Mono-Specific Data Deficient Saltmarsh Species Has Climate Relevant Carbon Stocks From the East Coast of India

Despite saltmarshes being considered as nature-based solutions (NbS) towards climate change mitigation, India’s saltmarsh ecosystems are least studied. This study quantifies the seasonal variation in carbon stocks mono-specific saltmarsh species (Porteresia coarctata) and its potential to play an important role in India’s climate change mitigation plans. Seasonal (pre-and post-monsoon) sampling of P. coarctata surface water, biomass and 30 cm sediment core was collected across four locations on the east coast of India to quantify sediment dry bulk density (DBD), organic matter (OM%), organic carbon (Corg%), Corg stocks of sediment and biomass, total carbon (C%) and nitrogen (N%) and stable isotopes of δ13C and δ15N. In general, the sediment DBD, OM and Corg of P. coarctata meadows was higher in post-monsoon and was influenced by salinity and pH changes. Isotope (δ¹³C) modelling of various sediment C sources suggested, particulate organic matter (POM) contribution was highest (0.04-0.79%) followed by P. coarctata (0.01–052%) and other macrophytes across our study locations. The seasonal variation of δ¹³C showed increased contribution of marine derived particulate and dissolved organic matter into P. coarctata meadows in post-monsoon season. Heavier δ¹⁵N values were observed in post-monsoon season suggesting anthropogenic input, that was utilized by P. coarctata to increase its above and below-ground biomass and shoot density. The combined ecosystem (30 cm sediment + biomass) Corg stocks of P. coarctata was 1.7-fold higher in post-monsoon (4021.20± 917 Mg C) compared to pre-monsoon (2297.36 ±647 Mg C) season among the four locations with a sediment Corg contribution >70%. Based on the International Panel for Climate Change Tier II assessment the P. coarctata meadows (443 ha) can help in avoiding the pre and post-monsoon emissions of 8431.34 and 14757.84 Mg CO₂ respectively. The combined price of the total CO₂ equivalent stored in P. coarctata meadows in pre- and post-monsoon is US$ 14.50 and US$ 25.38 million respectively. Further studies quantifying the NbS potential of P. coarctata mono-specific and mixed meadows of India’s coast is needed along with integration of saltmarsh ecosystems into India’s National Action Plan on Climate Change.

Shigellosis is a pathological condition that affects the digestive system to possibly cause diarrhoea. Shigella species, which are responsible for this disease, are highly contagious and spread through contaminated food and water. The increasing development of resistance by Shigella species necessitate the urgent need to search for new therapies against shigellosis causing diarrhoea. The scientific validation of medicinal plants, such as Diospyros gilletii, which is used for the traditional treatment of diarrhoeal conditions is worthwhile. The present study aims to investigate the antibacterial activity of extracts from D. gilletii against selected Shigella species. Extracts from D. gilletii stem bark were prepared by maceration using various solvents. An-tibacterial activity of D. gilletii extracts was evaluated on Shigella dysenteriae, S. flexneri, S. boydii, and S. sonnei using a microdilution method, whereas cytotoxicity test was performed on Vero and Raw cells using resazurin-based colorimetric assay. Bacterial membrane permeability studies were evaluated using pro-pidium iodide (PI) and 1-N-phenylnaphthylamine (NPN) uptake assays, whereas inhibition and eradica-tion tests on bacterial biofilms were carried out by spectrophotometry. As a result, methanol, ethanol and hydroethanol extracts of D. gilletii inhibited the growth of S. boydii, S. flexneri and S. sonnei with MIC values ranging from 125 to 500 µg/mL, without toxicity to Vero and Raw cells. Time kill kinetics revealed bacteri-cidal orientation at 2MIC and 4MIC, and bacteriostatic outcome at 1/2MIC. The mechanistic basis of anti-bacterial action revealed that D. gilletii extracts inhibited and eradicated Shigella biofilms, and promoted the accumulation of NPN and PI within the inner and outer membrane of bacteria to increase membrane per-meability, thereby causing membrane damage. This novel contribution toward the antibacterial and mech-anisms of action of D. gilletii extracts against Shigella species substantiate the use of this plant in the tradi-tional treatment of infectious diarrhoea.

Cirsium japonicum, a traditional herb, exhibits significant anti-inflammatory activity; however, the main components and potential mechanisms of C. japonicum remain unclear. The aim of this study is to investigate the anti-inflammatory mechanism of Cirsium japonicum through network pharmacology and cellular experiments. The effective components and potential targets for anti-inflammatory activity of C. japonicum were identified using traditional Chinese medicine systematic pharmacology database, TCMSP analysis platform, and GeneCards database. The drug-component-target-disease network diagram was constructed using Cytoscape3.8.0 software, while the protein interaction network diagram was created using STRING database and Cytoscape3.8.0 software. Gene ontology (GO) enrichment and KEGG pathway enrichment analysis were carried out using DAVID database. Molecular docking between key targets and active components was constructed with AutoDock software to determine the best binding target. Results revealed that 14 active components of C. japonicum targeted 171 anti-inflammatory proteins. GO function enrichment analysis yielded 173 items, while KEGG pathway enrichment analysis identified 48 signaling pathways related to inflammation regulation. Molecular docking showed that strong affinity between sitosterol, stigmasterol, and other components with key targets such as peroxisome proliferator-activated receptor α recombinant protein (PPARA) and cyclooxygenase-2 (PTGS2). Vanillin, one active ingredient of C. japonicum, inhibited the release of lipopolysaccharide (LPS)-induced inflammatory factors in RAW264.7 cells. These findings suggest that C. japonicum may exert its anti-inflammatory effects by modulating the phosphatidylinositol 3-kinase (PI3K)/ protein kinase B (Akt) signal pathway (PI3K-Akt) and apoptin signal pathway, highlighting the multi-component, multi-target, and multi-channel molecular mechanism underlying its anti-inflammatory properties. Finally, the anti-inflammatory effect of vanillin, an effective component in C. japonicum, was verified by cell experiments. This study provides a new understanding of the pharmacological mechanisms of C. japonicum in treatment of inflammatory conditions.

Hypophthalmichthys nobilis are widely distributed in the Yangtze River basin and its related lakes. They are an important economic fish species and are a famous cultured species known as the "Four Famous Domestic Fishes" in China. Currently, with the fishing ban in the Yangtze River basin, fishing for H. nobilis in natural water bodies of the Yangtze River basin has been completely prohibited. In order to identify the sources of H. nobilis appearing in the market, further control and accountability, it is necessary to trace the sources of H. nobilis in the Yangtze River basin and its related water bodies. Therefore, this study identified and traced different sources of H. nobilis through muscle element fingerprint analysis (EFA). The results show that H. nobilis from different stations have characteristic element compositions. The characteristic element of H. nobilis from Wuhan (WH) is Pb, which is significantly higher than that in other stations; the characteristic element from Anqing (AQ) is Hg, which is significantly higher than that in other stations; and the characteristic element from Taihu (TH) is Al, which is significantly higher than that in other water areas. Multivariate analysis selected different spatial distribution patterns of 4 discriminant elements ratios (Pb/Ca, Cr/Ca, Na/Ca, Al/Ca) in the muscle of H. nobilis in the Yangtze River basin and its related lakes. The study suggests that the screened discriminative elements can be used to visually distinguish different sources of H. nobilis, and to quickly trace and verify the origin of newly emerging samples. Therefore, the use of selected discriminant element fingerprint features to trace the origin of new samples has been proven to be feasible. By further discriminating and verifying the muscle element fingerprints of new samples, the discrimination rate is high. Therefore, Multivariate analysis for muscle element fingerprint can be used for origin traceability of samples of unknown origin in market supervision.

Heat stress is a serious threat to tomato production. The ultrastructure and element mapping of tomato chloroplast were investigated under different nitrogen levels to explore the regulatory effect of nitrogen on tomato chloroplast under heat stress. Experiments design two temperature treatments (25°C/15°C, as control, 40°C/30°C, as severe high-temperature, SHT ) and four nitrogen levels (0.0 kg hm-2, N1; 187.5 kg hm-2, N2; 250 kg hm-2, N3, as recommended nitrogen-application; 312.5kg hm-2 , N4) for a duration of 5 days in artificial climate chamber. The results showed that heat stress led to obvious damage to the ultrastructure of tomato chloroplast, including thylakoid stacking disorder, granum destruction, and starch grain aggregation and degradation. With the increase of nitrogen level, the ratio of long and short axes of tomato chloroplasts increased, the differentiation of chloroplasts was strengthened, the number of plastoglobuli was increased and aggregated, and the degree of damage to the ultrastructure of chloroplasts was reduced. The regulatory effect of nitrogen on the ultrastructure of tomato chloroplasts under heat stress was mainly reflected in the phosphorylation degradation process of starch granules in chloroplasts. There are obvious segmentation lines of Ca, P and Cl on both sides of the inner and outer chloroplasts, and enriched in the chloroplasts, which can outline the boundary of the chloroplasts. Element mapping analysis revealed that heat stress significantly increased the contents of calcium, phosphorus and chlorine in the chloroplast of tomato leaves, while the contents of potassium was decreased. The application of nitrogen fertilizer could significantly regulate the element distribution of tomato chloroplast under heat stress, especially the regulation of phosphorus, chlorine, calcium, magnesium. These results provide new insight into the role of nitrogen using TEM-EDX mapping in the protection of tomato chloroplast under heat stress and the mechanism of heat stress tolerance in plants.

Mulberry (Morus alba L), a significant fruit tree crop, requires magnesium (Mg) for its optimal growth and productivity. Nonetheless, our understanding of the molecular basis underlying magnesium stress tolerance in mulberry plants remains unexplored. In our previous study, we identified several differential candidate genes associated with Mg homeostasis via transcriptome analysis including xyloglucan endotransglucosylase/hydrolase (XTH) gene family. The XTH gene family is crucial for plant cell wall reconstruction and stress responses. These genes have been identified and thoroughly investigated in various plant species. However, there is no research pertaining to XTH genes within the M. alba plant. This research systematically examined the M. alba XTH (MaXTH) gene family at the genomic level using a bioinformatic approach. In total, 22 MaXTH genes were discovered and contained the Glyco_hydro_16 and XET_C conserved domains. The MaXTH’s were categorized into five distinct groups by their phylogenetic relationships. The gene structure possesses four exons and three introns. Furthermore, the MaXTH gene promoter analysis reveals a plethora of cis-regulatory elements; mainly stress responsiveness, growth and development, light responsiveness, and phytohormone responsiveness. GO analysis indicated that MaXTHs encode proteins that exhibit xyloglucan xyloglucosyl transferase and hydrolase activities in addition cell wall biogenesis as well as xyloglucan and carbohydrate metabolic processes. Moreover, a synteny analysis unveiled an evolutionary relationship between the XTH genes in M. alba and those in three other species: A. thaliana, P. trichocarpa, and Zea mays. Expression profiles from RNA-Seq data displayed distinct expression patterns of XTH genes in M. alba leaf tissue during Mg treatments. Real-time quantitative PCR analysis confirmed the expression of the MaXTH genes in Mg stress response. Overall, this research enhances our understanding of the characteristics of MaXTH gene family members and lays the foundation for future functional genomic study in M. alba.

Molecules sourced from marine environments hold immense promise for the development of novel therapeutic drugs, owing to their distinctive chemical compositions and valuable medicinal attributes. Notably, Talarolide A and Talaropeptides A-D have gained recent attention as potential candidates for pharmaceutical applications. This study aims to explore the chemical reactivity of Talarolide A and Talaropeptides A-D through the application of molecular modeling and computational chemistry techniques, specifically employing Conceptual Density Functional Theory (CDFT). By investigating their chemical behaviors, the study seeks to contribute to the understanding of the potential pharmacological uses of these marine-derived compounds. The investigation involves the utilization of molecular modeling and computational chemistry tools, with a specific focus on Conceptual DFT. These techniques are applied to examine the chemical reactivity of Talarolide A and Talaropeptides A-D, shedding light on their potential for interacting with various biological targets. The research reveals insights into the chemical reactivity of Talarolide A and Talaropeptides A-D, uncovering active reaction sites and potential biological targets. Talarolide A demonstrates robust anti-inflammatory properties and antitumor activity against diverse cancer cell lines, while Talaropeptide A displays antimicrobial efficacy against both bacteria and fungi. The study underscores the significance of comprehending the chemical reactivity of natural marine products for advancing pharmaceutical development. By employing computational tools such as Conceptual DFT, this research uncovers crucial information about the active sites and potential targets of Talarolide A and Talaropeptides A-D. Mono-crop plantation widely found in the world and dominated by few species such as Eucalyptus, Pinus and Tectona. While they lack diversified product, negative to environmental service and limited forest product for local use. The experiment was designed to evaluate the impact of mixed plantings on the early growth performance of the E. globulus tree species. The treatments included one monoculture of E. globulus and a combination of the plant with three other native and one foreign tree species. In four consecutive years after plantation, height, root collar diameter, DBH and leaf/branch count were measured. Following two years of out planting, the survival rates of seedlings were determined. Destructive sampling was applied to measure biomass of E. globulus. The growth performance of E. globulus in mixed plantations was compared to that of trees produced in pure plantations. The findings showed that there were no significant variations in growth parameters across the treatments. Following two years of out planting, the survival rates of seedlings were determined. The growth performance of E. globulus in mixed plantations was compared to that of trees produced in pure plantations. The findings showed that there were no significant variations in growth parameters across the treatments. The survival rate of E. globulus was good, ranging from 75 to 96.8%. Similarly, the growth performance of E. globulus's root collar diameter was larger even if the trial did not reveal a significant difference between the treatments for height. In comparison to other treatments, the one that included junipers and E. globulus had a higher mean height throughout the course of three years (1.28, 2.53, and 6.12 meters, respectively), as well as a lower RCD in the first and third years (1.3 and 5.34 cm). At year four, E. globulus attain higher in Height and DBH with high growth rate in mixed plantations compared to pure stands. The relative yield of E. globulus in pure stands compare to mixed stands greater than 1. Above and below ground biomass and carbon density of E, globulus is comparable in all treatment. Overall, the study's findings confirm that planting E. globulus improves its mingling with the examined tree species. Such studies ought to be conducted again over a longer time frame in order to examine the effects at a later stage and recommend technologies for broader implementation.

Utilising the metabolic processes (urease activity) of certain microorganisms that result in the precipitation of calcium carbonate (CaCO₃) as a sustainable method for manufacturing self-healing cementitious materials for use in the construction, soil stabilization, and wind-induced erosion industries has garnered considerable attention over the last twenty years. Despite extensive efforts for experimental characterization of the effect of numerous influential factors such as the bacteria type, nutrition type and quantity, and environmental conditions governing this phenomenon, computational modeling of this biochemical process has not advanced significantly due to its complexity and intertwined involved parameters. Among these parameters, pH is of special significance since the initial pH level has an immediate effect on the bacteria's urease activity, which then influences the rate at which calcium carbonate precipitates. Furthermore, during the CaCO₃ precipitation process, pH changes due to the generation of byproducts such as ammonium, which alters the velocity of the bacteria's urease activity continuously.The present study proposes a computational model for calcium carbonate precipitation by urease activity of Sporosarcina pasteurii using COMSOL Multiphysics^®. The theoretical background on governing parameters and chemical reactions involved in the process are discussed. The model takes into account the impact of calcium and urea concentrations, as well as the initial pH level and pH variations caused by the production of by-products during the process. The capability of the model to foresee CaCO3 concentration and ultimate pH level is evaluated by comparing the computational outcomes with empirical data obtained from established literature sources. The influence of the initial pH of the environment on the pH variation of the system during the precipitation process was simulated and compared to actual data available in the literature, suggesting that the model can accurately predict the kinetic. Finally, a parametric analysis is performed to identify the ideal initial environmental pH for calcium carbonate precipitation by the S. pasteurii bacterium.

Prophylactic Therapy with Emicizumab in Patients with Hemophilia A: Benefits and Challenges

This study presents a transversal investigation, that we performed at Fundeni hospital, into the therapeutic benefits and efficacy of Emicizumab, a non-factor therapy, in the context of hemophilia A. Ten patients diagnosed with hemophilia A were closely monitored, using clinical and laboratory resources, during the course of Emicizumab treatment, with an average of 12.8 months. Among these patients, six exhibited anti-factor VIII inhibitors, changing the medical approach and adding complexity to their clinical profiles. A comprehensive approach was adopted to assess the coagulation status of patients under Emicizumab therapy. The study employed several key coagulation monitoring tools, of which including thrombin generation time (TGT) and thrombelastography (TEG). These methodologies proved highly valuable results in evaluating the patients' coagulation profiles during the treatment regimen. Additionally, traditionally coagulation assays were utilized to gain a comprehensive understanding of the overall coagulation dynamics. Noteworthy, impressive outcomes emerged from the study. During prophylaxis with Emicizumab all patients had experienced a reduced number of bleeding events. Moreover, a subset of these patients underwent major surgical procedures (orthopedic joint replacements) with successful outcomes. These findings underscore the potential of Emicizumab therapy as an effective option for hemophilia A patients, including those that presented a prompt production of anti-factor VIII inhibitors. As for the outcomes, this transversal study sheds some light on the positive impact of Emicizumab therapy in addressing the coagulation challenges posed by hemophilia A. The utilization of global coagulation assays, such as thrombin generation time, thrombelastography and combined with traditional coagulation assays, used as monitoring tools highlights their significance once more in evaluating the therapeutic response. This research contributes by providing physicians with valuable insights to the field, offering a potential avenue for improved patients’ care and treatment strategies that translate in enhanced quality of life for hemophilia A patients undergoing Emicizumab therapy.
