artificial intelligence communities. The classical algorithm, Conflict-Based Search(CBS), is widely used in various real-world applications due to its ability to solve large-scale conflict-free paths. However, classical CBS assumes discrete time-space planning and overlooks physical constraints in actual scenarios, making it unsuitable for direct application in Unmanned Aerial Vehicle(UAV) swarm. Inspired by the decentralized planning and centralized conflict resolution ideas of CBS, we propose an optimal and efficient UAV swarm motion planner that integrates State Lattice with CBS, named SL-CBS. SL-CBS is a two-layer search algorithm: (1) The low-level search utilizes an improved State Lattice. We design emergency stop motion primitives to ensure complete UAV dynamics and handle spatio-temporal constraints from high-level conflicts. (2) The high-level algorithm defines comprehensive conflict types and proposes a motion primitive conflict detection method with linear-time complexity based on Sturm’s theory. Additionally, our modified Independence Detection(ID) technique is applied to enable parallel conflict processing. We validate the planning capabilities of SL-CBS in classical scenarios and compare it with State-Of-The-Art(SOTA) algorithms, showing great improvements in success rate, computation time, and flight time. Finally, we conduct large-scale tests to analyze the performance boundaries of SL-CBS+ID.

The skin, as the body's largest organ, is vital for protecting against environmental stressors, regulating temperature, and preventing water loss. Maintaining a healthy skin barrier is essential for overall skin health and function. We examined the potential of a mixture of five traditional Korean herbal extracts—Cimicifuga racemosa, Paeonia lactiflora, Phellodendron amurense, Rheum rhaponticum, and Scutellaria baicalensis— referred to as herbal medicine composite 5 (HRMC5) for enhancing skin health. High-performance liquid chromatography identified 14 bioactive compounds, including flavonoids, phenolic acids, anthraquinones, and alkaloids, known for their antioxidant, anti-inflammatory, and antimicrobial effects. The extract showed concentration-dependent effects on cell viability and provided UV protection at 6.25 g/L. It also significantly promoted wound healing, similar to the efficacy of epidermal growth factor. Furthermore, the mixture exhibited anti-inflammatory activity by downregulating Cox2 expression and upregulated key skin barrier proteins, including Involucrin, Claudin 1, and Collagen Type 1. These findings highlight the potential of this herbal mixture for addressing skin barrier dysfunction, inflammation, and facial redness, supporting its use in developing natural skincare products. Future studies should investigate the mechanisms behind these effects and assess its clinical applications.

Fusobacterium nucleatum (F. nucleatum, Fn), an oral commensal that can become pathogenic, is associated with periodontitis (PD), adverse pregnancy outcomes, and colorectal cancer (CRC). MicroRNAs (miRNAs) are conserved, non-coding RNAs that play a regulatory role in gene expression and are detected in microbial infections. The aim of this study was to characterize the global microRNA expression kinetics in the mandibles of C57BL/6J mice infected with F. nucleatum for 8 and 16 weeks, and to identify miRNAs as biomarkers of the PD process using high-throughput NanoString nCounter® miRNA panels. Mice were divided into four groups: 8 weeks of Fn infection, 16 weeks of Fn infection, and their respective sham infection. Fn-infected mice at both 8- and 16 weeks showed 100% bacterial colonization on the gingival surface, along with a significant increase in alveolar bone resorption (p<0.0001) and intravascular dissemination to heart, indicating its invasive potential. Out of 577 miRNAs analyzed, seven miRNAs (miR-361-5p, miR-99b-5p) were upregulated, and two miRNAs (miR-362-3p and miR-720) were downregulated in the 8-week Fn infection group. In the 16-week Fn infection group, seven miRNAs (miR-361-5p, miR-99b-5p) were upregulated miRNAs and 13 miRNAs (miR-323-3p, miR-488) were downregulated. Notably, miRNAs such as miR-205, miR-210, miR-199a-3p which were differentially expressed (DE) in the Fn-infected mice at 8 weeks and miR-28 at 16 weeks, have been previously reported in human periodontitis. The 13 miRNAs induced by F. nucleatum (miR-361-5p) are linked to multiple malignancies, including esophageal, gastric, pancreatic, and colorectal cancers. Additionally, the DE miR-126-5p in the 8-week infection group has been identified as a potential biomarker for patients with PD and cardiovascular disease. The results indicate that F. nucleatum acts as a potent oncomicrobe, inducing several miRNAs associated with PD and linking it to systemic comorbidities. Furthermore, the study revealed similar expressions between PD-associated miRNAs and nine CRC tumor-expressed miRNAs.

In this study, we demonstrate a proof of principle of an all-fiber random laser due to the plasmonic effect. This was achieved in a fiber co-doped with Bismuth/Aluminum/Yttria/Silver in which a microsphere (microcavity) at the fiber’s tip was made using a splicing machine. The presence in the fiber of Bismuth- and silver nanoparticles along with Bismuth-Aluminum phototropic centers stands behind the observed phenomenon. The effect can be attributed to in-pair functioning of this unit as an active medium and volumetric plasmonic feedback, with the result being lasing at 807 nm under 532-nm pumping with notably low (~2 mW) threshold.

Myokines are peptides released by the skeletal muscle that exert effects on peripheral tissues, activating metabolic or protective pathways against oxidative stress and inflammation. Many myokines increase their plasma levels after exercise, providing new insights into the role of physical activity in combating sarcopenia and preventing metabolic diseases, such as obesity, diabetes, and metabolic dysfunction associated with steatotic liver disease (MASLD). This review aims to discuss the roles of specific myokines in activating intracellular pathways in the liver to protect against steatosis and lipoperoxidation in MASLD. The progression of liver disease from steatosis to cirrhosis is partially driven by ferroptosis, an iron-dependent form of cell death linked to oxidative stress and lipoperoxidation. Exercise-induced myokines, such as irisin, have been shown to upregulate antioxidant enzymes such as glutathione (GSH) and glutathione peroxidase 4 (GPX4), which are crucial in reducing ferroptosis in hepatic tissues. This review provides a comprehensive perspective on the protective effects of myokines in the liver, particularly against lipid peroxidation and ferroptosis in MASLD, and highlights exercise as a therapeutic strategy to prevent the progression of this liver pathology.

Antibiotic-loaded bone cement has been traditionally advocated as a key step for the success of one-stage hip revision surgery, while cementless techniques have been re-cently proposed with and without the use of local antibacterials as an equally suc-cessful alternative. Aim of this review is to investigate the effective role of local anti-microbial protection for one-stage cemented and cementless hip revision surgery. Twelve studies reporting the results of cemented single-stage procedure at a minimum two years follow-up were reviewed. When pooling together the data, no infection re-currence was observed on average in 83.3% of the patients (range 75.0% to 100%). Only two papers, both from the same French group, included patients treated without the use of antibiotic-loaded bone cement, with an average infection control of 95.9% in a total of 195 patients. This figure appears better that the 80.7% infection control ob-tained by pooling together all the remaining studies. Concerning cementless one-stage revision, a total of 17 studies, reporting on 521 patients, showed an average 90.0% (range 56.8% to 100%) no infection recurrence, at a minimum two years follow-up. Eight papers reported the outcomes of cementless implants without any local antibac-terial protection and nine more described four different techniques for local antimi-crobial implant protection. No comparative study investigated cementless revision with or without local antibacterial protection. Pooled data showed an average infection control of 86.7%, without the application of local antibacterials, compared to 90.1% to 100% with local antimicrobial protection, depending on the technology used. Howev-er, due to the relatively low number of patients treated with each local antimicrobial protection, no statistical difference could be found, either considering local antibacte-rial strategies alone or pooled together. No side effect had been reported by any local antibacterial technique. This review points out that local antibacterial protection for one-stage hip revision surgery, although safe and largely performed in the clinical set-ting, appears still to rely mainly on experts’ opinion and on observational series with no prospective or comparative trial, hence no definitive conclusion can be drawn con-cerning its effective role in one-stage hip revision surgery.

The blood-brain barrier (BBB) strictly regulates the penetration of substances into the brain, which, although important for maintaining brain homeostasis, may delay drug development because of the difficulties in predicting pharmacokinetics/pharmacodynamics (PKPD), toxicokinetics/toxicodynamics (TKTD), toxicity, safety, and efficacy in the central nervous system (CNS). Moreover, BBB functional proteins show species differences; therefore, humanized in vitro BBB models are urgently needed to improve the predictability of preclinical studies. Recently, international trends in the 3Rs in animal experiments and the approval of the FDA Modernization Act 2.0 have accelerated the application of microphysiological systems (MPSs) in preclinical studies, and in vitro BBB models have become synonymous with BBB-MPSs. Recently, we developed an industrialized humanized BBB-MPS, BBB-NET. In our previous report, we reproduced transferrin receptor (TfR)-mediated transcytosis with high efficiency and robustness with hydrogels that include fibrin and collagen I microfibers (CMFs). Herein, we investigated how adding CMFs to the fibrin gel benefits BBB-NETs, showed that CMFs accelerated capillary network formation and maturation by promoting astrocyte (AC) survival, and clarified that integrin 1 is involved in the mechanism of CMFs. Our data suggest that quality control (QC) of CMFs is important for ensuring the stable production of BBB-NETs.

The etiology of autism spectrum disorder (ASD) has not yet been completely elucidated. Through time, multiple attempts have been made to uncover the causes of ASD. Different theories have been proposed, such as that it is caused by “Gods Wrath,” alterations in the gut–brain axis with an emphasis on gut dysbiosis, post vaccine complications, and genetic or even autoimmune causes. In this review we present data covering over 170 000 participants at age ranging from 2 to 14 years focusing on human cell changes rather than using animal models found in previous studies. The male/female ratio was 4:1. Data collection occurred in many countries covering ethnically diversified subjects. Moreover, we aimed to show how the progress in genetic techniques influences the explanation of medical White Papers in ASD area.

A study was conducted to investigate the occurrence of Trypanosoma vivax infection in 72 Murrah buffaloes from Soure, a municipality located on Marajó Island. The aim of the research was to identify the presence of T. vivax in the buffalo population. Blood samples were collected, followed by DNA extraction and polymerase chain reaction (PCR) analysis using the primers TviCatL1 and DTO 155. T. vivax DNA was detected in one 36-month-old female with a body condition score of 2 (on a scale of 1 to 5), although the animal exhibited no clinical signs of infection. These findings suggest that T. vivax is not frequently circulating among the tested buffaloes, or that the animals may be in the chronic phase of infection, which is often characterized by low parasitemia levels. The identification of T. vivax in a clinically asymptomatic animal underscores the importance of using molecular diagnostics for early detection and monitoring of the parasite in buffalo herds, especially in regions where the presence of T. vivax may be underestimated. This study contributes to understanding the epidemiological status of T. vivax in buffaloes on Marajó Island, highlighting the need for ongoing surveillance in endemic areas.

Plant diseases not only cause economic losses, but they also pose a health risk. In this regard, aflatoxins are linked to the development of some type of liver cancer, especially the hepatocellular carcinoma. Aflatoxins are usually produced by the fungi Aspergillus flavus and Parsiticus in figs, and their identification is usually carried out using invasive and complex methods or by visual inspection at an advanced stage. Given that Spain is one of the main producers of figs and Extremadura is the leading region in the country, it is necessary to explore alternative ways of detecting the presence of aflatoxins in figs and preventing them from entering the food chain. The aim of this research is to address the early detection of Aspergillus flavus fungus using non-invasive techniques with the aid of hyperspectral imaging and the application of artificial intelligence techniques, in particular deep learning. The images were taken after inoculation of the microtoxin using 3 different concentrations, related to three different classes and healthy figs (healthy controls). The analysis of the hyperspectral images was performed at the pixel level. Firstly, a full connected neural network was used to analyse the spectral signature associated with each pixel; second, the wavelet transform was applied to each spectral signature. The resulting images are fed to a convolutional neural network. The hyperparameters of the proposed models were adjusted based on the parameter tuning process also performed. The results are promising, with 83% accuracy, 82.75% recall and 83.25% F1-measure for the single neural network, and 77.25% accuracy, recall and F1 for the convolution neural network.

Cutaneous Squamous Cell Carcinoma (cSCC) is an increasingly common malignancy of the skin and the leading cause of death from skin cancer in adults over the age of 85. Fibroblast Growth Factor Receptor 2 (FGFR2) has been identified as an important effector of signaling pathways that lead to the growth and development of cSCC. In recent years, there have been numerous studies evaluating the role FGFR2 plays in multiple cancers, its contribution to resistance to anti-cancer therapy, and new drugs that may be used to inhibit FGFR2. This review will provide an overview of our current understanding of FGFR2 and potential mechanisms in which we can target FGFR2 in cSCC. The goals of this review are the following: 1) to highlight our current knowledge on the role of FGFR2 in healthy skin and contrast this with its role in the development of cancer; 2) to further explain the specific molecular mechanisms that FGFR2 uses to promote tumorigenesis; 3) to describe how FGFR2 contributes to more invasive disease, 4) to describe its immunosuppressive effects in skin, and 5) to evaluate its effect on current anticancer therapy and discuss therapies on the horizon to target FGFR2 related malignancy.

Phosphatidylserine (PS) has significant applications in various sectors, such as the medical and food industries. However, its production relies heavily on phospholipase D (PLD), a crucial tool that is hindered by issues like poor stability and irrecoverability. Immobilization presents itself as an effective solution to overcome these limitations. In this study, magnetic graphene oxide (MGO) modified with amino (-NH₂) were synthesized and utilized for PLD immobilization. The activity of the immobilized PLD (MGO-PLD) reached 3062 U/g_MGO, with a specific activity of 33.9 U/mg_PLD, virtually identical to that of the free PLD. MGO-PLD was utilized to synthesize PS efficiently in a biphasic system. Under optimal conditions, PS yield reached 18.66 g/L, with a conversion rate of 92.8% and a productivity of 3.11 g/L/h. Notably, MGO-PLD retained an impressive PS conversion rate of 77.4% even after seven repetitive usages. Moreover, MGO-PLD displayed enhanced thermal and pH resistance properties compared to free PLD, alongside an augmented storage stability. Post a 8-week preservation at 4°C, its residual activity was able to maintain at 76.3%. This study provides a sustainable and highly efficient pathway for biocatalytic synthesis of PS.

Introduction and importance: Congenital bicuspid aortic valve (BAV) is a common cardiac malformation predisposing individuals to aortic stenosis, regurgitation, and aortopathy. This risk persists even after aortic valve replacement (AVR). Case presentation: We report a case of a male in his 60s with a history of BAV who underwent bioprosthetic aortic valve replacement (BVR) and ascending aorta/root repair in 2014. The patient was noncompliant with follow-up and presented nine years later with severe bioprosthetic aortic stenosis and an 8.0 cm ascending aortic aneurysm with no symptoms. Emergent redo BVR and aneurysm repair were successful. Clinical discussion: It should be emphasized that successful outcomes for BAV patients require a lifelong commitment to surveillance, even after surgical intervention. Noncompliance can lead to life-threatening consequences like bioprosthetic valve deterioration or progressive aortic dilation. Conclusion: This case highlights the importance of early BAV diagnosis for ongoing monitoring, accessible follow-up care after complex cardiac procedures, and addressing patient barriers to treatment adherence.

For a left module RM over non-commutative ring R, the notion for the class of nilpotent elements (nilR(M)) was first introduced and studied by SSevviiri and Groenewald . Moreover, Armendariz and semicommutative modules are generalizations of reduced modules and nilR(M)=0 in the case of reduced modules. Thus, the nilpotent class plays a vital role in these modules. Motivated by this, we present the concept of nil-Armendariz modules as a generalization of reduced modules and a refinement of Armendariz modules, focusing on the class of nilpotent elements. Further, we demonstrate that the quotient module M/N is nil-Armendariz if and only if N is within the nilpotent class of RM. Additionally, we establish that the matrix module Mn(M) is nil-Armendariz over Mn(R) and explore conditions under which nilpotent classes form submodules. Finally, we prove that nil-Armendariz modules remain closed under localization.

Background/Objectives: Disrespectful care during childbirth has been observed as a global issue and a factor influencing maternal morbidity and mortality. While the United States has experienced worsening rates of maternal mortality, perceptions of respectful maternity care have been understudied. Methods A cross-sectional study was conducted Boston from April 2023 to January 2024 among 46 labor and delivery physicians, midwives, and nurses at Massachusetts General Hospital. The survey evaluated their observation of disrespectful care, performance of respectful care, and stress and support factors influencing respectfulness of care. Results: The most reported observed disrespectful behaviors were dismissing patients’ pain (87.0%), discriminatory care based on physical characteristics (67.4%) and race (65.2%), and uncomfortable vaginal examinations (65.2%). Respondents self-reported very high levels of respectful maternity care performance. Reported barriers to respectful care included workload (76.1%) and fatigue (60.9%). Conclusions: Disrespectful care in childbirth is an issue reported by health care providers. Implicit bias and working conditions of health care providers are factors in disrespectful care. This information can be used to strategize future training and other areas of intervention to improve maternity care.

Background: In 2006 to 2014, 10 cases of urolithiasis in yaks, with calcium carbonate uroliths, were confirmed in Switzerland and at the same time a 6-fold calcium overhang in roughage in an affected farm was shown. Methods: Blood samples from 99 animals were examined from 10 farms (n=6 problem farms; n=4 control farms). In addition, roughage, soil and water samples were analyzed. The farms are distributed within different regions in Switzerland. Results: The study revealed different Ca : P ratio from 1.56 to 7.74 : 1 in the forages and mild hypercalcemia in the animals of problem farms. In the univariate analysis, problem versus control farms, about 20 other significant factors emerged. The multivariate analysis has shown that altitude, CP (crude protein), P, Mg, NDF (neutral detergent fibre) and ADF (acid resistant detergent fibre) in the diet are important factors. Based on the analysis of the pedigree there was no evidence for an obvious genetic background of urolithiasis. Limitations: Limitation is the small number of Yaks in Switzerland. Conclusion: The question of whether urolithiasis in yaks in alpine Switzerland, poses a husbandry risk, can be answered affirmatively. The calcium-rich forages play a crucial role in this multifactorial process.

Background: Good reduction of the articular surface and restoration of the congruent joint space are crucial for promoting positive outcomes in ankle fracture treatment. A good understanding of the mechanism of ankle fractures is indispensable to achieve these goals. Unfortunately, until now, the mechanism has still not been fully elucidated, particularly regarding the posterior malleolar fractures (PMFs). This study aims to provide insight into the mechanism of PMFs, thereby grasping the key to proceeding with PMFs reduction and fixation. Methods: An FE model of a normal ankle in tiptoe posture with 9 collateral ligaments was constructed from CT images. On the model, lateral malleolar fractures were made below, through, and above the syndesmosis; the medial malleolus was broken at its base; and ligaments were suppressed to simulate ligament rupture. A force of 1000 Newtons was applied on the talus head along talus axis. The posterior malleolar fragment (PMF) displacement, whether the talus impacts the posterior malleolus, and the initial mechanism displacing the PMF, were recorded. Results: A total of 15 tests were conducted. The tests, 'all collateral ligaments rupture', 'LMFB', 'LMFC+AITFL rupture', 'ATFL+CFL+PTFL rupture', 'ATTL+PTTL+TCL rupture', 'MMF', and 'MMF+AITFL rupture', presented significant PMF displacement. In tests 'MMF' and 'ATTL+PTTL+TCL rupture', the talus hit the medial edge of the PMF; but in tests 'ATFL+CFL+PTFL rupture' and 'all collateral ligaments rupture', the talus impacted the center part of the PMF. Except tests 'all collateral ligaments rupture' and 'ATFL+CFL+PTFL rupture', where PMF displacement was initially caused by talus impact, the other tests demonstrated PMF displaced following PITFL drag. Conclusions: This research discovered that rotation is the initial and major mechanism causing PMF displacement. The subsequent impact of the talus on the posterior malleolus may be the reason for large/blocky PMF. The talus impacting the posterior malleolus is always combined with talus rotation. Without the medial ankle structure being broken, big/blocky PMF may not occur. These findings suggest that, in clinical practice, reconstructing both the medial and lateral ankle structures is critical for achieving ankle stability, and this procedure even takes precedence over posterior malleolar fracture fixation itself.

Gastrointestinal (GI) cancers, such as colorectal and gastric cancers, pose significant global health challenges due to their high rates of incidence and mortality. Even with advancements in treatment and early detection, many patients still face poor outcomes, highlighting the critical need for new biomarkers and therapeutic targets. Telomere length (TL) and telomerase activity (TA) have gained attention in this context. Telomeres, protective nucleotide sequences at chromo-some ends, shorten with each cell division, leading to cellular aging. Telomerase, a ribonucleoprotein enzyme, counteracts this shortening by adding telomeric repeats, a process tightly regulated in normal cells but often dysregulated in cancer. This review critically evaluates the role of TL and TA in the pathogenesis of GI cancers, examining their potential as diagnostic, prognostic, and predictive biomarkers. It explores how alterations in telomere biology contribute to the initiation and progression of GI tumors and assesses the therapeutic implications of targeting telomerase. By integrating findings from diverse studies, this review aims to elucidate the intricate relationship between telomere dynamics and gastrointestinal carcinogenesis, offering in-sights into how TL and TA could be leveraged to enhance the early detection, treatment, and prognosis of GI cancers.

Accurate fire severity mapping is essential for understanding the impacts of wildfires on vegetation dynamics in arid savannas. The frequent wildfires in these biomes often cause topkill, where vegetation experiences above-ground combustion, but the below-ground root structures survive, allowing for subsequent regrowth post-burn. Investigating post-fire regrowth is crucial for maintaining ecological balance, elucidating fire regimes, and enhancing the knowledge base of land managers regarding vegetation response. This study examined the relationship between bush fire severity and post-burn coppicing/regeneration events of woody vegetation in the Kalahari Desert of Botswana. Utilizing UAV-derived RGB imagery combined with a Random Forest (RF) classification algorithm, we aimed to enhance the precision of burn severity mapping at a fine spatial resolution. Our research focused on a 1 km2 plot within the Modisa Wildlife Reserve, extensively burnt by the Kgalagadi Transfrontier Fire of 2021. The UAV imagery, captured at various intervals post-burn, provided detailed orthomosaics and canopy height models, facilitating precise land cover classification and burn severity assessment. The RF model achieved an overall accuracy of 79.5% and effectively identified key burn severity indicators, including green vegetation, charred grass, and ash deposits. Our analysis revealed a &gt;50% probability of woody vegetation regrowth in high severity burn areas six months post-burn, highlighting the resilience of these ecosystems. This study demonstrates the efficacy of low-cost UAV photogrammetry for fine scale burn severity assessment and provides valuable insights into post-fire vegetation recovery, thereby aiding land management and conservation efforts in savannas.

(1) Background: This report aims to illustrate the development, progression, diagnosis and treatment of chronically present articular surface lesions; (2) Methods: In this report, two patients are described from the point of initial presentation of symptoms to surgical consultation based on radiologic findings. These patients underwent corrective surgery in the form of mosaicplasty to repair the lesion present at the articular surface and the underlying subchondral bone. (3) Discussion: Diagnosing juvenile OCD remains challenging due to variable clinical presentation and minute radiologic discoveries. X-rays are useful, however, the gold standard remains to be arthroscopy which can be both diagnostic and therapeutic. Future prospects include the use of novel sonographic methods and the use of artificial intelligence within the given modalities. (4) Conclusions: The detailed imaging provided by MRI, combined with the insights from X-rays and potentially other modalities, allows for a nuanced understanding of the disease. This comprehensive approach ensures that treatment decisions are well-informed, optimizing outcomes for young patients with this condition.

Several years now, the global scientific community has accepted and recognized the importance of Evidence-based practice (EBP) for Nursing science. The main factor for the implementation of EBP is the competence of undergraduate Nursing students towards EBP, so that they as active nurses be ready for its application in their clinical practice, in order to provide better care for their patients. The aim of the present study is to examine the level of evidence-based practice competency evaluated with the self-reported EBP-COQ questionnaire. A quantitative study based on cross-sectional design was conducted from February to June 2022 in a convenience sample of Greek undergraduate nursing students. The Evidence-Based Practice Competence Questionnaire (EBP-COQ) and a questionnaire on undergraduate students’ gender, age, academic year, and training in the field of research were used. The SPSS 26.0 program was used to perform descriptive, bivariate and multivariate analyses. A total of 175 undergraduate students participated at the Hellenic University Nursing (Greece), specifically from the 2nd and 3rd academic year. The mean score of EBP-COQ for nursing undergraduate students was 3.03 ± 0.26, indicating moderate EBP. Among the EBP-COQ dimensions, the mean scores were obtained 3.03 ± 0.32 for Attitude toward EBP, 3.01 ± 0.49 for Skills in EBP, and 3.03 ± 0.49 for Knowledge in EBP. They were not found significant differences among participants’ mean EBP-COQ scores regarding gender (p = 0.766), age (p = 0.400), academic year (p = 0.153), and training in the field of Research Methodology (p = 0.538). It appears that the level of readiness towards EBP is mediocre between undergraduate nursing students at a university in Greece. Therefore, it is necessary to carry out new studies in the future, so that there is a correct approach of all the elements that contribute to the readiness of Nursing undergraduate students regarding EBP.

Mercury is classified as one of the world’s most toxic and dangerous pollutants. It tends to bio-accumulate and biomagnify within the trophic chain and is persistent. Various approaches are available to remediate Hg-affected sites, including phytoremediation, which includes the use of plants to clean up contaminated environments. Phytoremediation of mercury contamination is at-tracting increasing attention because of its advantages: it is environmentally friendly, inexpensive, simple, and can improve soil fertility. In this report, VOSviewer, and Bibliometrix software were used to analyze 457 and 697 documents published from 2000 to 2023, retrieved from the data-bases WoS and Scopus, respectively. China, India, the United States, and Spain are the top four most productive countries. The largest topic area was environmental sciences, and the Chinese Academy of Sciences was the organization that contributed the most to the overall number of pub-lications. The keywords with the highest frequency, excluding phytoremediation and mercury in WoS, were heavy metals, accumulation, cadmium, soils, and phytoextraction. In Scopus, the most frequent keywords were bioremediation, heavy metals, soil pollution, bioaccumulation, biodeg-radation, and environmental. From the above analysis, we concluded that future research should focus on 1) finding native plants, 2) increasing remediation ability through assisted phytoremedi-ation, 3) studies of molecular mechanisms in plants with phytoremediation potential, and 4) ge-netic engineering applications. This study provides insight into trending themes and serves as a reference for future research.

Carbon nanodots (CNDs) have garnered significant attention as viable drug delivery vehicles in recent years, especially in the field of phytomedicine. Although there is much promise for therapeutic applications with phytomedicine, its effectiveness is frequently restricted by its low solubility, stability, and bioavailability. This paper offers a thorough synopsis of the developing field of phytomedicine drug delivery based on CND. It explores CND synthesis processes, surface functionalization strategies, and structural and optical characteristics. Additionally, the advantages and difficulties of phytomedicine are examined, with a focus on the contribution of drug delivery methods to the increased effectiveness of phytomedicine. The applications of CNDs in drug delivery are also included in the review, along with the mechanisms that underlie their improved drug delivery capabilities. Additionally, it looks at controlled release methods, stability augmentation, and phytomedicine loading tactics onto CNDs. The potential of polymeric carbon nanodots in drug delivery is also covered, along with difficulties and prospective directions going forward, such as resolving toxicity and biocompatibility issues. In summary, the present review highlights the encouraging contribution of CNDs to the field of drug delivery, specifically in enhancing the potential of phytomedicine for therapeutic purposes.

Background: The utilization of nutrient supplementation as a tool to address issues such as iron and vitamin A deficiencies among children in Sub–Saharan Africa (SSA) has been beneficial. Nevertheless, there is a dearth of evidence on the effect of vitamin A and iron supplementation on infectious diseases in pre–school children in SSA. We aimed to assess the association of vitamin A and iron supplementation on the incidence of major clinical features of infectious diseases (diarrhea, cough, and fever) in children aged 6–59 months in Ghana. Methods: A cross–sectional study that utilized data from the Ghana Demographic and Health Survey, involving a sample size of 2,408 children. The collected data was analyzed using binomial logistic regression. Results: Dual supplementation with vitamin A and iron was associated with an increased risk of fever (OR: 1.68 95% CI: 1.22–2.32) and cough (OR: 1.43, 95% CI: 1.02–1.99) in children 6–59 months. Iron supplementation alone was associated with an increased risk of fever (OR: 1.32, 95% CI: 1.01–1.72) while Vitamin A alone significantly increased the risk of diarrhea (OR: 1.73, 95% CI: 1.03–2.92). Conclusions: Dual supplementation of vitamin A and iron is associated with an increase in childhood morbidity, endorsement of combined supplementation should be done with caution.

Prescribed fire is increasingly utilized for conservation and restoration goals, yet there is limited empirical evidence supporting its effectiveness in reducing wildfire-induced damages to valued resources of significant worth—whether natural, cultural, or economic. This study evaluates the efficacy of prescribed fire in reducing wildfire severity to LANDFIRE-defined vegetation classes and valued resources impacted by the 2017 West Mims event. Wildfire severity, measured using the differenced normalized burn ratio (dNBR) index, was highly heterogeneous both within and between vegetation classes; however, profound differences between treated and untreated areas were evident. The beneficial effects of prescribed fire were most pronounced within ca. two-years post-treatment but remained evident beyond three-years relative to untreated areas. For example, actively managed areas that were treated with prescribed fire just one month prior to the West Mims event exhibited an 88% reduction in mean wildfire severity relative to unmanaged areas. When post-treatment duration reached 38 months, mean wildfire severity remained 17% lower than untreated areas. Findings from this study reveal significant reductions in both average and peak wildfire severities in areas treated with frequent fire, supporting burn rotations of one to two years to balance fuel reduction and ecological conservation.

Pet nutrition awareness has risen enormously, with an increasing interest in homemade diets aimed at controlling food composition. Literature in this field is scarce, especially regarding the homemade diet’s long-term effects on pathological conditions. This prospective study encompassed 167 healthy and sick dogs subjected to a customized dietary plan based on homemade food. After an initial visit, dog owners provided questionnaires with follow-up data on their perceptions of physical signs changes or symptom modifications. 104 (62%) subjects maintained the homemade diet, while 63 reverted to their previous diet. The median follow-up was 14 months. Out of 31 healthy dogs that continued the nutritional plan, 70% exhibited improvements in their coat condition and 47% a decrease in evacuation frequency. Regarding weight loss goals, 67% of dogs achieved their target. The 67 pathological dogs that completed the follow-up were primarily affected by gastrointestinal and dermatological disorders. Dogs with chronic enteropathy improved their symptoms in 95% of the cases, subjects with dermatological pathologies in 83%, and patients presenting both disorders in 100%. These clinical results are promising. Personalized and well-balanced homemade diets noticeably enhanced the overall pet’s health, with almost a complete remission of symptoms in pathological dogs.

In recent years, the research community has been interested in members of the Acinetobacter genus mainly because of their role as causative agents of nosocomial infections. However, this rich-in-species genus has been proven to play a significant role in different biotechnological processes, such as bioremediation and fermented foods production. To partially fill the lack of information on Acinetobacter’s dualistic nature, in this review, based on literature data, we attempt to summarize the available information on the different roles the members of the genus play by considering their genetic constitution and metabolic properties. We found pieces of evidence of genetic divergence between the pathogenic and non-pathogenic species and strains, which can be explained by their high adaptability to the different ecological niches. In turn, this adaptability could result from intrinsic genetic variability due to mechanisms of horizontal genetic transfer, as well as high mutability determined by the expression of error-prone DNA polymerases. Yet, further studies are needed, especially whole-genome sequencing of non-pathogenic isolates, which for the moment are relatively scarce.

Brain-Computer Interface (BCI) processes and converts brain signals to provide 20 commands to output devices to carry out certain tasks. The main purpose of BCI is to replace or 21 restore missing or damaged functions of disabled people including neuromuscular disorders like 22 Amyotrophic Lateral Sclerosis (ALS), cerebral palsy, stroke, or spinal cord injury. Hence, BCI does 23 not use neuromuscular output pathways. Scientists have used several techniques like 24 Electroencephalography (EEG), intracortical, and Electrocorticographic (ECoG) to collect brain 25 signals which are used to control robotic arms, prosthetics, wheelchairs, and several other devices. 26 The non-invasive method of EEG is used for collecting and monitoring the signals of the brain. 27 Implementing EEG-based BCI technology in home automation systems may facilitate a wide range 28 of tasks for people with disabilities. It is important to assist and empower individuals with paralysis 29 to engage with existing home automation systems and gadgets in this particular situation. This 30 paper proposed a home security system to control a door and a light using EEG-based BCI. The 31 system prototype consists of the EMOTIV Insight™ headset, Raspberry PI 4, servo motor to 32 open/close the door, and LED. The system can be very helpful for disabled people including arm 33 amputees who cannot close/open doors or use remote control to turn on/turn off doors. The system 34 includes an application made in Flutter to receive notifications on the smartphone related to the 35 status of the door and the LEDs. The disabled person can control the door as well as the LED using 36 his/her brain signals detected by the EMOTIV Insight™ headset.

This study investigates the cytotoxicity profile of superparamagnetic Fe3O4-Ag decorated nanoparticles against human fibroblasts (HFF-1) and breast cancer cells (MCF-7). The nanoparticles underwent comprehensive characterization employing scanning electron microscopy (SEM), X-ray diffraction (XRD) analysis, X-ray photoelectron spectroscopy (XPS), and magnetic assays including hysteresis curves and zero-field-cooled (ZFC) plots. Notably, the nanoparticles exhibited superparamagnetic behavior as evidenced by magnetic studies. Remarkably, cytotoxicity assays demonstrated that both HFF-1 and MCF-7 cells maintained nearly 100% viability upon nanoparticle exposure. These results underscore the outstanding biocompatibility of Fe3O4/Ag decorated nanoparticles, suggesting their potential utility in biomedical applications such as drug delivery and magnetic targeting without inducing significant cytotoxic effects. This investigation advances our understanding of the behavior of nanostructured materials in biological settings and highlights their promising prospects in biomedicine.

Due to their remarkable intrinsic physical properties, carbon nanotubes (CNTs) can enhance mechanical properties and confer electrical and thermal conductivity to polymers currently investigated for use in advanced applications based on thermal management. An epoxy resin filled with varying concentrations of CNTs (up to 3 wt.%) was produced and experimentally characterized. The electrical percolation curve identified two critical filler concentrations: 0.5 wt%, near the electrical percolation threshold, suitable for exploring mechanical and piezoresistive properties, and 3 wt% for investigating thermo-electric properties due to the Joule effect with applied voltages ranging from 70V to 200V. Numerical models were developed using multiphysics simulation software. Once validated with experimental data, these models were used to investigate additional physical properties of the composites. Furthermore, a statistical approach based on the design of experiments (DoE) was employed to examine the influence of certain thermal parameters on the final performance of the materials. The purpose of this research is to promote the use of contemporary statistical and computational techniques alongside experimental methods to enhance understanding of materials science. New materials can be identified through these integrated approaches, or existing ones can be more thoroughly examined.

The severity and invasiveness of clinical outcomes from organic responses to trauma are influenced by individual, surgical and anesthetic factors. A stress response elicits neuroendocrine and immune reactions that may lead to multi-organ dysfunction. The degree of neuroinflammatory reflex activation from trauma can increase pro-inflammatory cytokine production, leading to endothelial dysfunction, glycocalyx damage, neutrophil activation and multisystem tissue destruction. A shift in patient treatment towards a neuroinflammatory perspective has prompted a new evaluation protocol for surgical patients, required to understand surgical pathogenesis and its link to chosen anesthesia-surgical methods. This study aims to provide a description of the mechanisms involved in the immune and neuroendocrine response and below is a summary of the main mediators, focusing on video laparoscopic surgeries, by examining its frequency usage by researchers. We performed a narrative review of data from databases like PubMed, MEDLINE, EMBASE, high-impact journals, and studies from the past decade that used inflammatory mediator analysis to compare anesthetic and surgical strategies. Results showed preference for certain biomarkers more sensitive to tissue trauma. These markers display distinct behaviors, temporal fluctuations and their dosages serve as a tool to measure and validate therapeutic approaches in perioperative systemic inflammation.

COVID-19 infection in high-risk populations is fatal and has a poor prognosis, necessitating a test to determine the protectiveness of immune response. Antibody testing is necessary to determine the body's immune response to COVID-19 infection and also vaccination strategies. Among the various methods available, chemiluminescent immunoassay (CLIA) test is more widely used and accessible to determine antibody levels. This study aimed to determine the protection level of S-RBD SARS-CoV-2 IgG using CLIA compared to Surrogate Virus Neutralization Test (SVNT). The population of this study comprised all healthcare professionals who experienced S-RBD SARS-CoV-2 IgG antibody level examinations. S-RBD SARS-CoV-2 IgG antibody levels were examined using CLIA and SVNT. The cut-off was determined using Receiver Operating Characteristic (ROC) curve and AUC (Area Under the Curve) measurements were evaluated. The result showed a strong positive correlation between S-RBD SARS-CoV-2 IgG CLIA and SVNT with a value of r=0.933 and p &lt;0.001. The value ≥37.29 BAU/mL was determined as the cut-off based on SVNT 30% inhibition level with sensitivity, specificity, positive, and negative predictive values of 96.5%, 90.9%, 96.5%, and 90.9%, respectively. A titer of antibodies greater than or equal to 37.29 BAU/mL with CLIA showed the presence of protective antibodies compared to SVNT.

Recent advancements in communication technology have catalyzed the widespread adoption of realistic content, with augmented reality (AR) emerging as a pivotal tool for seamlessly inte-grating virtual elements into real-world environments. In construction, architecture, and urban design, the integration of mixed reality (MR) technology enables rapid interior spatial mapping, providing clients with immersive experiences to envision their desires. The rapid advancement of MR devices, or devices that integrate MR capabilities, offers users numerous opportunities for enhanced entertainment experiences. However, to support designers at a high level of expertise, it is crucial to ensure the accuracy and reliability of the data provided by these devices. This study explores the potential of utilizing spatial mapping within various methodologies for surveying architectural interiors. The objective is to identify optimized spatial mapping procedures and determine the most effective applications for their use. Experiments were conducted to evaluate the interior survey performance, using HoloLens 2, iPhone 13 Pro for spatial mapping, and photogrammetry. The findings indicate that HoloLens 2 is most well-suited for the tasks exam-ined in the scope of experiments. Nonetheless, based on the acquired parameters, the author also proposes approaches to apply the other technologies in specific real-world scenarios.

The water-soluble vitamin - vitamin B12, also known as cobalamin, plays a crucial role in cellular metabolism, particularly in DNA synthesis, methylation, and mitochondrial functionality. Its de-ficiency can lead to hematological and neurological disorders, however the manifestation of these clinical outcomes is relatively late. It leads to difficulties in its early diagnosis, often resulting in the development of hidden subclinical cobalamin deficiency. Prolonged lack of vitamin B12 con-dition may have severe consequences including increased morbidity to neurological and cardio-vascular diseases. Beyond inadequate dietary intake, vitamin B12 deficiency might be caused by insufficient bioavailability, blood transport disruptions, or impaired cellular uptake and metabo-lism. Despite nearly 70 years of knowledge since its isolation and characterization, there are still gaps in understanding its metabolic pathways. Thus, this review aims to gather current knowledge about the crucial proteins necessary to efficiently process vitamin B12 in humans, presenting it as a multi-protein mediated network. Based on these findings, we also highlight new potential methods for supporting the evaluation of the risk of epidemiological consequences of vitamin B12 deficiency or clinical warnings of increased risk of vitamin B12 deficiency based on already testing targeting specific moonlighting proteins engaged in vitamin B12 metabolic path-ways.

This study explores the potential of piloting desalination plants in a systematic way to evaluate energy improvements in water desalination processes. Pilot tests with the latest generation of reverse osmosis membranes in large desalination plants are becoming increasingly common, as small-scale experiments are carried out on the operation of new generation membranes with better salt rejection and lower energy consumption. This approach aims to achieve optimal water quality standard at the lowest possible cost. In this way, pilot tests are being carried out before the decision is made whether to change the reverse osmosis membranes, as this represents a significant investment. By carrying out these tests, we hope to minimize the risk of making a mistake to ensure the best results in terms of energy consumption, operating costs and reducing the environmental impact while complying with the required water quality. In this case, the Pareto analysis is used to identify the two or three most significant causes whose treatment affects more than 80% of the possible energy improvement to be implemented. This article introduces a novel approach to studying a total of 180 desalination plants at the territorial level in the Canary Islands. The global scarcity of freshwater resources has greatly contributed to the development of desalination technologies, in which reverse osmosis is one of the most widely used and highly regarded methods.

Although mask-wearing is now widespread, knowledge of how to quantify or improve its performance remains surprisingly limited and largely based on empirical evidence. The objective of this study was to visualize the expiratory airflows from facemasks and evaluate aerosol transmission between two persons. Different visualization methods were explored, including the Schlieren optical system, laser/LED-particle imaging system, thermal camera, and vapor-SarGel system. The leakage flows and escaped aerosols were quantified using a hotwire anemometer and a particle counter, respectively. The results show that mask-wearing reduces the exhaled flow velocity from 2~4 m/s (with no facemask) to around 0.1 m/s, thus decreasing droplet transmission speeds. Cloth, surgical, and KN95 masks showed varying leakage flows at the nose top, sides, and chin. The leakage rate also differed between inhalation and exhalation. The neck gaiter has low filtration efficiency and high leakage fractions, providing low protection efficiency. There was considerable deposition in the mouth-nose area, as well as the neck, chin, and jaw, which heightened the risk of self-inoculation through spontaneous face-touching. A face shield plus surgical mask greatly reduced droplets on the head, neck, and face, indicating that double face coverings can be highly effective when a single mask is insufficient. The vapor-SarGel system provided a practical approach to study interpersonal transmission under varying close contact scenarios or with different face coverings.

Over the past 10 years, developments in precision oncology have led to a rise in the demand for predictive tools that assist the screening and stratification of patients for treatment. The possibility digitizing tissue slide images has led to the creation of artificial intelligence (AI) and machine learning algorithms in digital pathology, which could ultimately improve patient care. In this context, we assessed a variety of AI-based computational models for digital pathology. In this review, we described the latest AI applications in oncology, current challenges and future prospects.

Porins are crucial proteins located in the outer membrane that directly influence antimicrobial resistance mechanisms and virulence in bacteria. In this study, a porin gene (Vp-porin) was cloned in V. parahaemolyticus, and the function of Vp-porin on biological characteristics and virulence was investigated. The results of sequence analysis showed that Vp-porin is highly conserved in Vibrio spp. and the predicted 3D structure showed it could form a transmembrane channel with 20 β barrel. Membrane permeabilization provides evidence that membrane integrity of ∆Vp-porin was damaged and the sensitivity to tetracycline, polymyxin B, rifampicin and cephalothin of ∆Vp-porin obvious increased. In addition, loss of Vp-porin damaged motility due to down-regulated flagellar synthesis. Besides, ∆Vp-porin exhibited attenuated cytotoxicity to Tetra-hymena. The relative survival rate of Tetrahymena infection with ∆Vp-porin was 86%, which is much high than that with WT (49%). Take together, Vp-porin in V. parahaemolyticus plays various roles in biological characteristics in membrane integrity, antimicrobial resistance and motility, and contributes to virulence.

The oral microbiome is a diverse and complex ecosystem essential for maintaining oral and systemic health. Our study is the first to define the oral microbial community in Egyptian young adults and investigate the effects of natural antimicrobials on the oral microbiome. SuperMint (SM) is a proprietary blend of peppermint, Japanese mint, bergamot mint, and spearmint essential oils encapsulated in a tiny soft beadlet. This work aimed to evaluate the effects of SM beadlets on the oral microbiome. This study recruited twenty healthy participants. A baseline investigation of the oral microbiome of the selected participants was performed by collecting saliva and swab samples before treatment. Treatment included chewing four SM beadlets twice a day for 7 days, and then post-administration saliva and swab samples were collected at the end of treatment. The oral microbiome samples were analyzed by high-throughput amplicon sequencing of 16S rRNA gene fragments, and the community composition was determined. The results showed that the abundance of some microbial genera and families decreased after using SM, including Prevotella, Streptococcus, Neisseria, and Haemophilus. However, some genera showed inconsistent patterns. We also found that the subject's gender and SM usage were significantly associated with diverse microbial composition. The results suggest that SM treatment decreased the abundance of several bacteria associated with halitosis and periodontal diseases, such as Actinomyces and Streptococcus. Furthermore, Corynebacterium species increased while Streptococcus decreased after SM usage. More research is needed to fully understand the antimicrobial effects of mint oils and their potential applications in maintaining good oral health.

Chronic rhinosinusitis (CRS) is a complex inflammatory condition affecting the nasal and paranasal sinus mucosa. Gastroesophageal reflux disease (GERD) has been implicated as a potential exacerbating factor in CRS, but the specific endoscopic features of nasopharyngeal pathology in this context remain poorly understood. Background and Objectives: CRS is a multifactorial disease with various underlying etiologies, including inflammation, anatomical factors, and environmental triggers. While GERD has been suggested as a potential contributor to CRS, the specific endoscopic features indicative of nasopharyngeal pathology in CRS patients with GERD symptoms have not been clearly elucidated. Our aim is to identify specific endoscopic features of nasopharyngeal pathology in patients with CRS associated with GERD symptoms, and to propose a method for assessing the influence of GERD on the mucosal layer of the nose and nasopharynx. Materials and Methods: We conducted a cross-sectional observational study involving 521 adult patients presenting with symptoms suggestive of CRS. From this cohort, 95 patients with the highest scores on the Reflux Symptom Index (RSI) and Reflux Symptom Score-12 (RSS-12) questionnaires were selected as the main group. Endoscopic examinations were performed to assess the nasal and nasopharyngeal mucosa. Results: Our study revealed significant alterations in the nasopharyngeal mucosa of patients with CRS associated with GERD symptoms. Increased vascularity of the nasopharyngeal mucosa was observed in 91 patients (95.7%), while hypertrophy was noted in 83 patients (87.4%). Mucus was present in the nasopharynx of 77 patients (81.1%), exhibiting varying characteristics of color and consistency. Asymmetric hypertrophy of the oropharyngeal mucosa was noted in 62 patients (65.3%). Conclusions: We propose a method for assessing the influence of GERD on the mucosal layer of the nose and nasopharynx, which may aid in diagnostic and management decisions. Further research is warranted to explore the potential impact of GERD symptoms on the course and severity of CRS exacerbations.

Numerous investigations have consistently underscored the impact of societal stigma on the well-being of transgender individuals, encompassing both those experiencing gender dysphoria (transgender) and those who have undergone gender-affirming interventions (transsexual). The primary objective of the current research is to translate and adapt the Attitudes Toward Transgendered Individuals Scale into the Greek language. This scale specifically assesses stigma, excluding components such as discreteness and violence, and is tailored to evaluate individuals within the general populace. Employing confirmatory factor analysis (CFA) and assessing gender metric equivalence, the analyses yielded highly favourable outcomes, demonstrating excellent scale fit, reliability, and content validity, reflecting the robustness of the adapted tool for the Greek population.

Recently, the Portuguese parliament approved the decriminalisation of euthanasia for incurable illnesses. As the experiences of other countries show us, it will be a matter of time before mental disorders are addressed. Studying the phenomenon, particularly in vulnerable groups, in advance is essential for proper law drafting. Therefore, instruments which allow an objective assessment and comparison between groups must be available. This study aims to explore the validation of Faria's Attitude about Euthanasia scale in Portuguese older adult patients with mixed anxiety-depressive disorder. A sample of 114 older adult patients with mixed anxiety-depressive disorder collected by convenience in the Psychiatry Department of Senhora da Oliveira Hospital in Portugal was included. The pre-final version of the scale was tested in a small group with good results. The validity of the internal structure was analysed using exploratory factorial analysis. The internal consistency study verified reliability. For construct validity, we assessed the correlation with other validated scales measuring attitudes toward euthanasia, cognitive performance, personality and empathy. Attitude about euthanasia scale showed good internal consistency. One factor was retained in the principal component analysis. Significant correlations verified construct validity. The results support the scale's usefulness and validity. This study makes available a unique instrument to assess the overall tendency of the attitudes towards euthanasia in European-Portuguese, which can be used, for example, to compare Portuguese with Brazilian older adult psychiatric patients suffering from the same disorder. Furthermore, the adapted scale paves the way for other cross-cultural translations, adaptations, validations, and comparative analyses.

Coronary artery disease (CAD) and hypertension significantly contribute to cardiovascular morbidity and mortality. MicroRNAs (miRNAs) have recently emerged as promising biomarkers and therapeutic targets for these conditions. This systematic review conducts a thorough analysis of the literature, with a specific focus on investigating miRNA expression patterns in patients with CAD and hypertension. The review encompasses an unspecified number of eligible studies that employed a variety of patient demographics and research methodologies, resulting in diverse miRNA expression profiles. This review highlights the complex involvement of miRNAs in CAD and hypertension and the potential for advances in diagnostic and therapeutic strategies. Future research endeavours are imperative to validate these findings and elucidate the precise roles of miRNAs in disease progression, offering promising avenues for innovative diagnostic tools and targeted interventions.

Bi1-xSbx crystal is one of the best n-type thermoelectric materials below 200 K, but its weak mechanical strength hinders practical applications for deep refrigeration. Herein, we adopted the mechanical enhancement method of hot extrusion to investigate the comprehensive mechanical and thermoelectric properties of Bi0.905Sb0.095. It revealed that reducing the grain size of the matrix and increasing the extrusion ratio can improve the gain size uniformity and mechanical properties. While the thermoelectric performance depends on the texture, grain size, and local composition. The extruded sample prepared by ingot with the high extrusion ratio of 9:1 generated a high texture degree of (001) plane and small grain sizes between ~4-40 mm, which resulted in the high bending strength of Bi1-xSbx ~130 Mpa and a high-power factor of ~ 68 μW·cm-1·K-2@173 K, as well as the relative high figure of merit of 0.25@173K. This work highlights the importance of the uniform distribution of the grain size and the compositions for Bi1-xSbx, as well as the required universal key parameter for the hot-extrusion method.

Background Neisseria gonorrhoeae can acquire antimicrobial resistance (AMR) through horizontal gene transfer (HGT) from other Neisseria spp. such as commensals like Neisseria subflava. The prevalence of Neisseria subflava in the oropharynx is close to 100%. Low doses of antimicrobials in food could select for AMR in N. subflava, which could then be transferred to N. gonorrhoeae. In this study, we aimed to determine the lowest concentration of ciprofloxacin that can induce ciprofloxacin resistance (minimum selection concentration – MSC) in N. subflava. Methods Neisseria subflava Co000790/2 was serially passaged on GC agar plates containing ciprofloxacin concentrations ranging from 1:100- to 1:10,000-below its ciprofloxacin MIC (0.006 µg/ml) for six days. Results After 6 days of serial passaging at ciprofloxacin concentrations 1/100th of the MIC, 24 colonies emerged on the 0.06 µg/ml ciprofloxacin plate. Their ciprofloxacin MICs were between 0.19 to 0.25 µg/ml, and whole genome sequencing revealed a missense mutation T91I in the gyrA gene, which has previously been found to cause reduced susceptibility to fluoroquinolones. Conclusion The N. subflava MSCde novo was determined to be 0.06 ng/mL or 1:100 below the MIC. The implications of this finding are that the low concentrations of antibiotics found in certain environmental samples and even the food we eat may be able to select for ciprofloxacin resistance in N. subflava.

We describe a family of paraxial and quasi-monochromatic optical wave packets with finite energy and smoothly shaped amplitude in space and time that develops a singularity in the intensity when spatiotemporally focused by imparting a converging spherical wave front and a negative temporal chirp. This singular behavior upon ideal focusing is manifested in actual focusing with finite apertures and in media with high-order dispersion in an "exploding" behavior featuring indefinitely increasing concentration of the energy when opening the aperture radius, thus exercising a continuous control on the focal intensity and spatial and temporal resolution. These wave packets offer a new way of focusing that outperforms what can be achieved with standard Gaussian wave packets in terms of focal intensity and resolution, and new possibilities in applications where energy concentration and its control are crucial.

The prevalence of diabetes mellitus (DM) continues to rise globally, posing significant health challenges and burdens. This metabolic disorder, characterized by elevated blood glucose levels, contributes to various complications including cardiovascular diseases, kidney failure, and stroke. While conventional diabetic drugs exist, they often come with adverse side effects, necessitating the exploration of safer and more cost-effective alternatives, particularly from natural sources like medicinal plants. This study investigates the potential of Neocarya macrophylla (Nm) Ethyl acetate leaf fraction in managing DM. Utilizing a high-fat diet/streptozotocin-induced diabetic rat model, the study explores several key aspects including effective dose determination, biological activity, impact on body mass index (BMI), and complete blood count. Statistical analysis was conducted using GraphPad Prism version 9.1. The determination of the minimum effective dose reveals that 120mg/kg of Ethyl acetate fraction of Nm reduced the blood glucose by 28.9% after 4 hours. In addition, ethyl acetate fraction (120 mg/kg) possessed the most hypoglycemic activity compared to the other fractions of Nm. The Ethyl acetate fraction of Nm at 120mg/kg significantly reduced fasting blood glucose levels compared to the diabetic control(p=0.001) and slightly decreased BMI. Furthermore, the fraction demonstrated significant improvement in the parameters of some of the blood count indices such as red blood cells (p<0.001), total white cells (p<0.01) and platelet (p<0.001) compared to the diabetic control. These findings underscore the potential of Nm as a holistic therapeutic option for managing DM and its haematological complications. In conclusion, this study sheds light on the hypoglycemic potential of Nm Ethyl acetate leaf fraction, offering valuable insights into its effectiveness, safety, and broader physiological impacts. Further research into the mechanisms underlying its therapeutic effects is warranted to fully harness its therapeutic potential in diabetes management.

The rapid spread of African swine fever virus (ASFV), causing severe and often lethal disease in domestic pigs and Eurasian wild boar, continues to be a threat for pig populations and dependent industries. Despite scientific achievements that deepen our understanding of ASFV pathogenesis, alternative transmission routes for ASFV remain to be elucidated. We previously demonstrated the efficient transmission of ASFV from infected boars to naïve recipient gilts via artificial insemination, thereby highlighting the importance of surveillance in boar semen prior to shipment. Since accurate and reliable detection of even low amounts of ASFV in boar semen is key for disease prevention and control, we established a suitable diagnostic pipeline to efficiently detect ASFV genome in boar semen. Here, we assessed sensitivity, specificity and overall performance of various routine nucleic acid extraction kits as well as qPCR protocols in detecting ASFV genome in blood and semen of boars. Feasibility of respective kits and methods for future use in boar studs was also considered. Variability in sensitivity mostly concerned samples with low to very low amounts of ASFV genome. Ultimately, we defined a well-suited pipeline to precisely detect ASFV genome in boar semen, as early as 2 days post ASFV infection.

The Huichang fault Basin in Jiangxi Province exhibits high terrestrial heat flow values and a dense concentration of hot springs. However, existing research is limited to individual geothermal fields without a comprehensive basin-wide analysis of geothermal water origins. This study investigates the source of geothermal water across five representative geothermal fields using hydrochemical components and isotopic analyses. The findings indicate: (1) The hydrochemical type transitions from bicarbonate sulphate-sodium-calcium in the north to bicarbonate sodium-calcium in the south; (2) Major chemical components of the geothermal water derive from evaporative salt rocks (Cretaceous sand conglomerate cover) and siliceous formations (granite/metamorphic rock basement); (3) The recharge elevation for geothermal water is approximately 1 km, sourced from adjacent hills. The central basin's geothermal water has an apparent age of about 26,000 years, with δ¹³C indicating participation in deep circulation; (4) Thermal reservoir temperatures range from 74 to 113℃, with the lowest temperatures at the basin's edges (Anzishan and Shangjin geothermal fields) and higher temperatures towards the center, suggesting increased warming and a circulation depth of 1.5 to 2.5 km; (5) Simulation results of reverse water-rock interaction indicate that mineral precipitation-dissolution processes are governed by the basin's main thermal control structures. The geothermal fields are predominantly influenced by reduction alterations associated with granite and are closely linked to paleo-geothermal waters in the enclosed basin, with the formation of these fields heavily reliant on the heat production or conduction from radioactive elements in granite.

Developing clinically meaningful nanomedicines for cancer therapy requires the drugs to be effective, safe, simple, cheap, and easy to store. In the present work, we report that a simple cationic Fe(III)-rich salt of [FeIIICl(TMPPH2)][FeIIICl4]2 (Fe-TMPP) exhibits a superior anticancer performance toward a broad spectrum of cancer cell lines, including breast, colorectal cancer, liver, pancreatic, prostate, and gastric cancers, with half maximal inhibitory concentration (IC50) values in the range of 0.098−3.97 μM (0.066−2.68 μg mL−1), and comparable to the best-reported medicines. Fe-TMPP can forms stand-alone nanoparticles in water without the need for extra surface modification or organic-solvent-assisted antisolvent precipitation. Critically, Fe-TMPP is TME-responsive (TME = tumor microenvironment), and can only elicit its function in the TME with overexpressed H2O2, converting H2O2 to the cytotoxic •OH to oxidize the phospholipid of the cancer cell membrane, causing ferroptosis, a programmed cell death process of cancer cells.

This study introduces a compact Raman spectrometer with a 1064 nm excitation laser coupled with a fiber probe and an inexpensive motorized stage, offering a promising alternative to widely used Raman imaging instruments with 785 nm excitation lasers. The benefits of 1064 nm excitation for biomedical applications include further suppression of fluorescence background and deeper tissue penetration. The performance of the 1064 nm instrument in detecting cancer in human bladder resectates is demonstrated. Raman images with 1064 nm excitation were collected ex vivo from 10 human tumor and non-tumor bladder specimens and the results are compared to previously published Raman images with 785 nm excitation. K-Means cluster (KMC) analysis is used after pre-processing to identify Raman signatures of control, tumor, necrosis, and lipid-rich tissues. Hierarchical cluster analysis (HCA) groups the KMC centroids of all specimens as input. The tools for data processing and hyperspectral analysis were compiled in an open source Python library called SpectraMap (SpMap). In conclusion, the 1064 nm Raman system can differentiate between tumor and non-tumor bladder tissues in a similar way to 785 nm Raman spectroscopy. These findings hold promise for future clinical hyperspectral imaging.

Microgrids (MGs) have evolved as critical components of modern energy distribution networks, providing increased dependability, efficiency, and sustainability. Effective control strategies are essential for optimising MG operation and maintaining stability in the face of changing environmental and load conditions. Traditional rule-based control systems are extensively used due to their interpretability and simplicity. However, these strategies frequently lack the flexibility for complex and changing system dynamics. This paper provides a novel method called hybrid intelligent control for adaptive MG that integrates basic rule-based-control and deep learning techniques, including gated recurrent units (GRU), basic recurrent neural network (RNN) and long short-term memory (LSTM). The main target of this hybrid approach is to improve MG management performance by combining the strengths of basic rule-based systems and deep learning techniques. These deep learning techniques readily enhance and adapt control decisions based on historical data and domain-specific rules, leading to increasing system efficiency, stability, and resilience in adaptive MG. Our results show that the proposed method optimises MG operation, especially under demanding conditions such as variable renewable energy supply and unanticipated load fluctuations. This study investigates special RNN architectures and hyper-parameter optimization techniques with the aim of predicting power consumption and generation within the adaptive MG system. Our promising results show the highest-performing models indicating high accuracy and efficiency in power prediction. The finest-performing model accomplishes an R2 value close to 1, representing a strong correlation between predicted and actual power values.

Wind turbines (WTs) are clean a renewable energy source that has been growing in popularity in recent years. Gearboxes are complex, expensive, and critical components of WT, which are subject to high maintenance costs and several stresses including high loads and harsh environments that can lead to failure with significant downtime and financial losses. This paper focuses on the development of a digital twin-based approach for the modelling and simulation of WT gearboxes with the aim to improve their design, diagnosis, operation, and maintenance by providing insights into their behavior under different operating conditions. Pow-erful commercial computer-aided design tools (CAD) and computer-aided engineering (CAE) software are embedded into a computationally efficient multi-objective optimization framework (modeFrontier) with the aim of maximizing the power density, compactness, performance, and reliability of the WT gearbox. This entails minimizing its weight, volume, and maximum stresses and strains achieved without compromising efficiency. The 3D CAD model of the WT gearbox is carried out using SolidWorks, the Finite Element Analysis (FEA) to obtain the stresses and strains fields are modelled using Ansys Workbench, while the multibody kinematic and dynamic system is analyzed using MSC Adams. The method has been successfully applied to different case studies to find the optimal design and analyze the performance of the WT gearboxes. Simulation results can be used to identify potential failure modes and optimize gearbox design to improve performance, extend service life and relia-bility, thereby ensuring proper operation over its lifetime and reducing maintenance costs.

Procedural Sedation and Analgesia (PSA) in the Emergency Department (ED) presents a crucial aspect of emergency medicine, enabling the execution of painful or distressing procedures with minimal patient discomfort. This narrative review delineates the pharmacological framework, methodologies, and clinical considerations integral to optimizing PSA, with a particular focus on pediatric and geriatric populations. Through a comprehensive review and analysis of current practices, this work evaluates the pharmacokinetics and pharmacodynamics of widely utilized sedatives and analgesics, including propofol, ketamine, dexmedetomidine, fentanyl, midazolam, etomidate, nitrous oxide, and remimazolam. Special attention is dedicated to the selection criteria based on patient-specific risk factors, procedural requirements, and the management of potential adverse effects. The manuscript also explores innovative sedation techniques and the integration of new pharmacological agents, emphasizing evidence-based approaches to enhance patient safety and outcome. Results underscore the significance of tailored sedation strategies, especially for vulnerable groups such as pediatric and geriatric patients, highlighting the need for meticulous pre-procedural assessment and monitoring to mitigate risks. Conclusions drawn advocate for a nuanced application of PSA, guided by current evidence and clinical guidelines, to improve the quality of care in emergency settings. This research reinforces the imperative for ongoing education, skill development, and adaptation of new evidence into clinical practice to advance procedural sedation and analgesia in the ED.

This study employs Geographic Information System (GIS) techniques to evaluate the walkability of Onaizah City, which serves as a representative model for a Saudi city situated in the Qassim region, north of the capital city, Riyadh. The goal is to evaluate how easily pedestrians can navigate the street network and identify areas that excel or need improvement in terms of walkability. The results offer insights for planning and development supporting sustainable initiatives. Approach; The study involves integrating GIS data, such as street layouts, land use patterns, and demographic information. By analyzing factors like integration values and choice values within the street network the study gauges’ walkability aspects. Furthermore, correlation analysis explores how spatial arrangements, influence walkability. By studying the case of a medium-sized Saudi city, which can then be applied to several Saudi cities. Cost distance analysis and cost-back correlation calculations are employed to determine pedestrian routes. Findings and Discussion; Through GIS techniques the study effectively assesses the walkability of Onaizah City by identifying three walking paths with lengths of (1401m, 1283m, and 1507m). The study reveals pedestrians' actual city distance. Weighted criteria assign 40% to heritage sites, 30% to green spaces, and 15% each to mosques/commercial sites. Preserving access to heritage sites and incorporating areas enhance walkability. Conclusion: Study enhances walkability understanding, offering data-driven planning suggestions. Urban planners prioritize pedestrian connectivity, considering GIS effectiveness and physical environment importance. Findings support walkability research and sustainable urban development.

Background: This study explored maternal gut, breast milk and infant gut microbiomes as possi-ble mediators of the observed effects of a relaxation intervention which reduced maternal stress and promoted infant weight gain. Methods: A randomized controlled trial was conducted in healthy Chinese primiparous mother-infant pairs. Mothers were randomly assigned to interven-tion group (IG, listening to relaxation meditation) or control group (CG). Outcomes were differ-ences in microbiome composition and diversity in maternal gut, breast milk and infant gut at 1- and 8-weeks between IG and CG, assessed by 16S rRNA gene amplicon sequencing of fecal and breastmilk samples. Results: 38 mother-infant pairs were included in this analysis (IG=19, CG=19). Overall microbiome community structure in the maternal gut was significantly different between IG and CG at 1-week and the difference was more significant at 8 weeks (Bray-Curtis distance R2=0.04 vs. R2=0.13). Post-intervention, the α-diversity was significantly lower in IG breast milk (observed features: CG=295 vs. IG=255, p=0.032); the Bifidobacterium genera presented higher relative abundance. Diffuse midline gliomas are among the deadliest human cancers and have had little progress in treatment in the last 50 years. Cell cultures of these tumors have been developed recently, but it is unknown the degree to which such cultures retain the characteristics of the source tumors. DNA methylation profiling offers a powerful tool to look at genome-wide epigenetic changes that are biologically meaningful and can help assess the similarity of cultured tumor cells to their in vivo progenitors. Paraffinized diagnostic tissue from three diffuse intrinsic pontine gliomas with H3 K27M mutations was compared with subsequent passages of neurosphere cell cultures from those tumors. Each cell line was passaged 3-4 times and analyzed with DNA methylation arrays and standard algorithms that provided comparison of diagnostic classification and cluster analysis. All samples tested maintained high classifier scores and clustered within the reference group of H3 K27M-mutant diffuse midline gliomas. There was a gain of 1q in all cell lines, with two cell lines initially manifesting the gain of 1q only during culture. In vitro cell cultures of H3 K27M-mutant gliomas maintain high degrees of similarity in DNA methylation profiles to their source tumor, confirming their fidelity even with some chromosomal changes.

The mechanisms of formation of induced intrinsic and impurity radiative states, which consist of intrinsic and impurity electron-hole trapping centers in irradiated Ca2P2O7-Mn and Ca2P2O7 phosphates, were investigated using thermoactivation and vacuum-ultraviolet spectroscopy methods. These centers are excited at photon energies of 4.0 eV and 4.5 eV, which are within the matrix's transparency region. New radiative induced states at 3.06 eV and 2.92 eV are demonstrated to be generated upon the excitation of anions by photons with energies of 5.0 and 5.64 eV. This process is due to charge transfer from the ion to the impurities, specifically Mn^2+ O^2- Mn^2+ and the neighboring ion O^2- (P2O7)^4-. Furthermore, upon the excitation of matrix anions with photon energies exceeding the band gap (8.0 - 8.25 eV), electron trapping by impurities such as Mn^2+ and (P2O7)^4- ions results.

Antimicrobial Peptides (AMPs), as immune effectors synthesized by a variety of organisms, not only constitute a robust defense mechanism against a broad spectrum of pathogens in the host, but also show promising applications as effective antimicrobial agents. Notably, insects are significant reservoirs of natural AMPs. However, the complex array of variations in types, quantities, antimicrobial activities, production pathways of AMPs, and evolution of AMPs across insect species presents a significant challenge for immunity system understanding and AMP applications. This review covers insect AMP discoveries, classification, common properties, and mechanisms of action. Additionally, we summarize the types, quantities, and activities of im-mune-related AMPs in each model insect. We also conducted the first comprehensive investigation into the diversity, distribution, and evolution of 20 types of AMPs in model insects, employing phylogenetic analysis to describe their evolutionary relationships and shed light on conserved and distinctive AMP families. Furthermore, we summarize the regulatory pathways of AMP production through classical signaling pathways and additional pathways associated with Nitric Oxide, insulin-like signaling, and hormones. This review advances our understanding of AMP as guardians in insect immunity system and also unlocks a gateway to insect AMP resources, facilitating the use of AMPs to address food safety concerns.

Introduction: Bovine tuberculosis is a zoonotic disease of considerable impact, especially in livestock countries, which can cause human disease. Despite its importance, Colombia is a country in which few studies analyse its behaviour, and none develop maps using geographic information systems to characterise it. Objective: To describe the temporal-spatial distribution of bovine tuberculosis in Colombia between 2001 and 2019. Methods: Retrospective cross-sectional descriptive study, based on reports by the Colombian Agricultural Institute, of tuberculosis surveillance in cattle on farms in Colombia from 2001 to 2019. The data were converted into databases in Microsoft Access 365®, and multiple epidemiological maps were generated with the Kosmo RC1®3.0 software coupled to shape files of all the country's departments. Results: During the study period, 5,273 tuberculosis cases in bovines were identified in the different departments of Colombia (mean 278 cases/year). Regarding its temporal distribution, the number of cases varied from a maximum of 903 cases (17.12% of the total) in 2015 to a minimum of 0 between 2001 and 2004 and between 2017 and 2019 (between 2005 and 2016, the minimum was 46 cases, 0.87%). Conclusions: GIS is an essential tool for understanding the temporospatial behaviour of zoonotic diseases in Colombia, as is the case of bovine tuberculosis, with its potential implications for Human and One Health.

Objective: Aims to investigate the effects of empagliflozin (EMPA), a sodium-glucose co-transporter 2 (SGLT2) inhibitor, on renal fibrosis in male diabetic Sprague-Dawley (SD) rat model. Methods: Streptozotocin(STZ) were injected to establish diabetes model, EMPA (10 mg/kg·d) or normal saline (NS) were administered for four weeks. The changes in renal fibrosis-related biomarkers, include vimentin (VIM), collagen types I(Col-I), collagen types III(Col-III), fibroblast-specific protein 1(FSP-1), connective tissue growth factor (CTGF), fibronectin (FN) were evaluated. Renal pathology, serum glucose levels and urinary protein excretion were also assessed. Results: Biomarkers of renal fibrosis were significantly reduced in the EMPA group compared to those in the NS group after 4-week treatment, including VIM (519.84±83.37 vs. 663.27±91.84 ng/ml, P<0.05), Col-I (21.27±1.46 vs. 25.71±2.82 ng/ml , P<0.05) and Col-III (7.06±1.12 vs. 9.43±1.36 ng/ml, P<0.05), FSP-1 (4.38±0.81 vs. 5.70±0.78 ng/ml, P<0.05), and CTGF (1147±217 vs. 1556±235 pg/ml, P<0.05),respectively. Nevertheless, compared to normal control (NC) group, the levels of renal fibrosis-related biomarkers in the EMPA group showed no significant differences except for VIM, which was VIM (336.66±28.87 vs. 519.84±83.37 ng/mL, P<0.001). NS group rats showed notable renal tubular dilation and casts, EMPA group exhibited only mild tubular dilation. Conclusion: These findings suggest that empagliflozin reverse renal fibrosis potentially through inhibiting CTGF-mediated epithelial-mesenchymal transition.

Landsat and Sentinel-2 acquisitions are among the most widely used medium-resolution optical data adopted for terrestrial vegetation applications, such as land cover and land use mapping, vegetation condition and phenology monitoring, and disturbance and change mapping. When combined, both data archives provide over 40 years, and counting, of continuous and consistent observations. Although the spatio-temporal availability of both data archives is well-known at the scene level, information on the actual availability of cloud-, snow-, and shade-free observations at the pixel level is lacking and should be explored individually for each study to correctly parametrize subsequent analyses. However, data exploration is time and resource consuming, thus is rarely performed a-priori. Consequently, the spatio-temporal heterogeneity of usable data is often inadequately accounted for in the analysis design, risking ill-advised selection of algorithms and hypotheses, and thus inferior quality of final results. Here we present precomputed data on the daily 1982‑2023 availability of usable Landsat and Sentinel-2 acquisitions across the globe, sampled at a pixel-level in a regular 0.18°-point grid. The dataset comprises separate Landsat- and Sentinel‑2‑specific data records, and is accompanied by a pixel-specific growing season information to facilitate data exploration. The dataset was derived based on freely available 1982-2023 Landsat surface reflectance (Collection 2) and Sentinel-2 top-of-the-atmosphere reflectance (pre‑Collection-1 and Collection-1) scenes from 2015 through 2023, following the methodology developed in the recent study on data availability over Europe [1]. Growing season information was derived based on 2001‑2019 time series of the yearly 500 m MODIS land cover dynamics product (MCD12Q2; Collection 6) [1]. As such, the dataset presents a unique overview of the spatio-temporal availability of usable daily Landsat and Sentinel-2 data at the global scale, hence offering much needed a-priori information aiding identification of appropriate methods and challenges for terrestrial vegetation analyses at local to global scale.

Background and Objectives: Trichomonas tenax is a protozoan, participating in the human oral microflora. It is considered as a potential paradontopathogen. This microorganism is also reported in the respiratory tract. We aimed to analyze the available literature about the prevalence of Trichomonas tenax in the population affected by periodontal disease. Materials and Methods: Тhe searching with keywords: ,,Trichomonas tenax” and “periodontal diseases’’ in Scopus, PubMed and ScienceDirect databases was able to identify several systematic reviews and original articles until July 2023. All studies with suffering from periodontal disease people, mentioned year of publication, country, specified detection methods, total number of tested samples as well as the percentage of those infected with Trichomonas tenax were included. Irrelevant articles were excluded. Results: We found 137 studies, but only 64 studies about distribution of Trichomonas tenax in patients with gum disease underwent qualitative analysis. The highest number of studies have been conducted in Iran, Poland and Iraq. Different methods have been used to detect the unicellular organism, each with different specificity and sensitivity. Conclusions: Interest in Trichomonas tenax has grown considerably since 2000. Because of its association with periodontal disease, Trichomonas tenax role in the inflammatory process shouldn’t be overlooked.

Indole is a versatile pharmacophore widely distributed in bioactive natural products. This privileged scaffold has been found in a variety of molecules isolated from marine organisms such as algae and sponges. Among these, indole alkaloids represent one of the biggest, most promising family of compounds, having shown a wide range of pharmacological properties, including anti-inflammatory, antiviral and anticancer activities. The aim of this review is to show the current scenario of marine indole alkaloid derivatives, covering not only the most common chemical structures, but also new synthetic routes developed during the last years as well as promising therapeutic applications.

There is sufficient knowledge of the beneficial effect of warm-dry storage on breaking the morpho-logical dormancy of the chincuya (Annona purpurea Moc. & Sessé ex Dunal) seed embryo. Chincuya is an underutilized, tropical wild species native to Mexico, with potential as a biopesticide. The impact of warm-dry storage and gibberellic acid treatment on seed viability and germination was determined to improve that knowledge further. Also, the content and kinetics of fatty acids in chincuya seeds were profiled during warm-dry storage. Seed treatments included storage time (0, 3, 6, 9, and 12 months), seed imbibition for 72 h in gibberellic acid at 350 mg L-1, and distilled water im-bibition for 72 h for the control treatment. Seed viability was determined using the TTZ classic test. Imbibed seeds were germinated on paper towels and incubated at 30 °C during the day and 25 °C at night in a 12:12 h dark:light photoperiod. The lipid profile was obtained by gas chromatography. During the storage treatment, seeds stayed in darkness at 25 ± 3 °C. Results showed that high viability remained up to month 9 in storage when it significantly decreased. Freshly harvested seeds germi-nated at a statistically significantly lower rate (26.1 %) than six-month-old seeds (65.6 %). The profile of fatty acids breaks down as palmitic acid (33.41 %), stearic acid (7.72 %), and arachidic acid (1.30 %), for a total of 42.44 % of saturated fatty acids. The detected unsaturated fatty acids (52.15 %) were palmitoleic acid (1.52 %), oleic acid (43.37 %), and linoleic acid (7.24 %). No significant changes were detected in fatty acid content; however, the highest fatty acid content (92.84 %) occurred with the highest germination rate. Regression analysis showed significant content changes: palmitoleic and palmitic acid increased while oleic acid decreased during dry storage. Arachidic acid showed a slight downward trend. Also, fatty acid content significantly differed between intact seeds (unstored and unincubated) and latent seeds (unstored, incubated, and ungerminated). The total fatty acids content for latent seeds was significantly lower. These results show that Annona purpurea seeds present morphophysiological dormancy. Also, we document the highest germination rate reported (65.6 %) and report the presence of arachidic and palmitoleic acids in chincuya seeds.

Acinetobacter baumannii is a multidrug-resistant and invasive pathogen associated with the etiopathology of both an increasing number of nosocomial infections and of relevance to poultry production systems. Multidrug-resistant Acinetobacter baumannii has been reported in connection to severe challenges to clinical treatment, mostly due to an increase rate of resistance to carbapenems. Amid the possible strategies aiming to reduce the insurgence of antimicrobial resistance, phage therapy has gained particular importance for the treatment of bacterial infections. This review summarises the different phage-therapy approaches currently in use for multiple-drug resistant Acinetobacter baumannii, including single phage therapy, phage cocktails, phage -antibiotic combination therapy, phage coding Acinetobacter baumannii and the novel phage enzyme treatment. Although phage therapy represents a potential treatment solution for multidrug-resistant Acinetobacter baumannii, further research is needed to unravel some unanswered questions especially in regard to its in vivo applications, before possible routine clinical use.

According to the IPCC guidelines, CO₂ emissions from biomass should be excluded from the entire amount of CO₂ emissions when calculating CO₂ emissions and should be separately reported due to the “carbon neutrality.” Sewage sludge is one of the representative biomass fuels. And it is mixed with fossil fuels in terms of greenhouse gas reduction or is used as fuel to replace fossil fuels by itself. According to the results of this study, biomass contents of both the sewage sludge and the sewage sludge incineration exhaust gases did not amount to 100%. At present, in many countries(South Korea, Japan, and Germany), when calculating greenhouse gas emissions from sewage sludge incinerator, all CO₂ emissions from sewage sludge are judged to be biomass and only those greenhouse gas emissions that correspond to Non-CO₂ gases are calculated as greenhouse gas emissions. However, since, according our results, the content of sewage sludge is not 100% biomass, if CO₂ emissions are excluded according to the existing greenhouse gas emission calculation method, the amount of emissions may be underestimated. Therefore, to accurately calculate greenhouse gas emissions from sewage sludge incinerator, CO₂ emissions should be calculated in consideration of the fossil carbon contents of sewage sludge.

The development effect of water flooding in ultra-low permeability reservoir was poor due to its poor physical property and high shale content, the experimental study of air flooding which help to complement producing energy was carried out. Based on the Accelerating Rate Calorimeter experimental results, the crude oil of N block in L oilfield can produce low-temperature oxidation reaction, which was the basic condition of air flooding. Three groups of experiment natural cylinder core were designed for oil displacement, water flooding and air flooding were used respectively, and the relationship of differential pressure, oil recovery, injection capacity with injection volume was investigated. It has been showed on the result that the recovery efficiency improved sharply than water flooding, the effect of depressurization and augmented injection was obvious, and the air displacement was validation.

Traumatic brain injury (TBI) causes multiple cerebrovascular disruptions and oxidative stress, resulting in pathophysiological mechanisms. These are often accompanied by serious impairment in cerebral blood flow autoregulation, as well as neuronal and glial degeneration. Most currently available early antioxidant therapies lack sufficient efficacy. However, the application of a newly developed class of biologically compatible carbon-based nanoclusters, namely poly-ethylene-glycol-functionalized hydrophilic carbon clusters (PEG-HCCs), mitigate secondary injury after a TBI in early studies. In the present pilot study, we explore the efficacy of PEG-HCCs in an open blast rat model of TBI. Here we report on the neuroprotective efficacy of PEG-HCCs.

Three-dimensional (3D) models with improved biomimicry are essential to reduce animal experimentation and drive innovation in tissue engineering. In this study, we investigate the use of alginate-based materials as polymeric inks for 3D bioprinting of osteogenic models using human bone marrow stem/stromal cells (hBMSCs). A composite bioink incorporating alginate, nanohydroxyapatite (nHA), type I collagen (Col) and hBMSCs was developed and for extrude-based printing. Rheological tests perfomed on crosslinked hydrogels confirm the formation of solid-like structures, consistently indicating a superior storage modulus in relation to the loss modulus. The swelling behaviour analysis showed that the addition of Col and nHA into an alginate matrix can enhance the swelling rate of the resulting composite hydrogels, which maximizes cell proliferation within the structure. The LIVE/DEAD assay outcomes demonstrate that the inclusion of nHA and Col did not detrimentally affect the viability of hBMSCs over seven days post-printing. PrestoBlueTM revealed a higher hBMSCs viability in the alginate-nHA-Col hydrogel compared to the remaining groups. Gene expression analysis revealed that alginate-nHA-col bioink favoured a higher expression of osteogenic markers, including secreted phosphoprotein-1 (SPP1) and collagen type 1 alpha 2 chain (COL1A2), in hBMSCs after 14 days, indicating the pro-osteogenic differentiation potential of the hydrogel. This study demonstrates that the incorporation of nHA and Col into alginate enhances osteogenic potential and therefore provides a bioprinted model to systematically study osteogenesis and the early stages of tissue maturation in vitro.

Background: There is a notable lack of protocols addressing extubation techniques in transoral robotic surgery (TORS) for obstructive sleep apnea (OSA). Methods. This retrospective cohort study enrolled patients who underwent TORS for OSA between March 2015 and December 2021 and were managed with different extubation approaches. The patients were divided into two groups: high-flow nasal cannula (HFNC) therapy and conventional oxygen therapy. The use of an airway exchange catheter (AEC) was investigated. Results: The only application of HFNC use versus conventional oxygen therapy led to a statistical reduction of extubation time (p=0.024); length of stay in the intensive care unit (ICU) and the episodes of desaturation below 95% were reduced, but data are non-statically significant. Similarly, the application of an AEC led to a reduction of extubation time in hours (p=0.008) and length of stay in the ICU (p=0.024). Conclusions: In patients with OSA who underwent TORS, the use of HFNC, with or without an AEC, resulted in a significant reduction in extubation time without major adverse events. Additionally, HFNC utilization may decrease desaturation episodes during extubation. Based on the findings of this preliminary investigation, the combination of HFNC and AEC emerges as a promising strategy for enhancing the safety and efficacy of extubation protocols in this patient population.

The presence of various pesticides in natural streams and wastewater is a significant environmental issue due to their high toxicity, which causes harmful consequences even at low quantities. One cost-effective method to remove these pollutants from water could be through adsorption using inexpensive, easily obtained adsorbent - biochar. This study demonstrates the effectiveness of using biochar produced from wheat grains to eliminate alachlor from water. The sorption properties of the biochar and the likely removal mechanisms have been defined. The study found that the biochar removed 76-94% of alachlor, depending on the initial concentration of the pesticide in water. The pseudo-second-order kinetic model (R² = 0.999) and the Langmuir isotherm model (R² = 0.996) effectively characterized the elimination of alachlor by wheat grains biochar. The analyzed biochar, with its micropores and various surface functional groups, can effectively adsorb alachlor and trap it within its structure.

The peptide hormone kisspeptin attenuates liver steatosis, metabolic dysfunction-associated stea-tohepatitis (MASH) and fibrosis in mouse models, by signaling via the kisspeptin 1 receptor (KISS1R). However, whether kisspeptin impacts fibrogenesis in the human liver is not known. We investigated the impact of a potent kisspeptin analog (KPA) on fibrogenesis using human precision cut liver slices (hPCLS) from fibrotic livers from male patients, in human hepatic stellate cells (HSCs), LX-2 and in primary mouse HSCs. In hPCLS, 48 h and 72 h of KPA (3 nM, 100 nM) treatment decreased collagen secretion, and lowered expression of fibrogenic and inflammatory markers. Immunohistochemical studies revealed that KISS1R is expressed and localized to HSCs in MASH/fibrotic livers. In HSCs, KPA treatment reduced transforming growth factor beta (TGFb)-induced expression of fibrogenic and inflammatory markers, in addition to decreasing TGFb induced collagen secretion, cell migration, proliferation and colony formation. Mechanistically, KISS1R signaling downregulated TGFb signaling by decreasing SMAD2/3 phosphorylation, via activation of protein-phosphatases PP2A, that dehosphorylates SMAD 2/3. This study revealed for the first time that kisspeptin reverses human hepatic fibrogenesis thus identifying it as a new therapeutic target to treat hepatic fibrosis.
